Hormonally active tumours are characterized by production and secretion of hormones, irrespective of endogenous feedback mechanisms. An adult llama had exuberant oestrous behaviour, infertility, elevated concentrations of oestradiol and a large ovarian mass. Necropsy revealed the presence of two large abdominal masses, one effacing the right ovary and one in the mesocolon. Considering the clinical and histopathological findings, we conclude that the llama was affected by a granulosa cell tumour. The case suggests that granulosa cell tumours in camelids are hormonally active, and the clinical presentation resembles that of other large animal species. To our knowledge, this is the first case report of an oestrogen‐producing, metastatic granulosa cell tumour in a llama. [ABSTRACT FROM AUTHOR]

Kisspeptin modulates GnRH secretion in mammals and peripheral administration of 10-amino acid fragment of kisspeptin (Kp10) induces LH release and ovulation in cattle. Experiments were done to determine if iv administration of kisspeptin will activate GnRH neurons (i.e., after crossing the blood-brain barrier) and if pre-treatment with a GnRH receptor blocker will alter kisspeptin-induced LH release (from gonadotrophs) and ovulation. In Experiment 1, cows (n = 3 per group) were given human-Kisspeptin10 (hKp10; 3 x 15 mg iv at 60-min intervals) or normal saline and euthanized 150 min after treatment was initiated. Every 20th free-floating section (50 μm thickness) from the preoptic area to hypothalamus was double immunostained to colocalize GnRH- (DAB) and activated neurons (cFOS; Nickel-DAB). Kisspeptin induced plasma LH release from 15 to 150 min (P = 0.01) but the proportion of activated GnRH neurons did not differ between groups (5.8% and 3.5%, respectively; P = 0.11). Immunogold electron microscopy detected close contacts between kisspeptin fibers and GnRH terminals in the median eminence. In Experiment 2, pubertal heifers (n = 5 per group) were treated with 1) hKp10 iv, 2) Cetrorelix (GnRH antagonist; im) + hKp10 iv or 3) saline on Day 6 of the follicular wave under low-progesterone condition. A rise in plasma LH concentration was detected from 15 to 240 min in the hKp10 group but not in cetrorelix or control group (P<0.001). Ovulations were detected only in the hKp10 group (4/5; P = 0.02). Cetrorelix treatment was associated with regression of the preovulatory dominant follicle and emergence of a new follicular wave 3.4±0.75 days after the treatment in all five heifers. Results support the hypothesis that the effect of peripheral kisspeptin is mediated downstream of GnRH synthesis and does not involve GnRH-independent LH release from gonadotrophs. As part of the development of a germplasm biobank to preserve the genetic diversity of threatened wood bison ( Bison bison athabascae ), a 2 × 2 factorial study was designed to determine the effects of ovulation induction agent and follicle maturity on the ovulatory response in wood bison during the anovulatory season. Bison ( n = 32) were assigned randomly to four groups ( n = 8/group) and treated with either pLH or hCG when a growing dominant follicle was either 8–9 mm or ≥10 mm. The ovaries were examined daily by ultrasonography to determine the timing of ovulation, and 7 days post-treatment to assess CL development. The proportion of bison that ovulated was greater in bison treated with hCG than pLH ([15/16] 94% vs. [8/16] 50%; P < 0.05), and when the dominant follicle was ≥10 mm vs. 8–9 mm at the time of treatment (88% vs. 56%; P < 0.05). The interval from treatment to ovulation was 37.0 ± 1.3 h and was not affected by induction agent or follicle size. However, synchrony of ovulation tended to be greater ( P = 0.10) in the ≥10 mm group vs. the 8–9 mm group, and the ensuing corpus luteum was larger (15.3 ± 0.43 mm vs. 13.4 ± 0.36; P < 0.05). In conclusion, both ovulation inducing agent and follicle size influenced the ovulatory response in bison during the anovulatory season. Treatment with hCG was more effective than pLH for inducing ovulation in wood bison, and the effect was greater when treatment was given when the growing dominant follicle was ≥10 mm. In an effort to develop an effective, minimum-handling protocol for the conservation of wood bison, the present study was designed to determine the effects of ovarian synchronization and superstimulation on cumulus oocyte complex (COC) collection and in vitro embryo production in wood bison during the ovulatory (Exp. 1) and anovulatory seasons (Exp. 2). We tested the hypotheses that COC collection and in vitro embryo production are 1) greater after follicular wave synchronization than at random stages of the follicular wave, 2) repeatable within individuals, 3) greater after ovarian superstimulation with a single dose of eCG than without treatment, and 4) greater during the anovulatory season than the ovulatory season. In Exp. 1, ultrasound-guided COC collection was performed on Day −1 in wood bison to induce follicular wave emergence the following day (Day = 0). Immediately after the COC collection on Day −1, bison were given a single im dose of 2500 IU eCG or saline (n = 6 per group). Subsequent COC collections were on Days 4 and 9. A similar design was used in Exp. 2, with an additional treatment group given 5000 IU eCG (n = 8 per group). In Exp. 1, compared to the saline-treated group, a single dose of 2500 IU eCG resulted in a greater number of ≥8 mm follicles at the time of the Day 4 COC collection (P = 0.03), but not at the Day 9. In Exp. 2, treatment with 5000 IU eCG resulted in a greater number of ≥8 mm follicles than 2500 IU eCG or the saline treatment (37.5 ± 6.9, 17.5 ± 2.0, 16.9 ± 2.0; P = 0.01, respectively). Although the number of embryos produced/COC submitted to IVM was not different among groups (mean = 18.6%), treatment with 5000 IU eCG produced more than twice as many embryos per bison as unstimulated bison (0.8 vs 1.9). In summary, embryo production rates were higher from COC collected subsequent to follicular wave synchronization vs random stages of the wave, and ovarian superstimulation with eCG resulted in a dose-related increase in the number of ≥8 mm follicles, COC collected, and embryos produced. Repeated COC collections after successive wave synchronization resulted in similar follicular counts and embryo production rates within individuals, and the greatest number of follicles aspirated, COC collected, and embryos produced was in the anovulatory season. Experiments were done to determine ovulation synchrony following a 4-day letrozole treatment (Exp 1), compare the efficacy of a letrozole-based protocol with other commonly used synchronization protocols for FTAI (Exp 2), and test a new intravaginal letrozole-releasing device (LRD) with and without pre-synchronization (Exp 3). In Exp 1, heifers and lactating cows at random stages of the estrous cycle were given an LRD for 4 days, PGF at LRD removal, and GnRH at 48 or 60 h after PGF, or no GnRH. In Exp 2, heifers and lactating cows were assigned to three FTAI groups: i) LRD, ii) estradiol+progesterone, iii) 5-d Co-synch+PRID. In Exp 3, heifers were pre-synchronized with PGF or not (control) 8 days before insertion of either an X-LRD or T-LRD for 3 days; FTAI was done 48 h after device removal. In Exp 1, the variation in interval to ovulation in cows, but not heifers, in the GnRH 48-h group was less than half that in other groups (P < 0.05). In Exp 2, the P/AI was lower (P < 0.001) in the LRD group compared to the other groups. In Experiment 3, the X-LRD increased (P < 0.0001) plasma letrozole concentrations compared to the T-LRD; the pregnancy rate was not affected by pre-synchronization or the type of LRD. Although ovulation synchrony was achieved following LRD treatment, the LRD group had the lowest P/AI compared to other protocols, perhaps because of too short an interval between LRD removal and GnRH/FTAI. Abstract: The objective was to evaluate the efficacy of three previously unreported ovarian superovulatory treatment protocols in wapiti. Protocols were initiated specifically at the time of ovarian follicular wave emergence, and intended to enable determination of the effects of frequency of treatment (i.e., animal handling) and LH supplementation on ovarian response. Thirteen parous wapiti hinds, 2 to 4 y of age, were used late in the anovulatory season (July). The ovaries were examined daily by transrectal ultrasonography. Hinds were given 5 mg estradiol 17-β im (day of treatment designated as Day 0) to induce a new wave of ovarian follicular development. On the expected day of wave emergence (Day 3), hinds were assigned randomly to three treatment groups and given: (1) 100 mg FSH im once a day for 4 days (N = 5); (2) 200 mg FSH sc on Day 3 and Day 5 (N = 4); or (3) 200 mg FSH plus 2.5 mg LH sc on Day 3 and Day 5 (N = 4). All hinds were given 10 mg LH im on Day 6 to induce ovulation. The mean (± SEM) number of ovulations per animal in the respective groups was 6.2 ± 2.0, 15.5 ± 5.9, and 14.8 ± 2.7. In conclusion, the technique of inducing follicular wave emergence to initiate superovulatory treatment at the time of wave emergence was effective in wapiti during the anovulatory season. The most efficient and effective method of ovarian superovulation in this study involved administration of estradiol 17-β on Day 0, followed by 200 mg FSH sc on Days 3 and 5, and induction of ovulation (10 mg of LH) on the evening of Day 6. Compared with conventional methods that require 14 days and handling the hinds six times, the protocol used herein reduced the treatment period to 8 days and the number of animal handlings to four. • The number of follicles ≥1 mm at wave emergence was predictive of the number of follicles recruited into subsequent waves. • Large AFC at wave emergence resulted in higher number of follicles and oocytes following superstimulation than small AFC. • Seven days of exogenous FSH treatments resulted in a greater number of large than small follicles than four days. • Prolonged follicular growth under exogenous FSH support requires control of endogenous LH release. • Ovarian response was positively associated with AFC at wave emergence in prepubertal calves. Antral follicle count (AFC) repeatability at the time of follicular wave emergence and duration of gonadotropin treatment in calves with small and large AFC affects the superstimulatory response of follicles. In Study I, the individual AFC was determined, calves were ranked as having a small, medium or large AFC, and a second count was performed prior to FSH treatments. There was a positive association between the number of follicles ≥1 mm after the first and second counts (r = 0.4; P = 0.003). In Study II, calves with small and large AFC were administered pFSH for 4 or 7 days, pLH 20 h after last pFSH administration and cumulus-oocyte-complexes (COC) were collected. In calves having large as compared with small AFC, number of follicles ≥6 mm were greater (P = 0.01) and COC collected were greater (P = 0.001). The proportion of large-sized follicles (>9 mm) was greater in the 7-day than in the 4-day gonadotropin treatment group (56.4 ± 8.3 and27.8 ± 7.5 %, respectively; P = 0.01). In Study III, there was a positive association between AFC and number of follicles ≥6 mm at the time of COC collection (r = 0.6; P = 0.003). In summary, the number of follicles at the time of follicular wave emergence was associated with the number of follicles recruited during subsequent waves of follicular development and ovarian response following gonadotropin superstimulation. • More than 80 % of follicles were between 3 and 8 mm on Day 4 of pFSH treatment • Higher cumulative pFSH dose resulted in a greater number of large-sized follicles • Longer duration of treatment resulted in greater number of follicles and ovulations The objective of the study was to determine relative effects of dose (200 or 350 mg) and duration (4 or 7 days) of superstimulatory treatment on the ovarian response in prepubertal calves. Calves with similar antral follicular counts at wave emergence (n = 24) were given eight doses of either 25 or 44 mg pFSH every 12 h for 4 days or 14 doses of either 14 or 25 mg pFSH for 7 days beginning at the time of follicular wave emergence and 12.5 mg of pLH im 12 h after the last FSH treatment. On Day 4 of pFSH treatment, calves given 14 mg had fewer follicles ≥3 mm than those given 25 mg (15.1 ± 1.9 and 27.9 ± 3.3, respectively; P = 0.04). At the end of treatment (24 h post-LH), number of follicles ≥9 mm was greater in calves of groups treated with 350 than 200 mg (13.5 ± 1.8 and 8.8 ± 1.3, respectively; P = 0.02) and calves of groups treated for 7 than 4 days (13.3 ± 1.8 and 9.0 ± 1.3, respectively; P = 0.03). The number of spontaneous ovulations was greater in calves of groups treated for 7 than 4 days as was the total number of ovulations (9.7 ± 0.9 and 6.9 ± 1.0, respectively; P ≤ 0.05). In summary, a dose of 25 mg of pFSH per treatment given twice daily for 7 days resulted in a greater ovarian response than other superstimulatory treatments in prepubertal calves. Background: A molecule identical to nerve growth factor, with ovulation-inducing properties has been discovered in the seminal plasma of South American camelids (ovulation-inducing factor/nerve growth factor; OIF/NGF). We hypothesize that the ovulatory effect of OIF/NGF is initiated at the level of the hypothalamus, presumably by GnRH neurons. The objective of the present study was to determine the structural relationship between GnRH neurons and neurons expressing high- and low-affinity receptors for NGF (i.e., TrkA and p75, respectively) in the hypothalamus. Methods: Mature llamas (n = 4) were euthanized and their hypothalamic tissue was fixed, sectioned, and processed for immunohistochemistry on free-floating sections. Ten equidistant sections per brain were double stained for immunofluorescence detection of TrkA and GnRH, or p75 and GnRH. Results: Cells immunoreactive to TrkA were detected in most hypothalamic areas, but the majority of cells were detected in the diagonal band of Broca (part of the ventral forebrain) and the supraoptic nuclei and periventricular area. The number of cells immunoreactive to p75 was highest in the diagonal band of Broca and lateral preoptic areas and least in more caudal areas of the hypothalamus (p < 0.05) in a pattern similar to that of TrkA. A low proportion of GnRH neurons were immunoreactive to TrkA (2.5% of total GnRH cells), and no co-localization between GnRH and p75 was detected. GnRH neuron fibers were detected only occasionally in proximity to TrkA immunopositive neurons. Conclusions: Results do not support the hypothesis that the effect of OIF/NGF is driven by a direct interaction with GnRH neurons, but rather provide rationale for the hypothesis that interneurons exist in the hypothalamus that mediate OIF/NGF-induced ovulation. Experiments were done to determine if inclusion of eCG and progesterone in the superstimulation protocol will increase the ovarian response and embryo production in wood bison, and to provide preliminary information regarding the effect of season. In Experiment 1 (anovulatory season), bison (n = 26) were synchronized by follicular ablation (Day −1) and given FSH on Days 0 and 2, and assigned to 3 groups: Progesterone (Days 0–4), eCG (Day 3), or progesterone + eCG. On Day 5, bison were given hCG and inseminated 12 and 24 h later. Ova/embryos were collected 8 days after hCG. In Experiment 2 (ovulatory season), bison (n = 24) were synchronized and assigned randomly to two groups in which superstimulation was induced with FSH, either with or without eCG, as in Experiment 1. No differences among groups were found in ovarian response or embryo production in either experiment. The follicular count at wave emergence was positively correlated with the number of large follicles at the end of superstimulation in all groups. A significantly greater number of follicles present at wave emergence in the anovulatory vs. ovulatory season was associated with a greater number of CL at the time of embryo collection, but only half the number of freezable embryos. In conclusion, the number of transferable embryos collected (1–2/bison) was higher than in any previous report, but was not attributable to the inclusion of eCG or progesterone in the superovulatory protocol. Experiments were done to compare the in vivo and in vitro maturational characteristics of cumulus-oocyte complexes (COC) collected from live wood bison. In Experiment 1 (anovulatory season), follicular ablation was done to synchronize follicle wave emergence among bison on Day −1, and FSH was given on Days 0 and 2. Bison were then assigned to 5 groups (n = 5/group) in which COC were collected by transvaginal follicle aspiration on Day 4 and either fixed immediately with no maturation (control), matured in vitro for 24 or 30 h, or collected on Day 5 after in vivo maturation for 24 or 30 h ( i.e ., after hCG treatment). In Experiment 2 (ovulatory season), bison were treated as described for Experiment 1, but PGF2α (cloprostenol) was given to control the luteal phase on Days −9 and 3. In both experiments, cumulus cell expansion was more extensive following in vivo than in vitro maturation, and the percentage of fully expanded COC was highest in the in vivo 30 h groups. Nuclear maturation occurred more rapidly in vitro ; 60–70% of oocytes were at the MII stage 24 h after in vitro maturation while only 25–27% of oocytes had reached the MII stage after 24 h of in vivo maturation. In conclusion, nuclear maturation occurred more rapidly during in vitro vs . in vivo maturation, but was associated with less cumulus expansion than in vivo maturation. In vivo oocyte maturation was more complete at 30 vs . 24 h after hCG treatment. Season had no effect on the maturational capacity of wood bison oocytes. [ABSTRACT FROM AUTHOR]

Experiments were done to determine the ovarian response and embryo production following superstimulation of wood bison. In Experiment 1 (Anovulatory season), the efficacy of pLH vs. hCG for inducing ovulation was compared in wood bison superstimulated with a single dose of pFSH in 0.5% hyaluronan and the effect of exogenous progesterone (PRID) on superovulatory response and embryo quality was examined. In Experiment 2 (Ovulatory season), the efficacy of pLH vs. hCG for the induction of ovulation was compared in wood bison superstimulated with pFSH in a single intramuscular dose vs. a two-dose regimen 48 h apart (split dose) in 0.5% hyaluronan. In Experiment 1, the number of CL was greater ( P < 0.05) in bison treated with hCG than pLH (6.6 ± 1.8 vs. 2.8 ± 0.8) and in those that were not given PRID (6.0 ± 1.5 vs. 2.7 ± 1.0). There was no effect of progesterone treatment on embryo quality. In Experiment 2, the number of CL was greater (P < 0.05) in bison treated with hCG than with pLH (6.3 ± 0.8 vs. 3.8 ± 1.2) and in bison superstimulated with split dose vs. single dose of FSH (7.1 ± 0.9 vs. 3.0 ± 0.8). The number of ova/embryos and freezable embryos did not differ among groups in either experiment. In conclusion, hCG induced a greater ovulatory response than pLH in both seasons. A 2×2 design was used to compare the ovarian response and oocyte collection characteristics in bison given a superstimulatory dose of eCG or FSH, with or without a follow-up dose of LH. Follicular wave emergence was synchronized by follicle ablation (Day −1) and bison were assigned randomly to two superstimulatory treatment groups (n =10 per group): (i) a single intramuscular dose of 2500IU of eCG given on Day 0, or (ii) two subcutaneous doses of 200mg of FSH given on Days 0 and 2. On Day 4, 200mg of LH was given intramuscularly in 5 bison in each superstimulatory treatment group. The study was done in two replicates (n =20 per replicate) involving a crossover design so that each animal was given the opposite superstimulatory treatment (eCG or FSH) during successive replicates. Cumulus-oocyte complexes (COC) were collected by transvaginal ultrasound-guided follicle aspiration, and were classified according to morphologic attributes as compact, expanded, or denuded. Superstimulatory treatment with FSH (vs. eCG) resulted in the development of more follicles ≥5mm (14.2±1.41 vs. 8.2±0.67; P <0.05; mean±SEM), and more follicles aspirated (12.4±1.3 vs. 6.3±0.6; P <0.04). Follow-up treatment with LH (vs. no LH) resulted in a greater proportion of expanded COC (37% vs. 15%; P <0.05), and a tendency for a higher COC collection rate (61% vs. 54%; P =0.08). In summary, superstimulation with FSH (vs. eCG) resulted in twice as many follicles available for aspiration and nearly twice as many COC collected in bison during the anovulatory season, and follow-up treatment with LH increased the proportion of expanded COC collected. Experiments were designed to validate the use of ultrasound bio-microscopy (UBM) as a method for assessing ovarian structures in rabbits. In Experiment 1, female New Zealand White (NZW) rabbits (n =4) were given an ovulation-inducing treatment and the ovaries were examined ex situ by UBM using a 25MHz oscillating sector transducer before being processed for histology. Pairwise correlations revealed strong relationships between UBM and histology in the number (Mean±SEM) of follicles ≥0.6mm (17.3±2.3 compared with19.0±1.6, respectively; r =0.96; P =0.040), CL (8.5±2.9 compared with 8.8±3.0; r =0.99; P =0.003), the diameter of follicles (1.1±0.05 compared with 1.1±0.03mm; r =0.96; P =0.035) and CL (2.1±0.7 compared with 1.8±0.6mm, r =0.99; P <0.001). In Experiment 2, the ovaries of NZW rabbits (n =12) were surgically translocated to a subcutaneous position in the flank region to permit serial examination of ovarian structures in vivo by UBM. Beginning 2 weeks after surgery, the ovaries were examined by UBM daily for at least 18 days, and again 2 months after surgery. Post-operative complications were minor, and both ovaries of each rabbit were identified consistently. The number and diameter of follicles ≥0.6mm were readily visualized during each examination. Multiple corpora lutea were detected in two rabbits, and serial follicular and luteal dynamics in these two rabbits were used to document the consistency of UBM and the retention of ovarian function after surgery. It is concluded that UBM is a valid tool for instant assessment of rabbit ovarian structures (follicles, corpora lutea, and cumulus-oocyte complexes) ex situ, and for serial assessment in vivo using a transcutaneous approach. Abstract: The development of assisted reproductive technologies such as embryo transfer (ET), artificial insemination (AI) and in vitro fertilization (IVF) in South American camelids is considerably behind that of other livestock species. Poor success of the embryo transfer technique has been related to a lack of an effective superstimulatory treatment, low embryo recovery rate, and the recovery of hatched blastocysts that are not conducive to the cryopreservation process. Superstimulation has been attempted using equine chorionic gonadotropin (eCG) and follicle stimulating hormone (FSH) during the luteal, or the sexually receptive phase, sometimes given at follicular wave emergence. The rationale for inducing a luteal phase prior to or during superstimulation in camelids is not clearly understood, but it may simply reflect an empirical bias to conventional methods used in other ruminants. The number of ovulations or CL varies widely among studies, ranging from 2 to more than 15 per animal, with the number of transferable embryos ranging from 0 to 4 per animal. The control of follicular growth combined with superstimulatory protocols has resulted in a more consistent ovarian response and a greater number of follicles available for aspiration and oocyte collection. Recent studies in llamas have demonstrated that the use of ovulation inducing treatments or follicle ablation can synchronize follicular wave emergence allowing the initiation of gonadotropin treatment in the absence of a dominant follicle resulting in a more consistent ovulatory response. Few studies in alpacas have been reported, but it appears from recent field studies that the ovarian response is more variable and that there is a greater number of poor responders than in llamas. A review of superstimulation protocols that have been used in llamas and alpacas in the last 15 years is provided, including a discussion of the potential of protocols designed to initiate treatment at specific stages of follicular growth. This study was designed to test the hypothesis that an immunoreactive chorionic gonadotropin (CG)-like substance is present in full-term dolphin placentas and to determine if CG immunoreactivity can be detected in corresponding serum and urine samples for potential application to diagnose pregnancy. Six placentas were collected immediately after parturition from four captive bottlenose dolphins in 2003, 2007 (Experiment 1), and 2011 (Experiment 2). Serum and urine were collected during early, middle, and late pregnancy from the same dolphins. In Experiment 1, an eCG radioimmunoassay (RIA) was used to analyze dilutions of supernatants from the homogenates of each placenta for eCG immunoreactivity, and a commercial hCG Enzyme-Linked Immunosorbent Assay (ELISA) was used to analyze individual serum samples and dilutions of pools of serum and urine for hCG immunoreactivity. Specific CG immunoreactivity was not detected above assay sensitivities in any of the supernatants of respective placental homogenates, including the highest concentrate (100 mg/mL), nor in any of the individual samples and pools of serum and urine. In Experiment 2, the highest placental homogenate was increased five-fold (500 mg/mL), sensitivity of the eCG RIA was increased six-fold, and a different combination of hCG antibodies was used in an alternative commercial "sandwich"-type ELISA. Despite the optimization, specific CG immunoreactivity in placental tissue and individual serum and urine samples was not detected above assay sensitivities. In conclusion, the hypothesis that an immunore-active CG-like substance is present in full-term dolphin placentas was not supported. In addition, non-immunoreactivity of a CG-like substance in serum and urine samples collected during various stages of pregnancy precluded the development and application of a CG-based immunoassay for diagnosing pregnancy status in dolphins.
