Abstract Introduction Alopecia areata (AA) is an autoimmune disease associated with high rates of emotional and psychosocial distress. The analysis reported here describes the evolution of measures assessing health-related quality of life (HRQoL) and symptoms of anxiety and depression up to week 104 in patients who achieved sustained scalp hair regrowth during treatment with baricitinib in the BRAVE-AA phase III trials. Methods This post-hoc analysis included data from the double-blind, parallel-group, randomized, placebo-controlled phase III trials BRAVE-AA1 (ClinicalTrials.gov number: NCT03570749) and BRAVE-AA2 (ClinicalTrials.gov number: NCT03899259). Adults with severe AA (defined as a Severity of Alopecia Tool [SALT] score ≥ 50) randomized to baricitinib 4 mg or baricitinib 2 mg at baseline who achieved SALT score ≤ 20 by week 36 and maintained SALT score ≤ 20 through week 104 on the same dose of baricitinib were included in this analysis of integrated data. Scalp hair regrowth (SALT score) and improvements in Skindex-16 AA Scale and Hospital Anxiety and Depression Scale (HADS) domain scores were analyzed over the 104-week period using descriptive statistics. Results In total, 131 patients (88 on baricitinib 4 mg and 43 on baricitinib 2 mg) were included in this analysis. Across the two groups, the mean age (standard deviation) was 37.2 years (12.7), and 84 (64.1%) patients were female. The interquartile range) for time to achieve a SALT score ≤ 20 for patients treated with baricitinib 4 mg and baricitinib 2 mg was 13.1 and 19.6 weeks, respectively. By week 104, 91% (baricitinib 2 mg) and 96% (baricitinib 4 mg) of patients had achieved a SALT score ≤ 10 on baricitinib treatment. In both groups, progressive improvements in the Skindex-16 AA and HADS domain scores were observed up to week 104. Conclusion This analysis of adults with severe AA treated with baricitinib revealed that achievement of sustained clinically meaningful scalp hair regrowth (SALT score ≤ 20) was associated with improvements in both measures of HRQoL and symptoms of anxiety and depression up to week 104.

Por medio de un análisis con perspectiva empresarial, el presente artículo analiza el auge que expe­rimentó la industria de producción cinematográfica mexicana a inicios de la década de 1930. Se propone que el auge debe ser entendido dentro del contexto de la transición internacional al cine sonoro y de efervescencia nacionalista en México. Del mismo modo, se propone que el auge fue resultado del aprovechamiento de dichos contextos por parte de la Compañía Nacional Productora de Películas, así como el entusiasmo generado por su cinta fundacional, Santa (Moreno, 1932). Los estudios sobre la industria cinematográfica mexicana han enfati­zado el papel de los elementos estéticos y sociales para entender el desarrollo del cine mexicano, sin embargo, el presente estudio busca contribuir a esta historiografía incorporando elementos microeconómicos, así como herramientas de análisis de la literatura sobre formación de industrias.

Toxoplasmosis is an infection caused by the parasite Toxoplasma gondii. One-third of the world’s population has come into contact with this parasite. In Mexico, the prevalence is between 15% and 50% in the general population and 34.9% in women with high-risk pregnancies. In pregnancy, the highest incidence of infection occurs in the third trimester and fetal damage is inversely proportional to gestational age. Maternal hormones play a fundamental role in the immune response. There are very few studies, with controversial results, on the levels of increased hormones and their relationship to the kinetics of T. gondii infections during pregnancy. The aim was to determine the serum levels of 17-β estradiol, prolactin, and progesterone, and their association with anti-T. gondii antibodies’ kinetics in pregnancy. Fifty-two pregnant patients were studied. A questionnaire with sociodemographic and clinical aspects was used. Afterward, 10 mL of venous blood was collected by venipuncture every trimester. The concentrations of 17-β estradiol, progesterone, and prolactin were measured, using the ELISA method. In addition, anti-Toxoplasma IgG and IgM antibodies were also determined in the first, second, and third trimester. The prevalence of anti-Toxoplasma IgG antibodies was 26.92% in the first and second trimester and 32.7% in the third trimester. In seropositive women, 17-β estradiol increased in the second and third trimesters of pregnancy. Progesterone increased significantly p < 0.039 in the third trimester in these women, while prolactin increased in the second trimester with a statistical significance of p < 0.021. In addition, 17-β estradiol, progesterone, and prolactin are associated with T. gondii infection during pregnancy. New studies are necessary to clarify the specific mechanisms of immune response related to these hormones during pregnancy.

Abstract Background The Severity of Alopecia Tool (SALT) is a standardized method for quantifying scalp hair loss in alopecia areata (AA). SALT scores can be used to guide treatment decisions and are widely used as eligibility criteria and endpoints for clinical trials in AA. However, clinicians may be unfamiliar with assessing and envisioning SALT scores in practice. Objectives To aid clinicians in the determination and application of SALT scores in a clinical setting, this manuscript seeks to contextualize SALT scores using patient images from a clinical trial of adults with severe AA. Methods Images from 722 patients enrolled in BRAVE‐AA1, a phase 2/3 study of baricitinib in adults with severe AA (SALT score ≥50; ≥50% scalp hair loss), were obtained at baseline and Weeks 12, 36, and 52 and compiled into a repository. Photographs were selected to represent SALT scores across the full range of disease (SALT scores 0–100) and to demonstrate the progression of SALT scores during the course of treatment. Results Images of six patients depict the range of SALT scores (0–100). Photographs are of male and female patients of different ages (21–69) and races (Asian, Black, White) with varying extent, density, and patterns of hair loss. Images of two additional patients demonstrate the use of SALT to monitor treatment progress, showing distinct patterns and timing of clinical response over 52 weeks of therapy. Conclusions The SALT is widely used in clinical trials for AA, but clinicians may lack familiarity. Presented patient images show SALT scores commonly used as eligibility criteria and endpoints in clinical trials, which may be useful in identifying patients eligible for systemic treatment and in visualizing therapeutic response.

ABSTRACT: Our objective was to determine the effect of a 200-µg dose of GnRH 25 d after previous artificial insemination (AI) in a Resynch-25 resynchronization program on ovulatory response, circulating progesterone (P4) concentrations before and after treatment, and pregnancy per AI (P/AI) compared with a 100-µg dose in lactating Holstein cows. Experimental d 0 was considered the day of the previous AI. Lactating dairy cows (n = 3,240) with an average of 126 d in milk (DIM) and between 1 and 6 services were randomly assigned to receive 100 µg or 200 µg of GnRH on d 25 (GnRH25). On d 32 after AI, cows diagnosed nonpregnant with the presence of a corpus luteum (CL) detected by ultrasound (n = 1,249) received PGF2α treatments on d 32 and 33, followed by a GnRH 32 h later and AI 16 h after this last GnRH. Blood samples were collected on d 25, 32, and 34 to evaluate serum P4 concentrations. Transrectal ultrasonographic examination was performed on d 25 and 27 to assess ovulatory response to GnRH25. Cows were checked for pregnancy on d 32, 46, and 88 after AI. The larger dose of GnRH increased the overall proportion of cows that ovulated to the GnRH25 (25.0% for the 100-µg dose vs. 32.5% for the 200-µg dose). However, when cows were evaluated separately according to the pregnancy status on d 32 after AI, we found no treatment effect within cows pregnant and nonpregnant. Even though treatment increased the proportion of cows with serum P4 ≤0.42 ng/mL at the last GnRH treatment (G2; 86.2% for the 100-µg dose vs. 93.0% for the 200-µg dose), it did not affect P/AI on d 32, 46, and 88. Furthermore, a greater proportion of cows without a functional CL at GnRH25 had circulating P4 concentrations ≥1.00 ng/mL on d 32 and lower than 0.42 ng/mL on G2. These cows also had a greater P/AI on d 32, 46, and 88. In summary, the larger dose of GnRH on d 25 after AI did not increase the ovulatory response in nonpregnant cows and P/AI on d 32, 46, and 88 after AI after the Resynch-25 program. Additionally, nonpregnant cows without a functional CL at GnRH25 were better synchronized after the Resynch-25 protocol and had greater P/AI on d 32, 46, and 88 after timed-AI.

Diurnal biting flies are strongly attracted to blue objects. This behaviour is widely exploited for fly control, but its functional significance is debated. It is hypothesized that blue objects resemble animal hosts; blue surfaces resemble shaded resting places; and blue attraction is a by-product of attraction to polarized light. We computed the fly photoreceptor signals elicited by a large sample of leaf and animal integument reflectance spectra, viewed under open/cloudy illumination and under woodland shade. We then trained artificial neural networks (ANNs) to distinguish animals from leaf backgrounds, and shaded from unshaded surfaces, in order to find the optimal means of doing so based upon the sensory information available to a fly. After training, we challenged ANNs to classify blue objects used in fly control. Trained ANNs could make both discriminations with high accuracy. They discriminated animals from leaves based upon blue-green photoreceptor opponency and commonly misclassified blue objects as animals. Meanwhile, they discriminated shaded from unshaded stimuli using achromatic cues and never misclassified blue objects as shaded. We conclude that blue-green opponency is the most effective means of discriminating animals from leaf backgrounds using a fly's sensory information, and that blue objects resemble animal hosts through such mechanisms.

Youth in the juvenile justice system evince high rates of mental health symptoms, including anxiety and depression. How these symptom profiles change after first contact with the justice system and - importantly - how they are related to re-offending remains unclear. Here, we use latent growth curve modeling to characterize univariate and multivariate growth of anxiety, depression, and re-offending in 1216 male adolescents over 5 years following their first arrest. Overall, the group showed significant linear and quadratic growth in internalizing symptoms and offending behaviors over time such that levels decreased initially after first arrest followed by a small but significant upturn occurring a few years later. Crucially, multivariate growth models revealed strong positive relationships between the rates of growth in internalizing symptoms and offending behaviors such that improvements in mental health related to greater decreases in offending, and vice versa. These results highlight the reciprocal nature of internalizing and externalizing problems in adolescence, underscoring the importance of considering mental health alongside offending in the juvenile justice system.

Background: As a result of botanical explorations carried out for the project Bromeliaceae of Mexico, we collected plants of the genus Hechtia within the Sierra de Huautla Biosphere Reserve, in Xochipala, municipality of Tlaquiltenango, Morelos, Mexico. However, this plant does not resemble any known species. Question: Do the Hechtia populations in Xochipala correspond to an undescribed taxon? Studied species: Species of Hechtia previously reported from Morelos and all the taxa with greater morphological similarity to the new proposed taxon. Study site and date: Xochipala, municipality of Tlaquiltenango, Morelos, Mexico, 2023. Methods: Our research involved field work, review of herbarium specimens, protologues, and type material of all the species of Hechtia distributed in Morelos, in the neighboring states of Puebla and Guerrero, and of the most morphologically similar taxa. Results: After a careful and detailed review of the living material and herbarium specimens, we conclude that those plants belong to an undescribed taxon that we propose here. A morphological description, images, and a distribution map of Hechtia cerrostlatilpae are included, as well as an identification key and a list of examined specimens of all Hechtia species known from the state of Morelos. Conclusions: The discovery of a new species of Hechtia in Xochipala, Morelos, Mexico increases the plant diversity of Morelos, particularly at the Sierra de Huautla Biosphere Reserve. Hechtia cerrostlatilpae presents unique vegetative and floral characters that allow its recognition as a new species within the genus.

Introducción: El asma es un trastorno inflamatorio crónico de las vías respiratorias, es tratada con corticoesteroides inhalatorios, que actúan sobre las vías respiratorias provocando efectos adversos sobre la salud bucodental. Objetivo: Evidenciar las afecciones bucales que se relacionan con el uso de corticosteroides para concientizar sobre las repercusiones de los mismos en la cavidad oral. Reporte de caso: Paciente de 4 años presenta lesión en el dorso de la lengua forma de placa blanquecina, refiere molestias ligeras, sensibilidad y ardor. La madre refiere que padece de asma y utiliza Salbutamol 100 mcg y Budesonida 200 mcg vía inhalatoria bucofaringea. Se retira la placa blanquecina quedando una zona eritematosa en el sector de la línea media de la lengua, rojiza, lisa y brillante con depapilación así también se evidencian lesiones cariosas en los incisivos primarios superiores. Se establece como diagnóstico candidiasis pseudomembranosa. Se indica enjuague con 5mL de nistatina 100.000 UI/mL por un minuto. A los 15 días, refiere gran disminución de las molestias y del ardor pero con persistencia de zonas lisas, brillantes y depapiladas, esta vez ubicadas enel sector anterior de la lengua, se indica continuar con el enjuague 1 vez al día antes de dormir durante 2 días más. A dos meses de haber iniciado el tratamiento se observa ausencia de candidiasis y se establece como diagnóstico secundario glositis migratoria benigna. Conclusión: El uso de corticoides repercute en la cavidad oral haciendo que el niño sea más propenso a padecer ciertas patologías orales como candidiasis y caries dental.

Chagas disease, caused by the parasite Trypanosoma cruzi, affects over 6 million people, mainly in Latin America. Two different clinical phases, acute and chronic, are recognised. Currently, 2 anti-parasitic drugs are available to treat the disease (nifurtimox and benznidazole), but diagnostic methods require of a relatively complex infrastructure and trained personnel, limiting its widespread use in endemic areas, and the access of patients to treatment. New diagnostic methods, such as rapid tests (RDTs) to diagnose chronic Chagas disease, or loop-mediated isothermal amplification (LAMP), to detect acute infections, represent valuable alternatives, but the parasite's remarkable genetic diversity might make its implementation difficult. Furthermore, determining the efficacy of Chagas disease treatment is complicated, given the slow reversion of serological anti-T. cruzi antibody reactivity, which may even take decades to occur. New biomarkers to evaluate early therapeutic efficacy, as well as diagnostic tests able to detect the wide variety of circulating genotypes, are therefore, urgently required. To carry out studies that address these needs, high-quality and traceable samples from T. cruzi-infected individuals with different geographical backgrounds, along with associated clinical and epidemiological data, are necessary. This work describes the framework for the creation of such repositories, following standardised and uniform protocols, and considering the ethical, technical, and logistic aspects of the process. The manual can be adapted according to the resources of each laboratory, to guarantee that samples are obtained in a reproducible way, favouring the exchange of data among different work groups, and their generalizable evaluation and analysis. The main objective of this is to accelerate the development of new diagnostic methods and the identification of biomarkers for Chagas disease.

Chagas disease (CD) is recognized as one of the 20 neglected tropical diseases by the World Health Organization (WHO), posing a significant global health challenge. The objective of this work was to conduct a systematic methodology review to explore the different classifications used to describe the presence and degree of organ involvement in patients with CD since the disease's description in 1909. We searched relevant electronic medical databases from their inception dates to July 2023. We also delved into historical variations and revisions of each classification, the necessary diagnostic methods, their prognostic value, and their uptake. Our study underscores the conspicuous absence of a universally accepted CD classification system for cardiac and digestive involvement, both in the context of clinical trials and within current clinical guidelines. This endeavour will facilitate cross-population comparisons if clinical manifestations and complementary test results are available for each patient, constituting a pivotal stride toward identifying precise prognoses and establishing a minimum data set requisite for a fitting CD classification, tailored to the test availability in both endemic and non-endemic regions.

The NHEPACHA Iberoamerican Network, founded on the initiative of a group of researchers from Latin American countries and Spain, aims to establish a research framework for Chagas disease that encompasses diagnosis and treatment. For this purpose, the network has created a questionnaire to gather relevant data on epidemiological, clinical, diagnostic, and therapeutic aspects of the disease. This questionnaire was developed based on a consensus of expert members of the network, with the intention of collecting high-quality standardized data, which can be used interchangeably by the different research centers that make up the NHEPACHA network. Furthermore, the network intends to offer a clinical protocol that can be embraced by other researchers, facilitating comparability among published studies, as well as the development of therapeutic response and progression markers.

Forest inventories are undergoing rapid changes due to an increasingly complex set of economic, environmental, and social policy objectives. Therefore, the objective is to identify, analyse, and discuss the main forest inventory methods at global, regional, and local levels, with an analytical perspective on the goals they seek to achieve in various forest ecosystems. For this review, information from 79 relevant studies related to the objectives and methods used in sampling forest resources in tropical, boreal, temperate, and arid ecosystems was considered. According to the analysed studies, forest inventories in different ecosystems face challenges and apply varied methods to assess forests. In the tropics, the focus is on monitoring biomass and carbon, but they show limitations in data quality and quantity limitations. To improve accuracy, robust sampling methods are suggested. In boreal ecosystems, LiDAR and data-driven models offer detailed biomass estimates. In temperate forests, diversified sampling techniques are employed to balance accuracy and efficiency. In arid ecosystems, non-plot methods are useful for mapping density and diversity of the forests. To board the specific challenges of each region, innovative approaches are needed. Inventories have been influenced by changes in environmental policies and technology; therefore, the need to estimate key forest variables and monitor their dynamics requires robust and technologically advanced sampling methods.

Many species of beetles from the family Scarabaeidae reflect light with near-circular polarization. In some cases, spectral narrow-band polarization phenomena result in a distinct color with a metallic shine. In other cases, broad-band features are seen, and these beetles have a silvery or goldish appearance. These features in the cuticles originate from helicoidal structures, so-called circular Bragg structures and also referred to as Bouligand structures. In this communication, Protaetia mirifica, exhibiting near-circular polarization properties in dual spectral regions, centered approximately at the wavelengths of 474 and 770 nm, is investigated in considerable detail using Mueller matrix spectroscopic ellipsometry (MMSE). From interference oscillations in the MMSE spectra, the pitch profile of the helicoidal structures in the beetle cuticle is extracted and further used in electromagnetic modeling of the cuticle structure, including the determination of epicuticle and exocuticle thicknesses (280 nm and 8.1 μm, respectively) and anisotropic optical properties. These findings are confirmed by scanning electron microscopy. The analysis shows that the uppermost 4 μm of the cuticle has a nearly constant pitch of 310 nm, which abruptly jumps to 440 nm and then gradually increases up to 575 nm. Sum decompositions of MMSE spectra reveal that the beetle cuticle reflects like a circular polarizer or like a dielectric mirror, depending on the spectral region.

BackgroundHabitat modification and land use changes impact ecological interactions and alter the relationships between humans and nature. Mexico has experienced significant landscape modifications at the local and regional scales, with negative effects on forest cover and biological biodiversity, especially in the Yucatan peninsula in southeastern Mexico. Given the close relationship between landscape modification and the transmission of zoonotic and vector-borne diseases, it is essential to develop criteria for identifying priority zoonoses in the south of the country.Methodology/principal findingsWe reviewed 165 published studies on zoonotic and vector-borne diseases in the region (2015-2024). We identified the most frequent vectors, reservoirs, and hosts, the most prevalent infections, and the factors associated with transmission risk and the anthropogenic landscape modification in urban, rural, ecotone, and sylvatic habitats. The most relevant pathogens of zoonotic risk included Trypanosoma cruzi, arboviruses, Leishmania, Rickettsia, Leptospira, and Toxoplasma gondii. Trypanosoma cruzi was the vector-borne agent with the largest number of infected vertebrate species across habitats, while Leishmania and arboviruses were the ones that affected the greatest number of people. Dogs, cats, backyard animals, and their hematophagous ectoparasites are the most likely species maintaining the transmission cycles in human settlements, while rodents, opossums, bats, and other synanthropic animals facilitate connection and transmission cycles between forested habitats with human-modified landscapes. Pathogens displayed different prevalences between the landscapes, T. cruzi, arbovirus, and Leptospira infections were the most prevalent in urban and rural settlements, whereas Leishmania and Rickettsia had similar prevalence across habitats, likely due to the diversity and abundance of the infected vectors involved. The prevalence of T. gondii and Leptospira spp. may reflect poor hygiene conditions. Additionally, results suggest that prevalence of zoonotic and vector-borne diseases is higher in deforested areas and agricultural aggregates, and in sites with precarious health and infrastructure services.ConclusionsSome hosts, vectors, and transmission trends of zoonotic and vector-borne diseases in the YP are well known but others remain poorly recognized. It is imperative to reinforce practices aimed at increasing the knowledge, monitoring, prevention, and control of these diseases at the regional level. We also emphasize the need to perform studies on a larger spatio-temporal scale under the socio-ecosystem perspective, to better elucidate the interactions between pathogens, hosts, vectors, environment, and sociocultural and economic aspects in this and many other tropical regions.

Robot manipulators play a crucial role in various industrial and research settings, requiring precise and controlled interactions with their surroundings. Achieving this goal with fewer sensors offers advantages not only in terms of cost and decreased risk of failure but also enhances accuracy and long-termreliability. In this paper,we introduce a nonlinear force/position controller that eliminates the requirement for velocity measurements. This controller provides versatility by facilitating the generation of bounded control actions during robot-environment interactions, ensuring a higher level of safety for both the robot and its environment during the execution of tasks necessitating physical contact between them. The proposed approach is underpinned by a stability analysis in the Lyapunov sense and has been validated through a series of simulation and experimental tests.

STUDY OBJECTIVES: Sleep duration and intraindividual variability in sleep duration undergo substantial changes in adolescence and impact brain and behavioral functioning. Although experimental work has linked acute sleep deprivation to heightened limbic responding and reduced regulatory control, there is limited understanding of how variability in sleep patterns might interact with sleep duration to influence adolescent functioning. This is important for optimal balancing of length and consistency of sleep. Here, we investigated how objective indices of sleep duration and variability relate to stress, restfulness, and intrinsic limbic network functioning in adolescents. METHODS: A sample of 101 adolescents ages 14-18 reported their stressors, after which they wore wrist actigraph watches to monitor their sleep and rated their restfulness every morning over a 2-week period. They also completed a resting-state fMRI scan. RESULTS: Adolescents reporting more stress experienced shorter sleep duration and greater sleep variability over the 2-week period. Longer nightly sleep duration was linked to feeling more rested the next morning, but this effect was reduced in adolescents with high cumulative sleep variability. Sleep variability showed both linear and quadratic effects on limbic connectivity: adolescents with high sleep variability exhibited more connectivity within the limbic network and less connectivity between the limbic and frontoparietal networks than their peers, effects which became stronger once variability exceeded an hour. CONCLUSIONS: Results suggest that cumulative sleep variability is related to stress and limbic network connectivity and shows interactive effects with sleep duration, highlighting the importance of balancing length and consistency of sleep for optimal functioning in adolescence.

Manta and devil rays (mobulids) are filter feeding elasmobranchs with extreme K-selective life histories found circumglobally from temperate to tropical waters. Their vulnerability to fisheries exploitation, bycatch, boat collisions, entanglement and unregulated tourism is exacerbated by their aggregative behavior. Studies have identified aggregation sites around the world for all nine mobulid species, with these groupings varying from a few individuals to thousands. However, the terminology used to define these aggregations and the drivers underpinning them remain unclear, hindering the development of effective management and conservation strategies. Here, we analyze aggregation behavior for mobulid species, providing consistent definitions for grouping events and summarizing the existing research on drivers and environmental factors triggering these events. We find that aggregation behaviors facilitate socializing and key life history functions in mobulids, including feeding, courtship and mating, predation avoidance, cleaning, and thermoregulation. Conservation threats and management mitigation opportunities associated with aggregations sites include fisheries, tourism, spatial protection, and climate change. Finally, we highlight knowledge gaps for future research prioritization and developments in the field for the identification of aggregation sites, the study of aggregation size and demographics and the functions and timing of aggregations.

Pancreatic cancer is characterized by extensive resistance to conventional therapies, making clinical management a challenge. Here we map the epigenetic dependencies of cancer stem cells, cells that preferentially evade therapy and drive progression, and identify SWI/SNF complex member SMARCD3 as a regulator of pancreatic cancer cells. Although SWI/SNF subunits often act as tumor suppressors, we show that SMARCD3 is amplified in cancer, enriched in pancreatic cancer stem cells and upregulated in the human disease. Diverse genetic mouse models of pancreatic cancer and stage-specific Smarcd3 deletion reveal that Smarcd3 loss preferentially impacts established tumors, improving survival especially in context of chemotherapy. Mechanistically, SMARCD3 acts with FOXA1 to control lipid and fatty acid metabolism, programs associated with therapy resistance and poor prognosis in cancer. These data identify SMARCD3 as an epigenetic modulator responsible for establishing the metabolic landscape in aggressive pancreatic cancer cells and a potential target for new therapies.

In adults, sleep disturbances are heritable and show strong genetic associations with brain morphology; however, little is known about the genetic architecture of childhood sleep and potential etiological links between sleep, brain development, and pediatric-onset psychiatric symptoms.MethodsUsing data from the Adolescent Brain Cognitive Development Study (n Phenotype = 4428 for discovery/replication, n Genetics = 4728; age 9-10 years), we assessed phenotypic relationships, heritability, and genetic correlations between childhood sleep disturbances (insomnia, arousal, breathing, somnolence, hyperhidrosis, sleep-wake transitions), brain size (surface area, cortical thickness, volume), and dimensional psychopathology.ResultsSleep disturbances showed widespread positive associations with multiple domains of childhood psychopathology; however, only insomnia showed replicable associations with smaller brain surface area. Among the sleep disturbances assessed, only insomnia showed significant heritability (h 2 SNP = 0.15, p < .05) and showed substantial genetic correlations with externalizing and attention-deficit/hyperactivity disorder symptomatology (r G s > 0.80, ps < .05). We found no evidence of genetic correlation between childhood insomnia and brain size. Furthermore, polygenic risk scores calculated from genome-wide association studies of adult insomnia and adult brain size did not predict childhood insomnia; instead, polygenic risk scores trained using attention-deficit/hyperactivity disorder genome-wide association studies predicted decreased surface area at baseline as well as insomnia and externalizing symptoms longitudinally.ConclusionsFindings demonstrate a distinct genetic architecture underlying childhood insomnia and brain size and suggest genetic overlap between childhood insomnia and attention-deficit/hyperactivity disorder symptomatology. Additional research is needed to examine how genetic risk manifests in altered developmental trajectories and comorbid sleep/psychiatric symptoms across adolescence.
