This study attempts to detect the differences in event-related potentials (ERPs) associated with two syntactic processes: the syntactic integration of discontinuous dependency and the detection of a violation of the syntactic island constraint. We recorded the electroencephalogram elicited by complex sentences in Japanese that included a dependency between a quantifier and its head noun, in which we changed the word order of the two words to manipulate the presence and absence of a syntactic integration and a syntactic island violation while keeping the lexical items and construction unchanged. We found significant negative and positive deflections for the syntactic integration only when a quantifier preceded its head noun. We also observed significant negative and positive deflections for the syntactic island violation, for which the negativity was more salient when a quantifier preceded its head noun. This study is the first to report a late positive ERP for a violation of the syntactic island constraints in Japanese, and the results showed that the ERP elicited by syntactic integration and that by syntactic island violation were different in terms of their latency, topography, and duration. More importantly, the ERPs elicited by the two syntactic processes were biphasic, and the amplitudes of the negative ERP and of the positive ERP were positively correlated. This positive correlation could be a characteristic of syntactic processing because it contrasted with the negative correlation reported for the ERP elicited by semantic anomalies in English. Furthermore, the amplitude of the ERP for syntactic integration was negatively correlated with the individual capacity of working memory (WM). That is, a reader with greater WM capacity showed smaller negativity and positivity for the syntactic integration, whereas the amplitude for the syntactic island violation showed no significant correlation with the individual capacity of WM. Our results suggested that linguistic ERPs functionally interacted with each other and that the ERP involving the retention and the retrieval of a distant word could be constrained by the individual differences in WM capacity. We discuss the possible reasons for the contrast between English and Japanese on the basis of the cross-linguistic differences in the two languages.

Objectives: It has been reported recently that gamma measures of the electroencephalogram (EEG) might provide information about the candidate biomarker of mental diseases like schizophrenia, Alzheimer&rsquo, s disease, affective disorder and so on, but as we know it is a difficult issue to induce visual and tactile evoked responses at high frequencies. Although a high-frequency response evoked by auditory senses is achievable, the quality of the recording response is not ideal, such as relatively low signal-to-noise ratio (SNR). Recently, auditory steady-state responses (ASSRs) play an essential role in the field of basic auditory studies and clinical uses. However, how to improve the quality of ASSRs is still a challenge which researchers have been working on. This study aims at designing a more comfortable and suitable evoked paradigm and then enhancing the quality of the ASSRs in healthy subjects so as to further apply it in clinical practice. Methods: Chirp and click stimuli with 40 Hz and 60 Hz were employed to evoke the gamma-ASSR respectively, and the sound adjusted to 45 dB sound pressure level (SPL). Twenty healthy subjects with normal-hearing participated, and 64-channel EEGs were simultaneously recorded during the experiment. Event-related spectral perturbation (ERSP) and SNR of the ASSRs were measured and analyzed to verify the feasibility and adaptability of the proposed evoked paradigm. Results: The results showed that the evoked paradigm proposed in this study could enhance ASSRs with strong feasibility and adaptability. (1) ASSR waves in time domain indicated that 40 Hz stimuli could significantly induce larger peak-to-peak values of ASSRs compared to 60 Hz stimuli (p &lt, 0.01**), ERSP showed that obvious ASSRs were obtained at each lead for both 40 Hz and 60 Hz, as well as the click and chirp stimuli. (2) The SNR of the ASSRs were &ndash, 3.23 &plusmn, 1.68, &ndash, 2.44 &plusmn, 2.90, &ndash, 4.66 &plusmn, 2.09, and &ndash, 3.53 &plusmn, 3.49 respectively for 40 Hz click, 40 Hz chirp, 60 Hz click and 60 Hz chirp, indicating the chirp stimuli could induce significantly better ASSR than the click, and 40 Hz ASSRs had the higher SNR than 60 Hz (p &lt, 0.01**). Limitation: In this study, sample size was small and the age span was not large enough. Conclusions: This study verified the feasibility and adaptability of the proposed evoked paradigm to improve the quality of the gamma-ASSR, which is significant in clinical application. The results suggested that 40 Hz ASSR evoked by chirp stimuli had the best performance and was expected to be used in clinical practice, especially in the field of mental diseases such as schizophrenia, Alzheimer&rsquo, s disease, and affective disorder.

Different cortical regions respond with distinct rhythmic patterns of neural oscillations to Transcranial Magnetic Stimulation (TMS). We investigated natural frequencies induced by TMS in left and right homologous dorsolateral prefrontal cortices (DLPFC) and related hemispheric differences. In 12 healthy young adults, single-pulse TMS was delivered in different blocks close to F3 and F4 channels to target left and right DLPFC. An occipital site near PO3 was stimulated as control. TMS-related spectral perturbation analyses were performed on recorded EEG data. A widespread unspecific increase in theta power was observed for all stimulation sites. However, occipital TMS induced greater alpha activity and a 10.58 Hz natural frequency, while TMS over the left and right DLPFC resulted in similar beta band modulations and a natural frequency of 18.77 and 18.5 Hz, respectively. In particular, TMS-related specific increase in beta activity was stronger for the right than the left DLPFC. The right DLPFC is more specifically tuned to its natural beta frequency when it is directly stimulated by TMS than with TMS over the left counterpart (or a posterior region), while the left DLPFC increases its beta activity more similarly irrespective of whether it is directly stimulated or through right homologous stimulation. These results yield important implications for both basic neuroscience research on inter-hemispheric prefrontal interactions and clinical applications.

In addition, the clinical development of new and more effective compounds is hampered by the lack of predictive criteria to judge their early clinical efficacy.In this context, electroencephalography (EEG) could be a sufficiently sensitive biomarker to identify in an early stage (i.e. Phase I) the therapeutic potential of a new molecule on cognitive functioning. In addition, the difficulty to detect subtle improvements in cognitive performance in Phase I (i.e. in healthy subjects) could be overcome by the development of experimental paradigms such as sleep deprivation (SD), to induce cognitive deficits, still reversible in healthy subjects.EEG and EEG coupled with sleep deprivation (SD) would be innovative and relevant strategies to determine and predict the clinical effectiveness of a molecule in Phase I.In this work, we try to determine the relevance of such strategies by identifying, in healthy subjects, EEG markers of cognitive functioning related to (1) either the taking of a cognitive drug, (2) or the induction of a reversible cognitive decline, (3) or concomitant effects of the two parameters. In order to do that, two studies were performed.In a first study, the effect of donepezil on cortical electrical activity was studied in 30 young, healthy adult volunteers. These volunteers were treated with donepezil (5 mg/day orally) (vs. placebo) for 15 days following a double-blind, randomized, cross-over trial.At the end of the treatment period, an EEG (58 electrodes) was performed during two attentional tasks (auditory and visual). Event-related potentials (ERP), the inter-trial coherence (ITC) and the event-related spectral perturbation (ERSP) were then calculated.In a second study, the effect of SD was studied in 36 young, healthy adult volunteers. The ERP, ITC and ERSP were then calculated.Through these two studies, we have found, at the group level, cognitive EEG markers related either to the induction of cognitive decline (SD) or pharmacological intervention targeting cholinergic system (donepezil) or several neurotransmitters (modafinil). All these markers would concern the modulation of cortical activity in the ventral frontoparietal network, known to regulate attentional and executive processes. We also confirmed that the network is underlying by δ/θ and α oscillatory activities. The cognitive efficiency would reflect the integrity of the network.We conclude that EEG is a sufficiently sensitive tool to detect subtle changes in neurocognitive processes of young, healthy adult volunteers, following the administration of a treatment of AD and more generally following the administration of a drug having an effect on cognition when cognitive decline is caused (SD).EEG and EEG coupled with SD could constitute additional tools to the current cognitive assessment for predicting the efficacy of new drug candidates for AD before initiation Phases II/III clinical trials. L’efficience cognitive serait le reflet de l’intégrité de ce réseau.Nous avons conclu que l’EEG est un outil suffisamment sensible pour détecter des changements subtils dans les processus neurocognitifs de participants adultes, jeunes et sains suivant l’administration d’un traitement de la MA et de manière plus générale suivant l’administration d’un médicament ayant un effet sur la cognition lorsqu’un déclin cognitif est provoqué (PS).Sous réserve de réplication des résultats et d’analyses complémentaires, l’EEG ainsi que l’EEG couplée à la PS pourraient constituer des outils additionnels à l’évaluation cognitive pour prédire l’efficacité de nouveaux candidat médicaments de la MA.

Objective To identify possible electroencephalographic (EEG) markers of donepezil’s effect on cortical activity in young, healthy adult volunteers at the group level. Methods Thirty subjects were administered a daily dose of either 5 mg donepezil or placebo for 15 days in a double-blind, randomized, cross-over trial. The electroencephalogram during an auditory oddball paradigm was recorded from 58 scalp electrodes. Current source density (CSD) transformations were applied to EEG epochs. The event-related potential (ERP), inter-trial coherence (ITC: the phase consistency of the EEG spectrum) and event-related spectral perturbation (ERSP: the EEG power spectrum relative to the baseline) were calculated for the target (oddball) stimuli. Results The donepezil and placebo conditions differed in terms of the changes in delta/theta/alpha/beta ITC and ERSP in various regions of the scalp (especially the frontal electrodes) but not in terms of latency and amplitude of the P300-ERP component. Conclusion Our results suggest that ITC and ERSP analyses can provide EEG markers of donepezil’s effects in young, healthy, adult volunteers at a group level. Significance Novel EEG markers could be useful to assess the therapeutic potential of drug candidates in Alzheimer’s disease in healthy volunteers prior to the initiation of Phase II/III clinical studies in patients.

Here, listeners were trained to judge the distances of English, Bengali, and backwards speech sources pre-recorded at near (2-m) and far (30-m) distances. Listeners' accuracy was tested before and after training. Improvements from pre-test to post-test were greater for forward speech, demonstrating a learning advantage for forward speech sounds. Independent component (IC) processes identified in electroencephalographic (EEG) data collected during pre- and post-testing revealed three clusters of ICs across subjects with stimulus-locked spectral perturbations related to learning and accuracy. One cluster exhibited a transient stimulus-locked increase in 4-8 Hz power (theta event-related synchronization; ERS) that was smaller after training and largest for backwards speech. For a left temporal cluster, 8-12 Hz decreases in power (alpha event-related desynchronization; ERD) were greatest for English speech and less prominent after training. In contrast, a cluster of IC processes centered at or near anterior portions of the medial frontal cortex showed learning-related enhancement of sustained increases in 10-16 Hz power (upper-alpha/low-beta ERS). The degree of this enhancement was positively correlated with the degree of behavioral improvements. Results suggest that neural dynamics in non-auditory cortical areas support distance judgments. Further, frontal cortical networks associated with attentional and/or working memory processes appear to play a role in perceptual learning for source distance.

However, findings have been inconsistent, both for behavioural and electrophysiological measures. METHODS: In this study, we examined event-related potentials (ERPs) in 20 6- to 14-year-old children with developmental dyslexia and 20 age-matched controls, divided into younger (6-11 years, n = 10) and older (11-14 years, n = 10) age bands. We focused on early (mismatch negativity; MMN) and late (late discriminative negativity; LDN) conventional mismatch responses and associated measures derived from time-frequency analysis (inter-trial coherence and event-related spectral perturbation). Responses were elicited using an auditory oddball task, whereby a stream of 1000-Hz standards was interspersed with rare large (1,200 Hz) and small (1,030 Hz) frequency deviants. RESULTS: Conventional analyses revealed no significant differences between groups in the size of the MMN to either large or small frequency deviants. However, the younger age band of children with dyslexia showed an enhanced inter-trial coherence in the theta frequency band over the time window corresponding to the MMN to small deviants. By contrast, these same children showed a reduced-amplitude LDN for the small deviants relative to their age-matched controls, whilst the older children with dyslexia showed a shorter and less intense period of event-related desynchronization over this time window. CONCLUSIONS: Initial detection and discrimination of auditory frequency change appears normal or even enhanced in children with dyslexia. Rather, deficits in late-stage auditory processing appear to be a feature of this population.

RESULTS: Conventional analyses revealed no significant differences between groups in the size of the MMN to either large or small frequency deviants. However, the younger age band of children with dyslexia showed an enhanced inter-trial coherence in the theta frequency band over the time window corresponding to the MMN to small deviants. By contrast, these same children showed a reduced-amplitude LDN for the small deviants relative to their age-matched controls, whilst the older children with dyslexia showed a shorter and less intense period of event-related desynchronization over this time window. CONCLUSIONS: Initial detection and discrimination of auditory frequency change appears normal or even enhanced in children with dyslexia. Rather, deficits in late-stage auditory processing appear to be a feature of this population.
