To study the cadmium (Cd)-accumulation characteristics of the post-grafting generation of Cyphomandra betacea (Cav.) Sendt., the seeds of C. betacea grafted on Solanum nigrum L., tomato, and eggplant rootstocks were collected and planted in Cd-contaminated soil. Compared with seeds of ungrafted seedlings, the S. nigrum rootstock increased the shoot biomass of the post-grafting generation of C. betacea seedlings, while eggplant and tomato rootstocks decreased or had no significant effects. Only the S. nigrum rootstock increased the chlorophyll a content, net photosynthetic rate, and stomatal conductance of the post-grafting generation of C. betacea seedlings, while the other rootstocks decreased or had no significant effects on these photosynthetic parameters. The S. nigrum rootstock enhanced the superoxide dismutase and catalase activities, and soluble protein content, of the post-grafting generation of C. betacea seedlings, while the other rootstocks reduced or had no significant effects on these parameters. The three rootstocks decreased the Cd contents in different organs of the post-grafting generation of C. betacea seedlings, and the order of treatments was ungrafted > tomato rootstock > eggplant rootstock > S. nigrum rootstock. Therefore, S. nigrum was the best rootstock to promote the growth and decrease the Cd contents in post-grafting generation of C. betacea, and this can benefit C. betacea production in Cd-contaminated areas. A pot experiment was conducted to study the effects of different rootstocks on the cadmium (Cd) accumulation characteristics of the post-grafting generations of Cd-hyperaccumulator Galinsoga parviflora plants. Five treatments, ungrafted and G. parviflora seedlings grafted on the rootstocks of Kalimeris indica, Senecio scandens, Conyza canadensis, and Artemisia sieversiana, were utilized. The four rootstock grafts decreased the shoot biomass of the G. parviflora post-grafting generation compared with ungrafted. The K. indica and S. scandens grafts increased the Cd concentration in shoots of the G. parviflora post-grafting generation by 15.06% and 14.40%, respectively, compared with ungrafted, while the C. canadensis and A. sieversiana grafts had no significant effects. K. indica grafts increased the amount of Cd extracted by shoots of the G. parviflora post-grafting generation by 10.59% compared with ungrafted, while the other treatments resulted in decreases. Compared with ungrafted, the different rootstocks had no significant effects on the photosynthetic pigment content of the G. parviflora post-grafting generation, and only C. canadensis grafts increased the superoxide dismutase activity level, while only K. indica grafts increased the peroxidase activity level. The aim of this study was to determine the effects of exogenous indole‐3‐butyric acid (IBA) on the growth and cadmium (Cd) accumulation in the Cd‐accumulator plant Stellaria media. Exogenous IBA was applied as a foliar spray to S. media sddelings growing in Cd‐contaminated soil. Statistical analyses of data were conducted using SPSS 13.0 (SPSS, Chicago, IL). Treatment with IBA (50, 75, and 100 mg L−1) increased the root, shoot, and whole plant biomasses of S. media, and also increased the photosynthetic pigment content, antioxidant enzyme activity, soluble protein content, and soluble sugar content of S. media, compared with their respective levels in controls. The maximum values for these parameters were at the 75 mg L−1 IBA treatment. Treatment with 50 mg L−1 IBA increased the Cd content in the roots and shoots of S. media compared with the control. Treatment with 75 mg L−1 IBA increased the amount of Cd extracted by roots and shoots compared with controls. The maximum Cd extraction by roots and shoots was in the 75 mg L−1 IBA treatment. Pot and field experiments were conducted to study the effects of interspecies rootstocks on cadmium (Cd) accumulation characteristics of the potential Cd-hyperaccumulator Solanum photeinocarpum postgrafting generation plants. Four treatments (ungrafted and S. photeinocarpum seedlings grafted on the rootstocks of eggplant, potato, and tomato) were utilized in the present study. In the 2 pot experiments, eggplant and potato reduced biomass of S. photeinocarpum postgrafting generation plants, whereas tomato increased shoot biomass compared with the control (ungrafted seedlings). Compared with ungrafted seedlings, tomato increased and eggplant decreased Cd contents in shoots of S. photeinocarpum postgrafting generation. Only tomato increased Cd extraction by shoots of S. photeinocarpum postgrafting generation compared with ungrafted seedlings. In the field experiment, only tomato increased shoot Cd content of S. photeinocarpum postgrafting generation by 8.31% and shoot Cd extraction by 9.30% compared with ungrafted seedlings. Therefore, use of tomato as rootstock could enhance the ability of S. photeinocarpum postgrafting generation plants to remediate Cd-contaminated soil. The effects of application of the straw of cadmium accumulating species (Youngia erythrocarpa (cadmium hyperaccumulator), Bidens pilosa (cadmium hyperaccumulator), Solanum photeinocarpum (potential cadmium hyperaccumulator), and Siegesbeckia orientalis (cadmium accumulator)) in cadmium contaminated soil on the growth and cadmium accumulation of Galinsoga parviflora were studied. The root biomass, stem biomass, shoot biomass, total biomass, and the antioxidant enzyme activity of G. parviflora were increased by application of the straw of Y. erythrocarpa. The straw of Y. erythrocarpa, S. orientalis, and B. pilosa reduced soil pH, increased the soil exchangeable Cd concentration, and increased the Cd contents in stems, leaves, and shoots of G. parviflora compared with the control. The straw of Y. erythrocarpa and B. pilosa increased the Cd extraction amount in stems, leaves, shoots and the whole plant of G. parviflora compared with the control, with the effect of Y. erythrocarpa higher than that of B. pilosa. Therefore, the straw of Y. erythrocarpa and B. pilosa improved the Cd extraction ability of G. parviflora from cadmium-contaminated soil, and the straw of Y. erythrocarpa demonstrated to be the best material. Fusarium oxysporum is the most important pathogen of potatoes which causes post-harvest destructive losses and deteriorates the market value of potato tubers worldwide. Here, F. oxysporum was used as a host pathogen model system and it was revealed that autophagy plays a vital role as a regulator in the morphology, cellular growth, development, as well as the pathogenicity of F. oxysporum. Previous studies based upon identification of the gene responsible for encoding the autophagy pathway components from F. oxysporum have shown putative orthologs of 16 core autophagy related-ATG genes of yeast in the genome database which were autophagy-related and comprised of ubiquitin-like protein atg3. This study elucidates the molecular mechanism of the autophagy-related gene Foatg3 in F. oxysporum. A deletion (∆) mutants of F. oxysporum (Foatg3∆) was generated to evaluate nuclear dynamics. As compared to wild type and Foatg3 overexpression (OE) strains, Foatg3∆ strains failed to show positive MDC (monodansylcadaverine) staining which revealed that Foatg3 is compulsory for autophagy in F. oxysporum. A significant reduction in conidiation and hyphal growth was shown by the Foatg3∆ strains resulting in loss of virulence on potato tubers. The hyphae of Foatg3∆ mutants contained two or more nuclei within one hyphal compartment while wild type hyphae were composed of uninucleate hyphal compartments. Our findings reveal that the vital significance of Foatg3 as a key target in controlling the dry rot disease in root crops and potato tubers at the postharvest stage has immense potential of disease control and yield enhancement. High levels of selenium (Se) uptakes negatively affect plant growth. In this study, the possible molecular mechanism for the effects of abscisic acid (ABA) on Se absorption, metabolism and toxicity in Cyphomandra betacea Sendt. (Solanum betaceum Cav.) young plants were investigated. Se+ABA treatment promoted significant Se absorption in C. betacea while impeding plant growth as compared to Se treatment. The expression levels of sulfate/phosphate transporter protein genes indicated that Se+ABA triggered more S/Se absorption and transportation into chloroplast. Furthermore, Se+ABA promoted higher metabolisms of inorganic sulfur (S)/Se and organic S/Se. The organic Se might be in several forms (SeCysth, SeCys and SeMet) in Se+ABA treatment, whereas SeCysth was the major organic form in Se treatment. More reactive oxygen species production was suggested in Se+ABA treatment from a series of genes involved in antioxidant enzymes and molecules, including superoxide dismutase, peroxiredoxin, glutathione sulfur-transferase and glutathione. Se+ABA further improved the expression levels of genes involved in biosynthesis and signaling transduction genes involved in stress-related phytohormones (jasmonic acid and salicylic acid). Combining with the data in ABA treatment, we hypothesized a model that ABA might first affect the biosynthesis and signaling transduction pathways of stress-related phytohormones, and subsequently altered the metabolic processes responding to Se stress. Although the underlying mechanisms of the albino phenotype have been deciphered in seedlings of model plants, leaf albinism of bud mutants in woody species has been less extensively studied. In this study, a bud mutant of jujube (Ziziphus jujuba Mill.) with non-lethal albino phenotype was characterized. Compared with the normal leaves, the albino leaves showed lower chlorophyll and carotenoid contents, and higher POD activity and proline content to respond to oxidative damage. The impaired chloroplasts with ruptured thylakoid membranes and vacuoles with excess autophagosomes were observed in albino leaves. The decreased photosynthetic capacity in albino leaves was manifested as negative net photosynthetic rate and reduced chlorophyll fluorescence. The molecular mechanism of the albino leaves during development was explored through transcriptome analysis. A total of 3213, 3348, 8072 differentially expressed genes (DEGs) were identified in leaf buds, young leaves and mature leaves of normal plant and albino mutant, respectively. Further analysis indicated that most DEGs related to the biosynthesis of chlorophyll and carotenoid, chloroplast development, and photosynthesis were down-regulated in albino leaf buds, which were likely the main factors responsible for albino phenotype at early stage. Contrarily, DEGs involved in chloroplast development, including PPR , FtsH , mTERF , were mainly up-regulated in albino mature leaves, suggesting different regulatory mechanism for defective chloroplast structure at various stages. In addition, DEGs were significantly enriched in biosynthesis of phenylpropanoids and flavonoids in young and mature leaves of the albino mutant Moreover, transcription factor family genes, involving bHLH, MYB_related, NAC, ERF, B3, and WRKY, were identified as DEGs at three stages, indicating the promoted secondary metabolism production and senescence of the albino leaves. Fruits provide abundant carotenoid nutrients for humans, whereas the understanding of the transcriptional regulatory mechanisms of carotenoids in fruits is still limited. Here, we identified a transcription factor AcMADS32 in kiwifruit, which was highly expressed in the fruit, correlated with carotenoid content and localized in the nucleus. The silencing expression of AcMADS32 significantly reduced the content of β-carotene and zeaxanthin and expression of β-carotene hydroxylase gene AcBCH1/2 in kiwifruit, while transient overexpression increased the accumulation of zeaxanthin, suggesting that AcMADS32 was an activator involved in the transcriptional regulation of carotenoid in fruit. When AcMADS32 was further stably transformed into kiwifruit, the content of total carotenoid and components in the leaves of transgenic lines significantly increased, and the expression level of carotenogenic genes was up-regulated. Moreover, Y1H and dual luciferase reporter experiments confirmed that AcMADS32 directly bound the AcBCH1/2 promoter and activated its expression. Through Y2H assays, AcMADS32 can interact with other MADS transcription factor AcMADS30, AcMADS64 and AcMADS70. • Melatonin application distinctly promoted anthocyanin accumulation in berry skin. • Melatonin application significantly increased sucrose content in berry. • Melatonin treatment promoted the absorption of mineral elements in berry. In this study, we investigated the effects of melatonin application on berry coloration, sugar accumulation, and nutrient absorption in 'Summer Black' grapes. Melatonin spraying at 100 μmol L−1 on grapes during veraison induced skin coloration earlier than that in controls, as well as higher transcript abundance of anthocyanin biosynthesis-related genes and transcription factors MYBA1 and MYBA2. Melatonin treatment increased the soluble sugar content, especially of sucrose, by promoting the activity of sucrose phosphate synthase, and also increased endogenous melatonin content and the concentrations of mineral nutrients N, K, Cu, Fe, and Zn in grape berries. Correlation analysis suggested that high sugar content promoted anthocyanin synthesis. Light is a key environmental factors affecting anthocyanin accumulation in plants. Ubiquitin E3 ligase COP1 has been proved to be a negative regulator involved in light-regulated plant development process, whereas the function and expression specificity of COP1 in anthocyanin biosynthesis in sweet cherry remains unclear. In the present study, we identified a COP1 in sweet cherry, named PacCOP1 , it exhibited apparent different expression patterns in red-colored 'Hongdeng' and bi-colored 'Satonishiki', with increasing trend largely in 'Satonishiki', but decreasing trend in 'Hongdeng' after veraison, which was contrary to their variation tendency of anthocyanin content. While the expression abundance of anthocyanin biosynthesis related genes were largely increased after veraison, in accordance with anthocyanin content. Correlation analysis proved that the expression of PacCOP1 was negative correlated with the major genes on anthocyanin accumulation in 'Hongdeng' and 'Satonishiki' fruit, in especial Pac DFR, Pac ANS, Pac MYBA and Pac bHLH33. Furthermore, over-expression of PacCOP1 in Arabidopsis displayed increased COP1 transcript level with negligible pigmentation and corresponding lower expression level of AtPAP1 , AtDFR , AtLDOX , and AtUFGT. Anthocyanins are important components in skins of red table grapes and contribute to the berries appearance, a key quality characteristic for customers. In recent years, exogenous foliage fertilizers has been applied to grapevines to improve the pigmentation of the fruit. The present study examines the effect on a biostimulant (SUNRED) pre-véraison application in the accumulation of anthocyanins in 'Red Globe' grapes, and investigates the related changes in expression of key genes and their enzyme activities in the flavonoid pathways. Additionally, abscisic acid (S-ABA) was also applied to grapevines to evaluate the comparative effect of SUNRED. Our analyses showed that total anthocyanin contents increased in both SUNRED and S-ABA treated grapes; for S-ABA, a 1% dilution (A100) of the commercially available stock solution treatments represented the greatest effect on pigmentation; for SUNRED, a 0.1% dilution (S1000) was most effective. The anthocyanin contents increased by 1.16-fold and 1.4-fold after A100 and S1000 treatments, respectively. The gene expression analyses showed that almost all genes involved in the anthocyanin biosynthesis pathway up-regulated after A100 and S1000 treatments, suggesting that the increment in total anthocyanin content was attributed to the increased expression level of related genes. Moreover, the activities of phenylalanine ammonia-lyase (PAL), chalcone isomerase (CHI), UDP glucose: flavonoid 3-o-glucosyl transferase (UFGT) and dihydroflavonol 4-reductase (DFR), key enzymes for biosynthesis of anthocyanin, were increased by the exogenous treatments. Plums ( Prunus salicina and Prunus domestica ) are prevalent in southwestern China, and have attracted interest owing to their delectable taste and exceptional nutritional properties. Therefore, this study aimed to investigate the nutritional and flavor properties of plum to improve its nutritional utilization. Specifically, we determined the soluble sugars, organic acids, and phenolic components in 86 accessions using high-performance liquid chromatography. Notably, glucose, fructose, malic, and quinic acids were the predominant sweetness and acidity in plums, with sucrose contributing more to the sweetness of the flesh than the peel. Moreover, The peel contains 5.5 fold more phenolics than flesh, epicatechin, gallic acid, and proanthocyanidins C1 and B2 were the primary sources of astringency. Correlation and principal component analyses showed eight core factors for plum flavor rating, and a specific rating criterion was established. Introduction: Exogenous melatonin (MT) can promote horticultural crops growth under stress conditions., Methods: In this study, the effects of exogenous MT on the accumulation of selenium (Se) in grape were studied under Se stress., Results and Discussion: Under Se stress, exogenous MT increased the biomass, content of photosynthetic pigments and antioxidant enzyme activity of grapevines. Compared with Se treatment, MT increased the root biomass, shoot biomass, chlorophyll a content, chlorophyll b content, carotenoids, superoxide dismutase activity, and peroxidase activity by 18.11%, 7.71%, 25.70%, 25.00%, 25.93%, 5.73%, and 9.41%, respectively. Additionally, MT increased the contents of gibberellin, auxin, and MT in grapevines under Se stress, while it decreased the content of abscisic acid. MT increased the contents of total Se, organic Se and inorganic Se in grapevines. Compared with Se treatment, MT increased the contents of total Se in the roots and shoots by 48.82% and 135.66%, respectively. A transcriptome sequencing analysis revealed that MT primarily regulated the cellular, metabolic, and bioregulatory processes of grapevine under Se stress, and the differentially expressed genes (DEGs) were primarily enriched in pathways, such as aminoacyl-tRNA biosynthesis, spliceosome, and flavonoid biosynthesis. These involved nine DEGs and nine metabolic pathways in total. Moreover, a field experiment showed that MT increased the content of Se in grapes and improved their quality. Increased selenium (Se) content in fruits can supply Se in human body, but the effects of teas on the Se uptake in fruit trees are unknown. The effects of infusions of four teas (green, black, dark, and white) on the Se uptake of grapevine were studied to promote the Se uptake in fruit trees in this study. However, only black tea infusion increased the biomass, photosynthetic pigment content, superoxide dismutase (SOD) activity, peroxidase (POD) activity, and soluble protein content of grapevine. Except for white tea infusion, other tea infusions also increased the catalase (CAT) activity of grapevine. Furthermore, the tea infusions increased the activities of adenosine triphosphate sulfurase (ATPS) and adenosine 5'-phosphosulfate reductase (APR), and decreased the activities of serine acetyltransferase (SAT) and selenocysteine methyltransferase (SMT). Only the dark and white tea infusions increased the shoot total Se content by 86.53% and 23.32%, respectively (compared with the control), and also increased the shoot inorganic Se content and shoot organic Se content. Notably, four tea infusions decreased the organic Se proportion and increased the inorganic Se proportion in grapevine. Correlation and grey relational analyses showed that the root total Se content, ATPS activity, and ARP activity were closely associated with the shoot total Se content. The principal component and cluster analyses also showed that the ATPS activity, APR activity, root total Se content, and shoot total Se content were classified into one category. Volatile compounds are important determinants affecting fruit flavor. Previous study has identified a bud mutant of 'Ehime 38' ( Citrus reticulata ) with different volatile profile. However, the volatile changes between WT and MT during fruit development and underlying mechanism remain elusive. In this study, a total of 35 volatile compounds were identified in the pulps of WT and MT at five developmental stages. Both varieties accumulated similar and the highest levels of volatiles at stage S1, and showed a downward trend as the fruit develops. However, the total volatile contents in the pulps of MT were 1.4-2.5 folds higher than those in WT at stages S2-S5, which was mainly due to the increase in the content of d-limonene. Transcriptomic and RT-qPCR analysis revealed that most genes in MEP pathway were positively correlated with the volatile contents, of which DXS1 might mainly contribute to the elevated volatiles accumulation in MT by increasing the flux into the MEP pathway. Moreover, temporal expression analysis indicated that these MEP pathway genes functioned at different developmental stages. To alleviate the selenium (Se) stress in fruit trees and improve its accumulation, the effects of exogenous indole-3-acetic acid (IAA) on the growth and Se accumulation of grapevine under Se stress were studied. The application of exogenous IAA increased the biomass of grapevine, and the concentration of exogenous IAA had a regression relationship with the biomass. The root and shoot biomass were the maximum at 60 mg L - 1 IAA, increasing by 15.61% and 23.95%, respectively, compared with the control. Exogenous IAA also increased the photosynthetic pigments and the activities of superoxide dismutase and peroxidase in grapevine. Moreover, exogenous IAA increased the contents of total Se, organic Se, and inorganic Se, and the concentration of exogenous IAA had a regression relationship with the total Se content. The highest contents of root total Se and shoot total Se were accumulated at 90 mg L - 1 IAA, increasing by 29.94% and 55.77% respectively,. In addition, the correlation and path analyses revealed that the carotenoid content and root total Se content were closely associated with the shoot total Se content. Therefore, the application of exogenous IAA can alleviate the stress of Se to grape and promote its uptake and the most effective amount for the uptake of Se is 90 mg L - 1 IAA., (© 2024. To identify suitable potassium fertilizers for grape ( Vitis vinifera L.) production and study their mechanism of action, the effects of four potassium-containing fertilizers (complex fertilizer, potassium nitrate, potassium sulfate, and potassium dihydrogen phosphate) on sugar and organic acid metabolism in grape fruits were investigated. Potassium-containing fertilizers increased the activity of sugar and organic acid metabolism-related enzymes at all stages of grape fruit development. During the later stages of fruit development, potassium-containing fertilizers increased the total soluble solid content and the sugar content of the different sugar fractions and decreased the titratable acid content and organic acid content of the different organic acid fractions. At the ripening stage of grape fruit, compared with the control, complex fertilizer, potassium nitrate, potassium sulfate, and potassium dihydrogen phosphate increased the total soluble solid content by 1.5, 1.2, 3.5, and 3.4 percentage points, decreased the titratable acid content by 0.09, 0.06, 0.18, and 0.17 percentage points, respectively, and also increased the total potassium content in grape fruits to a certain degree. Transcriptome analysis of the differentially expressed genes (DEGs) in the berries showed that applying potassium-containing fertilizers enriched the genes in pathways involved in fruit quality, namely, carbon metabolism, carbon fixation in photosynthetic organisms, glycolysis and gluconeogenesis, and fructose and mannose metabolism. Potassium-containing fertilizers affected the expression levels of genes regulating sugar metabolism and potassium ion uptake and transport. Overall, potassium-containing fertilizers can promote sugar accumulation and reduce acid accumulation in grape fruits, and potassium sulfate and potassium dihydrogen phosphate had the best effects among the fertilizers tested.

Kiwifruit, a nutrient-dense fruit, has become increasingly popular with consumers in recent decades. However, kiwifruit trees are prone to stunted growth after a few years of planting, called early tree decline. In this study, melatonin (MT), pollen polysaccharide (SF), 14-hydroxyed brassinosteroid (14-HBR) were applied alone or in combination to investigate their influence on plant growth, nutrition absorption and rhizosphere bacterial abundance in kiwifruit seedlings. The results revealed that MT, SF and 14-HBR alone treatments significantly increased leaf chlorophyll content, photosynthetic capacity and activities of dismutase and catalase compared with the control. Among them, MT treatment significantly increased the dry root biomass by 35.7%, while MT+14-HBR treatment significant enhanced the dry shoot biomass by 36.9%. Furthermore, both MT and MT+14-HBR treatments markedly improved the activities of invertase, urease, protease and phosphatase in soil, as well as the abundance of Proteobacteria and Acidobacteria in rhizosphere microorganisms based on 16S rDNA sequencing. In addition, MT treatment improved the content of available K and organic matter in soil, and increased the uptake of P, K and Fe by seedlings. In summary, 14-HBR and MT combined had the best effect on promoting rhizosphere bacterial distribution, nutrient absorption and plant growth. The novel loquat cultivar 'Chunhua No.1' (CH1) is a promising commercial cultivar. However, CH1 has texture characteristics different from those of common loquat, and its formation mechanism remains unclear. Here, we first identified the phenolic compounds of CH1 and its parent ('Dawuxing', DWX) and the effect on texture formation. The special presence of stone cells explained the flavor differences in CH1. Chlorogenic acid, neochlorogenic acid, and coniferyl alcohol were the main phenolic compounds in loquat, and the high content of coniferyl alcohol was a potential factor for the rough texture of CH1. Transcriptome reveals that phenylpropanoid metabolism was activated during CH1 fruit texture formation. Kyoto Encyclopedia of Genes and Genomes (KEGG) identified 51 structural genes involved in phenylpropanoid biosynthesis, and Weighted Gene Co-expression Network Analysis (WGCNA) identified four structural genes and 88 transcription factors. Pathogenic oomycetes infect a wide variety of organisms, including plants, animals, and humans, and cause massive economic losses in global agriculture, aquaculture, and human health. Salicylic acid (SA), an endogenous phytohormone, is regarded as an inducer of plant immunity. Here, the potato late blight pathogen Phytophthora infestans was used as a model system to uncover the inhibitory mechanisms of SA on pathogenic oomycetes. In this research, SA significantly inhibited the mycelial growth, sporulation, sporangium germination, and virulence of P. infestans . Inhibition was closely related to enhanced autophagy, suppression of translation initiation, and ribosomal biogenesis in P. infestans , as shown by multiomics analysis (transcriptomics, proteomics, and phosphorylated proteomics). Monodansylcadaverine (MDC) staining and Western blotting analysis showed that SA promoted autophagy in P. infestans by probably targeting the TOR signaling pathway. These observations suggest that SA has the potential to control late blight caused by P. infestans .

Most red-fleshed kiwifruit cultivars, such as Hongyang, only accumulate anthocyanins in the inner pericarp; the trait of full red flesh becomes the goal pursued by breeders. In this study, we identified a mutant "H-16" showing a red color in both the inner and outer pericarps, and the underlying mechanism was explored. Through transcriptome analysis, a key differentially expressed gene AcGST1 was screened out, which was positively correlated with anthocyanin accumulation in the outer pericarp. The result of McrBC-PCR and bisulfite sequencing revealed that the SG3 region (-292 to -597 bp) of AcGST1 promoter in "H-16" had a significantly lower CHH cytosine methylation level than that in Hongyang, accompanied by low expression of methyltransferase genes ( MET1 and CMT2 ) and high expression of demethylase genes ( ROS1 and DML1 ). Transient calli transformation confirmed that demethylase gene DML1 can activate transcription of AcGST1 to enhance its expression. Overexpression of AcGST1 enhanced the anthocyanin accumulation in the fruit flesh and leaves of the transgenic lines. These results suggested that a decrease in the methylation level of the AcGST1 promoter may contribute to accumulation of anthocyanin in the outer pericarp of "H-16".
