Long term residues of organochlorine pesticides (OCPs) in soils are of great concerning because they seriously threaten food security and human health. This article focuses on isolation of OCP-degrading strains and their performance in bioremediation of contaminated soil under ex situ conditions. A bacterium, Chryseobacterium sp. PYR2, capable of degrading various OCPs and utilizing them as a sole carbon and energy source for growth, was isolated from OCP-contaminated soil. In culture experiments, PYR2 degraded 80-98% of hexachlorocyclohexane (HCH) or 1,1,1-trichloro-2,2-bis (4-chlorophenyl) ethane (DDT) isomers (50 mg L(-1)) in 30 days. A pilot-scale ex situ bioremediation study of highly OCP-contaminated soil augmented with PYR2 was performed. During the 45-day experimental period, DDT concentration was reduced by 80.3% in PYR2-augmented soils (35.37 mg kg(-1) to 6.97 mg kg(-1)) but by only 57.6% in control soils. Seven DDT degradation intermediates (metabolites) were detected and identified in PYR2-augmented soils: five by GC/MS: 1,1-dichloro-2,2-bis (4-chlorophenyl) ethane (DDD), 1,1-dichloro-2,2-bis (4-chlorophenyl) ethylene (DDE), 1-chloro-2,2-bis (4-chlorophenyl) ethylene (DDMU), 1-chloro-2,2-bis (4-chlorophenyl) ethane (DDMS), and dichlorobenzophenone (DBP); and two by LC/MS: 4-chlorobenzoic acid (PCBA) and 4-chlorophenylacetic acid (PCPA). Levels of metabolites were fairly stable in control soils but varied greatly with time in PYR2-augmented soils. Levels of DDD, DDMU, and DDE in PYR2-augmented soils increased from day 0 to day 30 and then decreased by day 45. A DDT biodegradation pathway is proposed based on our identification of DDT metabolites in PYR2-augmented systems. To contribute to the use of the tropical brown mussel Perna perna as a sentinel species for organochlorine pesticides (OCP) and polychlorinated biphenyls (PCB), the present study reports data on the toxicokinetics of these compounds in P. perna. Specifically, the authors present data on OCP and PCB bioaccumulation for eight sampling months from three bays (SE Brazil) and two transplant experiments (each 1 month long). Although seasonality is observed in the total lipid content of the whole soft tissue, with summer samples showing higher values, no such seasonality is observed in the OCP and PCB concentrations bioaccumulated by the mussel P. perna. Because no seasonal effect is observed in the annual OCP and PCB concentrations bioaccumulated by P. perna, the use of this species as a sentinel organism to monitor organochlorinated compounds is encouraged. One month of transplantation is not enough to allow the transplanted specimens to reach the concentrations observed in animals reared at the destination site. Nevertheless, P. perna showed a clear tendency to depurate the DDT metabolites p,p'-DDD and p,p'-DDE after 1 month of transplantation.

Soil samples were collected at various depths (0.5-21.5 m) from ten boreholes that were drilled with a SH-30 Model Rig, four of which were at a dicofol production site while six were at a dichlorodiphenyltrichloroethane (DDT) production site. In industrial sites, the shallow soils at depths of 0-2 m were mostly backfill soils, which cannot represent the contamination situation of the sites. The contaminated levels in the deep original soil can represent the situation in contaminated sites. All the soil samples investigated at the DDT and dicofol production sites were found to be seriously polluted. The contents of both DDT (0.6-6071 mg/kg) and dicofol (0.5-1440 mg/kg) were much higher at the dicofol production site than at the DDT production site (DDTs, 0.01-664.6 mg/kg; dicofol, <0.1 mg/kg), even in the deep soil. DDTs had a different distribution in the soil of the pesticide production site from that in the soil outside the sites and that in agricultural soils. The results of the investigation revealed that DDTs were easily enriched in cohesive soil and in the bottom zone of aquifers, where the concentration was higher than in above the layers. DDTs were found to be hard to degrade, and their degradation speed was slower than their vertical migration, despite the fact that hydrophobic DDTs did not migrate easily in soils. In the dicofol production site, the value of DDE/DDD cannot indicate the degradation condition of DDTs, nor can the value of (DDE + DDD)/DDT identify how long DDTs have remained in the soil. It is debatable that the half-life of DDT inputted to soils is about 20-30 years, maybe longer than the generally recognized time.

Organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) were assessed in blubber from 35 dead Southern Right Whales (SRW - Eubalaena australis) stranded at Península Valdés, Argentina. The life cycle includes a feeding period in high productivity areas of the South West Atlantic and a reproductive period in coastal template waters of Argentina. Organochlorine pesticides showed higher concentrations (22.6±13.8 ng·g(-1)ww) than PCBs (7.5±10 ng·g(-1)ww). Among pesticides, HCHs, DDTs, endosulfans, dieldrin, chlordans, heptachlor epoxide, and trans-nonachlor were detected. p,p'-DDE and p,p'-DDT were present in 69% and 26% of samples, respectively. p,p'-DDT/p,p'-DDE ratio showed low values (<0.33) as a result of aged DDT inputs. Concentrations of polychlorinated biphenyls (PCBs), dichlorodiphenyl trichloroethane and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), hexachlorobenzene (HCB) and chlordane compounds (CHLs) were determined in the blubber of males (20-25 years old) of Antarctic minke whales, Balaenoptera bonaerensis, from the International Whaling Commission (IWC) management Areas IV (70°-130°E) and V (130°E-170°W), south 60°S. The ranges of concentrations (ng g(-1) lipid wt.) for each compound were, PCBs: 7.7-89; DDTs: 29-340; HCHs: 0.20-4.3; HCB: 75-430; CHLs: 10-120, which were much lower than those in common minke whales, Balaenoptera acutorostrata, from the northern hemisphere. The levels of PCBs, HCHs, HCB and CHLs in Area IV were significantly higher than those in Area V, while the levels of DDTs in both areas were similar. For comparing the fate among four pesticides in the Antarctic Ocean avoiding the effect of variance due to food intake, the ratios of the pesticides to PCBs, which has an extremely high chemical stability and environmental persistence, were examined. The HCHs/PCBs ratio decreased by a factor of about 20 in a span of 16 years in both Areas IV and V, while temporal trends of DDTs/PCBs, HCB/PCBs and CHLs/PCBs ratios were not observed. These results indicate that PCBs, DDTs, HCB and CHLs levels did not vary or slightly decreased in Areas IV and V during the study period. However HCHs levels clearly decreased. Biotransformation plays an important role in the bioaccumulation and toxicity of a chemical in biota. Dichlorodiphenyltrichloroethane (DDT) commonly co-occurs with its metabolites (dichlorodiphenyldichloroethane [DDD] and dichlorodiphenyldichloroethylene [DDE]), in the environment; thus it is a challenge to accurately quantify the biotransformation rates of DDT and distinguish the sources of the accumulated metabolites in an organism. The present study describes a method developed to quantitatively analyze the biotransformation of p,p'-DDT in the benthic polychaete, Nereis succinea. The lugworms were exposed to sediments spiked with DDT at various concentrations for 28 d. Degradation of DDT to DDD and DDE occurred in sediments during the aging period, and approximately two-thirds of the DDT remained in the sediment. To calculate the biotransformation rates, residues of individual compounds measured in the bioaccumulation testing (after biotransformation) were compared with residues predicted by analyzing the partitioning of the parent and metabolite compounds between gut fluid and tissue lipid (before biotransformation). The results suggest that sediment ingestion rates decreased when DDT concentrations in sediment increased. Extensive biotransformation of DDT occurred in N. succinea, with 86% of DDT being metabolized to DDD and <2% being transformed to DDE. Of the DDD that accumulated in the lugworms, approximately 70% was the result of DDT biotransformation, and the remaining 30% was from direct uptake of sediment-associated DDD. In addition, the biotransformation was not dependent on bulk sediment concentrations, but rather on bioaccessible concentrations of the chemicals in sediment, which were quantified by gut fluid extraction. The bioremediation of a long-term contaminated soil through biostimulation and surfactant addition was evaluated. The concentrations of 1,1,1-trichloro-2,2-bis(4-chlorophenyl) ethane (DDT) and its metabolites 1,1-dichloro-2,2-bis(4-chlorophenyl) ethane (DDD) and 1,1-dichloro-2,2-bis(4-chlorophenyl) ethylene (DDE) were monitored during an 8-week remediation process. Physicochemical characterization of the treated soil was performed before and after the bioremediation process. The isolation and identification of predominant microorganisms during the remediation process were also carried out. The efficiency of detoxification was evaluated after each bioremediation protocol. Humidity and pH and the heterotrophic microorganism count were monitored weekly. The DDT concentration was reduced by 79% after 8 weeks via biostimulation with surfactant addition (B+S) and 94.3% via biostimulation alone (B). Likewise, the concentrations of the metabolites DDE and DDD were reduced to levels below the quantification limits. The microorganisms isolated during bioremediation were identified as Bacillus thuringiensis, Flavobacterium sp., Cuprivadius sp., Variovorax soli, Phenylobacterium sp. and Lysobacter sp., among others. Analysis with scanning electron microscopy (SEM) allowed visualization of the colonization patterns of soil particles. The toxicity of the soil before and after bioremediation was evaluated using Vibrio fischeri as a bioluminescent sensor. A decrease in the toxic potential of the soil was verified by the increase of the concentration/effect relationship EC50 to 26.9% and 27.2% for B+S and B, respectively, compared to 0.4% obtained for the soil before treatment and 2.5% by natural attenuation after 8 weeks of treatment., (Copyright © 2014. The uptake of organic pollutants by plants is an important process for the exposure of humans to toxic chemicals. The objective of this study was to calibrate the parameters of a common plant uptake model by comparison to experimental results from literature. Radish was grown in contaminated soil (maximum concentration 2.9 mg/kg dw) and control plot. Uptake of HCHs, HCB, PCBs, and DDT plus metabolites was studied (log K(ow) 3.66 to 7.18). Measured BCF roots-to-soil were near 1 g/g dw on the control plot and about factor 10 lower for the contaminated soil. With default data set, uptake into roots of most substances was under predicted up to factor 100. The use of site-specific data improved the predictions. Consideration of uptake from air into radish bulbs was relevant for PCBs. Measured BCF shoots ranged from <0.1 to >10 g/g dw and were much better predicted by the standard model. The results with default data and site-specific data were similar. Deposition from air was the major uptake mechanism into shoots. Transport from soil with resuspended particles was only relevant for the contaminated plot. The calculation results (in dry weight) were most sensitive to changes of the water content of plant tissue.

A plasmid transfer-mediated bioaugmentation method for the enhancement of dichlorodiphenyltrichloroethane (DDT) degradation in soil was developed using the catabolic plasmid pDOD from Sphingobacterium sp. D-6. The pDOD plasmid could be transferred to soil bacteria, such as members of Cellulomonas, to form DDT degraders and thus accelerate DDT degradation. The transfer efficiency of pDOD was affected by the donor, temperature, moisture, and soil type. Approximately 50.7% of the DDT in the contaminated field was removed 210 days after the application of Escherichia coli TG I (pDOD-gfp). Long-beaked common dolphins (Delphinus capensis) in the Gulf of California have been exposed to persistent contaminants that originated in large agricultural areas near the coast. Live common dolphins were sampled by remote dart biopsies to determine concentrations of tDDT in blubber. Life stage and initial gender identification was determined by field observations. Gender was confirmed by genetic analysis of the skin. Concentration of tDDT in blubber was analyzed by gas chromatography. The 16 samples collected consisted of: 2 adult males, 6 adult females, and 8 juveniles. 4,4'-DDE was detected in most of the samples with 4,4'-DDD and 4,4'-DDT under detection levels. Concentrations of DDE varied from non-detectable to 87.3 µg/g lipid weight with a median of 16 µg/g lipid weight. The highest concentration was detected in an immature female. No differences were detected between gender or life stage but this could be attributed to small sample size. We recommend continued sampling of D. capensis blubber biopsies from the Gulf of California in order to relate these levels with affected in vitro biomarkers such as mixed function oxidase activity.

In the present work, persistent organic pollutants (POPs), such as polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane (DDT) and metabolites, polybrominated diphenyl ethers (PBDEs), and hexachlorocyclohexane (HCH) were analyzed in three Antarctic notothenioids fish species: Trematomus newnesi (TRN), Notothenia coriiceps (NOC) and Notothenia rossii (NOR). The contribution of each POP-family to the total load was as follows: ΣPCB (40%)>ΣDDT (27%)>ΣPBDEs (23%)>ΣHCH (10%). Among the 23 PCB congeners analyzed, penta-CBs homologues were the prevalent group, followed by hexa-CBs and hepta-CBs. DDT and its metabolites presented the following trend: p,p'-DDT>p,p'-DDE~p,p'-DDD. PBDE profile was dominated by BDE-47 and BDE-99 congeners, followed by BDE-100>BDE-28>BDE-154, BDE-153. Among HCHs, the γ-HCH isomer was detected in all samples, constituting 69% total HCH load, while α-HCH and β-HCH contributions were 15% and 16%, respectively. The levels of POPs reported here suggest that NOR and NOC are more susceptible to accumulate the analyzed contaminants than TRN, a species not previously analyzed for POPs. Distribution of POPs among different tissues of the three species (muscle, liver, gonads, and gills) was also investigated. Considering lipid weight, the general pattern of POPs distribution in tissues indicated that while gonads showed higher levels of PCBs, DDTs and HCH, the most significant PBDE concentrations were recorded in gills. The residues and enantiomeric fractions of organochlorine pesticides (OCPs) were measured in 11 mollusk species collected from the coastal areas along the Yangtze River Delta to evaluate the status, potential sources, and health risks of pollution in these areas. The concentrations of DDTs, HCHs, and chlordanes ranged from 6.22 to 398.19, 0.66-7.11, and 0.14-4.08 ng g(-1) based on wet weight, respectively; DDTs and HCHs have the highest values, globally. The DDTs increased and the HCHs decreased compared to historical data. Both the box-and-whisker plots and the one-way ANOVA tests indicated that the OCP levels varied little between sampling locations and organism species. The compositions of the DDTs and HCHs suggested a cocktail input pattern of fresh and weathered technical products. The comparative EF values for the α-HCH between the sediments and mollusks, as well as the lack of any discernible difference in the relative proportions of HCH isomers among different species from the same sampling site implied that the HCH residues in the mollusks came directly from the surrounding environment. However, the biotransformation of DDTs in mollusks cannot be precluded. The assessments performed based on several available guidelines suggested that although no significant human health risks were associated with the dietary intake of OCPs, the concentrations of DDTs exceeded the maximum residual limits of China and many developed nations. Moreover, an increased lifetime cancer risk from dietary exposure to either DDTs or HCHs remains a possibility. Because non-racemic OCP residues are common in the mollusk samples, our results suggest a need to further explore the levels and toxicity of the chiral contaminants in mollusks and other foodstuff to develop the human risk assessment framework based on chiral signatures., (Copyright © 2014 Elsevier Ltd. In this study, the levels of DDT and its metabolite residues were determined in chewable parts of Catha edulis plants grown in the southern part of Ethiopia. The levels of p,p'-DDT and p,p'-DDE were found to be in the range of 10.8-19.7 and 3.5-18.6 μg/kg, respectively. These data revealed that the residue being detected is from recent applications. The estimated daily intake of total DDT from C. edulis consumption was calculated to be in the range between 0.0278 and 0.0747 μg/kg, which is significantly lower than the FAO/WHO guideline. However, this may not guarantee safety, as the application of DDT extends to vegetables as well. Even though the use of DDT was banned in Ethiopia for agriculture purposes, detectable levels are still being observed. The results of the study necessitate the need for awareness creation among the people in the community.

Indo-Pacific humpback dolphins (Sousa chinensis) are apex predators in the Pearl River Estuary waters (PRE) of China. PCBs, DDTs and other organochlorine pesticides (OCPs) (e.g., HCHs, HCB, mirex and dieldrin) were analysed in blubber samples of 45 dolphins and 10 prey fishes of S. chinensis collected from 2004 to 2013 in the PRE region to investigate the bioaccumulation and potential biomagnification of persistent organic pollutants (POPs). DDTs were the most abundant residue in the dolphins, with an average of 6,2700 ng g(-1) wet weight (ww), followed by PCBs (average: 1,790 ng g(-1) ww) and other OCPs including ∑HCHs, mirex, endrin, ∑chlordanes, HCB, dieldrin, aldrin, heptachlor, and pentachlorobenzene. The concentrations of PCBs and DDTs in male dolphins significantly increased with age and length. In contrast, female dolphins did not show obvious bioaccumulation trends with age and body length, possibly due to the lactational and parturitional transfer of these compounds. Compared with the POP residues in the prey fishes, the concentrations of PCBs, DDTs, and HCHs in the dolphin blubber increased by factors of 99, 212, and 5, respectively, whereas the residue levels of the other OCPs increased 2-185 times, indicating a potentially significant biomagnification in the top predators. In the 1950s and 60s, discharges from a DDT manufacturing plant contaminated a tributary system of the Tennessee River near Huntsville, Alabama, USA. Regulatory action resulted in declaring the area a Superfund site which required remediation and extensive monitoring. Monitoring data collected from 1988, after remediation, through 2011 showed annual decreases approximating first-order decay in concentrations of total DDT and its six principal congeners (p,p'-DDT, o,p'-DDT, p,p'-DDD, o,p'-DDD, p,p'-DDE and o,p'-DDE) in filets from three species of fish. As of 2013, these concentrations met the regulatory requirements of 5 mg/kg or less total DDT for each fish tested. The enantiomer fractions (EF) of chiral o,p'-DDD in smallmouth buffalo and channel catfish were always below 0.5, indicating preferential decay of the (+)-enantiomer of this congener; this EF did not change significantly over 15 years. Organochlorine contaminants in upper trophic-level consumers inhabiting Puget Sound are consistently higher than in those species inhabiting other west coast locations. We analyzed persistent organic pollutants (POPs) in the six most common fish prey of rhinoceros auklets breeding on Protection Island (Puget Sound), Tatoosh Island (WA coast), and Destruction Island (WA coast). The aim of this study was to evaluate the impact of selected electron donors and electron acceptors on the anaerobic biodegradation of DDT and its major metabolites in a muck soil with a long history of exposure to the pesticide. Loss of DDT was measured in anaerobic microcosms supplemented with H2, lactate, and acetate. The greatest loss of DDT (approximately 87 %) was observed in microcosms amended with lactate and no additional electron acceptor compared to the no additional electron donor or acceptor sets. An increase in measureable concentrations of DDx was observed in un-amended microcosms. In larger scale mesocosms, significant increases in dissolved organic carbon (DOC) corresponded with low redox potentials. Increases in DOC corresponded with sharp increases in measured concentrations of DDx, followed by a decrease in measured DDT concentrations in lactate-amended mesocosms. Our studies indicate that sorbed DDx is released upon anaerobic incubation, and that indigenous microorganisms capable of DDx degradation respond to lactate additions. Both the potential for release of sorbed DDx and the potential for biodegradation of DDx should be considered during remediation of DDx-contaminated organic soils at low redox potentials.

A 1,1,1-trichloro-2,2-bis(4-chlorophenyl)ethane (DDT) degrading bacterium strain IITR03 producing trehalolipid was isolated and characterized from a pesticides contaminated soil. The strain IITR03 was identified as a member of the genus Rhodococcus based on polyphasic studies. Under aqueous culture conditions, the strain IITR03 degraded 282 μM of DDT and could also utilize 10mM concentration each of 4-chlorobenzoic acid, 3-chlorobenzoic acid and benzoic acid as sole carbon and energy source. The catechol 1,2-dioxygenase enzyme activity resulted in conversion of catechol to form cis,cis-muconic acid. Cloning and sequencing of partial nucleotide sequence of catechol 1,2-dioxygenase gene (cat) from strain IITR03 revealed its similarity to catA gene present in Rhodococcus sp. strain Lin-2 (97% identity) and Rhodococcus strain AN22 (96% identity) degrading benzoate and aniline, respectively. While a number of studies have reported residual levels of persistent halogenated compounds (PHCs) in crocodilia, there is still a dearth of information on the Chinese alligator, a critically endangered crocodilian species. In the present study, several PHCs, including polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), and dichlorodiphenyltrichloroethane and its metabolites (DDTs), were detected in the adult tissues, neonates, and eggs of captive Chinese alligators from China. The concentrations of ΣPBDEs, ΣPCBs, and ΣDDTs in Chinese alligators ranged from 0.11 to 16.1, 1.12 to 22.2, and 6.03 to 1020ngg(-1) wet weight, respectively, with higher levels of ΣPCBs and ΣDDTs in the neonates and eggs than in muscle tissues. Thirty-two light-vented bulbuls (Pycnonotus sinensis) were collected from six sampling sites in Guangdong Province, South China to investigate the geographical variation on the occurrence of polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane and its metabolites (DDTs). Concentrations of PCBs and DDTs in the pectoral muscle of light-vented bulbul ranged from 140 to 73,000 ng/g lipid weight (lw) and 12 to 4600 ng/g lw, respectively. PCB concentrations were significantly higher in birds from e-waste site compared to other sampling sites (mean, 18,000 vs 290 ng/g lw, p<0.0001), implying that PCBs mainly came from e-waste recycling activities. No significant differences for DDT levels were observed among the sampling sites (p=0.092). Differences in PCB homologue profiles among the sampling sites were found and can be probably ascribed to different local contamination sources. p,p'-DDE (>80%) was the most abundant component of DDTs in birds. Compositional pattern of DDTs suggested that historical residue was the main source of DDT. The toxic equivalent (TEQ) concentrations had significant positive correlations with PCB concentrations, indicating that elevated PCB levels may have adverse effects on light-vented bulbuls., (Copyright © 2014 Elsevier B.V. Persistent organic pollutants (POPs) are toxic man-made chemicals that bioaccumulate and biomagnify in food webs, making them a ubiquitous threat to the marine environment. Although many studies have determined concentrations of POPs in top predators, no studies have quantified POPs in stranded cetaceans within the last 30 years around the Hawaiian Islands. A suite of POPs was measured in the blubber of 16 cetacean species that stranded in the tropical Pacific, including Hawai'i from 1997 to 2011. The sample set includes odontocetes (n=39) and mysticetes (n=3). Median (range) contaminant concentrations in ng/g lipid for the most representative species category (delphinids excluding killer whales [n=27]) are: 9650 (44.4-99,100) for ∑DDTs, 6240 (40.8-50,200) for ∑PCBs, 1380 (6.73-9520) for ∑chlordanes, 1230 (13.4-5510) for ∑toxaphenes, 269 (1.99-10,100) for ∑PBDEs, 280 (2.14-4190) for mirex, 176 (5.43-857) for HCB, 48.1 (<5.42-566) for ∑HCHs, 33.9 (<2.42-990) for ∑HBCDs, 1.65 (<0.435-11.7) for octachlorostyrene and 1.49 (<2.07-13.1) for pentachlorobenzene. ∑PCB concentrations in these Pacific Island cetaceans approach and sometimes exceed proposed toxic threshold values. Backward stepwise multiple regressions indicated the influence of life history parameters on contaminant concentrations when performed with three independent variables (species category, year of stranding, and sex/age class). No temporal trends were noted (p>0.063), but sex/age class influences were evident with adult males exhibiting greater contaminant loads than adult females and juveniles for ∑DDT, ∑PCBs, ∑CHLs, and mirex (p≤0.036). POP concentrations were lower in mysticetes than odontocetes for many compound classes (p≤0.003). p,p'-DDE/∑DDTs ratios were greater than 0.6 for all species except humpback whales, suggesting exposure to an old DDT source. Organochlorine pesticides have been used in Mexico in malaria control programs and against ectoparasites. The objective of this study was to compare the levels of organochlorine pesticides: HCB, α-β-γ-HCH, pp'-DDE, op'-DDT and pp'-DDT in adipose tissue of female inhabitants from three Mexican states: Veracruz, Puebla and Tabasco. Data analyses indicated higher β-HCH levels in Puebla inhabitants. When comparing the mean values of the pp'-DDE concentrations among the three states, no statistically significant differences were noted. A trend of increasing concentrations of op'-DDT from Veracruz to Puebla and Tabasco was observed. Significantly higher pp'-DDT concentrations in Veracruz as compared to Puebla and Tabasco were determined. Using factorial analysis of three age categories (>30, 31-50, <51) organochlorine pesticide concentrations increases with age of participants, indicating time of exposure as a principal factor of organochlorine pesticides accumulation in adipose tissue.

Adding organic amendments to stimulate the biodegradation of pesticides is a subject of ongoing interest. The effect of sewage sludge on the bioremediation of dichlorodiphenyltrichloroethane (DDT) and hexachlorocyclohexane (HCH) contaminated soil was investigated in bench scale experiments, and intermittent aeration strategy was also used in this study to form an anaerobic-aerobic cycle. Bioremediation of DDT and HCH was enhanced with the addition of sewage sludge and the intermittent aeration. The removal rates of HCH and DDT were raised by 16.8%-80.8% in 10 days. Sewage sludge increased the organic carbon content from 6.2 to 218 g/kg, and it could also introduce efficient degradation microbes to soil, including Pseudomonas sp., Bacillus sp. and Sphingomonas sp. The unaerated phase enhanced the anaerobic dechlorination of DDT and HCH, and anaerobic removal rates of β-HCH, o,p'-DDT and p,p'-DDT accounted for more than 50% of the total removal rates, but the content of α-HCH declined more in the aerobic phase., (Copyright © 2014. Background: Increased insecticide detoxification mediated by cytochrome P450s is a common mechanism of insecticide resistance. Although Cyp6a2 has been observed to be overexpressed in many 4,4'-dichlorodiphenyltrichloroethane (DDT)-resistant strains of Drosophila melanogaster, how Cyp6a2 is regulated and whether its overproduction confers DDT resistance remain elusive., Results: Molecular analysis identified five Cyp6a2 alleles (Cyp6a2(Canton) (-S-1) , Cyp6a2(Canton) (-S-2) , Cyp6a2(91-C) , Cyp6a2(91-R) and Cyp6a2(Wisconsin) (-) (WD) ) from four D. melanogaster strains, notably differing in the presence or absence of an intact Nrf2/Maf (a transcription factor) binding site in the 5'-promoter core region, a 'G1410' frameshift deletion mutation in the heme-binding region and a long terminal repeat (LTR) of transposable element 17.6 in the 3'-untranslated region (UTR). Linkage analysis confirmed that DDT resistance was genetically linked to a Nrf2/Maf-binding-site-containing, LTR-lacking functional allele of Cyp6a2 (Cyp6a2(91-R) ). The qRT-PCR results showed that overexpression of functional Cyp6a2 was consistently associated with DDT resistance. Luciferase reporter gene assays revealed that an intact Nrf2/Maf binding site in the 5'-promoter core region enhanced the constitutive transcription of Cyp6a2., Conclusion: The results suggest that the Nrf2/Maf binding-site-containing functional Cyp6a2 allele is associated with DDT resistance in the D. melanogaster strains under study., (© 2013 Society of Chemical Industry.)

We investigated occurrence, transport pathways, and effects of polybrominated diphenyl ether (PBDE) flame retardants and other endocrine disrupting chemicals (EDCs) in aquatic media and the foodweb in the lower Columbia River. In 2009 and 2010, foodweb sampling at three sites along a gradient of contaminant exposure near Skamania (Washington), Columbia City (Oregon) and Longview (Washington) included water (via passive samplers), bed sediment, invertebrate biomass residing in sediment, a resident fish species (largescale suckers [Catostomus macrocheilus]), and eggs from osprey (Pandion haliaetus). This paper primarily reports fish tissue concentrations. In 2009, composites of fish brain, fillet, liver, stomach, and gonad tissues revealed that overall contaminant concentrations were highest in livers, followed by brain, stomach, gonad, and fillet. Concentrations of halogenated compounds in tissue samples from all three sites ranged from <1 to 400nanograms per gram of wet tissue. Several chemical classes, including PBDEs, organochlorine pesticides, and polychlorinated biphenyls (PCBs), were detected at all sites and in nearly all fish tissues sampled. In 2010, only fish livers were sampled and inter-site concentration differences were not as pronounced as in 2009. Chemical concentrations in sediments, fish tissues, and osprey eggs increased moving downstream from Skamania to the urbanized sites near Columbia City and Longview. Numerous organochlorine (OC) pesticides, both banned and currently used, and PBDEs, were present at each site in multiple media and concentrations exceeded environmental quality benchmarks in some cases. Frequently detected OC compounds included hexachlorobenzene, pentachloroanisole, dichlorodiphenyltrichloroethane (DDT) and its degradates, chlorpyrifos, and oxyfluorofen. Biomagnification of BDE47, 100, 153, and 154 occurred in largescale suckers and osprey eggs. Pooled yellow European eel (Anguilla anguilla (L.)) samples, consisting of 3-10 eels, collected between 2000 and 2009 from 60 locations in Flanders (Belgium) were investigated for persistent contaminants, such as polybrominated diphenyl ethers (PBDEs), hexabromocyclododecanes (HBCDs), polychlorinated biphenyls (PCBs) and dichlorodiphenyltrichloroethane and its metabolites (DDTs). The current study expands the knowledge regarding these contaminant concentrations, their patterns and distribution profiles in aquatic ecosystems. PBDEs, HBCDs, PCBs, and DDTs were detected in all eel samples and some samples had high concentrations (up to 1400, 9500, 41,600 and 7000ng/g lw, respectively). PCB levels accounted for the majority of the contamination in most samples. The high variability in PBDE, HBCD, PCB and DDT concentrations reported here is likely due to the variety in sampling locations demonstrating variable local pollution pressures, from highly industrialised areas to small rural creeks. Among PBDEs, BDE-47 (57% contribution to the sum PBDEs), -100 (19%) and -99 (15%) were the predominant congeners, similar to the composition reported in the literature in eel samples. For HBCDs, α-HBCD (74%) was predominant followed by γ-(22%) and β-HBCD (4%) isomers in almost all eel samples. CB-153 (19%) was the most dominant PCB congener, closely followed by CB-138 (11%), CB-180 (9%), CB-187 (8%) and CB-149 (7%). The contribution to the total human exposure through local wild eel consumption was also highly variable. Intake of PBDEs and HBCDs, through consumption of wild eel, was below the RfD values for the average population (consuming on average 2.9g eel/day). At 16 out 60 sites, eels exceeded largely the new EU consumption threshold for PCBs (300ng/g ww for the sum of 6 indicator PCBs). A combined quantum mechanics/molecular mechanics (QM/MM) computation of the detoxifying mechanism of an epsilon class glutathione transferases (GSTs) toward organochlorine insecticide DDT, 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane, has been carried out. The exponential average barrier of the proton transfer mechanism is 15.2 kcal/mol, which is 27.6 kcal/mol lower than that of the GS-DDT conjugant mechanism. It suggests that the detoxifying reaction proceeds via a proton transfer mechanism where GSH acts as a cofactor rather than a conjugate. The study reveals that the protein environment has a strong effect on the reaction barrier. The experimentally proposed residues Arg112, Glu116 and Phe120 were found to have a strong influence on the detoxifying reaction. The influence of residues Pro13, Cys15, His53, Ile55, Glu67, Ser68, Phe115, and Leu119 was detected as well. It is worth noticing that Ile55 facilitates the detoxifying reaction most. On the basis of the structure of DDT, structure 2, (BrC6H4)2CHCCl3, is the best candidate among all the tested structures in resisting the detoxification of enzyme agGSTe2.

Contamination of organochlorine pesticides (OCPs) such as DDT and its metabolites (ΣDDs), γ-HCH (hexachlorocyclohexane), HCB (hexachlorobenzene), aldrin, endrin, isodrin and trans-nonachlor were investigated in wild mussels (Mytilus galloprovincialis) collected from Galician Rías (Rías of Ferrol, A Coruña, Muros, Arousa, Pontevedra and Vigo) during the period 1998-2012. Biological parameters, lipid content, shell length and condition index, were also studied. The OCPs levels in the wild mussel were in the order ΣDDs>γ-HCH>HCB. The other pesticides, aldrin, endrin, isodrin and trans-nonachlor, were not detected or were below the analytical detection limit. All concentrations found in these samples were below the allowable limits for human consumption (Regulation (EC) no. 396/2005). Univariate analysis confirmed that levels of some compounds presented significant relation with biological parameters. Multivariate analysis of the OCPs levels revealed significantly differences between studied Rías, samples from Ría of Ferrol had the highest levels of these compounds (values of ΣDDs ranged from 3.9 to 4.2 ng g(-1) ww) and samples from Ría of Arousa, the lowest levels (values of ΣDDs from 1.3 to 2.4 ng g(-1) ww). PCB and DDT levels were evaluated in populations of endemic Iberian barbel (Barbus bocagei) in the Jarama River in Spain via a pollution gradient from well-preserved areas upstream to contaminated downstream areas. Age structure, abundance, recruitment and levels of morphological abnormalities and ectoparasites were assessed. Upstream to downstream PCB concentrations ranged from 3.4 to 101.4 ng/g (ww) and from 0.9 to 19.6 ng/g ww for DDTs. The PCB pattern was dominated by the PCB 153, 138 and 180 congeners, and the less chlorinated ones had a relatively high contribution upstream. Barbels exposed to low PCB and DDT levels had a well-balanced population with a predominant cohort of young fish, indicating good recruitment. The most contaminated sites displayed a disrupted age distribution, where the proportion of young fish was clearly under-represented. Recruitment and total density of barbel populations decreased downstream where the highest PCB and DDT levels were found. In addition, a higher incidence of abnormalities and ectoparasites was observed at these sites. High concentrations of PCBs and DDTs most likely contribute to the reduction of Iberian barbel reproductive performance in the most contaminated sites, as shown by the disrupted age-distribution found in our study., (Copyright © 2014 Elsevier B.V. Contamination of organohalogen pollutants (OHPs), including dichlorodiphenyl trichloroethane and its metabolites (DDTs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), decabromodiphenylethane (DBDPE), hexabromocyclododecanes (HBCDs), and dechlorane plus (DP) in three metropolises of China, Beijing, Wuhan, and Guangzhou, and a reference rural site were determined using terrestrial residential passerine species as bioindicator. DDTs dominated in Wuhan whereas flame retardants dominated in Guangzhou and Beijing. No geographical variation was found for PCB levels but it exhibited different homologue profiles among different sites which could be attributed to different dietary sources of birds. Industry characteristics of the sampling location contributed to the geographical differences in the occurrence and contamination profile of OHPs. Human serum samples (n = 113) from Bizerte, northern Tunisia, collected between 2011 and 2012 were analyzed for 8 organochlorine pesticides (OCPs) including p,p'-dichlorodiphenyltrichloroethane (DDT) and its metabolites, hexachlorobenzene (HCB), hexachlorocyclohexane isomers, dieldrin, and heptachlor and 12 polychlorinated biphenyls (PCBs) congeners. Concentrations of these residues in serum were determined by gas chromatography with electron capture detector and total cholesterol (CHOL) and triglycerides (TG) levels were evaluated by enzymatic colorimetric method. HCB, p,p'-DDE, PCB-138, PCB-153, and PCB-180, were the most abundant organochlorine compounds (OCs) detected in >95 % of the study subjects. The mean levels of p,p'-DDE and HCB in serum were 168.8 and 49.1 ng g(-1) lipid, respectively. The sum PCBs concentrations ranged from 37.5 to 284.6 ng g(-1) lipid in the samples, with mean and median value of 136.1 and 123.2 ng g(-1) lipid, respectively. The PCB profile consisted of persistent congeners, such as PCB-138, PCB-153, and PCB-180 which contributed for approximately 82.7 % to the ∑PCBs. Statistical analysis showed that most OCs correlated significantly with age, considering all samples together or with gender differentiation. The present study shows that the levels of p,p'-DDE and ∑DDTs were significantly higher in females than in males (p < 0.05), while PCBs levels were significantly higher in male (p < 0.05) than in females. No statistically significant association was found between body mass index and concentration of any organochlorine pesticide or PCB congeners 153, 138, 180, or ∑PCBs.

Analysis of DDT isomers and breakdown products, DDD and DDE, in over 3500 bivalve samples collected from more than 300 locations along the continental United States indicates that concentrations are decreasing. Overall average concentrations for the East (45.8±24.6 ng/gdw), Gulf (42.4±21.1 ng/gdw), and West (90.9±43.3ng/gdw) coasts are declining with an environmental half-life between 10 and 14 ears and are predicted to decrease below 10% of today's concentrations by 2050. Geographically, areas with high and low levels are well identified. Bivalves yielding the highest concentrations were collected in areas linked to areas of DDT production or heavy usage. These areas are clustered in the southern California and San Francisco area, on the West coast; Delaware and Hudson/Raritan Estuary, on the East coast; and in Alabama and northwestern Florida, on the Gulf of Mexico. Prenatal life is the most sensitive stage of human development to environmental pollutants. Early exposure to persistent organic pollutants (POPs) may increase the risk of adverse health effects during childhood. The mechanisms of transference of POPs during pregnancy are still not well understood. The present study is aimed to investigate the transfer of POPs between mother and fetus. The concentrations of 14 organochlorine pesticides, 7 polychlorinated biphenyls (PCBs) and 14 polybromodiphenyl ether (PBDEs) congeners have been measured in 308 maternal serum samples, their respective umbilical cords and 50 placental tissues from a mother-infant cohort representative of Spanish general population. In general, the adjusted lipid-basis concentrations were higher in maternal serum than in cord serum and placenta. The concentrations of most pollutants between maternal serum and cord serum and between maternal serum and placenta were significantly correlated. These distributions were consistent with a predominant maternal source that transfers the pollutants into the placenta and the fetus. However, this distribution did not correspond to passive diffusion of these compounds between these tissues according to lipid content. The compounds more readily metabolized were higher in newborns, which suggest that differences in metabolic capabilities may be responsible of the observed variations in POP distributions between mother and newborns. Prenatal exposure to 4,4'-DDT and some PBDEs such as BDE 99 and BDE 209 is much higher than it could be anticipated from the composition of maternal serum. Analysis of metabolic mechanisms of dichlorodiphenyltrichloroethane (DDT) accumulation and degradation in microorganisms, which could be used to reduce its hazard to higher organisms at the higher in the food chain, have not been investigated. Robust resistance to DDT (grows well in 256 mg/L DDT) and a surprising ability to degrade DDT (more than 70% DDT within 4h) were found in the ciliated protozoan Tetrahymena thermophila. A P450 gene (CYP5013C2) was found to respond specifically to DDT treatment. In the presence of 256 mg/L DDT, cells with overexpressing CYP5013C2 (p450-OE) grew faster and degraded DDT more efficiently than wild-type (WT) cells, while cells with CYP5013C2 partially knocked down (p450-KD) grew slower and exhibited reduced ability to degrade DDT compared to WT cells. Both dichlorodiphenyldichloroethylene (DDE) and dichlorodiphenyldichloroethane (DDD) were detected in cells after exposure to DDT, and the concentration of DDD in the p450-OE strain gradually decreased from 0.5 to 4h. Thus, we argue that this P450 gene (CYP5013C2), by efficiently degrading DDT to DDD, is associated with robust resistance to DDT in Tetrahymena, and that a strain overexpressing this gene has the potential to serve as bioreactor that degrades environmental DDT., (Copyright © 2014 Elsevier B.V. Little data is available on the bioaccumulation of persistent organic pollutants (POPs) in marine organisms from South China Sea (SCS). Five marine fish species were collected from Yongxing Island, SCS to investigate the presence of polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs), dichlorodiphenyltrichloroethane and its metabolites (DDTs). PBDEs, PCBs, and DDTs concentrations ranged from 2.0-117, 6.3-199, and 9.7-5831 ng g(-1) lw, respectively. In general, contaminants measured in this study were at the lower end of the global range. Concentrations of PBDEs and PCBs were significantly correlated in fish samples, implying that PBDEs are as prevalent as PCBs in Yongxing Island. Among the five fish species studied, yellow striped goatfish had the highest concentrations of PBDEs, PCBs, and DDTs, probably attributed to its different living and feeding habits. The contaminant distribution pattern indicated that agrochemical source is more important than industrial source in Yongxing Island, SCS. The average estimated daily intakes of PBDEs, PCBs, and DDTs via fish consumption by local residents in the coastal areas of South China ranged from 1.42-5.91, 3.20-13.3, and 8.08-33.6 ng d(-1), which were lower than those in previous studies, suggesting that consumption of marine fish in Yongxing Island, SCS, might not subject local residents to significant health risk as far as POPs are concerned. Background & Objectives: The enzyme glutathione-s-transferases (GSTs) are associated with detoxification of DDT, as experimentally proved in Anopheles gambiae. Insect GSTs are classified into six classes and among them Delta and Epsilon class GSTs have been implicated in detoxification of organochlorine insecticides. Both Delta and Epsilon GSTs produce, in total, 24 transcripts that result in the production of corresponding enzyme proteins. However, the conventional assay estimates the level of total GSTs and relates to development of resistance to DDT. Hence, it would be more reliable to estimate the level of the specific class GSTs that shows higher affinity with DDT. This would also lead to design a specific molecular tool for resistance diagnosis., Methods: Of the 24 GSTs, computational models for 23 GSTs, which are available in Swiss-Prot database, were retrieved and for the remaining one, D7-2, for which no model is available in the data bank, a structural model was developed using the sequence of An. dirus B with a PDB ID of 1R5A as the template. All the models were docked with DDT in the presence of reduced glutathione., Results: The energy output showed that Delta, D6 has the highest interaction affinity with DDT. Hence, this particular GST (D6) is likely to get elevated on exposure of mosquitoes to DDT., Interpretation & Conclusion: It would be, therefore, possible to design a specific molecular assay to determine the expression level of such high affinity transcript(s) and to use for resistance diagnosis reliably in the vector surveillance programme.

Hydrophobic organic contaminants in soils may pose toxicity or transfer to food chains after their uptake to soil biota. However, uptake data for earthworms are usually limited, as: (a) only fixed exposure times are studied instead of whole uptake kinetics and (b) studies including compounds with different environmental properties and more than two soils of different properties are quite rare. In our study, five persistent organic pollutants (phenanthrene, pyrene, lindane, p,p'-dichlorodiphenyltrichloroethane (p,p'-DDT), and polychlorinated biphenyl congener No. 153 (PCB 153)) were added to six soils of a wide range of soil properties. Detailed kinetics of uptake to earthworms Eisenia fetida were measured. Results indicated that an equilibrium of concentrations for p,p'-DDT and PCB 153 was reached after 11 days of exposure in all soils. Uptake of phenanthrene, pyrene, and lindane was strongly influenced by the decrease in concentrations in the soils, resulting in peak-shaped accumulation curves. Only in soils with the highest total organic carbon content (7.9 and 20.2%), the equilibrium of lindane concentrations was achieved (after 17 and 5 days of exposure, respectively). Organochlorine compounds are highly persistent in the environment, causing bioaccumulation and biomagnification through the marine food chain. To verify the bioaccumulation pattern of DDT and its metabolites, as well as PCBs, Mirex, HCHs and HCB, samples of subcutaneous adipose tissue from 25 Guiana dolphins (Sotalia guianensis), two Fraser's dolphins (Lagenodelphis hosei), two spinner dolphins (Stenella longirostris), one spotted dolphin (Stenella frontalis), and one striped dolphin (Stenella coeruleoalba) were analyzed. The collected specimens were found stranded in northeastern Brazil between 2005 and 2011. The concentrations were reported in μg·g(-1) lip, varying from 0.02 to 17.3 for ΣPCB, from 0.003 to 5.19 for ΣDDT, from 0.005 to 0.16 for ΣHCH, from 0.002 to 0.16 for HCB, and from 0.02 to 2.38 for Mirex. The PCBs dominated the bioaccumulation pattern for both the Guiana and spinner dolphins; for the other species, DDT was the major compound. A discriminant function analysis revealed the differences in the organochlorine accumulation patterns between the Guiana dolphin (S. guianensis) and the continental shelf/oceanic dolphins (S. frontalis and S. longirostris). The results also show that the concentrations of organochlorine compounds found in the delphinids in northeastern Brazil were lower than the delphinids found in other regions of Brazil, as well as other locations worldwide with intense agroindustrial development and/or a larger population., (Copyright © 2013 Elsevier B.V. Improved approaches are needed to assess bioavailability of hydrophobic organic compounds in contaminated soils. Performance of thin-film solid-phase extraction (TF-SPE) using vials coated with ethylene vinyl acetate was compared to earthworm bioassay (Lumbricus terrestris). A DDT and dieldrin contaminated soil was amended with four organic carbon materials to assess the change in bioavailability. Addition of organic carbon significantly lowered bioavailability for all compounds except for 4,4'-DDT. Equilibrium concentrations of compounds in the polymer were correlated with uptake by earthworms after 48d exposure (R(2) = 0.97; p < 0.001), indicating TF-SPE provided an accurate uptake simulation. Bioavailability of residues in soil was compared with a spiked soil aged for 90d in laboratory. Dieldrin and DDX were respectively 18% and 11% less bioavailable in contaminated soil relative to spiked soil despite >40yr of aging. Exposure to contaminants, often pesticides, has been implicated as a major factor contributing to decreases in bat populations. Bats provide essential ecosystem services and a sustained, thriving population is vital for ecosystem health. Understanding issues threatening their survival is crucial for their protection and conservation. This paper provides the first review for 12years on organic pollutants in bats and aims to investigate trends and any new issues impacting bat resilience. Organochlorine (OC) pesticides have been reported most often, especially in the older literature, with the dichlorodiphenyltrichloroethane (DDT) metabolite, dichlorodiphenyldichloroethylene (DDE), present at highest concentrations in tissues analyzed. The OC pesticide concentrations reported in bat tissues have declined significantly since the late 1970s, presumably as a result of restrictions in use. For example, DDE study mean concentrations over time periods 1970-1980, 1981-1999 and 2000-2013 ranged from 2.6-62, 0.05-2.31, 0.08-0.19ppm wet weight, respectively. Exposure, however, still occurs from remaining residues, many years after the compounds have been actively used. In recent years (2000-2013), a range of other organic chemicals have been reported in bat tissues including brominated flame retardants (polybrominated diphenyl ether at a mean concentration of 2.9ppm lipid weight) and perfluorinated compounds (perfluorooctanyl sulfonate at a mean concentration 0.09ppm wet weight). The persistent organic compounds concentrate in tissues with higher fat content notably back-depot fat. Numerous factors influence exposure, residues detected and concentrations in different individuals, species and tissues which must be understood to provide meaningful assessment of the impacts of exposure. Exposure can lead to not only acute and lethal impacts, but also physiological sub-lethal and chronic effects, often linked to the annual cycle of fat deposition and withdrawal. Current challenges for bat conservation include collation of a more extensive and standardized database of bat exposure, especially to current use pesticides and emerging contaminants, and better prediction and definition of toxicity end points notably for the sub-lethal effects. Persistent organic pollutants (POPs) levels in resident and migratory birds collected from an urbanized coastal region of South Korea were investigated. As target species, resident birds that reside in different habitats-such as inland and coastal regions-were selected and their POP contamination status and accumulation features evaluated. Additionally, winter and summer migratory species were analysed for comparison with resident birds. Black-tailed gull and domestic pigeon were selected as the coastal and inland resident birds, respectively, and pacific loon and heron/egret were selected as the winter and summer migratory birds, respectively. The overall POP concentrations (unit: ng/g lipid) in resident birds were 14-131,000 (median: 13,400) for PCBs, 40-284,000 (11,200) for DDTs, <1.0-2850 (275) for CHLs, 23-2020 (406) for HCHs, 2-1520 (261) for HCB, <0.2-48 (5) for pentachlorobenzene (PeCB), 71-7120 (1840) for PBDEs, and <1.8-2300 (408) for HBCDs. In resident birds, the overall level of POPs was higher in seagull compared to pigeon. The stable isotope ratio of nitrogen and carbon indicates that seagull occupies a higher trophic position in the environment than pigeon. However, the POP accumulation profiles in these species differed. Pigeon tends to accumulate more recently used POPs such as PBDEs than seagull. The high-brominated BDE congeners, γ-HBCDs and γ-HCH (also called lindane) were enriched in pigeon compared to seagull, implying the widespread use of Deca-BDE, technical HBCDs, and lindane in the terrestrial environment of South Korea. The different accumulation profile of POPs in both resident species would be related to their habitat difference and trophic positions. For urban resident bird such as pigeon, an intentional intake of dust or soils during feeding is likely to be an additional route of exposure to POPs. Increasing evidence supports that harmful chemicals accumulating in the human body may pose a significant threat to infant health through foetal exposure. Persistent organic pollutants (POPs) are thought to enhance the risk for later development of allergic disease like atopic dermatitis (AD). However, few studies have evaluated the effect of foetal exposure to various POPs on the development of AD in early infancy. Here, we describe the impact of foetal exposure to a number of POPs on the occurrence of AD in 7-month-old infants. The participants were 81 infants with or without AD who participated in a birth cohort study, where the concentrations of 15 polychlorinated biphenyl (PCBs) congeners, dichlorodiphenyltrichloroethane (p,p'-DDT), dichlorodiphenyldichloroethylene (p,p'-DDE), β-hexachlorocyclohexane (β-HCH), hexachlorobenzene (HCB), cis-nonachlor, trans-nonachlor, mirex, oxychlordane, and 27 polybrominated diphenyl ether (PBDEs) congeners were measured in the umbilical cord tissues collected immediately after birth. At 7 months, 27 of the 81 infants (33.8%) were diagnosed with AD. Of all POPs examined, total concentrations of 27 PBDE congeners were associated with a significantly decreased incidence of AD. Notably, the concentration of 27 PBDEs was significantly lower in AD infants than in non-AD infants (P<0.01), and the risk of AD development decreased with increasing PBDE levels. In an attempt to explore complex metabolic interactions between toxicants present in polluted freshwater, hepatic metabolism of benzo(a)pyrene (BaP) and tributyltin (TBT) in fish was investigated when these compounds were administrated alone, mixed together and along with dichlorodiphenyltrichloroethane (DDT). Ten Rhamdia quelen per group were treated with a single intra-peritoneal (IP) dose (5-day experiment) or three successive doses (15-day experiment) either containing BaP (0.3; 3 or 30mgkg(-1)) or TBT (0.03; 0.3 or 3mgkg(-1)) or a combination of BaP+TBT, BaP+DDT, TBT+DDT and BaP+TBT+DDT under their respective lower doses, with DDT dose kept at 0.03mgkg(-1). Tetrahydroxy-benzo(a)pyrene (BaP-tetrol-I), and dibutyltin (DBT) and monobutyltin (MBT) were analyzed to assess BaP and TBT hepatic metabolism, respectively. A significant difference in BaP-tetrol-I concentration was observed in liver and bile between the lowest and the highest doses of BaP in both 5 and 15-day experiments. In the 15-day experiment, the presence of TBT with BaP reduced the amount of BaP-tetrol-I in bile compared to the BaP alone. The time of exposure and the number of doses affected BaP-tetrol-I concentration in the bile of fish exposed to BaP 0.3mgkg(-1) and BaP+DDT. TBT and its metabolites concentrations showed a dose-dependent increase in the liver in both experiments and in the bile in the 5-day experiment. TBT at its lowest dose was completely metabolized into DBT and MBT in the liver in the 15-day experiment. No TBT metabolites were detected in the bile of fish exposed to the mixtures in the 5-day experiment, except for a small MBT amount found in BaP+TBT+DDT. The tissue and organs (muscle, brain, liver, and gill) of four species of freshwater fish from Lake Baiyangdian were analyzed for hexachlorocyclohexanes (HCHs) and dichloro-diphenyl-trichloroethanes (DDTs). The distribution characteristics were analyzed for HCHs and DDTs in various tissue and organs, which determined the health risks for humans. The research results showed that the wet weight content of all HCHs (∑HCHs) ranged from 0.05 ∼ 14.53 ng g(-1), with a mean of 3.47 ng g(-1). The wet weight content of all DDTs (∑DDTs) ranged from ND to 8.51 ng g(-1), with a mean of 2.41 ng g(-1). For the various species of fish, the residual level of ∑HCHs was relatively higher in chub and grass carp and lowest in snakehead. The residual level of ∑DDTs was the highest in snakehead and did not exhibit a significant variance in the other three species. For the various tissues and organs, the contents of HCHs and DDTs were both highest in the fish liver, second highest in the fish gill, and lowest in the fish brain and muscle. Among the four types of isomers, the residual level of γ-HCH was relatively higher, while the residual level of α-HCH was the lowest. The content of p,p'-DDE was significantly greater to other forms of DDT and its isomer. The residual levels of HCHs and DDTs in fish were both below the national standard. However, the carcinogenic risk from the HCHs in parts of the tissue and organs of four fish species in Lake Baiyangdian exceeded the screen value threshold set by USEPA.

Organic contaminant and total mercury concentrations were compared in four species of lamniform sharks over several age classes to examine bioaccumulation patterns and gain insights into trophic ecology. Contaminants found in young of the year (YOY) sharks were assumed to be derived from maternal sources and used as a proxy to investigate factors that influence maternal offloading processes. YOY white (Carcharodon carcharias) and mako (Isurus oxyrinchus) sharks had comparable and significantly higher concentrations of PCBs, DDTs, pesticides, and mercury than YOY thresher (Alopias vulpinus) or salmon (Lamna ditropis) sharks. A significant positive relationship was found between YOY contaminant loads and maternal trophic position, suggesting that trophic ecology is one factor that plays an important role in maternal offloading. Differences in organic contaminant signatures and contaminant concentration magnitudes among species corroborated what is known about species habitat use and may be used to provide insights into the feeding ecology of these animals., (Copyright © 2013 The Authors. The concentrations and biomagnifications of dichlorodiphenyltrichloroethane (DDT) and its metabolites were examined in four fish species (Clarias gariepinus, Oreochromis niloticus, Tilapia zillii, and Carassius auratus) from Lake Ziway, Rift Valley, Ethiopia. Paired stomach content analysis, and stable isotope ratio of nitrogen (δ(15)N, ‰) and carbon (δ(13)C, ‰) were used to study the trophic position of the fish species in the lake. 4,4'-DDE, 4,4'-DDT and 4,4'-DDD were the main DDTs identified in the fish samples, with 4,4'-DDE as the most predominant metabolite, with mean concentration ranging from 1.4 to 17.8 ng g(-1) wet weight (ww). The concentrations of DDTs found in fish from Lake Ziway were, in general lower than those found in most studies carried out in other African Lakes. However, the presence of DDT in all tissue samples collected from all fish species in the lake indicates the magnitude of the incidence. Moreover, the observed mean 4,4'-DDE to 4,4'-DDT ratio below 1 in C. auratus from Lake Ziway may suggest a recent exposure of these species to DDT, indicating that a contamination source is still present. 4,4'-DDE was found to biomagnify in the fish species of the lake, and increases with trophic level, however, the biomagnification rate was generally lower than what has been reported from other areas. Solid-phase microextraction (SPME) has often been used to estimate the freely dissolved concentration (Cfree ) of organic contaminants in sediments. A significant limitation in the application of SPME for Cfree measurement is the requirement for attaining equilibrium partition, which is often difficult for strongly hydrophobic compounds such as DDT. A method was developed using SPME with stable isotope-labeled analogues as performance reference compounds (PRCs) to measure Cfree of DDT and metabolites (DDTs) in marine sediments. Six (13) C-labeled or deuterated PRCs were impregnated into polydimethylsiloxane (PDMS) fiber before use. Desorption of PRCs from PDMS fibers and absorption of DDTs from sediment were isotropic in a range of sediments evaluated ex situ under well-mixed conditions. When applied to a historically contaminated marine sediment from a Superfund site, the PRC-SPME method yielded Cfree values identical to those found by using a conventional equilibrium SPME approach (Eq-SPME), whereas the time for mixing was reduced from 9 d to only 9 h. The PRC-SPME method was further evaluated against bioaccumulation of DDTs by Neanthes arenaceodentata in the contaminated sediment with or without amendment of activated carbon or sand. Strong correlations were consistently found between the derived equilibrium concentrations on the fiber and lipid-normalized tissue residues for DDTs in the worms. Results from the present study clearly demonstrated the feasibility of coupling PRCs with SPME sampling to greatly shorten sampling time, thus affording much improved flexibility in the use of SPME for bioavailability evaluation., (Copyright © 2013 SETAC.)

The present study aimed to evaluate the occurrence of persistent organic pollutants (POPs: (PCBs, PBDEs, DDTs, HCHs, CHLs and HCB) in sediments and biota from the middle Congo River Basin (CRB) and to investigate their trophic transfer through the aquatic food web using nitrogen stable isotope ratios. To our knowledge, no data on levels of POPs in sediment and biota from the CRB are present in the literature, and studies on trophic transfer and biomagnification profiles of POPs using δ(15)N are scarce in tropical regions. POP levels in the sediment and biota were low, with exception of total PCB levels found in fish from the Itimbiri River (1.4 to 44ng/g ww). Compared to concentrations found in fish from pristine to relatively industrial developed areas, the ∑PCB levels in fish from the Itimbiri were high, indicating the presence of a local PCB contamination source in this catchment. Based on minimum risk level criteria formulated by ATSDR, the consumption of PCB contaminated fish from the Itimbiri river poses a potential risk for humans. The POP levels in biota were not significantly related to the POP levels in sediments, and the BSAF concept (Biota-Sediment Accumulation Factor) was found to be a poor predictor of the bioavailability and bioaccumulation of environmental pollutants in the present study. With increasing trophic levels, a significant increase in PCB 95, 101, 110, 138, 146, 149, 153, 174, 180 & 187 and p,p'-DDT in Itimbiri and BDE 47 & 99 in Itimbiri, Aruwimi & Lomami river basins was observed. In situ measurements of hydrophobic organic chemicals in sediment porewater, a central component in assessing the bioavailability and mobility of chemicals in sediment, have been scarce. Here, we introduce a multisection passive sampler with low-density polyethylene (LDPE) as the sorbent phase, which is appropriate for measuring vertical concentration profiles of chemicals in sediment porewater. This sampler is composed of a series of identical sampling cells insulated with seclusion rings. In each section, sorption of chemicals into LDPE is diffusion-controlled through the water layer separated from the sediment by a glass fiber filtration membrane and a porous stainless steel shield. Pilot laboratory testing indicated that the sampler can roughly determine the porewater concentrations of 1,1-dichloro-2,2-bis-(chlorophenyl)ethane (p,p'-DDD) and 1,1-dichloro-2,2-bis-(chlorophenyl)ethylene (p,p'-DDE), comparable to those yielded through centrifugation/liquid-liquid extraction, a conventional technique for sampling sediment porewater. Field deployment of the sampler was performed in an urbanized coastal region to measure the depth profiles of dichlorodiphenyltrichloroethane and its metabolites in sediment porewater. Sampling rate-calibrated and performance reference compound-calibrated concentrations were calculated, which were consistent with those obtained by the centrifugation/liquid-liquid extraction method. These results verified the utility of the sampler for measuring depth profiles of sediment porewater chemicals.
