A copper-catalyzed asymmetric cascade cyclization via C(sp2)–O bond cleavage is disclosed, affording a range of chromeno[3,4-c]pyrroles bearing a triaryl oxa-quaternary carbon stereocenter in high yields and enantioselectivities.

Hepatic fibrosis is a major global health problem and considered a leading cause of liver-related morbidity and mortality worldwide. Although previous studies have suggested that transient receptor potential vanilloid-1 (TRPV1) is protective against cardiac and renal fibrosis, its functional role in hepatic fibrosis has remained elusive. Herein, we characterize the effects of TRPV1 on carbon tetrachloride- (CCl4-) induced mice, in vitro transforming growth factor-β- (TGF-β-) treated hepatic stellate cells (HSCs), and human fibrosis specimens. Finally, our results demonstrated the significant TRPV1 downregulation in human liver fibrosis tissues. Knocking out TRPV1 significantly increased the expression of various hepatic fibrosis markers, while the expression of these biomarkers declined markedly in capsaicin-activated mice. Moreover, our study revealed that knocking down TRPV1 would enhance the promotive effect of TGF-β on HSC proliferation, cell cycle, cell apoptosis, and ECM expression. Also, such promotive effect can be partially reversible by capsaicin, an exogenous activator of TRPV1. Collectively, the obtained data suggest that TRPV1 may alleviate CCl4-induced hepatic fibrosis and attenuate the effect of TGF-β on HSC activation, proliferation, and apoptosis, which overall implies that targeting TRPV1 channel activity may be an effective therapeutic strategy for treating hepatic fibrosis.

Background Brahma-related gene 1 (BRG1) is essential for embryogenesis and cellular metabolism. A deficiency of BRG1 in vivo decreases lipid droplets, but the molecular mechanism underlying its role in lipid metabolism associated with hepatocellular carcinoma (HCC) remains unknown. Aims We aimed to determine the role of BRG1 in lipid metabolism in HCC. Methods We assessed the differential expression of BRG1 in HCC and adjacent non-tumorous tissues using tissue microarrays. We stained lipid droplets in HCC cells with Bodipy fluorescence and Oil Red O, and verified BRG1 binding to the promoter region of glycosylated lysosomal membrane protein (GLMP) using chromatin immunoprecipitation. Results The expression of GLMP, a potential lipid metabolism regulator, was suppressed by BRG1 via transcriptional activity. Knockdown of BRG1 decreased lipid droplets, increased GLMP expression and altered the phosphoinositide-3-kinase adaptor protein 1 (PIK3AP1)/phosphatidylinositol-3 kinase (PI3K)/protein kinase B (AKT) pathway in HCC, which further GLMP knockdown partially restored. Thus, GLMP knockdown increased lipid droplets and differentially altered the PI3K/AKT pathway. Conclusions Downregulating BRG1 decreased lipid droplet deposition in HCC cells by upregulating GLMP and altering the PI3K/AKT pathway. Both BRG1 and GLMP might serve as therapeutic targets for disorders associated with dysregulated lipid metabolism, such as NAFLD and NAFLD-associated HCC.

The fungal genus Microcera consists of species mostly occurring as parasites of scale insects, but are also commonly isolated from soil or lichens. In the present study, we surveyed the diversity and assess the taxonomy of entomopathogenic fungi in Sichuan Province, China. Two new species of Microcera, viz. M. chrysomphaludis and M. pseudaulacaspidis, were isolated from scale insects colonising walnut (Juglans regia). Maximum Likelihood and Bayesian Inference analyses of ITS, LSU, tef1-α, rpb1, rpb2, acl1, act, tub2, cmdA and his3 sequence data provide evidence for the validity of the two species and their placement in Nectriaceae (Hypocreales). Microcera pseudaulacaspidis primarily differs from similar species by having more septate and smaller cylindrical macroconidia, as well as DNA sequence data. Meanwhile, Microcera chrysomphaludis has elliptical, one-septate ascospores with acute ends and cylindrical, slightly curved with 4–6 septate macroconidia up to 78 µm long. Morphological descriptions with illustrations of the novel species and DNA-based phylogeny generated from analyses of multigene dataset are also provided to better understand species relationships.

Two new species of Scytinostroma viz. S. acystidiatum and S. macrospermum, are described from southwest China. Phylogeny based on ITS + nLSU dataset demonstrates that samples of the two species form two independent lineages and are different in morphology from the existing species of Scytinostroma. Scytinostroma acystidiatum is characterized by resupinate, coriaceous basidiomata with cream to pale yellow hymenophore, a dimitic hyphal structure with generative hyphae bearing simple septa, the absence of cystidia, and amyloid, broadly ellipsoid basidiospores measuring 4.7–7 × 3.5–4.7 μm. Scytinostroma macrospermum is characterized by resupinate, coriaceous basidiomata with cream to straw yellow hymenophore, a dimitic hyphal structure with generative hyphae bearing simple septa, numerous cystidia embedded or projecting from hymenium, and inamyloid, ellipsoid basidiospores measuring 9–11 × 4.5–5.5 μm. The differences between the new species and morphologically similar and phylogenetically related species are discussed.

Recent development within the field of medium voltage wide-bandgap semiconductor devices are drawing the attention from both researchers and industries, due to the demanding requirements for more efficient high-power energy conversion. The rapid development has entailed an increased awareness of the negative impact of increased rate of change in voltage, dv/dt, and its derived issues caused by the inevitable parasitic capacitive couplings. This paper is dedicated to present an overview of the parasitic capacitive couplings in high-power medium voltage power electronic converter systems, using an example reference system enabled by medium voltage SiC MOSFETs. The definitions of capacitive couplings are presented and the impacts raised by parasitic capacitive couplings are reviewed. In addition, the similarities of different capacitive couplings introduced by components in practical medium voltage systems are presented and summarized. Lastly, the challenges and future work with respect to parasitic capacitive couplings in medium voltage power electronics converter systems are shared from the authors’ perspective.

The Langmuir-Blodgett (LB) technique, in which monolayers are commonly transferred from a liquid/gas interface to a solid surface, allows convenient fabrication of highly ordered thin films with molecular-level precision. This method is widely applicable to substances ranging from organic molecules to nanomaterials. Therefore, LB methods have provided a critical toolbox for researchers to engineer nanoarchitectures. The LB fabrication process is also compatible with numerous substrate materials over large areas, which is advantageous for practical application. Despite its wide applicability, the LB strategy has not been extensively employed in battery studies. The versatility of LB film, along with the accumulated knowledge associated with this technique, makes it a promising platform for promoting battery chemistry evolution. This Review summarizes recent advances of LB methods for high-performance battery development, including preparation of electrode materials, fabrication of functional layers, and battery diagnosis and thus illustrates the high utility of LB approaches in battery research.

Alternative splicing (AS) is one of the major post-transcriptional regulation mechanisms that contributes to plant responses to various environmental perturbations. Darkness and heat are two common abiotic factors affecting plant growth, yet the involvement and regulation of AS in the plant responses to these signals remain insufficiently examined. In this study, we subjected Arabidopsis seedlings to 6 h of darkness or heat stress and analyzed their transcriptome through short-read RNA sequencing. We revealed that both treatments altered the transcription and AS of a subset of genes yet with different mechanisms. Dark-regulated AS events were found enriched in photosynthesis and light signaling pathways, while heat-regulated AS events were enriched in responses to abiotic stresses but not in heat-responsive genes, which responded primarily through transcriptional regulation. The AS of splicing-related genes (SRGs) was susceptible to both treatments; while dark treatment mostly regulated the AS of these genes, heat had a strong effect on both their transcription and AS. PCR analysis showed that the AS of the Serine/Arginine-rich family gene SR30 was reversely regulated by dark and heat, and heat induced the upregulation of multiple minor SR30 isoforms with intron retention. Our results suggest that AS participates in plant responses to these two abiotic signals and reveal the regulation of splicing regulators during these processes.

The polypores with shallow pores from tropical Asia and America are studied. Our molecular phylogeny based on the internal transcribed spacer (ITS), the large subunit nuclear ribosomal RNA gene (nLSU), the translation elongation factor 1-α gene (TEF1), and the largest subunit of RNA polymerase II (RPB1) demonstrates six clades are formed among Porogramme and related genera. Two new genera, Cyanoporus and Pseudogrammothele, are established, and the six clades represent Porogramme, Cyanoporus, Grammothele, Epithele, Theleporus, and Pseudogrammothele, respectively. The molecular clock analyses estimate the divergence times of the six clades based on a dataset (ITS + LSU + TEF1 + RPB1 + RPB2), and we recognize the mean stem ages of the six genera are earlier than 50 Mya. Three new species in Porogramme were morphologically and phylogenetically confirmed, and they are described as P. austroasiana, P. cylindrica, and P. yunnanensis. Phylogenetic analysis shows that type species of Tinctoporellus and Porogramme are nested in the same clade, and Tinctoporellus is treated as a synonym of Porogramme. Based on our phylogeny, twelve new combinations are proposed, and the differences between the new species and similar or related species are discussed.

Two new species of Antrodia, A. aridula and A. variispora, are described from western China. Phylogeny based on a six-gene dataset (ITS + nLSU + nSSU + mtSSU + TEF1 + RPB2) demonstrates that samples of the two species form two independent lineages within the clade of Antrodia s.s. and are different in morphology from the existing species of Antrodia. Antrodia aridula is characterized by its annual and resupinate basidiocarps with angular to irregular pores of 2–3 mm each and oblong ellipsoid to cylindrical basidiospores measuring 9–12 × 4.2–5.3 μm, growing on gymnosperm wood in a dry environment. Antrodia variispora is characterized by its annual and resupinate basidiocarps with sinuous or dentate pores with a size of 1–1.5 mm each and oblong ellipsoid, fusiform, pyriform, or cylindrical basidiospores measuring 11.5–16 × 4.5–5.5 μm, growing on the wood of Picea. The differences between the new species and morphologically similar species are discussed in this article.

Two new species in Hymenochaetaceae, Fulvifomes acaciae and Pyrrhoderma nigra, are illustrated and described from tropical Asia and America based on morphology and phylogenetic analyses. F. acaciae is characterized by perennial, pileate, and woody hard basidiomata when fresh; ash gray to dark gray, encrusted, concentrically sulcate, and irregularly cracked pileal surface; circular pores of 7–8 per mm with entire dissepiments; a dimitic hyphal system in trama and context; absence of setal element and presence of cystidioles; and broadly ellipsoid, yellowish brown, thick-walled, and smooth basidiospores measuring 5–6 μm × 4–5 μm. P. nigra is characterized by perennial and resupinate basidiomata with dark gray to almost black pore surface when fresh; small and circular pores of 7–9 per mm, a monomitic hyphal system with generative hyphae simple septate, hyphoid setae dominant in subiculum but not in tube trama, and absence of cystidia; and ellipsoid, hyaline, thin-walled basidiospores measuring 4–5 μm × 3–3.6 μm. The differences between the new species and morphologically similar and phylogenetically related species are discussed. Keys to Fulvifomes and Pyrrhoderma have also been provided.

Boletes are favored by consumers because of their unique flavor, rich nutrition and delicious taste. However, the different nutritional values of each species lead to obvious price differences, so shoddy products appear on the market, which affects food safety. The aim of this study was to find a rapid and effective method for boletes species identification. In this paper, 1,707 samples of eight boletes species were selected as the research objects. The original Mid-Infrared (MIR) spectroscopy data were adopted for support vector machine (SVM) modeling. The 11,949 spectral images belong to seven data sets such as two-dimensional correlation spectroscopy (2DCOS) and three-dimensional correlation spectroscopy (3DCOS) were used to carry out Alexnet and Residual network (Resnet) modeling, thus we established 15 models for the identification of boletes species. The results show that the SVM method needs to process complex feature data, the time cost is more than 11 times of other models, and the accuracy is not high enough, so it is not recommended to be used in data processing with large sample size. From the perspective of datasets, synchronous 2DCOS and synchronous 3DCOS have the best modeling results, while one-dimensional (1D) MIR Spectrum dataset has the worst modeling results. After comprehensive analysis, the modeling effect of Resnet on the synchronous 2DCOS dataset is the best. Moreover, we use large-screen visualization technology to visually display the sample information of this research and obtain their distribution rules in terms of species and geographical location. This research shows that deep learning combined with 2DCOS and 3DCOS spectral images can effectively and accurately identify boletes species, which provides a reference for the identification of other fields, such as food and Chinese herbal medicine.

An organocatalytic intramolecular (4 + 2) annulation of enals with ynamides is disclosed, providing a convenient way to axially chiral indolines. The synthesized axially chiral skeleton was proved to be potentially applicable as a chiral ligand.

Sidera, belonging to the Rickenella clade of Hymenochaetales, is a worldwide genus with mostly poroid hymenophore of wood-inhabiting fungi. Two new species in the genus, Sidera americana and S. borealis, are described and illustrated from China and North America based on morphological and molecular evidence. They were mainly found growing on rotten wood of Abies, Picea and Pinus. S. americana is characterized by annual, resupinate basidiomata with silk sheen when dry, round pores (9–11 per mm), a dimitic hyphal system, and allantoid basidiospores measuring 3.5–4.2 × 1 μm. S. borealis is characterized by annual, resupinate basidiomata with cream to pinkish buff dry pore surface, angular pores (6–7 per mm), a dimitic hyphal system, and allantoid basidiospores measuring 3.9–4.1 × 1–1.1 μm. Phylogenetic analysis based on a combined 2-locus dataset [ITS1-5.8S-ITS2 (ITS) + nuclear large subunit RNA (nLSU)] shows that the two species are members of Sidera, and they are compared with morphologically similar and phylogenetically related species, respectively. An identification key to 18 accepted species of Sidera in worldwide is provided.

Nowadays, wild edible bolete mushrooms are more and more attractive among consumers due to their natural health, nutrition, and delicious characteristics. Appropriate analytical techniques together with multivariate statistics analysis are required for the quality control and evaluation of these edible mushrooms. Ultraviolet-visible (UV-Vis) and infrared (IR) technologies have the advantages of time-saving, low-cost, and environmentally friendly, are now prominent among major analytical technologies for quality evaluation of bolete mushrooms. Chemometrics methods have been developed to solve classification and regression issues of bolete mushrooms in combination with spectrum. This paper reviewed the most recent applications of UV-Vis and IR technology coupled with chemometrics in wild edible bolete mushrooms, including the identification of species, origin, and storage duration, fraud detection, and antioxidant properties evaluation, and discussed the limitations and prospects of spectroscopy technologies in the researches of bolete mushrooms, excepting to provide a reference for further research and practical application of wild edible bolete mushrooms.
