We describe the career of John Birks as a pioneering scientist who has, over a career spanning five decades, transformed palaeoecology from a largely descriptive to a rigorous quantitative science relevant to contemporary questions in ecology and environmental change. We review his influence on students and colleagues not only at Cambridge and Bergen Universities, his places of primary employment, but also on individuals and research groups in Europe and North America. We also introduce the collection of papers that we have assembled in his honour. The papers are written by his former students and close colleagues and span many of the areas of palaeoecology to which John himself has made major contributions. These include the relationship between ecology and palaeoecology, late-glacial and Holocene palaeoecology, ecological succession, climate change and vegetation history, the role of palaeoecological techniques in reconstructing and understanding the impact of human activity on terrestrial and freshwater ecosystems and numerical analysis of multivariate palaeoecological data. The varved sediment of Lake Suigetsu (central Japan) provides a valuable opportunity to obtain high-resolution, multi-proxy palaeoenvironmental data across the last glacial/interglacial cycle. In order to maximize the potential of this archive, a well-constrained chronology is required. This paper outlines the multiple geochronological techniques being applied - namely varve counting, radiocarbon dating, tephrochronology (including argon-argon dating) and optically stimulated luminescence ( OSL) - and the approaches by which these techniques are being integrated to form a single, coherent, robust chronology. Importantly, we also describe here the linkage of the floating Lake Suigetsu ( SG06) varve chronology and the absolute ( Int Cal09 tree-ring) time scale, as derived using radiocarbon data from the uppermost (non-varved) portion of the core. This tie-point, defined as a distinct (flood) marker horizon in SG06 (event layer B-07-08 at 1397.4 cm composite depth), is thus derived to be 11 255 to 11 222 Int Cal09 cal. years BP (68.2% probability range). Calibration is a fundamental stage of the radiocarbon (14C) dating process if one is to derive meaningful calendar ages from samples' 14C measurements. For the first time, the IntCal09 calibration curve (Reimer et al. 2009) provided an internationally ratified calibration data set across almost the complete range (0 to 50,000 cal BP) of the 14C timescale. However, only the last 12,550 cal yr of this record are composed of terrestrial data, leaving approximately three quarters of the 14C timescale necessarily calibrated via less secure, marine records (incorporating assumptions pertaining to the temporally variable "marine reservoir effect"). The predominantly annually laminated (varved) sediment profile of Lake Suigetsu, central Japan, offers an ideal opportunity to derive an extended terrestrial record of atmospheric 14C across the entire range of the method, through pairing of 14C measurements of terrestrial plant macrofossil samples (extracted from the sediment) with the independent chronology provided through counting of its annual laminations. This paper presents new data (182 14C determinations) from the upper (largely non-varved) 15 m of the Lake Suigetsu (SG06) sediment strata. These measurements provide evidence of excellent coherence between the Suigetsu 14C data and the IntCal09 calibration curve across the last ~12,000 cal yr (i.e. the portion of IntCal based entirely on terrestrial data). Such agreement demonstrates that terrestrial plant material picked from the Lake Suigetsu sediment provides a reliable archive of atmospheric 14C, and therefore supports the site as being capable of providing a high-resolution extension to the "wholly terrestrial" (i.e. non-reservoir-corrected) calibration curve beyond its present 12,550 cal BP limit. The oxygen and carbon isotope composition of sedimentary carbonates formed in Lake Hayq, a closed-basin lake in northern Ethiopia, suggests marked variation in the balance between precipitation and evaporation during the last 2000 years. Diatom stratigraphy shows that the lake remained deep enough to support planktonic taxa, and was never saline. The record suggests that rainfall was higher than that of the twentieth century during most of this time, especially during two short intervals centred on AD 700 and AD 1300. Climate was drier only around AD 800, and from AD 1750–1900. Similar, but slightly moister climate than today, with high interdecadal variability, prevailed from AD 800 to AD 1200, equivalent to the European ‘Mediaeval Warm Period’. A period of high effective precipitation followed, from AD 1200 to AD 1700, during the ‘Little Ice Age’. Pollen evidence suggests that increased forest cover from AD 1400 to AD 1750 was a response to higher rainfall; forest cover may have increased evaporative loss from the catchment, contributing to a subsequent trend of increasing lakewater δ18O values. Acquired drug resistance is the major cause for disease recurrence in cancer patients, and this is particularly true for patients with metastatic melanoma that carry a BRAF V600E mutation. To address this problem, we investigated cyclic membrane-active peptides as an alternative therapeutic modality to kill drug-tolerant and resistant melanoma cells to avoid acquired drug resistance. We selected two stable cyclic peptides (cTI and cGm), previously shown to have anti-melanoma properties, and compared them with dabrafenib, a drug used to treat cancer patients with the BRAF V600E mutation. The peptides act via a fast membrane-permeabilizing mechanism and kill metastatic melanoma cells that are sensitive, tolerant, or resistant to dabrafenib. Melanoma cells do not become resistant to long-term treatment with cTI, nor do they evolve their lipid membrane composition, as measured by lipidomic and proteomic studies. In vivo studies in mice demonstrated that the combination treatment of cTI and dabrafenib resulted in fewer metastases and improved overall survival. The development and application of lacustrine paleotemperature proxies based on microbial membrane lipid structures, including the TEX 86 and branched glycerol dialkyl glycerol tetraether (brGDGT) paleothermometers, have greatly advanced our understanding of the late-glacial and postglacial temperature history of Africa. However, the currently available records are from equatorial and southern hemisphere sites, limiting our understanding of the spatial patterns of temperature change. Here we use the brGDGT paleotemperature proxy to reconstruct Late Pleistocene and Holocene temperatures from Lake Tana, Ethiopia (12°N, 37°E). Following the termination of Heinrich Stadial 1 at ~ 15 ka, Lake Tana experienced a 3.7 °C oscillation over 1.2 ky. Temperatures then increased abruptly by nearly 7 °C between 13.8 and 13.0 ka, followed by a slow warming trend that peaked during the mid Holocene. Temperatures subsequently cooled from ~ 6 ka to ~ 0.4 ka. These data indicate that temperature at Lake Tana was sensitive to climate changes caused by variations in the Atlantic Meridional Overturning Circulation during the Late Pleistocene, as well as to regional hydroclimatic changes and reorganizations of the monsoons. Our record suggests that late-glacial temperature changes in northeast Africa were linked to high-latitude northern hemispheric climate processes, but that subsequent post-glacial temperature variations were strongly influenced by tropical hydrology.
