High mortalities and highly variable results during the subsequent development of post-thaw larvae have been widely considered as key issues restricting the application of cryopreservation techniques to support genetic improvement programs and hatchery production in farmed marine bivalve species. To date, few studies have been undertaken to investigate the effects of cryodamage at the molecular level in bivalves. This study is the first to evaluate the effect of larval cryopreservation on the epigenetics of the resultant progenies of the Pacific oyster Crassostrea gigas. The results show that the level of DNA methylation was significantly (p < 0.05) higher and lower than that of the control when the trochophore larvae were revived and when they developed to D-stage larvae (day 1 post-fertilization), respectively, but the level returned to the control level from day 8 post-fertilization onwards. The expression of the epigenetic regulator genes DNMT3b, MeCP2, JmjCA, KDM2 and OSA changed significantly (p < 0.05) when the trochophore larvae were thawed, and then they reverted to the control levels at the D- and later larval developmental stages. However, the expression of other epigenetic regulator genes, namely, MBD2, DNMT1, CXXC1 and JmjD6, did not change at any post-thaw larval developmental stage. For the newly thawed trochophore larvae, the amount of methylated H3K4Me1 and H3K27Me1 significantly changed, and the expression of all Jumonji orthologs, except that of Jumonji5, significantly (p < 0.05) decreased. These epigenetic results agree with the data collected on larval performances (e.g., survival rate), suggesting that the effect period of the published cryopreservation technique on post-thaw larvae is short in C. gigas. Oysters have the potential to be a part of more sustainable farming systems, such as multitrophic systems integrated into biofloc systems, due to their filtration activity, which enables them to act as organic consumers. However, the stress experienced by animals in a system with a high organic load can compromise their productive performance. The objective of this study was to evaluate the biological responses of Crassostrea gasar oysters when exposed to different concentrations of total suspended solids in biofloc systems. The oysters were exposed to four different concentrations of solids for 28 days. Hall effect sensors were installed on the outside of the shells to detect the movement of the oyster valves. Also, biochemical and histological analyses were conducted to assess the biological responses of the oysters to exposure to varying levels of solids. A difference in valve opening detected by the Hall sensors was observed from the second week of culture, indicating a relationship between shell closure and higher concentrations of suspended solids present in the system. In terms of biochemical analysis, a significant increase in lipid damage was observed in treatments with medium and high levels of total suspended solids compared with the control group. Conversely, no changes were observed in the gill structure of the oysters caused by the concentrations of suspended solids in the system when compared with the control. According to the analyses of gill activity and biochemistry, it is suggested that C. gasar should be cultured with total suspended solids at less than 200 mg/L. Oysters cultivated in a biofloc system keep their shells closed when subjected to high concentrations of total suspended solids; concentrations of total suspended solids below 200 mg/L do not induce oxidative stress, changes in behavior or histological alterations in C. gasar oysters cultivated in a biofloc system. Coastal warming, acidification, and deoxygenation are progressing primarily due to the increase in anthropogenic CO 2. Coastal acidification has been reported to have effects that are anticipated to become more severe as acidification progresses, including inhibiting the formation of shells of calcifying organisms such as shellfish, which include Pacific oysters (Crassostrea gigas), one of the most important aquaculture resources in Japan. Moreover, there is concern regarding the combined impacts of coastal warming, acidification, and deoxygenation on Pacific oysters. However, spatiotemporal variations in acidification and deoxygenation indicators such as pH, the aragonite saturation state (Ωarag), and dissolved oxygen have not been observed and projected in oceanic Pacific oyster farms in Japan. To assess the present impacts and project future impacts of coastal warming, acidification, and deoxygenation on Pacific oysters, we performed continuous in situ monitoring, numerical modeling, and microscopic examination of Pacific oyster larvae in the Hinase area of Okayama Prefecture and Shizugawa Bay in Miyagi Prefecture, Japan, both of which are famous for their Pacific oyster farms. Our monitoring results first found Ωarag values lower than the critical level of acidification for Pacific oyster larvae in Hinase, although no impact of acidification on larvae was identified by microscopic examination. Our modeling results suggest that Pacific oyster larvae are anticipated to be affected more seriously by the combined impacts of coastal warming and acidification, with lower pH and Ωarag values and a prolonged spawning period, which may shorten the oyster shipping period and lower the quality of oysters. Background: Apoptosis regulates normal development, homeostasis, immune tolerance and response to environmental stress by eliminating unwanted or diseased cells, and plays a key role in non-specific immunity of invertebrates. The exogenous pathway mediated by death receptors and death ligands is a very important pathway for cell apoptosis. Death ligands are mainly members of the tumour necrosis factor (TNF) family, of which FasL is an important member. The deep involvement of FasL in vertebrates cell apoptosis and immunity has been reported many times, but there is limited research on the FasL gene in shellfish, and its functional importance in oyster cell apoptosis and immunity remains unclear. Methods: The full length of ChFasL was identified and cloned based on the genome of Crassostrea hongkongensis. Quantitative PCR was used to detect the relative expression of ChFasL in different developmental stages and tissues, as well as the changes of relative expression in hemocytes after bacterial infection. The expression position of ChFasL in HEK293T cells was also located by subcellular localization, and the effect of increased recombinant protein content on the activity of reporter genes p53 and p21 was studied by dualfluorescence reporter gene. Finally, the changes of apoptosis rate in hemocytes after ChFasL silencing was identified by RNA interference technology. Results: We identified a novel FasL gene from C. hongkongensis and named it ChFasL. We found that ChFasL has potential N-linked glycosylation site, a transmembrane domain and a TNF region, which was a typical characteristics of TNF family. ChFasL was expressed in all developmental stages of larvae and in all tissues of oysters. After stimulation by V. alginolyticus or S. haemolyticus, its relative expression in hemocytes increased significantly, suggesting that ChFasL was deeply engaged in the immune response process of C. hongkongensis to external microbial stimulation. The results of subcellular localization showed that ChFasL was mainly distributed in the cytoplasm of HEK293T cells. With the overexpression of the recombinant protein pcDNA3 1- ChFasL, the activity of p53 and p21 significantly increased, showing a positive regulatory effect. Moreover, after dsRNA successfully reduced the relative expression of ChFasL, the apoptosis rate of hemocytes was significantly lower than that the dsGFP group. Conclusion: These results comprehensively confirmed the important role of ChFasL in the apoptosis process of C. hongkongensis, which provided the basis and premise for the in-depth understanding of the immune function of apoptosis in molluscs, and also contributed to the research on the pathogenic death mechanism and disease resistance breeding of marine bivalves. β-Galactosidases (EC 3.2.1.23) are exoglycosidases that catalyze the cleavage of glycoconjugates with terminal β-D-galactose residues in β1,3-, β1,4- or β1,6-linkage. Although this family of exoglycosidases has been extensively studied in vertebrates, plants, yeast, and bacteria, little information is available for mollusks. Mollusks are a diverse and highly successful group of animals that play many different roles in their ecosystems, including filter feeders and detritivores. Here, the first β-galactosidase from the Pacific oyster, Crassostrea gigas was discovered, biochemically characterized, and compared to our previously characterized slug enzyme from Arion vulgaris (UniProt Ref. Nr.: A0A0B7AQJ9). Overall, the mussel enzyme showed similar biochemical parameters to the snail enzyme. The enzyme from C. gigas was most active in an acidic environment (pH 3.5) and at a reaction temperature of 50 °C. Optimal storage conditions were up to 37 °C. In contrast to the enzyme from A. vulgaris, the supplementation of cations (Ni2+, Co2+, Mn2+, Mg2+, Ca2+, Cu2+, Ba2+) increased the activity of the enzyme from C. gigas. Substrate specificity studies of the β-galactosidases from the mussel, C. gigas, and the slug, A. vulgaris, revealed activity towards terminal β1,3- and β1,4-linked galactose residues for both enzymes. Using the same substrates in labeled and unlabeled form, we were able to detect the effect of labeling on the β-galactosidase activity using MALDI-TOF MS, HPTLC, and HPLC. While lactose was cleaved by the enzymes in an unlabeled or labeled state, galacto-N-biose was not cleaved as soon as a 2-amino benzoic acid label was added. In this study we present the biochemical characterization of the first recombinantly expressed β-galactosidase from the Pacific oyster, C. gigas, and we compare different analytical methods for the determination of β-galactosidase activity using the enzyme from C. gigas and A. vulgaris. Parallel diversification provides a proper framework for studying the role of natural selection in evolution. Yet, empirical studies from ecological 'non‐model' species of invertebrates are limited at the whole genome level. Here, we presented a chromosome‐scale genome assembly for Crassostrea angulata and investigated the parallel genomic evolution in oysters. Specifically, we used population genomics approaches to compare two southern–northern oyster species pairs (C. angulata–C. gigas and southern–northern C. ariakensis) along the coast of China. The estimated divergence time of C. angulata and C. gigas is earlier than that of southern and northern C. ariakensis, which aligns with the overall elevated genome‐wide divergence. However, the southern–northern C. ariakensis FST profile represented more extremely divergent "islands". Combined with recent reciprocal hybridization studies, we proposed that they are currently at an early stage of speciation. These two southern–northern oyster species pairs exhibited significant repeatability in patterns of genome‐wide differentiation, especially in genomic regions with extremely high and low divergence. This suggested that divergent and purifying selection has contributed to the genomic parallelism between southern and northern latitudes. Top differentiated genomic regions shared in these two oyster species pairs contained candidate genes enriched for functions in energy metabolism, especially adipogenesis, which are closely related to reproductive behaviours. These genes might be good candidates for further investigation in vivo. In conclusion, our results suggest that similar divergent selection and shared genomic features could predictably transform standing genetic variation within one species pair into differences in another. Summer mortality of Crassostrea gigas occurs in Brazilian crops, with predominance in the final phase of cultivation, when they reach the commercial size, generating significant losses for local producers. In order to mitigate this problem, a technical solution, whose principle is based on long-term confinement at low temperatures (10 −12°C) for inducing a similar basal metabolism regime, has been developed. According to the current literature, if the spawning process is not triggered, a substantial reduction in the mortality rate can be achieved. This work evaluates not only the effectiveness of this technological solution to reduce summer mortality but also the quality associated to commercial attributes of the oysters as a final product. A ninety-one day experiment has been carried out and has shown that the proposed solution is able to maintain Crassostrea gigas diploid oysters alive and healthy. [ABSTRACT FROM AUTHOR]

The Crassostrea gasar (C. gasar) are economically important bivalves. The study investigated the physico-chemistry, the morphometrics and microbial profiles of C. gasar, water, and sediment from the Lagos Lagoon. The physico-chemistry and morphometry of C. gasar were determined using standard methods while the microbial were identified by Vitek 2 automated system. The mean Total Heterotrophic Bacterial Counts (Log CFU/mL) of C. gasar flesh and shells ranged from 5.28 ± 0.3 to 5.41 ± 0.3 and 5.49 ± 0.1 to 5.62 ± 0.3 respectively. The mean total coliform counts of sediment ranged from 4.34 ± 0.2 to 4.39 ± 0.2 Log CFU/mL, while the highest mean faecal coliform counts and total fungal counts of water samples were 2.41 ± 0.3 and 2.41 ± 0.1 Log CFU/mL, respectively. Vibrio spp, Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus spp, Streptococcus faecalis, Escherichia coli and Aspergillus niger were the species found in all samples in varying percentages. The median-sized C. gasar had the highest values of 7.5 – 12.4 cm (TL); 4.5 – 7.4 cm (MW); and 80.5 – 160.4 g (TW). The highest mean values for surface water temperature, pH, salinity, Chemical Oxygen Demand and Phosphate were 27.3 ±1.51oC, 29.8 ± 1.94, 16.5 ± 0.39 ‰. 7.26 ±0.01 mg/L and 0.39 ± 0.01 mg/L respectively. This study revealed that C. gasar from the riparian swampy areas harboured some potential pathogenic organisms of significant public health concern and can contribute to the spread of illnesses when consumed. To achieve a non-destructive and rapid detection of oyster freshness, an intelligent method using deep learning fused with malondialdehyde (MDA) and total sulfhydryl groups (SH) information was proposed. In this study, an "MDA-SH-storage days" polynomial fitting model and oyster meat image dataset were first built. AleNet-MDA and AlxNet-SH classification models were then constructed to automatically identify and classify four levels of oyster meat images with overall accuracies of 92.72% and 94.06%, respectively. Next, the outputs of the two models were used as the inputs to "MDA-SH-storage days" model, which ultimately succeeded in predicting the corresponding MDA content, SH content and storage day for an oyster image within 0.03 ms. Furthermore, the interpretability of the two models for oyster meat image were also investigated by feature visualization and strongest activations techniques. The evaluation of oyster marinating (Crassostrea gasar) and pasteurization process in vacuum packaging during storage. The aim of this study was to establish better conditions for marinating and pasteurization for oysters (Crassostrea gasar) and to evaluate their shelf life. Microbiological, physicochemical and sensory analyzes were carried out on the oysters and on the optimized product. In the vacuum-packed product, under refrigeration, pH, water activity (aw), total volatile bases (N-TVB) and texture analyzes were performed, every 7 days for 35 days. Oysters had a high protein, mineral and amino acid content and a low lipid content. Planning 23 and 221 hour, 2.5% acetic acid, 1% NaCl and pasteurization was 62°C/2 min. Sensory analysis achieved acceptability above 80%. Marinated oysters remained suitable for consumption for 21 days, being an alternative to extend the shelf life and add value. In the "Blue Bay Remediation Action" project, the oyster reef plays the dual role of ecological restoration and wave suppression, and the study of the effect of oyster reef wave dissipation forms the basis of relevant projects. Taking the Binzhou oyster reef ecological restoration project as its object, this paper studied the wave-dissipating effect of the oyster reef, using the SWAN model. The simulation results showed that after the deployment of oyster reefs in extreme high-water-level conditions, they could produce a wave-dissipation effect, with a wave-dissipation rate between 8% and 21%. Significant wave-dissipation effects can be produced under the designed high-water-level conditions, with a wave-height dissipation rate of 42% to 65%. Among them, the wave-dissipation effect of oyster reefs was more significant for waves coming from the ENE, NE, N, and NNE directions. The regeneration of bone defects is a major challenge for clinical orthopaedics. Herein, we designed and prepared a new type of bioactive material, using stingray skin collagen and oyster shell powder (OSP) as raw materials. A stingray skin collagen/oyster osteoinductive composite scaffold (Col-OSP) was prepared for the first time by genipin cross-linking, pore-forming and freeze-drying methods. These scaffolds were characterized by ATR-FTIR, SEM, compression, swelling, cell proliferation, cell adhesion, alkaline phosphatase activity, alizarin red staining and RT-PCR etc. The Col-OSP scaffold had an interconnected three-dimensional porous structure, and the mechanical properties of the Col-OSP composite scaffold were enhanced compared with Col, combining with the appropriate swelling rate and degradation rate, the scaffold was more in line with the requirements of bone tissue engineering scaffolds. The Col-OSP scaffold was non-toxic, promoted the proliferation, adhesion, and differentiation of MC3T3-E1 cells, and stimulated the osteogenesis-related genes expressions of osteocalcin (OCN), collagen type I (COL-I) and RUNX2 of MC3T3-E1 cells. Predator–prey interactions are a key feature of ecosystems and often chemically mediated, whereby individuals detect molecules in their environment that inform whether they should attack or defend. These molecules are largely unidentified, and their discovery is important for determining their ecological role in complex trophic systems. Homarine and trigonelline are two previously identified blue crab (Callinectes sapidus) urinary metabolites that cause mud crabs (Panopeus herbstii) to seek refuge, but it was unknown whether these molecules influence other species within this oyster reef system. In the current study, homarine, trigonelline, and blue crab urine were tested on juvenile oysters (Crassostrea virginica) to ascertain if the same molecules known to alter mud crab behavior also affect juvenile oyster morphology, thus mediating interactions between a generalist predator, a mesopredator, and a basal prey species. Oyster juveniles strengthened their shells in response to blue crab urine and when exposed to homarine and trigonelline in combination, especially at higher concentrations. This study builds upon previous work to pinpoint specific molecules from a generalist predator's urine that induce defensive responses in two marine prey from different taxa and trophic levels, supporting the hypothesis that common fear molecules exist in ecological systems. Pacific oyster (Crassostrea gigas) aquaculture has been economically impacted in many countries by Pacific oyster mortality syndrome (POMS), a disease initiated by Ostreid herpesvirus 1. The objectives of this study were to determine whether naturally exposed, adult C. gigas could act as reservoirs for OsHV-1 and explain the recurrent seasonal outbreaks of POMS and to test whether or not they were resistant to OsHV-1. In a laboratory infection experiment using thermal shock, OsHV-1 replication was not reactivated within the tissues of such oysters and the virus was not transmitted to naïve cohabitating spat. The adult oysters were resistant to intramuscular injection with a lethal dose of OsHV-1 and had 118 times lower risk of mortality than naïve oysters. Considered together with the results of other studies in C. gigas, natural exposure or laboratory exposure to OsHV-1 may result in immunity during subsequent exposure events, either in the natural environment or the laboratory. While adult C. gigas can carry OsHV-1 infection for lengthy periods, reactivation of viral replication leading to mortality and transmission of the virus to naïve oysters may require specific conditions that were not present in the current experiment. Further investigation is required to evaluate the mechanisms responsible for resistance to disease in oysters previously exposed to OsHV-1, whether immunity can be exploited commercially to prevent POMS outbreaks and to determine the source of the virus for recurrent seasonal outbreaks. Two native species of Crassostrea cupped oysters, C. rhizophorae and C. gasar, are economically important in Brazil. However, C. gasar genetic resources are poorly characterized. This study assesses and characterizes genetic variation within and between oyster banks along the southeast coast of the state of São Paulo, Brazil, to provide baseline data useful for conservation and management. To correctly distinguish C. gasar from C. rhizophorae, we screened PCR–RFLP markers based on variation of the AluI restriction digest banding pattern. Genetic variation within C. gasar was assessed using a panel of nine microsatellite DNA markers. Intrapopulation analysis revealed low allelic richness (4.50 to 5.31 alleles per locus), deficit of heterozygotes, high rates of inbreeding, and segregation of null alleles in all populations. At one of the five sampling sites, a bottleneck effect was inferred in the only population that exhibited no private alleles. In silico analysis revealed four genetic groups using DAPC and DEST genetic differentiation index, showing moderate to high population genetic structure (DEST 0.05–0.34) with gene flow. The Mantel test did not show a significant correlation (P = 0.27) between geographic and genetic distances. The results indicate that the stocks of C. gasar have low genetic variability with moderate genetic structuring. The establishment of an exotic species (Saccostrea cucullata) may have reduced Crassostrea populations in the oyster banks studied, and loss of genetic diversity may be indicators that further study and timely action are needed to preserve native genetic resources of this essential marine food resource. Chama coralloides Reeve, 1846 is a commercially important mollusk in Acapulco, Mexico. There is currently no fisheries regulation or protection. Therefore, the objective of the present work was to estimate the growth parameters of C. coralloides using the Schnute model cases. Shell length data collected from March 2019 to December 2021 were used. A multinomial analysis was performed to determine the modal groups monthly. Using the mean lengths of each modal group, a modal progression was performed to define the time increments ∆t, which were taken as equivalent representations of the data from direct age estimation methods (mark-recapture). These data were used to estimate the individual growth parameters using the five cases of the Schnute model and to define the case that best describes the growth. 2804 organisms were analyzed, with lengths ranging from 7.61 to 116.30 mm SL. Multinomial analysis showed one to four modal groups per month and 14 cohorts were identified. The best-fit case was case 5 (wi = 0.72) with an L∞ = 114.2 mm, followed by case 1 (wi = 0.28) with an L∞= 109.8 mm. Both cases (5 and 1) were modeled and because the best model did not reach 90%, a mean model was estimated. C. coralloides presented a type of asymptotic growth, with an estimated L∞ that can be reached nearly 22 years. [ABSTRACT FROM AUTHOR]

Recently, three proton channels (HV) have been identified and characterized in Aplysia californica (AcHV1‐3). Focusing on AcHV1 and AcHV2, analysis of Transcriptome Shotgun Assembly and genomic databases of 91 molluscs identified HV homologous channels in other molluscs: channels homologous to AcHV1 and to AcHV2 were found in 90 species (56 full‐length sequences) and in 33 species (18 full‐length sequences), respectively. Here, we report the discovery of a fourth distinct proton channel family, HV4. This new family has high homology to AcHV1 and AcHV2 and was identified only in bivalvian molluscs (13 species, 12 full‐length sequences). Typically, these channels possess an extracellular S1–S2 loop of intermediate size (~ 20 amino acids) compared to the shorter loops of molluscan HV1 channels (~ 13 amino acids) and the much larger loops of molluscan HV2 channels (> 65 amino acids). The characteristic voltage‐sensor motif in S4 possesses only two arginine residues with the common third arginine being replaced by a lysine. Moreover, HV4 channels are much smaller with only around 200 amino acids in total length. The smallest functional channel found so far in nature (189 amino acids) is expressed in the pacific oyster Crassostrea gigas (CgHV4) and might be considered an archetypical minimal proton channel. Functional expression and electrophysiological characterization demonstrated that CgHV4 shares distinctive hallmarks of other investigated proton channels as high proton selectivity, slow activation, and pH‐ and voltage‐regulated gating. The identification of native mangrove oyster species of the genus Crassostrea in Brazil is a recurring issue for specialists. Studies have recognized the presence of two species in the littoral zone: C. rhizophorae and C. gasar. However, other names may be used, such as C. mangle for C. rhizophorae and C. brasiliana or C. tulipa for C.gasar. As in most oyster taxonomic studies, their identification requires genetic evaluation for the precise determination of taxa. Moreover, previous revisions have proposed some morphological traits by which each species could be precisely identified, leading to a series of debates about the best model for species identity. The aim of this study was to apply an integrative taxonomic approach to native Crassostrea mangrove oyster species on the Brazilian coast to assert the correct species names, delimitations, and concise descriptions. Molecular tracing of cultivated specimens, along with wild specimens of Crassostrea collected from the Brazilian coast, was used to confirm the morphological identity of the taxa. The wild specimens were tentatively identified as either C. gasar or C. rhizophorae based on previous morphological revisions. PCR-RFLP and DNA analyses of the mitochondrial 16S gene mitochondrial rDNA from wild specimens were used to confirm taxa and re-evaluate morphological boundaries and descriptions of the species. We confirmed two patterns in the RFLP and sequence analyses: C. gasar and C. rhizophorae. Conchological and anatomical evaluations of 1006 identified Crassostrea specimens confirmed the inherent polymorphism between the native species, preventing reliable identification of the species by morphology alone. A clear exception, however, is found for specimens whose shell height is greater than 110 mm with internal view of white-colored valves and without an umbonal cavity, fitting the description of C. paraibanensis , which belongs to C. gasar. C. brasiliana and C. mangle are synonyms of C. gasar and C. rhizophorae , respectively. C. tulipa represents a species disconnected from any described mangrove oyster species from Atlantic seas. Sponges, especially Calcarea, are minor components of benthic associations, especially during the Mesozoic. In the Lower Cretaceous of the Neuquén Basin, small calcareous sponges have been found building a small monospecific meadow. It is restricted to a marlstone lens-shaped bed in a quiet outer-ramp setting in the Cerro Marucho Locality (Picún Leufú depocentre), above a shell bed of small exogyrid oysters; oysters and sponges were the only preserved macrobenthic faunal elements. Individual sponges were small, under 4 cm high, and presented a sub-cylindrical morphology with one or more rounded, apical osculi, many inhalant openings and triactine spicules. Specimens studied here were assigned to Endostoma sp. aff. Endostoma nodosa. These sponges are quite commonly encrusted by exogyrid oysters, serpulids, sabellids, agglutinating foraminifers and cyclostome bryozoans. Overgrowths among sclerobionts were common, though no undoubtedly in vivo interaction has been recorded. Disarticulated left oyster valves were frequently bioclaustrated by the sponges, showing that in vivo settlement upon sponges was common. Many oysters settled in the periphery of the osculum suggesting a commensal relationship. The study of this sponge meadow and its sclerobiont community allowed the identification of different stages of ecological succession. The pioneer stage was characterised by sponge settlement on oyster valves, within an otherwise soft consistency bottom. High sedimentation or high nutrient inputs, either individually or in combination, could explain the great abundance of oysters. During the climax stage, sponges thrived and harboured several sclerobiont taxa, developing a relatively dynamic palaeocommunity. Finally, an intensification in either sedimentation rates or nutrient input (or both) past the tolerable threshold for sponges may have been the cause(s) of the meadow's demise. Endostoma and similar forms were up to now reported mostly from the Jurassic and Cretaceous of Europe as accessory builders, or as accompanying fauna in reefal settings. Massive mortality outbreaks affecting Pacific oysters (Crassostrea gigas) spat/juveniles are often associated with the detection of a herpesvirus called ostreid herpesvirus type 1 (OsHV-1). In this work, experimental infection trials of C. gigas spat with OsHV-1 were conducted using two contrasted Pacific oyster families for their susceptibility to viral infection. Live oysters were sampled at 12, 26, and 144 h post infection (hpi) to analyze host-pathogen interactions using comparative proteomics. Shotgun proteomics allowed the detection of seven viral proteins in infected oysters, some of them with potential immunomodulatoy functions. Viral proteins were mainly detected in susceptible oysters sampled at 26 hpi, which correlates with the mortality and viral load observed in this oyster family. Concerning the Pacific oyster proteome, more than 3,000 proteins were identified and contrasted proteomic responses were observed between infected A- and P-oysters, sampled at different post-injection times. Gene ontology (GO) and KEGG pathway enrichment analysis performed on significantly modulated proteins uncover the main immune processes (such as RNA interference, interferon-like pathway, antioxidant defense) which contribute to the defense and resistance of Pacific oysters to viral infection. In the more susceptible Pacific oysters, results suggest that OsHV-1 manipulate the molecular machinery of host immune response, in particular the autophagy system. This immunomodulation may lead to weakening and consecutively triggering death of Pacific oysters. The identification of several highly modulated and defense-related Pacific oyster proteins from the most resistant oysters supports the crucial role played by the innate immune system against OsHV-1 and the viral infection. Our results confirm the implication of proteins involved in an interferon-like pathway for efficient antiviral defenses and suggest that proteins involved in RNA interference process prevent viral replication in C. gigas. Overall, this study shows the interest of multi-omic approaches applied on groups of animals with differing sensitivities and provides novel insight into the interaction between Pacific oyster and OsHV-1 with key proteins involved in viral infection resistance.

Since its discovery in 2004, graphene has been used in a wide variety of fields including biomedicine, electronics, filtration materials, and surface coatings. The rapidly expanding consumer market for graphene family nanomaterials (GFNs), such as graphene oxide (GO), raises concern regarding their environmental toxicity. The aim of this study was to evaluate the effects of GO exposures in a marine filter-feeding bivalve (Crassostrea virginica) using sublethal biomarker approaches that can contribute to the development of an adverse outcome pathway (AOP). A 14-day study was conducted to identify tissue-specific molecular markers of GO toxicity using a static renewal design. Elevated lipid peroxidation and changes in glutathione-s-transferase (GST) activities were observed in gills and digestive gland tissues of the GO-exposed oysters. These cellular changes were noted for 2.5 and 5 mg/L GO exposures in seawater. Based on our results, reactive oxygen species (ROS)-induced oxidative damage is identified as a key event in the proposed AOP. Additionally, detoxification enzymes, such as GST, are thought to be involved in stress signaling leading to adverse effects on cellular health. This study is a part of our two-tier approach towards the identification of short- and long-term effects of GO exposures. This work, together with our previous 72 h exposure, represents the application of biomarker-based investigations in the process of AOP development for graphene family nanomaterials.

This study was performed to improve the ability to predict the concentrations of Escherichia coli in oyster meat and estuarine waters by using environmental parameters, and microbiological and heavy metal contamination from shellfish growing area in southern Thailand. Oyster meat (n = 144) and estuarine waters (n = 96) were tested for microbiological and heavy metal contamination from March 2016 to February 2017. Prevalence and mean concentrations of E. coli were 93.1% and 4.6 × 103 most probable number (MPN)/g in oyster meat, and 78.1% and 2.2 × 102 MPN/100 mL in estuarine water. Average 7-day precipitation, ambient air temperature, and the presence of Salmonella were associated with the concentrations of E. coli in oyster meat (p < 0.05). Raw data (MPN/g of oyster meat and MPN/100 mL of estuarine water) and log-transformed data (logMPN/g of oyster meat and logMPN/100 mL of estuarine water) of E. coli concentrations were examined within two contrasting regression models. However, the more valid predictions were conducted using non-log transformed values. These findings indicate that non-log transformed data can be used for building more accurate statistical models in microbiological food safety, and that significant environmental parameters can be used as a part of a rapid warning system to predict levels of E. coli before harvesting oysters.

Graphene is a 2-dimensional nanomaterial with unique mechanical, thermal, electrical, and optical properties. With increasing applications of graphene-family nanomaterials (GFNs) in electronics, biomedicine, and surface coatings, concern for their impacts on aquatic ecosystems is rising. Current information on the toxicity of GFNs, including graphene oxide, is scarce. Filter-feeding bivalves, such as eastern oysters, are good models for nanomaterial exposure studies. We present results from a 72-h static renewal oyster study using 1 and 10 mg/L graphene oxide, which, to our knowledge, is the first report on in vivo effects of graphene oxide exposures in marine bivalves. Water samples were analyzed for graphene oxide concentration and size assessments. Gill and digestive gland tissues were evaluated for lipid peroxidation and glutathione-S-transferase (GST) activity. In addition, gill sections were fixed for histopathological analyses. Elevated lipid peroxidation was noted in oysters exposed to 10 mg/L graphene oxide. No significant changes in GST activity were observed, but reduced total protein levels were found in digestive gland tissues of exposed oysters at both concentrations. Loss of mucous cells, hemocytic infiltration, and vacuolation were observed in gills of exposed oysters. The results indicate that short-term graphene oxide exposures can induce oxidative stress and epithelial inflammation and adversely affect overall oyster health. Further investigations regarding the fate and sublethal effects of graphene oxide are critical to understanding the risks associated with a rapidly growing graphene consumer market. The editorial "Spiralian genomics in a changing world" discusses the importance of studying spiralians, a diverse group of animal species, in the context of climate change. Despite being crucial to ecosystems and human culture and economy, spiralians have been understudied compared to other phyla. Advances in sequencing technologies have enabled researchers to generate genomic and transcriptomic data to better understand spiralian biology and their responses to climate change.
