The ultrasound-assisted extraction technique was used for extracting polysaccharides of Tricholoma matsutake (PTM) in this work. The effects of different parameters, i.e., solid-to-liquid ratio, ultrasonic power, extraction time, and extraction temperature on the PTM yield were optimized using a rotatable, orthogonal central composite design. In the derived optimal conditions, the PTM yield was 16.97 ± 0.06%, which corresponded well with the regression model predicted value of 16.85%. The extracted PTM was purified, and the structural characteristics and antioxidant activities were investigated. The PTM was found to be composed mainly of fructose, mannose, glucose, and galactose with a molar molecular ratio of 1:2.25:9.35:5, and an average molecular weight of 90,100 Da. High antioxidant effects of PTM were seen in the tests of DPPH, hydroxyl, and ABTS radical scavenging, along with a moderate superoxide radical scavenging rate.

Abstract Herbivorous insects employ an array of salivary proteins to aid feeding. However, the mechanisms behind the recruitment and evolution of these genes to mediate plant-insect interactions remain poorly understood. Here, we report a potential horizontal gene transfer (HGT) event from bacteria to an ancestral bug of Eutrichophora. The acquired genes subsequently underwent duplications and evolved through co-option. We annotated them as horizontal-transferred, Eutrichophora-specific salivary protein (HESPs) according to their origin and function. In Riptortus pedestris (Coreoidea), all nine HESPs are secreted into plants during feeding. The RpHESP4 to RpHESP8 are recently duplicated and found to be indispensable for salivary sheath formation. Silencing of RpHESP4-8 increases the difficulty of R. pedestris in probing the soybean, and the treated insects display a decreased survivability. Although silencing the other RpHESPs does not affect the salivary sheath formation, negative effects are also observed. In Pyrrhocoris apterus (Pyrrhocoroidea), five out of six PaHESPs are secretory salivary proteins, with PaHESP3 being critical for insect survival. The PaHESP5, while important for insects, no longer functions as a salivary protein. Our results provide insight into the potential origin of insect saliva and shed light on the evolution of salivary proteins.

Abstract Background Diabetic kidney disease (DKD) is a prevalent complication of diabetes and the leading cause of end-stage renal disease. Recent evidence suggests that total flavonoids of Astragalus (TFA) has promising effects on diabetes; however, its influence on DKD and the underlying mechanism remains unclear. Methods In this study, we induced the DKD model using streptozotocin (STZ) in male C57BL/6J mice and utilized glomerular endothelial cell (GEC) lines for in vitro investigations. We constructed a network pharmacology analysis to understand the mechanism of TFA in DKD. The mechanism of TFA action on DKD was investigated through Western blot analysis and multi-immunological methods. Results Our findings revealed that TFA significantly reduced levels of urinary albumin (ALB). Network pharmacology and intracellular pathway experiments indicated the crucial involvement of the PI3K/AKT signaling pathway in mediating these effects. In vitro experiments showed that TFA can preserve the integrity of the glomerular filtration barrier by inhibiting the expression of inflammatory factors TNF-alpha and IL-8, reducing oxidative stress. Conclusion Our findings demonstrated that TFA can ameliorates the progression of DKD by ameliorating renal fibrosis and preserving the integrity of the kidney filtration barrier. These results provide pharmacological evidence supporting the use of TFA in the treatment of kidney diseases.

Species of the family Prosthogonimidae are considered the most pathogenic trematodes of poultry and wild birds worldwide, causing heavy economic losses in many countries. Prosthogonimosis was a common parasitic disease of Grus japonensis (Müller, 1776) which caused inflammation of the cloaca and bursa of Fabricius and even death. Morphological identifications of Prosthogonimus species are easily confusing; therefore, molecular characterization is used for discrimination. The present study was conducted to identify Prosthogonimus species at Zhalong National Nature Reserve, northeast of China. Considering the morphological variability and wide host range of individual Prosthogonimus species, a combination of both morphological and molecular analyses is indispensable for the valid identification of this parasite and the internal transcribed spacer (ITS) region was amplified for the sequence analysis and phylogenetic analysis. The results of molecular analysis together with phylogenetic reconstruction indicated that the Prosthogonimus pellucidus (von Linstow, 1873) in this study form a single cluster with P. pellucidus, revealing potentially high diversity within the genus Prosthogonimus. Classification of Prosthogonimus species seems to be unrelated to the host and may be related to geographical location. These data provide a significant resource of molecular markers for studying the taxonomy, population genetics, and systematics of Prosthogonimidae.

Silent information regulator sirtuin 1 (SIRT1) is an NAD+-dependent deacetylase with potent anti-arterial aging activities. Its protective function in aging-related diseases has been extensively studied. In the microcirculation, SIRT1 plays a crucial role in preventing microcirculatory endothelial senescence by suppressing inflammation and oxidative stress while promoting mitochondrial function and optimizing autophagy. It suppresses hypoxia-inducible factor-1α (HIF-1α)-mediated pathological angiogenesis while promoting healthy, physiological capillarization. As a result, SIRT1 protects against microvascular dysfunction, such as diabetic microangiopathy, while enhancing exercise-induced skeletal muscle capillarization and energy metabolism. In the brain, SIRT1 upregulates tight junction proteins and strengthens their interactions, thus maintaining the integrity of the blood−brain barrier. The present review summarizes recent findings on the regulation of microvascular function by SIRT1, the underlying mechanisms, and various approaches to modulate SIRT1 activity in microcirculation. The importance of SIRT1 as a molecular target in aging-related diseases, such as diabetic retinopathy and stroke, is underscored, along with the need for more clinical evidence to support SIRT1 modulation in the microcirculation.
