Abstract

Recent advances in end-to-end continuous-flow synthesis are rapidly expanding the capabilities of automated customized syntheses of small-molecule pharmacophores, resulting in considerable industrial and societal impacts; however, many hurdles persist that limit the number of sequential steps that can be achieved in such systems, including solvent and reagent incompatibility between individual steps, cumulated by-product formation, risk of clogging and mismatch of timescales between steps in a processing chain. To address these limitations, herein we report a strategy that merges solid-phase synthesis and continuous-flow operation, enabling push-button automated multistep syntheses of active pharmaceutical ingredients. We demonstrate our platform with a six-step synthesis of prexasertib in 65% isolated yield after 32 h of continuous execution. As there are no interactions between individual synthetic steps in the sequence, the established chemical recipe file was directly adopted or slightly modified for the synthesis of twenty-three prexasertib derivatives, enabling both automated early and late-stage diversification., Although strategies for the automated assembly of compounds of pharmaceutical relevance is a growing field of research, the synthesis of small-molecule pharmacophores remains a predominantly manual process. Now, an automated six-step synthesis of prexasertib is achieved by multistep solid-phase chemistry in a continuous-flow fashion using a chemical recipe file that enables automated scaffold modification through both early and late-stage diversification.

Abstract

Butyrolactone I (BTL-I), a butenolide compound isolated from land or marine-derived fungi, has been reported to show diverse activities. To further study the pharmaceutical potential of BTL-I, transcriptome and bioinformatics analysis of BTL-I treated HepG2 cells were taken. BTL-I was revealed with lipid metabolism regulatory activity and confirmed by increasing the mRNA expression of related genes, such as LXRα and its target gene UGT1A1. However, the obvious chemical carcinogenesis of BTL-I was also disclosed. BTL-I could significantly increase the mRNA and protein levels of oncogenes such as CYP1A1. Molecular docking of BTL-I and its analogs were performed to understand the active or toxic effects. Abstract

By using the methods such as literature review, expert interview, video analysis and mathematical statistics, this paper analyzes the application of VAR in 240 matches of 30 rounds of China League One in 2019 season. Through the analysis of factors such as the number of reviews, time of review, classification, accuracy rate and the influence on the teams’ results of VAR technology, it is found that the accuracy rate of key decisions is significantly improved, and VAR intervention boundaries are not so clear. The wrong judgment of goals and penalty kicks was corrected in time, and fouls involving red cards could receive effective penalties. VAR technology has no great impact on the flow of the competition. However, for the use of VAR technology in China League One, intervention boundaries are not accurately got hold of, VAR is not operated well, referees are not very meticulous and comprehensive in the VAR viewing process, and communication between VAR and referees is not standardized, which needs to be further improved.

Abstract

Transition-metal-catalyzed homogeneous dehydrogenation and isomerization are common organic molecular activation reactions. Palladium hydrides are good olefin isomerization catalysts but are usually short-lived species under redox-active dehydrogenation conditions. Here, we show that Pd-H in the presence of an N-heterocyclic carbene ligand and an alkene regulator enables transfer-dehydroaromatization, avoiding the homo-disproportionation pathway. The desired product is obtained with up to 99:1 selectivity, and the exo-to-endo olefin isomerization can be carried out in one pot. In contrast to previously reported methods that rely on the efficient removal of Pd-H, the approach reported herein benefits from the steric effects of the N-heterocyclic carbene and the choice of alkene to regulate the competing reactivity of allylic C‒H activation and hydropalladation. This method circumvents the challenges associated with tedious olefin separation and a low exo-to-endo olefin isomerization ratio and expands the scope to include challenging endo- and exo-cyclic olefins under mild, neutral, and oxidant-free conditions. Overall, herein, we provide a strategy to synthesize (hetero)aromatic compounds via chemoselective dehydrogenation of cyclic alkenes over ketones and the dehydrogenative Diels-Alder reaction of a cyclic enamine. Aromatic compounds can be prepared via dehydrogenation of cyclic compounds. Here the authors report the dehydroaromatization of endocyclic and exocyclic olefins via chain-walking isomerization and transfer-dehydrogenation catalyzed by palladium N-heterocyclic carbene complexes in the presence of alkenes as sacrificial reagents. Abstract

The combination of surface plasmon resonance (SPR) effect with hetero‐p–n structure shows promising benefits to optical linear and nonlinear properties. In this study, Au nanoparticles (NPs) decorated p–n hetero‐structured Co3O4/Bi2WO6 composite was synthesized and characterized in terms of the optical linear and nonlinear and magnetic properties, morphology, electron transition, charge transfer, energy band gap, polarizability, SPR effect, and oxygen vacancies using scanning electron microscopy, X‐ray photoelectron spectroscopy, electron paramagnetic resonance spectroscopy, Z‐scan, ultraviolet–visible spectra, and vibrating sample magnetometer. The combination of Co3O4 provided active 3d electrons transition and charge transfer which increased carriers' concentration and reduced the energy band gap. Au SPR enhanced the internal polarization and strengthened the built‐in electric field, yielding strong nonlinear behavior. In addition, magnetic Co3O4 endowed sample with room‐temperature ferromagnetism which was obviously strengthened by Au NPs. The obtained sample is promising for laser and photonics applications.
