Abstract

Transcription factor pancreatic and duodenal homeobox-1 (PDX-1) plays an essential role in pancreatic development, β-cell differentiation, maintenance of normal β-cell function and tumorigenesis. PDX-1 expression is tightly controlled through a variety of mechanisms under different cellular contexts. We report here that overexpression of Notch1 intracellular domain (NICD), an activated form of Notch1, enhanced PDX-1 expression in both PDX-1 stable HEK293 cells and mouse insulinoma β-TC-6 cells, while NICD shRNA inhibited the enhancing effect. NICD-enhanced PDX-1 expression was accompanied by increased insulin expression/secretion and cell proliferation in β-TC-6 cells, which was reversed by NICD shRNA. Cre activation-induced specific expression of NICD in islet β cells of transgenic βNICD+/+ mice induced increased expression of PDX-1, insulin and proliferating cell nuclear antigen (PCNA) and decreased expression of p27 with accompanied fasting hyperinsulinemia and hypoglycemia and altered responses to intraperitoneal glucose tolerance test. Systemically delivered NICD shRNA suppressed islet expression of PDX-1 and reversed the hypoglycemia and hyperinsulinemia. Moreover, expression levels of NICD were correlated with those of PDX-1 in human pancreatic neuroendocrine tumor. Thus, Notch1 acts as a positive regulator for PDX-1 expression, cooperates with PDX-1 in the development of insulin overexpression and islet cell neoplasia and represents a potential therapeutic target for islet neoplasia.

Abstract

Melatonin (MT) is a growth regulator and antioxidant that can resist peroxidation damage on plants caused by environmental stresses. In this study, the alleviation effects of melatonin on alfalfa under salt stress were investigated in terms of photosynthesis, antioxidant enzymes, and osmoregulation. The alfalfa seedlings were cultured in 200 mM NaCl Hoagland solution. Five levels of MT (0, 0.1, 0.2, 0.3, and 0.4 mM) were applied as a foliar spray. Generally, the foliar spray of MT increased root length, root surface area, height, leaf length and width, aerial and root biomass, SPAD readings, the content of proline and soluble protein, and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT). Malonaldehyde (MDA) content was decreased by MT foliar spray. The beneficial effects of MT on alfalfa under salt stress were dosage-dependent, and excessive MT levels inhibited alfalfa growth. The alleviating effects of MT on salt stress were more pronounced at 0.3 mM MT. This study suggested that exogenous MT foliar spray at appropriate levels can ameliorate the adverse effects of salt stress on alfalfa seedlings. [ABSTRACT FROM AUTHOR]

Abstract

Fertilizer management is one of the easiest and most practical ways of combating salt stress. This study was done to evaluate the alleviative effects of nitrogen and phosphorus on the growth and salt tolerance of salt-affected sorghum. A controlled study organized in a randomized block design with three replications was conducted, testing three nitrogen rates (N0: 0 kg ha−1, N1: 180 kg ha−1, N2: 360 N kg ha−1) and phosphorus rates (P0: 0 P2O5 kg ha−1, P1: 60 P2O5 kg ha−1, P2: 120 P2O5 kg ha−1). Nitrogen and phosphorus application had positive effects on morphological indexes (plant height, stem diameter), some physiological and biochemical attributes (the content of proline and soluble protein, and the activities of superoxide dismutase, catalase, peroxidase, and ascorbate peroxidase), and aerial biomass (fresh and dry weight) of sorghum grown in saline soils. Reactive oxygen species accumulation and cell membrane damage were decreased with the application of nitrogen and phosphorus. Compared with sole fertilizer, the combined application of nitrogen and phosphorus showed better performance in alleviating salt damage on sorghum. Despite the fact that the maximum of most of the measured parameters and the minimum of reactive oxygen species accumulation and cell membrane damage were generally obtained at N1P1 and N2P2 treatment, N1P1 was recommended to be the suitable treatment considering economic benefits and environmental protection. Abstract

Plant density is one of the most important factors influencing the physiological, morphological characteristics, and yield of sesame. Sesame is an important oilseed and cash crop production in Sudan. The effects of different rows spacing on the growth attributes, yield and yield compound on three sesame seeds varieties were investigated at rainfall conditions in semi-arid regions. Two field experiments were conducted in 2014-2015 and 2015-2016. Three varieties were planted at four rows spacing [5(control), 10, 20 and 30cm]. The experiment was laid out in RCBD in split plots with three replications. There were significant differences between row spacing, varieties and interaction between row spacing and varieties for all parameters except the 1000 seeds weight. Moreover, Abo Nama has recorded the highest plants, number of branches and number of capsules per plant, while the abo Radom recorded the highest number of seeds per capsule and seed yield, and abo sofa had the highest weight of 1000 seeds. Planted varieties at the 5 cm between rows can increase the seed yield (210.18 kg/h). The interaction between Abo Radom and control recorded the high seed yield by 265.30 kg per h-1. This study suggests that the appropriate rows spacing can actively increase the yield of the sesame plant.

98. Foliar application of 5-aminolevulinic acid alleviated high temperature and drought stresses on wheat plants at seedling stage

Abstract

Wheat (Triticum aestivum L.) seedling establishment is very sensitive to temperature and drought stresses. The present study was conducted to investigate the effects of foliar exogenous 5-aminolevulinic acid (5-ALA) application on wheat seedling under different temperature (25, 30, and 35 ℃) and water regimes (normal and 65% watering). Five different levels of 5-ALA (0, 25, 50, 100 and 150 mg L-1) were applied as foliar application. Leaf area, stem diameter, shoot and root lengths, fresh and dry weights of shoot and root, and physiological parameters were measured. Drought stress (W2) decreased leaf area by 14.9% compared with normal watering (W1). High temperature (35 ℃) increased malondialdehyde (MDA) content by 72.1% compared with control (25 ℃). 35 ℃ with W2 reduced root and shoot dry weights by 71.8% and 51.9% relative to 25 ℃ and W1. At 35 ℃, 25 mg L-1 5-ALA increased root length by 17.1% and 3.4% at W1 and W2, respectively. 150 mg L-1 5-ALA increased root fresh weight by 101.0% compared with 0 mg L-1 5-ALA at W1 and 35 ℃. The highest protein content (8.35 mg g-1 FW) was achieved at the interaction between W2, 35 ℃ and 50 mg L-1 5-ALA. At W2 and 35 ℃, 150 mg L-1 5-ALA increased proline content by 66.5% relative to 0 mg L-1. The results of the present study suggest that foliar application of 5-ALA could be useful to alleviate the temperature and/or drought stresses of wheat seedling.
