53. Neutrophils in pancreatic cancer: Potential therapeutic targets

Abstract

Pancreatic cancer is a digestive system malignancy and poses a high mortality worldwide. Traditionally, neutrophils have been thought to play a role in acute inflammation. In contrast, their importance during tumor diseases has been less well studied. Generally, neutrophils are recruited into the tumor microenvironment and exert inflammation and tumor-promoting effects. As an essential part of the tumor microenvironment, neutrophils play diverse roles in pancreatic cancer, such as angiogenesis, progression, metastasis and immunosuppression. Additionally, neutrophils can be a new potential therapeutic target in cancer. Inhibitors of cytokines, chemokines and neutrophil extracellular traps can exert antitumor effects. In this review, we describe the role of neutrophils in the development and progression of pancreatic cancer, discuss their potential as therapeutic targets, and aim to provide ideas for improving the prognosis of patients with this malignant tumor disease.

Abstract

Cordycepin is a nucleoside molecule found in Cordyceps sinensis and can be obtained through chemical synthesis and biotransformation. Cordycepin has been extensively studied and has been shown to have antitumour activity. This activity includes effects on the autophagy process and inhibition of the MAPK/ERK and Hedgehog pathways. Ultimately, the inhibitory effect of cordycepin on tumour cells is due to the interplay of these effects. Cordycepin was shown to enhance the therapeutic effects of radiotherapy. There is increasing evidence indicating that cordycepin plays an anticancer role in the treatment of various cancers. The present review aims to provide a clear understanding of the antitumour mechanisms of cordycepin and discuss its present application in the treatment of tumours. This information can be an important theoretical basis and provide clinical guidance for the further development of cordycepin as an antitumour drug. [ABSTRACT FROM AUTHOR]

Abstract

Objective: To investigate the expression, function and clinical significance of maternal leucine zipper kinase (MELK) in pancreatic cancer. Methods: The expression of MELK in pancreatic cancer was analyzed and the possible molecular mechanism of MELK in pancreatic cancer was predicted by bioinformatics. The clinical data of 45 patients with pancreatic cancer undergoing surgical treatment in the First Hospital of Lanzhou University were retrospectively analyzed. Immunohistochemistry was used to explore the MELK expression levels in pancreatic cancer tissues and normal para-cancerous tissues, and its relationship with clinical pathological characteristics was also analyzed. The postoperative recurrence time of patients was collected through outpatient records and telephone follow-up to explore the relationship between MELK expression and postoperative recurrence time. Results: MELK mRNA was highly expressed in pancreatic cancer tissues and correlated with the overall survival rate and postoperative recurrence time. Multivariate Cox analysis showed that the MELK gene could be used as an independent prognostic factor in patients with pancreatic cancer. Functional enrichment suggests that MELK is involved in multiple cancer-related functional pathways and is positively correlated with the pathogenic molecules of pancreatic cancer. Immunohistochemistry results showed that MELK in pancreatic cancer tissues was significantly higher than that in adjacent normal tissues. The expression of MELK in pancreatic cancer tissues was correlated with tumor size. Conclusion: The MELK gene is highly expressed in pancreatic cancer tissues and is associated with patients' prognosis. MELK plays a role in the progression of pancreatic cancer and is expected to become a potential biomarker and therapeutic target of pancreatic cancer. [ABSTRACT FROM AUTHOR]
