101. Progressive cavitating leukoencephalopathy: A novel childhood disease

Abstract

We report 19 patients with a previously undelineated neurodegenerative syndrome characterized by episodic acute onset of irritability or neurological deficits between 2 months and 3.5 years of age, followed by steady or intermittent clinical deterioration. Seven children died between 11 months and 14 years of age. Cranial magnetic resonance imaging (MRI) shows patchy leukoencephalopathy with cavities, and vascular permeability, in actively affected regions. Early lesions affect corpus callosum and centrum semiovale, with or without cerebellar or cord involvement. After repeated episodes, areas of tissue loss coalesce with older lesions to become larger cystic regions in brain or spinal cord. Diffuse spasticity, dementia, vegetative state, or death ensues. Gray matter is spared until late in the course. In some, incomplete clinical or MRI recovery occurs after episodes. The clinical course varies from rapid deterioration to prolonged periods of stability that are unpredictable by clinical or MRI changes. Elevated levels of lactate in brain, blood, and cerebrospinal fluid, abnormal urine organic acids, and changes in muscle respiratory chain enzymes are present but inconsistent, without identifiable mitochondrial DNA mutations or deletions. Pathological studies show severe loss of myelin sparing U-fibers, axonal disruption, and cavitary lesions without inflammation. Familial occurrence and consanguinity suggest autosomal recessive inheritance of this distinct entity.

Abstract

A novel G8363A mutation in the mtDNA tRNA(Lys) gene was associated, in two unrelated families, with a syndrome consisting of encephalomyopathy, sensorineural hearing loss, and hypertrophic cardiomyopathy. Muscle biopsies from the probands showed mitochondrial proliferation and partial defects of complexes I, III, and IV of the electron-transport chain. The G8363A mutation was very abundant (>95%) in muscle samples from the probands and was less copious in blood from 18 maternal relatives (mean 81.3% +/- 8.5%). Single-muscle-fiber analysis showed significantly higher levels of mutant genomes in cytochrome (c) oxidase-negative fibers than in cytochrome (c) oxidase-positive fibers. The mutation was not found in >200 individuals, including normal controls and patients with other mitochondrial encephalomyopathies, thus fulfilling accepted criteria for pathogenicity.

G.B., D.C.K., and S.M.F. acknowledge support from NSF grant AST-08-08133 and NASA grant HST-GO-12060.10A., We present a catalog of visual-like H-band morphologies of ~50.000 galaxies (H_f160w < 24.5) in the 5 CANDELS fields (GOODS-N, GOODS-S, UDS, EGS, and COSMOS). Morphologies are estimated using Convolutional Neural Networks (ConvNets). The median redshift of the sample is < z> 1.25. The algorithm is trained on GOODS-S, for which visual classifications are publicly available, and then applied to the other 4 fields. Following the CANDELS main morphology classification scheme, our model retrieves for each galaxy the probabilities of having a spheroid or a disk, presenting an irregularity, being compact or a point source, and being unclassifiable. ConvNets are able to predict the fractions of votes given to a galaxy image with zero bias and ~10% scatter. The fraction of mis-classifications is less than 1%. This work is based on observations taken by the 3D-HST Treasury Program (GO 12177 and 1232) with the NASA/ESA HST, which is operated by the Association of Universities for Research in Astronomy, Inc, under NASA contract NAS5-26555., We quantify the morphological evolution of z ~ 0 massive galaxies (M_*/M_∼ ∼10^11.2 +/- 0.3) from z ~ 3 in the 5 CANDELS fields. The progenitors are selected using abundance matching techniques to account for the mass growth. At z < 1, the population matches the massive end of the Hubble sequence, with 30% of pure spheroids, 50% of galaxies with equally dominant disk and bulge components, and 20% of disks. At z ~ 2-3 however, there is a majority of irregular systems (~ 60%-70%) with still 30% of pure spheroids. We then analyze the stellar populations, star formation rates (SFRs), gas fractions, and structural properties for the different morphologies independently. Our results suggest two distinct channels for the growth of bulges in massive galaxies. Around ~ 30%-40% were already bulges at z ~ 2.5, with low average SFRs and gas fractions (10%-15%), high Sersic indices (n > 3-4), and small effective radii (R_e ∼1 kpc), pointing toward an even earlier formation through gas-rich mergers or violent disk instabilities. Between z ~ 2.5 and z ~ 0, they rapidly increase their size by a factor of ~ 4-5, are quenched, and slightly increase their Sersic indices (n ~ 5) but their global morphology remains unaltered. The structural evolution is independent of the gas fractions, suggesting that it is driven by ex situ events. The remaining 60% experience a gradual morphological transformation, from clumpy disks to more regular bulge +disk systems, essentially happening at z > 1. This results in the growth of a significant bulge component (n ~ 3) for 2/3 of the systems, possibly through the migration of clumps, while the remaining 1/3 retain a rather small bulge (n ~ 1.5-2). Thanks to the anonymous referee for a constructive and quick report., We quantify the evolution of the stellar mass functions (SMFs) of star-forming and quiescent galaxies as a function of morphology from z ∼ 3 to the present. Our sample consists of ∼50 000 galaxies in the CANDELS fields (∼880 arcmin^2), which we divide into four main morphological types, i.e. pure bulge-dominated systems, pure spiral disc-dominated, intermediate two-component bulge+disc systems and irregular disturbed galaxies. At z ∼ 2, 80 per cent of the stellar mass density of star-forming galaxies is in irregular systems. However, by z ∼ 0.5, irregular objects only dominate at stellar masses below 10^9 M_⊙. A majority of the star-forming irregulars present at z ∼ 2 undergo a gradual transformation from disturbed to normal spiral disc morphologies by z ∼ 1 without significant interruption to their star formation. Rejuvenation after a quenching event does not seem to be common except perhaps for the most massive objects, because the fraction of bulge-dominated star-forming galaxies with M^*/M_⊙ > 10^10.7 reaches 40 per cent at z < 1. Quenching implies the presence of a bulge: the abundance of massive red discs is negligible at all redshifts over 2 dex in stellar mass. However, the dominant quenching mechanism evolves. At z > 2, the SMF of quiescent galaxies above M^* is dominated by compact spheroids. Quenching at this early epoch destroys the disc and produces a compact remnant unless the star-forming progenitors at even higher redshifts are significantly more dense. At 1 < z < 2, the majority of newly quenched galaxies are discs with a significant central bulge. This suggests that mass quenching at this epoch starts from the inner parts and preserves the disc. At z < 1, the high-mass end of the passive SMF is globally in place and the evolution mostly happens at stellar masses below 10^10 M_⊙. Abstract

The European Space Agency's Euclid mission will provide high-quality imaging for about 1.5 billion galaxies. A software pipeline to automatically process and analyse such a huge amount of data in real time is being developed by the Science Ground Segment of the Euclid Consortium; this pipeline will include a model-fitting algorithm, which will provide photometric and morphological estimates of paramount importance for the core science goals of the mission and for legacy science. The Euclid Morphology Challenge is a comparative investigation of the performance of five model-fitting software packages on simulated Euclid data, aimed at providing the baseline to identify the best-suited algorithm to be implemented in the pipeline. In this paper we describe the simulated dataset, and we discuss the photometry results. A companion paper is focussed on the structural and morphological estimates. We created mock Euclid images simulating five fields of view of 0.48 deg2 each in the IE band of the VIS instrument, containing a total of about one and a half million galaxies (of which 350 000 have a nominal signal-to-noise ratio above 5), each with three realisations of galaxy profiles (single and double Sérsic, and 'realistic' profiles obtained with a neural network); for one of the fields in the double Sérsic realisation, we also simulated images for the three near-infrared YE, JE, and HE bands of the NISP-P instrument, and five Rubin/LSST optical complementary bands (u, g, r, i, and z), which together form a typical dataset for an Euclid observation. The images were simulated at the expected Euclid Wide Survey depths. Abstract

The European Space Agency's Euclid mission will provide high-quality imaging for about 1.5 billion galaxies. A software pipeline to automatically process and analyse such a huge amount of data in real time is being developed by the Science Ground Segment of the Euclid Consortium; this pipeline will include a model-fitting algorithm, which will provide photometric and morphological estimates of paramount importance for the core science goals of the mission and for legacy science. The Euclid Morphology Challenge is a comparative investigation of the performance of five model-fitting software packages on simulated Euclid data, aimed at providing the baseline to identify the best-suited algorithm to be implemented in the pipeline. In this paper we describe the simulated dataset, and we discuss the photometry results. A companion paper is focussed on the structural and morphological estimates. The Euclid Morphology Challenge is a comparative investigation of the performance of five model-fitting software packages on simulated Euclid data, aimed at providing the baseline to identify the best suited algorithm to be implemented in the pipeline. In this paper we describe the simulated data set, and we discuss the photometry results. A companion paper (Euclid Collaboration: Bretonni\`ere et al. 2022) is focused on the structural and morphological estimates. We created mock Euclid images simulating five fields of view of 0.48 deg2 each in the $I_E$ band of the VIS instrument, each with three realisations of galaxy profiles (single and double S\'ersic, and 'realistic' profiles obtained with a neural network); for one of the fields in the double S\'ersic realisation, we also simulated images for the three near-infrared $Y_E$, $J_E$ and $H_E$ bands of the NISP-P instrument, and five Rubin/LSST optical complementary bands ($u$, $g$, $r$, $i$, and $z$). To analyse the results we created diagnostic plots and defined ad-hoc metrics. Five model-fitting software packages (DeepLeGATo, Galapagos-2, Morfometryka, ProFit, and SourceXtractor++) were compared, all typically providing good results. (cut), Comment: 29 pages, 33 figures. 116. A dual-team approach benefits standard-volume surgeons, but has minimal impact on outcomes for a high-volume surgeon in AIS patients

Abstract

Study design: Retrospective chart review of prospectively collected data. Objective: This study seeks to evaluate the effect of number of surgeons, surgeon experience, and surgeon volume on AIS surgery. Summary of background data: Recent literature suggests that utilizing two surgeons for spine deformity correction surgery can improve perioperative outcomes. However, the surgeon’s experience and surgical volume are likely as important. Methods: AIS patients undergoing PSF from 2009 to 2019 were included. Patient demographics, X-ray and perioperative outcomes were collected and collated based on primary surgeon. Analysis was performed for single versus dual surgeons, surgeon experience (≤ 10 years in practice), and surgical volume (less/greater than 50 cases/year). Median (IQR) values, Wilcoxon Rank Sums test, Kruskal–Wallis test, and Fisher’s exact test were utilized. Results: 519 AIS cases, performed by 4 surgeons were included. Two surgeons were highly experienced, 1 of whom was also high volume. Five cohorts were studied: a single senior high volume (S1) (n= 302), dual-junior surgeons (DJ) (n= 73), dual senior–junior (SJ) (n= 36), dual-senior (DS) (n= 21) and a single senior, standard-volume surgeon alone (S2) (n= 87). Radiographic parameters were similar between the groups (p> 0.05). Preoperative Cobb was significantly higher for DS compared to S1 (p= 0.034) Pre- and post-op kyphosis were similar (p> 0.05). Cobb correction was similar (p> 0.05). Levels fused, fixation points, anesthesia and surgical times were similar (p> 0.05). When the standard-volume surgeon operated with a second surgeon, radiographic parameters were similar (p> 0.05), but anesthesia time, surgical time, and hospital length of stay were significantly shorter (p&lt; 0.05). Additionally, DJ had significantly shorter anesthesia and operative times (p&lt; 0.001) and length of stay (p&lt; 0.001) compared to S2. Conclusion: Standard-volume surgeons have better outcomes with a dual surgeon approach. Junior surgeons benefit operating with an experienced surgeon. A high-volume surgeon, however, does not benefit from a dual surgeon approach. Level of evidence: Level II.

Abstract

Dysregulation of the networking of RNA-binding proteins (RBPs) and RNAs drives many human diseases, including cancers, and the targeting of RNA–protein interactions (RPIs) has emerged as an exciting area of RNA-targeted drug discovery. Accordingly, methods that enable the discovery of cell-active small molecule modulators of RPIs are needed to propel this emerging field forward. Herein, we describe the application of live-cell assay technology, RNA interaction with protein-mediated complementation assay (RiPCA), for high-throughput screening to identify small molecule inhibitors of the pre-let-7d–Lin28A RPI. Utilizing a combination of RNA-biased small molecules and virtual screening hits, we discovered an RNA-binding small molecule that can disrupt the pre-let-7–Lin28 interaction demonstrating the potential of RiPCA for advancing RPI-targeted drug discovery.

118. Historical Perspective on Mitochondrial Medicine

Abstract

In this review, we trace the origins and follow the development of mitochondrial medicine from the premolecular era (1962-1988) based on clinical clues, muscle morphology, and biochemistry into the molecular era that started in 1988 and is still advancing at a brisk pace. We have tried to stress conceptual advances, such as endosymbiosis, uniparental inheritance, intergenomic signaling and its defects, and mitochondrial dynamics. We hope that this historical review also provides an update on mitochondrial medicine, although we fully realize that the speed of progress in this area makes any such endeavor akin to writing on water.

119. Student-Designed Enzyme-Linked Metabolite Assay Kits

Abstract

The extensive use of commercial kits in molecular biology and biochemistry has prompted us to design a series of practical sessions to help students become familiar with the uses and limitations of pre-packaged assay systems. To facilitate an understanding of these assay systems and to promote reflection on their appropriate use, students manufacture their own kit for one of four enzyme-linked metabolite assays. To do this they must investigate the role of each of the components in the assay, optimize the conditions where possible, check for cross reactivity, and then work this assay up for a few related "real" samples. Students make up all the buffers, the standard solution, instructions, and even the packaging. The kit is checked for accuracy by the producers, "marketed" to other students, and evaluated by their peers. By going through this process, students learn the benefits and pitfalls of commercial kits as well as reinforcing the basic principles of metabolite measurement and gaining experience with assay design, troubleshooting, and problem solving.

Abstract

Genetic mutation of the leucine-rich repeat kinase 2 (LRRK2) protein has been associated with Parkinson’s disease (PD), a disabling and progressive neurodegenerative disorder that is devoid of efficacious disease-modifying therapies. Herein, we describe the invention of an amidoisoquinoline (IQ)-derived LRRK2 inhibitor lead chemical series. Knowledge-, structure-, and property-based drug design in concert with rigorous application of in silicocalculations and presynthesis predictions enabled the prioritization of molecules with favorable CNS “drug-like” physicochemical properties. This resulted in the discovery of compound 8, which was profiled extensively before human ether-a-go-go (hERG) ion channel inhibition halted its progression. Strategic reduction of lipophilicity and basicity resulted in attenuation of hERG ion channel inhibition while maintaining a favorable CNS efflux transporter profile. Further structure- and property-based optimizations resulted in the discovery of preclinical candidate MK-1468. This exquisitely selective LRRK2 inhibitor has a projected human dose of 48 mg BID and a preclinical safety profile that supported advancement toward GLP toxicology studies.
