Epicardial cells regulate heart growth by secreting numerous growth factors and undergoing lineage specification into other cardiac lineages. However, the lack of specificmarker genes for epicardial cells has hindered the understanding of this cell type in heart development. Through the analysis of a cardiac single cell mRNA sequencing dataset, we identified a novel epicardial gene named Keratin 19 (Krt19). Further analysis of the expression patterns of Krt19 and Wt1, a well-known epicardial gene, revealed their preferences in major cardiac cell types. Using lineage-tracing analysis, we analyzed Krt19-CreER labeled cells at multiple time windows and found that it labels epicardial cells at both embryonic and neonatal stages. Furthermore, we studied the function of epicardial cells using a diphtheria toxin A chain (DTA)-based cell ablation system. We discovered that Krt19-CreER labeled cells are essential for fetal heart development. Finally, we investigated the function of Krt19-CreER and Wt1-CreER labeled cells in neonatal mouse development. We observed that the Krt19-CreER; Rosa-DTA mice displayed a smaller size after tamoxifen treatment, suggesting the potential importance of Krt19-CreER labeled cells in neonatal mouse development. Additionally, we found that Wt1-CreER; Rosa-DTA mice died at early stages, likely due to defects in the kidney and spleen. In summary, we have identified Krt19 as a new epicardial cell marker gene and further explored the function of epicardial cells using the Krt19-CreER and Wt1-CreER-mediated DTA ablation system. Combinations of synthetic polymers, such as poly(N-isopropylacrylamide) (PNIPAM), with natural biomolecules, such as keratin, show potential in the field of biomedicine, since these hybrids merge the thermoresponsive properties of PNIPAM with the bioactive characteristics of keratin. This synergy aims to produce hybrids that can respond to environmental stimuli while maintaining biocompatibility and functionality, making them suitable for various medical and biotechnological uses. In this study, we exploit keratin derived from wool waste in the textile industry, extracted via sulfitolysis, to synthesize hybrids with PNIPAM microgel. Utilizing two distinct methods—polymerization of NIPAM with keratin (HYB-P) and mixing preformed PNIPAM microgels with keratin (HYB-M)—resulted in hybrids with 20% and 25% keratin content, respectively. Dynamic light scattering (DLS) and transmission electron microscopic (TEM) analyses indicated the formation of colloidal systems with particle sizes of around 110 nm for HYB-P and 518 nm for HYB-M. The presence of keratin in both systems, 20% and 25%, respectively, was confirmed by spectroscopic (FTIR and NMR) and elemental analyses. Distinct structural differences were observed between HYB-P and HYB-M, suggesting a graft copolymer configuration for the former hybrid and a complexation for the latter one. Furthermore, these hybrids demonstrated temperature responsiveness akin to PNIPAM microgels and pH responsiveness, underscoring their potential for diverse biomedical applications. Acute and chronic wounds present a significant healthcare challenge, requiring innovative solutions for effective treatment. The exploitation of natural by-products with advanced cell regeneration potential and plant-based materials, which possess bioactive properties, is an innovative topic in wound management. This study investigates the potential of donkey gelatin and keratin for blending with natural bioactive extracts such as sumac, curcumin, and oak acorn to fabricate antioxidant and antimicrobial nanofibers with accelerated wound healing processes. The fabricated nanofibers possess good in vitro biocompatibility, except for the sumac-based donkey nanofibers, where cell viability significantly dropped to 56.25% (p < 0.05 compared to non-treated cells). The nanofiber dimensions showed structural similarities to human extracellular matrix components, providing an ideal microenvironment for tissue regeneration. The donkey nanofiber-based sumac and curcumin extracts presented a higher dissolution in the first 10 min (74% and 72%). Curcumin extract showed similar antimicrobial and antifungal performances to rivanol, while acorn and sumac extracts demonstrated similar values to each other. In vitro tests performed on murine fibroblast cells demonstrated high migration rates of 89% and 85% after 24 h in the case of acorn and curcumin nanofibers, respectively, underscoring the potential of these nanofibers as versatile platforms for advanced wound care applications. Recently, declining petroleum resources worldwide, increasing demand of developing economies for petroleum, and political and environmental concerns about fossil-based resources have led to an urgent need for environmentally friendly, renewable, sustainable, cost-effective, and biological resources in the packaging industry. Effective use of hide trimming and chicken feather waste is one of the sustainable ways to recycle solid waste from the leather and poultry industry. This research aimed to synthesize and characterize collagen hydrolysate (CH), chitosan (CS), and keratin (K) ternary films with different blend compositions (CH/CS/K, 50/50/0, 45/45/10, 40/40/20, 35/35/30, and 30/30/40). SDS-PAGE and FTIR results confirmed that CH isolated from hide trimming wastes was in type I structure, while the molecular weight of the extracted K samples was in the range of 42.5–60 kDa. The mechanical results of this study showed that the increase in the K ratio in the ternary-based films caused a significant increase in thickness and tensile strength (p < 0.05) and a significant decrease in the elongation at break (%). This study demonstrated that hide trimming and chicken feather waste could be used to produce a biodegradable film, which provides a way to maximize recycling and achieve sustainable development in the leather and poultry industry. Extraction parameters are important in determining the physicochemical properties of extracts. This work aimed to investigate the effect of extraction parameters on the physicochemical properties of keratin powder obtained from chicken feather wastes. Keratin was extracted using alkaline hydrolysis under different experimental conditions viz: concentration of sodium hydroxide, retention time and temperature. Extracted keratin powder was subjected to physicochemical, bulk powder and microbiological investigations. The powder was also subjected to high-resolution analyses such as scanning electron microscopy (SEM), Fourier transform infrared (FTIR) spectroscopy and differential scanning calorimetry (DSC). An optimal yield of 62% keratin was obtained from 3.0 M NaOH solution at 37°C and 24 hours’ reaction time. Extracted keratin powder was odourless, light-brown and insoluble in neutral and acidic solutions but soluble in alkaline medium. It exhibited a moisture sorption capacity of 98% with fair to good flowability. SEM revealed a rough surface powder particle with a mean diameter of 100 µm. FTIR spectroscopy revealed the presence of amide groups while DSC showed a melting point of 110°C. Extraction conditions impacted yield, physicochemical and powder properties of keratin from chicken feathers. AP collagen peptides (APCPs) are enzymatically decomposed collagen peptides that have been shown to promote hair growth and improve hair cuticle structure and thickness in mice. However, their impact on the elasticity and gloss of human hair has not been thoroughly investigated. Therefore, this study sought to identify the factors affecting hair elasticity and gloss in APCPs-treated human hair follicles (hHFs). Using Time-of-Flight Secondary Ion Mass Spectrometry (ToF-SIMS) analysis, changes in amino acid content related to hair elasticity and lipid components associated with hair gloss were measured. As a result, APCP treatment led to increases in proline and cysteine amino acid, as well as in lipid components such as 18-methyleicosanoic acid (18-MEA), lauric acid, oleic acid, 11,13-eicosenoic acid, behenic acid, methyleicosanoic acid and squalene. Fourier-transform infrared spectroscopy (FT-IR) analysis of hair cross-sections indicated an increase in the intensity of bonds such as S=O, C=O, cystine, C–N and C–H in APCPs-treated hHFs. Additionally, APCPs elevated the expression of Keratin 17, integral to the inner root sheaths (IRS) and outer root sheaths (ORS), as well as Keratins 31, 85, and 86, components of the hair cortex and medulla. APCPs treatment also resulted in increased bonded water and moisture content in hHFs, along with a reduction in cuticle surface roughness. These findings suggest that APCPs may have the potential to enhance hair elasticity and gloss.

* Context.--Differentiated vulvar intraepithelial neoplasia (dVIN) is a human papillomavirus-independent lesion with the potential for rapid progression to invasive squamous cell carcinoma (SCC). The histopathologic features of dVIN are diverse, have overlapping characteristics with lichen sclerosus (LS) and lichen simplex chronicus (LSC), and may be diagnosed by dermatopathologists or gynecologic pathologists because of the vulva's anatomic location. Objectives.--To identify the salient histopathologic features of dVIN, particularly those that predict progression to SCC, and to evaluate interobserver agreement in diagnosing dVIN within the same subspecialty and across subspecialties. Design.--One general surgical pathologist, 2 pathology-trained dermatopathologists, and 1 gynecologic pathologist blinded to the final diagnoses were asked to record 20 histopathologic features and to provide their final interpretations on cases of dVIN (n = 65), LS (n = 126), LSC (n = 112), and LS with LSC (n = 6). Results.--Interobserver agreement for the 4 diagnoses and 10 histopathologic features was moderate. Logistic regression analysis indicated that keratin pearls, basal pleomorphism, and basal layer disarray were independent variables for diagnosing dVIN (coefficients 1.95,1.97, and 0.91, respectively; P < .001) and progression to SCC (coefficients 1.96,1.20, and 1.08, respectively; P < .001). Conclusions.--There is no single histopathologic feature pathognomonic for dVIN; however, the presence of keratin pearls, basal pleomorphism, and basal layer disarray should raise high suspicion for dVIN and concurrent SCC. Bead-free nanofibrous scaffolds composed of PVOH/keratin/chitosan were prepared using electrospinning after optimising the process parameters using a Box–Behnken experimental design. Two quadratic models were developed to optimise the fibre diameter and the diameter of fibre beads considering the voltage, flow rate, spinning distance, and amount of biopolymer as independent variables. All independent variables were found to be significant in determining responses, although not all interactions among these were significant. The models were highly effective in describing responses, with an R2 of 98.58 and 99.67%. The optimum conditions were determined to be 15.82 kV voltage, 0.25 mL/h flow rate, 105 mm spinning distance, and 30% biopolymers. The accuracy of the models was verified and found to be within an acceptable range. The bead-free nanofibrous scaffold exhibited no cytotoxicity to Human Aneuploid Immortal Keratinocyte (HaCaT) and Normal Human Dermal Fibroblast (NHDF) cell lines, enabling cell adhesion and proliferation. Both cell lines remained attached with perfect cell morphology when co-cultured on the scaffold for 30 days, indicating the scaffold’s potential for biomedical applications.

Abstract Hair is an advantageous biological sample due to its recordable, collectable, and storable nature. Hair's primary components are keratin and keratin-associated proteins. Owing to its abundance of cystine, keratin possesses impressive mechanical strength and chemical stability, formed by creating disulfide bonds as crosslinks within the protein peptide chain. Furthermore, keratin is cross-linked with keratin-associated proteins to create a complex network structure that provides the hair with strength and rigidity. Protein extraction serves as the foundation for hair analysis research. Bleaching hair causes damage to the structure between keratin and keratin-associated proteins, resulting in texture issues and hair breakage. This article outlines various physical treatment methods and lysate analysis that enhance the efficiency of hair protein extraction. The PLEE method achieves a three-fold increase in hair protein extraction efficiency when using a lysis solution containing SDS and combining high temperatures with intense shaking, compared to previous methods found in literature. We utilized the PLEE method to extract hair from both normal and damaged groups. Normal samples identified 156–157 proteins, including 51 keratin and keratin-associated proteins. The damaged group consisted of 155–158 identified proteins, of which 48–50 were keratin and keratin-associated proteins. Bleaching did not cause any notable difference in the protein identification of hair. However, it did reduce coverage of keratin and keratin-associated proteins significantly. Our hair protein extraction method provides extensive coverage of the hair proteome. Our findings indicate that bleaching damage results in subpar hair quality due to reduced coverage of protein primary sequences in keratin and keratin-associated proteins.

The human vaginal epithelium is a crucial component of numerous reproductive processes and serves as a vital protective barrier against pathogenic invasion. Despite its significance, a comprehensive exploration of its molecular profiles, including molecular expression and distribution across its multiple layers, has not been performed. In this study, we perform a spatial transcriptomic analysis within the vaginal wall of human fetuses to fill this knowledge gap. We successfully categorize the vaginal epithelium into four distinct zones based on transcriptomic profiles and anatomical features. This approach reveals unique transcriptomic signatures within these regions, allowing us to identify differentially expressed genes and uncover novel markers for distinct regions of the vaginal epithelium. Additionally, our findings highlight the varied expressions of keratin ( KRT) genes across different zones of the vaginal epithelium, with a gradual shift in expression patterns observed from the basal layer to the surface/superficial layer. This suggests a potential differentiation trajectory of the human vaginal epithelium, shedding light on the dynamic nature of this tissue. Furthermore, abundant biological processes are found to be enriched in the basal zone by KEGG pathway analysis, indicating an active state of the basal zone cells. Subsequently, the expressions of latent stem cell markers in the basal zone are identified. In summary, our research provides a crucial understanding of human vaginal epithelial cells and the complex mechanisms of the vaginal mucosa, with potential applications in vaginal reconstruction and drug delivery, making this atlas a valuable tool for future research in women’s health and reproductive medicine.

Changes in keratin gene expression and spatiotemporal regulation determine the compositional content and cellular localization of wool keratin, thereby affecting wool traits. Therefore, keratin gene family member 32 (KRT32) was selected for a study using RT-qPCR, immunofluorescence, and penta-primer amplification refractory mutation system (PARMS) techniques. The results showed that KRT32 mRNA was highly expressed in the skin and localized to the inner root sheath (IRS), outer root sheath (ORS) and dermal papilla (DP). Sequencing results identified eight SNPs in KRT32, and association analyses revealed that the variations were significantly associated with multiple traits in wool (p < 0.05), including MFD, CF and MFC. The constructed haplotype combination H2H3 has higher CF and smaller MFD than other haplotype combination (p < 0.05). In conclusion, KRT32 can be used as a candidate gene for molecular genetic improvement of wool in Gansu Alpine Fine-wool sheep.

Although their favorable performance, appearance, and comfort attributes, woolen garments have the tendency to felt unenviably during washing. In this work, an eco-friendly, energy-saving, non-deteriorative process for the fabrication of machine-washable wool tops (WTs) was proposed. WTs were exposed to atmospheric air or an argon plasma treatment at different powers for various durations, followed by coating with a protein biopolymer, namely keratin, using the pad-dry-cure technique. The felting resistance of the finished WTs was evaluated, and the results revealed that the finished sample has an enhanced felting resistance to the extent of machine-washable wool (no felt ball is formed), whereas the untreated sample forms a felt ball with a diameter of 2.166 cm after undergoing the felting test. The treated WTs maintained their felting resistance after 20 wash cycles. The surface composition of the plasma-treated WTs was examined using X-ray photoelectron spectroscopy. The change in the chemical structure of the treated WTs was monitored by determining the cystine content, alkali solubility, base-combining capacity, and affinity to anionic dye. The dye exhaustion of the untreated and finished samples after 60 min was 66% and 98.8%, respectively. Scanning electron microscopy was used to study the wool fibers’ topography.

In this study, a novel and sustainable triboelectric material is successfully designed and developed by extracting keratin from recycled fur and combining it with chitosan to create a keratin/chitosan triboelectric nanogenerator (CK‐TENG) device. The device is designed for the simultaneous treatment of waste and the generation of green and clean energy, crucial for addressing global challenges. CK is prepared through a freeze‐drying method to overcome the weak mechanical properties of keratin and achieve a high surface contact area, which is important for triboelectric properties. The results showed that the output voltage is improved by 375% compared to the TENG without keratin, with values reaching 322 V under a 6 N. It produced a high power density of 14.4 W m−2 and excellent cyclic stability for more than 8000 cycles, which are crucial for practical applications. Further, the potential applications are also studied, and the results showed that it can turn on over 250 light‐emitting diodes (LEDs), display liquid crystal display (LCD), and charge capacitors under 6 N. Moreover, it is successful in harvesting energy from waste vibrations, acoustic energy, and as a shoe, and the electrical output is utilized to power electronic devices. Furthermore, the power density harvested by the device is higher than that of TENG devices made with keratin‐rich materials. Therefore, this study paves the way for the use of keratin‐based bio‐waste materials for clean and green energy production, which can play significant roles in applications such as the charging of electronic devices, biomedicine, sensors, and smart textiles. Introduction: The new topical formula is urgent needed to meet clinical needs for majority mild patients with psoriasis. Deucravacitinib exerts outstanding antipsoriatic capacity as an oral TYK2 inhibitor; however, single therapy is insufficient to target the complicated psoriatic skin, including excessive reactive oxygen species (ROS) and persistent inflammation. To address this need, engineered smart nano-therapeutics hold potential for the topical delivery of deucravacitinib. Methods: hydrophobic Deucravacitinib was loaded into polyethylene glycol block-polypropylene sulphide (PEG-b-PPS) for transdermal delivery in the treatment of psoriasis. The oxidative stress model of HaCaT psoriasis was established by TNF-a and IL-17A in vitro. JC-1 assay, DCFH-DA staining and mtDNA copy number were utilized to assess mitochondrial function. 0.75% Carbopol®934 was incorporated into SPMs to produce hydrogels and Rhb was labeled to monitor penetration by Immunofluorescence. In vivo, we established IMQ-induced psoriatic model to evaluate therapeutic effect of Car@Deu@PEPS. Results: Deu@PEPS exerted anti-psoriatic effects by restoring mitochondrial DNA copy number and mitochondrial membrane potential in HaCaT. In vivo, Car@Deu@PEPS supramolecular micelle hydrogels had longer retention time in the dermis in the IMQ-induced ROS microenvironment. Topical application of Car@Deu@PEPS significantly restored the normal epidermal architecture of psoriatic skin with abrogation of splenomegaly in the IMQ-induced psoriatic dermatitis model. Car@Deu@PEPS inhibited STAT3 signaling cascade with a corresponding decrease in the levels of the differentiation and proliferative markers Keratin 17 and Cyclin D1, respectively. Meanwhile, Car@Deu@PEPS alleviated IMQ-induced ROS generation and subsequent NLRP3 inflammasome-mediated pyroptosis. Conclusion: Deu@PEPS exerts prominent anti-inflammatory and anti-oxidative effects, which may offers a more patient-acceptable therapy with fewer adverse effects compared with oral deucravacitinib. Fossil feathers have transformed our understanding of integumentary evolution in vertebrates. The evolution of feathers is associated with novel skin ultrastructures, but the fossil record of these changes is poor and thus the critical transition from scaled to feathered skin is poorly understood. Here we shed light on this issue using preserved skin in the non-avian feathered dinosaur Psittacosaurus. Skin in the non-feathered, scaled torso is three-dimensionally replicated in silica and preserves epidermal layers, corneocytes and melanosomes. The morphology of the preserved stratum corneum is consistent with an original composition rich in corneous beta proteins, rather than (alpha-) keratins as in the feathered skin of birds. The stratum corneum is relatively thin in the ventral torso compared to extant quadrupedal reptiles, reflecting a reduced demand for mechanical protection in an elevated bipedal stance. The distribution of the melanosomes in the fossil skin is consistent with melanin-based colouration in extant crocodilians. Collectively, the fossil evidence supports partitioning of skin development in Psittacosaurus: a reptile-type condition in non-feathered regions and an avian-like condition in feathered regions. Retention of reptile-type skin in non-feathered regions would have ensured essential skin functions during the early, experimental stages of feather evolution. The evolution of feathers is associated with the evolution of related skin microstructures. Context: Improving the utilisation of alternative protein ingredients in livestock production will reduce feeding costs and improve industry sustainability. Feather meal (FM) is an abundant, alternative protein source with a high protein content but poor amino acid (AA) digestibility. Aim: This study evaluated strategies for improving AA digestibility of FM. Methods: Experiment 1 examined the effects of physical treatment with ultrasound, microwave and autoclaving on FM AA profile and digestibility. Experiment 2 evaluated the dietary addition of a protease (Ronozyme ProAct, 200 and 600 mg/kg; RPA) and a probiotic (BioPlus 400, 1500 mg/kg) on FM AA digestibility. Apparent ileal digestibility was determined by feeding each treatment to four replicate groups of six birds in Experiment 1 and five replicate groups of seven birds in Experiment 2, and then collecting the contents of the lower half of the ileum. Key results: None of the physical treatments improved (P > 0.05) the AA profile or ileal AA digestibility of FM. Dietary supplementation with RPA at 200 mg/kg or BioPlus 400 at 1500 mg/kg did not significantly (P > 0.05) influence the apparent ileal AA digestibility of FM. However, the higher concentration of RPA (600 mg/kg) significantly (P < 0.05) increased the apparent ileal AA digestibility of FM. Conclusion: The increased digestibility of FM by the protease and numerical increase (P < 0.1) by the probiotic (1500 mg/kg) presumably reflects keratinase activity of both feed supplements. Implications: The results of this study indicated that there is scope for further improvement in the nutritive value of FM for broilers. Feather meal (FM) is a major by-product of the poultry industry and is potentially a valuable source of crude protein and amino acids. However, the major constraint to its use is the feather protein keratin that reduces FM digestibility. The aim of this study was to explore technologies to improve FM digestibility. The frontiers of antibacterial materials in the biomedical field are constantly evolving since infectious diseases are a continuous threat to human health. In this work, waste-wool-derived keratin electrospun nanofibers were blended with copper by an optimized impregnation procedure to fabricate antibacterial membranes with intrinsic biological activity, excellent degradability and good cytocompatibility. The keratin/copper complex electrospun nanofibers were multi-analytically characterized and the main differences in their physical–chemical features were related to the crosslinking effect caused by Cu2+. Indeed, copper ions modified the thermal profiles, improving the thermal stability (evaluated by differential scanning calorimetry and thermogravimetry), and changed the infrared vibrational features (determined by infrared spectroscopy) and the chemical composition (studied by an X-ray energy-dispersive spectroscopy probe and optical emission spectrometry). The copper impregnation process also affected the morphology, leading to partial nanofiber swelling, as evidenced by scanning electron microscopy analyses. Then, the membranes were successfully tested as antibacterial materials against gram-negative bacteria, Escherichia coli. Regarding cytocompatibility, in vitro assays performed with L929 cells showed good levels of cell adhesion and proliferation (XTT assay), and no significant cytotoxic effect, in comparison to bare keratin nanofibers. Given these results, the material described in this work can be suitable for use as antibiotic-free fibers for skin wound dressing or membranes for guided tissue regeneration. [ABSTRACT FROM AUTHOR]

Background: The immune microenvironment impacts tumor growth, invasion, metastasis, and patient survival and may provide opportunities for therapeutic intervention in pancreatic ductal adenocarcinoma (PDAC). Although never studied as a potential modulator of the immune response in most cancers, Keratin 17 (K17), a biomarker of the most aggressive (basal) molecular subtype of PDAC, is intimately involved in the histogenesis of the immune response in psoriasis, basal cell carcinoma, and cervical squamous cell carcinoma. Thus, we hypothesized that K17 expression could also impact the immune cell response in PDAC, and that uncovering this relationship could provide insight to guide the development of immunotherapeutic opportunities to extend patient survival. Methods: Multiplex immunohistochemistry (mIHC) and automated image analysis based on novel computational imaging technology were used to decipher the abundance and spatial distribution of T cells, macrophages, and tumor cells, relative to K17 expression in 235 PDACs. Results: K17 expression had profound effects on the exclusion of intratumoral CD8+ T cells and was also associated with decreased numbers of peritumoral CD8+ T cells, CD16+ macrophages, and CD163+ macrophages (p < 0.0001). The differences in the intratumor and peritumoral CD8+ T cell abundance were not impacted by neoadjuvant therapy, tumor stage, grade, lymph node status, histologic subtype, nor KRAS, p53, SMAD4, or CDKN2A mutations. Conclusions: Thus, K17 expression correlates with major differences in the immune microenvironment that are independent of any tested clinicopathologic or tumor intrinsic variables, suggesting that targeting K17-mediated immune effects on the immune system could restore the innate immunologic response to PDAC and might provide novel opportunities to restore immunotherapeutic approaches for this most deadly form of cancer. Most reduced organic matter entering activated sludge systems is particulate (1–100-µm diameter) or colloidal (0.001–1-µm diameter), yet little is known about colonization of particulate organic matter by activated sludge bacteria. In this study, colonization of biopolymers (chitin, keratin, lignocellulose, lignin, and cellulose) by activated sludge bacteria was compared with colonization of glass beads in the presence and absence of regular nutrient amendment (acetate and ammonia). Scanning electron microscopy and quantitative PCR revealed chitin and cellulose were most readily colonized followed by lignin and lignocellulose, while keratin and glass beads were relatively resistant to colonization. Bacterial community profiles on particles compared to sludge confirmed that specific bacterial phylotypes preferentially colonize different biopolymers. Nitrifying bacteria proved adept at colonizing particles, achieving higher relative abundance on particles compared to bulk sludge. Denitrifying bacteria showed similar or lower relative abundance on particles compared to sludge. A wide variety of biomaterials has been developed to assist in wound healing, including acellular animal and human‐derived protein matrices. However, millions of patients worldwide still suffer from non‐healing chronic wounds, demonstrating a need for further innovation in wound care. To address this need, a novel biomaterial, the human keratin matrix (HKM), was developed, characterised, and tested in vitro and in vivo. HKM was found to be degradation‐resistant, and a proteomics analysis showed it to be greater than 99% human keratin proteins. PCR revealed adult human epidermal keratinocytes (HEKa) grown in contact with HKM showed increased gene expression of keratinocyte activations markers such as Epidermal Growth Factor (EGF). Additionally, a cytokine microarray demonstrated culture on HKM increased the release of cytokines involved in wound inflammatory modulation by both HEKa cells and adult human dermal fibroblasts (HDFa). Finally, in a murine chronic wound model, full‐thickness wounds treated weekly with HKM were smaller through the healing process than those treated with human amniotic membrane (AM), bovine dermis (BD), or porcine decellularized small intestinal submucosa (SIS). Trichostasis spinulosa is a disorder of hair follicles characterized by the retention of vellus telogen club hair, leading to the formation of comedo‑like lesions. It usually presents over the face and is frequently asymptomatic. We report a 53‑year‑old female who presented with multiple itchy, discrete, bluish‑black, 2–3 mm comedo‑like follicular papules and pustules on her breast and lower abdomen for the past 2 years. In‑vivo dermoscopy showed keratotic plugs with a tuft of hair. Extraction dermoscopy yielded a cystic structure filled with keratin and multiple vellus telogen club hairs. Histology showed a cyst lined by squamous epithelium containing abundant laminated keratinous debris and a vellus hair shaft. Truncal or breast involvement, as seen in the present case, is relatively rare, and can be pruritic, causing significant morbidity due to itching and secondary bacterial infections. Background: Keratin pearls are foci of central keratinization within concentric layers of squamous cells that can form under the clitoral prepuce and cause pain (clitorodynia); in-office removal of keratin pearls may reduce clitoral pain and improve sexual function. Aim: This study aims to investigate clitoral pain and sexual function in women with partial clitoral phimosis and keratin pearls before and after in-office lysis of clitoral adhesions with keratin pearl excision (LCA-KPE). Methods: A pre-post interventional study evaluated patients who underwent LCA-KPE between January 2017 and February 2023 in 2 metropolitan gynecology clinics specializing in vulvar pain. Patients presenting with keratin pearls and partial clitoral phimosis identified through retrospective chart review were asked to complete postprocedure questionnaires and provide subjective responses on clitoral discomfort, sexual function, sexual distress, and their experience with in-office LCA-KPE. Bivariate analyses with paired t tests were conducted to determine the effect of LCA-KPE. Qualitative data were analyzed with thematic coding. Outcomes: An 11-point pain visual analog scale was utilized to determine pre- and postprocedure clitoral discomfort and difficulty with orgasm. Female sexual dysfunction was measured with the Female Sexual Function Index (FSFI) and Female Sexual Distress Scale–Revised. Results: A total of 32 of 74 patients who met inclusion criteria completed postprocedure surveys (43% response rate). Mean clitoral pain for respondents was 6.91 at baseline and 2.50 after LCA-KPE (P <.001). Mean difficulty with orgasm was significantly decreased from 5.45 at baseline to 3.13 after LCA-KPE (P <.001). Participants had a mean FSFI total score of 17.68 after treatment compared with a mean total baseline FSFI of 12.12 (P =.017). The mean FSFI score for pain was 2.43 at follow-up compared with 1.37 at baseline (P =.049). There was no significant difference in the mean Female Sexual Distress Scale–Revised score before vs after the procedure (P =.27). Qualitative themes described the procedure as painful but worthwhile, with 77% of participants reporting the overall experience as positive. Recurrence rate overall was 28%, with a median of 2 repeat procedures. Clinical Implications: Recognizing keratin pearls as a structural cause of clitoral pain and offering in-office treatment is an important tool in addressing clitorodynia and improving sexual function. Strengths and Limitations: This is the largest study to date documenting the occurrence, identifying associated pain conditions, and evaluating procedural outcomes for clitoral keratin pearls. Keratin extracted (KE) from chicken feathers was used for the production of composite films comprising poly(e-caprolactone) (PCL) and keratin (PCL/KE films). The process involved the extraction of keratin from chicken feathers using a 0.1 M NaOH solution, followed by characterization via sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The PCL was synthesized through the ring-opening polymerization (ROP) of e-caprolactone (ε-CL) with Sn(Oct)2 as a catalyst. Films were prepared via solvent casting, including pure PCL films and those enriched with different weight percentages of KE (10%, 15%, 25%, and 30%). The films were characterized by differential scanning calorimetry (DSC), thermogravimetric analysis (TG), and scanning electron microscopy (SEM). SEM analysis revealed a more uniform incorporation of KE within the PCL matrix in the case of the 15% keratin-enriched film (PCL/KE15) as compared to other keratin percentages. The thermal analysis showed a positive influence of keratin on the thermal stability of the films. Keratinocytes viability and proliferation tests on the PCL/KE15 film demonstrated compatibility with cells. Simple Summary: Our society is increasingly concerned about the well-being of animals like dolphins, which can be affected by life under professional human care or by anthropogenic disturbances in the wild. To better understand and improve their welfare, scientists are exploring new, non-invasive methods to study the response to stressors in these animals. This research introduces an approach for measuring the stress response by analysing cortisol from epidermis samples, a method that does not require invasive procedures. We developed and tested a reliable technique to extract and measure cortisol levels from the epidermis of common bottlenose dolphins and belugas. Our findings show that this method works well and is accurate, even with very small epidermis samples. We also found that the amount of stress hormones can vary depending on the individual animal but not necessarily where the body sample was taken. This study is a step forward in non-invasively studying and enhancing the welfare of cetaceans, offering insights into their stress levels in a way that is safer and more comfortable for them, which could ultimately lead to better care and conservation practices for these species. Society is showing a growing concern about the welfare of cetaceans in captivity as well as cetaceans in the wild threatened by anthropogenic disturbances. The study of the physiological stress response is increasingly being used to address cetacean conservation and welfare issues. Within it, a newly described technique of extracting cortisol from epidermal desquamation may serve as a non-invasive, more integrated measure of a cetacean's stress response and welfare. However, confounding factors are common when measuring glucocorticoid hormones. In this study, we validated a steroid hormone extraction protocol and the use of a commercial enzyme immunoassay (EIA) test to measure cortisol concentrations in common bottlenose dolphin (Tursiops truncatus) and beluga (Delphinapterus leucas) epidermal samples. Moreover, we examined the effect of sample mass and body location on cortisol concentrations. Validation tests (i.e., assay specificity, accuracy, precision, and sensitivity) suggested that the method was suitable for the quantification of cortisol concentrations. Cortisol was extracted from small samples (0.01 g), but the amount of cortisol detected and the variability between duplicate extractions increased as the sample mass decreased. In common bottlenose dolphins, epidermal skin cortisol concentrations did not vary significantly across body locations while there was a significant effect of the individual. One of the primary complications in generating physiologically representative skin tissue is the inability to integrate vasculature into the system, which has been shown to promote the proliferation of basal keratinocytes and consequent keratinocyte differentiation, and is necessary for mimicking representative barrier function in the skin and physiological transport properties. We created a 3D vascularized human skin equivalent (VHSE) with a dermal and epidermal layer, and compared keratinocyte differentiation (immunomarker staining), epidermal thickness (H&E staining), and barrier function (transepithelial electrical resistance (TEER) and dextran permeability) to a static, organotypic avascular HSE (AHSE). The VHSE had a significantly thicker epidermal layer and increased resistance, both an indication of increased barrier function, compared to the AHSE. The inclusion of keratin in our collagen hydrogel extracellular matrix (ECM) increased keratinocyte differentiation and barrier function, indicated by greater resistance and decreased permeability. Surprisingly, however, endothelial cells grown in a collagen/keratin extracellular environment showed increased cell growth and decreased vascular permeability, indicating a more confluent and tighter vessel compared to those grown in a pure collagen environment. The development of a novel VHSE, which incorporated physiological vasculature and a unique collagen/keratin ECM, improved barrier function, vessel development, and skin structure compared to a static AHSE model. Background: Radical mastoidectomy is the outstanding standard technique for management of cholesteatoma. Long term studies showed increasing incidence of discharging cavities (1). Modified radical mastoidectomy was described to prevent recurrent cholesteatoma along with reconstruction of the ossicular hearing mechanismre. Exteriorization of the disease bearing areas is essential for effective follow up (2). However, there are many recesses which cannot be exteriorized regarding its anatomical location such as the mesotympanum. It will be a source of discharge or accumulation of keratin. Covering these recesses by grafting is an essential rather than optional step in all radical mastoidectomies to prevent problematic cavities. Aim of work: Demonstrate the necessity of grafting in radical mastoidectomy for obtaining dry cavity. Methods: A prospective, comparative study conducted from March 2016 to October 2019 on 60 cholesteatoma patients. Patients were randomized into 2 equal groups of 30 patients each. In group A, patients undergone modified radical mastoidectomy with grafting, while patients in group B have undergone radical mastoidectomy without grafting. Regular follow-up was done monthly for 1 year for both groups. Results: In group A, 22 patients have intact grafts, and dry well epithelialized cavities, 2 patients had perforated grafts, 5 patients had small discharging granulations and 1 patient had a keratin pearl under the graft. In group B, 19 patients suffered persistent discharging cavity, 8 patients had less discharge with keratin debris and granulations and 3 patients had dry cavities. Hair is an advantageous biological sample due to its recordable, collectable, and storable nature. Hair's primary components are keratin and keratin-associated proteins. Owing to its abundance of cystine, keratin possesses impressive mechanical strength and chemical stability, formed by creating disulfide bonds as crosslinks within the protein peptide chain. Furthermore, keratin is cross-linked with keratin-associated proteins to create a complex network structure that provides the hair with strength and rigidity. Protein extraction serves as the foundation for hair analysis research. Bleaching hair causes damage to the structure between keratin and keratin-associated proteins, resulting in texture issues and hair breakage. This article outlines various physical treatment methods and lysate analysis that enhance the efficiency of hair protein extraction. The PLEE method achieves a three-fold increase in hair protein extraction efficiency when using a lysis solution containing SDS and combining high temperatures with intense shaking, compared to previous methods found in literature. We utilized the PLEE method to extract hair from both normal and damaged groups. Normal samples identified 156–157 proteins, including 51 keratin and keratin-associated proteins. The damaged group consisted of 155–158 identified proteins, of which 48–50 were keratin and keratin-associated proteins. Bleaching did not cause any notable difference in the protein identification of hair. However, it did reduce coverage of keratin and keratin-associated proteins significantly. Our hair protein extraction method provides extensive coverage of the hair proteome. Chicken feathers represent a viable material for producing biochar adsorbents. Traditional slow pyrolysis methods often result in sulfur element losses from chicken feathers, whereas hydrothermal reactions generate substantial amounts of nutrient-rich hydrothermal liquor. Magnesium-modified high-sulfur hydrochar MWF was synthesized through magnesium modification, achieving a S content of 3.68%. The maximum equilibrium adsorption amounts of MWF for Cd2+ and Pb2+ were 25.12 mg·g−1 and 70.41 mg·g−1, respectively, representing 4.00 times and 2.75 times of WF. Magnesium modification elevated the sulfur content, pH, ash content, and electronegativity of MWF. The primary mechanisms behind MWF's adsorption of Cd2+ and Pb2+ involve magnesium ion exchange and complexation with C=O/O=C–O, quaternary N, and S functional groups. MWF maintains robust stability and antioxidative properties, even with low aromaticity levels. Given the lower energy consumption during hydrochar production, MWF offers notable carbon sequestration benefits. The hydrothermal solution derived from MWF is nutrient-rich. Following supplementation with inorganic fertilizer, the hydrothermal solution of MWF significantly enhanced bok choy growth compared to the control group. In general, adopting magnesium-modified hydrothermal reactions to produce hydrochar and converting the resultant hydrothermal solution into water-soluble fertilizer proves a viable strategy for the eco-friendly utilization of chicken feathers. Programmed death ligand 1 (PD-L1) is an immune checkpoint molecule that plays a crucial role in regulating antitumor immune responses. Canine mammary carcinomas (CMCs) are common tumors of dogs. Despite extensive studies on the heterogeneity of CMCs, there is still a lack of effective precision therapies for the treatment of CMCs. In this study, we aimed to investigate the correlation between PD-L1 mRNA and protein expression in CMCs and explore its association with histopathological grade and molecular markers, including the estrogen receptor, epidermal growth factor receptor 2, and cytokeratin 5/6 (CK5/6). Formalin-fixed paraffin-embedded samples were evaluated for PD-L1 mRNA expression using RNA in situ hybridization and PD-L1 protein expression using immunohistochemistry. We observed no substantial correlation between PD-L1 mRNA and protein expression in CMCs; however, PD-L1 mRNA levels were significantly higher in grade 3 than in grade 1 tumors (P =.001). In addition, we observed a positive correlation between PD-L1 protein expression and CK5/6 expression in CMCs (P =.032). These findings suggest that PD-L1 expression in CMCs is heterogeneous and may be regulated post-transcriptionally. Further studies are needed to explore the prognostic and therapeutic implications of PD-L1 expression in different molecular subtypes of CMCs and their potential as predictive biomarkers for immunotherapy. Background/Aim: Epitope H contains an O-linked N-acetylglucosamine (O-GlcNAcH) residue in a specific conformation or environment, recognized by a site-specific monoclonal mouse IgM antibody H. O-GlcNAcH occurs in several normal and pathological cells and in several polypeptides, including keratin-8 and vimentin, on the latter in cells under stress. Materials and Methods: In this work, we studied the distribution of O-GlcNAcH on cells of endocervical mucosa in 60 specimens of endocervical curettings, 10 of which contained 15 inflamed polyps. Results: In our results, expression of O-GlcNAcH was weak in the mucosa with <5% mucin-secreting cells and up to 30% of the polyps staining positively. All non-ciliated, non-mucin-secreting cells, normal and hyperplastic 'reserve' cells, as well as the cells of immature squamous metaplasia, showed strong diffuse cytoplasmic staining for O-GlcNAcH. In mature squamous epithelium, fewer than 5% of basal cells and all the intermediate and superficial cells showed cytoplasmic staining for O-GlcNAcH, whereas parabasal cells were negative. All ciliated cells showed patchy or diffuse cytoplasmic staining. Nuclear staining for O-GlcNAcH was weak with fewer than 5% of hyperplastic 'reserve' and ciliated cells staining positively. Moreover, mucosal fibroblasts were negative, whereas all stromal cells of the polyps showed strong cytoplasmic staining for O-GlcNAcH. Conclusion: O-GlcNAcH is: a) differentially expressed among the cellular elements of mucosa and polyps, b) upregulated in mucin-secreting cells of polyps, c) induced in stromal cells of inflamed polyps, and d) can be used as a marker to differentiate between 'reserve' (positive) and parabasal (negative) cells, which have similar morphology using conventional cytological stains. To develop and evaluate a novel human stratum corneum (SC) mimetic phospholipid vesicle-based permeation assay (PVPASC) model for in vitro permeation studies. Due to the increasing restrictions on the use of human and animal skins, artificial skin models have attracted substantial interest in pharmaceuticals and cosmetic industries. In this study, a modified PVPASC model containing both SC lipids and proteins was developed. The PVPASC model was optimized by altering the lipid composition and adding keratin in the formulation of large liposomes. The barrier properties were monitored by measuring the electrical resistance (ER) and permeability of Rhodamine B (RB). The modified PVPASC model was characterized in terms of the surface topography, solvent influence and storage stability. The permeation studies of the active components in Compound Nanxing Zhitong Plaster (CNZP) were performed to examine the capability of PVPASC in the application of skin penetration. The ER and Papp values of RB obtained from the optimized PVPASC model indicated a similar barrier property to porcine ear skin. Scanning electron microscope analysis demonstrated a mimic 'brick-and-mortar' structure. The PVPASC model can be stored for three weeks at −20 °C, and withstand the presence of different receptor medium for 24 h. The permeation studies of the active components demonstrated a good correlation (r2 = 0.9136) of Papp values between the drugs' permeation through the PVPASC model and porcine ear skin. We studied some fibrotic aspects of chronic interstitial pneumonitis in the lungs of dogs infected with Leishmania infantum. The lungs of eleven naturally infected dogs, twelve experimentally infected with two distinct strains of L. infantum (BH401 and BH46), and six uninfected (controls) dogs, were analyzed by histological, parasitological, and immunohistochemical studies. Conventional histology (HE), collagen deposition (Gomori's silver staining for reticulin collagen fibers), and immunohistochemistry for myofibroblast characterization were carried out based on the cellular expression of alpha-smooth muscle actin, vimentin, cytokeratin, E-cadherin, snail antigen homologue 1 (SNAI1) (Snail), and the cytokine expression of transforming growth factor-beta (TGF-β). Parasitological screening was carried out using conventional polymerase chain reaction (PCR) and the immunohistochemical reaction of streptavidin–peroxidase for visualizing Leishmania amastigotes. Dogs naturally infected with L. infantum and experimentally infected with L. infantum BH401 strains showed intense interstitial pneumonitis characterized by thickening of the alveolar septa as a consequence of an intense diffuse and focal (plaques) chronic exudate of mononuclear cells associated with fibrogenesis. The expression of alpha-actin, vimentin, and TGF-β was higher in the lung interstitium of all infected dogs than in the other two groups (BH46 strain and controls). Moreover, in both the naturally and experimentally infected dog (BH401 strain) groups, the expression of Snail was moderate to intense in contrast to the other groups. Based on these immunohistochemical results, we concluded that mesenchymal cells are active in promoting changes in the extracellular matrix in the lungs of dogs naturally and experimentally infected with L. infantum, but it depends on the virulence of the parasite. A simple and sensitive colorimetric method for organophosphorus pesticides (Ops) detection was proposed via a multi-enzyme cascade reaction. The coordination compound of wool keratin and Cu (WK-Cu) was synthesized by self-assembly and used as a key catalyst in the cascade reaction. The enzymatic kinetics curve was established to explore the catalytic property, and the obtained Km and Vmax proved that WK-Cu has a good peroxidase-like activity. Ops could inhibit acetylcholinesterase activity and trigger the multienzyme cascades process, resulting in an amplified response signal. The key conditions were optimized, such as enzyme-linked reaction time and the concentration of each reactant in Ops detection. The detection range of phoxim was 10-1000 µg/L with a detection limit of 2.31 µg/L. Compared with other colorimetric detections, the proposed method exhibited a wider detection range and a relatively lower detection limit. In addition, this colorimetric method showed good universality and anti-interference ability for the analysis of phoxim in apple, cabbage and cauliflower samples. Improving eco‐friendly coating‐based materials for wool artifacts, such as kilim fabric can inspire new‐fashion design avenues for conserving ancient cultural heritage. Thus, this research addresses an efficient treatment agent from natural rosin acids (R) and dapsone (D) with variable sublethal proportions of synthesized titanium dioxide nanoparticles (TiO2 NPs) (i.e., 0.1%, 0.3%, and 0.6%) for modifying kilim fabric (KF) using a dip‐coating method. The nanostructure of prepared TiO2 nanoparticles and their particle size were examined by x‐ray diffraction (XRD) and Zetasizer analyzer. The data showed that the majority of prepared TiO2 oriented to a rutile‐type structure with an average hydrodynamic diameter of 48.30–49.65 nm. The KF‐R/D‐0.3% TiO2 NPs specimen provided the best tensile properties reached ~3192 N compared with the untreated KF (i.e., 1125 N). This finding confirmed by SEM observations. Whereas, the specimen containing decorated TiO2 NPs (i.e., 0.6%) possessed higher flammability resistance property compared with other specimens. Moreover, XRD and FTIR results for treated KF specimens demonstrated the significant role of TiO2 NPs, which react with KF keratin either by new linkages or through chelating geometry to form a compact KF matrix. This contributed to enhance the tensile and flammability properties. Additionally, a great achievement in antibacterial properties occurred when TiO2 NPs used and their inhibitory zones were 18–36 mm, depending on the bacteria and specimen type. Volumetric loss is one of the challenging issues in muscle tissue structure that causes functio laesa. Tissue engineering of muscle tissue using suitable hydrogels is an alternative to restoring the physiological properties of the injured area. Here, myogenic properties of type I collagen (0.5%) and keratin (0.5%) were investigated in a mouse model of biceps femoris injury. Using FTIR, gelation time, and rheological analysis, the physicochemical properties of the collagen (Col)/Keratin scaffold were analyzed. Mouse C2C12 myoblast-laden Col/Keratin hydrogels were injected into the injury site and histological examination plus western blotting were performed to measure myogenic potential after 15 days. FTIR indicated an appropriate interaction between keratin and collagen. The blend of Col/Keratin delayed gelation time when compared to the collagen alone group. Rheological analysis revealed decreased stiffening in blended Col/Keratin hydrogel which is favorable for the extrudability of the hydrogel. Transplantation of C2C12 myoblast-laden Col/Keratin hydrogel to injured muscle tissues led to the formation of newly generated myofibers compared to cell-free hydrogel and collagen groups (p < 0.05). In the C2C12 myoblast-laden Col/Keratin group, a low number of CD31+ cells with minimum inflammatory cells was evident. Western blotting indicated the promotion of MyoD in mice that received cell-laden Col/Keratin hydrogel compared to the other groups (p < 0.05). Despite the increase of the myosin cell-laden Col/Keratin hydrogel group, no significant differences were obtained related to other groups (p > 0.05). Exudate plays a crucial role in wound healing, but an excessive amount can lead to over-hydration of tissue and aggravating the infection and the injury. On the other hand, inadequate exudate can cause scarring and hinder healing. Traditional wound dressings are unable to regulate exudate levels based on the specific needs of the wound. To address this issue, a liquid-gated trilayered fibrous wound dressing capable of pumping fluid in a temperature-dependent manner is developed. This dressing comprises a hydrophilic cotton layer, a thermo-sensitive (TPPU) layer, and a hydrophobic layer of polyurethane (PU) nanofibers. The TPPU layer is constituted by nanofibers that are composed of an upper critical solution temperature (UCST)-type polymer, PU, and silver nanoparticles. The intermediate TPPU layer exhibits changes in its wettability upon heating, adjusting the thickness of the hydrophobic layer and ultimately achieving the appropriate structure for guiding spontaneous fluid transport. The positive effect of this novel dressing on diabetic wounds is observed, as it enhanced epithelialization and collagen synthesis while reducing inflammation, ultimately accelerating the wound healing process. Recent findings have greatly improved our understanding of mysticete oral filtration, and have upended the traditional view of baleen filtration as a simple process. Flow tank experiments, telemetric tag deployment on whales, and other lab and field methods continue to yield new data and ideas. These suggest that several mechanisms arose from ecological, morphological, and biomechanical adaptations facilitating the evolution of extreme body size in Mysticeti. Multiple lines of evidence strongly support a characterization of baleen filtration as a conceptually dynamic process, varying according to diverse intraoral locations and times of the filtration process, and to other prevailing conditions. We review and highlight these lines of evidence as follows. First, baleen appears to work as a complex metafilter comprising multiple components with differing properties. These include major and minor plates and eroded fringes (AKA bristles or hairs), as well as whole baleen racks. Second, it is clear that different whale species rely on varied ecological filtration modes ranging from slow skimming to high-speed lunging, with other possibilities in between. Third, baleen filtration appears to be a highly dynamic and flow-dependent process, with baleen porosity not only varying across sites within a single rack, but also by flow direction, speed, and volume. Fourth, findings indicate that baleen (particularly of balaenid whales and possibly other species) generally functions not as a simple throughput sieve, but instead likely uses cross-flow or other tangential filtration, as in many biological systems. Fifth, evidence reveals that the time course of baleen filtration, including rate of filter filling and clearing, appears to be more complex than formerly envisioned. Flow direction, and possibly plate and fringe orientation, appears to change during different stages of ram filtration and water expulsion. Sixth, baleen's flexibility and related biomechanical properties varies by location within the whole filter (=rack), leading to varying filtration conditions and outcomes. Seventh, the means of clearing/cleaning the baleen filter, whether by hydraulic, hydrodynamic, or mechanical methods, appears to vary by species and feeding type, notably intermittent lunging versus continuous skimming. Together, these and other findings of the past two decades have greatly elucidated processes of baleen filtration, and heightened the need for further research. Many aspects of baleen filtration may pertain to other biological filters; designers can apply several aspects to artificial filtration, both to better understand natural systems and to design and manufacture more effective synthetic filters. Understanding common versus unique features of varied filtration phenomena, both biological and artificial, will continue to aid scientific and technical understanding, enable fruitful interdisciplinary partnerships, and yield new filter designs. Background: The status of the sentinel lymph nodes (SLNs) was an important prognostic factor in varies cancers. A one-step nucleic acid amplification (OSNA) assay, a molecular-based whole-node analysis method based on CK19 mRNA copy number, was developed to diagnose lymph node metastases. We aimed to evaluate the value of OSNA for the diagnosis of sentinel lymph node metastasis in CK19 positive cancers. CK19 mRNA and protein expression for pan-caner analysis were obtained from TCGA and the Human protein atlas database. Methods: Two researchers independently searched the PubMed, Cochrane Library and Web of Science databases for qualified articles published before December 1, 2023. A meta-analysis was performed using MetaDisc and STATA. Risk bias and quality assessments of the included studies were evaluated, and a subgroup analysis was performed. Ten cancer types were found to be CK19 positively expressed and 7 of 10 had been reported to use OSNA for SLN detection. Results: After literature review, there were 61 articles included in the meta-analysis, which consisted of 7115 patients with 18007 sentinel lymph nodes. The pooled sensitivity and specificity of OSNA were 0.87 and 0.95 in overall patients. Moreover, we found the background CK19 expression in normal tissue affected the diagnostic accuracy of OSNA. In breast cancer, we performed subgroup analysis. OSNA exhibited to be a stable method across different population groups and various medical centers. In addition, when 250 copies/μl was chosen as the cutoff point of CK19 mRNA, there were a relatively higher sensitivity and AUC in detecting SLN micro-metastasis than 5000 copies/μl. Discussion: OSNA can predict the occurrence of SLN metastasis accurately in CK19 positive cancers, especially in breast cancer, colorectal cancer, lung cancer, gastric cancer and endometrial cancer. The massive amount of water-soluble urea used leads to nutrient loss and environmental pollution in both water and soil. The aim of this study was to develop a novel lignin-based slow-release envelope material that has essential nitrogen and sulfur elements for plants. After the amination reaction with a hydrolysate of yak hair keratin, the coating formulation was obtained by adding different loadings (2, 5, 8, 14 wt%) of aminated lignin (AL) to 5% polyvinyl alcohol (PVA) solution. These formulations were cast into films and characterized for their structure, thermal stability, and mechanical and physicochemical properties. The results showed that the PVA-AL (8%) formulation had good physical and chemical properties in terms of water absorption and mechanical properties, and it showed good degradation in soil with 51% weight loss after 45 days. It is suitable for use as a coating material for fertilizers. Through high-pressure spraying technology, enveloped urea particles with a PVA-AL (8%) solution were obtained, which showed good morphology and slow-release performance. Compared with urea, the highest urea release was only 96.4% after 30 days, conforming to Higuchi model, Ritger–Peppas model, and second-order dynamic model. The continuous nitrogen supply of PVA-AL coated urea to Brassica napus was verified by potting experiments. Therefore, the lignin-based composite can be used as a coating material to produce a new slow-release nitrogen fertilizer for sustainable crop production. [ABSTRACT FROM AUTHOR]

In tissue engineering and regenerative medicine applications, the utilization of bioactive materials has become a routine tool. The goal of tissue engineering is to create new organs and tissues by combining cell biology, materials science, reactor engineering, and clinical research. As part of the growth pattern for primary cells in an organ, backing material is frequently used as a supporting material. A porous three-dimensional (3D) scaffold can provide cells with optimal conditions for proliferating, migrating, differentiating, and functioning as a framework. Optimizing the scaffolds' structure and altering their surface may improve cell adhesion and proliferation. A keratin-based biomaterials platform has been developed as a result of discoveries made over the past century in the extraction, purification, and characterization of keratin proteins from hair and wool fibers. Biocompatibility, biodegradability, intrinsic biological activity, and cellular binding motifs make keratin an attractive biomaterial for tissue engineering scaffolds. Scaffolds for tissue engineering have been developed from extracted keratin proteins because of their capacity to self-assemble and polymerize into intricate 3D structures. In this review article, applications of keratin-based scaffolds in different tissues including bone, skin, nerve, and vascular are explained based on common methods of fabrication such as electrospinning, freeze-drying process, and sponge replication method. In the realm of military applications, the effective management of ice accumulation on strategic equipment without compromising the integrity of the electromagnetic signal transmission is a perennial challenge. To confront this issue, a revolutionary trilayered material composite known as MPE (metal-polyimide-electric heating layer) has been developed, demonstrating a sophisticated balance between deicing functionality and electromagnetic transparency. The MPE composite is ingeniously architected in a trilayer configuration, comprising a frequency-selective wave-transmissive stratum, an interjacent insulating dielectric interlayer, and an electric heating layer. The former is the result of a pioneering surface grafting modality of keratin, subsequently metallized with a nickel (Ni) coat via a chemical plating technique, which concurrently imparts the composite with a temperature-sensitivity range between −10 and 80 °C. The latter is formulated from a cyanate ester (CE) resin with organic conductive fillers, endowing the material with a high thermal threshold of up to 220 °C. Experimental evaluations of the MPE material have yielded a remarkable 88% transmissivity at the designated resonant frequency, a significant improvement over traditional graphene heating layer. This high level of performance, combined with the material's inherent deicing properties and the capacity for remote control via integrated sensing technology, positions the MPE as a substantial breakthrough for military operations. Keratinocyte dysdifferentiation and proinflammatory cytokine production play a central role in psoriatic inflammation. According to recent studies, the Rh family C glycoprotein (RHCG) enhances cell proliferation and disrupts cell differentiation. However, the specific role of RHCG psoriasis development remains unclear. We here explored the effect of RHCG on keratinocytes under psoriatic inflammation. The cell counting kit‑8 assay was conducted to assess proliferation. RHCG protein expression was assessed through western blotting and enzyme-linked immunosorbent assays. The expression of proinflammatory cytokines and differentiation markers was analyzed through a quantitative reverse-transcription polymerase chain reaction. Both RHCG mRNA and protein levels increased in psoriatic skin. Notably, cultured keratinocytes treated with an M5 cocktail, which mimics psoriatic inflammation, exhibited higher RHCG expression. Furthermore, RHCG overexpression promoted keratinocyte proliferation, accompanied by an increase in the production of interleukin (IL)-1β, IL-6, and IL-8, and tumor necrosis factor-α. RHCG overexpression also resulted in higher expression of keratin 17, a differentiation marker. Conversely, RHCG gene knockdown reduced keratinocyte proliferation and cytokine secretion. RHCG inhibition in cells recovered both keratin 1 and loricrin expression. Additionally, RHCG overexpression facilitated the phosphorylation of nuclear factor-kappa B and extracellular signal-regulated protein kinase signaling pathways. Importantly, when these signaling pathways were inhibited, the effect of RHCG on keratinocytes was attenuated. Promoting soil suppressiveness against soil borne pathogens could be a promising strategy to manage crop diseases. One way to increase the suppression potential in agricultural soils is via the addition of organic amendments. This microbe-mediated phenomenon, although not fully understood, prompted our study to explore the microbial taxa and functional properties associated with Rhizoctonia solani disease suppression in sugar beet seedlings after amending soil with a keratin-rich waste stream. Soil samples were analyzed using shotgun metagenomics sequencing. Results showed that both amended soils were enriched in bacterial families found in disease suppressive soils before, indicating that the amendment of keratin-rich material can support the transformation into a suppressive soil. On a functional level, genes encoding keratinolytic enzymes were found to be abundant in the keratin-amended samples. Proteins enriched in amended soils were those potentially involved in the production of secondary metabolites/antibiotics, motility, keratin-degradation, and contractile secretion system proteins. We hypothesize these taxa contribute to the amendment-induced suppression effect due to their genomic potential to produce antibiotics, secrete effectors via the contractile secretion system, and degrade oxalate—a potential virulence factor of R. solani —while simultaneously possessing the ability to metabolize keratin.
