The volume of the hippocampus is a biomarker of incipient Alzheimer's disease and is reduced in schizophrenia, major depression and mesial temporal lobe epilepsy. Whereas many brain imaging phenotypes are highly heritable, identifying and replicating genetic influences has been difficult, as small effects and the high costs of magnetic resonance imaging (MRI) have led to underpowered studies. Here we report genome-wide association meta-analyses and replication for mean bilateral hippocampal, total brain and intracranial volumes from a large multinational consortium. The intergenic variant rs7294919 was associated with hippocampal volume (12q24.22; N = 21,151; P = 6.70 x 10(-16)) and the expression levels of the positional candidate gene TESC in brain tissue. Additionally, rs10784502, located within HMGA2, was associated with intracranial volume (12q14.3; N = 15,782; P = 1.12 x 10(-12)). We also identified a suggestive association with total brain volume at rs10494373 within DDR2 (1q23.3; N = 6,500; P = 5.81 x 10(-7)).

Het doel van het onderzoek is om inzicht te krijgen in motivationele processen tijdens organisatieverandering. Gebaseerd op het JD-R model wordt een positieve samenhang verwacht tussen persoonlijke en werkhulpbronnen, werkbeleving en organisatie uitkomsten. Verder wordt gekeken naar de modererende rol van persoonlijke hulpbronnen op de relatie tussen werkhulpbronnen en werkbeleving en de medierende rol van werkbeleving op de relatie tussen werkhulpbronnen en gedrag van werknemers. 98 werknemers uit verschillende sectoren die te maken hadden met verschillende organisatieveranderingen vulden de online vragenlijst in. Regressieanalyse liet zien dat de werkhulpbron open-systeem-waarden cultuur positief samenhangt met bevlogenheid en bevlogenheid positief samenhangt met team veranderprestatie. De voornaamste conclusie is dat organisatiecultuur een belangrijke voorspeller is voor bevlogenheid, wat positief samenhangt met team veranderprestatie. Dit biedt aanknopingspunten voor vervolgonderzoek naar motivationele processen binnen organisatieverandering. De verwachte moderatie en mediatie effecten kunnen niet bevestigd worden.

According to the JD-R model, job resources have been linked to several positive organizational outcomes (e.g. work engagement). This study examined possible mediators in the relationship between the change resource participation and two positive organizational outcomes (work engagement and adaptive performance). The hypothesized mediators consisted of one change attitude (willingness to change) and one change behavior (job crafting) which were sequentially analyzed. The data was collected via an online survey. The final sample consisted of 665 Dutch employees in different sectors all of which who were confronted with organizational change. The attitude willingness to change mediated the relationship between the change resource of participation and the change behavior of job crafting. Job crafting, in turn, mediated the relationship between willingness to change and work engagement and adaptive performance. Theoretical and practical implications of those findings are discussed.

Here, we report on the identification and characterization of a 22-amino acid CPP derived from the human milk protein, lactoferrin. The peptide exhibits a conformation-dependent uptake efficiency that is correlated with efficient binding to heparan sulfate and lipid-induced conformational changes. The peptide contains a disulfide bridge formed by terminal cysteine residues. At concentrations exceeding 10 mum, this peptide undergoes the same rapid entry into the cytoplasm that was described previously for the arginine-rich CPPs nona-arginine and Tat. Cytoplasmic entry strictly depends on the presence of the disulfide bridge. To better understand this conformation dependence, NMR spectroscopy was performed for the free peptide, and CD measurements were performed for free and lipid-bound peptide. In solution, the peptides showed only slight differences in secondary structure, with a predominantly disordered structure both in the presence and absence of the disulfide bridge. In contrast, in complex with large unilamellar vesicles, the conformation of the oxidized and reduced forms of the peptide clearly differed. Moreover, surface plasmon resonance experiments showed that the oxidized form binds to heparan sulfate with a considerably higher affinity than the reduced form. Consistently, membrane binding and cellular uptake of the peptide were reduced when heparan sulfate chains were removed.

The Default Mode Network (DMN) is a network of brain areas that is engaged when people are at rest. The medial prefrontal cortex, posterior cingulate cortex and the inferior parietal lobe are thought to be the core of the DMN. Different functional processes have been linked to the DMN. However it remains difficult to define the cognitions that are at work when one is at rest. Recently more studies have focussed on the changes of the DMN in childhood and old age. To understand the development of the DMN will expand our understanding of its function. In this review an overview will be given from the literature about the development of the DMN from infancy to old age and about the DMN in Autism Spectrum Disorders (ASD) and Alzheimer’s disease (AD). The findings of the development of the DMN are supported by results from other areas of research such as underlying brain anatomy. Finally, implications of the development of the DMN on the functionality will be discussed, along with the criticism directed at the concept of a default mode of brain function.

Item does not contain fulltext, The aim of this study was to investigate the physicochemical and biological properties of a newly developed calcium phosphate cement (CaP cement) implanted in cortical bone. CaP cement was injected as a paste into tibia cortical bone defects in goats. Polymethylmethacrylate (PMMA) bone cement was used as a control. The animals were killed after 3 days, 2, 8, 16 and 24 weeks. X-ray diffraction and Fourier transform infrared spectroscopy performed at retrieved samples showed that the CaP cement had set as a carbonate apatite and remained stable over time. Light microscopic evaluation showed that after 2 weeks the cement was in tight contact with the bone without any inflammatory reaction or fibrous encapsulation. At later time points, the CaP cement implants were totally covered by a thin layer of bone and osteoclasts, present at the interface, which were clearly resorbing the cement. At locations where CaP cement was resorbed, new bone was deposited. Transmission electron microscopy revealed that indeed a seamless contact existed between CaP cement and bone, as characterized by the occurrence of an electron dense line of 50-60 nm thick that covered the CaP cement. Osteoblasts, in contact with the cement, were depositing new bone. Although the bulk of the material was still in situ after 24 weeks, the progressive osteoclast resorption of the cement followed by new bone formation suggests that all of the material may be replaced eventually. In contrast to the CaP cement, the PMMA reference cement was always surrounded by a thin fibrous capsule. The results indicate that the investigated CaP cement is biocompatible, osteoconductive as well as osteotransductive and is a candidate material for use as a bone substitute.

RIVM rapport:The impact of the Betuwe-line, a new rail line from the harbour of Rotterdam to Germany, on CO2 and NOx emissions from the Netherlands depends on: *- Emission rates from rail lines versus alternative transport modes *- The amount of extra goods transported via the Netherlands *- The mode of transport of goods transported via the Netherlands. Now and in 2010/2015 emission factors for CO2 and NO2 per tonne kilometer are much higher for lorries than for inland shipping and rail transport. NOx emission factors for rail transport are even lower than for inland shipping. Of all goods that will be transported on the Betuwe line a maximum of 50% will be transported through other countries if the Betuwe-line is not constructed. The Betuwe-line will attract more goods from inland shipping than from road haulage. Depending on the shift between inland shipping, lorries and other countries in a situation without the Betuwe-line, total CO2 and NOx emissions from the transport of goods on the Dutch territory will be diminished by 0-2.5% as a result of the construction of the Betuwe-line.
