Abstract

The baculovirus vector expression system is considered a useful insect cell-based platform for large-scale production of recombinant biopharmaceutical glycoproteins. The GA733 antigenic protein is a 40 k Da cell surface glycoprotein that is overexpressed in human colorectal carcinoma. Three GA733-expressing baculovirus vectors were constructed: (i) GA733 fused to the Fc fragment of human immunoglobulin G ( GA733- Fc); (ii) GA733- Fc tagged with the endoplasmic reticulum ( ER) retention signal KDEL ( GA733- Fc K); and (iii) GA733- Fc K without the signal peptide [GA733-FcK (w/o SP)]. These three recombinant proteins were expressed in Fall Armyworm ( S podoptera frugiperda) Sf9 insect cells. GA733- Fc purified from insect culture medium ( CM) and cell lysate ( CL) ( GA733I- Fc CM and GA733I- Fc CL, respectively), and GA733- Fc K and GA733- Fc K (w/o SP) purified from CL [GA733I-FcCL and GA733I-FcKCL (w/o SP), respectively] were injected intraperitoneally into BALB/c mice [three times at 1 μg, with or without an adjuvant (aluminum hydroxide)]. Surface plasmon resonance ( SPR) analysis (involving a chip with the immobilized GA733 antigen) showed that, in general, the serum samples from mice immunized with the adjuvant yielded stronger SPR signals than did those from mice immunized without an adjuvant. The serum samples from mice after the second injection yielded stronger signals than did those from mice before the second injection. The group immunized with GA733I- Fc KCL (glycosylated with an oligomannose) showed the strongest SPR signals. The SPR signals from the GA733I- Fc KCL group were stronger than or similar to those from mice immunized with mammalian-cell-expressed GA733- Fc ( GA733M- Fc). Abstract

GA733-2, an epithelial cell adhesion molecule highly expressed at the human colorectal carcinoma cell surface, is a candidate protein for the development of a colorectal cancer preventive vaccine. In this study, a signal peptide andc-terminal transmembrane domain truncated GA733-2 was fused to a His-tag, and the recombinant protein (rGA733-2) was expressed inEscherichia coliRosetta (DE3) pLysS. rGA733-2 was expressed in the form of inclusion bodies, and the protein was purified using either a serial dialyzed urea refolding method or metal ion exchange column after 8 M urea solubilization. The efficiency of rGA733-2 purification was increased three times using the urea refolding method compared to the column purification. A polyclonal antibody against the rGA733-2 was generated in rabbit using the refolded rGA733-2 as an antigen. The polyclonal antibodies (pAb rGA733-2) sensitively detected rGA733-2, which was heterologously expressed in bacteria and plants, and also detected native GA733-2 proteins in mouse and human cancer cells. pAb rGA733-2 showed high specificity against tobacco-expressed rGA733-2 in an immunoprecipitation assay. rGA733-2 and pAb rGA733-2 could serve as useful tools toward the development of a plant-derived colorectal cancer vaccine. Abstract

The baculovirus-insect cell expression system is widely used to produce recombinant proteins for various biomedical applications. Our previous study demonstrated that Ep CAM, a colorectal cancer vaccine candidate protein, can be expressed in the baculovirus-insect cell expression system. However, its functionality (the ability to elicit an immune response), which is important for its possible use as a colorectal cancer vaccine for immunotherapy, still needed to be confirmed. In this study, we examined the ability of recombinant Ep CAM to induce maturation of immature dendritic cells ( DCs) derived from CD34+ cells isolated from human umbilical cord blood. We demonstrated that Ep CAM induces the expression of four DC maturation markers: CD80, CD83, CD86 and MHC II. These results suggest that Ep CAM produced in the baculovirus-insect cell expression system is functional in terms of its ability to trigger maturation of human DCs. [ABSTRACT FROM AUTHOR]

Abstract

The baculovirus-insect cell expression system has been used to produce functional recombinant proteins. The antigen GA733 is a cell-surface glycoprotein highly expressed on most human colorectal carcinoma cells. Conditions for the expression of GA733 fused to the human immunoglobulin Ig G Fc fragment ( GA733- Fc) were optimized in the baculovirus expression system. Several variable factors were adjusted to optimize expression, including the cell line ( Sf9 and High Five), multiplicity of infection ( MOI) value (0.05, 0.1, 0.5, 1 and 3), post-infection time (48, 72 and 96 h) and harvested sample (cell culture media ( CM) or cell lysate ( CL)). In addition, two p Fast Bac Dual vectors carrying the GA733- Fc gene were constructed to express GA733- Fc with or without an endoplasmic reticulum ( ER) retention sequence KDEL and used to generate recombinant baculoviruses. Western blot showed that expression depended on the conditions used to express the recombinant proteins. The protein production level and secretion capability differed in each cell line. In Sf9 cells, the highest expression in the CM and CL was obtained with GA733- Fc at 96 h post-infection at 0.1 MOI and with GA733- FcK at 96 h post-infection at 3 MOI, respectively. In High Five cells, the highest expression in the CM and CL was obtained with GA733- Fc at 48 h post-infection at 1 MOI and with GA733- Fc K at 48 h post-infection at 3 MOI, respectively. These results suggest that the MOI value, post-infection time and subcellular localization affect expression, and that these conditions can be modified to optimize protein expression in the baculovirus-insect cell system.
