Background and Aims: Although acute exhaustive exercise is known to increase liver reactive oxygen species (ROS) production and aerobic training has shown to improve the antioxidant status in the liver, little is known about mitochondria adaptations to aerobic training. The main objective of this study was to investigate the effects of the aerobic training on oxidative stress markers and antioxidant defense in liver mitochondria both after training and in response to three repeated exhaustive swimming bouts., Methods: Wistar rats were divided into training (n = 14) and control (n = 14) groups. Training group performed a 6-week swimming training protocol. Subsets of training (n = 7) and control (n = 7) rats performed 3 repeated exhaustive swimming bouts with 72 h rest in between. Oxidative stress biomarkers, antioxidant activity, and mitochondria functionality were assessed., Results: Trained group showed increased reduced glutathione (GSH) content and reduced/oxidized (GSH/GSSG) ratio, higher superoxide dismutase (MnSOD) activity, and decreased lipid peroxidation in liver mitochondria. Aerobic training protected against exhaustive swimming ROS production herein characterized by decreased oxidative stress markers, higher antioxidant defenses, and increases in methyl-tetrazolium reduction and membrane potential. Trained group also presented higher time to exhaustion compared to control group., Conclusions: Swimming training induced positive adaptations in liver mitochondria of rats. Increased antioxidant defense after training coped well with exercise-produced ROS and liver mitochondria were less affected by exhaustive exercise. Therefore, liver mitochondria also adapt to exercise-induced ROS and may play an important role in exercise performance.

The aim of this study was to analyze the responses of women to the repeated bout effect (RBE) and to a short eccentric training program. Twenty-four young females were randomly assigned to a training group (TG, n = 14) or a control group (CG, n = 10). They performed two identical acute eccentric bouts (120 repetitions at 70% of 1RM) in a leg-press device in an 8 weeks interval. TG followed a 4-week-eccentric-training program between the bouts. Maximal isometric contraction, range of motion, peak power and quadriceps muscle soreness were compared between and within groups before and after the two acute eccentric bouts. TG and CG presented significant losses of isometric strength and peak power, and an increment in soreness after the first bout. Isometric strength and peak power were recovered faster in CG after the second bout (p < 0.05) compared with TG, which showed a similar recovery of these parameters after the second bout compared with the first one. A decrease in soreness and a faster recovery of range of motion were found in TG (p < 0.05) following the second bout compared with the first one, but not in CG. Data indicate that a 4-week eccentric training program may prevent the RBE over those adaptations related with muscle damage (e.g. strength loss), but it may increase RBE impact on inflammatory processes (e.g. soreness). Key pointsAn acute bout of eccentric exercise induces losses of strength, peak power and range of motion, and increases muscle soreness in young active women.When the acute eccentric bout is repeated by young women, the losses of strength and power are smaller, indicating less muscle damage. However, muscle pain and range of motion do not present any difference with the results obtained after the first bout, which would indicate that the repeated bout effect does not affect inflammatory response after acute eccentric exercise.Four weeks of eccentric training is enough to increase maximal isometric strength, but not dynamic strength (1RM) or peak power. Furthermore, this training seems to prevent those adaptations provided by the repeated bout effect related with muscle damage. On the other hand, the eccentric training seems to be a positive tool to decrease muscle soreness, and thus the inflammatory response, associated to a repeated acute eccentric bout.

Although soccer is one of the most widely played sports around the world, studies about young players and the success factors in soccer are still scarce. Therefore, the purpose of this study was to offer some insight into the factors contributing to success in this sport and to describe how physiological and technical performance evolves in young soccer players. Soccer technical skills during match play, maximum voluntary isometric contraction and power of lower limbs, jumping ability and endurance parameters were assessed in 30 prepubescent male soccer players with the same experience in soccer training. Subjects were divided into 2 groups of 15 children, a younger group (YG), aged 9.4 +/- 0.3 years, and an older group (OG), aged 11.8 +/- 0.2 years. Correlations between technical and physiological parameters were also described. Significant difference was set at p < 0.05. Differences between YG and OG appeared in physiological performance, mainly in VO(2)peak expressed in absolute values, VO(2) at different speeds, perceived exertion in treadmill test, jump performance, strength, and peak power of lower limbs. Among the technical skills measured, significant differences were found only in heading. The differences found between groups showed that most physical capacities that were measured here have an important increase during the first stages of puberty, pointing out that a specific training at these ages is necessary to get an appropriate basis for future performance. Besides, over 30% of the technical performance measured in this study can be explained with the physiological parameters. The data shown in this paper help to determine the most important capacities in youth soccer, which can facilitate the development of more appropriate selection models and trainings.

This study aimed to examine short-term resistance training effects of resting period length between sets on maximal number of repetitions and mean velocity over a moderate-intensity (60% of the maximum voluntary isometric contraction [MVIC]) set to failure on elbow-flexor muscles. The MVIC and surface electromyographic activity (sEMG) were also measured. Twenty-one untrained subjects were divided into 3 groups: short rest between sets (1 minute; SR), long rest between sets (4 minutes; LR), and nontraining control group (CG). The SR and LR performed 3 sets to failure in an arm-curl machine, 2 days per week for 5 weeks, with moderate loads (60-75% of the MVIC). The LR completed a significantly higher (31.6%, p < 0.05) total training volume than the SR. Both training groups enhanced the maximal number of repetitions to failure, with no significant differences in the magnitude of gains. The posttraining average velocity achieved by the SR at 40, 50, 60, 70, 80, and 90% of the total number of repetitions completed was significantly higher (p < 0.05) than the corresponding average velocity achieved on pretraining conditions, whereas no significant differences were observed in the LR. No significant changes in the MVIC or sEMG were observed in any group. We conclude that short-term elbow-flexor resistance training to failure, allowing 1 or 4 minutes of rest between sets, induces similar gains concerning local muscular endurance. Nevertheless, only the SR training approach reduced the rate of decline in the average repetition velocity during a set to failure. This can be of some importance in sport modalities in which not only the maximal number of repetitions (e.g., muscle endurance), but also a greater maintenance of high repetition velocities, may be critical for performance.

The purpose of this study was to examine whether and how cycle time duration affects energy expenditure and substrate utilization during whole-body vibration (WBV). Nine men performed 3 squatting exercises in execution frequency cycles of 6, 4, and 2 seconds to 90 degrees knee flexion with vibration (Vb+) (frequency was set at 30 Hz and the amplitude of vibration was 4 mm) and without vibration (Vb-) during 3 minutes, each with an additional load of 30% of the subject's body weight. A 2-way analysis of variance for VO2 revealed a significant vibration condition main effect (p < 0.001) and a cycle time duration effect (p < 0.001). When differences were analyzed by Fisher's LSD test, cycle time duration of 2 seconds was significantly different from 4 and 6 seconds, both in Vb+ and Vb-. Total energy expenditure (EE(tot)), carbohydrate oxidation rate (EE(cho)), and fat oxidation rate (EE(fat)) demonstrated a significant vibration condition main effect (EE(tot): p < 0.01; EE(cho): p < 0.001; EE(fat): p < 0.001) and cycle time duration main effect (EE(tot) and EE(cho): p < 0.001; EE(fat): p < 0.01). EE(tot), EE(cho), and EE(fat) post hoc comparisons indicated that values for the 2-second test significantly differed from 4 and 6 seconds when compared in the same vibration condition. VO2 and EE values were greater in Vb+ than in Vb- conditions with the same cycle time duration. Our study confirms that squatting at a greater frequency helps to maximize energy expenditure during exercise with or without vibration. Therefore, cycle time duration must be controlled when vibration exercise is prescribed.

Rare cases of metastasis to uterine polyps have been reported in English literature but not, to the best of our knowledge, to uterine adenomyomas. All these cases are represented by breast cancer, most of them involving tamoxifen-associated polyps. We first report a case of cutaneous malignant melanoma metastatic to uterine adenomyoma. A computed tomography scan did not reveal any further evidence of disease, suggesting that this metastatic localization may represent something more than a fortuitous case. Based on these observations it is suggested that a subset of malignant melanoma and breast cancer cells share a sort of "homing'' phenomenon to polypoid lesions of uterus, due probably to the presence of some chemokines and their specific receptors. Pathologists should be aware of this possibility in order to look carefully for metastatic implants in similar lesions. It is proposed that chemokine profile of neoplastic cells can be a useful tool in predicting metastatic targets.

Purpose: The p53 gene plays a critical role in cellular response to DNA damage and has been implicated in the response to platinum compounds in ovarian carcinoma patients. Because taxanes could induce p53-independent apoptosis, we assessed the relevance of p53 gene status to response in ovarian carcinoma patients receiving paclitaxel and platinum-containing chemotherapy., Patients and Methods: Forty-eight previously untreated patients with advanced disease received standard paclitaxel/platinum-based chemotherapy. In tumor specimens collected at the time of initial surgery, before therapy, p53 gene status and expression were examined by single-strand conformation polymorphism, sequence analysis, and immunohistochemical analysis. Microsatellite instability analysis was performed on available samples from 30 patients., Results: Thirty-four (71%) of the 48 patients had a clinical response. Pathologic complete remission was documented in 13 (27%) of 48 patients. p53 mutations were detected in 29 (60%) of 48 tumors. Among the patients with mutant p53 tumors, 25 patients (86%) responded to chemotherapy. Only nine (47%) of 19 patients with wild-type p53 tumors responded to the same treatment. The overall response rate and the complete remission rate were significantly higher among patients with mutant p53 tumors than among patients with wild-type p53 tumors (P: =.008). Most of the tested tumors not associated with complete remission (10 of 12 tumors) were also characterized by microsatellite instability. The complete remission rate was higher among patients with tumors without microsatellite instability (five of seven patients)., Conclusion: In contrast to the limited efficacy of treatment with paclitaxel in combination with standard platinum doses against wild-type p53 ovarian tumors, patients with mutant p53 ovarian tumors were more responsive to paclitaxel-based chemotherapy. The pattern of response to chemotherapy containing paclitaxel is different from that associated with high-dose cisplatin therapy. Determining p53 mutational status can be useful in predicting therapeutic response to drugs effective in ovarian carcinoma.

Purpose: To analyze the time-dependent prognostic role of the investigated variables, considered, when appropriate, on a continuous scale, for the purpose of evaluating and describing the interrelationships between clinically relevant patient and tumor characteristics (age, size and histology, and estrogen receptor [ER] and progesterone receptor content) and the risk of new disease manifestation., Patients and Methods: We applied a flexible statistical model to a case series of 1,793 patients with axillary lymph node-negative breast cancer with a minimal potential follow-up of 10 years. To avoid a potential confounding effect of adjuvant treatment, only patients given local-regional therapy until relapse were considered., Results: ER content and tumor size (adjusted for all the other covariates) showed a time-dependent relationship with the risk of new disease manifestations. In particular, ER content failed to show a prognostic effect within the first years of follow-up; thereafter, a positive association with risk of relapse was observed. For tumor size, within the first years of follow-up, the risk of relapse was directly related to size for only tumors up to 2.5 cm in diameter; thereafter, the impact on prognosis progressively decreased., Conclusion: The availability of a long follow-up on a large breast cancer series, as well as the use of innovative statistical approaches, allowed us to explore the functional relation between steroid receptors and clinical outcome and to generate a hypothesis on the involvement of ER in favoring long-term metastasis development.

The p53 protein is a multifunctional transcriptional regulator involved in cellular response to DNA damage and has been implicated as a putative determinant of sensitivity of tumor cells to cytotoxic agents. Since the p53 gene becomes inactivated in over one-half of advanced ovarian carcinoma, in this study we have examined the relationships between p53 gene alterations, p53 immunoreactivity, and response to cisplatin-based chemotherapy in ovarian cancer patients. All patients had advanced (FIGO stage III or IV) ovarian carcinoma and, with one exception, were untreated at the time of collection of tumor specimens. After initial debulking surgery, patients received high-dose cisplatin therapy. Tumor samples were analyzed for p53 gene mutations and for p53 protein accumulation, and the findings were correlated with tumor responsiveness. Of the 33 tumors examined, p53 gene mutations were found in 20 cases, including 15 missense mutations, 2 deletions, 2 nonsense mutations, and a base substitution at splice site. Twenty tumors showed positive immunostaining for p53. Only missense mutations were associated with positive immunostaining. In addition, p53 overexpression was detected in five tumors in the absence of mutations. Most (12 of 14) of the missense mutations associated with p53 protein stabilization were found refractory to therapy, as well as tumors overexpressing wild-type p53 (4 of 5). A significant correlation has been found between p53 accumulation, type of mutation (i.e., missense mutations), and pathological response to cisplatin-based therapy. In conclusion, the present results are consistent with a role of p53 as a determinant of chemosensitivity of ovarian carcinoma.

Aims: The study evaluated the safety and efficacy of granisetron as an antiemetic drug in ovarian cancer patients treated with cisplatin-based chemotherapy., Study Design: Two groups of consecutive patients were considered: the first (Group A) with advanced disease, receiving 4-day cisplatin therapy in a 40 mg/m2 daily dose; the second (B), with minimal disease after radical surgery at high risk of recurrence, treated by single-day chemotherapy with a 90 mg/m2 dose. In both groups, 3.0 mg of granisetron was administered as a 10 min. intravenous infusion, 30 min. before cisplatin infusion. The treatment schedule included in all patients the administration of 125 mg i.v. of methylprednisolone 2 h before chemotherapeutic infusion. No further doses of granisetron were allowed within each 24 h study period for breakthrough nausea and vomiting. Assessment for nausea and vomiting was made at 24 h intervals through the 6-day study period for both groups using a diary card., Results and Conclusion: In group A, 25 patients were collected and evaluated; in group B, 25 were recruited and 13 evaluated. In both groups, excellent control of nausea and vomiting was achieved, since in group A we had a global major antiemetic efficacy of granisetron in 69.2% of patients (54.4% complete control and 14.8% major control); in group B, global major efficacy was present in 83.3% of cases (31.6% complete control and 51.7% major control). The antiemetic effect in the days following antiblastic treatment lasted longer in group A.

This work comparatively evaluates the results of therapy for CIN (in terms of recovery persistence and relapse) in 362 women submitted to electrocauterization, laser vaporization, laser conization, diathermy loop conization, cold knife conization, from January 1980 to December 1989. Parameters as age, CIN grade, time elapsed between diagnosis and therapy, contraceptive exposition, have been considered. The results show a slight difference in the persistence and relapse rates between conization globally considered (traditional, laser, loop) and local surgery (cauterization, laser) in favour of the latter. Nevertheless the difference was too little to suggest abandoning one treatment in favour of the others. Moreover this work indicates that the results of therapy are independent from the type of contraceptive used. Furthermore, the finding of relapses during follow up only in CIN grade 2 indicates that this grade of CIN must be considered at risk, requesting adequate consideration and follow up.
