In vitro cultures of primary cortical neurons are widely used to investigate neuronal function. However, it has yet to be fully investigated whether there are significant differences in development and function between cultured rodent and primate cortical neurons, and whether these differences influence the utilization of cultured cortical neurons to model pathological conditions. Using in vitro culture techniques combined with immunofluorescence and electrophysiological methods, our study found that the development and maturation of primary cerebral cortical neurons from cynomolgus monkeys were slower than those from mice. We used a microelectrode array technique to compare the electrophysiological differences in cortical neurons, and found that primary cortical neurons from the mouse brain began to show electrical activity earlier than those from the cynomolgus monkey. Although cultured monkey cortical neurons developed slowly in vitro, they exhibited typical pathological features-revealed by immunofluorescent staining-when infected with adeno-associated viral vectors expressing mutant huntingtin (HTT), the Huntington's disease protein. A quantitative analysis of the cultured monkey cortical neurons also confirmed that mutant HTT significantly reduced the length of neurites. Therefore, compared with the primary cortical neurons of mice, cultured monkey cortical neurons have longer developmental and survival times and greater sustained physiological activity, such as electrophysiological activity. Our findings also suggest that primary cynomolgus monkey neurons cultured in vitro can simulate a cell model of human neurodegenerative disease, and may be useful for investigating time-dependent neuronal death as well as treatment via neuronal regeneration. Nanoarchitectured electroactive materials can boost rates of Li insertion/extraction, showing genuine potential to increase power output of Li-ion batteries. However, electrodes assembled with low-dimensional nanostructured transition metal oxides by conventional approach suffer from dramatic reductions in energy capacities owing to sluggish ion and electron transport kinetics. Here we report that flexible bulk electrodes, made of three-dimensional bicontinuous nanoporous Cu/MnO2 hybrid and seamlessly integrated with Cu solid current collector, substantially optimizes Li storage behavior of the constituent MnO2. As a result of the unique integration of solid/nanoporous hybrid architecture that simultaneously enhances the electron transport of MnO2, facilitates fast ion diffusion and accommodates large volume changes on Li insertion/ extraction of MnO2, the supported MnO2 exhibits a stable capacity of as high as ∼1100 mA h g-1 for 1000 cycles, and ultrahigh charge/discharge rates. A simple and sensitive reversed-phase high-performance liquid chromatography method with UV detection is developed and validated for the determination of 6,7-dimethoxycoumarin in rat plasma and comparative analysis of its pharmacokinetics after intragastric administration of 6,7-dimethoxycoumarin and three different decoctions of Yinchenhao Tang. The plasma samples are deproteinated with acetonitrile. The components are separated on a Kromasil C18 column (250 × 4.6 mm, 5 µm,) with methanol–1% acetic acid solution–tetrahydrofuran (30:63:7, v/v/v) as the mobile phase, and the UV detector is set at 340 nm. Coumarin is used as an internal standard. The linear calibration curve is obtained in the concentration range of 25~2500 ng/mL. The lower limit of quantitation of the method is 25 ng/mL. The intra- and inter-day precision are less than 12%, and the accuracy determined with relative error ranges from –2.9% to 1.7%. The data obtained from rat plasma are analyzed with Topfit 2.0 Pharmacokinetic Software. With pharmacokinetic analysis, the main parameters after intragastric administration of 6,7-dimethoxycoumarin, Herba Artemisiae Scopariae decoction, Artemisiae Scopariae decoction plus Radix et Rhizoma Rhei and Fructus Gardeniae decoction, Yinchenhao Tang are as follows: T1/2 is 0.29, 1.30, 1.07, and 1.75 h, AUCÆt is 919.1, 1215.0, 2035.3, and 2537.9 ng·h/mL, AUC0Æ is 928.5, 1325.9, 2094.4, and 2612.6 ng·h/mL, respectively. Aptamers are specific nucleic acid sequences that can bind to a wide range of nucleic acid and non-nucleic acid targets with high affinity and specificity. Nucleic acid aptamers are selected in vitro from single stranded DNA or RNA ligands containing random sequences of up to a few hundred nucleotides. Systematic evolution of ligands by exponential enrichment (SELEX) was used to select and PCR amplify DNA sequences (aptamers) capable of binding to and detecting Listeria monocytogenes, one of the major food-borne pathogens. A simplified affinity separation approach was employed, in which L. monocytogenes in exponential (log) phase of growth was used as the separation target. A fluorescently-labeled aptamer assay scheme was devised for detecting L. monocytogenes. This report described a novel approach to the detection of L. monocytogenes using DNA aptamers. Aptamers were developed by nine rounds of SELEX. A high affinity aptamer was successfully selected from the initial random DNA pool, and its secondary structure was also investigated. One of aptamers named e01 with the highest affinity was further tested in aptamer-peroxidase and aptamer-fluorescence staining protocols. This study has proved the principle that the whole-cell SELEX could be a promising technique to design aptamer-based molecular probes for dectection of pathogenic microorganisms without tedious isolation and purification of complex markers or targets.
