The pollution of heavy metal ions can cause damage to the human body through food, so developing a new biocompatible material that can remove the damage of heavy metal ions has a good application prospect. In this study, we obtained a new homogeneous polysaccharide composed of seven monosaccharides from Zingiber strioatum by using the method of separation and purification of polysaccharide. The results of adsorption behavior showed that the concentration, temperature and pH value could affect the adsorption effect of Zingiber strioatum polysaccharide (ZSP). Through model fitting of the data of adsorption time and metal concentration, the pseudo second-order kinetic model can well describe the kinetics of the adsorption process, and the adsorption isotherm data fit well with the Langmuir model. In the preliminary research results of adsorption mechanism, SEM showed the appearance of ZSP as flake and porous surface; EDX analysis confirmed the metal adsorption capacity of ZSP. Meanwhile, XPS and FT-IR further clarified the adsorption based on functional groups composed of C and O. The cells preprotected by ZSP can resist heavy metal ions. The above results show that ZSP can be used as a new macromolecule to bind heavy metal ions, which can broaden the research scope of polysaccharides in contaminated food systems. In this study, we investigated the protective efficacy of extracellular polysaccharide from Cordyceps militaris (CEP-I) in liver and kidney and their regulating effect on gut microbiota against Pb-induced toxicity in vivo. The results indicated that CEP-I could reduce the Pb2+ content and organ index of liver and kidney in mice. Besides, biochemical analysis showed that CEP-I could improve the activity of glutathione peroxidase (GSH-Px), malondialdehyde (MDA) and superoxide dismutase (SOD) in serum and organs, restore the physiological indexes of total protein (TP), albumin (ALB), blood urea nitrogen (BUN) and creatinine (CRE) in serum and decrease the enzyme activity of lactate dehydrogenase (LDH) and aspartate aminotransferase (AST) in the liver and kidney of mice poisoned by Pb2+. This indicated that CEP-I has a protective effect on organs against damage in mice. In addition, CEP-I could regulate the expression of key proteins in the Nrf2 signaling pathway, including NF-E2-related factor 2 (Nrf2), Kelch-like ECH-associated protein-1 (Keap1), Heme oxygenase (HO-1) and NAD(P)H: quinone oxidoreductase 1 (NQO1). Furthermore, the intestinal flora analysis results indicated that CEP-I also has the capacity to regulate the intestinal flora imbalance caused by Pb2+ in poisoned mice. In conclusion, we hope that this study can provide theoretical basis for the treatment of tissue damage induced by Pb2+.

Trichoderma asperellum ZZY has good tolerance to Pb 2+ , but the tolerance mechanism is not clear. The manuscript aimed to clarify the tolerance mechanism from the perspective of the response changes of related active ingredients. The synthesis of polysaccharides, proteins and thiol compounds in Trichoderma asperellum can be accelerated with Pb 2+ stress. Under Pb 2+ stress, Trichoderma asperellum can synthesize oxalic acid and secrete it extracellularly. In addition, high concentration of Pb 2+ can inhibit the synthesis and extracellular secretion of formic acid and malic acid. The tolerance of Trichoderma asperellum to Pb 2+ is the results of multiple reactions. The Pb 2+ can promote the synthesis of polysaccharides, proteins, thiol compounds and oxalic acid. In the early stage of Pb 2+ stress, Trichoderma asperellum can rapidly initiate an extracellular emergency mechanism, synthesize oxalic acid in mycelia and secrete it extracellularly to remove free Pb 2+ and alleviate the toxicity of Pb 2+ to cells. With the transport of Pb 2+ into cells, it can promote the synthesis of polysaccharides, proteins, thiol compounds to adsorb and transform the Pb 2+ and ease the damage to the cells. Mimicking Nature by using synthetic molecules that resemble natural products may open avenues to key knowledge that is difficult to access by using substances from natural sources. In this context, a novel N-acetylchitooligosaccharide analogue, beta-1,3-N-acetamido-gluco-pentasaccharide, has been designed and synthesized by using aminoglucose as the starting material. A phthalic group has been employed as the protecting group of the amine moiety, whereas a thioalkyl was used as the leaving group on the reducing end. The conformational properties of this new molecule have been explored and compared to those of the its chito analogue, with the beta-1,3 linkages, by a combined NMR spectroscopic/molecular modeling approach. Furthermore, the study of its molecular recognition properties towards two proteins, a lectin (wheat germ agglutinin) and one enzyme (a chitinase) have also been performed by using NMR spectroscopy and docking protocols. There are subtle differences in the conformational behavior of the mimetic versus the natural chitooligosaccharide, whereas this mimetic is still recognized by these two proteins and can act as a moderate inhibitor of chitin hydrolysis.
