Orientador: Lucas Rios do Amaral Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Engenharia Agrícola Resumo: O principal gargalo para acelerar a procriação e liberação de novas variedades de cana-de-açúcar com características desejáveis para atender as demandas do setor sucroenergético e adaptação às mudanças climáticas é a ausência de métodos de fenotipagem de plantas de alto rendimento para avaliação das variedades em campo. Métodos tradicionais de fenotipagem de campo dependem de especialistas treinados, são lentos, trabalhosos e subjetivos, além de apresentarem riscos para os avaliadores que muitas vezes tem de fazer a avaliação caminhando por dosséis completamente formados. Atualmente, os Veículos Aéreos Não Tripulados (VANT) são considerados por alguns autores a melhor plataforma para a aquisição de imagens no sensoriamento remoto de campos de melhoramento genético vegetal, tornando-se ferramenta crítica na fenotipagem baseada em campo (FBC). O objetivo deste trabalho foi desenvolver uma metodologia para determinação do nível de severidade das ferrugens alaranjada e marrom da cana-de-açúcar por meio de processamento de imagens de alta resolução espacial com sensores acoplados em VANT. Mais especificamente, o capítulo três, referente ao primeiro artigo, trata da exploração de dados multiespectrais de sensor embarcado em VANT para fenotipagem de campo de variedades de cana-de-açúcar em relação a resistência à infecção pelas ferrugens alaranjada e marrom. Os resultados mostraram que apesar dos dados de refletância serem sensiveis aos sintomas das duas doenças, não permitiram a determinação da severidade das mesmas como substituição direta das notas de infecção atribuidas visualmente ou por meio de processamento de imagens das folhas. Porém, é possível classificar as variedades em relação a resistência geral à infecção pelos dois tipos de ferrugens simultaneamente, com alta acurácia, em três classes: resistentes, moderadamente resistentes e susceptiveis. Portanto, as imagens aéreas multiespectrais não permitiram a distinção entre os dois tipos de ferrugens. Já o capítulo quatro, referente ao segundo artigo, trata do processamento de imagens RGB de folhas infectadas para identificação dos sintomas foliares dentre os dois tipos de ferrugens, com alta acurácia mostrada nos resultados. Esse diagnóstico não é uma tarefa simples na rotina de fenotipagem e se mostrou como alternativa uma vez que não foi possível a distinção dentre as duas doenças em imagens aéreas. Os resultados mostram que os dados fenotípicos obtidos a partir de imagens aéreas podem melhorar a eficiência e a qualidade na coleta de dados a nível de campo em relação a infecção pelas ferrugens, com potencial de ser escalável para dezenas de milhares de parcelas dentro de um programa moderno de melhoramento de plantas, porém, a distinção de infecção pelas duas doenças necessita de observação próxima das folhas Abstract: The main bottleneck for obtainning new sugarcane varieties with desirable characteristics to meet the demands of the sugarcane industry and adaptation to climate change is the absence of high-throuput phenotyping methods for plants varieties evaluation in the field. Traditional field phenotyping methods rely on trained specialists, and are slow, laborious and subjective; in addition, it presents risks for evaluators who often have to carry out the evaluation by walking through fully formed canopies. Currently, Unmanned Aerial Vehicles (UAV) are considered by some authors to be the best platform for image acquisition in remote sensing of plant breeding fields, becoming a critical tool in field-based phenotyping (FBC). The objective of this work was to develop a methodology to determine the severity level of orange and brown sugarcane rust by processing high spatial resolution images with sensors coupled to UAV. More specifically, chapter three, referring to the first article, deals with the exploitation of multispectral data from an UAV embedded sensor for field phenotyping of sugarcane varieties in relation to resistance to infection by orange and brown rust. The results showed that despite the reflectance data being sensitive to the symptoms of the two diseases, they did not allow the determination of their severity as a direct replacement of the infection scores attributed visually or through image processing of the leaves. However, it is possible to classify the varieties in relation to general resistance to infection by both types of rust simultaneously, with high accuracy, in three classes: resistant, moderately resistant and susceptible. Therefore, the multispectral aerial images did not allow the distinction between the two types of rust. Chapter four, referring to the second article, deals with the processing of RGB images of infected leaves to identify leaf symptoms between the two types of rust, with high accuracy shown in the results. This diagnosis is not a simple task in the phenotyping routine and proved to be an alternative since it was not possible to distinguish between the two diseases in aerial images. The results show that phenotypic data obtained from aerial images can improve efficiency and quality in collecting data at the field level regarding rust infection, with the potential to be scalable to tens of thousands of plots within a modern plant-breeding program. Many single-celled eukaryotes have complex cell morphologies defined by cytoskeletal elements comprising microtubules arranged into higher-order structures. Trypanosoma brucei (T. brucei) cell polarity is mediated by a parallel array of microtubules that underlie the plasma membrane and define the auger-like shape of the parasite. The subpellicular array must be partitioned and segregated using a microtubule-based mechanism during cell division. We previously identified an orphan kinesin, KLIF, that localizes to the division plane and is essential for the completion of cytokinesis. To gain mechanistic insight into how this novel kinesin functions to complete cleavage furrow ingression, we characterized the biophysical properties of the KLIF motor domain in vitro. We found that KLIF is a non-processive dimeric kinesin that dynamically crosslinks microtubules. Microtubules crosslinked in an antiparallel orientation are translocated relative to one another by KLIF, while microtubules crosslinked parallel to one another remain static, resulting in the formation of organized parallel bundles. In addition, we found that KLIF stabilizes the alignment of microtubule plus ends. These features provide a mechanistic understanding for how KLIF functions to form a new pole of aligned microtubule plus ends that defines the shape of the new posterior, which is a unique requirement for the completion of cytokinesis in T. brucei.

Hot-dip galvanized steel is one of the most used materials in equipment and metallic structures of the Brazilian electric sector. Although carbon steel is the main substrate in the galvanizing hot-dip process, recently, weathering steel has been used as an alternative material to be galvanized. In the transmission line segment of the Brazilian electric sector, for instance, compact towers made of galvanized weathering steel have been installed to conduct energy through urban sites. It is well known that weathering steel, depending on wet and dry cycles and on the pollutants present in the atmosphere, develops a protective patina made of its corrosion products. The patina is dense and strongly adhered to the substrate, blocking the active surface and, thus, reducing the corrosion rate of the base metal. However, when the weathering steel is galvanized, the substrate surface has a layer of zinc and zinc-iron intermetallic alloys. When the sacrificial layer is consumed by atmospheric corrosion, critical questions remain to be answered regarding the underlying substrate. Will the patina of weathering steel be formed? In what condition? Does the hot-dip galvanizing process modify the weathering steel microstructure? The present work carried out an experimental research to shed light on the anticorrosive behavior of hot-dip galvanized weathering steel, after the zinc layer is corroded. This was done by a controlled pickling process, where the zinc layer was removed simulating its consumption during real corrosion processes. The results, obtained through electrochemical techniques and different accelerated corrosion tests, showed that galvanizing weathering steel is a promising technology to enhance the lifetime of structures used in the Brazilian electric sector.

O presente artigo intende debater como os romances O manual dos inquisidores (1998) e A morte de Carlos Gardel (1994), do escritor português António Lobo Antunes, constroem e desenvolvem, através das personagens João e Álvaro, respectivamente, uma compreensão da realidade social onde estão inseridos por meio de um cotidiano objetivado. Essa hipótese será debatida com o auxílio de pressupostos da Teoria da Literatura e da Sociologia do Conhecimento de Peter Berger e Thomas Luckmann (1991), procurando, ainda, problematizar como os processos de memória interferem nos discursos que institucionalizam a realidade, transformando, inclusive, memórias em objetos cotidianos.

Allosteric signaling within multidomain proteins is a driver of communication between spatially distant functional sites. Understanding the mechanism of allosteric coupling in large multidomain proteins is the most promising route to achieving spatial and temporal control of the system. The recent explosion of CRISPR-Cas9 applications in molecular biology and medicine has created a need to understand how the atomic level protein dynamics of Cas9, which are the driving force of its allosteric crosstalk, influence its biophysical characteristics. In this study, we used a synergistic approach of nuclear magnetic resonance (NMR) and computation to pinpoint an allosteric hotspot in the HNH domain of the thermostable GeoCas9. We show that mutation of K597 to alanine disrupts a salt-bridge network, which in turn alters the structure, the timescale of allosteric motions, and the thermostability of the GeoHNH domain. This homologous lysine-to-alanine mutation in the extensively studied mesophilic S. pyogenes Cas9 similarly alters the dynamics of the SpHNH domain. We have previously demonstrated that the alteration of allostery via mutations is a source for the specificity enhancement of SpCas9 (e SpCas9). Hence, this may also be true in GeoCas9.

Unmanned Aerial Vehicles (UAVs) have potentially significant application in agriculture and, with the emergence of the digital farming era and Agriculture 4.0, this platform has become increasingly important. UAV imagery may improve or even replace routine data surveys, as well as optimize phytosanitary product application. High-spatial resolution imagery makes UAVs attractive for several applications where traditional satellite sensing is still unsuitable. With the significant recent development of data science techniques, UAVs have a prominent position in assisting farmers for more efficient decision-making and automating agricultural processes. Thus, this work addresses the main agricultural applications of UAVs into five major topics: topographic survey, physiological assessments, biophysical assessments, monitoring of biological targets, and spraying of phytosanitary products and application of bio inputs. RESUMO As potencialidades de uso de UAVs na agricultura são enormes e, com a entrada na era da digitalização das lavouras e da Agricultura 4.0, essa plataforma tem ganhado cada vez mais importância. Levantamento rotineiros de dados em campo podem ser melhorados ou mesmo substituídos por imagens obtidas via UAVs e as aplicações de produtos fitossanitários podem ser otimizados. A altíssima resolução espacial das imagens torna os UAVs atrativos para diversas aplicações que o tradicional sensoriamento via satélite ainda não atendem. Ainda, com o enorme desenvolvimento recente das técnicas e aplicações de ciência dos dados, os UAVs têm posição de destaque para auxiliar o agricultor na tomada de decisão mais eficiente e na viabilização da automação de processos agrícolas. Assim, neste trabalho abordaremos as principais aplicações dos UAVs na agricultura, dividindo o texto em cinco grandes tópicos: Levantamento topográfico, Avaliações fisiológicas, Avaliações biofísicas, Monitoramento de alvos biológicos, e Pulverização de produtos fitossanitários e aplicação de bioinsumos.

Orientador: Zanoni Dias Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Computação Resumo: Uma das ferramentas mais importantes no campo da bioinformática e biologia computacional é o alinhamento múltiplo de sequências (MSA, do inglês Multiple Sequence Alignment). Sua função abrange a criação de árvores filogenéticas, identificação de motifs e domínios conservados, assim como a predição de funções das proteínas e de suas estruturas secundárias e terciárias. Algoritmos genéticos são heurísticas de busca local, utilizadas para problemas de otimização, inspirados nos conceitos da seleção natural: uma popularção de possíveis soluções é criada e iterativamente cruzada e selecionada buscando um resultado mais adequado para o problema sendo tratado. Esta dissertação descreve o estudo realizado no uso de algoritmos genéticos para a resolução do problema de alinhamento múltiplo de proteínas. Como resultado deste trabalho, duas ferramentas foram desenvolvidas: ALGAe, um ambiente para a execução de um algoritmo genético visando o alinhamento múltiplo de proteínas e o Anubis, um visualizador de alinhamentos que permite a comparação entre dois alinhamentos para o mesmo conjunto de proteínas, visando identificar as discrepâncias entre eles Abstract: MSA, that stands for Multiple Sequence Alignment, is one of the most important tools on bioinformatics and computational biology fields. Its usage includes phylogenetic trees creation, motifs and conservated domains prediction, as well as protein secondary and tertiary structure and function prediction. Genetic algorithms are heuristics for local search used on optimization problems, inspired on the natural selection concepts: a population of possible solution is created and iterativelly mate and selected in order to search for a suitable solution to the target problem. This dissertation describes the study we proceed on using genetic algorithms to solve the multiple sequence alignment problem. As results of this work, two tools were developed: ALGAe, a genetic algorithm execution environment for MSA solving, and Anubis, a multiple sequence alignment viewer that allows comparing a given MSA against a benchmark, in order to find discrepancies among them Mestrado Ciência da Computação Mestre em Ciência da Computação

Light charged particles emitted by the compound nucleus $^{158}\mathrm{Er}$ produced in the reaction $^{32}\mathrm{S}$ $(180 \mathrm{MeV})+^{126}\mathrm{Te}$, at the excitation energy ${E}_{x}=92$ MeV, have been measured at Laboratori Nazionali di Legnaro in coincidence with fission fragments and evaporation residues. The $4\ensuremath{\pi}$ detector array $8\ensuremath{\pi}\mathrm{LP}$ coupled to a system of parallel-plate avalanche counters to detect evaporation residues has been used. Data have been analyzed in the framework of the statistical model of evaporation with the code pace2_n11. This enlarged version of the code pace2 has been used to reproduce the large set of observables measured in the fusion-evaporation and fusion-fission channels along with experimental prescission neutron multiplicity and fission cross section taken from literature. It is found that the simultaneous reproduction of the prescission neutron, proton, and $\ensuremath{\alpha}$-particle multiplicities can be obtained with zero fission delay without dynamical effects. However, the same set of model input parameters does not allow us to reproduce proton and $\ensuremath{\alpha}$-particle multiplicities in the evaporation channel. Extensive calculations, with different sets of parameters, show the limits of the statistical model in reproducing the whole set of data. This work evidences the importance of measuring a large set of observables in order to obtain a reliable description of the decay of the compound nucleus, and in particular of the fission process.

This contribution reports on the time of flight implementation in the FAZIA Si-Si-CsI(Tl) telescopes, focusing on the basic ideas of the method and on some results from recent experiments at LNS.
