- Abstract
The stories about the future have never been neutral narratives or mere literary games, in the images about tomorrow what is mainly being projected are the challenges and values of the socioeconomic context of the moments in which they are written. Urban agriculture has been an element present in reference works, as a daily activity, an aspiration, an element of hope or an island of resistance. A reciprocal influence can be observed between the cultural and environmental experimentalism of different periods and the connotations that this activity acquires in science fiction literature. This can be seen in the utopias of the 19th century in reaction to the industrial city, in the aspiration to an ecological society in the 70s, and in the dystopias and recent stories of social reconstitution after environmental collapses., Los relatos sobre el futuro nunca han sido narraciones neutrales o meros juegos literarios, pues en las imágenes sobre el mañana lo que principalmente se proyecta son los retos y valores del contexto socioeconómico del momento en que se escribe.

- Abstract
Objectives To identify genetic determinants of susceptibility to clinical vertebral fractures, which is an important complication of osteoporosis. Methods Here we conduct a genome-wide association study in 1553 postmenopausal women with clinical vertebral fractures and 4340 controls, with a two-stage replication involving 1028 cases and 3762 controls. Potentially causal variants were identified using expression quantitative trait loci (eQTL) data from transiliac bone biopsies and bioinformatic studies. Results A locus tagged by rs10190845 was identified on chromosome 2q13, which was significantly associated with clinical vertebral fracture (P=1.04×10−9) with a large effect size (OR 1.74, 95% CI 1.06 to 2.6). Bioinformatic analysis of this locus identified several potentially functional SNPs that are associated with expression of the positional candidate genes TTL (tubulin tyrosine ligase) and SLC20A1 (solute carrier family 20 member 1). Three other suggestive loci were identified on chromosomes 1p31, 11q12 and 15q11. All these loci were novel and had not previously been associated with bone mineral density or clinical fractures. Conclusion We have identified a novel genetic variant that is associated with clinical vertebral fractures by mechanisms that are independent of BMD. Further studies are now in progress to validate this association and evaluate the underlying mechanism.

[EUS] Munduko biztanleriaren %10ak dislexia pairatzen du, eta Lehen Hezkuntzako geletan gero eta gehiago ematen ari den nahastea da. IKTek (Informazio eta Komunikaziorako Teknologiak) dislexia duten umeekin lan egiteko baliabideak eta abantailak eskaintzen dituzte. Horregatik, lan honen helburu nagusia irakasleek dislexiari buruz eta teknologiek eskaintzen dituzten aukerei buruz dituzten ezagutzak aztertzea da. Ikerketa hau aurrera eramateko galdetegi bat erabili da, LHko lehen zikloko 9 irakaslek erantzun duena. Lortutako emaitzek agerian uzten dute irakasle gehienek ez dituztela dislexiaren ezaugarri nagusiak ezagutzen eta ez dutela formakuntza nahikorik IKTetan. Beraz, etorkizunerako proposamen bat egiten da, irakasleek IKTek eskaintzen dituzten abantailei buruz duten formakuntza hobetzeko helburuarekin. [EN] 10% of the world population suffers from dyslexia, and is an increasingly common disorder in Primary Education classrooms. ICT (Information and Communication Technologies) offer resources and advantages to work with children who suffer from dyslexia. Therefore, the aim of this project is to analyse the knowledge teachers have about dyslexia and options offered by ICT. To that end, a survey has been answered by 9 teachers of the first cycle of primary. - Abstract
Hypophosphatasia (HPP) is a rare inherited disorder characterized by rickets and low circulating concentrations of total alkaline phosphatase (ALP) caused by mutations in ALPL. Severe HPP presents in childhood but milder forms can present in adulthood. The prevalence and clinical features of adult HPP are poorly defined. The aim of this study was to evaluate the prevalence and clinical significance of low serum total alkaline phosphatase (ALP) levels in a clinic‐based population of adult osteoporotic patients. We searched for patients with low ALP in a cohort of 3285 patients referred to an osteoporosis clinic over a 10‐year period and performed mutation screening of ALPL in those with low ALP (≤40 U/L) on two or more occasions. These individuals were matched with four clinic controls with a normal ALP. We also evaluated the prevalence of low ALP and ALPL mutations in 639 individuals from the general population from the same region. We identified 16/3285 (0.49%) clinic patients with low ALP and 14 (87.5%) had potentially pathogenic variants in ALPL. Eight of these individuals were heterozygous for mutations previously described in HPP and 2 were heterozygous for novel mutations (p.Arg301Trp and p.Tyr101X). These mutations were not found in clinic controls or in the general population. Eight patients with low ALP, including 4 with ALPL mutations, were treated with bisphosphonates for an average of 6.5 years. In these individuals, the rate of fractures during treatment was comparable to that in normal ALP clinic controls who were treated with bisphosphonates. We conclude that heterozygous loss‐of‐function mutations in ALPL are common in osteoporosis patients with low ALP. Further studies are required to determine how best these individuals should be treated. © 2019 The Authors. Bone Mineral Density (BMD) is a highly heritable trait, but genome-wide association studies have identified few genetic risk factors. Epidemiological studies suggest associations between BMD and several traits and diseases, but the nature of the suggestive comorbidity is still unknown. We used a novel genetic pleiotropy-informed conditional False Discovery Rate (FDR) method to identify single nucleotide polymorphisms (SNPs) associated with BMD by leveraging cardiovascular disease (CVD) associated disorders and metabolic traits. By conditioning on SNPs associated with the CVD-related phenotypes, type 1 diabetes, type 2 diabetes, systolic blood pressure, diastolic blood pressure, high density lipoprotein, low density lipoprotein, triglycerides and waist hip ratio, we identified 65 novel independent BMD loci (26 with femoral neck BMD and 47 with lumbar spine BMD) at conditional FDR < 0.01. Many of the loci were confirmed in genetic expression studies. Genes validated at the mRNA levels were characteristic for the osteoblast/osteocyte lineage, Wnt signaling pathway and bone metabolism. The results provide new insight into genetic mechanisms of variability in BMD, and a better understanding of the genetic underpinnings of clinical comorbidity.

Paget's disease of bone (PDB) has a strong genetic component. Here we investigated possible associations between genetic variants that predispose to PDB and disease severity. METHODS: Allelic variants identified as predictors of PDB from genome wide association studies were analysed in 1940 PDB patients from the UK, Italy, Western Australia and Spain. A cumulative risk allele score was constructed by adding the variants together and related to markers of disease severity, alone and in combination with SQSTM1 mutations. RESULTS: In SQSTM1 negative patients, risk allele scores in the highest tertile were associated with a 27% increase in disease extent, compared with the lowest tertile (p < 0.00001) with intermediate values in the middle tertile (20% increase; p = 0.0007). The effects were similar for disease severity score which was 15% (p = 0.01) and 25% (p < 0.00001) higher in the middle and upper tertiles, respectively. Risk allele score remained a significant predictor of extent and severity when SQSTM1 positive individuals were included, with an effect size approximately one-third of that observed with SQSTM1 mutations. A genetic risk score was developed by combining information from both markers, which identified subgroups of individuals with low, medium and high levels severity with a specificity of 70% and sensitivity of 55%. CONCLUSIONS: Risk allele scores and SQSTM1 mutations both predict extent and severity of PDB. It is possible that with further refinement, genetic profiling may be of clinical value in identifying individuals at high risk of severe disease who might benefit from enhanced surveillance and early intervention.

- Abstract
Osteoporosis pseudoglioma syndrome (OPPG) is a rare autosomal recessive condition of congenital blindness and severe childhood osteoporosis with skeletal fragility, caused by loss-of-function mutations in the LRP5 (Low-density lipoprotein receptor-related protein 5) gene. We report the first case of atypical (subtrochanteric) femoral fracture (AFF) in OPPG, occurring in a 38 year old man within the context of relatively low bone turnover and trabecular osteoporosis on bone histology. We identify two novel LRP5 mutations: R752W is associated with low bone mass density (BMD), as demonstrated by the heterozygous carriage identified in his 57 year old mother; however, the combination of this R752W mutation with another novel W79R mutation, causes a severe case of compound heterozygous OPPG. We undertake three-dimensional homology modelling of the four extracellular YWTD β-propeller/EGF-like domains (E1-E4) of LRP5, and show that both novel mutations destabilise the β-propeller domains that are critical for protein and ligand binding to regulate Wnt signalling and osteoblast function. Whilst AFFs have been reported in other rare bone diseases, this is the first in a genetic condition of primary osteoblast dysfunction. The relatively low bone turnover observed, and knowledge of LRP5 function, implicates impaired bone remodelling in the pathogenesis of AFF. © 2014 American Society for Bone and Mineral Research.

Body fat distribution is a heritable trait and a well-established predictor of adverse metabolic outcomes, independent of overall adiposity. To increase our understanding of the genetic basis of body fat distribution and its molecular links to cardiometabolic traits, here we conduct genome-wide association meta-analyses of traits related to waist and hip circumferences in up to 224,459 individuals. We identify 49 loci (33 new) associated with waist-to-hip ratio adjusted for body mass index (BMI), and an additional 19 loci newly associated with related waist and hip circumference measures (P

- Abstract
[EUS] Munduko biztanleriaren %10ak dislexia pairatzen du, eta Lehen Hezkuntzako geletan gero eta gehiago ematen ari den nahastea da. IKTek (Informazio eta Komunikaziorako Teknologiak) dislexia duten umeekin lan egiteko baliabideak eta abantailak eskaintzen dituzte. Horregatik, lan honen helburu nagusia irakasleek dislexiari buruz eta teknologiek eskaintzen dituzten aukerei buruz dituzten ezagutzak aztertzea da. Ikerketa hau aurrera eramateko galdetegi bat erabili da, LHko lehen zikloko 9 irakaslek erantzun duena. Lortutako emaitzek agerian uzten dute irakasle gehienek ez dituztela dislexiaren ezaugarri nagusiak ezagutzen eta ez dutela formakuntza nahikorik IKTetan. The results show that most teachers do not know the main characteristics of dyslexia and the lack of training in ICT. So, a proposal for the future is made in order to improve teacher training in the area of ICT.

Arquitectura - Abstract

Urban agriculture is beginning to gain importance in Spain. Nevertheless there are several difficulties for its recognition in municipal regulation, especially regarding community gardens. This perpetuates the insecurity and precariousness of these projects. Comparative analysis of urban agriculture regulation (as defined in law, urban planning, zoning codes, and other legal instruments) in western cities, has allowed us to identify operational tools and processes that work in other countries and can be adapted to Spanish urban planning. This requires a wider municipal commitment, and recognition of the potential of urban agriculture. Tentative progress are not up to par with another countries regulating framework development.

- Abstract

Herein, we present a fast, efficient and general one-pot method for the synthesis of
11 C-labelled compounds via the Negishi cross-coupling reaction. Our approach, based on the in situ formation of [11 C]CH3 ZnI and subsequent reaction with aryl halides or triflates, has proven efficient to synthesize [11 C]thymidine, a biologically relevant compound with potential applications as a proliferation marker. Theoretical calculations have shown irreversible formation of a tetracoordinated nucleophilic11 C–Zn(ii) reagent and electronic requirements for an efficient Negishi coupling.

- Abstract
Sexual dimorphism in various bone phenotypes, including bone mineral density (BMD), is widely observed; however, the extent to which genes explain these sex differences is unclear. To identify variants with different effects by sex, we examined gene-by-sex autosomal interactions genome-wide, and performed expression quantitative trait loci (eQTL) analysis and bioinformatics network analysis. We conducted an autosomal genome-wide meta-analysis of gene-by-sex interaction on lumbar spine (LS) and femoral neck (FN) BMD in 25,353 individuals from 8 cohorts. In a second stage, we followed up the 12 top single-nucleotide polymorphisms (SNPs; p?1?X?10-5) in an additional set of 24,763 individuals. Gene-by-sex interaction and sex-specific effects were examined in these 12 SNPs. We detected one novel genome-wide significant interaction associated with LS-BMD at the Chr3p26.1-p25.1 locus, near the GRM7 gene (male effect?=?0.02 and p?=?3.0?X?10-5; female effect?=?-0.007 and p?=?3.3?X?10-2), and 11 suggestive loci associated with either FN- or LS-BMD in discovery cohorts. However, there was no evidence for genome-wide significant (p?5?X?10-8) gene-by-sex interaction in the joint analysis of discovery and replication cohorts. Despite the large collaborative effort, no genome-wide significant evidence for gene-by-sex interaction was found to influence BMD variation in this screen of autosomal markers. If they exist, gene-by-sex interactions for BMD probably have weak effects, accounting for less than 0.08% of the variation in these traits per implicated SNP. Agricultura ,Urbanismo - Abstract

Derecho ,Agricultura ,Historia - Abstract

Medio Ambiente ,Arquitectura ,Urbanismo - Abstract

- Abstract
In a multi-target complex network, the links (L-ij) represent the interactions between the drug (d(i)) and the target (t(j)), characterized by different experimental measures (K-i, K-m, IC50, etc.) obtained in pharmacological assays under diverse boundary conditions (c(j)). In this work, we handle Shannon entropy measures for developing a model encompassing a multi-target network of neuroprotective/neurotoxic compounds reported in the CHEMBL database. The model predicts correctly >8300 experimental outcomes with Accuracy, Specificity, and Sensitivity above 80%-90% on training and external validation series. Indeed, the model can calculate different outcomes for >30 experimental measures in >400 different experimental protocolsin relation with >150 molecular and cellular targets on 11 different organisms (including human). Hereafter, we reported by the first time the synthesis, characterization, and experimental assays of a new series of chiral 1,2-rasagiline carbamate derivatives not reported in previous works. The experimental tests included: (1) assay in absence of neurotoxic agents; (2) in the presence of glutamate; and (3) in the presence of H2O2. Lastly, we used the new Assessing Links with Moving Averages (ALMA)-entropy model to predict possible outcomes for the new compounds in a high number of pharmacological tests not carried out experimentally.
