101. Random Seismic Response Analysis of Long-Span Steel-Concrete Composite Structure

Abstract

Considering the randomness of seismic input and the nonlinear characteristic of long-span structures, the probability density evolution method is used to study the dynamic response of the long-span steel-concrete composite structure under random earthquake. Three-dimensional structural model is established in the finite element software SAP2000, and then the displacements between layers and the response rule of vertical displacements of truss roof of the composite structure are obtained. The results show that under the input of three-dimensional earthquake, the vertical displacement of the grid roof of the isolated long-span steel-concrete composite structure is an order of magnitude larger than the horizontal direction of each layer. The probability density evolution method can offer comprehensive probability information of nodal displacements, and provide a reasonable way for the seismic reliability analysis of long-span isolated structures.

112. A novel guide device improves the accuracy of pedicle screw placement

Abstract

Pedicle screw placement in thoracic and lumbar spine is technically demanding and involves radiation exposure. Commercially available systems for pedicle screw insertion are expensive and cannot be widely used for those less-developed areas. The purpose of our study was to develop a simple guide device assisting pedicle screw placement and improving the accuracy in the operation. A simple guide device was developed with the aim of improving accuracy of pedicle screw technique based on anatomical structures of the spine. We retrospectively collected clinical data of 111 consecutive patients who received treatment of pedicle screw internal fixation between January 2013 and September 2014. In total, 518 screws were inserted by the same surgeon, among which 280 screws were implanted in 60 patients (33 males and 27 females) with conventional freehand technique and 238 screws were implanted in 51 patients (27 males and 24 females) using the guide device. According to postoperative CT evaluation, screws were classified into different grades and accuracy rates were compared between the two groups. A total of 518 screws were placed (238 in the guide group, 280 in the conventional group). No intra-operative or postoperative complications such as infection, vessel and nerve injuries occurred. After postoperative CT evaluation, 215 from guide group and 236 from the conventional group were classified in Grade 1. Only screws in Grade 1 were considered as accurate insertion. Thus, the accuracy rates were 84.3% for the conventional group and 90.3% for the guide group respectively. Through statistical test, there was significant difference in the accuracy rate between two groups. The simple guide device significantly improves the accuracy of pedicle screw placement. Combined with the advantage of cheapness, it can be widely used in spinal internal fixations, especially suitable for those inexperienced spine surgeons in less-developed areas.

Abstract

The purpose of the present study was to identify the key long non-coding (lnc)RNAs in the occurrence and development of osteoporosis (OP) and to explore the associated molecular mechanism. First, the Gene Expression Omnibus (GEO) datasets, with key words 'osteoporosis' and 'HG-133A', were screened. RankProd R package was used to calculate the dysregulated lncRNAs in OP. Following this, bone marrow mesenchymal stem cells (BM-MSCs) harvested from 3-week-old Sprague Dawley rats were employed for detection of osteoblast differentiation. Following overexpression or interference with X-inactive specific transcript (XIST), osteogenesis-associated genes and proteins in BM-MSCs were detected using reverse transcription-quantitative polymerase chain reaction and western blot analysis. Alkaline phosphatase (ALP) and Alizarin Red S staining were also performed to measure the osteogenic ability of BM-MSCs. Results from the two datasets indicated that 6 lncRNAs were dysregulated in OP. Notably, XIST is key lncRNA in diverse diseases, and was subsequently selected for analysis. It was revealed that XIST was significantly upregulated in plasma and monocytes from patients with OP compared with the normal controls. Furthermore, results indicated that overexpression of XIST significantly inhibited osteoblast differentiation in BM-MSCs, as evidenced by the decreased expression of ALP, bone γ-carboxyglutamic acid-containing protein and runt related transcription factor 2, reduced ALP activity and a decreased number of calcium deposits. However, interference of XIST exhibited the opposite biological effects in BM-MSCs. Taken together, XIST was highly expressed in the serum and monocytes of patients with OP. Abstract

A fiber optic fluorescence biosensor for choline is introduced in this paper. Choline is an important neurotransmitter in mammals. Due to the growing needs for on-site clinical monitoring of the choline, much effect has been devoted to develop choline biosensors. Fiber-optic fluorescence biosensors have many advantages, including miniaturization, flexibility, and lack of electrical contact and interference. The choline fiber-optic biosensor we designed implemented a bifurcated fiber to perform fluorescence measurements. The light of the blue LED is coupled into one end of the fiber as excitation and the emission spectrum from sensing film is monitored by fiber-spectrometer (S2000, Ocean Optics) through the other end of the fiber. The sensing end of the fiber is coated with Nafion film dispersed with choline oxidase and oxygen sensitive luminescent Ru(II) complex (Tris(2,2'-bipyridyl)dichlororuthenium(II), hexahydrate). Choline oxidase catalyzes the oxidation of choline to betaine and hydrogen peroxide while consuming oxygen. The fluorescence intensity of oxygen- sensitive Ru(II) are related to the choline concentration. The response of the fiber-optic sensor in choline solution is represented and discussed. Abstract

Mycobacterium tuberculosisis the causative agent of tuberculosis (TB), which is still the leading cause of mortality from a single infectious disease worldwide. The development of novel anti-TB drugs and vaccines is severely hampered by the complicated and time-consuming genetic manipulation techniques for M. tuberculosis.Here, we harnessed an endogenous type III-A CRISPR/Cas10 system of M. tuberculosisfor efficient gene editingand RNA interference (RNAi). This simple and easy method only needs to transform a single mini-CRISPR array plasmid, thus avoiding the introduction of exogenous protein and minimizing proteotoxicity. We demonstrated that M. tuberculosisgenes can be efficiently and specifically knocked in/out by this system as confirmed by DNA high-throughput sequencing. This system was further applied to single- and multiple-gene RNAi. Moreover, we successfully performed genome-wide RNAi screeningto identify M. tuberculosisgenes regulating in vitroand intracellular growth. This system can be extensively used for exploring the functional genomics of M. tuberculosisand facilitate the development of novel anti-TB drugs and vaccines.

Abstract

[Display omitted] • A new snail shell structure is used as the prototype of the optimal design. • The multi-objectives are changed into one objective through the weight coefficient. • The designed labyrinth seal has good load-bearing and heat insulation performance. Labyrinth seals are widely used in turbomachinery devices to prevent the leakage of fluid medium in seal structures. Under high parametrical environments, the heat generated by frictional resistance of the turbulence between the stator and rotor walls not only increases the temperature of the fluid and the solid, but also reduces the operation efficiency of the device. Therefore, it is necessary to implement the fluid–solid-thermal multi-field analysis of labyrinth seals and propose a comprehensive optimization design of the load-bearing and heat insulation. In the present study, inspired by a bionic snail shell, a sandwich structure with load-bearing material on both upper and lower surfaces and the heat insulation material in the middle is obtained through the genetic algorithm optimization. Then, the luid-solid-thermal multi-field analysis of the labyrinth seal with the sandwich structure is carried out. It is verified that the layer arrangement and layer thickness ratio of the bionic structure ensured the optimal performance of load-bearing and heat insulation. After changing the layer arrangement or layer thickness ratio, the comprehensive performance of load-bearing and heat insulation is obviously weakened. The result shows that the bionic multilayer sandwich structure can be applied to the labyrinth seal to achieve optimal comprehensive performance of load-bearing and heat insulation.
