Abstract

Molecular cytogenetic techniques, such as in situ hybridization methods, are admirable tools to analyze the genomic structure and function, chromosome constituents, recombination patterns, alien gene introgression, genome evolution, aneuploidy, and polyploidy and also genome constitution visualization and chromosome discrimination from different genomes in allopolyploids of various horticultural crops. Using GISH advancement as multicolor detection is a significant approach to analyze the small and numerous chromosomes in fruit species, for example, Diospyros hybrids. This analytical technique has proved to be the most exact and effective way for hybrid status confirmation and helps remarkably to distinguish donor parental genomes in hybrids such as Clivia, Rhododendron, and Lycoris ornamental hybrids. The genome characterization facilitates in hybrid selection having potential desirable characteristics during the early hybridization breeding, as this technique expedites to detect introgressed sequence chromosomes. This review study epitomizes applications and advancements of genomic in situ hybridization (GISH) techniques in horticultural plants.

116. Cytogenetic assessment of Haworthiausing flow cytometry and fluorescence in situ hybridization

Abstract

The purpose of this study was to investigate cytogenetic variations between the nine Haworthiaspecies necessary for subsequent breeding. Flow cytometric analysis of H. badia‘Murasaki’, H. splendens, H. truncata‘Seiko’, H. cooperivar. obtusa, H. cymbiformis, H. mirabilisvar. mundula, and H. retusashowed a wide range of DNA content from 10.17 to 12.17 pg/1C, whereas two tetraploids, H. limifoliaand H. angustifoliavar. baylisii, showed higher DNA content of 16.81 and 12.64 pg/1C, respectively. Seven diploid Haworthiaexpressed three karyotype formulas, whereas two tetraploid Haworthiaeach expressed a different karyotype formula. Secondary constriction was observed in H. truncata‘Seiko’ and H. cooperivar. obtusa. Results of fluorescence in situ hybridization (FISH) showed a major difference between the diploid and tetraploid Haworthiain the distribution of 5S and 18S ribosomal DNA (rDNA) loci. There were eight loci in two out of seven diploids and both tetraploid Haworthia, while H. cymbiformisand H. mirabilisvar. mundulaeach showed seven 18S rDNA loci. H. badia‘Murasaki’, H. splendens, and H. retusahad four, three, and five 18S rDNA loci, respectively. In addition, a single pair of 5S rDNA loci was observed in all diploid Haworthiaexcept H. cooperivar. obtusa, which had four 5S rDNA loci. Each tetraploid Haworthiahad four 5S rDNA loci, with two strong and two weak signals. All 5S and 18S rDNA loci were observed on the long arm, in the subtelomeric or telomeric region, while a significant variation in chromosome length was observed among the Haworthia. This cytological analysis provides the necessary genetic information to elucidate the genetic makeup of newly introduced Haworthiahybrids.

117. LD50determination and phenotypic evaluation of three Echeveriavarieties induced by chemical mutagens

Abstract

Objective: The study aims to determine the Lethal Dose 50 (LD50) of Echeveriavarieties as induced by chemical mutagens. Methods: Three cultivated varieties from Echeveria species, namely ‘Brave,’ ‘Viyant,’ and ‘Snow bunny,’ were induced with chemical mutagens: colchicine, ethyl methanesulfonate (EMS), methyl methanesulfonate (MMS), and sodium azide (NaN3). Each mutagen was diluted to different concentrations: colchicine (0.2%, 0.4%, 0.6%, 0.8%, 1.0%), NaN3(0.02%, 0.04%, 0.06%, 0.08%, 0.1%), EMS, and MMS (0.1%, 0.2%, 0.3%, 0.4%, 0.5%). Soaking durations for each concentration level were 3, 6, 9, and 12 h. The survival rate and phenotypic data for mutated plants per variety in response to chemical mutagens were collected. Results: The LD50evaluation revealed maximum concentration and treatment duration vary per varieties. Regardless of varieties, EMS-treated leaf cuttings had the highest survival rate. However, upon phenotypic evaluation, the results revealed that mutagenic plants were only taken from those treated with colchicine. Conclusion: The use of colchicine to produce mutated succulents should be further investigated at the molecular level. The results of the study are highly beneficial for mutation breeding programs for other succulent varieties or other related crops.

118. Morphological characterization under different ecological habitats and physical mapping of 5S and 45S rDNA in Lilium distichum with fluorescence in situ hybridization

Abstract

BACKGROUND: This study was performed to investigate the phenotypic, karyomorphological, and habitat environment characteristics of Lilium distichum that grows naturally in South Korea. Currently, this species follows limited distribution areas and its natural populations are at the brink of extinction mainly due to fragmentation or destruction its natural habitat. RESULTS: This species was distributed between approximately 1,000 and 1,500 m above sea level with an average temperature of 22°C. The soil characteristics surrounding the natural habitats included loamy and silt loam soils having organic matter content (10.82%), pH (5.22), electrical conductivity (EC) (0.37 dS/m), total nitrogen (0.45%), and cation exchange capacity (34.3 cmol+/kg). The peak period of blossoming was between 27 July and 1 August. The maximum number of flowers was observed in Mount Deogyu (2.8), whereas the minimum number of flowers was observed in Mount Jiri (1.3) and Mount Seorak (1.2). Results regarding the number of verticillate leaf, bract counts, and verticillate leaf length and width were highest in Mount Odae, while lowest in the Mount Seorak region. The chromosome complement of L. distichum is 2n = 2x = 24; the length of somatic metaphase chromosomes ranges from 17.01 ± 0.32 μm (chromosome 10) to 32.06 ± 0.35 μm (chromosome 1) with a total length/genome of 261.92 μm. In L. distichum, the presence of 1 pair (two loci) of 5S ribosomal DNA (rDNA) and 8 pairs (16 loci) of 45S rDNA was revealed on metaphase chromosomes. One pair of 5S rDNA signal was observed in interstitial region of long arm of chromosome 3 which co-occurred with 45S rDNA. Among the eight pairs of 45SrDNA, three pairs of 45S rDNA signals were observed in short arm of chromosome (chromosome 2, 6, and 7) which were located close to centromere. The other five pairs of 45S rDNA signals were positioned in the interstitial region of long arm (chromosome 3, 4, 5, 10, and 11. CONCLUSIONS: This study provides baseline information regarding the effective exploitation and use of L. distichum resources for breeding research to be used as cut flower and potted plants.

Abstract

Adsorption of Hg(II) on polyaminated highly porous chitosan (PEI-CS) from washing solutions of refuse incinerator flue gases is technically feasible. Equilibrium isotherms for adsorption Hg(II) on PEI-CS were measured by a batch method. The experimental data correlate well with the Langmuir equation in the range of about 5 to 70 mol/m3 Hg(II) aqueous solution. When HCl exists in Hg(II) solution, the saturation capacity and equilibrium constant for adsorption of Hg(II) on PEI-CS decrease with increasing concentration of HCl, CHCl. When NaCl is in Hg(II) solution, the equilibrium constant is constant, but the saturation capacity decreases with an increase in the concentration of NaCl. Breakthrough curves for adsorption of 10 mol/m3 Hg(II) were measured using a PEI-CS packed column at Re′ = 5. The intraparticle effective diffusivities Deff of Hg(II) are determined from the experimental breakthrough curves. When HCl exists in Hg(II) solution, Deff increases with increasing C0, HCl when C0, HCl < 180 mol/m3. When C0, HCl > 180 mol/m3, it shows a constant value, which is about 8 times larger than that measured in pure Hg(II) aqueous solution. When NaCl is in Hg(II) solution, the shapes of the breakthrough curves are similar to those in pure Hg(II) solution. Deff is kept constant in the range of 0 ≤ C0, NaCl ≤ 1000 mol/m3.

139. Elegant lady a pink longiflorum lily cultivar suitable for cut flower forcing

Abstract

A new pink longiflorum (Lilium longiflorum x (L. longiforum x L. rubellum)) lily cultivar was obtained by integrated intersectional hybridization techniques. The F1 intersectional hybrid was obtained by crossing between L. longiflorum 'Gelria' and wild type L. rubellum. Since the F1 hybrid was absolutely sterile, amphidiploid of F1 hybrid was made by using in vitro mitotic chromosome doubling technique to restore its fertility. The amphidiploid of F1 interspecifc hybrid had relatively high frequency of pollen viability and was used for the subsequent backcrossing progeny. Backcrossing to the L. longiflorum 'Snow Queen using pollen of F1 hybrid produced a number of BC1 seedlings. Flower color of F1 hybrid (L. longiflorum x L. rubellum) was dark pink due to its male parent (L. rubellum) and BC1 progeny was mainly the same flower color as soft pink with similar flower shape and plant type. Chromosome analysis was performed to analyze the ploidy value and the variation at chromosome level. The ploidy level of the F1 hybrid was diploid (2n=2x=24) as expected. Each genome was transferred into F1 hybrid from both parents, and the amphidiploid after chromosome doubling was tetraploid (2n=4x=48) without any chromosomal variation. Genomic in situ hybridization (GISH) result shows that all BC1 plant derived from backcrossing was triploid (2n=3x=36) containing two copies of L.longiflorum chromosomes and one copy of L. rubellum chromosomes. Forcing days of F1 hybrid was intermediate between parents, and by back crossing the forcing days was delayed but earlier than its maternal parent L. longiflorum under standard growing conditions. On the other hand, Fusarium resistance was relatively decreased when compared to L. longiflorum. It is the first pink longiflorum as a commercial cultivar through intersectional hybridization between L. longiflorum and L. rubellum and already in favor on the Japanese market

Abstract

Sharp line structure is observed in the low energy shoulder of the 3000‐A absorption band of crystalline nitrates that have been cooled to 4°K. Single crystals of normal and of nitrogen‐15 isotopically substituted sodium and potassium nitrates were examined spectrographically. Correlation between the spacings, polarization and orientation properties, isotope effect, and crystal field splitting of the spectra permitted their analysis. In terms of the D3h point symmetry of a free nitrate ion the excited electronic state is E′. This electronic configuration is one of three considered likely upon the basis of a LCAO molecular orbital treatment of the ion. The two asymmetric E′ vibrations of the ion predominantly contribute to the band structure. A large Franck‐Condon shift, an extensive weakening of force constants, and the character of the excited vibrations indicate that the excited ion is planar but expanded with perhaps some, but not a great amount of asymmetric distortion. Predissociation appears at a level corresponding to a dissociation energy of 3.76 ev for the nitrate ion.
