A growing literature demonstrates the impact of helminths on their host gut microbiome. We investigated whether the stickleback host microbiome depends on ecoevolutionary variables by testing the impact of exposure to the cestode parasite Schistocephalus solidus with respect to infection success, host genotype, parasite genotype, and parasite microbiome composition. We observed constitutive differences in the microbiome of sticklebacks of different origin, and those differences increased when sticklebacks exposed to the parasite resisted infection. In contrast, the microbiome of successfully infected sticklebacks varied with parasite genotype. More specifically, we revealed that the association between microbiome and immune gene expression increased in infected individuals and varied with parasite genotype. In addition, we showed that S. solidus hosts a complex endomicrobiome and that bacterial abundance in the parasite correlates with expression of host immune genes. Within this comprehensive analysis we demonstrated that (i) parasites contribute to modulating the host microbiome through both successful and unsuccessful infection, (ii) when infection is successful, the host microbiome varies with parasite genotype due to genotype-dependent variation in parasite immunomodulation, and (iii) the parasite-associated microbiome is distinct from its host and impacts the host immune response to infection.

In the past 50 years, enormous progress has been made in the diagnosis, treatment and control of taeniid cestode infections/diseases and in the scientific understanding thereof. Most interest in this group of parasites stems from the serious diseases that they cause in humans. It is through this lens that we summarize here the most important breakthroughs that have made a difference to the treatment of human diseases caused by these parasites, reduction in transmission of the taeniid species associated with human disease, or understanding of the parasites’ biology likely to impact diagnosis or treatment in the foreseeable future. Key topics discussed are the introduction of anti-cestode drugs, including benzimidazoles and praziquantel, and the development of new imaging modalities that have transformed the diagnosis and post-treatment monitoring of human echinococcoses and neurocysticercosis. The availability of new anti-cestode drugs for use in dogs and a detailed understanding of the transmission dynamics of Echinococcus granulosus sensu lato have underpinned successful programs that have eliminated cystic echinococcosis in some areas of the world and greatly reduced the incidence of infection in others. Despite these successes, cystic and alveolar echinococcosis and neurocysticercosis continue to be prevalent in many parts of the world, requiring new or renewed efforts to prevent the associated taeniid infections. Major advances made in the development of practical vaccines against E. granulosus and Taenia solium will hopefully assist in this endeavour, as might the understanding of the parasites’ biology that have come from an elucidation of the nuclear genomes of each of the most important taeniid species causing human diseases.

The Lecanicephalidea represents one of 11 currently recognized elasmobranch-hosted tapeworm lineages. It is quite speciose, with nearly 100 validly recognized species to date. Yet spermatozoon features have been fully characterized for only a single species; spermatozoon characters for a second species were previously included in a data matrix used to infer phylogenetic relationships among cestode orders, but data are limited and no images were provided. Specimens of Tetragonocephalum sp. were collected from the whipray, Urogymnus asperrimus 1, from the Solomon Sea off the Solomon Islands. The mature spermatozoa of Tetragonocephalum sp. are distinctly different from the other lecanicephalidean species for which spermatozoon ultrastructure has been thoroughly investigated. Tetragonocephalum sp. spermatozoa represent the Type IV (sensu Levron, 2010) morphology possessing a single axoneme, crested bodies, cortical microtubules running parallel to the axoneme, and nucleus, which is helical to the axoneme. Although spermatozoa for both lecanicephalideans examined to date are of the Type IV spermatozoon morphology, they differ in the number of crested bodies and cortical microtubules, the relative size of the spermatozoon-specifically as it relates to the nuclear region-and the presence or absence of the anterior spiral structure. Significant spermatozoon character variation is present across these 2 representatives of 2 lecanicephalidean families, indicating that additional studies of representatives of the other families in the order are needed to better understand the extent of character variation present in the Lecanicephalidea. Overall, spermatozoon ultrastructure is understudied in elasmobranch-hosted tapeworm lineages compared to other vertebrate-hosted tapeworm orders at both the species and family levels.

We report molecular identification of an adult Spirometra mansoni tapeworm retrieved from a crab-eating fox (Cerdocyon thous) in Colombia, confirming presence of this parasite in South America. This tapeworm is the causative agent of human sparganosis, commonly reported from Southeast Asia, and represents the second congeneric species with known zoonotic potential in the Americas.

Spirometra infection is aneglected food- and waterborne disease with worldwide distribution.The present study aims to estimate the global prevalence of Spirometra species in snakes, frogs, dogs and cats.Multiple databases (PubMed, Scopus, ProQuest, Web of Science and Google Scholar) were searched for relevant literatures published up to March 2022.Among 131 data sets (including 113 articles) that met the inclusion, 15 investigations reported Spirometra infection in snakes, 23 in frogs, 41 in dogs and 52 in cats. The pooled prevalence (95% confidence interval) in intermediate hosts and definitive hosts was found to be 0.313% and 0.089%, respectively. Based on continent, the infection was most prevalent in Asia for studies on snakes (0.696%) and frogs (0.181%), while Africa (0.224%) and Oceania (0.203%) were the regions with the highest pooled prevalence rates of the infection in dogs and cats, respectively. Among different diagnostic methods, the highest pooled prevalence was related to morphological method for studies on snakes, frog and cats with rate of 0.665%, 0.189% and 0.104%, respectively. Regarding studies on dogs, the highest pooled prevalence was observed for molecular technique (0.101%).The results presented here revealed the importance of establishing a prevention and control measure focused on protection of aquaculture systems from being contaminated with faeces of dogs and cats, and raising awareness of parasitic zoonotic diseases to decrease the transmission risk.

The mechanisms enabling fish tapeworms to avoid proteolytic attacks by digestive enzymes of their fish host have been studied in less detail compared with mammalian cestodes. This study aimed to assess the inhibitory ability towards trypsin and chymotrypsin in Eubothrium rugosum, an intestinal parasite of burbot Lota lota, and establish its localization in the tapeworm. To this end, the worms were treated with Triton X-100 followed by differential centrifugation to isolate the tegumental brush border membrane. The protease inhibitory abilities of the worms were mostly determined by their excretory/secretory products released into the incubation medium. These inhibitory abilities proved to be linked mainly with the brush border fractions. Notably, the per cent inhibition of both studied digestive enzymes (trypsin and chymotrypsin) hardly depended on the duration of the parasite exposure in the incubation medium, probably due to intermittent glycocalyx renewal. Improved knowledge on functions of the excretory/secretory proteins produced by fish tapeworms may contribute to a better understanding of host-parasite relations and development of new approaches to the treatment and prevention of diseases caused by pathogenic helminths.

Two new species of Scalithrium Ball, Neifar & Euzet, 2003 were found in rajiform batoids of the Argentine Sea: Scalithrium kirchneri sp. nov., parasite of the Rio skate Rioraja agassizii (Müller & Henle) from waters off Buenos Aires Province, and Scalithrium ivanovae sp. nov., parasite of the La Plata skate Atlantoraja platana (Günther) from waters off Río Negro Province. Scalithrium kirchneri sp. nov. and S. ivanovae sp. nov. differ from their congeners by having a particular combination of features including: total length, number of loculi on the distal bothridial surface, absence of bilobed bothridia and number of testes. Additionally, S. kirchneri sp. nov. can be distinguished from S. ivanovae sp. nov. by the presence of a vaginal sphincter and seminal receptacle, by the position of the entrance of the vas deferens into the cirrus sac, and by the microthrix pattern on the proximal bothridial surface. Based on this and previous studies, we conclude that some morphological traits may not be considered as key characters for the generic diagnosis of Scalithrium due to their variability within the genus. These characters include the musculature disposition in bothridial transverse septa and position of the entrance of the vas deferens into the cirrus sac, which certainly complement other diagnostic characters at a specific level. The discovery of these two species represents the first record of the genus Scalithrium in temperate waters of the Southern Hemisphere. In addition, this study is the first to undoubtedly identify batoids from the order Rajiformes as hosts for Scalithrium.

The Rhoptrobothriidae are one of the more enigmatic families of cestodes of elasmobranchs. Opinions on the taxonomic status of the family’s three original genera (i.e., Myzophyllobothrium, Rhoptrobothrium, and Myzocephalus) have varied over the 115 years since they were erected. Some authors have considered all three valid, others have considered Rhoptrobothrium to be a synonym of Myzopyllobothrium or a genus inquirendum, yet others have considered Myzocephalus to be a synonym of the phyllobothriid genus Thysanocephalum. All three genera were established for specimens collected from eagle rays off Sri Lanka. The erection of Mixophyllobothrium for two specimens from a cowtail stingray off India three decades ago added additional confusion to the situation, with some authors considering it valid and others a synonym of Myzocephalus. These disagreements stem largely from differences in interpretation of the complex morphology of the scolex of members of these genera. Furthermore, with the exception of Rhoptrobothrium comprising four species, each genus is monotypic. All but Rhoptrobothrium has not been considered in detail for nearly a century, largely because of a lack of available material. The taxonomic status of these genera is assessed here based on light and scanning electron microscopy, and molecular data generated from new material collected from eagle rays off Indonesian and Malaysian Borneo, Japan, Sri Lanka, and Viet Nam. Morphological work indicates that the genera differ largely only in the degree of folding of the four remi that extend from the cephalic peduncle. A molecular phylogeny based on sequence data for the D1–D3 region of the 28S rRNA gene, which include new data for eight specimens of four species, indicates that Myzophyllobothrium, Myzocephalus, and Rhoptrobothrium are not mutually monophyletic. The latter two genera and Mixophyllobothrium are considered synonyms of Myzophyllobothrium and five species are transferred to that genus. Myzophyllobothrium okamuri n. comb. is considered a species inquirendum. Myzophyllobothrium nagasawai n. sp. is described from Aetobatus narutobiei off Japan. Myzophyllobothrium narinari n. comb. is re-described based on newly collected cestodes from the type host and locality (i.e., Aetobatus ocellatus off Sri Lanka). Despite consisting of only a single genus, the family status of the group is retained in recognition of the unusual configuration of the scolex, which bears four biloculate bothridia and four remi extending from the cephalic peduncle. The ordinal placement of the family remains uncertain, but affinities with the Phyllobothriidea, rather than “Tetraphyllidea” are considered.

Red foxes (Vulpes vulpes) are the most prevalent wild carnivores in the world and definitive hosts of many pathogenic parasites for humans and farm animals. These animals travel great distances in search of prey and nests, and cause contamination of large geographic areas with parasites. For this reason, monitoring the parasitic pathogens of red foxes is particularly important in terms of public and animal health. The goal of this study was to determine the intestinal helminths and molecular characterization of Taenia species of red foxes in Turkey. In this study, 103 red fox intestines obtained from 29 provinces of Turkey were examined with sedimentation and counting technique. Collected helminths were diagnosed according to their morphologic features. Additionally, further molecular analysis (PCR and DNA sequencing) was performed for the identification of Taeniid cestodes. At the end of the study, it was determined that 87.37% (90/103) of red foxes were infected with at least one helminth species. Detected helminths and their prevalence’s were Mesocestoides sp. (56.31%), Joyeuxiella echinorhynchoides (33%), Taenia polyacantha (15.53%), Dipylidium caninum (0.97%), Pterygodermatites affinis (51.45%), Toxascaris leonina (45.63%), Uncinaria stenocephala (33%), Oxynema numicidum (20.38%), Toxocara canis (14.56%), Ancylostoma caninum (12.62%), and Trichuris vulpis (1.94%), respectively. Additionally, Pachysentis sp. (37.69%), Centrorhynchus sp. (0.97%) (Acantocephala), and nymphs of Linguatula serrata (20.38%) (Arthropoda) were also detected in the same intestinal samples. This is the most comprehensive study that has been conducted on the intestinal helminthes of red foxes in Turkey. To the best of our knowledge, molecular characterization of T. polyacantha and the detection of O. numicidum, A. caninum, Pachysentis sp., and Centrorhynchus sp. are the first reports in red foxes in Turkey. Praziquantel is the drug of choice for treating the tapeworm Dipylidium caninum in dogs; however, resistance is possible, and regular, non-targeted administration of praziquantel may select for anthelminthic-resistant populations.The zinc sulfate fecal floatation procedure was conducted. Gross visualization was used to identify Dipylidium spp. segments, and capsule endoscopy was used to visualize adult tapeworms within the intestinal tract.An 18-month-old spayed female terrier mix was presented due to diarrhea, hematochezia and weight loss. The dog received appropriate anthelmintic therapy for Giardia spp., Ancylostoma spp. and Dipylidium spp. The dog's clinical signs resolved, and elimination of Ancylostoma spp. was confirmed by subsequent fecal analysis. However, Dipylidium spp. segments were repeatedly present in the stool. Observation of the segments confirmed the presence of adult Dipylidium spp in feces. Treatment with praziquantel and epsiprantel were unsuccessful in eliminating the organism but was apparently successful in flea prevention. A single dose of nitrosconate was administered and eliminated Dipylidium spp. infection in the dog.Nitrosconate can be an effective treatment for praziquantel-resistant dipylidiasis in dogs. The novel application of capsule endoscopy confirmed the anthelmintic efficacy of this treatment.

Based on morphological and molecular data, a new species of tapeworm, Bothriocephalus kupermani n. sp., is described from pumpkinseed, Lepomis gibbosus (Linnaeus, 1758) (type host), and green sunfish, L. cyanellus Rafinesque, 1819 (Actinopterygii: Centrarchidae) in the United States. The new species differs from its North American congeners mainly in the shape of its scolex, which is almost rectangular in dorsoventral and lateral views (nearly parallel margins in the bothrial region), with shallow, wide bothria, and by an extensive field of vitelline follicles that are also variably present, albeit more sparsely, medially in the cortex of the ovarian and uterine areas. This tapeworm appears to be typically associated with Lepomis sunfishes. Examination of museum specimens of Bothriocephalus species from Lepomis spp. indicates that the tapeworm also parasitizes bluegill, Lepomis macrochirus, longear sunfish, Lepomis megalotis, and that previous records of Bothriocephalus cuspidatus and Bothriocephalus claviceps in these fishes are of the new species, B. kupermani.

Cestode infections is widely reported in rodents, however species identification remains problematic due to the genetic or interspecies variation. Therefore, this study was aimed to verify the Cyclophyllidean parasites recovered from wild rats captured from different forest types using molecular based methods. Maximum-likelihood (ML) and neighbour-joining (NJ) trees were constructed inferred from 18 small subunit nuclear ribosomal RNA gene (18SrDNA) and mitochondrial cytochrome c oxidase subunit one gene (COX1) sequences of cestode worms recovered from 124 individuals from four rat species. Sequences obtained from both Hymenolepis diminuta and Hydatigera parva represents the first records in Malaysia. All the sequences were successfully amplified with product with total length of 205 and 1202 base pairs (bp), respectively. Three cestode species from the Family Hymenolepididae (Hymenolepis diminuta) and Family Taeniidae (Hydatigera parva; Hydatigera taeniaeformis) were successfully characterized using phylogenetic analyses and haplotype networking. Phylogenetic analysis showed that H. diminuta, Hydatigera parva (Hy. parva) and Hydatigera taeniaeformis (Hy. taeniaeformis) formed its own monophyletic clade in 18SrDNA analyses. Results also showed that Hy. taeniaeformis shared the same haplotype group with Hy. taeniaeformis from China (COX1) and linked with Hy. taeniaeformis from Japan (18SrDNA) while the Malaysian H. diminuta clearly formed a separate haplotype and networked with other regions. The Malaysian Hy. parva isolation, on the other hand, appeared to be genetically distinct from the European Hy. parva (Spain) strain, but closely linked to the local isolates. Molecular methods employed successfully improved in the detection of complex species in this group. The findings showed that molecular data can be useful to deeply study intra-specific variation in other cestode worms.

The phylum Platyhelminthes shares a unique population of undifferentiated cells responsible for the proliferation capacity needed for cell renewal, growth, tissue repair and regeneration. These cells have been extensively studied in free-living flatworms, whereas in cestodes the presence of a set of undifferentiated cells, known as germinative cells, has been demonstrated in classical morphology studies, but poorly characterized with molecular biology approaches. Background Matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry (MS) has become a widely used technique for the rapid and accurate identification of bacteria, mycobacteria and certain fungal pathogens in the clinical microbiology laboratory. Thus far, only few attempts have been made to apply the technique in clinical parasitology, particularly regarding helminth identification. Methods We systematically reviewed the scientific literature on studies pertaining to MALDI-TOF MS as a diagnostic technique for helminths (cestodes, nematodes and trematodes) of medical and veterinary importance. Readily available electronic databases (i.e. PubMed/MEDLINE, ScienceDirect, Cochrane Library, Web of Science and Google Scholar) were searched from inception to 10 October 2018, without restriction on year of publication or language. The titles and abstracts of studies were screened for eligibility by two independent reviewers. Relevant articles were read in full and included in the systematic review. Results A total of 84 peer-reviewed articles were considered for the final analysis. Most papers reported on the application of MALDI-TOF for the study of Caenorhabditis elegans, and the technique was primarily used for identification of specific proteins rather than entire pathogens. Since 2015, a small number of studies documented the successful use of MALDI-TOF MS for species-specific identification of nematodes of human and veterinary importance, such as Trichinella spp. and Dirofilaria spp. However, the quality of available data and the number of examined helminth samples was low. Conclusions Data on the use of MALDI-TOF MS for the diagnosis of helminths are scarce, but recent evidence suggests a potential role for a reliable identification of nematodes. Future research should explore the diagnostic accuracy of MALDI-TOF MS for identification of (i) adult helminths, larvae and eggs shed in faecal samples; and (ii) helminth-related proteins that are detectable in serum or body fluids of infected individuals. The structural response and plasticity of the cestode tegument in response to the influence of the host organism is not yet well understood. The main aims of our in vitro study were to analyse the ultrastructural mechanisms and kinetics of tegumental secretion in two cestode species, Dibothriocephalus dendriticus and Ligula interrupta, in response to the influence of fish host blood serum. The incubation of plerocercoids in the culture medium, which contained fish host blood serum, resulted in an increased number of secretory products on the tegumental surface. Our study is the first to experimentally demonstrate the formation of plerocercoid protective layers influenced by the host's internal environment factors. The mechanism of the generation of the protective layer included the following: the intensive formation of organelles in the tegumental cytons and their transfer to the distal cytoplasm of the tegument; increases in extracellular vesicles and vacuoles released on the tegumental surface; arrangement of secretory products and fine-dispersed extracellular matrix in layers; and formation of the protective layer. The structural tegumental response included increases in the glycocalyx layer and structural changes. Our study revealed that the universal mechanism of protective layer formation was intrinsic to different tapeworms. We hypothesize that plerocercoids of cestodes parasitizing fish may use tegumental secretion in the formation of a protective layer and in the release of immunoregulator molecules to evade the host's immune response.
