Information on the ground fissures induced by coal mining is important to the safety of coal mine production and the management of environment in the mining area. In order to identify these fissures timely and accurately, a new method was proposed in the present paper, which is based on an unmanned aerial vehicle (UAV) equipped with a visible light camera and an infrared camera. According to such equipment, edge detection technology was used to detect mining-induced ground fissures. Field experiments show high efficiency of the UAV in monitoring the mining-induced ground fissures. Furthermore, a reasonable time period between 3:00 am and 5:00 am under the studied conditions helps UAV infrared remote sensing identify fissures preferably. The Roberts operator, Sobel operator, Prewitt operator, Canny operator and Laplacian operator were tested to detect the fissures in the visible image, infrared image and fused image. An improved edge detection method was proposed which based on the Laplacian of Gaussian, Canny and mathematical morphology operators. The peak signal-to-noise rate, effective edge rate, Pratt’s figure of merit and F-measure indicated that the proposed method was superior to the other methods. In addition, the fissures in infrared images at different times can be accurately detected by the proposed method except at 7:00 am, 1:00 pm and 3:00 pm.

Gemstones are minerals of gem qualities used for adornment and decoration with the attributes of beauty, durability and rarity. Traditionally, although China has been regarded as the most important source for nephrite, over the past decades, a large variety of gemstone resources have been newly discovered in China owing to continuous exploration works. The vast land with various geological and geochemical backgrounds is rich in gemstone resources with potential for new deposits discoveries. In pegmatites, gemstones are related to granitic magma events and mainly occur in pegmatitic cavities, such as tourmaline, aquamarine, spodumene, spessartine, moonstone, quartz, apatite, and topaz. The eruption of Tertiary basaltic magma provides gem-quality sapphire, spinel, olivine, garnet, and zircon. The supergene oxidation zones of some copper and iron deposits in Hubei and Anhui province host gem-quality turquoise and malachite. Moreover, the formation of the nephrite deposit in China is mostly related to the carbonatite and serpentinite rocks involved in the metamorphic-metasomatic processes. This paper comprehensively introduces the distribution of gemstones deposits, as well as the gemological and mineralogical characteristics of gemstones in China. Our present investigation provides insights into the gemstone potential of China for further exploitation.

Four new cytochalasans, phychaetoglobins A–D (1–4), together with twelve known cytochalasans (5–16), were isolated from a mangrove-associated fungus Chaetomium globosum kz-19. The new structures were elucidated on the basis of extensive 1D and 2D NMR, HR ESIMS spectroscopic analyses, and electronic circular dichroism (ECD) calculations. The absolute configuration of 2 was established by application of Mosher’s method. Compounds 4–8 exhibited moderate cytotoxicities against A549 and HeLa cell lines with the IC50 values less than 20 μM.

The stability of inclined shaft lining structure (ISLS) in complex water-rich strata is affected by many factors, such as water pressure, joint, soft rock, lining corrosion and so on. The instability of the ISLS will affect the safe and efficient coal mine production. Bathe sed on the geological conditions of the Xiaobaodang coal mine, this paper tested the evolution characteristics of concrete composition in long-term water seepage areas and revealed the influence mechanism of corrosion weakening of shaft lining (SL) in water-rich strata. Meanwhile, transient electromagnetic, ground penetrating radar, and infrared monitoring are used to detect the water-rich zones, and damage zones of surrounding rock and lining water seepage zones, and a three-level safety evaluation model for the instability risk of ISLS is constructed. Water abundance of the surrounding rock, surrounding rock deterioration, and shaft lining seepage were the specific indicators in the model. The main inclined shaft (MIS) in the studied coal mine is divided into three levels: non instability risk zone, potential instability risk zone, and high instability risk zone. According to the evaluation results, comprehensive prevention and control measures of “hydrophobic hole drainage” and “back-lining grouting” are adopted for the water inrush source and the surrounding rock micro-crack water channel. The precise prevention and control of ISLS is realized. The research results also provide a reference for the stability evaluation of ISLS and the accurate prevention and control under similar conditions. [ABSTRACT FROM AUTHOR]

Heparinase has an important application in the preparation of low-molecular-weight heparins and the deheparinization of heparin-treated blood. To increase the soluble expression of heparinase I (HepI) in recombinant Escherichia coli, the hepA gene (coding for HepI) from Flavobacterium heparinum was obtained through chemical synthesis and fused with the gene encoding hexahistidine, small ubiquitin-like modifier (SUMO), a flexible peptide linker (G4S) and a bovine enterokinase site (D4K). The constructed fusion protein (SUMO–HepI) was expressed in E. coli BL21 (DE3), and then purified with Ni2+-chelating affinity chromatography. The fermentation conditions were optimized and the enzymatic properties were also analyzed. As the results showed, the recombinant SUMO–HepI was successfully expressed in E. coli BL21 (DE3) and purified with affinity chromatography. The recombinant protein reached the highest soluble expression when the expression strain was induced by 0.6 mmol/L isopropyl β-D-thiogalactoside and grown at 30 °C with a shaking speed of 150 r/min for 9 h. The fusion protein could exhibit high enzyme activity without requirements of in vitro refolding and SUMO-tag releasing process, and the optimum enzyme activity was obtained at 30 °C, pH 7.0 and 10 mmol/L Ca2+ in the reaction buffer. This work provided a novel simple approach for the soluble expression of HepI in E. coli, and might establish a foundation for the following production and application of HepI in the industry.

Abstract Background Prostate cancer is known to have ethnic and regional differences. The study aimed to clinically evaluate the detection rate of prostate cancer on transrectal ultrasonography (TRUS)-guided prostate biopsy and analyze its characteristics among the northern Han Chinese population at a single center. Methods Between October 2009 and September 2016, a total of 1027 Chinese men, who had undergone TRUS-guided prostate biopsy at Qingdao Municipal Hospital, were retrospectively analyzed. Prostate biopsies were performed in the case of an abnormally elevated serum PSA level, and/or abnormal digital rectal examination (DRE) findings, and/or suspicious prostatic imaging findings. Results Of the 1022 men enrolled in the analysis, 438 patients (42.8%) were diagnosed with prostate adenocarcinoma histologically. When serum PSA levels were divided into five subgroups (less than 4.0, 4.0 to 10.0, 10.0 to 20.0, 20.0 to 100.0, and ≥ 100.0 ng/ml), the detection rates of prostate cancer were 12.4, 15.9, 34.1, 66.2, and 93.8%, respectively. With serum PSA levels of 4.0 to 10.0 ng/ml, the cancer detection rates for a normal DRE and a suspect DRE finding were 13.5 and 58.2%, respectively. Accordingly, the cancer detection rates for a normal imaging and a suspect imaging finding were 13.5 and 58.2%, respectively. Besides, a large proportion of the patients were in the clinically advanced stage. Conclusions The present study data reported a relatively higher prostate cancer detection rate of 42.8% and that the majority of the patients presented with clinically advanced prostate cancers within a local clinical urologic practice. An early detection and screening program for prostate cancer is of great need to reduce the burden from this disease among the northern Han Chinese population.

Abstract Vascular endothelial growth factor A (VEGF-A) is a pivotal player in angiogenesis. It is capable of influencing such cellular processes as tubulogenesis and vascular smooth muscle cell (VSMC) proliferation, yet very little is known about the actual signaling events that mediate VEGF-A induced VSMC phenotypic switch. In this report, we describe the identification of an intricate VEGF-A-induced signaling cascade that involves VEGFR2, STAT3, and Myocardin. We demonstrate that VEGF-A promotes VSMC proliferation via VEGFR2/STAT3-mediated upregulating the proliferation of markers like Cyclin D1 and PCNA. Specifically, VEGF-A leads to nitrosylation of Myocardin, weakens its effect on promoting the expression of contractile markers and is unable to inhibit the activation of STAT3. These observations reinforce the importance of nitric oxide and S-nitrosylation in angiogenesis and provide a mechanistic pathway for VEGF-A-induced VSMC phenotypic switch. In addition, Myocardin, GSNOR and GSNO can create a negative feedback loop to regulate the VSMC phenotypic switch. Thus, the discovery of this interactive network of signaling pathways provides novel and unexpected therapeutic targets for angiogenesis-dependent diseases.

Estrogen receptor alpha (ERα)-negative cells, which are enriched during endocrine therapy, are associated with metastatic relapse of breast cancer. Here the authors show that ERα inhibits breast cancer metastasis and suggest that ERα suppresses the amoeboid-like migration of breast cancer cells by upregulating vinculin.

Dark matter may interact with Standard Model (SM) particle through the exchange of a massive spin-2 graviton producing signals that can be detected. In this work we examine the γ-ray emission signals, including the line emission and the continuous spectrum component in such a massive graviton-mediated dark matter model. The constraints of LHC data, dark matter relic density as well as the dark matter indirect detection data have been applied to narrow down the parameter space. We focus on the vector dark matter model which could produce detectable γ-ray line signal. It is found that the γ-ray line data is effective on constraining the model parameters and the ongoing and upcoming space or ground-based γ-ray experiments can constrain the model further. As for the continuous γ-ray emission, the total effective annihilation cross section is ∼10−26 cm3s−1 except at the region where dark matter mass is around the graviton mass or half of it, which is consistent with current observational data and will be reliably probed by the upcoming CTA.

Histone acetylation influences the speed of seed germination. Here, Wang et al. show that loss of the SNL1/SNL2 histone deacetylase binding factors accelerates seed germination and provide evidence that they act by regulating the expression of AUX1which in turn influences cell division.

Traditional Chinese Medicines (TCMs) have been used for more than 3,000 years. However, most TCMs are used based on previous experience, and their exact mechanisms are still unclear. Proteomics, as a major component of systems biology, is an appropriate tool that could bring breakthroughs in the study of pharmacological mechanisms of TCMs. Researchers made substantial progress by using the proteomics approach to investigate the mechanism of TCMs. In this review, we summarize the current applications of proteomics in the mechanistic study of TCMs. Proteomics technologies and strategies that might be used in the future to enhance the study of TCMs are also discussed.

Gardeniae Fructus (GF) and carbonized GF (GFC) have been shown to exert a gastrointestinal protective effect and are frequently used in clinical practice for the treatment of hemorrhage and brown stool. In this study, we employed a combination of pharmacological methods and metabolomics in a rat model of ethanol-induced acute stomach ulcer to investigate the gastroprotective effect of GF and GFC water extracts and the potential mechanism involved in this process. The levels of nitric oxide (NO) and interleukin 6 (IL-6) in the plasma of rats were determined. The results showed that both GF and GFC reduced the ethanol-induced gastric lesions and expression of NO and IL-6 in these rats. Of note, 16 and 11 feature metabolites were filtered and identified in the GF and GFC groups, respectively. Both GF and GFC act by restoring the biosynthesis of valine, leucine, and isoleucine, and the metabolism of glycerophospholipids. Moreover, histological evaluation revealed that heat processing of GF to create GFC enhanced the gastric mucosa protective effect. Furthermore, heat processing converted the main pathway from alanine, aspartate, and glutamate metabolism, associated with GF, to histidine metabolism, associated with GFC. GF and GFC ameliorated gastric mucosa lesions in rats via reductions in NO production and inflammatory cytokine secretion, and the induction of prostaglandin E2.

Background: Depression has been linked to vitamin D deficiency. However, little attention was paid to the neural substrate underlying this association.Methods: Fifty patients with major depressive disorder (MDD) were enrolled in this study. High-resolution structural magnetic resonance imaging was performed to calculate total intracranial volume (TIV). Peripheral venous blood samples were collected to measure serum vitamin D concentration. Hamilton Rating Scale for Depression (HAMD) was used to assess severity of depression symptoms. The relationship among TIV, serum vitamin D concentration, and HAMD score was examined using correlation, linear regression, and mediation analyses.Results: In patients with MDD, HAMD score was negatively correlated with TIV and serum vitamin D concentration, and TIV was positively correlated with serum vitamin D concentration. Linear regression analyses showed that TIV and serum vitamin D concentration were significant predictors of HAMD score. Importantly, mediation analysis revealed that TIV significantly mediated the relationship between serum vitamin D concentration and HAMD score.Conclusion: Our findings suggest that TIV may serve as a potential neural biomarker for monitoring responses to adjuvant therapy of vitamin D in patients with MDD.

Gardeniae Fructus (GF), prepared GF (GFP), and carbonized GF (GFC) are widely used in China for the treatment of fever. However, the involved antipyretic mechanism has not been fully elucidated. In this study, rectal temperature and pyrogenic cytokines were used to evaluate the antipyretic effect of raw and processed GF in rats with dry-yeast-induced pyrexia. Reverse phase and hydrophilic interaction liquid chromatography and ultra-high-performance liquid chromatography/mass spectrometry were used to acquire the metabolomics profile of GF, GFP, and GFC in rats with pyrexia. The results showed that the rectal temperature of rats treated with GF, GFP, and GFC was suppressed after 6 h (P < 0.05), compared with that observed in pyrexia model rats. The enzyme-linked immunosorbent assay showed that the expression of tumor necrosis factor α and interleukin 6 were suppressed by GF, GFP, and GFC. Moreover, GFC suppressed the expression of interleukin 6 significantly (P < 0.01). Of note, 11, 15, and 25 feature metabolites were identified in the GF, GFP, and GFC groups. Pathway analysis showed that GF mainly regulated the biosynthesis of valine, leucine, and isoleucine. Notably, GFP was involved in glycerophospholipid metabolism, while GFC was linked to glycerophospholipid and sphingolipid metabolism. These results suggested that GF, GFP, and GFC maintained their antipyretic effect despite heat processing. However, heat processing altered endogenous feature metabolites and certain pathways of GF, GFP, and GFC in rats with yeast-induced pyrexia to exert an antipyretic effect.

Highwall mining (HWM) technology is an efficient method for exploiting residual coal resources in Chinese open-pit coal mines. However, on-site personnel and equipment can be damaged by the instability of the highwall mining residual coal pillars and subsidence of final end-walls. This paper considers the geological conditions of an open-pit mine in Shendong Coal Field (China) in order to prevent overlying rock fall accidents; the Mark-Bieniawski formula and the FLAC3D numerical simulation are used to analyse reasonable coal pillar widths outside and under the road, which were determined to be 1.7 m and 1.3 m, respectively. Using the EBH132 cantilever excavator for remote control mining, the field experiment shows that the recovery ratio of highwall residual coal resources was over 67%; hence, safety, efficiency and high recovery ratio of highwall mining were achieved for the residual coal resources of an open-pit mine.

This paper presents the results of an evaluation study of gas drainability in the Bulli seam in the Southern Coalfield of the Sydney Basin, NSW, Australia, where the coal seam gas (CSG) contains a high proportion of carbon dioxide (CO2). Historically the gas drainability in some particular areas of this coal seam was found to be particularly poor, which posed a significant challenge to gas predrainage. As a result, a large volume of greenhouse gases were released to the atmosphere during mining of the coal seam. Furthermore, the high gas content associated with the CO2-rich composition also increased the risks of coal and gas outburst incidents, affecting the safety of mining. After systematic literature review of evaluation factors affecting gas drainability, this evaluation study comprehensively analyzed the main critical factors, including the geology of the area, the coal cleat system, coal microstructure, coal permeability, coal sorption capacity, gas content, and gas composition. Field geology analysis showed geological variations that affected the variations of the coal cleat system and CO2 content in the coal seam. Scanning Electron Microscope (SEM) tests showed the tight and less-porous features in hard-to-drain coal samples. The colliery gas database analysis was carried out to assess the impact of gas content and gas composition on the drainability of the coal seam. Laboratory tests showed that the coal seam had a permeability of less than 1 mD and also showed that the coal seam was highly undersaturated, especially with high CO2 content.

The coal permeability is known to be influenced by the pore pressure and effective stress in coal mines. In this study, the characteristics of the bituminous coal permeability response to the pore pressure and effective shear stress in the Xutuan coal mine in Huaibei Coalfield in China were investigated under different stress conditions. For this purpose, gas seepage tests with various stress levels were conducted via the original gas flow and displacement testing apparatus using bituminous coal samples from the Xutuan coal mine. The pore pressure effect on the permeability under different stress conditions was assessed by varying the pore pressure in coal samples and simulating different in situ stresses. The axial and radial pressures were controlled to study the response of coal permeability to the effective shear stress. The experimental results revealed that with an increase in pore pressure, the permeability of coal in different stress environments firstly drops and then rises. The permeability increased gradually with the effective shear stress, which trend became more pronounced when the effective shear stress exceeded zero. In case of the axial pressure exceeding the radial one, the cross shear slip was observed, for which the permeability of coal samples increased with the effective shear stress. In the opposite case, the separated shear slip was observed, with the reverse trend.

Knowledge of the gas sorption characteristics of a coal not only helps to explain the mechanism of enhanced coalbed methane recovery but also provides an important basis for simultaneous coal and gas extraction. In consequence, the pure and binary gas excess sorption capacity of methane, carbon dioxide, and nitrogen of bituminous coal samples derived from the Xutuan Coal Mine in Huaibei coalfield, in Anhui Province in China, was measured using the volumetric method. The fitting analysis of the pure gas Langmuir adsorption model was carried out. The binary gas excess sorption measurement showed that the final sorption capacity of bituminous samples was the same no matter what the gas adsorption order of competitive adsorption and displacement adsorption. Hence, coal gas adsorption is physical adsorption, i.e. the different adsorption and desorption process of gas molecules does not affect the final adsorption amount of coal to each component of gas. Using the fitting parameters obtained by the Langmuir equation, the extended Langmuir equation was used to predict the adsorption capacity for each component of the binary gas. The comparison between predicted adsorption capacity and measured adsorption capacity showed that the extended Langmuir equation can better describe the trend of the adsorption isotherm curves of a binary gas under different pressures. The separation coefficient and displacement coefficient were defined from Langmuir adsorption theory. The separation coefficient involves the proportion of each component in the free phase and the proportion of each component in the adsorption phase. The displacement coefficient involves the displacement ability of gas molecules at adsorption sites by free gas molecules.

Moisture content in coal is an important factor affecting the coal seam gas extraction. It directly affects the storage and flow of gas in bituminous coal. In this paper, the cylindrical bituminous coal cores of Xutuan coal mine in Huaibei coal mine group were studied as experimental objects, using the laboratory self-designed experimental device Gas Adsorption and Strain Testing Apparatus system . The influence of the bituminous coal moisture content on gas adsorption characteristics was studied. Drying experiments of coal samples showed that they lose the original moisture content following the exponential decay function of time. At wetting, the saturated moisture content in coal samples increased following the Exponential Association function of time. The experimental results show that the average original moisture content and average saturated moisture content of raw coal samples are 1.3 and 2.4%, respectively. On this basis, the gas adsorption experiments on samples with different moisture contents under different gas pressures were carried out. With the moisture content increase, the gas adsorption capacity and saturation value decreased and the decrease rate gradually reduced. The single exponential decay function describes the gas adsorption capacity dependence on moisture content. Moisture content also affects the adsorption deformation of bituminous coal. At high moisture content, the adsorption deformation of bituminous coal is less.

Seven important grain traits, including grain length (GL), grain width (GW), grain perimeter (GP), grain area (GA), grain length/width ratio (GLW), roundness (GR), and thousand-grain weight (TGW), were analyzed using a set of 139 simple sequence repeat (SSR) markers in 130 hexaploid wheat varieties and 193 Aegilops tauschii accessions worldwide. In total, 1 612 alleles in Ae. tauschii and 1 360 alleles in hexaploid wheat (Triticum aestivum L.) were detected throughout the D genome. 197 marker-trait associations in Ae. tauschii were identified with 58 different SSR loci in 3 environments, and the average phenotypic variation value (R2) ranged from 0.68 to 15.12%. In contrast, 208 marker-trait associations were identified in wheat with 66 different SSR markers in 4 environments and the average phenotypic R2 ranged from 0.90 to 19.92%. Further analysis indicated that there are 6 common SSR loci present in both Ae. tauschii and hexaploid wheat, which are significantly associated with the 5 investigated grain traits (i.e., GA, GP, GR, GL, and TGW) and in total, 16 alleles derived from the 6 aforementioned SSR loci were shared by Ae. tauschii and hexaploid wheat. These preliminary data suggest the existence of common alleles may explain the evolutionary process and the selection between Ae. tauschii and hexaploid wheat. Furthermore, the genetic differentiation of grain shape and thousand-grain weight were observed in the evolutionary developmental process from Ae. tauschii to hexaploid wheat.
