When shield machine traverses through hard rock mass, disc cutters will be abraded to different degrees. Excessive height difference exists between adjacent disc cutters, which critically affects the rock-breaking efficiency. Relying on a shield section of hard rock mass in Qingdao, the disc cutter abrasions are measured on site, and the reason for low construction efficiency is analysed. Subsequently, the discrete element method is used to study the rock-breaking law under different height differences of disc cutters. The ultimate cutter height difference for rock breaking is obtained. According to this, the cutter adjustment scheme is proposed to reduce the cutter height differences on site. Comparing shield tunnelling parameters before and after the cutter adjustment, it is found that the construction efficiency is significantly improved. By using a combination of genetic, pharmacological, and anatomical approaches, we show that the melanocortin 4 receptor (MC4R), implicated in the control of food intake and energy expenditure, also modulates erectile function and sexual behavior. Evidence supporting this notion is based on several findings: (i) a highly selective non-peptide MC4R agonist augments erectile activity initiated by electrical stimulation of the cavernous nerve in wild-type but not Mc4r-null mice; (ii) copulatory behavior is enhanced by administration of a selective MC4R agonist and is diminished in mice lacking Mc4r; (iii) reverse transcription (RT)-PCR and non-PCR based methods demonstrate MC4R expression in rat and human penis, and rat spinal cord, hypothalamus, brainstem, pelvic ganglion (major autonomic relay center to the penis), but not in rat primary corpus smooth muscle cavernosum cells; and (iv) in situ hybridization of glans tissue from the human and rat penis reveal MC4R expression in nerve fibers and mechanoreceptors in the glans of the penis. Collectively, these data implicate the MC4R in the modulation of penile erectile function and provide evidence that MC4R-mediated proerectile responses may be activated through neuronal circuitry in spinal cord erectile centers and somatosensory afferent nerve terminals of the penis. Our results provide a basis for the existence of MC4R-controlled neuronal pathways that control sexual function.

The ability of the alpha-subunit of G proteins to dissociate from the plasma membrane was supported in a DNA sequencing analysis involving G protein strains derived from rats. This interaction may be partly be explained by the depalmitoylation of amino-terminal cysteine residue. Experimental results further reveal that the dissociation process influences the activity of cytoskeletal proteins, which in turn, creates a substantial impact on receptor G protein coupling.

Abstract Background Previous research showed that individuals have a natural tendency to conform to others. This study investigated the temporal characteristics of neural processing involved in social conformity by recording participants’ brain potentials in performing a line judgment task. After making his initial choice, a participant was presented with the choices of four same-sex group members, which could be congruent or highly or moderately incongruent with the participant’s own choice. The participant was then immediately given a second opportunity to respond to the same stimulus. Results Participants were more likely to conform to the group members by changing their initial choices when these choices were in conflict with the group’s choices, and this behavioral adjustment occurred more often as the level of incongruence increased. Electrophysiologically, group choices that were incongruent with the participant’s choice elicited more negative-going medial frontal negativity (MFN), a component associated with processing expectancy violation, than those that were congruent with the participant’s choice, and the size of this effect increased as the level of incongruence increased. Moreover, at both levels of incongruence, the MFN responses were more negative-going for incongruent trials in which participants subsequently performed behavioral adjustment than for trials in which they stuck to their initial choices. Furthermore, over individual participants, participants who were more likely to conform to others (i.e., changing their initial choices) exhibited stronger MFN effect than individuals who were more independent. Conclusions These findings suggest that incongruence with group choices or opinions can elicit brain responses that are similar to those elicited by violation of non-social expectancy in outcome evaluation and performance monitoring, and these brain signals are utilized in the following behavioral adjustment. The present research complements recent brain imaging studies by showing the temporal characteristics of neural processing involved in social conformity and by suggesting common mechanisms for reinforcement learning in social and non-social situations.
