Plant acyl-CoA-binding proteins (ACBPs) form a highly conserved protein family that binds to acyl-CoA esters as well as other lipid and protein interactors to function in developmental and stress responses. This protein family had been extensively studied in non-leguminous species such as Arabidopsis thaliana (thale cress), Oryza sativa (rice), and Brassica napus (oilseed rape). However, the characterization of soybean (Glycine max) ACBPs, designated GmACBPs, has remained unreported although this legume is a globally important crop cultivated for its high oil and protein content, and plays a significant role in the food and chemical industries. In this study, 11 members of the GmACBP family from four classes, comprising Class I (small), Class II (ankyrin repeats), Class III (large), and Class IV (kelch motif), were identified. For each class, more than one copy occurred and their domain architecture including the acyl-CoA-binding domain was compared with Arabidopsis and rice. The expression profile, tertiary structure and subcellular localization of each GmACBP were predicted, and the similarities and differences between GmACBPs and other plant ACBPs were deduced. A potential role for some Class III GmACBPs in nodulation, not previously encountered in non-leguminous ACBPs, has emerged. Interestingly, the sole member of Class III ACBP in each of non-leguminous Arabidopsis and rice had been previously identified in plant-pathogen interactions. As plant ACBPs are known to play important roles in development and responses to abiotic and biotic stresses, the in silico expression profiles on GmACBPs, gathered from data mining of RNA-sequencing and microarray analyses, will lay the foundation for future studies in their applications in biotechnology.

Salinity is a major environmental factor that constrains soybean yield and grain quality. Given our past observations using the salt-sensitive soybean (Glycine max [L.] Merr.) accession C08 on its early responses to salinity and salt-induced transcriptomic modifications, the aim of this study was to assess the lipid profile changes in this cultivar before and after short-term salt stress, and to explore the adaptive mechanisms underpinning lipid homeostasis. To this end, lipid profiling and proteomic analyses were performed on the leaves of soybean seedlings subjected to salt treatment for 0, 0.5, 1, and 2 h. Our results revealed that short-term salt stress caused dynamic lipid alterations resulting in recycling for both galactolipids and phospholipids. A comprehensive understanding of membrane lipid adaption following salt treatment was achieved by combining time-dependent lipidomic and proteomic data. Proteins involved in phosphoinositide synthesis and turnover were upregulated at the onset of salt treatment. Salinity-induced lipid recycling was shown to enhance jasmonic acid and phosphatidylinositol biosyntheses. Our study demonstrated that salt stress resulted in a remodeling of membrane lipid composition and an alteration in membrane lipids associated with lipid signaling and metabolism in C08 leaves.

Mobile edge computing is a new computing paradigm, which pushes cloud computing capabilities away from the centralized cloud to the network edge to satisfy the increasing amounts of low-latency tasks. However, challenges such as service interruption caused by user mobility occur. In order to address this problem, in this article, we first propose a multiple service placement algorithm, which initializes the placement of each service according to the user’s initial location and their service requests. Furthermore, we build a network model and propose a based on Lyapunov optimization method with long-term cost constraints. Considering the importance of user mobility, we use the Kalman filter to correct the user’s location to improve the success rate of communication between the device and the server. Compared with the traditional scheme, extensive simulation results show that the dynamic service migration strategy can effectively improve the service efficiency of mobile edge computing in the user’s mobile scene, reduce the delay of requesting terminal nodes, and reduce the service interruption caused by frequent user movement.

Ovarian cancer (OC) is a lethal disease occurring in women worldwide. Due to the lack of obvious clinical symptoms and sensitivity biomarkers, OC patients are often diagnosed in advanced stages and suffer a poor prognosis. Circulating tumor cells (CTCs), released from tumor sites into the peripheral blood, have been recognized as promising biomarkers in cancer prognosis, treatment monitoring, and metastasis diagnosis. However, the number of CTCs in peripheral blood is low, and it is a technical challenge to isolate, enrich, and identify CTCs from the blood samples of patients. This work develops a simple, effective, and inexpensive strategy to capture and identify CTCs from OC blood samples using the folic acid (FA) and antifouling-hydrogel-modified fluorescent-magnetic nanoparticles. The hydrogel showed a good antifouling property against peripheral blood mononuclear cells (PBMCs). The FA was coupled to the hydrogel surface as the targeting molecule for the CTC isolation, held a good capture efficiency for SK-OV-3 cells (95.58%), and successfully isolated 2–12 CTCs from 10 OC patients’ blood samples. The FA-modified fluorescent-magnetic nanoparticles were successfully used for the capture and direct identification of CTCs from the blood samples of OC patients.

Based on the electrical conductivity model built for graphene oxide, the thermal crosstalk effects of resistive random access memory (RRAM) with graphene electrode and Pt electrode are simulated and compared. The thermal crosstalk effects of Pt-RRAM with different metal oxides of TiOx, NiOx, HfOx, and ZrOx are further simulated and compared to guide its compatibility design. In the Pt-RRAM array, the distributions of oxygen vacancy density and temperature are obtained, and the minimum spacing between adjacent conduction filaments to avoid device operation failure is discussed. The abovementioned four metal oxides have different physical parameters such as diffusivity, electrical conductivity, and thermal conductivity, from which the characters of the RRAMs based on one of the oxides are analyzed. Numerical results reveal that thermal crosstalk effects are severe as the spacing between adjacent conduction filaments is small, even leading to the change of logic state and device failure.

Tetralogy of Fallot (TOF) is a congenital heart disease characterized by abnormal cardiomyocyte differentiation in the right ventricular outflow tract (RVOT), and HA117 is a novel long noncoding RNA (lncRNA) with anti-differentiation roles.To investigate the potential association of HA117 with TOF, we collected 84 RVOT tissues from patients with TOF. We determined the expression of HA117 in RVOT samples from TOF patients and collected clinical data to conduct a cross-sectional and short-term follow-up study.McGoon ratio, Nakata index, and left ventricular end-diastolic volume index (LVEDVI) were negatively correlated with the expression of HA117 based on subgroup analysis, correlation analysis and logistic regression analysis. Additionally, cardiopulmonary bypass (CPB) time and ICU stay were longer in patients with higher expression of HA117 than in patients with lower expression of HA117. Furthermore, percentage improvement in SPO2 was significantly reduced in patients with increased HA117 expression at 6 months after surgery.Our results suggested that the increased expression of the novel lncRNA HA117 is a risk factor for unfavorable McGoon ratio, Nakata index and LVEDVI in TOF patients. Abstract Background Residential instability during pregnancy has been linked to poor health outcomes. As a first step toward providing better health care to pregnant migrant women, the size and characteristics of this population and factors associated with mobility during pregnancy should be studied. Methods Using the “Monitoring Data of Chinese Migrants” for 2012, from the Chinese National Population and Family Planning Commission, this study explored mobility patterns during pregnancy and associated factors among migrants within China. From a library of 158,556 participants, two subsamples were selected. Percentages, with chi-squared tests, and means and standard deviations, with ANOVAs, were adopted to describe mobility patterns during pregnancy (always staying in sending area, mainly staying in sending area, mainly staying in receiving area, and always staying in receiving area) and delivery location choice. Logistic regression analysis was used to explore the associated factors. Results We found that the percentage of migrants always or mainly staying in receiving areas during pregnancy rose from nearly 40% in 1985 to more than 80% in 2012, while the percentage of migrants who were mobile between receiving and sending areas during pregnancy fluctuated between 30 and 40% before 1995, and between 40 and 45% after 1995, decreasing to around 40% after 2008. The percentage of respondents who chose to deliver in receiving areas fluctuated but increased from 10% in 1985 to more than 50% in 2011. Among respondents who had delivered during the last year of the survey period, families with older pregnant women (OR = 1.09, 95% CI 1.05–1.13), their own housing (OR = 5.66, 95% CI 2.45–13.05), longer time in the receiving area (OR = 1.14, 95% CI 1.09–1.20), and strong will to integrate (OR = 1.32, 95% CI 1.15–1.51) always stayed in the receiving area during pregnancy, rather than the sending area, and families with broadly similar characteristics were inclined to choose the receiving area for their delivery. Conclusions The mobility patterns of pregnant migrant women in China have been changing in recent years, with the percentage of them staying in receiving areas during pregnancy and delivering there increasing. Individual and family characteristics were also associated with mobility patterns and delivery location choice.

To mitigate global warming and the resulting climate risk, many countries have accelerated the optimization of industrial structures and mixture of energy type in an attempt to achieve carbon neutrality by the second half of the 21st century. Here, we present the first assessment of the quantitative benefits of population exposure to extreme heat (defined by the heat index) during 2040–2049 under two scenarios of global carbon neutrality by 2060 and 2050, i.e. moderate green (MODGREEN) and strong green (STRGREEN) recovery scenarios, relative to the baseline scenario of Shared Socioeconomic Pathway (SSP) 2–4.5. Global mean extreme heat days increase by 12.1 d yr ^−1 (108%) during 2040–2049 under the SSP2-4.5 scenario relative to the historical period (1995–2014). The aggravating extreme heat events could be mitigated by as much as 12% and 18% during 2040–2049 under the MODGREEN and STRGREEN scenarios, respectively. Following the changes in extreme heat days, global population exposure to extreme heat is mitigated by 27.3 billion person-days (7%) in the MODGREEN scenario and 39.9 billion person-days (11%) in the STRGREEN scenario during 2040–2049 relative to the SSP2-4.5 scenario. Such benefits from these low-carbon policies are larger in regional hotspots, including India and Northern Africa, which have experienced high population growth and have extremely limited medical infrastructure. Moreover, an early carbon neutrality (2050 vs 2060) could avoid 12.6 billion person-days exposure to extreme heat during 2040–2049. Our results provide an important scientific support for governments to drive early policymaking for climate change mitigation.

The land ecosystems of China are estimated to provide an important sink for the increased atmospheric carbon dioxide (CO _2 ), but are undermined by severe ozone (O _3 ) pollution. Mitigation of O _3 damage to ecosystems remains a challenge considering that O _3 precursors are emitted from a wide range of anthropogenic sectors and O _3 formations are also affected by regional transport. Here, we combine chemical transport and dynamic vegetation models to quantify the benefits of sectoral and regional emission controls for the recovery of gross primary productivity (GPP) in the Yangtze River Delta (YRD). For sectoral emission controls, the largest mitigation of O _3 damage to GPP in YRD by 3.1 ± 0.4 and 2.2 ± 0.2 Gg[C] d ^−1 with 50% reductions in the emissions from industry and transportation sectors, respectively. For regional emission controls, reducing 50% anthropogenic emissions outside YRD can mitigate GPP losses by 18.6 ± 3.5 Gg[C] d ^−1 , larger than the recovery of 10.1 ± 1.6 Gg[C] d ^−1 by the 50% reductions of anthropogenic emissions within YRD. Moreover, summer months, especially July are the best period for GPP recovery from anthropogenic emission controls. Our results highlight the importance of sectoral and regional emission controls to mitigate O _3 damage to ecosystem productivities in YRD.

Conventional dealloying methods to produce bicontinuous open nanoporous structures for catalysis are limited to very few alloys and produce chemical waste. Here, the authors develop a green process, vapor-phase dealloying, to selectively remove high partial vapor pressure components and create nanoporosity in a wide range of alloys.

Tuning the ratio of facets in materials can affect catalytic activity but methods to achieve this can be difficult to control. Here, using surfactants during dealloying, the authors prepare nanoporous gold with controlled facet ratios and show how it can significantly improve electrocatalytic activity.

The transfer of mobile genetic elements (MGEs) has been known as a strategy adopted by organisms for survival and adaptation to the environment. The rhizosphere, where microbes and plants coexist, is a hotspot of MGE transfers. In this review, we discuss the classic mechanisms as well as novel mechanisms of MGE transfers in the rhizosphere. Both intra-kingdom and cross-kingdom MGE transfers will be addressed. MGE transfers could be ancient events which drove evolution or recurrent events which regulate adaptations. Recent findings on MGE transfers between plant and its interacting microbes suggest gene regulations brought forth by such transfers for symbiosis or defense mechanisms. In the natural environment, factors such as temperature and soil composition constantly influence the interactions among different parties in the rhizosphere. In this review, we will also address the effects of various environmental factors on MGE transfers in the rhizosphere. Besides environmental factors, plant root exudates also play a role in the regulation of MGE transfer among microbes in the rhizosphere. The potential use of microbes and plants for bioremediation will be discussed.

This study assessed three commercially available cell-free DNA (cfDNA) extraction kits and the impact of a PEG-based DNA cleanup procedure (DNApure) on cfDNA quality and yield. Six normal donor urine and plasma samples and specimens from four pregnant (PG) women carrying male fetuses underwent extractions with the JBS cfDNA extraction kit (kit J), MagMAX Cell-Free DNA Extraction kit (kit M), and QIAamp Circulating Nucleic Acid Kit (kit Q). Recovery of a PCR product spike-in, endogenous TP53, and Y-chromosome DNA was used to assess kit performance. Nucleosomal-sized DNA profiles varied among the kits, with prominent multi-nucleosomal-sized peaks present in urine and plasma DNA isolated by kits J and M only. Kit J recovered significantly more spike-in DNA than did kits M or Q (p < 0.001) from urine, and similar amounts from plasma (p = 0.12). Applying DNApure to kit M- and Q-isolated DNA significantly improved the amplification efficiency of spike-in DNA from urine (p < 0.001) and plasma (p ≤ 0.013). Furthermore, kit J isolated significantly more Y-chromosome DNA from PG urine compared to kit Q (p = 0.05). We demonstrate that DNApure can provide an efficient means of improving the yield and purity of cfDNA and minimize the effects of pre-analytical biospecimen variability on liquid biopsy assay performance.

Frequent and severe PM _2.5 pollution over China seriously harms natural environment and human health. Changes in meteorological conditions in recent decades have been recognized to contribute to the long-term increase in PM _2.5 pollution in North China (NC). However, the dominant climatic factors driving the interdecadal changes of the weather conditions conducive to PM _2.5 pollution remain unclear. Here we identify a potential global teleconnection mechanism: the decadal reduction in European aerosol emissions since the 1980s may have partially contributed to the interdecadal increase in weather conditions conducive to PM _2.5 pollution in NC, measured by an Emission-weighted Air Stagnation Index (ASI _E ) that increases at a rate of 6.2% decade ^−1 (relative to the 1981–1985 level). By regression analysis, we show that the decreased European aerosol loadings can warm the lower atmosphere and induce anomalous ascending motion in Europe, which potentially stimulates two anomalous Rossby wave trains in the upper troposphere travelling eastward across Eurasia. The teleconnection patterns project on NC by weakening the near-surface horizontal dispersion, which may be favorable to the increase in local ASI _E and air pollution build-up. The suggested mechanism is further supported by the results from a set of large-ensemble simulations, showing that the European aerosol emission decline since the 1980s excites similar local heating and ascending motion and leads to increasing trends of 0.1–0.5 μ g m ^−3 (38 year) ^−1 in surface sulfate concentrations over most of NC. This proposed ‘West-to-East Aerosol-to-Aerosol’ teleconnection mechanism helps resolve opposite views on the impact of global versus local aerosol forcing on PM _2.5 pollution weather in NC. The policy implication is that the sustained decline in European aerosol emissions in coming decades, in conjunction with unabated global and regional warming, could further exacerbate air pollution in NC, thus imposing stronger pressure to reduce local emission sources quicker and deeper.

The soybean (Glycine max (L.) Merr.) is an important oil and food crop. Its growth and development is regulated by complex genetic networks, and there are still many genes with unknown functions in regulation pathways. In this study, GmNMHC5, a member of the MADS-box protein family, was found to promote flowering and maturity in the soybean. Gene expression profiling in transgenic plants confirmed that the 35S:GmNMHC5 T3 generation had early flowering and precocity. We used CRISPR-Cas9 to edit GmNMHC5 and found that late flowering and maturity occurred in Gmnmhc5 lines with stable inheritance. Remarkably, in the 35S:GmNMHC5 plants, the expression of flowering inhibitors GmFT1a and GmFT4 was inhibited. In addition, overexpression of GmNMHC5 in ft-10 (a late flowering Arabidopsis thaliana mutant lacking Flowering Locus T (FT) function) rescued the extremely late-flowering phenotype of the mutant A. thaliana. These results suggest that GmNMHC5 is a positive transcription factor of flowering and maturity in the soybean, which has a close relationship with FT homologs in the flowering regulation pathway. This discovery provides new ideas for the improvement of the flowering regulation network, and can also provide guidance for future breeding work.

Hirschsprung Disease (HSCR) and/or hypoganglionosis are common pediatric disorders that arise from developmental deficiencies of enteric neural crest cells (ENCCs). Retinoid acid (RA) signaling has been shown to affect neural crest (NC) development. However, the mechanisms underlying RA deficiency-induced HSCR or hypoganglionosis are not well-defined. In this report, we found that in HSCR patient bowels, the RA nuclear receptor RARα and its interacting coregulator CREB-binding protein (CBP) were expressed in enteric neural plexuses in the normal ganglionic segment. However, the expression of these two genes was significantly inhibited in the pathological aganglionic segment. In a Xenopus laevis animal model, endogenous RARα interacted with CBP and was expressed in NC territory. Morpholino-mediated knockdown of RARα blocked expression of the NC marker genes Sox10 and FoxD3 and inhibited NC induction. The morphant embryos exhibited reduced nervous cells in the gastrointestinal anlage, a typical enteric nervous deficiency-associated phenotype. Injection of CBP mRNA rescued NC induction and reduced enteric nervous deficiency-associated phenotypes. Our work demonstrates that RARα regulates Sox10 expression via CBP during NC induction, and alteration of the RA-CBP signaling pathway may contribute to the development of enteric nervous system disorders.

Objective: To evaluate the frequency and clinical features of restless legs syndrome (RLS) in a group of Chinese patients with amyotrophic lateral sclerosis (ALS).Methods: 109 Patients included in this study fulfilled the revised El Escorial diagnostic criteria for clinically definite, probable and lab-supported probable ALS, and a group of 109 control subjects was matched for age and sex to the ALS group. Disease severity was assessed by the revised ALS functional rating scale (ALSFRS-R). The diagnosis of RLS was made according to the criteria of the International RLS Study Group. Other characteristics including sleep quality, excessive daytime sleepiness (EDS), REM sleep behavior disorder (RBD), depression and anxiety were also evaluated in ALS patients.Results: RLS was significantly more frequent in ALS patients than in control subjects (14.6 vs. 0.9%; P < 0.05). Compared to those without RLS, ALS patients with RLS reported a higher frequency of anxiety and EDS. ALS patients with RLS showed more severe legs dysfunction. EDS and legs function scores of the ALSFRS-R were independent factors significantly associated with RLS in ALS patients.Conclusions: Our findings suggest that Chinese ALS patients exhibit a high frequency of RLS symptoms and that these patients may benefit from recognition of the condition and optimized management of its symptoms. Moreover, ALS patients might cause circadian rhythms disturbance and our study further supports that ALS is a heterogeneous disorder involving multiple systems; further studies are needed to confirm these preliminary findings.

Realization of open online control of diverse in-situ sensors is a challenge. This paper proposes a Cyber-Physical Geographical Information Service-enabled method for control of diverse in-situ sensors, based on location-based instant sensing of sensors, which provides closed-loop feedbacks. The method adopts the concepts and technologies of newly developed cyber-physical systems (CPSs) to combine control with sensing, communication, and computation, takes advantage of geographical information service such as services provided by the Tianditu which is a basic geographic information service platform in China and Sensor Web services to establish geo-sensor applications, and builds well-designed human-machine interfaces (HMIs) to support online and open interactions between human beings and physical sensors through cyberspace. The method was tested with experiments carried out in two geographically distributed scientific experimental fields, Baoxie Sensor Web Experimental Field in Wuhan city and Yemaomian Landslide Monitoring Station in Three Gorges, with three typical sensors chosen as representatives using the prototype system Geospatial Sensor Web Common Service Platform. The results show that the proposed method is an open, online, closed-loop means of control.

Stargardt disease is the most common cause of juvenile macular dystrophy. Five subjects from a two-generation Chinese family with Stargardt disease are reported in this study. All family members underwent complete ophthalmologic examinations. Patients of the family initiated the disease during childhood, developing progressively impaired central vision and bilateral atrophic macular lesions in the retinal pigmental epithelium (RPE) that resembled a "beaten-bronze" appearance. Peripheral venous blood was obtained from all patients and their family members for genetic analysis. Exome sequencing was used to analyze the exome of two patients II1, II2. A total of 50709 variations shared by the two patients were subjected to several filtering steps against existing variation databases. Identified variations were verified in all family members by PCR and Sanger sequencing. Compound heterozygous variants p.Y808X and p.G607R of the ATP-binding cassette, sub-family A (ABC1), member 4 (ABCA4) gene, which encodes the ABCA4 protein, a member of the ATP-binding cassette (ABC) transport superfamily, were identified as causative mutations for Stargardt disease of this family. Our findings provide one novel ABCA4 mutation in Chinese patients with Stargardt disease.
