Gyrfalcon (Falco rusticolus) is the largest falcon in the world. It inhabits a wide range of climate zones in the Northern Hemisphere, from boreal forests in the south of its range to the arid polar deserts of the High Arctic. In Greenland, because of the harsh, remote environments in which gyrfalcons live, research related to the contemporary and pre-modern periods has been limited to the north-west, central west and central east coasts, with no specific investigations being conducted for the north-east. Here, we report the first pre-modern record of a gyrfalcon in north-east Greenland, located at 80.4°N in Kronprins Christian Land. Skin tissue from a decaying gyrfalcon’s body was radiocarbon dated to 769–944 CE (common era) using a terrestrial-only calibration curve, and 1182–1456 CE using a marine-only calibration curve. Since the gyrfalcon has a mixed terrestrial/marine diet, the actual age can be said to belong between these two groups. This limited data, therefore places the presence of the gyrfalcon in north-east Greenland during a period of prolonged elevated temperatures and climate stress associated with the Medieval Climate Anomaly. Whether the gyrfalcon was part of a larger population or a straggler, and whether the species survived the whole of the Medieval Climate Anomaly in north-east Greenland, is unknown.

Asperphenamate is a small peptide natural product that has gained much interest due to its antitumor activity. In the recent years numerous bioactive synthetic asperphenamate analogs have been reported, whereas only a handful of natural analogs either of microbial or plant origin has been discovered. Herein we describe a UHPLC-HRMS/MS and amino acid supplement approach for discovery and design of novel asperphenamate analogs. Chemical analysis of Penicillium astrolabium, a prolific producer of asperphenamate, revealed three previously described and two novel asperphenamate analogs produced in significant amounts, suggesting a potential for biosynthesis of further asperphenamate analogs by varying the amino acid availability. Subsequent growth on proteogenic and non-proteogenic amino acid enriched media, revealed a series of novel asperphenamate analogs, including single or double amino acid exchange, as well as benzoic acid exchange for nicotinic acid, with the latter observed from a natural source for the first time. In total, 22 new asperphenamate analogs were characterized by HRMS/MS, with one additionally confirmed by isolation and NMR structure elucidation. This study indicates an extraordinary nonribosomal peptide synthetase (NRPS) flexibility based on substrate availability, and therefore the potential for manipulating and designing novel peptide natural products in filamentous fungi.

A temporary discontinuation (drug holiday) of high-dose antiresorptive (AR) agents has been proposed to reduce the risk of medication-related osteonecrosis of the jaw (MRONJ). The aim of this systematic review was to answer the question: Is high-dose AR drug holiday, at the time of tooth extraction or dentoalveolar surgery, necessary to prevent the development of MRONJ in patients with cancer? This protocol was registered in the PROSPERO database. Medline, Embase, and the Cochrane Central Register of Controlled Trials (CENTRAL) were searched for relevant studies up to and including April 2019. Randomized controlled trials (RCTs), cohort and cross-sectional studies, surveys, and case reports with more than five patients were included. Records were imported into www.covidence.org. Electronic searches were supplemented by manual searches and reference linkage. The Preferred Reporting Items for Systematic Reviews and Meta Analysis (PRISMA) were followed. Although only one study fitted the population, intervention, comparison, outcome (PICO) framework, valuable information on AR drug holiday could be extracted from 14 of 371 reviewed articles. Among these, 3 were prospective and 11 were retrospective studies. These studies described or evaluated high-dose AR drug holidays. In 2 studies, patients were being treated with denosumab, but neither showed that a drug holiday was effective. The remaining 12 studies evaluated bisphosphonate treatment and 2 of these studies found no reason to use AR drug holiday before surgery. Three studies recommended drug holidays, whereas most of the studies recommended assessing each patient separately. The only paper that fitted the PICO approach was a non-randomized, prospective study with a control group. This study concluded that drug holiday was not necessary. Thus, there are no evidence for using drug holiday, but it is also clear that caused by a limited numbers of eligible patients, and a great variation in between these patient, high-level evidence for using AR drug holiday is almost impossible to obtain.

Novofumigatonin is a meroterpenoid found in the fungus Aspergillus novofumigatus. Here, the authors elucidate the biosynthetic pathway of novofumigatonin and show that the endoperoxidase NvfI and the endoperoxide isomerase NvfE are involved in it.

Purpose: To compare in a randomized controlled trial the 5-year clinical outcomes of (1) 3-unit fiber-reinforced resin-bonded fixed dental prostheses (RBFDPs) to 3-unit metal-ceramic RBFDPs; and (2) 3-unit fiber-reinforced RBFDPs to 2-unit cantilevered metal-ceramic RBFDPs.Materials and Methods: A consecutive sample of 50 young adult patients with tooth agenesis or trauma in the anterior region was included. The recruitment period was from 2005 to 2009, during which a total of 62 RBFDPs were inserted. In group A, 27 participants were randomly treated with 18 metal-ceramic 3-unit RBFDPs and 15 3-unit fiber-reinforced RBFDPs. In group B, 23 participants were randomly treated with 16 metal-ceramic 2-unit RBFDPs and 13 3-unit fiber-reinforced RBFDPs. All patients were followed up at baseline registration and at 1, 3, and 5 years. The primary outcome parameter was survival rate of the RBFDPs, and the secondary outcome parameters were periodontal and technical/esthetic outcomes and patient-reported evaluation of the RBFDPs.Results: Seven patients dropped out of the study during the 5-year observation period. In group A, the fiber-reinforced RBFDPs had a cumulative survival rate of 23% after 5 years, which was significantly less than for the 3-unit metal-ceramic RBFDPs. In group B, the cumulative 5-year survival rate was 36% for the fiber-reinforced RBFDPs and 91% for the 2-unit cantilevered metal-ceramic RBFDPs.Conclusion: The 3- and 2-unit metal-ceramic RBFDPs had a significantly higher 5-year survival rate than the 3-unit fiber-reinforced RBFDPs. The accessibility for oral hygiene practice was better with 2-unit compared to 3-unit RBFDPs, and the degree of gingivitis was lower. Purpose: Stress and achievement pressure constitute factors affecting young people’s mental health, especially among girls. Leisure participation holds the potential to be a collective space where young people can respond to stressors together. This study explores how girls collectively construct responses to daily stressors within the context of leisure participation. Methods: Nine focus groups were conducted with 16 girls aged 14–21 who were active members in two sport organizations in northern Sweden. Data was collected by using participatory observations and photo-elicited focus group discussions. Results: Our findings from the inductive thematic analysis were interpreted by combining the stress process model with social practice theory, resulting in three subthemes or responses: sharing sites of responsibility, resisting norms related to (gendered) youth and focused distraction. The subthemes were abstracted into the central theme of trustful belonging as a resource for collective responses, representing what pre-conditions need to be in place to make the responses possible. Conclusion: Leisure participation is an important relational space for young people to respond to stressors by making use of everyday routines, and the agency these social practices hold. However, the effort needed to respond to these stressors brought additional pressure in terms of responsibilities, and achievements.

Mosses from the genus Polytrichum have been shown to contain rare benzonaphthoxanthenones compounds, and many of these have been reported to have important biological activities. In this study, extracts from Polytrichum formosum were analyzed in vitro for their inhibitory properties on collagenase and tyrosinase activity, two important cosmetic target enzymes involved respectively in skin aging and pigmentation. The 70% ethanol extract showed a dose-dependent inhibitory effect against collagenase (IC50 = 4.65 mg/mL). The methanol extract showed a mild inhibitory effect of 44% against tyrosinase at 5.33 mg/mL. Both extracts were investigated to find the constituents having a specific affinity to the enzyme targets collagenase and tyrosinase. The known compounds ohioensin A (1), ohioensin C (3), and communin B (4), together with nor-ohioensin D (2), a new benzonaphthoxanthenone, were isolated from P. formosum. Their structures were determined by mass spectrometry and NMR spectroscopy. Compounds 1 (IC50 = 71.99 µM) and 2 (IC50 = 167.33 µM) showed inhibitory activity against collagenase. Compound 1 also exhibited inhibition of 30% against tyrosinase activity at 200 µM. The binding mode of the active compounds was theoretically generated by an in-silico approach against the 3D structures of collagenase and tyrosinase. These current results present the potential application from the moss P. formosum as a new natural source of collagenase and tyrosinase inhibitors.
