Objective: To investigate changes of brain functional activity in patients with acute unilateral vestibulopathy (AUVP) using functional magnetic resonance imaging (fMRI). Methods: We studied 32 AUVP patients and 30 healthy controls (HC) who received resting-state fMRI scanning. Methods of voxel-based amplitude of low-frequency fluctuation (ALFF) and seed-based functional connectivity (FC) were adopted to compare the difference in brain function between the two groups. In addition, we evaluated the associations between abnormal neuroimaging results and clinical data in AUVP patients. Results: Compared with HC, patients with AUVP showed lower ALFF in brain regions of bilateral insular, right precentral gyrus, left inferior frontal gyrus and right middle frontal gyrus, as well as higher ALFF in left cerebellar anterior lobe. Using these abnormal brain areas as seeds, we observed decreased FC between left insular and left precuneus in AUVP patients. Furthermore, AUVP patients showed increased FC between left insular and left supplementary motor area. Results of correlation analysis indicated that ALFF value (z-value) in left insular was negatively correlated with the canal paresis value (p = 0.005, r = −0.483), and the FC (z-value) between left insular and left precuneus was negatively correlated with dizziness handicap inventory score (p = 0.012, r = −0.438) in patients with AUVP. Conclusion: Patients with AUVP during acute period showed altered functional activity and connectivity in brain regions mainly involved in motor control and vestibular information processing. These changes in brain functional activity and connectivity were potentially attributed to decreased vestibular input resulting from unilateral peripheral vestibular impairment. Simple Summary: Earlier studies confirmed that the pyridineamide insecticide flonicamid inhibits the Kir1 channel in Nilaparvata lugens (Stål) (Hemiptera: Delphacidae), thereby reducing insect feeding. This led to the hypothesis that Kir channels are the molecular target of flonicamid. This hypothesis was later contested by researchers who found that flumetnicam, a metabolite of flonicamid, strongly inhibits nicotinamidase (Naam). This inhibition causes nicotinamide accumulation in neurons, impairing the negative geotaxis behavior of D. melanogaster. As a result, many peers have now accepted Naam as the target of flonicamid. In our study, we observed that the reduced expression of Kir genes increased tolerance to both flonicamid and its metabolite, suggesting that Kir channels play a significant role in flonicamid's toxicity to D. melanogaster. Additionally, we discovered that flonicamid is more toxic to flies than its metabolite, even though it does not inhibit insect nicotinamidase, raising questions about the assumed role of nicotinamidase as the primary target. Inwardly rectifying potassium (Kir) channels regulate essential physiological processes in insects and have been identified as potential targets for developing new insecticides. Flonicamid has been reported to inhibit Kir channels, disrupting the functions of salivary glands and renal tubules. However, the precise molecular target of flonicamid remains debated. It is unclear whether flonicamid directly targets Kir channels or acts on other sites involved in the activation of transient receptor potential vanilloid (TRPV) channels. In this study, we observed that flonicamid is more toxic to flies than its metabolite, flumetnicam. This higher toxicity is difficult to reconcile if nicotinamidase is the active target, as flonicamid does not inhibit nicotinamidase. An alternative explanation is that flonicamid and flumetnicam may have distinct targets or act on multiple targets. Furthermore, reducing the expression of three individual Kir genes in the salivary glands of D. melanogaster significantly decreased the flies' susceptibility to both flonicamid and flumetnicam. The double knockdown of Kir1 with Kir3 or Kir2 with Kir3 further reduced the flies' sensitivity to both compounds. These findings confirm the involvement of Kir channels in mediating the toxic effects of flonicamid on flies. The lack of representative nasopharyngeal carcinoma (NPC) models has seriously hampered research on EBV carcinogenesis and preclinical studies in NPC. Here we report the successful growth of five NPC patient-derived xenografts (PDXs) from fifty-eight attempts of transplantation of NPC specimens into NOD/SCID mice. The take rates for primary and recurrent NPC are 4.9% and 17.6%, respectively. Successful establishment of a new EBV-positive NPC cell line, NPC43, is achieved directly from patient NPC tissues by including Rho-associated coiled-coil containing kinases inhibitor (Y-27632) in culture medium. Spontaneous lytic reactivation of EBV can be observed in NPC43 upon withdrawal of Y-27632. Whole-exome sequencing (WES) reveals a close similarity in mutational profiles of these NPC PDXs with their corresponding patient NPC. Whole-genome sequencing (WGS) further delineates the genomic landscape and sequences of EBV genomes in these newly established NPC models, which supports their potential use in future studies of NPC.

Introduction: AS2/LOB genes, a class of transcription factors ubiquitously existing in plants, are vital for plant growth, development, and stress tolerance. Despite the availability of the physic nut genome, information regarding the expression profiles and evolutionary histories of its AS2/LOB genes remains scarce. Methods: An elaborate exploration of the AS2/LOB gene family was conducted, including phylogeny, exon-intron structure, chromosomal location, conserved domain characteristics, conserved motifs, promoter cis-acting elements, protein interaction, and expression profiles under normal growth and abiotic stress conditions. Results: In this study, 28 AS2/LOB genes (JcASLs) were identified in the physic nut genome. Phylogenetic analysis, based on homologs from Arabidopsis, classified the 28 JcASLs genes into two groups (calss I and II). Chromosome localization indicated that the 28 JcASLs genes were unevenly distributed across nine chromosomes. RNA-seq and qRT-PCR results revealed that the majority of the 28 JcASLs genes exhibited differential expression in tissues such as roots, cortex stems, leaves, and seeds. Notably, JcASL8 and JcASL13 were exclusively expressed in seeds, and 16 JcASLs genes responded to drought and salt stress at least at one time point under at least one treatment condition. Discussion: These results establish a basis for future investigations into the molecular mechanism by which the JcASLs genes regulate physic nut's response to drought and salt stress and their role in modulating the growth and development of physic nut. In this article, an extended‐phase‐shift‐based pulse train control (EPS‐PT) for an intrinsically safe dual active bridge (DAB) converter is proposed to satisfy the safe and reliable power supply in harsh environments such as gas stations, chemical plants, and mining tunnels in complex power systems. A discrete pulse switching control method is introduced, and the input voltage and output current are sampled to calculate the primary target phase shift angle of the DAB converter, followed by the inner phase shift angle, while the secondary phase shift angle is rapidly adjusted by the voltage feedback. In order to meet the requirements of effective control and safe power supply in harsh environments, the system parameters are designed by comprehensively considering the control performance and intrinsically safe requirements. This study investigates the Chinese market's physicochemical properties and sensory attributes of 14 original-cut potato chip brands. Color characteristics, compositional analysis, sugar content, acrylamide levels, and textural properties were examined alongside sensory evaluations. Significant variations were observed across all the parameters. Color analysis revealed diverse L*, a*, and b* values, with total color difference (ΔE) strongly correlating with sensory scores (r = 0.73, p < 0.01). A compositional analysis showed substantial differences in protein (5.19–8.51%), fat (27.91–40.16%), and moisture (0.67–3.78%) contents. Acrylamide levels varied widely (166.7–1101.78 mg/kg), positively correlating with the sucrose content (r = 0.57, p < 0.05). A textural analysis demonstrated significant variations in hardness (379.38–1103.6 gf) and fracturability (167.5–857.77 gf), with fracturability negatively correlating with sensory scores (r = −0.75, p < 0.01). A sensory evaluation revealed distinct brand preferences, with the total scores ranging from 65 to 85. This comprehensive analysis provides valuable insights into the complex interplay between the physicochemical properties and consumer perception of potato chips in the Chinese market and offers potential directions for product optimization and quality control in the snack food industry, inspiring hope and innovation among industry professionals. The aims of this study were to investigate clinical factors associated with encephalitis relapse and chronic epilepsy development, and to evaluate the effectiveness of immunotherapy on encephalitis relapse. Methods: Patients with autoimmune encephalitis diagnosed as positive for neuronal surface antibodies in five general hospitals were included. A minimum 12-month follow-up period was conducted, and binary logistic regression analysis was used to identify predictors of encephalitis relapse and chronic epilepsy development. Additionally, decision curve analysis (DCA) was employed to assess the clinical net benefit of predicting encephalitis relapse and chronic epilepsy. Results: The study encompassed 65 patients with autoimmune encephalitis. The one-year relapse rate for encephalitis was 13.9%. The CASE score (P=0.045) was associated with encephalitis relapse, with subsequent immunotherapy proving beneficial in enhancing outcomes. Chronic epilepsy prevalence at one year was 26.2%, particularly higher among patients with positive LGI1 antibodies. Although adjustments in antiseizure medications were partially effective, 41.2% of patients developed drug-resistant epilepsy (DRE). DCA confirmed that the predictive models provided significant net clinical benefit in assessing the risk of encephalitis relapse and chronic epilepsy. Notably, the presence of diffuse cortical atrophy, medial temporal lobe atrophy, or cerebellar hemisphere atrophy was linked to relapsing encephalitis and chronic epilepsy. Conclusion: Most cases of autoimmune encephalitis are effectively managed, however, a minority of patients experience relapse or chronic epilepsy. The CASE score and LGI1 antibodies are independent risk factors for encephalitis relapse and chronic epilepsy development, respectively. Immunotherapy remains beneficial for relapsing patients, yet a portion may progress to DRE. Background: Perfluorooctane sulfonate (PFOS), an industrially synthesized persistent organic pollutant (POP), is intricately intertwined with human production and daily life. It has been discovered that PFOS is related to an elevated incidence of birth defects in fetuses. In contrast, melatonin (MLT), a hormone secreted by the pineal gland, has been demonstrated to exert a protective effect on reproductive development. Methods: This paper investigates the protective effect of MLT against PFOS‐induced reproductive toxicity by simultaneously orally administering MLT to pregnant mice exposed to PFOS. The therapeutic effect was evaluated through the monitoring of pregnancy outcomes, histological changes in the placenta, apoptosis and proliferation of placental spongiotrophoblast, as well as the expression of antioxidant enzyme genes, anti‐aging genes, anti‐inflammatory genes and other relevant genes. Results: The results of the study demonstrated that MLT treatment reversed the adverse pregnancy outcomes caused by toxic PFOS, including a low number of implanted fetuses, low neonatal fetal weight, and an increased number of resorbed fetuses. MLT treatment decreased the levels of MDA, an oxidation product generated by PFOS in the placenta of pregnant mice, and increased the levels of the antioxidant enzyme SOD. Additionally, MLT was able to maintain the normalization of placental structure, reduce apoptosis and sustain the proliferation of placental spongiotrophoblast by upregulating the expression of antioxidant genes (Nrf2, CAT) and anti‐aging gene (Klotho), anti‐inflammatory gene (Hsd11b2), thereby counteracting the oxidative stress caused by PFOS in the placenta, moreover, it also reduced the expression of inflammatory genes (Pycard) in the placenta. Conclusions: The findings firmly establish the effectiveness of MLT in mitigating the harmful impacts of tainted PFOS on reproductive development during pregnancy. To satisfy the requirements for transferring signals in an inductively coupled power transfer system, a novel power and signal composite modulation strategy is proposed using the binary frequency shift keying technology. The information‐carrying inverter control signal is generated by the logical combination of a signal carrier and the inverter control pulse using AND operation. Then a binary frequency shift keying method is used to modulate the signal carrier, and the resulting voltage ripples are injected to the bus at the output side as the signal carrier, to realize the signal and power synchronous transmission. At the signal receiver side, the output voltage ripple is sampled and filtered, and fast Fourier transform (FFT) is adopted for signal demodulation. The experimental test reveals that the proposed synchronous transmission method can achieve signal modulation and injection by modulating the frequency of power switches, and the transmitted signal can be demodulated by using FFT method with small time delay. This research can provide a new method for realizing power and signal synchronous transmission in an inductively coupled power transfer system. [ABSTRACT FROM AUTHOR]

Improving the reproductive ability and fertility of male ruminants is a central concern in animal husbandry. Phytogenic feed additives, known for their anti-inflammatory, antioxidant, and immunomodulatory properties, are commonly used in animal feed. Icariin (ICA), the primary flavonoid glucoside derived from Epimedium, is a traditional tonic in Chinese herbal medicine. However, its potential to enhance the reproductive performance of male ruminants remains unclear. In this study, twelve healthy adult male dairy goats were divided into two groups. The goats received oral administration of ICA at doses of 0 (control) and 50 mg/kg body weight daily for a consecutive period of 80 days during the breeding season. The effects of ICA on the reproductive performance was analyzed by histological examinations, semen quality analysis, and ELISA experiments. ELISA results showed a progressive increase in serum levels of GnIH, LH, and testosterone with the prolonged ICA treatment (p < 0.05). However, the serum concentration of GnRH in the ICA group initially increased, followed by a subsequent decrease (p < 0.05). The hypothalamic concentrations of dopamine (DA) and 5-hydroxytryptamine (5-HT) were significantly higher in the ICA group compared to the control group (p < 0.01). The CASA system analyzed sperm kinematics and revealed that ICA increased ejaculate volume, with both total motile and progressive motile sperm gradually increasing over time (p < 0.05). ICA did not affect the body weight of the goats but significantly increased the organ coefficient of the testes (p < 0.01). Additionally, there was an upregulation of hormone receptor expression in testicular tissue and an improvement in the antioxidant capacity of the testes after ICA treatment (p < 0.01). Furthermore, ICA was implicated in testosterone synthesis by modulating the expression of key enzymes associated with steroidogenesis and promoting the differentiation of spermatogonial stem cell to enhance spermatogenesis. In conclusion, our results indicate that icariin, as a phytogenic feed additive incorporated into the diet of ruminants, offers potential benefits in improving the reproductive performance of male dairy goats. Porcine reproductive and respiratory syndrome (PRRS) is a highly contagious viral disease that causes significant economic losses to the swine industry worldwide. PRRS virus (PRRSV) infection is a receptor-mediated endocytosis and replication process. The purpose of this study was to determine the localization and expression of four important PRRSV receptors in immunological organs of piglets. After piglets were infected with PRRSV, Hematoxylin and Eosin staining, immunofluorescence, and Western blot were used to perform histopathological examination and receptors distribution analysis. The results showed that PRRSV caused severe damage to the piglets' immune organs, including atrophy of the thymus and swelling of lymph node. Histopathological lesions were mainly observed in the lung and lymph node and were characterized by interstitial pneumonia, collapsed follicles, exhaustion of germinal centers, and extensive hemorrhage. Immunofluorescence staining and Western blot results showed that the receptors of CD163 and NMHCII-A were mainly distributed in the thymus, hilar lymph nodes, and mesenteric lymph nodes. However, Sn and vimentin receptors were expressed at low levels in the immune organs of piglets. The distribution of the four receptors in the immune organs was more concentrated in the cortex but was more scattered in the medulla. Compared to the control group, the relative expression of the four receptors increased significantly in most immune organs after viral infection. In conclusion, our study examined the distribution and expression of four PRRSV receptors in immunological organs. We observed a significant increase in the expression of Sn, CD163, and vimentin following viral infection.
