In many species, including mice, females show strikingly different pup-directed behaviors based on their reproductive state1,2. Naïve wild female mice often kill pups while lactating females are dedicated to pup caring3,4. The neural mechanisms that mediate infanticide and its switch to maternal behaviors during motherhood remain unclear. Here, based on the hypothesis that maternal and infanticidal behaviors are supported by distinct and competing neural circuits5,6, we used the medial preoptic area (MPOA), a key site for maternal behaviors7–11, as a starting point and identified three MPOA-connected brain regions that drive differential negative pup-directed behaviors. Further functional manipulation andin vivorecording revealed that estrogen receptor alpha (Esr1) expressing cells in the principal nucleus of the bed nucleus of stria terminalis (BNSTprEsr1) are necessary, sufficient, and naturally activated during infanticide in female mice. Furthermore, MPOAEsr1and BNSTprEsr1neurons form reciprocal inhibition and change their excitability in opposite directions with reproductive state. The shift in balance between BNSTprEsr1and MPOAEsr1cell activity is likely a key mechanism for the behavioral switch during motherhood.

For the Fe-based catalysts in Fischer-Tropsch synthesis, the reduction and activation process of α-Fe2O3 precursor has a significant effect on the catalytic performance. As a crystalline material, the reduction and activation of α-Fe2O3 is assuredly influenced by the exposed crystal plane; however, there is a lack of necessary research in this regard. In this work, α-Fe2O3 nanocrystals of three different morphologies, viz., pseudo-cubic, hexagonal-plate and rhombohedra, were synthesized, which mainly expose the crystal planes of (102), (001) and (104), respectively. The evolution of α-Fe2O3 crystal structure was then investigated in CO atmosphere by using the Operando Raman spectroscopy (ORS). The results show that the α-Fe2O3 (001) plane has a better reductive activity in comparison to the (104) and (102) planes. The SEM, TEM, XPS and XRD characterization and DFT calculation results reveal that CO2 desorption is a decisive step for the reduction of α-Fe2O3; owing to the weak binding ability of (001) crystal plane to oxygen atoms, the desorption of CO2 on the (001) crystal plane is much easier, which can promote the reduction process.

Several medications are available for the treatment of cancer, and monoclonal antibodies that target PD-1 and PD-L1 represent first-line options for cancer. PD-1 promotes the ability of the immune system to recognize and attack cancer cells by activating T cells. PD-1 also activates the autoimmune system. This activation causes healthy cells in the body to be attacked by the immune system, resulting in immune-related adverse events (irAE). The objective of this study was to comprehensively evaluate the adverse events of rejection reactions in real-world solid organ transplant patients using monoclonal antibodies that target PD-1/PD-L1.Data from 2016-2021 were extracted from the U.S. Food and Drug Administration(FDA) Adverse Reporting System (FAERS) to describe the rejection reaction in patients with solid organ transplantation cases after using PD-1/PD-L1 inhibitors approved by the FDA. The reporting odds ratio (ROR) with 95% confidence interval (CI) for rejection reaction was calculated for each PD-1/PD-L1 inhibitor. A disproportionality signal was defined when the lower limit of 95% CI1.The FAERS database recorded 11,935 adverse events related to solid organ transplantation. Among these reports, 117 showed that various PD-1/PD-L1 inhibitors exhibited a strong correlation with solid organ transplantation rejection. The 3 medicines with the incidence of rejection reaction include avelumab (1), nivolumab (79) and pembrolizumab (37). The average time of solid organ transplantation rejection associated with PD1 / PD-L1 inhibitors was 40.64 days. Of those patients who experienced solid organ transplant rejection, a total of 24.79% died.This study found that PD-1/PD-L1 inhibitor use in patients with solid organ transplantation was associated with donor organ rejection. This information serves as a pharmacovigilance signal that we need to continue to track in the real world.

The anatomy of the right posterior portal vein (RPPV) plays an important role in planning hepatic resection, living transplantation and interventional radiological procedures, yet the incidence of variations of RPPV without a common trunk in Chinese persons is still unclear. Therefore, we conducted this study and discussed its clinical implications.A retrospective analysis of multidetector computed tomography (MDCT) scans was performed in 1,933 patients with various abdominal pathologies between September 28, 2018 through May 23, 2019. After excluding 930 patients, a total of 1,003 patients were included in this study. Variations of the RPPV without a common trunk were classified according to classification standards.A total of 1,003 patients were included. RPPV without a common trunk was found in 216 (21.54%, 216/1,003) patients. Among them, we identified three variations of the origin from the right portal vein (RPV): first separate origin of P6, P7, or simultaneous separate origin of P6 and P7, and the incidences of these three variations were 1.50% (15/1,003), 6.58% (66/1,003) and 13.46% (135/1,003), respectively. Among 1,003 patients included in this study, 787 patients (78.46%, 787/1,003) showed that RPPV normally divided into P6 and P7 branches.Variations of the RPPV without a common trunk were not rare in Chinese population. Knowledge of this anatomic variation of the RPPV is extremely important for hepatic and transplant surgeons and interventional radiologists.

Phosphodiesterase is an enzyme that degrades the phosphodiester bond in the second messenger molecules cAMP and cGMP, and regulates the localization, duration, and amplitude of cyclic nucleotide signaling within subcellular domains. Sildenafil is a potent and selective inhibitor of the type 5 cGMP-specific phosphodiesterase used clinically to treat erectile dysfunction and pulmonary arterial hypertension. In this study, we examined the effect of sildenafil on human colorectal cancer HT-29 cells. Our data showed that sildenafil induced cell cycle arrest and apoptosis, and cotreatment with a ROS scavenger N-acetyl-L-cysteine partially reversed cell apoptosis caused by sildenafil in human colorectal cancer HT-29 cells. Overall, our study suggests that sildenafil appears to be a promising new treatment option for colorectal cancer.

Currently the standard surgical treatment for bladder defects is augmentation cystoplasty with autologous tissues, which has many side effects. Biomaterials such as small intestine submucosa (SIS) can provide an alternative scaffold for the repair as bladder patches. Previous studies have shown that SIS could enhance the capacity and compliance of the bladder, but its application is hindered by issues like limited smooth muscle regeneration and stone formation since the fast degradation and poor mechanical properties of the SIS. Procyanidins (PC), a natural bio-crosslinking agent, has shown anti-calcification, anti-inflammatory and anti-oxidation properties. More importantly, PC and SIS can crosslink through hydrogen bonds, which may endow the material with enhanced mechanical property and stabilized functionalities. In this study, various concentrations of PC-crosslinked SIS (PC-SIS) were prepared to repair the full-thickness bladder defects, with an aim to reduce complications and enhance bladder functions. In vitro assays showed that the crosslinking has conferred the biomaterial with superior mechanical property and anti-calcification property, ability to promote smooth muscle cell adhesion and upregulate functional genes expression. Using a rabbit model with bladder defects, we demonstrated that the PC-SIS scaffold can rapidly promote in situ tissue regrowth and regeneration, in particular smooth muscle remodeling and improvement of urinary functions.
