Moxa wool is a traditional Chinese herbal medicine, which can warm channels to dispel coldness. At present, there is no unified index to evaluate the purity and growing years of moxa wool in the market. Terpineol is one of the effective substances in the volatile oil of moxa wool. Here, we characterize the purity and growing years of moxa wool by studying terpineol. Gas chromatography-mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC) are the methods for monitoring terpineol at present, all of which have defects of complicated procedures. We established linear fitting to distinguish the different purities of moxa wool through the intensities (areas) of terpineol, the characteristic peaks, and the consequence presented; the coefficient of determination (R2) was higher than 0.90. Furthermore, based on the characteristic peak position of standard terpineol, the correlation model with the purity and growing year of moxa wool was set up, thereby differentiating the quality of moxa wool. We have built the partial least squares (PLS) model of the growing years of moxa wool with high accuracy, and the determination coefficient is greater than 0.98. In addition, we compare the quantitative accuracy of Raman spectroscopy with terahertz technology. Finally, a new method of terahertz spectroscopy to evaluate quality of moxa wool was found. It provides a new idea for the identification of inferior moxa wool in the market and a new method for identifying the quality of moxa wool in traditional Chinese medicine.

In order to abstract away a network model from some real-world networks, such as navigation satellite networks and mobile call networks, we proposed an Isochronal-Evolution Random Matching Network (IERMN) model. An IERMN is a dynamic network that evolves isochronally and has a collection of edges that are pairwise disjoint at any point in time. We then investigated the traffic dynamics in IERMNs whose main research topic is packet transmission. When a vertex of an IERMN plans a path for a packet, it is permitted to delay the sending of the packet to make the path shorter. We designed a routing decision-making algorithm for vertices based on replanning. Since the IERMN has a specific topology, we developed two suitable routing strategies: the Least Delay Path with Minimum Hop (LDPMH) routing strategy and the Least Hop Path with Minimum Delay (LHPMD) routing strategy. An LDPMH is planned by a binary search tree and an LHPMD is planned by an ordered tree. The simulation results show that the LHPMD routing strategy outperformed the LDPMH routing strategy in terms of the critical packet generation rate, number of delivered packets, packet delivery ratio, and average posterior path lengths.

As China’s first operational second-generation geostationary satellite, Fengyun-4B carries the newly developed Advanced Geostationary Radiation Imager (AGRI), which adds a low-level water vapor detection channel and an adjusted spectrum range of four channels to improve the quality of observation. To characterize biases of the infrared (IR) channels of Fengyun-4B/AGRI, RTTOV was applied to simulate the brightness temperature of the IR channels during the period of Fengyun-4B trial operation (from June to November 2022) under clear-sky conditions based on ERA5 reanalysis, which may provide beneficial information for the operational applications of Fengyun-4B/AGRI, such as data assimilation and severe weather monitoring. The results are as follows: (1) due to the sun’s influence on the satellite instrument, the brightness temperature observations of the Fengyun-4B/AGRI 3.75 μm channel were abnormally high around 1500 UTC in October, although the data producer made efforts to eliminate abnormal data; (2) the RTTOV simulations were in good agreement with the observations, and the absolute mean biases of the RTTOV simulations were less than 1.39 K over the ocean, and less than 1.77 K over land, for all IR channels under clear-sky conditions, respectively; (3) for the variation of spatial distribution bias over land, channels 12–15 were more obvious than channels 9–11, which indicates that the skin temperature of ERA-5 reanalysis and surface emissivity may have greater spatial uncertainty than the water vapor profile; (4) the biases and standard deviations of Fengyun-4B/AGRI channels 9–15 had negligible dependence on the satellite zenith angles over the ocean, while the standard deviation of channels 8 and 12 had a positive correlation with satellite zenith angles when the satellite zenith angles were larger than 30°; and (5) the biases and standard deviations of Fengyun-4B/AGRI IR channels showed scene brightness temperature dependence over the ocean.

An inter-satellite communication network of satellite swarm (ICNS) is created if the members of a satellite swarm communicate with each other via inter-satellite links (ISLs). ICNS can be constructed using the theory of complex networks. A link community is defined as two satellites between which the ISL has been established. The satellite swarm, whose members have not established ISLs, is modeled as a pre-link network (PLN). The edge of a PLN is described as a candidate for the link community. Consequently, an ICNS can be constructed by collecting combinations of candidates for link communities (CCLC) based on PLN and then by selecting one of these. An algorithm is designed to take a sample of all CCLCs. A new structural entropy of networks is developed to evaluate a CCLC. The CCLC with a maximum structural entropy in the CCLC sample will be selected to become the edge set of the ICNS. An improvement method was proposed to ensure that the ICNS remains a dynamic connected network by preventing each satellite from establishing an ISL with the same satellite. The simulations demonstrated that the proposed method outperformed the benchmark methods, and it is necessary to adopt the improvement method.

The complete chloroplast genome sequence of Podocarpus macrophyllus was determined in this study. According to the results, the genome is 134,239 bp in length. The GC content of the whole chloroplast genome is 37.0%. The complete chloroplast genome of P. macrophyllus encodes a total of 120 genes, including 34 tRNA genes, 4 rRNA genes and 82 protein-coding genes. Like other conifers chloroplast genomes, P. macrophyllus has no inverted repeat sequences. To reveal the phylogenetic relationship of P. macrophyllus, we constructed phylogenetic trees using other species of Podocarpaceae, and the phylogenetic analysis showed that P. macrophyllus is evolutionarily closest to Podocarpus longifoliolatus.

Emotion information from speech can effectively help robots understand speaker's intentions in natural human-robot interaction. The human auditory system can easily track temporal dynamics of emotion by perceiving the intensity and fundamental frequency of speech, and focus on the salient emotion regions. Therefore, speech emotion recognition combined with the auditory mechanism and attention mechanism may be an effective way. Some previous studies used auditory-based static features to identify emotion while ignoring the emotion dynamics. Some other studies used attention models to capture the salient regions of emotion while ignoring cognitive continuity. To fully utilize the auditory and attention mechanism, we first investigate temporal modulation cues from auditory front-ends and then propose a joint deep learning model that combines 3D convolutions and attention-based sliding recurrent neural networks (ASRNNs) for emotion recognition. Our experiments on the IEMOCAP and MSP-IMPROV datasets indicate that the proposed method can be effectively used to recognize the emotions of speech from temporal modulation cues. The subjective evaluation shows that the attention patterns of the attention model are basically consistent with human behaviors in recognizing the emotions.

Abstract Background The optimal number of retrieved lymph nodes (LNs) in gastric cancer (GC) is still debatable and previous studies proposing new classification alternatives mostly focused on the number of retrieved LNs without proper consideration on the anatomic nodal groups’ location. Here, we assessed the impact of retrieved LNs from different nodal location groups on the survival of GC patients. Methods Stage I–III gastric cancer patients who had radical gastrectomy were investigated. LN grouping was determined according to the 13th edition of the JCGC. The optimal cut-off values of retrieved LNs in different LN groups (Group 1 and 2) were calculated, based on which a proposed nodal classification (rN) simultaneously accounting the optimal number and location of retrieved LNs was proposed. The performance of rN was then compared to that of LN ratio, log-odds of metastatic LNs (LODDs) and the 8th edition of the Union for International Cancer Control/American Joint Committee on Cancer (UICC/AJCC) N classification. Results The optimal cut-off values for Group 1 and 2 were 13 and 9, respectively. The 5-year overall survival (OS) was higher for patients in retrieved Group 1 LNs > 13 (vs. Group 1 LNs ≤ 13, 63.2% vs. 57.9%, P = 0.005) and retrieved Group 2 LNs > 9 (vs. Group 2 LNs ≤ 9, 72.5% vs. 60.7%, P = 0.009). Patients staged as pN0–3b were sub classified using this Group 1 and 2 nodal analogy. The OS of pN0–N2 patients in retrieved Group 1 LNs > 13 or Group 2 LNs > 9 were superior to those in retrieved Group 1 LNs ≤ 13 and Group 2 LNs ≤ 9 (All P 13 and > 9 LNs for Group 1 and Group 2, respectively, could represent an alternative lymph node retrieval approach in radical gastrectomy for more precise survival prognostication and minimizing staging migration, especially if > 16 LNs is found to be difficult.

PbxSr1-xTiO3 (x = 0.30, 0.35, 0.40, 0.45, 0.50 and 0.55) ceramics were fabricated by a solid-state reaction route. X–ray diffraction data at room temperature show PST samples shift from cubic to tetragonal phase with the increase of Pb2+ content. The microstructures were observed by scanning electron microscopy. Dielectric measurement was employed to investigate the ferroelectric–paraelectric phase transition behavior. Temperature dependent polarization–electric field hysteresis loops were conducted to study the electrocaloric effect (ECE) of the ferroelectric ceramics by indirect methods over a wide temperature range. Direct measurement of temperature change (ΔT) at room temperature for all samples can achieve 0.79–1.86 K. What's more, a giant ECE (ΔT = 2.05 K, EC strength (ΔT/ΔE) = 0.51 × 10−6 K m/V, under 40 kV/cm) was obtained in the sample of x = 0.35 near phase transition temperature. Our results suggest that the ceramics are promising cooling materials with excellent EC properties for energy related applications. In one study on vocal emotion recognition using noise-vocoded speech (NVS), the high similarities between modulation spectral features (MSFs) and the results of vocal-emotion-recognition experiments indicated that MSFs contribute to vocal emotion recognition in a clean environment (with no noise and no reverberation). Other studies also clarified that vocal emotion recognition using NVS is not affected by noisy reverberant environments (signal-to-noise ratio is greater than 10 dB and reverberation time is less than 1.0 s). However, the contribution of MSFs to vocal emotion recognition in noisy reverberant environments is still unclear. We aimed to clarify whether MSFs can be used to explain the vocal-emotion-recognition results in noisy reverberant environments. We analyzed the results of vocal-emotion-recognition experiments and used an auditory-based modulation filterbank to calculate the modulation spectrograms of NVS. We then extracted ten MSFs as higher-order statistics of modulation spectrograms. As shown from the relationship between MSFs and vocal-emotion-recognition results, except for extremely high noisy reverberant environments, there were high similarities between MSFs and the vocal emotion recognition results in noisy reverberant environments, which indicates that MSFs can be used to explain such results in noisy reverberant environments. We also found that there are two common MSFs (MSKTk (modulation spectral kurtosis) and MSTLk (modulation spectral tilt)) that contribute to vocal emotion recognition in all daily environments.

Background and Aims: It has recently emerged the concept of “obesity paradox,” a term used to describe an inverse association between obesity and clinical outcomes in cardiovascular diseases and stroke. The purpose of this study was to investigate the association between body mass index (BMI) and the risk of intracranial aneurysm rupture.Methods: In this study, we conducted a retrospective analysis of a prospectively maintained database of patients with intracranial aneurysms from 21 medical centers in China. A total of 3,965 patients with 4,632 saccular intracranial aneurysms were enrolled. Patients were separated into unruptured (n = 1,977) and ruptured groups (n = 1,988). Univariable and multivariable logistic regression analyses were performed to determine the association between BMI and intracranial aneurysm rupture.Results: Compared to the patients with normal BMI (18.5 to < 24.0 kg/m2), the odds of intracranial aneurysm rupture were significantly lower in patients with BMI 24.0 to < 28.0 kg/m2 (OR = 0.745, 95% CI = 0.638–0.868, P = 0.000) and patients with BMI ≥ 28.0 kg/m2 (OR = 0.628, 95% CI = 0.443–0.890, P = 0.009). Low BMI (60y, 95% CI = 0.586–0.928, P = 0.009) and the BMI ≥ 28.0 kg/m2 (OR = 0.517 for aged 50–60y, 95% CI = 0.281–0.950, P = 0.0034; OR = 0.535 for aged >60y, 95% CI = 0.318–0.899, P = 0.0018) was associated with a lower rupture risk in patients aged ≥50 years, whereas the association was not significant in patients aged

ObjectiveA novel immunochromatographic test strip method was developed to detect tissue parathyroid hormone (PTH) using the immune colloidal gold technique (ICGT). The accuracy and application value of this method for intraoperative parathyroid identification were evaluated.MethodsSerum samples were collected to measure PTH by both ICGT and electrochemiluminescence immunoassay (ECLIA). Patients who underwent unilateral and total thyroidectomy were enrolled to evaluate the feasibility and clinical efficacy of rapid intraoperative identification of parathyroid glands via PTH determination using ICGT. Two sample preparation methods, fine needle aspiration (FNA) and tissue block homogenate (TBH), were used for PTH-ICGT analysis.ResultsBablok analysis showed a linear relationship between the serum PTH measurements obtained by ICGT and ECLIA. Non-parathyroid tissues had much lower PTH concentrations (14.8 ± 2.1 pg/ml, n = 97) detected by ICGT, compared to the parathyroid gland tissues (955.3 ± 16.1 pg/ml, n = 79; P < 0.0001), With biopsy results as the standard, ICGT showed higher diagnosis rates as compared with direct visual inspection, for identifying both parathyroid glands (97.4 vs. 78.2%) and non-parathyroid tissues (100 vs. 68.9%). The cut-off values for parathyroid identification by FNA and TBH methods were 63.99 and 136.30 pg/ml, respectively. The detection time was 2 min by TBH method for in vitro tissue detection and 6 min by FNA method for in situ tissue detection, both of which were faster than traditional intraoperative cryopathological examination (usually >30 min). Intraoperative application of ICGT method was associated with higher postoperative serum calcium and blood PTH levels at 1 and 3 months as well as a lower incidence of postoperative transient hypocalcemia, as compared with direct visual inspection.ConclusionPTH-ICGT assay shows high potential as a rapid, novel alternative for intraoperative parathyroid identification.

Background: Environmental exposure to toxic metals is an important risk factor to human health. Traditional methods have examined associations between a health endpoint and exposure to heavy metals by either univariate or multiple regression. In the setting of ubiquitous heterogeneous environmental exposures, statistical methods that incorporate mixed exposures are increasingly relevant and may provide new insight into the association between metal exposure and important cardiovascular, renal and respiratory outcomes. Objective: The objective of this study was to classify the population of National Health and Nutrition Examination Survey (NHANES) into different exposure subgroups using modern unsupervised clustering methods based on lead, cadmium, mercury, and arsenic measured in urine or whole blood, and to assess the association between the identified exposure groups and twelve important health endpoints. Methods: We analyzed a sub-cohort of 9662 subjects participating in the 6 cycles (2003–2004 to 2013–2014) of NHANES study. The urine levels of 3 heavy metals (total arsenic, lead, cadmium) and blood levels of 3 heavy metals (lead, cadmium and mercury) were analyzed using a two-step approach. In the first step, we stratified the population into subgroups using unsupervised clustering (k-medoids) based on levels of metals either in urine or in blood. Then, we examine the association between 12 health endpoints and identified exposure subgroups while controlling for age, sex, race/ethnicity, education, smoking status, BMI, and urinary creatinine. Results: The k-medoids algorithm clustered NHANES population into 2 groups based on either blood or urinary levels of heavy metals. Developing effective and green methods for food analysis and separation has become an urgent issue regarding the ever-increasing concern of food quality and safety. Ionic liquids (ILs) are a new chemical medium and soft functional material developed under the framework of green chemistry and possess many unique properties, such as low melting points, low-to-negligible vapor pressures, excellent solubility, structural designability and high thermal stability. Combining ILs with extraction techniques not only takes advantage of ILs but also overcomes the disadvantages of traditional extraction methods. This subject has attracted intensive research efforts recently. Here, we present a brief review of the current research status and latest developments regarding the application of IL-assisted microextraction, including dispersive liquid–liquid microextraction (DLLME) and solid-phase microextraction (SPME), in food analysis and separation. The practical applications of ILs in determining toxic and harmful substances in food specimens with quite different natures are summarized and discussed. The critical function of ILs and the advantages of IL-based microextraction techniques over conventional extraction techniques are discussed in detail. Additionally, the recovery of ILs using different approaches is also presented to comply with green analytical chemistry requirements.

Ferroptosis is a type of programmed cell death dependent on iron and characterized by the accumulation of lipid peroxides. In this study, we explore the combination of a ferroptosis activator with an oncolytic vaccinia virus in tumor models. Erastin induced cell death in hepatoma, colon, and ovarian cancer cells, but not in melanoma cancer cells. Erastin, not the oncolytic vaccinia virus (OVV), induced the expression of key marker genes for ferroptosis in cancer cells. In hepatocellular carcinoma and colon cancer models, either erastin or OVV inhibited tumor growth, but a combination of the two yielded the best therapeutic effects, as indicated by inhibited tumor growth or regression and longer host survival. Immunological analyses indicate that erastin alone had little or no effect on systemic immunity or local immunity in the tumor. However, when combined with OV, erastin enhanced the number of activated dendritic cells and the activity of tumor-infiltrating T lymphocytes as indicated by an increase in IFN-γ+CD8+ and PD-1+CD8+ T cells. These results demonstrate that erastin can exert cytotoxicity on cancer cells via ferroptosis, but has little effect on immune activity by itself. However, when combined with an OVV, erastin promoted antitumoral immunity and efficacy by increasing the number of activated dendritic cells and promoting the activities of tumor specific CD8+ T cells in the tumor.

Regenerating islet-derived type 4 (REG4), a member of the calcium-dependent lectin gene superfamily, is abnormally expressed in various cancers, such as colorectal, gastric, gallbladder, pancreatic, ovarian, prostate, and lung cancer. REG4 is associated with a relatively unfavorable prognosis and clinicopathologic features in cancers, including advanced tumor and nodal stage, histological differentiation, and liver and peritoneal metastasis. Moreover, REG4-positive cancer cells show more frequent resistance to chemoradiotherapy, especially 5-FU-based chemotherapy. REG4 participates in many aspects of carcinogenesis, including cell proliferation, apoptosis, cell cycle, invasion, metastasis, and drug resistance. The underlying mechanisms are complex and involve a series of signaling mediators and multiple pathways. Thus, REG4 may be a potential diagnostic and prognostic biomarker as well as a candidate therapeutic target in cancer patients. In this review, we systematically summarize the advances about the clinical significance, biological functions, and mechanisms underlying REG4 in cancer to provide new directions for future cancer research.

The elderly population has rapidly increased in past years, bringing huge demands for elderly serving devices, especially for those with mobility impairment. Present assistant walkers designed for elderly users are primitive with limited user interactivity and intelligence. We propose a novel smart robotic walker that targets a convenient-to-use indoor walking aid for the elderly. The walker supports multiple modes of interactions through voice, gait or haptic touch, and allows intelligent control via learning-based methods to achieve mobility safety. Our design enables a flexible, initiative and reliable walker due to the following: (1) we take a hybrid approach by combining the conventional mobile robotic platform with the existing rollator design, to achieve a novel robotic system that fulfills expected functionalities; (2) our walker tracks users in front by detecting lower limb gait, while providing close-proximity walking safety support; (3) our walker can detect human intentions and predict emergency events, e.g., falling, by monitoring force pressure on a specially designed soft-robotic interface on the handle; (4) our walker performs reinforcement learning-based sound source localization to locate and navigate to the user based on his/her voice signals. Experiment results demonstrate the sturdy mechanical structure, the reliability of multiple novel interactions, and the efficiency of the intelligent control algorithms implemented. Due to their advantages of good flexibility, low cost, simple operations, and small equipment size, electrochemical sensors have been commonly employed in food safety. However, when they are applied to detect various food or drug samples, their stability and specificity can be greatly influenced by the complex matrix. By combining electrochemical sensors with molecular imprinting techniques (MIT), they will be endowed with new functions of specific recognition and separation, which make them powerful tools in analytical fields. MIT-based electrochemical sensors (MIECs) require preparing or modifying molecularly imprinted polymers (MIPs) on the electrode surface. In this review, we explored different MIECs regarding the design, working principle and functions. Additionally, the applications of MIECs in food and drug safety were discussed, as well as the challenges and prospects for developing new electrochemical methods. The strengths and weaknesses of MIECs including low stability and electrode fouling are discussed to indicate the research direction for future electrochemical sensors.

Abstract Membrane fluidity, essential for cell functions, is obviously affected by copper, but the molecular mechanism is poorly understood. Here, we unexpectedly observed that a decrease in phospholipid (PL) bilayer fluidity caused by Cu2+ was more significant than those by Zn2+ and Ca2+, while a comparable reduction occurred in the last two ions. This finding disagrees with the placement in the periodic table of Cu just next to Zn and far from Ca. The physical nature was revealed to be an anomalous attraction between Cu+ cations, as well as the induced motif of two phospholipids coupled by Cu-Cu bond (PL-diCu-PL). Namely, upon Cu2+ ion binding to a negatively charged phosphate group of lipid, Cu2+ was reduced to Cu+. The attraction of the cations then caused one Cu+ ion simultaneously binding to two lipids and another Cu+, resulting in the formation of PL-diCu-PL structure. In contrast, this attraction cannot occur in the cases of Zn and Ca ions. Remarkably, besides lipids, the phosphate group also widely exists in other biological molecules, including DNA, RNA, ADP and ATP. Our findings thus provide a new view for understanding the biological functions of copper and the mechanism underlying copper-related diseases, as well as lipid assembly.

Current simulation models are not only represented in the form of traditional data or formula for pure theory analysis but expanded to be simulation modeling assets that are featured with complicated structure, diverse behaviors, and abundant semantics. Semantic composability, therefore, receives a constantly growing attention in recent years. So far, one of the popular solutions to enhance semantic expressiveness is domain specific modeling based on general metamodeling (GMM) facilities. But for some particular domains, researchers identified the need of deeper semantic expressiveness therefore proposed domain specific metamodeling (DSMM). Hence, this paper aims to explore the underlying methodologies of DSMM for supporting deep semantic composability. Compared with several usual alternatives based on GMM, this paper applies the multi-level metamodeling architecture to create a set of metamodeling primitives using an example named SEvent. In fact, SEvent is a novel formalism that slightly extends Petri net to support continuous states transition and continuous event triggering. As a proof of concept, we concentrate on developing the textual syntax of SEvent and using it to represent torpedo's behaviors.

Abstract Background Ethanol photosynthetic production based on cyanobacteria cell factories utilizing CO2 and solar energy provides an attractive solution for sustainable production of green fuels. However, the scaling up processes of cyanobacteria cell factories were usually threatened or even devastated by biocontaminations, which restricted biomass or products accumulations of cyanobacteria cells. Thus it is of great significance to develop reliable biocontamination-controlling strategies for promoting ethanol photosynthetic production in large scales. Results The scaling up process of a previously developed Synechocystis strain Syn-HZ24 for ethanol synthesis was severely inhibited and devastated by a specific contaminant, Pannonibacter phragmitetus, which overcame the growths of cyanobacteria cells and completely consumed the ethanol accumulation in the cultivation systems. Physiological analysis revealed that growths and ethanol-consuming activities of the contaminant were sensitive to alkaline conditions, while ethanol-synthesizing cyanobacteria strain Syn-HZ24 could tolerate alkaline pH conditions as high as 11.0, indicating that pH-increasing strategy might be a feasible approach for rescuing ethanol photosynthetic production in outdoor cultivation systems. Thus, we designed and evaluated a Bicarbonate-based Integrated Carbon Capture System (BICCS) derived pH-rising strategy to rescue the ethanol photosynthetic production in non-sterilized conditions. In lab scale artificially simulated systems, pH values of BG11 culture medium were maintained around 11.0 by 180 mM NaHCO3 and air steam, under which the infection of Pannonibacter phragmitetus was significantly restricted, recovering ethanol production of Syn-HZ24 by about 80%. As for outdoor cultivations, ethanol photosynthetic production of Syn-HZ24 was also successfully rescued by the BICCS-derived pH-rising strategy, obtaining a final ethanol concentration of 0.9 g/L after 10 days cultivation. Conclusions In this work, a novel product-consuming biocontamination pattern in cyanobacteria cultivations, causing devastated ethanol photosynthetic production, was identified and characterized. Physiological analysis of the essential ethanol-consuming contaminant directed the design and application of a pH-rising strategy, which effectively and selectively controlled the contamination and rescued ethanol photosynthetic production. Our work demonstrated the importance of reliable contamination control systems and strategies for large scale outdoor cultivations of cyanobacteria, and provided an inspiring paradigm for targeting effective solutions.

Background. Proximal gastrectomy is used for the treatment of primary gastric cancer by open or laparoscopic surgery in the upper third of the stomach. Esophagogastrostomy (EG) or jejunal interposition (JI) is widely used in various reconstruction methods after proximal gastrectomy. We conducted a meta-analysis of EG and JI for treatment of gastric cancer. Materials and Methods. Background. Pseudomyxoma peritonei (PMP) is a rare neoplasm involving the peritoneum. Most PMPs are low-grade appendicular mucinous neoplasms (LAMNs). There have been no reports of PMP originating from a transverse colonic mucinous adenocarcinoma and causing metastatic mucinous adenocarcinoma. Case Presentation. We report a 46-year-old woman who presented with a right abdominal mass of more than 4-month duration. Transverse colonic mucinous adenocarcinoma, PMP, and ovarian metastatic mucinous adenocarcinoma were diagnosed. The patient’s diet was normal, and she had no abdominal pain or bloating. The abdomen mass increased in the month before treatment. After chemotherapy, the transverse colon mass and ovarian giant cyst were resected and about 2000 mL of gelatinous tumor tissue was removed. Postoperative histology confirmed PMP from the transverse colonic mucinous adenocarcinoma, ovarian metastatic mucinous adenocarcinoma, and mesocolon metastatic cancer. Multiple lung metastases appeared 8 months after surgery. The patient died 29 months after surgery because of an inability to eat and poor nutrition. A systematic literature review of the management and outcome of all known similar cases is also presented. Conclusions. This is the first report of PMP originating from a transverse colonic mucinous adenocarcinoma. It was diagnosed during resective surgery, involved ovarian metastasis, and survival was short. We did an extensive literature review in order to describe the clinical characteristics, histopathological findings, genetic profile, and potential treatments of PMP caused by nonappendiceal mucinous adenocarcinoma.

To achieve model reuse in combat effectiveness simulation systems development, cognitive decision behaviors are usually implemented using a scripting language, which is separate from the programming language used to implement simulation models. Therefore, it is desirable to establish a much better grounding for cognitive behaviors modeling. In the context of domain specific modeling, metamodeling from scratch for designing such a scripting language poses some limitations, among which is the issue of integrating various models that are represented by various customized languages with different syntax and semantics, together with a large expenditure of designing, implementing, and maintaining these languages and their supporting resources. Instead, UML profile-based metamodeling is adopted, as a lightweight extension to capture the cognitive domain specific concepts, relationships, and constraints. Moreover, a unifying framework is proposed to guide the cognitive domain specific profiles design. Upon this framework, the development process is shown through constructing an anti-submarine tactical profile in combat effectiveness simulation systems domain and the feasibility of the domain specific language is illustrated with an armed escort scenario.

Microalgae and cyanobacteria have shown significant potential for the development of the next biofuels innovation because of their own characteristics as photosynthetic microorganisms. However, it is confronted with a lot of severe challenges on the economic scaling-up of the microalgae- and cyanobacteria-based biofuels production. One of these major challenges is the lack of a reliable preventing and controlling culture system of biological contamination, which can attack the cell growth or product accumulation causing crashing effects. To increase the commercial viability of microalgae- and cyanobacteria-based biofuels production, overcoming the biological contaminations should be at the top of the priority list. Here, we highlight the importance of two categories of biological contaminations and their controlling strategies in the mass cultivations of microalgae and cyanobacteria, and outline the directions that should be exploited in the future.

Oncolytic viruses (OVs) are potent anti-cancer biologics with a bright future, having substantial evidence of efficacy in patients with cancer. Bi- and tri-specific antibodies targeting tumor antigens and capable of activating T cell receptor signaling have also shown great promise in cancer immunotherapy. In a cutting-edge strategy, investigators have incorporated the two independent anti-cancer modalities, transforming them into bi- or tri-specific T cell engager (BiTE or TriTE)-armed OVs for targeted immunotherapy. Since 2014, multiple research teams have studied this combinatorial strategy, and it showed substantial efficacy in various tumor models. Here, we first provide a brief overview of the current status of oncolytic virotherapy and the use of multi-specific antibodies for cancer immunotherapy. We then summarize progress on BiTE and TriTE antibodies as a novel class of cancer therapeutics in preclinical and clinical studies, followed by a discussion of BiTE- or TriTE-armed OVs for cancer therapy in translational models. In addition, T cell receptor mimics (TCRm) have been developed into BiTEs and are expected to greatly expand the application of BiTEs and BiTE-armed OVs for the effective targeting of intracellular tumor antigens. Future applications of such innovative combination strategies are emerging as precision cancer immunotherapies.

Purpose. Upper or proximal gastric cancer occurs in the upper third of the stomach between the cardia and a line connecting the greater and lesser curvatures. As it differs from other gastric cancers in pathology and prognosis, we evaluated patient and disease characteristics that might guide improved treatment and survival of upper gastric cancer. Methods. We conducted a retrospective analysis of 649 patients with upper gastric cancer and 1551 patients with lower gastric cancer and R0 radical surgery at our institution between January 1980 and December 2012. A fluorescence strategy for alkaline phosphatase (ALP) assay in complicated samples with high sensitivity and strong stability is developed based on an allosteric probe (AP). This probe consists of two DNA strands, a streptavidin (SA) aptamer labeled by fluorophore and its totally complementary DNA (cDNA) with a phosphate group on the 5′ end. Upon ALP introduction, the phosphate group on the cDNA is hydrolyzed, leaving the unhydrolyzed cDNA sequence for lambda exonuclease (λ exo) digestion and releasing SA aptamer for binding to SA beads, which results in fluorescence enhancement of SA beads that can be detected by flow cytometry or microscopy. We have achieved a detection limit of 0.012 U/mL with a detection range of 0.02~0.15 U/mL in buffer and human serum. These figures of merit are better than or comparable to those of other methods. Because the fluorescence signal is localized on the beads, they can be separated to remove fluorescence background from complicated biological systems. Notably, the new strategy not only applies to ALP detection with simple design, easy operation, high sensitivity, and good compatibility in complex solution, but also can be utilized in ALP-linked immunosorbent assays for the detection of a wide range of targets.

We describe a facile and low-cost approach for a flexibly integrated surface-enhanced Raman scattering (SERS) substrate in microfluidic chips. Briefly, a SERS substrate was fabricated by the electrostatic assembling of gold nanoparticles, and shaped into designed patterns by subsequent lift-up soft lithography. The SERS micro-pattern could be further integrated within microfluidic channels conveniently. The resulting microfluidic SERS chip allowed ultrasensitive in situ SERS monitoring from the transparent glass window. With its advantages in simplicity, functionality and cost-effectiveness, this method could be readily expanded into optical microfluidic fabrication for biochemical applications.

Aim. Gastrectomy with lymph node dissection is standard treatment in gastric cancer. This study aimed to explore whether preoperative investigation finds could predict lymph node metastatic scope in gastric carcinoma so that the optimal surgical procedure could be selected. Materials and Methods. Radical gastrectomy patients (n=378) were separated into two groups according to the lymph node metastatic scope. Univariate and multivariate analyses of preoperative examination results were performed to identify the predictors of metastatic scope. ROC curves were constructed, and the area under the curve (AUC) was calculated to estimate diagnostic values. Results. Serum CEA (OR: 3.73; 95% CI: 1.84–7.56; P≤0.001), tumor size (OR: 2.07; 95% CI: 1.08–3.98; P=0.03), and CT examination results (OR: 17.81; 95% CI: 9.18–34.55; P≤0.001) were identified as independent predictors. The AUC proved that they possessed significant diagnostic value. When CT examination was negative, the combination of serum CEA and tumor size showed high specificity (95.3%; 164/172), negative predictive value (92.7%; 164/177), and accuracy (89.0%; 170/191). Conclusions. Preoperative serum CEA, tumor size, and CT examination are independent predictors of lymph node metastatic scope and can be used for selecting the appropriate lymphadenectomy pattern in gastric cancer patients.

This study aimed to investigate the diversity of the Butyrivibrio group bacteria in goat rumen and its response to garlic oil (GO) supplementation as revealed by molecular analysis of cloned 16S rRNA genes. Six wethers fitted with ruminal fistulas were assigned to two groups for a cross-over design with 28-d experimental period and 14-d interval. Goats were fed a basal diet without (control) or with GO ruminal infusion (0.8 g/d). Ruminal contents were used for DNA extraction collected before morning feeding on d 28. A total bacterial clone library was firstly constructed by nearly full-length 16S rRNA gene cloned sequences using universal primers. The resulting plasmids selected by Butyrivibrio-specific primers were used to construct a Butyrivibrio group-specific bacterial clone library. Butyrivibrio group represented 12.98% and 10.95% of total bacteria in control and GO group, respectively. In libraries, clones were classified to the genus Pseudobutyrivibrio, Butyrivibrio and others within the family Lachnospiraceae. Additionally, some specific clones were observed in GO group, being classified to the genus Ruminococcus and others within the family Ruminococcaceae. Based on the criterion that the similarity was 97% or greater with database sequences, there were 29.73% and 18.42% of clones identified as known isolates (i.e. B. proteoclasticus and Ps. ruminis) in control and GO groups, respectively. Further clones identified as B. fibrisolvens (5.41%) and R. flavefaciens (7.89%) were specifically found in control and GO groups, respectively. The majority of clones resembled Ps. ruminis (98% to 99% similarity), except for Lachnospiraceae bacteria (87% to 92% similarity) in the two libraries. The two clone libraries also appeared different in Shannon diversity index (control 2.47 and GO group 2.91). Our results indicated that the Butyrivibrio group bacteria had a complex community with considerable unknown species in the goat rumen.

Treatments for triple-negative breast cancer (TNBC) are limited; intermediate-conductance calcium-activated potassium (SK4) channels are closely involved in tumor progression, but little is known about these channels in TNBC. We aimed to investigate whether SK4 channels affect TNBC. First, by immunohistochemistry (IHC) and western blotting (WB), increased SK4 protein expression in breast tumor tissues was detected relative to that in non-tumor breast tissues, but there was no apparent expression difference between various subtypes of breast cancer (p>0.05). In this study, a 3D coupled thermo-mechanical finite element model is developed to predict and analyze the defect formation during friction stir welding based on coupled Eulerian Lagrangian method. The model is validated by comparing the estimated welding temperature, processed zone shape and void size with those obtained experimentally. The results compared indicate that the simulated temperature and the data measured are in good agreement with each other. In addition, the model can predict the plasticized zone shape and the presence of a void in the weld quite accurately. However, the void size is overestimated. The effects of welding parameters and tool pin profile are also analyzed. The results reveal that welding at low welding speed or high tool rotational speed could produce a smaller void. Moreover, compared to a smooth tool pin, a featured tool pin can enhance plastic flow in the weld and achieve defect-free weldment. The results are helpful for the optimization of the welding process and the design of welding tools.

INTRODUCTION: LDH-A, the enzyme responsible for transforming pyruvate into lactate, has been demonstrated to be up-regulated in many types of cancer and to give rise to more aggressive behavior by regulating proliferation and anti-apoptosis. However, its expression in gastric cancer (GC) has not been characterized thoroughly. The purpose of this study was to clarify the expression and potential impact of LDH-A in GC. METHODS: We examined LDH-A expression by immunohistochemistry on GC tissue microarray (TMA) and using Western blot on fresh GC tissues and cell lines. Prognostic value and correlation with other clinicopathologic factors were evaluated. We transfected siRNA into GC cells against LDH-A. LDH-A was analyzed by Western blotting and real-time RT-PCR. Cell growth was evaluated in vitro and in vivo. Lactate and ATP production by cells were determined. RESULTS: There was significantly higher LDH-A expression in carcinoma than in non-neoplastic mucosa (NNM). There was a positive correlation of LDH-A expression with age, histological type and Lymph node metastases. Survival analysis demonstrated that high expression of LDH-A in GC was associated with lower overall survival (OS). When stratified by Lauren grade and histological classification, significance appeared in diffuse type and undifferentiated type GC. ObjectiveFlotillin gene is known as a tumor promoter or suppressor, depending on the tumor type or tumor stage. We aimed to investigate the clinical significance of flotillin2 protein expression in gastric cancer.MethodsWe examined flotillin2 and erbB2 levels in tissue microarray of 282 gastric cancer samples and analyzed the association between flotillin2 levels, clinicopathologic factors and prognosis. The regulation of erbB2 by flotillin2 was examined with flotillin2 siRNA-transfected gastric cancer cells.ResultsFlotillin2 partially co-localized with erbB2 at the plasma membrane as detected by confocal microscopy, levels of erbB2 were reduced after flotillin knockdown in SGC-7901 cancer cells, and the expression of flotillin2 was positively correlated with that of erbB2. In non-neoplastic gastric mucosa, flotillin2 was not expressed in the epithelial compartment. In gastric cancer, positive staining of flotillin2 was shown in 129 (45.7%) of 282 cases, also, it was significantly associated with a Lauren grade, histologic type, lymphovascular invasion and tumor location. OBJECTIVE: Legumain, a novel asparaginyl endopeptidase, has been observed to be highly expressed in several types of tumors, which may play a vital role in carcinogenesis. However, there is no study investigating the relationship among Legumain expression, clinicopathologic, biological variables and patient prognosis in gastric carcinoma. METHODS: In this study, a tissue microarray (TMA) containing 282 samples of primary gastric cancer was assessed for Legumain expression by immunohistochemistry. The TMA included 98 lymph node metastasis samples. The protein expression levels of Legumain were evaluated by Western blot analysis. RESULTS: Cytoplasmic immunoreactivity of Legumain was over-expressed in gastric cancer compared with paired normal gastric mucosa. Increased Legumain levels were significantly correlated with clinical stage, presence of distant metastasis. Legumain was significantly over-expressed in primary gastric cancer with metastasis than without metastasis. Patients with Legumain-positive localized tumors had lower 5-year overall survival (OS) than those with Legumain-negative tumors. Multivariate survival analysis showed that Legumain was an independent prognostic marker for OS (HR 1.459, 95% CI 1.251-1.703, P = 0.007). CONCLUSIONS: Legumain expression could serve as a prognostic biomarker in patients at risk of developing metastasis or recurrence with gastric carcinoma.

Friction stir lap welding of 7B04 aluminum alloy was conducted in the present paper, and the effect of welding speed on the defect features and mechanical performance of lap joints was investigated. The results indicate that the hook defect at the advancing side (AS) can reduce the effective thickness of the top sheet, and the sheet thinning level is gradually lowered by increasing the welding speed. The cold lap defect at the retreating side (RS) can result in effective thickness reduction in both top and bottom sheets, and the total height of the cold lap defect varies slightly with the welding speed. The tensile properties of the lap joints are largely related to the sheet thinning levels caused by the defects. The fracture strength of AS-loaded lap joints is progressively increased with increasing welding speed, while that of RS-loaded lap joints evolves slightly with welding speed. It is found that the affecting characteristic of loading configuration on the joint performance is also dependent on the welding speed. At lower welding speeds, the AS-loaded lap joints show lower fracture strength than the RS-loaded lap joints. When the welding speed is high, the AS-loaded lap joints present superior tensile properties to RS-loaded lap joints.

BackgroundAnterior gradient homolog 2 (AGR2) is a functional protein with critical roles in a diverse range of biological systems, including vertebrate tissue development, inflammatory tissue injury responses, and cancer progression. Clinical studies have shown that the AGR2 protein is overexpressed in a wide range of human cancers, including carcinomas of the esophagus, pancreas, breast, prostate, and lung, making the protein as a potential cancer biomarker. However, the general biochemical functions of AGR2 in human cells remain undefined, and the signaling mechanisms that drive AGR2 to inhibit p53 are still not clearly illustrated. Therefore, it is of great interest to develop molecular probes specifically recognizing AGR2 for its detection and for the elucidation of AGR2-associated molecular mechanism.Methodology/principal findingsThrough a bead-based and flow cytometry monitored SELEX technology, we have identified a group of DNA aptamers that can specifically bind to AGR2 with K(d) values in the nanomolar range after 14 rounds of selections. Aptamer C14B was chosen to further study, due to its high binding affinity and specificity. The optimized and shortened C14B1 has special G-rich characteristics, and the G-rich region of this binding motif was further characterized to reveal an intramolecular parallel G-quadruplex by CD spectroscopy and UV spectroscopy. Our experiments confirmed that the stability of the G-quadruplex structure was strongly dependent on the nature of the monovalent ions and the formation of G-quadruplex structure was also important for the binding capacity of C14B1 to the target. Furthermore, we have designed a kind of allosteric molecule beacon (aMB) probe for selective and sensitive detection of AGR2.Conclusion/significanceIn this work, we have developed new aptamer probes for specific recognition of the AGR2. Structural study have identified that the binding motif of aptamer is an intramolecular parallel G-quadruplex structure and its structure and binding affinity are strongly dependent on the nature of the monovalent ion. Furthermore, with our design of AGR2-aMB, AGR2 could be sensitively and selectively detected. This aptamer probe has great potential to serve as a useful tool for early diagnosis and prognosis of cancer and for fundamental research to elucidate the biochemical functions of AGR2.

BACKGROUND: Glioblastoma is the most common and most lethal form of brain tumor in human. Unfortunately, there is still no effective therapy to this fatal disease and the median survival is generally less than one year from the time of diagnosis. Discovery of ligands that can bind specifically to this type of tumor cells will be of great significance to develop early molecular imaging, targeted delivery and guided surgery methods to battle this type of brain tumor. METHODOLOGY/PRINCIPAL FINDINGS: We discovered two target-specific aptamers named GBM128 and GBM131 against cultured human glioblastoma cell line U118-MG after 30 rounds selection by a method called cell-based Systematic Evolution of Ligands by EXponential enrichment (cell-SELEX). These two aptamers have high affinity and specificity against target glioblastoma cells. They neither recognize normal astraglial cells, nor do they recognize other normal and cancer cell lines tested. Clinical tissues were also tested and the results showed that these two aptamers can bind to different clinical glioma tissues but not normal brain tissues. More importantly, binding affinity and selectivity of these two aptamers were retained in complicated biological environment. CONCLUSION/SIGNIFICANCE: The selected aptamers could be used to identify specific glioblastoma biomarkers. Methods of molecular imaging, targeted drug delivery, ligand guided surgery can be further developed based on these ligands for early detection, targeted therapy, and guided surgery of glioblastoma leading to effective treatment of glioblastoma.

BACKGROUND: Centromere protein A (CENP-A) plays important roles in cell-cycle regulation and genetic stability. Herein, we aimed to investigate its expression pattern, clinical significance, and biological function in hepatocellular carcinoma (HCC). METHODOLOGY/PRINCIPAL FINDINGS: CENP-A expression at the mRNA and protein levels was examined in 20 pairs of fresh HCCs and corresponding nontumor liver tissues. Immunohistochemistry for CENP-A was performed on 80 paraffin-embedded HCC specimens, and the clinical significance of its expression was analyzed. A human HCC cell line HepG2 with high abundance of CENP-A was used to study the effects of manipulating CENP-A on HCC growth. Quantitative real-time polymerase chain reaction arrays and Western blot analysis were employed to identify the cell-cycle control- and apoptosis-related genes regulated by CENP-A. The results showed that CENP-A was aberrantly overexpressed in HCCs relative to adjacent nontumor tissues. This overexpression was significantly associated with positive serum HBsAg status, increased histological grade, high Ki-67 index and P53 immunopositivity. Knockdown of CENP-A in HepG2 cells reduced cell proliferation, blocked cell cycle at the G1 phase, and increased apoptosis. The antiproliferative effects of CENP-A silencing were also observed in vivo. Conversely, CENP-A overexpression promoted HCC cell growth and reduced apoptosis. Furthermore, many genes implicated in cell-cycle regulation and apoptosis, including CHK2, P21waf1, P27 Kip1, SKP2, cyclin G1, MDM2, Bcl-2, and Bax, were deregulated by manipulating CENP-A. CONCLUSIONS/SIGNIFICANCE: Overexpression of CENP-A is frequently observed in HCC. Targeting CENP-A can inhibit HCC growth, likely through the regulation of a large number genes involved in cell-cycle progression and apoptosis, and thereby represents a potential therapeutic strategy for this malignancy.

Biodiesel is a renewable alternative to petroleum diesel fuel that can contribute to carbon dioxide emission reduction and energy supply. Biodiesel is composed of fatty acid alkyl esters, including fatty acid methyl esters (FAMEs) and fatty acid ethyl esters (FAEEs), and is currently produced through the transesterification reaction of methanol (or ethanol) and triacylglycerols (TAGs). TAGs are mainly obtained from oilseed plants and microalgae. A sustainable supply of TAGs is a major bottleneck for current biodiesel production. Here we report the de novo biosynthesis of FAEEs from glucose, which can be derived from lignocellulosic biomass, in genetically engineered Escherichia coli by introduction of the ethanol-producing pathway from Zymomonas mobilis, genetic manipulation to increase the pool of fatty acyl-CoA, and heterologous expression of acyl-coenzyme A: diacylglycerol acyltransferase from Acinetobacter baylyi. An optimized fed-batch microbial fermentation of the modified E. coli strain yielded a titer of 922 mg L(-1) FAEEs that consisted primarily of ethyl palmitate, -oleate, -myristate and -palmitoleate.
