The spider mite, Tetranychus pueraricola (Ehara & Gotoh; Acari: Tetranychidae), is a serious pest in agriculture and horticulture. Application of chemical pesticides is the main mode of this pest control. Due to pesticide residues and resistance-induced resurgence of pests, there is a need to discover alternatives for spider mite management. GC16 comprises a mixture of calcium chloride (CaCl2, 45%) and lecithin (55%), which was recently found to have acaricidal properties. We evaluated the sublethal effects of GC16 on T. pueraricola using life table and enzyme [catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), carboxylesterase (CarE), glutathione S-transferases (GST), and Ca2+-ATPase (Ca2+-ATP)] activity assays. The results showed that fecundity of T. pueraricola increased at LC30 but decreased at LC50 of GC16. The intrinsic rate of increase (r) of T. pueraricola decreased under the LC30 and LC50 of GC16. GC16 concentration and exposure time significantly influenced the activities of CAT, POD, CarE, GST, and Ca2+-ATP in adult mites. Twelve hours later after the treatment, GST and Ca2+-ATP activities were significantly inhibited by LC30 but enhanced by LC50. Moreover, the demographic parameter r and enzyme activities were negatively correlated. In sum, sublethal amounts of GC16 had an adverse effect on mites, and there was a trade-off between developmental performance and physiological enzyme activity of mites under GC16 stress, and GC16 showed an acaricidal potential for T. pueraricola. This work provides guidance for the application of GC16 to control T. pueraricola.

Lead halide perovskite nanocrystals (PNCs) are attractive in light-emitting applications with their extraordinary photoluminescent property. However, the inherent instability of mixed Br/I ions poses a major hurdle to the practical application of yellow-red emitting PNCs. In this work, we report the growth of crystalline ZnO on the surface of CsPb(Br/I)3 nanocrystals through a ligand-mediated in situ surface reaction route, which forms monodispersed CsPb(Br/I)3/ZnO heterostructure nanocrystals (PZNCs). The unique heterostructure endows the PZNCs with largely enhanced stability against air, moisture, and polar solvents. Notably, the red-emitting PZNCs exhibit high color quality and superior stability compared with classical CsPb(Br/I)3 PNCs. The PZNCs retain 95% of initial luminescence after 500 h of illumination, and the PZNC-converted red-light LEDs show no obvious change in the peak position and light intensity after continuous operation for 100 h, demonstrating the promising prospect of the materials in light-emitting applications.

This study aimed to investigate the effect of norisopoldine (NOR) on acute lung injury in septic mice. Lipopolysaccharide (LPS) was used to establish sepsis induced acute lung injury (ALI) in mice. The dry and wet weight of mice lung was detected, and the pathological changes of lung were observed by hematoxylin and eosin (H&E) staining. Bronchoalveolar lavage fluid (BALF) was detected. Inflammatory factors in BALF were detected by enzyme-linked immunosorbent assay (ELISA). The polarization of macrophages in lung tissue was detected by flow cytometry. The markers of M1 and M2 macrophages were detected by RT-PCR. LPS induced RAW264.7 cells were treated with NOR. Inflammatory response, macrophage polarization, glycolysis, and M2 pyruvate kinase (PKM2)/hypoxia inducible factor-1α (HIF-1α)/peroxisome proliferator activated receptor-γ co-activator 1-α (PGC-1α) signaling pathway were detected. NOR could effectively alleviate sepsis induced ALI, and reduce the number of total cells, total protein concentration, neutrophils, macrophages in BALF. NOR decreased the level of inflammatory factors and promoted macrophages from M1 to M2 type in vivo and vitro. Moreover, NOR could activated PKM2, and inhibited PKM2 from cytoplasm to nuclear, attenuated HIF-1α expression, and increased PGC-1α and peroxisome proliferator-activated receptor (PPAR)-γ expression. In addition, NOR inhibited glycolysis and promoted oxidative phosphorylation in RAW264.7 cells. Furthermore, PKM2 inhibitors could reverse the effect of NOR on PKM2/HIF-1α/PGC-1α signaling pathway in RAW264.7 cells. NOR alleviated sepsis induced AIL in mice, inhibited the inflammatory response, promote M2 polarization of macrophages through regulating PKM2/HIF-1α/PGC-1α signaling pathway.

BackgroundDrug-coated balloon (DCB) angioplasty has been studied for reducing the occurrence of restenosis after treatment for intracranial atherosclerotic stenosis (ICAS), but no comparison has been published of the use of drug-coated and non-coated balloons in angioplasty for ICAS. We aim to compare the safety and efficacy of DCB angioplasty with conventional balloon (CB) angioplasty for the treatment of symptomatic ICAS.MethodsOne hundred cases with symptomatic ICAS treated with DCB (n=49) and CB (n=51) angioplasty were retrospectively analyzed. 1:1 propensity score matching (PSM) was completed to eliminate bias in the patients selected for further analysis. The periprocedural events and follow-up outcomes between the two groups were compared.ResultsThere were 32 cases in each group after PSM. Technical success (ConclusionsCompared with CB angioplasty, DCB angioplasty can effectively reduce the incidence of restenosis. Further studies are needed to validate the role of DCB angioplasty in the management of symptomatic ICAS.

Background The effects of PSENEN mutations in patients with acne inversa (AI) are poorly understood. Hyperproliferation of follicular keratinocytes and resulting occlusion may constitute the initial pathophysiology. Objective To investigate the effects of PSENEN knockdown on γ-secretase subunits, biological behaviors, and related signaling pathways in keratinocytes. Methods HaCaT cells were divided into an experimental group (PSENEN knock down), a negative control group, and a blank control group. Whole transcriptome sequencing was used to measure differences in mRNA expression of the whole genome; real-time PCR and Western blotting were performed to determine the interference efficiency and the effects of interference on the components of γ-secretase and related molecules. CCK-8 was used to measure cell proliferation, and flow cytometry was used to measure apoptosis and the cell cycle. Results A comparison of five healthy controls with three patients with PSENEN mutation (c.66delG, c.279delC, c.229_230insCACC) revealed decreased expression of mRNA and protein in skin lesions of the experimental group. In this group, expression of the other components of γ-secretase presenilin C-terminal fragment decreased, expression of immature nicastrin increased, expression of mature nicastrin decreased, and expression of anterior pharynx defective-1 remained unchanged. KEGG analysis revealed that differentially expressed molecules were enriched in m-TOR signaling pathways. Subsequent verification confirmed that differences in PI3K-AKT-mTOR signaling pathway molecules, cell proliferation, apoptosis, cell cycle and the expression levels of Ki-67, KRT1, and IVL between the groups were not statistically significant. Conclusions PSENEN mutations alone may be insufficient to cause the development of AI, or they may only induce a mild phenotype of AI.

Atmospheric particulate matter (APM) is an environmental hazard that endangers human health and causes a variety of diseases. In this work, the microbial community composition, chemical element composition and antimicrobial resistance gene (ARG) prevalence, along with their relationships with environmental parameters were analysed using APM samples collected in Jinan, China. Pathogenic Klebsiella and Aeromonas were found to be significantly correlated with PM2.5 and temperature, suggesting their proliferation on APM. PM2.5 and PM10 have similar microbial community compositions but different chemical element compositions, suggesting they have different origins, which have little impact on microbial community structures. This finding, together with analysis of the timing of microbial community structure changes, suggests that microbial community composition is impacted by anthropic activities. Further investigations showed that rare metals including lanthanides are significantly negatively correlated with pathogens in APM, suggesting their inhibitory role. ARGs were observed for every class of antibiotic except for carbapenems in APM, suggesting high ARG prevalence in APM, and APM functions in transmission of antimicrobial resistance. Results obtained in this study suggest that APM can act as a transmission vehicle for pathogenic bacteria and ARGs and lead to the implication of a new transmission route for bacterial pathogenesis by APM.

Curcumin has been employed as a photosensitizer agent during photodynamic therapy (PDT). Cutibacterium acnes (C. acnes) can cause an inflammatory response in human keratinocytes; however, no research has been conducted to determine whether curcumin and its photodynamic properties can prevent this inflammatory reaction.We hypothesized that curcumin may control the C. acnes biofilm-induced inflammatory response in keratinocytes, either alone or in combination with blue light photodynamic therapy.Following C. acnes biofilm stimulation, human primary keratinocytes were treated with 20 μM curcumin solution alone or 5 μM curcumin with combined blue light irradiation. The amount of secreted protein was measured using an ELISA kit. The expression levels of Toll-like receptor 2 (TLR2) and its downstream proteins were determined using western blot.Treatment with 20 μM curcumin, but not 5 μM curcumin, reduced the inflammatory response to C. acnes biofilms in keratinocytes by blocking the TLR2/MAPK/NF-κB pathway. Interestingly, 5 μM curcumin combined with blue light also reduced the C. acnes biofilm-induced inflammation indicated above by blocking the TLR2/MAPK/NF-κB pathway.Curcumin alone, in sufficient concentrations, or low-concentration curcumin with blue light had anti-inflammatory activity on keratinocytes stimulated by C. acnes biofilms through inhibition of MAPK and NF-κB signaling pathways by downregulating TLR2 expression.

The enhancer of zeste homolog 2 (EZH2) represents a potential target for periodontitis treatment; however, its role in the development of periodontitis remains unclear. The current study aimed to elucidate the role of EZH2 in osteoclasts (OCs) growth as well as the mechanism underpinning the related process. The potential interaction among EZH2, microRNA-101 (miR-101), and vascular cell adhesion molecule 1 (VCAM-1) was evaluated using chromatin immunoprecipitation and dual-luciferase reporter gene assay. The expressions of EZH2 and miR-101 in OCs were examined by Western blot analysis and reverse transcription squantitative polymerase chain reaction. Loss- and gain-function assays were then performed to determine the role of EZH2/miR-101/VCAM-1 in periodontitis and OCs proliferation, followed by OC growth and proliferation detected using tartrate resistant acid phosphatase (TRAP) and 5-ethynyl-2'-deoxyuridine staining. Enzyme-linked immunoassay was conducted to determine the expression of interleukin 1β (IL-1β) and tumor necrosis factor-α (TNF-α). A periodontitis rat model was established to investigate the effect of EZH2 and VCAM-1 in vivo. EZH2 was overexpressed, while miR-101 was downregulated in the OCs of periodontitis. Silencing of EZH2, VCAM-1 repression, or miR-101 elevation suppressed the growth and proliferation of OC while acting to encumber the release of IL-1β and TNF-α. EZH2 negatively targeted miR-101, while miR-101 negatively targeted VCAM-1. Moreover, silencing of EZH2 or VCAM-1 was observed to attenuate periodontitis which was evidenced by an increase in BMD, BV/TV, and BS/BV as well as reduction in TRAP and cathepsin K in vivo. Taken together, the key findings of the current study demonstrate that EZH2 knockdown inhibited OC formation by elevating the expression of miR-101 via suppression of VCAM-1, ultimately attenuating periodontitis.

Objective:. Acne inversa is a multifactorial chronic debilitating disease. Genetic factors are involved in 40% of patients, especially the nicastrin (NCSTN) gene. However, the role of the mutated NCSTN gene in the pathogenesis of acne inversa remains unclear. Retinoic acid is recommends to treat moderate to severe acne inversa, therefor we conduct this in vitro research to study the association between NCSTN gene mutation and the retinoic acid signaling pathway in human immortalized skin keratinocyte (HaCaT) cells. Methods:. HaCaT cells were infected with a lentivirus-mediated short hairpin RNA (shRNA) expression plasmid specifically targeting the NCSTN gene. Real-time polymerase chain reaction (PCR) and Western blotting were used to detect the interference efficiency of NCSTN. RNA sequencing was used to detect differential genes in the NSCTN-deficient HaCaT cells. Based on bioinformatics analysis and clinical treatment data, the retinoic acid signal pathway was selected for screening. Quantitative PCR was used to verify the changes in the expressions of retinoic acid signaling pathway-related receptors and molecules in the HaCaT cell line after NCSTN silencing. The Student t test and one-way analysis of variance were used to evaluate intergroup differences. Results:. Sequencing showed that the NCSTN-shRNA lentiviral recombinant expression plasmid was successfully constructed. After lentivirus infection of HaCaT cells, real-time PCR results showed significantly reduced NCSTN mRNA expression in the interference group compared with the negative control group, and the interference efficiency was 75.0%. Western blotting showed that the inhibition rate of NCSTN protein expression in the shRNA group was 71.7%. RNA sequencing revealed significant differential expression of some genes, and changes in signaling pathways. A series of bimetallic nickel cobalt sulfides with hierarchical micro/nano architectures were fabricated via a facile synthesis strategy of bimetallic micro/nano structure precursor construction-anion exchange via solvothermal method. Among the nickel cobalt sulfides with different Ni/Co contents, the coral-like Ni1.01Co1.99S4 (Ni/Co, 1/2) delivers ultrafast and stable Na-ion storage performance (350 mAh·g−1 after 1,000 cycles at 1 A·g−1 and 355 mAh·g−1 at 5 A·g−1). The remarkable electrochemical properties can be attributed to the enhanced conductivity by co-existence of bimetallic components, the unique coral-like micro/nanostructure, which could prevent structural collapse and self-aggregation of nanoparticles, and the easily accessibility of electrolyte, and fast Na+ diffusion upon cycling. Detailed kinetics studies by a galvanostatic intermittent titration technique (GITT) reveal the dynamic change of Na+ diffusion upon cycling, and quantitative kinetic analysis indicates the high contribution of pseudocapacitive behavior during charge-discharge processes. Moreover, the ex-situ characterization analysis results further verify the Na-ion storage mechanism based on conversion reaction. This study is expected to provide a feasible design strategy for the bimetallic sulfides materials toward high performance sodium-ion batteries.

A simple, yet efficient synthetic approach for the construction of vinylene-linked conjugated porous networks was developed. Based on the Horner–Wadsworth–Emmons reaction, the condensation polymerization for the formation of an sp2 carbon-linkage can be achieved at room temperature. The resulting vinylene-linked frameworks exhibit a promising porous nature with the best surface area of up to 1373 m2 g−1. Their intrinsic conjugated architectures and semiconducting properties lead to photocatalytic evolution of hydrogen from water under visible light irradiation. This new synthesis method provides a facile means to prepare attractive sp2 carbon linked porous organic frameworks.

Protein methyltransferase arginine 7 (PRMT7) is the sole member among protein arginine methyltransferases (PRMTs) that participates in the monomethylarginine formation involved in tumor biology, DNA damage repair, and development. In this study, we report that PRMT7 is downregulated in prostate cancer cell lines in vitro and its overexpression suppresses migration and invasion in the prostate cancer cell lines LNCaP and PC3, as determined by wound healing, migration, and invasion assays. To identify the potential downstream mediator of PRMT7, we performed whole-genome transcriptome and proteome sequencing analysis, and found that the metastasis suppressor gene KISS1R was up-regulated both at the transcriptional and translational level by PRMT7. Knockdown of KISS1R rescued the tumor-suppressive phenotype. Collectively, we uncovered the anti-oncogenic role of PRMT7 in prostate cancer cells and further demonstrated that KISS1R is a downstream effector molecule of PRMT7 in modulating tumor cell migration and invasion and PRMT7 may be effective target for the clinical treatment of prostate cancer.

BACKGROUND: Hemorrhagic transformation (HT) of acute ischemic stroke (AIS) is associated with poor outcome. Previous studies only reported the association of mean ischemic severity or total infarct volume with HT after endovascular therapy (EVT). We aimed to investigate the predictive value of preoperative apparent diffusion coefficient (ADC) map for HT by combinated ischemic severity and corresponding volume in AIS after successful recanalization with EVT. METHODS: We retrospectively analyzed 119 consecutive cases of AIS with large vessel occlusion of anterior circulation within 24 hours after symptom onset and successful recanalization after EVT. Reactive oxygen species (ROS) are increasingly recognized as non-enzymatic players in the processes of radicle elongation growth and endosperm weakening during seed germination. NADPH oxidases (EC 1.6.3.1), also known as respiratory burst oxidase homologues (Rbohs), are key enzymes for the production of ROS. We previously reported that ROS are involved in the radicle elongation growth and endosperm weakening during lettuce seed germination. However, the function of the gene(s) encoding Rbohs during lettuce seed germination remains to be elucidated. In this study, one lettuce Rboh gene LsRbohC1 was cloned, and over-expression and RNAi-lines of this gene were generated. It was found that LsRbohC1 was abundantly expressed in germinating seeds, especially in the endosperm cap and hypocotyl. Over-expression/knock-down of this gene observably increased/decreased the production of superoxide radicals in the radicle and endosperm cap, and significantly promoted/delayed the germination process. The results suggest that LsRbohC1 plays a role in lettuce seed germination.

To enhance the clinical diagnosis rate of acute respiratory distress syndrome (ARDS) and provide a reliable basis for follow-up treatment, in this research, the hemodynamic indexes of patients with severe respiratory diseases are monitored by ultrasonic imaging. A total of 64 patients with ARDS in the department of respiratory medicine of XXX hospital from June 2018 to December 2018 are listed as study subjects (including 45 males and 19 females), and are divided into control group (CG) and experimental group (EG) in random. In the CG, central venous pressure is used to detect the patients, while in the EP, ultrasonic imaging is used to detect the patients, and the ultrasonic image is denoised by wavelet transform. Respiratory parameters (respiratory rate, oxygenation index, and positive end-expiratory pressure ventilation) and hemodynamic parameters (heart rate, mean arterial pressure, pulmonary static compliance, and airway plateau pressure) are compared on the first and 10th day of admission. The fluid volume of the two groups of patients was managed according to the hemodynamic parameters. After three months, the prognosis (duration of ICU treatment, duration of mechanical ventilation, incidence of heart failure and mortality) of the patients in both groups were calculated. The results show that wavelet transform can effectively reduce the noise of ultrasonic images, so as to obtain real and reliable data. Compared with the first day of admission, respiratory parameters and hemodynamics of the patient change significantly after 10 days (P < 0.05). The average daily fluid intake in the EP is significantly lower than that in the CG (P < 0.05). The duration of treatment in ICU and mechanical ventilation in the EG is significantly lower than that in the CG (P < 0.05), which indicates that ultrasonic imaging can detect and diagnose the hemodynamics of patients, thus providing reliable guidance for patients’ fluid management and avoiding patients’ cardiac failure induced by fluid overload. This provides an experimental scheme for hemodynamic monitoring of patients with severe respiratory system.

The parasitoid of whiteflies Encarsia formosa has been widely applied to reduce whitefly-mediated damage on vegetables and ornamental plants grown in greenhouses. Although its chemosensory behavior has been described, the mechanism by which E. formosa recognizes chemical volatiles at the molecular level remains unknown. In this study, we obtained 66,632 unigenes from antennae transcriptomic architecture of E. formosa, of which 19,473 (29.2%) were functionally annotated. All that matters is that we manually identified 39 odorant-binding proteins (OBPs) from above dataset, and further investigated the tissue and stage-specific expression profiles of all identified OBP genes by real-time quantitative PCR. Among these OBP genes, 32 were enriched in antennae, and 2 in body. In addition, 4 OBPs were highly expressed in pupae, and 32 in 6-hour-age adults after eclosion. In addition to identifying OBP genes from E. formosa, this study provides a molecular basis for further functional studies of OBPs and the interactions of hosts and parasitic wasps.

It is a challenge to promote aqueous-phase precise conversion of renewable lignin to valuable chemical building blocks via engineering transition metal catalysts. Motivated by the water effect in promoting catalytic activity, a Ni/Nb2O5 catalyst assembled from plenty of Nb2O5 nanoplates enabled efficient selective hydrodeoxygenation of lignin-derived phenols to alcohols in the aqueous phase. Taking advantage of hierarchical plate features, the interactions between Ni and Nb2O5 plates were enhanced, with the generation of additional interfacial Ni–NbOx sites. The presence of water significantly enhanced the selective hydrodeoxygenation performance by promoting hydrogenation and hydrolysis, and retarding the dehydration process. Through this strategy, the overall yields of alcohols for lignin oil upgrading reached 73.5 wt% in water at 220 °C.

Water imbibition is a key factor affecting the flowback regime of shale gas wells after volume fracturing. In this study, a set of experimental apparatus and corresponding test and evaluation methods were developed to analyze the laws of forcible imbibition of water in a shale reservoir, characterize the initiation time of microfractures induced by shale hydration quantitatively, and optimize the shut-in time of shale gas wells; the imbibition depths of different pore types are quantitatively calculated based on the multiple pore imbibition analytical model. The experimental results show that: according to imbibition saturation growth rate, the shale forcible imbibition can be divided into three periods, imbibition diffusion, imbibition transition, and imbibition balance. Among them, the imbibition diffusion period is the main period for imbibition capacity rise. The reason for this phenomenon is that due to the fluid pressure difference effect, the shale fills its large pores and microfractures rapidly in the early stage, and in the percolation transition period, the percolation rate decreases continuously due to the gradual increase of fluid saturation. Due to the Jamin effect, it is difficult for the fluid to enter the small pores and the fluid fills the pore roar channel, the seepage saturation tends to stabilize, and the seepage equilibrium period appears. In the early period of shut-in, the imbibition capacity of shale increases significantly under the action of fluid pressure, providing a large amount of imbibition fluid for the spontaneous imbibition later. The imbibition depth of a clay pore was much greater than that of a brittle mineral pore and an organic pore. The reservoir confining pressure has prohibition on shale imbibition, but even under reservoir confining pressure, imbibition can still improve the fracturing effect of the reservoir, resulting in an increase in porosity of 0.42–1.63 times and an increase in permeability of 17.6–67.3 times. Under the experimental conditions, the initiation time of induced microfractures is 98.5 h on average and is in negative correlation with imbibition capacity. On this basis, the optimized shortest shut-in time of a shale gas well is 5 days. The study results can provide a scientific basis for the optimization of the flowback regime of shale gas reservoirs.

Blood vessels in the brain tissue form a compact vessel structure and play an essential role in maintaining the homeostasis of the neurovascular system. The low dosage of photodynamic intervention (PDT) significantly affects the expression of cellular biomarkers. To understand the impact of photodynamic interventions on cerebrovascular endothelial cells, we evaluated the dosage-dependent impact of porfimer sodium-mediated PDT on B.END3 cells using flow cytometer, comet assay, RNA sequencing, and bioinformatics analysis. To examine whether PDT can induce disorder of intracellular organelles, we did not observe any significance damage of DNA and cellular skeleton. Moreover, expression levels of cellular transporters-related genes were significantly altered, implying the drawbacks of PDT on cerebrovascular functions. To address the potential molecular mechanisms of these phenotypes, RNA sequencing and bioinformatics analysis were employed to identify critical genes and pathways among these processes. The gene ontology (GO) analysis and protein-protein interaction (PPI) identified 15 hub genes, highly associated with cellular mitosis process (CDK1, CDC20, MCM5, MCM7, MCM4, CCNA2, AURKB, KIF2C, ESPL1, BUB1B) and DNA replication (POLE2, PLOE, CDC45, CDC6). Gene set enrichment analysis (GSEA) reveals that TNF-α/NF-κB and KRAS pathways may play a critical role in regulating expression levels of transporter-related genes. To further perform qRT-PCR assays, we find that TNF-α/NF-κB and KRAS pathways were substantially up-regulated, consistent with GSEA analysis. The current findings suggested that a low dosage of PDT intervention may be detrimental to the homeostasis of blood-brain barrier (BBB) by inducing the inflammatory response and affecting the expression of surface biomarkers.

The development of novel redox-active polymers for sustainable sodium ion batteries (SIBs) has captured growing attention, but battery performance has been significantly limited by poor reversible specific capacities, where the majority of aromatic C6-benzene linkages are redox inactive. Here, a simple, yet efficient approach to improve sodium (Na) storage on these C6-benzene rings within a porous polymeric framework by rationally regulating their steric hindrance is reported. Decreasing intrinsic hindrance affords a significant improvement in redox reaction kinetics within the porous architecture, thereby facilitating the acceptance of Na ions on these functionalized benzene rings and boosting the SIB performance. As a result, the modulate porous framework exhibits an exceptional battery capacity of 376 mAh g-1 after 1000 cycles at 1.0 A g-1 , which is ≈1.5 times larger than that of the pristine framework. Furthermore, the performance can reach as high as 510 mAh g-1 at 0.1 A g-1 , comparable to that of the best-performing polymeric electrodes. The simple modulation approach not only enables Na storage modulation on functionalized C6-benzene rings, but also simultaneously provides a means to extend the understanding of the structure-property relationship and facilitate new possibilities for organic SIBs.
