106. Bactericidal Effect and Application of DMDC on Pseudomonas Planktonic and Biofilm Bacteria.

Abstract

To explore the bactericidal effect of dimethyl dicarbonate (DMDC) on foodborne harmful microorganism, the bactericidal effect of DMDC on the planktonic and biofilm bacteria of Pseudomonas fluorescens and Pseudomonas lund of spoilage organism in fresh food were comparatively evaluated in the present study. Effects of DMDC on the bacterial activity, intracellular enzyme activity and mature biofilm of Pseudomonas were measured by using API ZYM kit, XTT kits and confocal microscope. Meanwhile, antibacterial activity of DMDC at different mass concentration in tomato and lettuce were performed. The results showed that DMDC had stronger bactericidal action against the two Pseudomonas compared with other three pathogens. The two Pseudomonas were inactivated by 250 mg/L DMDC treatment for 5 min, which was attributed to the decreasing activities of alanine araminase and naphthol -AS -BI -phosphohydrolas. In addition, mature biofilm of P. fluorescein and P. lund could be effectively destroyed. The population in biofilm cell decreased by 4.60 lg (CFU/cm²) and 4.15 lg (CFU/cm²), respectively, and the cell activity significantly dropped, resulting in the increase of dead cell and the thinner biofilm structure, when treated by 250 mg/L DMDC. Moreover, the aerobic plant count (APC), Pseudomonas number (PA), and mold and yeast count (MYC) of tomato and lettuce were significantly reduced by treatment of DMDC at 250 mg/L. The APC and PA in tomato was reduced by 2.60 lg (CFU/cm²) and 1.73 lg (CFU/ cm²), and in lettuce was decreased by 2.56 lg(CFU/cm²)and 1.98 lg(CFU/cm²), respectively, for 15 min treatment, accompanied by no detection of mold and yeast after 5 min treatment. Sensory qualities were not affected. Thus, DMDC significantly killed the planktonic and mature biofilm of two Pseudomonas, and has a good bactericidal effects when applied to the surface of two kinds of fruits and vegetables, which is expected to become a new cleaning agent in fruit and vegetable. Abstract

Neural stem/progenitor cells (NSPCs) persist in the mammalian subventricular zone throughout life, where they can be activated in response to physiological and pathophysiological stimuli. A recent study indicates metabotropic glutamate receptor 4 (mGluR4) is involved in regulating NSPCs behaviors. Therefore, defining mGluR4 function in NSPCs is necessary for determining novel strategies to enhance the intrinsic potential for brain regeneration after injuries. In this study, mGluR4 was functionally expressed in SVZ‐derived NSPCs from male Sprague‐Dawley rats, in which the cyclic adenosine monophosphate concentration was reduced after treatment with the mGluR4‐specific agonist VU0155041. Additionally, lateral ventricle injection of VU0155041 significantly decreased 5‐bromo‐2'‐deoxyuridine (BrdU)+ and Ki67+ cells, while increased Doublecortin (DCX)/BrdU double‐positive cells in SVZ. In cultured NSPCs, mGluR4 activation decreased the ratio of BrdU+ cells, G2/M‐phase cells, and inhibited Cyclin D1 expression, whereas it increased neuron‐specific class III β‐tubulin (Tuj1) expression and the number of Tuj1, DCX, and PSA‐NCAM‐positive cells. However, pharmacological blocking mGluR4 with the antagonist MSOP or knockdown of mGluR4 abolished the effects of VU0155041 on NSPCs proliferation and neuronal differentiation. Further investigation demonstrated that VU0155041 treatment down‐regulated AKT phosphorylation and up‐regulated expression of the phosphatase and tensin homolog protein (PTEN) in NSPCs culture. Moreover VU0155041‐induced proliferating inhibition and neuronal differentiating amplification in NSPCs were significantly hampered by VO‐OHpic, a PTEN inhibitor. We conclude that activation of mGluR4 in SVZ‐derived NSPCs suppresses proliferation and enhances their neuronal differentiation, and regulation of PTEN may be involved as a potential intracellular target of mGluR4 signal. 140. Effects of ultra-high pressure on gel properties of big head croaker (Collichthys lucidus) surimi

Abstract

The objective of this study was to evaluate the effect of ultra-high pressure (UHP) treatment on gel properties of Collichthys lucidus surimi. Three factors of UHP treatment which were pressure,time and temperature were investigated. The textural parameters such as hardness,springiness,cohesiveness,chewiness and gel strength of surimi gel were obtained by texture analyzer. With the gel strength as index,the optimum UHP treatment process was optimized with orthogonal test as follows: pressure of 300 MPa,time of 15 min,temperature of 20 ℃ and under the above conditions,the gel strength was 363.15 g·cm. It was found that surimi formed gelation at 200 MPa,gel properties had no significant difference when UHP time was more than 5 min,and as UHP temperature increased,gel properties decreased significantly. The effect order of UHP treatment conditions on gel strength was pressuretemperaturetime. When pressure was higher than 300 MPa and time was more than 5 min,gel cohesiveness had no significant difference under those conditions. Cohesiveness decreased significantly with the increasing temperature when the temperature was higher than 20 ℃.Compared with heat treatment,we found that the gel strength of surimi by UHP treatment was better than heat treatment,was 2.2 times of heat treatment. But the hardness under UHP treatment was lower,only 67% of heat treatment. As for the springiness,cohesiveness,chewiness,water holding capability and whiteness,the effects of UHP treatment were better,the springiness was 1.6 times of heat treatment,particularly. From the two cooperative processing methods of UHP and heat treatment,it showed that the effect of heat treatment after UHP was similar to the two-stage heat treatment while UHP treatment after heat treatment could damage gel structure obviously. Those results indicated that UHP treatment had an effect on the formation of surimi gel. UHP treatment could modify the quality of processed muscle-based products. This study suggested that UHP treatment exhibits a potential for surimi processing.

Abstract

Bladder cancer, the second most common genitourinary malignancy, severely endangers the human health. Rising evidence suggests that metabotropic glutamate receptors (mGluRs) are involve in tumor progression. In this study, we observed that metabotropic glutamate receptor 4 (mGluR4) was functionally expressed in normal and cancerous bladder cells and its expression was positively correlated with high bladder cancer grading. We further confirmed that the activation of mGluR4 by VU0155041, an mGluR4‐specific agonist, decreased cyclic adenosine monophosphate (cAMP) concentration and cell viability, promoted apoptosis and inhibited proliferation in bladder cancer cells, whereas MSOP (group III mGluR antagonist) or mGluR4 knockdown eliminated the effects of mGluR4 activity. Western blotting revealed the decreased cyclin D1 expression, increased procaspase‐8/9/3 cleavage, and unbalanced Bcl‐2/Bax expression in bladder cancer cell lines after mGluR4 activation, and likewise MSOP and mGluR4 knockdown abrogated the actions of mGluR4 activity. In vivo study showed that mGluR4 activation significantly inhibited tumor growth of bladder cancer via suppressing proliferation and promoting apoptosis. Furthermore, upregulation of phosphatase and tensin homolog (PTEN) and inhibition of Akt phosphorylation were also observed after mGluR4 activation. Similar with VU0155041, the Akt‐specific inhibitor markedly promoted apoptosis and inhibited proliferation. Nevertheless, the PTEN‐specific inhibitor significantly abolished the mGluR4 activation‐induced cell apoptosis and proliferative inhibition in bladder cancer cell lines. These results indicate that mGluR4 can regulate the switch between survival and death via the cAMP/PTEN/AKT signaling pathway in bladder cancer cells.
