Abstract

Adult progeria, Werner syndrome (WS), is an autosomal recessive disorder that develops accelerated aging-associated symptoms after puberty. Refractory skin ulcer of limbs, which is one of the symptoms specific to WS, is seriously painful and sometimes results in amputation. In recent years, cell therapy using mesenchymal stem cells (MSCs) has been attracting attention; however, the effect of WS-derived MSCs on skin ulcers is still unclear. In this study, we generated iPS cells from a patient with WS and a normal subject, differentiated them into MSCs (WS- and NM-iMSC, respectively), and performed cell therapy to a refractory skin ulcer mouse model. As a result, WS-iMSC recapitulated premature senescence phenotypes in vitro. Upon subcutaneous injection around the wounds of mice, WS-iMSC was significantly inferior in wound healing effect compared to NM-iMSC. Proteome and transcriptome analysis revealed altered expression of genes related to angiogenesis, inflammation, and proliferation in WS-iMSC with remarkable downregulation of VEGF, a potent angiogenic factor. In addition, simultaneous administration of recombinant human VEGF and WS-iMSC improved the wound healing effect in vivo. These results indicate that the expression of angiogenic factors is reduced in WS-iMSC, and its supplementation restores the wound healing ability. This finding may pave the way to develop the treatment of intractable skin ulcers of WS.

120. Health Monitoring Tests of Heritage Structures: Application of MEMS Accelerometers to Two Multi Tier Pagodas

Abstract

The paper introduces a health monitoring system based on MEMS (micro electromechanical systems) accelerometers. The used MEMS accelerometers were recently developed by one of the authors. They excel in cost efficiency, calibration accuracy and robustness. The health monitoring system was installed in two multi-tiered pagodas. They were a two-tiered pagoda, Pashupatinath temple and a five-tiered pagoda, Nyatapola temple for four months. Both pagodas are located in Nepal. In both cases, a number of earthquakes were captured by the installed accelerometers, as it permitted estimation of the fundamental mode of the tested pagodas. The paper deals with two tasks. The first task introduces the used MEMS accelerometers. The features of the accelerometers are discussed. The second task discusses the dynamic behaviour of the pagodas identified by the installed monitoring system. The paper presents adequacy of the developed MEMS accelerometers for health monitoring test of heritage structures.

Abstract

This study investigated the effectiveness of treatment with Janus kinase (JAK) inhibitors in rheumatoid arthritis (RA) assessed by ultrasonography (US) activity, and the influence of patient characteristics and previous treatments. This prospective study assessed 60 treatment initiations among 53 Japanese patients diagnosed with RA who underwent treatment with JAK inhibitors during June 2013 to February 2020. Of the 53 patients, seven patients were enrolled in duplicate because they were treated with two different JAK inhibitors at different periods. For each case, the improvement rate on the power Doppler (PD) score was assessed at 6 month follow-up. Median improvement rate of PD score was used to classify cases as either US responders or non-responders, and patient characteristics were compared between the two groups. All indicators of clinical disease activity and US activity showed a significant improvement at 3 months compared with baseline. Although the JAK inhibitor-cycler group and the interleukin-6 (IL-6) inhibitor inadequate response (IR) group tended to show a later improvement for US activity, all indicators of clinical disease activity and US activity showed a significant improvement at 6 months compared with baseline for both groups. Multivariate analysis showed that concomitant methotrexate use and an IR to the previous biologic or targeted-synthetic disease-modifying anti-rheumatic drug (b/tsDMARD) treatment were independently and significantly associated with US responders. Use of a JAK inhibitor in combination with methotrexate and an absence of IR to any previous b/tsDMARDs demonstrated superior effectiveness for patients with RA. [ABSTRACT FROM AUTHOR]

Abstract

The ability of skeletal muscle to regenerate declines significantly with aging. The expression of aryl hydrocarbon receptor nuclear translocator (ARNT), a critical component of the hypoxia signaling pathway, was less abundant in skeletal muscle of old (23-25 months old) mice. This loss of ARNT was associated with decreased levels of Notch1 intracellular domain (N1ICD) and impaired regenerative response to injury in comparison to young (2-3 months old) mice. Knockdown of ARNT in a primary muscle cell line impaired differentiation in vitro. Skeletal muscle-specific ARNT deletion in young mice resulted in decreased levels of whole muscle N1ICD and limited muscle regeneration. Administration of a systemic hypoxia pathway activator (ML228), which simulates the actions of ARNT, rescued skeletal muscle regeneration in both old and ARNT-deleted mice. These results suggest that the loss of ARNT in skeletal muscle is partially responsible for diminished myogenic potential in aging and activation of hypoxia signaling holds promise for rescuing regenerative activity in old muscle.

Abstract

We have investigated the detailed magnetization reversal in 10-nm-thick Ni--Fe elliptical dots with several dot sizes using magnetic field sweeping (MFS)-magnetic force microscopy (MFM). At the dot edges, the shape of the phase (the stray field) curve versus magnetic field changes from a stepped hysteresis loop to a almost nonstepped hysteresis loop with increasing dot size. On the other hand, at the center within the dot, sharp or weak decreases in phase are observed as the magnetic field is varied. From these results, it is found that, in the magnetization reversal of a 10-nm-thick Ni--Fe elliptical dot with several dot sizes, the dominant factor changes from the change in the magnetic domain configuration to the domain wall motion as the length of major axis increases. Abstract

We have studied the effect of the dot separation on the switching behavior of Ni-Fe elliptical dot arrays with major axial distances between the adjacent dots. The magnetization process and the magnetic configuration in the dot arrays apparently differ according to the distance between the adjacent dots. These differences are due to the fact that the dipole-dipole interaction becomes stronger while decreasing the major axial distance between the adjacent dots. From these results, it is evident that the optimal distance at which the magnetization between the adjacent dots couples by the dipole-dipole interaction is below 30 nm.
