In this study, iron oxide nanoparticles supported on Bermuda grass-based activated carbon (Fe3O4/AC) was prepared, characterized and utilized to remove aqueous Cr(VI). By examining the crystal structure of loaded nanoparticles, the iron oxide nanoparticles contained FeO, Fe0 and Fe3O4. Among them, the Fe3O4 was proved to have the greatest influence on Cr(VI) removal. Kinetic and isotherm models revealed that the above-synthesized materials had ultrafast removal rate (3.08 mg/g/min) and excellent adsorption capacity (430.7 mg/g) for aqueous Cr(VI). Moreover, reusability of Fe3O4/AC was assessed and the results indicated that after third cycles, the absorbent maintained a 2.03-times higher adsorption capacity when using NaOH solution regeneration compared to HCl. A further increase of adsorption capacity was observed under ultraviolet conditions, which might be due to the oxidation of part Fe3O4 to γ-Fe2O3. The Cr(VI) removal mechanisms involved the reduction of Cr(VI) into Cr(III), electrostatic attraction, co-precipitation, ligand exchange and chelation.

In this study, Fe-Mn co-embed sodium alginate carbon spheres (Fe-Mn/SA-C) were prepared as a cost-effective adsorbent for aqueous Cd(II) removal by a facile pyrolysis method. The morphology and chemical composition of Fe-Mn/SA-C were explored with various characterized techniques. The results indicate that Fe-Mn/SA-C possess porous structures and abundant oxide-based functional groups. Fe-Mn/SA-C exhibits excellent adsorption properties for aqueous Cd(II) and the adsorption process is pH-dependent. The adsorption data could fit pseudo second-order and Langmuir models well, suggesting that chemisorption is a dominant adsorption mechanism and the maximum adsorption capacity is 280.11 mg−1. Thermodynamic analysis demonstrates that the adsorption is spontaneous and endothermic. The adsorption mechanisms of Cd(II) include the complexation of Cd(II) with oxygen-based functional groups, precipitation, and ion exchange. Finally, the granulated and magnetic Fe-Mn/SA-C is an efficient and practical adsorbent for cadmium-containing wastewater treatment.

Diesel supercritical combustion can effectively improve engine efficiency and reduce harmful emissions. Therefore, studying the trans-critical injection characteristics of diesel is of great significance for further understanding of diesel supercritical combustion. In this work, a diesel trans-critical injection experiment is first carried out to test the liquid and vapor jet structure. Then the six-component surrogate of diesel is used to represent the thermo-physical properties of diesel in the range from subcritical state to supercritical state. Based on these thermo-physical properties, the dense fluid approximation method based on Large Eddy Simulation is applied to simulate the trans-critical injection processes of diesel. The results show that there is only a short continuous cold liquid core near the nozzle exit in the diesel trans-critical injection. The dense fluid approximation method based on diesel six-component surrogate and Large Eddy Simulation can accurately predict the diesel trans-critical injection properties. In order to accurately simulate the diesel trans-critical injection, it is necessary to develop a reliable diesel surrogate that can accurately emulate diesel various properties. Generally, this work is helpful to understand the characteristics of diesel trans-critical injections, and the numerical model is valuable for the optimal design of diesel engines.

Blockchain technology, as the supporting technology in digital currency, has brought considerable changes to e-commerce sector with its decentralised, tamper-proof and anonymous working methods. Based on the characteristics and the tamper-proof traceability of the block chain, this study constructs the application architecture of the e-commerce platform integration system, which not only realises the collaborative computing on the multi-party secure data platform, but also maintains the confidentiality of the calculation verification and input data. Additionally, the actual consumption process of consumers is introduced for case analysis to verify the feasibility of the method, which guides the collaborative application of blockchain technology and multi-party security computing technology in the field of e-commerce.

Fear extinction forms a new memory but does not erase the original fear memory. Exposure to novelty facilitates transfer of short-term extinction memory to long-lasting memory. However, the underlying cellular and molecular mechanisms are still unclear. Using a classical contextual fear-conditioning model, we investigated the effect of novelty on long-lasting extinction memory in rats. We found that exposure to a novel environment but not familiar environment 1 h before or after extinction enhanced extinction long-term memory (LTM) and reduced fear reinstatement. However, exploring novelty 6 h before or after extinction had no such effect. Infusion of the β-adrenergic receptor (βAR) inhibitor propranolol and glucocorticoid receptor (GR) inhibitor RU486 into the CA1 area of the dorsal hippocampus before novelty exposure blocked the effect of novelty on extinction memory. Propranolol prevented activation of the hippocampal PKA-CREB pathway, and RU486 prevented activation of the hippocampal extracellular signal-regulated kinase 1/2 (Erk1/2)-CREB pathway induced by novelty exposure. These results indicate that the hippocampal βAR-PKA-CREB and GR-Erk1/2-CREB pathways mediate the extinction-enhancing effect of novelty exposure. Infusion of RU486 or the Erk1/2 inhibitor U0126, but not propranolol or the PKA inhibitor Rp-cAMPS, into the CA1 before extinction disrupted the formation of extinction LTM, suggesting that hippocampal GR and Erk1/2 but not βAR or PKA play critical roles in this process. These results indicate that novelty promotes extinction memory via hippocampal βAR- and GR-dependent pathways, and Erk1/2 may serve as a behavioral tag of extinction.

Individual differences in regulation of fear and extinction memory play significant roles in the aetiology development of post-traumatic stress disorder (PTSD). Previous animal based studies showed that the activity of β-adrenergic receptors (β-ARs) are involved in memory modulation. However in humans it is not clear that whether genetic variability in β-ARs contributes to individual differences of fear and extinction memory. In the current study, we investigated the role of a common single-nucleotide polymorphism of β2-adrenergic receptor (ADRB2) gene in fear memory acquisition, fear memory extinction, extinction recall and fear generalization in human participants. Ninety-one male participants were exposed to a Pavlovian fear conditioning and their fear responses were assessed by the skin conductance response. Participants were genotyped for a polymorphism (rs2400207) located within the promoter region of the human ADRB2. Differences between genotypes were observed in the extinction memory recall test but not in fear acquisition, extinction learning and fear generalization. Particularly, A-allele carriers of rs2400707 displayed successful retention of extinction memory and prevented the return of fear during recall test. The results revealed the involvement of human noradrenergic system in the retention of extinction memory and genetic variability in this system may underlie individual differences in PTSD. Furthermore, rs2400207 polymorphism of ADRB2 gene may play a key role in the treatment efficacy of PTSD and can be a basis for future studies investigating a personalized medicine for fear memory related disorders.

Fear is prone to return following extinction that is the basis of exposure therapy for fear-related disorders. Manipulations that enhance the extinction process can be beneficial for treatment. Animal studies have shown that fasting or caloric restriction can enhance extinction and inhibit the return of fear. The present study examined the effects of fasting on fear acquisition, extinction, and the return of fear in humans. One hundred and twenty-five male participants were randomized into a fasting group and food group and exposed to a Pavlovian fear conditioning paradigm. Changes in plasma cortisol and ghrelin levels were examined using enzyme-linked immunosorbent assays. One-night fasting had no effect on fear acquisition but enhanced fear extinction retention and prevented the return of fear, and this effect persisted for at least 6 months. This procedure was also effective for remote fear memory. Plasma ghrelin levels were elevated after fasting and had a negative relationship with the fear response in spontaneous recovery test. However, overnight fasting did not affect cortisol levels. These findings indicate that fasting enhances extinction retention and prevents the return of fear, without influencing fear memory formation. We propose that this novel procedure may open new avenues for promoting extinction-based therapies for fear-related disorders.

Rasmussen encephalitis (RE) is a rare neurological disorder characterized by unilateral inflammation of cerebral cortex and other structures, most notably the hippocampus, progressive cognitive deterioration, and pharmacoresistant focal epilepsy. The pathogenesis of RE with unilateral cortical atrophy and focal seizures is still enigmatic. Activation of adenosine A1 receptors (A1R) has been proven to prevent the spatial spread of seizures. We hypothesized that the epileptogenic mechanisms underlying RE are related to changes in neuronal A1R expression. Immunnohistochemistry was used to examine the expression of A1R and adenosine kinase (ADK) in cortical specimens from RE (n = 12), and compared with control cortical tissue. The quantification of A1R and ADK expression was evaluated by Western blot. A1R was predominantly localized in perinuclear of neurons and not in astrocytes or microglia. Upregulation of neuronal A1R was observed in the lesions of RE. Reactive astrocytes and subpopulation of remaining neurons demonstrated over-expression of the ADK within the lesions of RE. Significant increase of A1R and ADK expression in RE compared with controls was confirmed by Western blot. These results suggest that over-expression of ADK is a common pathologic hallmark of RE, and that upregulation of neuronal A1R in RE is crucial in preventing the spread of seizures.

Early-life stress in adolescence has a long-lasting influence on brain function in adulthood, and it is mostly recognized as a predisposing factor for mental illnesses, such as anxiety and posttraumatic stress disorder. Previous studies also indicated that adolescent predictable chronic mild stress (PCMS) in early life promotes resilience to depression- and anxiety-like behaviors in adulthood. However, the role of PCMS in associated memory process is still unclear. In the present study, we found that adolescent PCMS facilitated extinction and inhibited fear response in reinstatement and spontaneous recovery tests in adult rats, and this effect was still present 1 week later. PCMS in adolescence increased the activity of brain-derived neurotrophic factor (BDNF)-extracellular signal-regulated kinase 1/2 (ERK1/2) signaling in infralimbic cortex (IL) but not prelimbic cortex in adulthood. Intra-IL infusion of BDNF antibody and the ERK1/2 inhibitor U0126 reversed PCMS-induced enhancement of fear extinction. Moreover, we found that PCMS decreased DNA methylation of the Bdnf gene at exons IV and VI and elevated the mRNA levels of Bdnf in the IL. Our findings indicate that adolescent PCMS exposure promotes fear memory extinction in adulthood, which reevaluates the traditional notion of adolescent stress.

Background Nicotine craving and relapse often occurs after reactivation of nicotine reward memories. We recently developed a memory retrieval-reconsolidation interference procedure in which reactivation of nicotine reward memories by acute exposure to nicotine (the unconditioned stimulus, UCS) and then pharmacologically interfering with memory reconsolidation decreased relapse to nicotine seeking in rats and nicotine craving in smokers. Here we investigated underlying mechanisms. Methods In the first series of experiments, we trained rats for nicotine-induced conditioned place preference (CPP) or nicotine self-administration and ventricularly microinjected them with the protein synthesis inhibitor anisomycin immediately after UCS-induced memory retrieval. In the second series of experiments, we used TAI-FISH (tyramide-amplified immunohistochemistry–fluorescence in situ hybridization) to examine neural ensembles in basolateral amygdala (BLA) reactivated by nicotine conditioned stimulus (CS)- or UCS-induced memory retrieval. We then used the Daun02 chemogenetic inactivation procedure to selectively inhibit the nicotine UCS-reactivated BLA neuronal ensembles. Results Ventricular injections of the anisomycin immediately after nicotine UCS memory retrieval inhibited subsequent nicotine CPP and relapse to operant nicotine seeking after short or prolonged abstinence. More importantly, within the BLA, distinct neuronal ensembles encoded Pavlovian CPP and operant self-administration reward memories and nicotine (the UCS) injections in the homecage reactivated both neuronal ensembles. Daun02 chemogenetic inactivation of the nicotine-reactivated ensembles prevented both nicotine CPP and relapse to nicotine seeking. Conclusions Results demonstrate that the nicotine UCS-induced memory retrieval manipulation reactivates multiple nicotine reward memories that are encoded by distinct BLA neuronal ensembles that play a role in nicotine preference and relapse.

Deep brain stimulation (DBS) is an important effective treatment for pharmacoresistant epilepsy, but its mechanism is still not clear. In clinical application, the possible effects and mechanisms DBS in treatment of epilepsy are different according to different stimulation parameters. We reviewed the mechanisms of DBS in different frequencies including high frequency and low frequency in the treatment of epilepsy, covering the inhibition or excitation of synaptic, pathological neuronal and network activity, pathological rhythms or oscillation and neurotransmitter systems. As the onset and progression of seizure, it’s more frequent accompanied with cognitive and psychiatric comorbidities in patients with epilepsy. We review the possible mechanism of DBS in treating epilepsy associated comorbidities including cognitive and memory disorders. With the further development of clinical application and basic research of DBS, the processing of clinical individualized therapy of DBS in epilepsy and evaluation of effects of DBS in associated comorbidities may contribute to extend the understanding of the mechanisms of DBS for epilepsy and associated comorbidities.

Several preclinical studies have reported the rapid antidepressant effects of N-methyl-D-aspartate receptor (NMDAR) antagonists, although the underlying mechanisms are still unclear. Death-associated protein kinase 1 (DAPK1) couples GluN2B subunits at extrasynaptic sites to regulate NMDAR channel conductance. In the present study, we found that chronic unpredictable stress (CUS) induced extracellular glutamate accumulation, accompanied by an increase in the DAPK1-NMDAR interaction, the high expression of DAPK1 and phosphorylated GluN2B at Ser1303, a decrease in phosphorylated DAPK1 at Ser308 and synaptic protein deficits in the rat medial prefrontal cortex (mPFC). CUS also enhanced GluN2B-mediated NMDA currents and extrasynaptic responses that were induced by bursts of high-frequency stimulation, which may be associated with the loss of astrocytes and low expression of glutamate transporter-1 (GLT-1). The blockade of GLT-1 in the mPFC was sufficient to induce depressive-like behavior and cause similar molecular changes. Selective GluN2B antagonist, DAPK1 knockdown by adeno-associated virus-mediated short-hairpin RNA or a pharmacological inhibitor, and the uncoupling of DAPK1 from the NMDAR GluN2B subunit produced rapid antidepressant-like effects and reversed CUS-induced alterations in the mPFC. The inhibition of DAPK1 and its interaction with GluN2B subunit in the mPFC also rescued CUS-induced depressive-like behavior 7 days after treatment. A selective GluN2B antagonist did not have rewarding effects in the conditioned place preference paradigm. Altogether, our findings suggest that the DAPK1 interaction with the NMDAR GluN2B subunit acts as a critical component in the pathophysiology of depression and is a potential target for new antidepressant treatments.

Cognition is impacted by aging. However, the mechanisms that underlie aging-associated cognitive impairment are unclear. Here we showed that cognitive decline in aged rats was associated with changes in DNA methylation of protein kinase Mζ (PKMζ) in the prelimbic cortex (PrL). PKMζ is a crucial molecule involved in the maintenance of long-term memory. Using different behavioral models, we confirmed that aged rats exhibited cognitive impairment in memory retention test 24 h after training and overexpression of PKMζ in the PrL rescued cognitive impairment in aged rats. After fear conditioning, the protein levels of PKMζ and the membrane expression of GluR2 increased in the PrL in young and adult rats but not in aged rats and the levels of methylated PKMζ DNA in the PrL decreased in all age groups, whereas the levels of unmethylated PKMζ DNA increased only in young and adult rats. We also found that environmentally enriched housing reversed the hypermethylation of PKMζ and restored cognitive performance in aged rats. Inactivation of PKMζ prevented the potentiating effects of environmental enrichment on memory retention in aged rats. These results indicated that PKMζ might be a potential target for the treatment of aging-related cognitive impairment, suggesting a potential therapeutic avenue.

In this paper, metabolic fingerprints were obtained from 102 nasopharyngeal carcinoma (NPC) patients and 107 healthy adults by gas chromatography-mass spectrometry (GC/MS). Partial least squares-discriminant analysis (PLS-DA) has revealed a pattern recognition discriminating the patients from controls, which sensitivity is 89.72% (96/107) and specificity is 85.29% (87/102). Furthermore, double-blind experiment was carried out and satisfactory results were obtained (total correct rate 87.30%). In addition, metabolites that most strongly influence this separation were obtained. The results indicated that a metabonomic approach is feasible and efficient and deserves further evaluation as a potential novel strategy for the detection of nasopharyngeal carcinoma.

Free fatty acids (FFAs), which are considered to be closely related with type 2 diabetes mellitus (T2DM), are not only the main energy source as nutrients, but also signaling molecules in insulin secretion. In this study, gas chromatography–mass spectrometry (GC–MS) coupled with two chemometric resolution methods, heuristic evolving latent projections (HELP) and selective ion analysis (SIA), was successfully applied to investigate plasma FFAs profiling of T2DM. Totally, twenty-three FFAs were identified and quantified. The results showed that HELP and SIA methods could be used to effectively handle overlapping peaks of GC–MS data and hence improve the qualitative and quantitative accuracy. Furthermore, a newly proposed competitive adaptive reweighted sampling (CARS) method coupled with partial least squares linear discriminant analysis (PLS-LDA) was introduced to seek the potential biomarkers. Finally, three fatty acids, oleic acid (OLA C18:1n-9), α-linolenic acid (ALA C18:3n-3), and eicosapentaenoic acid (EPA C20:5n-3), were identified as the potential biomarkers of T2DM for their powerful discriminant ability of T2DM patients from healthy controls. The study indicated that GC–MS combining with chemometric methods was a useful strategy to analyze metabolites and further screen the potential biomarkers of T2DM.

Volatile compositions of different parts (stem, branch and root barks) of cortex Magnolia officinalis, cultivated in China, were investigated for the first time by GC-MS with the help of heuristic evolving latent projection (HELP). Identification of components was conducted by similarity matching to NIST mass library but also assisted by comparison of temperature-programmed retention indices (PTRIs) with the data web available. A total of 90, 82 and 76 volatile compounds in the essential oils of the three samples taken from the same batch aforementioned were qualitatively and quantitatively determined, representing 84.03, 83.68 and 83.10% of the total content, respectively. Among the constituents determined, there were 50 components coexisting. Eudesmol and its isomers were shown to be the principal compounds in the studied samples, accounting for 47.66, 36.74 and 36.31%, respectively. The three kinds of isomers (alpha-, beta- and gamma-eudesmol) in houpo volatile oils have been tentatively qualified and quantified simultaneously for the first time. By comparative analysis, significant qualitative and semi-quantitative differences and similarities were observed among the three samples. The results achieved provide a scientific evidence for further exploitation of Magnolia bark and clinical medication.

Gas chromatography–mass spectrometry based metabolic profiling was used to characterize metabolic profiles in type 2 diabetes mellitus. We found distinct differences between type 2 diabetic patients and healthy controls, type 2 diabetic patients before and after treatment of the plasma metabolic profiles. Furthermore, levels of lactate, a-hydroxyisobutyric acid, phosphate, 1-monopalmitin and 1-monostearin were closely correlated with the disease state and may be considered as potential biomarkers for type 2 diabetes mellitus. The results demonstrated that plasma GC–MS metabolic profiling combined with multi- and uni-variate statistical analysis techniques provided an efficient way of depicting metabolic perturbations of diseases, and it may also be able to assist disease and treatment surveillance.

Focal cortical dysplasia (FCD) is a common cause of pharmacologically-intractable epilepsy, however, the precise mechanisms underlying the epileptogenicity of FCD remains to be determined. Neuropeptide Y (NPY), an endogenous anticonvulsant in the central nervous system, plays an important role in the regulation of neuronal excitability. Increased expression of NPY and its receptors has been identified in the hippocampus of patients with mesial temporal lobe epilepsy, presumed to act as an endogenous anticonvulsant mechanism. Therefore, we investigated whether expression changes in NPY receptors occurs in patients with FCD. We specifically investigated the expression of seizure-related NPY receptor subtypes Y1, Y2, and Y5 in patients with FCD versus autopsy controls. We found that Y1R and Y2R were up-regulated at the mRNA and protein levels in the temporal and frontal lobes in FCD lesions. By contrast, there was no significant change in either receptor detected in parietal lesions. Notably, overexpression of Y5R was consistently observed in all FCD lesions. Our results demonstrate the altered expression of Y1R, Y2R and Y5R occurs in FCD lesions within the temporal, frontal and parietal lobe. Abnormal NPY receptor subtype expression may be associated with the onset and progression of epileptic activity and may act as a therapeutic candidate for the treatment of refractory epilepsy caused by FCD.

Importance A relapse into nicotine addiction during abstinence often occurs after the reactivation of nicotine reward memories, either by acute exposure to nicotine (a smoking episode) or by smoking-associated conditioned stimuli (CS). Preclinical studies suggest that drug reward memories can undergo memory reconsolidation after being reactivated, during which they can be weakened or erased by pharmacological or behavioral manipulations. However, translational clinical studies using CS-induced memory retrieval-reconsolidation procedures to decrease drug craving reported inconsistent results. Objective To develop and test an unconditioned stimulus (UCS)-induced retrieval-reconsolidation procedure to decrease nicotine craving among people who smoke. Design, Setting, and Participants A translational rat study and human study in an academic outpatient medical center among 96 male smokers (aged 18- 45 years) to determine the association of propranolol administration within the time window of memory reconsolidation (after retrieval of the nicotine-associated memories by nicotine UCS exposure) with relapse to nicotine-conditioned place preference (CPP) and operant nicotine seeking in rats, and measures of preference to nicotine-associated CS and nicotine craving among people who smoke. Intervention The study rats were injected noncontingently with the UCS (nicotine 0.15 mg/kg, subcutaneous) in their home cage, and the human study participants administered a dose of propranolol (40 mg, per os; Zhongnuo Pharma). Main Outcomes and Measures Nicotine CPP and operant nicotine seeking in rats, and preference and craving ratings for newly learned and preexisting real-life nicotine-associated CS among people who smoke. Results Sixty-nine male smokers completed the experiment and were included for statistical analysis: 24 in the group that received placebo plus 1 hour plus UCS, 23 who received propranolol plus 1 hour plus UCS, and 22 who received UCS plus 6 hours plus propranolol. In rat relapse models, propranolol injections administered immediately after nicotine UCS-induced memory retrieval inhibited subsequent nicotine CPP and operant nicotine seeking after short (CPP, d = 1.72, 95% CI, 0.63-2.77; operant seeking, d = 1.61, 95% CI, 0.59-2.60) or prolonged abstinence (CPP, d = 1.46, 95% CI, 0.42-2.47; operant seeking: d = 1.69, 95% CI, 0.66-2.69), as well as nicotine priming-induced reinstatement of nicotine CPP ( d = 1.28, 95% CI, 0.27-2.26) and operant nicotine seeking ( d = 1.61, 95% CI, 0.59-2.60) after extinction. Among the smokers, oral propranolol administered prior to nicotine UCS-induced memory retrieval decreased subsequent nicotine preference induced by newly learned nicotine CS (CS1, Cohen d = 0.61, 95% CI, 0.02-1.19 and CS2, d = 0.69, 95% CI, 0.10-1.28, respectively), preexisting nicotine CS ( d = 0.57, 95% CI, −0.02 to 1.15), and nicotine priming (CS1, d = 0.82, 95% CI, 0.22-1.41 and CS2, d = 0.78, 95% CI, 0.18-1.37, respectively; preexisting nicotine CS, d = 0.92, 95% CI, 0.31-1.52), as well as nicotine craving induced by the preexisting nicotine CS ( d = 0.64, 95% CI, 0.05-1.22), and nicotine priming ( d = 1.15, 95% CI, 0.52-1.76). Conclusions and Relevance In rat-to-human translational study, a novel UCS-induced memory retrieval-reconsolidation interference procedure inhibited nicotine craving induced by exposure to diverse nicotine-associated CS and nicotine itself. This procedure should be studied further in clinical trials.

This paper discussed the results of stated preference and choice models in express industry research. We established a binary logit model with three most influential factors of express service, such as transit time, delivery price, and distance between the demand point and express service point. Our analysis based on the survey data showed that different express choice behaviors in China are influenced by not only the three express service factors mentioned above but also a wide variety of customer personal factors, including gender, age, income, delivery frequency each month and goods type. Finally, the model is suggested to estimate express market share in china.

Maladaptive memories elicited by exposure to environmental stimuli associated with drugs of abuse are often responsible for relapse among addicts. Interference with the reconsolidation of drug memory can inhibit drug seeking. Previous studies have indicated that the dephosphorylation of the eukaryotic initiation factor 2 α-subunit (eIF2α) plays an important role in synaptic plasticity and long-term memory consolidation, but its role in the reconsolidation of drug memory remains unknown. The amygdala is required for the reconsolidation of a destabilized drug memory after retrieval of drug-paired stimuli. Here, we used conditioned place preference (CPP) and self-administration procedures to determine whether amygdala eIF2α dephosphorylation is required for the reconsolidation of morphine and cocaine memories in rats. We found that the levels of eIF2α phosphorylation (Ser51) and activating transcription factor 4 (ATF4) were decreased after reexposure to a previously morphine- or cocaine-paired context (i.e., a memory retrieval procedure) in the basolateral amygdala (BLA) but not in the central amygdala. Intra-BLA infusions of Sal003, a selective inhibitor of eIF2α dephosphorylation, immediately after memory retrieval disrupted the reconsolidation of morphine- or cocaine-induced CPP, leading to a long-lasting suppression of drug-paired stimulus-induced craving. Advanced knockdown of ATF4 expression in the BLA by lentivirus-mediated short-hairpin RNA blocked the disruption of the reconsolidation of morphine-induced CPP induced by Sal003 treatment. Furthermore, inhibition of eIF2α dephosphorylation in the BLA immediately after light/tone stimulus retrieval decreased subsequent cue-induced heroin-seeking behavior in the self-administration procedure. These results demonstrate that eIF2α dephosphorylation in the BLA mediates the memory reconsolidation of drug-paired stimuli.

Extinction therapy has been suggested to suppress the conditioned motivational effect of drug cues to prevent relapse. However, extinction forms a new inhibiting memory rather than erasing the original memory trace and drug memories invariably return. Perineuronal nets (PNNs) are a specialized extracellular matrix around interneurons in the brain that have been suggested to be a permissive factor that allows synaptic plasticity in the adolescent brain. The degradation of PNNs caused by chondroitinase ABC (ChABC) may generate induced juvenile-like plasticity (iPlasticity) and promote experience-dependent plasticity in the adult brain. In the present study, we investigated the effect of removing PNNs in the amygdala of rat on the extinction of drug memories. We found that extinction combined with intra-amygdala injections of ChABC (0.01 U/side) prevented the subsequent priming-induced reinstatement of morphine-induced and cocaine-induced, but not food -induced, conditioned place preference (CPP). Intra-amygdala injections of ChABC alone had no effect on the retention, retrieval, or relearning of morphine-induced CPP and storage of acquired food-induced CPP. Moreover, we found that the procedure facilitated the extinction of heroin- and cocaine-seeking behavior and prevented the spontaneous recovery and drug-induced reinstatement of heroin- and cocaine-seeking behavior. We also found that the effect of PNNs degradation combined with extinction may be mediated by the potentiation of several plasticity-related proteins in the amygdala. Altogether, our findings demonstrate that a combination of extinction training with PNNs degradation in the amygdala erases drug memories and suggest that ChABC may be an attractive candidate for the prevention of relapse.

Background: Currently approved medications for opioid addiction have shown clinical efficacy, but undesired side effects, dependence induced by the medications themselves, and low treatment compliance necessitate the need for novel therapies. Methods: A novel morphine-keyhole limpet hemocyanin conjugate vaccine was synthesized with 6-glutarylmorphine as the hapten and a lengthened linker of 6 carbon atoms. The titer and specificity of the triggered antibody were assessed by enzyme-linked immunosorbent assay. The effects of the vaccine on the morphine-induced elevation of dopamine levels in the nucleus accumbens were determined by high-performance liquid chromatography. The effects of the vaccine on morphine-induced locomotor sensitization and heroin-primed reinstatement of heroin self-administration were also assessed. Results: After subcutaneous administration in rats, the vaccine triggered a high antibody titer, with comparable specificity for morphine, 6-acetylmorphine, and heroin, but no interaction with dissimilar therapeutic opioid compounds, including buprenorphine, naloxone, and nalorphine, was observed. The vaccine significantly prevented the elevation of dopamine levels in the nucleus accumbens induced by a single morphine challenge. Moreover, the vaccine prevented the expression of morphine-induced locomotor sensitization and heroin-primed reinstatement of heroin seeking, suggesting its potential for preventing relapse. Conclusion: These results demonstrate that active immunization with the present vaccine induces a robust morphine/heroin-specific antibody response in rats and attenuates the behavioral effects of morphine and heroin.
