53. Light Spectrum Impacts on Growth, Molting, and Oxidative Stress Response of the Mud Crab Scylla paramamosain

Abstract

An 8 weeks trial was performed to test the effects of light spectra [full-spectrum, violet (405 nm), blue (470 nm), cyan (500 nm), green (525 nm), yellow (570 nm), and red (625 nm)] on growth performance, molting, antioxidant capacity, stress response and expression of molting, and apoptosis-related genes in Scylla paramamosain. Results showed that spectrum had a significant effect on S. paramamosain physiology. Compared to blue light, crabs exposed to violet light had a significantly lower survival rate (79.5 ± 3.6% vs. 94.9 ± 3.6%), weight gain (49.2 ± 5.4 vs. 67.6 ± 6.7), molt frequency (4.2 ± 0.2 vs. 4.5 ± 0.1), and extended intermolt intervals between instar 1 and 2 stages (C1–C2) (6.3 ± 0.3 vs. 5.0 ± 0.1 days). Expression of the molt-inhibiting hormone (mih) gene was upregulated in crabs reared under violet light. According to the regression analysis, maximum SGR would be at 449.97 nm. Crabs exposed to blue light also had lower melatonin levels than under full-spectrum and lower cortisol levels than violet and yellow groups. Regarding oxidative stress, crabs in full-spectrum had lower H2O2 and MDA contents, however, no significant difference was found in total antioxidant capacity (T-AOC), superoxide dismutase (SOD), and catalase (CAT) in hepatopancreas from crabs under different spectra. Gene expression of hsp40, hsp70, hsp90 were down-regulated in crabs exposed to the full-spectrum light group. Regarding apoptosis-related genes, bcl-2 gene expression in crabs under cyan and the cox IV and caspase 3 in green were upregulated, suggesting cyan light may inhibit, while green light may promote apoptosis. Taken together, these results suggest that blue or cyan light would promote growth performance, while full-spectrum light could reduce stress response in S. paramamosain.

Abstract

Identifying the response of Portunus trituberculatus to ocean acidification (OA) is critical to understanding the future development of this commercially important Chinese crab species. Recent studies have reported negative effects of OA on crustaceans. Here, we subjected swimming crabs to projected oceanic CO2 levels (current: 380 μatm; 2100: 750 μatm; 2200: 1500 μatm) for 4 weeks and analyzed the effects on survival, growth, digestion, antioxidant capacity, immune function, tissue metabolites, and gut bacteria of the crabs and on seawater bacteria. We integrated these findings to construct a structural equation model to evaluate the contribution of these variables to the survival and growth of swimming crabs. Reduced crab growth shown under OA is significantly correlated with changes in gut, muscle, and hepatopancreas metabolites whereas enhanced crab survival is significantly associated with changes in the carbonate system, seawater and gut bacteria, and activities of antioxidative and digestive enzymes. In addition, seawater bacteria appear to play a central role in the digestion, stress response, immune response, and metabolism of swimming crabs and their gut bacteria. We predict that if anthropogenic CO2 emissions continue to rise, future OA could lead to severe alterations in antioxidative, immune, and metabolic functions and gut bacterial community composition in the swimming crabs through direct oxidative stress and/or indirect seawater bacterial roles. Abstract

Three salinities (4‰, 12‰ and 25‰) were selected to determine the effects of different salinities on the growth, survival, nutrition and energy metabolism of Scylla paramamosain during indoor overwintering. Growth performance (survival rate, condition factor, hepatopancreatic index, weight gain rate and specific growth rate), nutrient composition (ash, moisture, crude fat, crude protein and total nitrogen), fatty acids, energy metabolites (glycogen, glucose, triglycerides, cholesterol and lactic acid) and energy metabolic enzymes (hexokinase, pyruvate kinase, lactate dehydrogenase and succinate dehydrogenase) were measured and calculated. The results showed that S. paramamosain exhibited the best growth and survival at 25‰ and worst at 4‰ after overwintering. In addition to consuming crude fat, excess protein was also broken down to provide energy in the 4‰ and 12‰ groups. The content of unsaturated fatty acids in the 25‰ group was higher after overwintering. Glycogen and glucose consumption and triglycerides, cholesterol and lactic acid production were lowest in the 25‰ group. The activity of hexokinase, pyruvate kinase, lactate dehydrogenase and succinate dehydrogenase was highest in the 4‰ group and lowest in the 25‰ group. The activity of hexokinase, pyruvate kinase, lactate dehydrogenase and succinate dehydrogenase in the hepatopancreas was higher than in the muscle. In conclusion, S. paramamosain uses less energy, has more unsaturated fatty acids and has a higher survival rate at a salinity of 25‰ after overwintering. Abstract

The geographical origin of mud crab (Scylla paramamosain) has been arousing increasing interest to consumers due to their flavour and market price. In this study, an NMR-based metabolomics method was used to characterize the differences in the chemical composition of muscle samples of mud crabs caught in four different geographical areas of China. The results showed that a statistically significant separation existed between each of the groups of mud crabs in terms of their geographical origins. The major metabolites responsible for differentiation included inosine 5'-monophosphate (IMP), adenosine 5'-monophosphate (AMP), and some amino acids. A gradual increase in the AMP level in conjunction with a declining IMP level was closely associated with the growth latitude of the mud crab, which means that these metabolites could potentially be used to characterize the specific geographic origin of mud crabs. Abstract

Abstract: BACKGROUND: Marinated meat in soy sauce is one of the most popular traditional cured meat products in China. Its taste quality is directly related to primary and secondary metabolites. Herein, the change of metabolite composition of marinated meat in soy sauce during processing was systematically characterised using 1H NMR and multivariate data analysis. RESULTS: The marinated meat in soy sauce metabonome was dominated by 26 metabolites, including amino acids, sugars, organic acids, nucleic aides and their derivatives. PC1 and PC2 explained a total of 78.6% and 16.6% of variables, respectively. Amino acids, sugars, acetate, succinate, uracil and inosine increased during marinating, while lactate, creatine, inosine‐5′‐monophosphate (5′‐IMP) and anserine decreased (P < 0.05). After marinating, most of the metabolites decreased except for acetate and alanine (P < 0.05). There was a negative effect on the taste of marinated meat in soy sauce during the late stage of dry‐ripening. CONCLUSION: These findings indicated that the potential of NMR‐based metabonomics is of importance for taste quality of marinated meat in soy sauce, which could contribute to a better understanding of the changes of taste compounds in meat products during processing.
