Universal design knowledge has been addressed in universities, but students cannot imagine how age affects the needs of senior citizens physically and psychologically. The teaching practice in this study adopted a dyadic teaching method whereby students observed the needs of individuals in a nursing care center for elderly people. It also used cooperative learning methods for nursing students to discuss the needs of senior citizens and to help design students understand the lifestyles of senior citizens through the use of simulation devices. Through the feedback from the learning process, statistical analysis was used to assess the learning performance of students. The design studio used these two teaching methods, and the respective work was entered into an open design competition to verify whether teaching performance was successful. Approximately 70% of the works earned design awards. This provided a preliminary validation of the success of the respective teaching practice method.

AIMS: Hypertension (HTN) can lead to heart and kidney damage. The gut microbiota has been linked to HTN, although it is difficult to estimate its significance due to the variety of other features known to influence HTN. In the present study, we used germ-free (GF) and colonized (COL) littermate mice to quantify the impact of microbial colonization on organ damage in HTN. METHODS AND RESULTS: Four-week-old male GF C57BL/6J littermates were randomized to remain GF or receive microbial colonization. HTN was induced by subcutaneous infusion with angiotensin (Ang) II (1.44 mg/kg/d) and 1% NaCl in the drinking water; sham-treated mice served as control. Renal damage was exacerbated in GF mice, whereas cardiac damage was more comparable between COL and GF, suggesting that the kidney is more sensitive to microbial influence. Multivariate analysis revealed a larger effect of HTN in GF mice. Serum metabolomics demonstrated that the colonization status influences circulating metabolites relevant to HTN. Importantly, GF mice were deficient in anti-inflammatory fecal short-chain fatty acids (SCFA). Flow cytometry showed that the microbiome has an impact on the induction of anti-hypertensive myeloid-derived suppressor cells and pro-inflammatory Th17 cells in HTN. In vitro inducibility of Th17 cells was significantly higher for cells isolated from GF than conventionally raised mice. CONCLUSIONS: Microbial colonization status of mice had potent effects on their phenotypic response to a hypertensive stimulus, and the kidney is a highly microbiota-susceptible target organ in HTN. The magnitude of the pathogenic response in GF mice underscores the role of the microbiome in mediating inflammation in HTN. TRANSLATION PERSPECTIVE: To assess the potential of microbiota-targeted interventions to prevent organ damage in hypertension, an accurate quantification of microbial influence is necessary. We provide evidence that the development of hypertensive organ damage is dependent on colonization status and suggest that a healthy microbiota provides anti-hypertensive immune and metabolic signals to the host. In the absence of normal symbiotic host-microbiome interactions, hypertensive damage to the kidney in particular is exacerbated. We suggest that hypertensive patients experiencing perturbations to the microbiota, which are common in CVD, may be at a greater risk for target-organ damage than those with a healthy microbiome.

Located in tropical and subtropical region, Taiwan is an island with high algal species diversity. In this study, a number of fungal strains were isolated from marine macroalgae collected from northeastern intertidal zone of Taiwan. Preliminary anti-inflammatory screening has shown that the methanolic extracts of solid fermented products of the red alga Mastophora rosea-derived fungal strain Acremonium sp. NTU492 exhibited significant bioactivity. In an attempt to disclose the active principles from this fungal strain, a series of separation and purification was thus undertaken, which has led to the isolation and characterization of seven compounds including four new peptides, namely acrepeptins A-D (1-4), along with previously reported destruxin B (5), guangomide A (6), and guangomide B (7). Their structures were elucidated by spectroscopic analysis and compared with literatures. This investigation was designed to evaluate the healing response of 9.3-μm CO2 laser-assisted periodontal therapy. Five patients presenting with moderate to severe periodontitis, with an initial pocket depth (PD) ≥ 5 mm and with teeth predetermined to be surgically extracted, were enrolled and consented to treatment with full-mouth CO2 laser-assisted therapy. The laser treatment was carried out in the Ultraguide Mode at a setting of 0.25-mm spot size, with an average power of 0.65 to 1.15 watts and 20% mist. The laser tip was passed from the gingival margin and down apically to the base of the pocket with a sweeping L motion. The teeth were intensely scaled with piezoultrasonic instrumentation afterwards. A second pass of the laser tip was performed for the study teeth. At 9 months, all patients were clinically reevaluated. For sites with an initial pocket depth of ≥ 7 mm, a mean PD reduction of 3.97 ± 1.36 mm and a mean clinical attachment level gain of 3.54 ± 1.54 mm were achieved, resulting in a mean PD of 3.91 ± 0.77 mm. En bloc biopsy samples of four teeth were obtained and analyzed; two demonstrated histologic evidence of new bone formation while the other two healed with a long junctional epithelium with minimal inflammatory infiltrate. Further long-term clinical studies are needed to investigate the treatment stability obtained with a 9.3-μm CO2 laser compared to conventional surgical therapy. Nevertheless, the encouraging clinical results indicated that adjunctive use of the 9.3-μm CO2 laser-assisted periodontal therapy can be beneficial for treatment of periodontally compromised patients.

Selective inhibitors of gut bacterial β-glucuronidases (GUSs) are of particular interest in the prevention of xenobiotic-induced toxicities. This study reports the first structure-activity relationships on potency and selectivity of several iminocyclitols (2-7) for the GUSs. Complex structures of Ruminococcus gnavus GUS with 2-7 explained how charge, conformation, and substituent of iminocyclitols affect their potency and selectivity. N1 of uronic isofagomine (2) made strong electrostatic interactions with two catalytic glutamates of GUSs, resulting in the most potent inhibition (Ki ≥ 11 nM). C6-propyl analogue of 2 (6) displayed 700-fold selectivity for opportunistic bacterial GUSs (Ki = 74 nM for E. coli GUS and 51.8 μM for RgGUS). In comparison with 2, there was 200-fold enhancement in the selectivity, which was attributed to differential interactions between the propyl group and loop 5 residues of the GUSs. The results provide useful insights to develop potent and selective inhibitors for undesired GUSs.

Background/Aim: Casein kinase 1 epsilon (CK1ε) is a member of the casein kinase 1 family, which includes highly conserved and ubiquitous serine/threonine protein kinases. Recent research has revealed that CK1ε plays an important role in a variety of human cancer types; however, its role in human melanoma remains unclear. The aim of this study was to elucidate the clinical role of CK1ε in patients with melanoma. Patients and Methods: Samples from 34 patients with melanoma were analyzed by immunohistochemical staining. Formalin-fixed paraffin-embedded tissue microarrays were also examined by two histopathologists to assess CK1ε protein expression in humans. Results: Cytoplasmic CK1ε protein expression was significantly lower in tumor tissue than in normal tissue. Lack of cytoplasmic CK1ε protein was significantly correlated with distant metastasis (p=0.022) and poorer survival (p=0.030). However, Kaplan-Meier survival analysis revealed that elevated expression of cytoplasmic CK1ε protein was not significantly associated with the overall survival of patients with melanoma. Background This study aimed to collect data regarding patient perception and knowledge of dental implants. It was conducted with the hope that the data would provide dental professionals and policymakers with a better understanding of ways to promote implant therapy. Methods An anonymous online survey with 10 questions was distributed through 12 dental offices in Tokyo and provincial cities in Japan to assess patient perception and knowledge of dental implants. Harvard Medical School’s IRB approved this study. Results We collected data from 1172 patients (59% female, 41% male). The most common perceptions of implant therapy were that it was “expensive,” “advanced,” and “scary”. Patients’ implant knowledge came primarily from magazines or books, while professional dental societies/associations were the least sought out source of information. Patients believed that the purpose of dental implants was to avoid dentures and improve chewing function. Their primary concerns about dental implants were the cost and longevity. Approximately 12% of patients with dental implants and 61% of patients without implants did not know that bone grafts may be required and that sedation during surgery was an option. For patients who experienced sedation during the procedure, 60% of them want it for future surgeries. Patients also had limited knowledge of bone-graft materials and the effects of CBCT radiation; 75% of the patients expressed concerns over the safety of graft materials and radiation exposure. For patients with a history of dental implant therapy, 80% of them would recommend dental implants to their family and friends. Conclusions Overall, patients’ experiences with dental implant therapy were positive, but there was a lack of patient education regarding dental implants and their associated procedures. Dental professionals need to take the initiative to improve patient education.

Endozoicomonas, a core bacterial group in corals, may also be a coral symbiont. Endozoicomonas communities often decrease rapidly in corals under heat stress. However, how the bacteria respond to changes in temperature and coral host during heat stress is unknown. Here, we employed the cultivable, dominant species E. montiporae as a working organism to explore how Endozoicomonas responds to heat stress. We designed two experiments to clarify the extent to which E. montiporae is influenced by temperature and coral host. We detected differentially expressed protein (DEP) profiles in this bacterium at 31°C and 33°C compared to 25°C by tandem mass tags-based quantitative proteome analysis. Fifty DEPs, including many heat shock proteins, were detected when the temperature changed. The expression of antioxidant defense proteins and key pyruvate synthase proteins decreased, suggesting that E. montiporae were in a physiology of stress at 33°C. Furthermore, some proteins were differentially expressed because of the heat-stress-treated coral lysate specifically, suggesting that not only heat but also heat-induced host factors can affect the protein expression of the bacterium. This study provides an in-depth analysis of how the molecular mechanisms of Endozoicomonas are affected by heat stress and coral host.

We investigate the impact of aggressive low-precision representations of weights and activations in two families of large LSTM-based architectures for Automatic Speech Recognition (ASR): hybrid Deep Bidirectional LSTM - Hidden Markov Models (DBLSTM-HMMs) and Recurrent Neural Network - Transducers (RNN-Ts). Using a 4-bit integer representation, a na\"ive quantization approach applied to the LSTM portion of these models results in significant Word Error Rate (WER) degradation. On the other hand, we show that minimal accuracy loss is achievable with an appropriate choice of quantizers and initializations. In particular, we customize quantization schemes depending on the local properties of the network, improving recognition performance while limiting computational time. We demonstrate our solution on the Switchboard (SWB) and CallHome (CH) test sets of the NIST Hub5-2000 evaluation. The growing prevalence and computational demands of Artificial Intelligence (AI) workloads has led to widespread use of hardware accelerators in their execution. Scaling the performance of AI accelerators across generations is pivotal to their success in commercial deployments. The intrinsic error-resilient nature of AI workloads present a unique opportunity for performance/energy improvement through precision scaling. Motivated by the recent algorithmic advances in precision scaling for inference and training, we designed RaPiD1, a 4-core AI accelerator chip supporting a spectrum of precisions, namely, 16 and 8-bit floating-point and 4 and 2-bit fixed-point. The 36mm2 RaPiD chip fabricated in 7nm EUV technology delivers a peak 3.5 TFLOPS/W in HFP8 mode and 16.5 TOPS/W in INT4 mode at nominal voltage. Using a performance model calibrated to within 1% of the measurement results, we evaluated DNN inference using 4-bit fixed-point representation for a 4-core 1 RaPiD chip system and DNN training using 8-bit floating point representation for a 768 TFLOPs AI system comprising 4 32-core RaPiD chips. Our results show INT4 inference for batch size of 1 achieves 3 - 13.5 (average 7) TOPS/W and FP8 training for a mini-batch of 512 achieves a sustained 102 - 588 (average 203) TFLOPS across a wide range of applications.

How to accurate predict the topology of a constituted wafer and its warpage would be critical in improving processing reliabilities. Traditionally, Stoney equation has been widely used to correlate film stress and wafer warpage. However, it only works under ideal situation and could be deviated from real situation significantly. Many previous studies thus have been performed to revise the relation but these analytical-based formulations usually take single factor into consideration. In reality, multiple imperfection issues are usually simultaneously existed and pure analytical approach would be too challenging to yield useful results. Instead, data-driven methods such as artificial neural network might be feasible to achieve effective black box mapping to evaluate the problem. Specifically, the stress state of bi-layer structures with thicker, viscoelastic, and multi-layer films are investigated in this work to demonstrate the feasibility. The multilayer perception model is chosen and the effects of thick film, viscoelasticity, and multiple layers on film stress are individually investigated subsequently. Finally, all three factors are simultaneously considered under the same MLP structure and a 99% successful rate can be achieved based on a 5% deviation threshold with 2300 simulation data. Meanwhile, a program is designed to reconstruct and visualize the deformed wafer surface from local curvatures as the preparation for final real 3D reconstitute structure study in the future.

Objectives.The objective of this retrospective clinical study was to assess the accuracy of a method of predicting post-movement root position during orthodontic treatment, using a 3D digital crown/root model (3DCRM), which was created with pre-movement records of both CBCT and dental-arch digital scans.Material & Methods.Pre-movement CBCT scans, post-movement CBCT scans, and dental-arch digital scans of five patients who had completed orthodontic treatments were used in this study with the approval of internal review board of Iwate Medical University. The 3DCRM was superimposed onto post-movement scanned dental-arch to identify the post-movement root position (test method). Post-movement CBCT (referenced as current method) served as the control used to identify the actual post-movement root position. The predicted root positions by the two methods were compared using color displacement mapping and 3D coordinate XYZ analysis.Results.Color displacement mapping showed that the average root displacement of five cases was -0.16 mm ±0.05 mm. 3D coordinate analysis revealed no significant differences between test and current methods in X and Y axis. However, the discrepancy in Z axis (especially in cases of intrusion) was greater than all other directions (mesial, distal, labial, palatal and extrusion direction) for all three tooth types examined (central incisor, lateral incisor and canine, p

Sheet metal-based products serve as a major portion of the furniture market and maintain higher quality standards by being competitive. During industrial processes, while converting a sheet metal to an end product, new defects are observed and thus need to be identified carefully. Recent studies have shown scratches, bumps, and pollution/dust are identified, but orange peel defects present overall a new challenge. So our model identifies scratches, bumps, and dust by using computer vision algorithms, whereas orange peel defect detection with deep learning have a better performance. The goal of this paper was to resolve artificial intelligence (AI) as an AI landing challenge faced in identifying various kinds of sheet metal-based product defects by ALDB-DL process automation. Therefore, our system model consists of multiple cameras from two different angles to capture the defects of the sheet metal-based drawer box. The aim of this paper was to solve multiple defects detection as design and implementation of Industrial process integration with AI by Automated Optical Inspection (AOI) for sheet metal-based drawer box defect detection, stated as AI Landing for sheet metal-based Drawer Box defect detection using Deep Learning (ALDB-DL). Therefore, the scope was given as achieving higher accuracy using multi-camera-based image feature extraction using computer vision and deep learning algorithm for defect classification in AOI. We used SHapley Additive exPlanations (SHAP) values for pre-processing, LeNet with a (1 × 1) convolution filter, and a Global Average Pooling (GAP) Convolutional Neural Network (CNN) algorithm to achieve the best results. It has applications for sheet metal-based product industries with improvised quality control for edge and surface detection. The results were competitive as the precision, recall, and area under the curve were 1.00, 0.99, and 0.98, respectively. Successively, the discussion section presents a detailed insight view about the industrial functioning with ALDB-DL experience sharing.

Background: Antinuclear antibody pattern recognition is vital for autoimmune disease diagnosis but labor-intensive for manual interpretation. To develop an automated pattern recognition system, we established machine learning models based on the International Consensus on Antinuclear Antibody Patterns (ICAP) at a competent level, mixed patterns recognition, and evaluated their consistency with human reading. Methods: 51,694 human epithelial cells (HEp-2) cell images with patterns assigned by experienced medical technologists collected in a medical center were used to train six machine learning algorithms and were compared by their performance. Next, we choose the best performing model to test the consistency with five experienced readers and two beginners. Results: The mean F1 score in each classification of the best performing model was 0.86 evaluated by Testing Data 1. For the inter-observer agreement test on Testing Data 2, the average agreement was 0.849 (κ) among five experienced readers, 0.844 between the best performing model and experienced readers, 0.528 between experienced readers and beginners. The results indicate that the proposed model outperformed beginners and achieved an excellent agreement with experienced readers. Conclusions: This study demonstrated that the developed model could reach an excellent agreement with experienced human readers using machine learning methods.

This survey paper summarizes recent progress of deep learning circuits and systems technologies and contains four topics: hardware-centric deep learning algorithms, digital architectures, analog architectures, and system demonstrations. We present an overview of these four areas and introduce key contributions of papers in this special issue.

Sulfuric-acid neutralization by motor oils in the presence of ethanol was investigated for the first time by quantitatively visualizing the fate of acid drops in five off-the-shelf commercial motor oils via capillary video-microscopy. For all the oils at room temperature, there was neutralization when ethanol was absent, whereas little neutralization was exhibited when ethanol was present. A very different behavior was exhibited at 100 °C, with rate of acid neutralization being much faster in the presence of ethanol than in absence. With 1% ethanol and temperature of 100 °C, acid droplets were neutralized in 2 min, whereas at room temperature droplets expanded ∼35% in diameter after 60 min with little neutralization. The reported phenomena may affect engine behavior and tribology.

A model liquid hydrocarbon (n-hexadecane) was used to gain insight on the mobilization of non-aqueous phase liquids (NAPLs) trapped in porous formations, important for both enhanced oil recovery (EOR) and oil spill remediation in porous media. Food-grade surfactants lecithin from soy and tween 80 were compared to commercial dispersant Corexit 9500A, for their ability to mobilize the hydrocarbon originally trapped in a water-filled cryolite porous medium. Red dye was added to the n-Hexadecane to improve visualization, and the aqueous phase incorporated different ratios of the surfactants Lecithin and Tween 80 to seek synergistic benefits when the two surfactants are combined. Visual-microscopic flooding experiments carried on a miniature packed bed produced images which were then analyzed using grayscale image analysis. It was determined that a ratio 40:60 by weight of Lecithin to Tween 80 has the best performance in the mobilization of n-hexadecane from the porous media. Furthermore, any (non-optimal) mixture of lecithin and tween 80 exhibited a better (synergistic) performance in n-Hexadecane recovery from the porous media than when applying either surfactant separately.

This letter presents a multi-TOPS AI accelerator core for deep learning training and inference. With a programmable architecture and custom ISA, this engine achieves >90% sustained utilization across the range of neural network topologies by employing a dataflow architecture to provide high throughput and an on-chip scratchpad hierarchy to meet the bandwidth demands of the compute units. A custom 16b floating point (fp16) representation with 1 sign bit, 6 exponent bits, and 9 mantissa bits has also been developed for high model accuracy in training and inference as well as 1b/2b (binary/ternary) integer for aggressive inference performance. At 1.5 GHz, the AI core prototype achieves 1.5 TFLOPS fp16, 12 TOPS ternary, or 24 TOPS binary peak performance in 14-nm CMOS.

Preterm labor is associated with inflammation and infection. The mechanisms underlying the role of omega-3 fatty acid in inflammasome activation and prevention of preterm labor remain unknown. We hypothesized that omega-3 fatty acid can reduce the rate of preterm birth induced by infection and trophoblast inflammation. In the present study, we found that inflammasome-related molecules and IL-1β in trophoblasts were activated by TNF-α derived from lipopolysaccharide (LPS)-stimulated THP-1 cell-conditioned medium (CM) and recombinant TNF-α protein. The results demonstrated that stimulation with TNF-α caused lysosomal rupture in trophoblasts, which accelerated cathepsin S (CTSS) diffusion from lysosomes to the cytosol and activated NLRP1 (nacht domain-leucine-rich repeat, and pyd-containing protein 1) and absent in melanoma 2 (AIM2) inflammasomes, thereby increasing IL-1β secretion. Moreover, in response to LPS challenge, TNF-α increased trophoblast cell death and decreased cell viability through inflammasome and CTSS activation. Stearidonic acid (SDA; 18:4n−3) and docosahexaenoic acid (DHA; 22:6n−3) inhibited inflammasome-related molecule synthesis and CTSS and caspase-1 activation, which further reduced the preterm delivery rate of pregnant mice induced by LPS (92.9 compared with 69.7% (DHA); 92.9 compared with 53.5% (SDA)). Higher expression of TNF-α, IL-1β, prostaglandin E2, and CTSS, but lower resolvin D1 expression, was observed in preterm pregnant mice than in controls. Similarly, resolvin D1 was highly expressed in women with term delivery compared with women with preterm delivery. Thus, SDA and DHA may attenuate macrophage-derived TNF-α inducing CTSS and inflammasome activation, IL-1β secretion, and placental trophoblast cell death. These functions are implicated in the preventive effects of SDA and DHA on preterm labor.

This work highlights the fabrication of nickel‑copper/titanium nitride (Ni-Cu/TiN) for the use in direct methanol fuel cells. Ni-Cu nanoparticles are coated on the surface of TiN film by electrochemical methods. The formation of Ni-Cu/TiN is confirmed by a variety of means such as field-emission scanning electron microscopy and atomic force microscopy. The presence of Ni-Cu on TiN film is further confirmed by cyclic voltammetry. For the electrochemical oxidation of methanol, the electrocatalytic activity of the prepared Ni-Cu/TiN is higher than those of Cu/TiN, Ni/TiN, Ni-Mn/TiN, and Ni-Co/TiN. The stability for the electrochemical oxidation of methanol at Ni-Cu/TiN is better than that of Ni-Cu/glassy carbon electrode. The proposed Ni-Cu/TiN might be beneficial to develop high performance and low manufacturing cost of direct methanol fuel cells.

Low-precision computation is the key enabling factor to achieve high compute densities (T0PS/W and T0PS/mm2) in AI hardware accelerators across cloud and edge platforms. However, robust deep learning (DL) model accuracy equivalent to high-precision computation must be maintained. Improvements in bandwidth, architecture, and power management are also required to harness the benefit of reduced precision by feeding and supporting more parallel engines to achieve high sustained utilization and optimize performance within a given product power envelope. In this work, we present a 4-core AI chip in 7nm EUV technology that exploits cutting-edge algorithmic advances for iso-accurate models in low-precision training and inference [1, 2] and aggressive circuit/architecture optimization to achieve leading-edge power-performance. The chip supports fp16 (DLFIoat16 [8]) and hybrid-fp8(hfp8) [1] formats for training and inference of DL models, as well as int4 and int2 formats for highly scaled inference.

This work proposes a multi-criteria artificial bee colony (MABC) algorithm to optimize the energy consumption problem in wireless sensor networks. The approach uses the artificial bee colony (ABC) algorithm to discover sensor nodes in a network as a cluster header combination. Different nodes are dynamically selected according to their current status in the network. The purpose is to cluster sensor nodes in the network in such a way that nodes can transmit packets to their cluster header, and then identify the most energy efficient packet routing from the cluster headers to the Internet of Things (IoT) base station. The routing strategy takes into account nodes’ residual energy and energy consumption, routing distance, number of hops, and frequency, in order to assign decision scores to help the algorithm discover a better solution. The use case shows that the MABC algorithm provides energy-efficient packet routing, and thus extends the wireless sensor network lifespan, which is confirmed by the multi-criteria analysis evaluation of the candidate routing. The contribution of this research is its use of swarm intelligence algorithms in wireless sensor network routing, with a multi-criteria artificial bee colony algorithm used in a wireless sensor network to address the problem of fast convergence of the algorithm.

The goal of this multicenter randomized controlled study was to evaluate the effectiveness of a newly developed ionic-sonic electric toothbrush in terms of plaque removal and reduction of gingival inflammation. A total of 78 subjects from three dental centers were invited to join the study. They were randomized to receive either a manual toothbrush (control group) or an ionic-sonic electric brush (test group). Full-mouth prophylaxis and oral hygiene instructions based on the stationary bristle technique were provided 1 week prior to the baseline visit. At baseline and at each follow-up appointment, Plaque Index (PI) and Gingival Index (GI) were recorded. In addition, probing depth (PD) and bleeding on probing were recorded at baseline and at the last appointment (week 5). At completion of the study, subjects in the test group were given a questionnaire regarding their satisfaction with the toothbrush. Sixty-four subjects completed the study (control: 28; test: 36). The mean age of the subjects was 36.90 ± 12.19 years. No significant difference between the baseline and 5-week PD was found. Plaque removal efficacy and reduction in gingival inflammation were more significant for the test group at week 2. Both the control and test groups showed statistically significant improvement in PI and GI from baseline to week 5. The ionic-sonic toothbrush was more effective than manual toothbrush after a 1-week application.

In vitro evaluations are essential to gaining a better understanding of re-osseointegration, while reducing animal use and the overall costs of peri-implantitis studies. This pilot study evaluated preosteoblast migration from 3-D-printed scaffolds to decontaminated titanium microimplants, creating a system that tries to mimic the bone–implant interface. Smooth (S) and minimally rough (R) titanium microimplants were incubated in Escherichia coli cultures and divided into six groups according to the decontamination protocol applied: EDTA gel (EDTA); chlorhexidine (CHL); chlorhexidine-soaked gauze (GCHL); scaling (SC); titanium brush (TiB); and implantoplasty (IP). Pristine S and R microimplants were used as the controls (C). After the decontamination procedures, the microimplants were inserted in 3-D-printed polyurethane-based scaffolds previously inoculated with preosteoblast cell cultures. Cellular migration was assessed after 24, 72 and 120 hours by ATP quantification. At the 120-hour time point, there was no statistically significant difference between S-C, S-EDTA, S-CHL, S-GCHL and S-SC ( p > 0.05), and between R-C, R-EDTA and R-GCHL ( p > 0.05). The in vitro model developed in this pilot study successfully demonstrated cell migration on the different decontaminated surfaces. This methodology suggests that on smooth microimplants, EDTA, GCHL, SC and TiB decontamination may have a reduced impact on preosteoblast migration, while on minimally rough microimplants, EDTA and GCHL decontamination affected cell migration the least. However, when selecting a decontamination protocol, the effectiveness of the decontamination per se must also be considered.

The aim of this study was to evaluate the biological efficacy of a unique perpendicular protrusion of type-I collagen (Col-I) from TiO2 nanotubes (NT-EPF surface). We hypothesized that the NT-EPF surface would play bifunctional roles in stimulating platelet-mediated fibroblast recruitment and anchoring fibroblast-derived Col-I to form a perpendicular collagen assembly, mimicking the connective tissue attachment around natural teeth for the long-term maintenance of dental implants. Ti surface modification was accomplished in two steps. First, TiO2 nanotubes (NT) array was fabricated via anodization. Diameters and depths of NTs were controlled by applied voltage and duration. Subsequently, an electrophoretic fusion (EPF) method was applied to fuse Col-I into nanotube arrays in a perpendicular fashion. Surface wettability was assessed by contact angle measurement. The bioactivity of modified TiO2 surfaces was evaluated in terms of NIH3T3 fibroblast attachment, platelet activation, and collagen extension. Early attachment, aggregation, and activation of platelets as well as release of platelet-related growth factors were demonstrated on NT-EPF surfaces. Platelet-mediated NIH3T3 cells migration toward NT-EPF was significantly increased and the attached cells showed a typical fibrous morphology with elongated spindle shape. A direct linkage between pseudopod-like processes of fibroblasts to NT-EPF surfaces was observed. Furthermore, the engineered EPF collagen protrusion linked with cell-derived collagen in a perpendicular fashion. Within the limitation of this in vitro study, the TiO2 nanotube with perpendicular Col-I surface (NT-EPF) promoted better cell attachment, induced a strong platelet activation which suggested the ability to create a more robust soft tissue seal.

The goal of the present study was to evaluate human histologic healing of dental implants with a unique triangular neck design that is narrower than the implant body. Four patients in need of full-mouth reconstruction were recruited and received several implants to support a full-arch prosthesis. In each patient, two additional customized reduced-diameter implants were placed, designated to be harvested after 6 months of submerged healing. The eight harvested implants were all placed in healed edentulous maxillary or mandibular ridges. These implants were O 3.5 × 8 mm in size, and the final osteotomy drill allowed for the creation of a gap up to 0.2 mm in size between the coronal aspect of the triangular implant neck and the surrounding bone. At the end of the healing period, the implants were retrieved with the surrounding bone. Microcomputed tomography (μCT) was performed before processing the biopsy samples for undecalcified histologic exampination. Bone-to-implant contact (BIC) was measured from the μCT data and from buccolingual/buccopalatal and mesiodistal central histologic sections. All implant gaps were filled by mature remodeled bone. The mean BICs of the BL/BP and MD sections were 64.45% ± 6.86% and 65.39% ± 10.44%, respectively, with no statistically significant difference. The mean 360-degree 3D BIC measured all over the implant surface was 68.58% ± 3.76%. The difference between the BIC measured on the μCT and on the histologic sections was not statistically significant. The positive histologic results of the study confirmed the efficacy of this uniquely designed dental implant.

Introduction: The gut microbiota is suspected to play a role in hypertension and hypertensive end organ damage. In the present study, we used germ-free mice to demonstrate that microbial colonization modulates the response to a hypertensive stimulus. Methods: Four-week-old male germ-free C57BL6/J littermates were randomized to remain germ-free (GF) or to receive microbiota transfer from SPF donor mice to achieve full colonization status (COL). At 12 weeks, Angiotensin (Ang) II was infused s.c. for 14 days (1.44mg/kg/d, osmotic minipumps) and 1% NaCl added to the drinking water; sham-treated mice served as control. After 14 days of AngII we assessed inflammation and organ damage. Results: Fecal bacterial load in COL mice was similar to SPF donor mice (qPCR). Shotgun metagenomic sequencing of fecal samples revealed hypertension-induced alterations in microbiome composition confirming previous reports. Serum metabolome analysis ( Biocrates MxP Quant 500) confirmed the absence of microbiota-dependent metabolites in GF. Interestingly, microbiota-dependent metabolites relevant for cardiovascular risk (TMAO, indoxyl sulfate) were elevated in hypertensive COL mice compared to sham-treated. Hypertensive kidney damage was aggravated in GF mice. However, marker genes for tubular damage ( Lcn2 ), inflammation ( Ccl2 ), and fibrosis ( Col1a3 ) showed a stronger increase in GF mice (fold changes [fc] COL vs. GF: 7.5 vs 11.0, 1.2 vs 3.3, 1.3 vs 2.2, respectively). Albuminuria (fc 2 vs 25) and histology for kidney fibrosis (fc 1.1 vs 1.4) confirmed the aggravated kidney damage in GF mice. Similarly, we observed an aggravated cardiac damage in GF mice. Flow cytometry of splenic lymphocytes showed that the adaptive immune response to AngII + 1% NaCl, as evidenced by Th17 (fc 1.4 vs 2) and CD8+ central memory cells, was intensified in GF mice. In vitro , naïve T cells isolated from GF mice more readily polarized into Th17 (26 ± 5%) compared to T cells from SPF mice (19 ± 1%). Conclusion: The bacterial colonization status has potent effects on the phenotypic response to a hypertensive stimulus, evident to varying degrees in hearts and kidneys. The inflammatory response and the end organ damage in GF compared to COL mice demonstrates the importance of the gut microbiota in hypertension.

This cross-sectional study aimed to examine the oral hygiene behaviors in the general population and identify factors affecting oral hygiene behaviors and plaque removal efficacy. A survey was distributed to patients through 11 dental practices in Japan, and each patient’s plaque index score (PIS) was recorded. In total, 1184 patients participated (521 women and 660 men), with 84.04% using manual toothbrushes (MTBs) and 15.96% using electric toothbrushes (ETBs). ETB users had a significantly lower PIS compared to MTB users (p = 0.0017). In addition, a statistically significant difference in the PIS was detected in relation to the frequency of brushing per day (≥2 times) and time spent on brushing (≥1 min). Some MTB users spent less than 1 min brushing, while all ETB users spent at least 1 min brushing, and extended brushing periods significantly improved the PIS for the MTB users. MTB users tend to replace brush heads more frequently than ETB users, and the frequency of replacement affected the PIS significantly (p < 0.01) for the MTB users. The status of dental treatment (first visit, in treatment versus recall) also significantly affected the PIS (p < 0.01). The ETB was more effective than the MTB in terms of better plaque removal and reduced frequency of brush head replacement.

This paper presents the fabrication of polyaniline and graphene oxide aerogel composite (PGA) thin film-based sensors for H2S gas sensing application. The optical and morphological properties of these composite films were characterised correspondingly by UV–visible spectroscopy and scanning electron microscopy. To prepare the gas sensors, thin films of PANI and PGA composites were deposited onto interdigitated electrodes by spin coating method. The response of these sensors towards H2S gas was evaluated by monitoring the change in electrical conductivity at room temperature under two relative humidity values of 60% RH and 80% RH. The PGA composite thin film-based gas sensors showed a higher response compared with the pure PANI-based sensor. The gas sensor made of PGA-3 showed a good response of 3.87 in 67 s (60% RH), which is 4.6 times higher than that of pure PANI and 2.5 times higher than that of PGA-1 at 1 ppm. The first-principles simulations were carried out to investigate the H2S sensing mechanism of PGA heterostructure. The adsorption of H2S at different locations of pure PANI and PGA composites was analysed. The band gap for PGA showed more significant change after adsorption than that of pure PANI, which indicated the higher sensitivity for detecting H2S.

The use of zirconia for implants and abutments has become more prevalent in implant dentistry as an alternative to the commonly used titanium implants, and peri-implant disease can still affect them. The erbium, chromium-doped:yttrium-scandium-gallium-garnet (Er, Cr:YSGG) laser has emerged as a promising treatment modality. The purposes of this in vitro study were to (1) determine the effects of the laser on the surface roughness of zirconia discs; (2) determine the extent of removal of a single species biofilm, E. coli, on the zirconia discs after applying the laser; (3) determine the amount of cell adhesion and proliferation utilizing fibroblasts on zirconia discs after treatment with the laser. All treatments will be compared with the commonly used ultrasonic instrumentation and hand scalers. For the first aim, gross examination revealed noticeable surface damage on the discs when using ultrasonic and scalers but not for the laser group. For surface roughness, the mean roughness was Pa= 0.623±0.185 μm, 0.762±0.421 μm, 0.740±0.214 μm, and 0.724±0.168 μm for control discs, and discs treated with either the Er,Cr:YSGG laser, ultrasonic instrumentation, and hand scalers respectively. There was no statistical significance among the groups (p=0.628). For bacteria decontamination, there was a statistical significance among the groups (p0.0001). Statistical significance was seen between the control group and each of the three treatment groups, favoring the treatment groups (p0.0001). Statistical significance was seen when comparing ultrasonic instrumentation and hand scalers (p= 0.000) as well as when comparing the Er,Cr:YSGG laser to hand scalers (p= 0.007), favoring both the ultrasonic instrumentation and Er,Cr:YSGG laser. No significance between the Er,Cr:YSGG laser group and the ultrasonic instrumentation group was noted (p =0.374). When comparing the cell attachment following treatment in each of the three groups and also without treatment (control), there was a statistical significance among the groups (p0.0001) in terms of total cell count, favoring the control and the laser groups. Further evaluations with SEM showed differences in cell morphology indicating more adherent cells on Er,Cr:YSGG laser-treated surfaces. In conclusion, gross examination of the discs show clear surface changes when using ultrasonic instrumentation and hand scalers compared to the Er,Cr:YSGG laser group. The Er,Cr:YSGG laser was able to effectively ablate bacteria from zirconia disc. Fibroblast attachment on the surfaces of the zirconia discs shows more adherence when treated with Er,Cr:YSGG laser.

A processor core is presented for AI training and inference products. Leading-edge compute efficiency is achieved for robust fp16 training via efficient heterogeneous 2-D systolic array-SIMD compute engines leveraging compact DLFloat16 FPUs. Architectural flexibility is maintained for very high compute utilization across neural network topologies. A modular dual-corelet architecture with a shared scratchpad and a software-controlled network/memory interface enables scalability to many-core SoCs and large-scale systems. The 14nm AI core achieves fp16 peak performance of 3.0 TFLOPS at 0.62V and 1.4 TFLOPS/W at 0.54V.

Slit proteins have been reported to act as axonal repellents in Drosophila; however, their role in the placental microenvironment has not been explored. In this study, we found that human placental multipotent mesenchymal stromal cells (hPMSCs) constitutively express Slit2. Therefore, we hypothesized that Slit2 expressed by hPMSCs could be involved in macrophage migration during placental inflammation through membrane cognate Roundabout (Robo) receptor signaling. In order to develop a preclinical in vitro mouse model of hPMSCs in treatment of perinatal infection, RAW 264.7 cells were used in this study. Slit2 interacted with Robo4 that was highly expressed in RAW 264.7 macrophages: their interaction increased the adhesive ability of RAW 264.7 cells and inhibited migration. Lipopolysaccharide (LPS)-induced CD11bCD18 expression could be inhibited by Slit2 and by hPMSC-conditioned medium (CM). LPS-induced activation of p38 and Rap1 was also attenuated by Slit2 and by hPMSC-CM. Noticeably, these inhibitory effects of hPMSC-CM decreased after depletion of Slit2 from the CM. Furthermore, we found that p38 siRNA inhibited LPS-induced Rap1 expression in RAW 264.7 cells, indicating that Rap1 functions downstream of p38 signaling. p38 siRNA increased cell adhesion and inhibited migration through reducing LPS-stimulated CD11bCD18 expression in RAW 264.7 cells. Thus, hPMSC-derived Slit2 may inhibit LPS-induced CD11bCD18 expression to decrease cell migration and increase adhesion through modulating the activity and motility of inflammatory macrophages in placenta. This may represent a novel mechanism for LPS-induced placental infection.
