Granulocyte-colony stimulating factor (G-CSF) serves as an important cytokine in haematopoiesis; released at both physiological and pathological conditions by a range of cells. We hypothesized that the systemic administration of G-CSF would produce an accelerated fracture-healing rate in non-union bone defects; thus, potentially leading to useful clinical applications. Ten male adult Katjang goats, weighing about 15-26 kilograms were randomly chosen and a tibial bone defect was induced in each animal. The defect was maintained by internal fixation with a titanium plate and reinforced by an external fiberglass cast. Post-operative radiographs were performed twice weekly and radiographic assessments were performed by evaluating the bridging and union measurements through a validated method. In the treatment group, the time for bridging and union exhibited statistically significant differences when compared with a control group. The outcomes of the present study establishing a notion that administration of G-CSF besides inducing haematopoiesis, promotes healing of fractures and non-union bone defects as well.

Branched glycerol dialkyl glycerol tetraethers (brGDGTs) are a family of bacterial lipids which have emerged over time as robust temperature and pH paleoproxies in continental settings. Nevertheless, it was previously shown that other parameters than temperature and pH, such as soil moisture, thermal regime or vegetation can also influence the relative distribution of brGDGTs in soils and peats. This can explain a large part of the residual scatter in the global brGDGT calibrations with mean annual air temperature (MAAT) and pH in these settings. Despite improvements in brGDGT analytical methods and development of refined models, the root-mean-square error (RMSE) associated with global calibrations between brGDGT distribution and MAAT in soils and peats remains high (∼5 °C). The aim of the present study was to develop a new global terrestrial brGDGT temperature calibration from a worldwide extended dataset (i.e. 775 soil and peat samples, i.e. 112 samples added to the previously available global calibration) using a machine learning algorithm. Statistical analyses highlighted five clusters with different effects of potential confounding factors in addition to MAAT on the relative abundances of brGDGTs. The results also revealed the limitations of using a single index and a simple linear regression model to capture the response of brGDGTs to temperature changes. A new improved calibration based on a random forest algorithm was thus proposed, the so-called random Forest Regression for PaleOMAAT using brGDGTs (FROG). This multi-factorial and non-parametric model allows to overcome the use of a single index, and to be more representative of the environmental complexity by taking into account the non-linear relationships between MAAT and the relative abundances of the individual brGDGTs. The FROG model represents a refined brGDGT temperature calibration (R2 = 0.8; RMSE = 4.01 °C) for soils and peats, more robust and accurate than previous global soil calibrations while being proposed on an extended dataset. This novel improved calibration was further applied and validated on two paleo archives covering the last 110 kyr and the Pliocene, respectively.

Hafnia is of interest in thermal and environmental barrier coatings, but little is known about its response to molten silicate attack. This article investigates that response using two model silicate melts, compares it with pure ZrO2 and examines the effect of YO1.5 additions. HfO2 was found to form HfSiO4 with acidic melts but undergoes grain boundary penetration in basic melts, which do not exhibit reactive crystallization. The latter can be exacerbated by microcracking resulting from the thermal expansion anisotropy of monoclinic HfO2. Y additions generally degrade the ability to form hafnon (and zircon), and exacerbate grain boundary penetration, especially in HfO2 where Y is present as a fluorite second phase. The fluorite controls grain growth in monoclinic HfO2 and suppresses microcracking, but dissolves faster, especially in basic melts. The results are presented in the context of the relevant thermodynamics and kinetics. The implications for coating applications are discussed.

Accurate measurements of absorption data are required for the development and validation of inversion algorithms for upcoming hyperspectral ocean color imaging sensors, such as the NASA Phytoplankton, Aerosol, Cloud, and ocean Ecosystem mission. This study aims to provide uncertainty estimates associated with leading approaches to measure hyperspectral absorption coefficients in complex coastal waters. Absorption spectra were collected at 12 different stations, all located in the Indian River Lagoon, Florida, USA, between 09 January 2017 and 13 January 2017. Measurements included spectral absorption coefficients in the visible range (400–700 nm) associated with dissolved, a CDOM, total particulate, a p, and total nonwater, a nw, fractions, and were made both in situ and from discrete samples. Discrete sample approaches included dual-beam spectrophotometer, liquid waveguide capillary cell, point-source integrating cavity absorption meter (PSICAM) for dissolved matter absorption samples, and quantitative filter technique ICAM measurements and the dual-beam spectrophotometer with center-mounted integrating sphere filter pad technique, while the Turner Designs ICAM, and WET Labs AC-s, and AC-9 instruments were used to determine absorption coefficients in situ. The Gershun approach, determining absorption from measurement of the irradiance quartet with respect to depth was also assessed in situ. Measurement uncertainties and relative accuracies were quantified for each of these approaches. Results showed generally strong agreements between different discrete sample methods, with average percent absolute error %δ abs < 7% for a CDOM and < 9% for a p. In situ approaches showed higher variability and reduced accuracy. For a nw, %δ abs deviation relative to PSICAM data was on average 12% to 20%. Results help identify remaining technological gaps and need for improvements in the different absorption measurement approaches.

SUMMARYAn underground nuclear explosion (UNE) couples mechanical energy into crustal rock, which propagates as seismic and acoustic waves. These different physical phenomena transport, by different pathways, to standoff detectors at varying distances. The transport pathways attenuate the original signal but in different ways. Enabled by correct statistical weighting, signal attenuation models can be used to combine these disparate sensor data to estimate the yield of an UNE. Contemporaneous statistical models, used in yield estimation, can be improved with an advanced partition of error for these physical signal propagation models. We present an advanced multivariate approach to error modelling of multiphenomenology physical signatures. In addition to measurement error, our error model represents physical model biases as random with a physics-based covariance structure. To illustrate this proposed framework, we demonstrate the estimation of explosion yield using openly available seismic and acoustic data from chemical single-point explosions.

The degradation of thermal barrier coatings (TBCs) by molten silicates (CMAS) represents a fundamental barrier to progress in gas turbine technology, requiring a mechanistic understanding of the problem to guide the development of improved coatings. This article investigates the dissolution of yttria-stabilized zirconia (7YSZ and 20YSZ) into two model silicate melts at 1300–1400 °C. The approach involves the 1D dissolution of YSZ into a semi-infinite melt, characterizing the dissolution rates of YSZ and the diffusion rates of Zr4+ and Y3+ therein. The assessed kinetics of YSZ dissolution and diffusion were then applied to modeling the same phenomena on TBC-relevant length scales. These findings provide fundamental insight into (i) the dissolution mechanism of YSZ, (ii) the subsequent reprecipitation upon saturation, (iii) the quantitative effects of temperature and melt composition on the dissolution and diffusion kinetics, and (iv) how the measured kinetics manifests on the scale of flow channels present in TBCs.
