The understanding of the association between salt intake and precancerous lesions may contribute to clarify the causal relation with gastric cancer. We systematically reviewed 17 articles addressing the association between dietary salt exposure and gastric intestinal metaplasia and conducted meta-analyses for quantitative synthesis (random effects model). Salt exposure was estimated assessing salted/salty food consumption, preference for salted/salty foods, use of table salt, or sodium urinary excretion. Heterogeneity was also large regarding food items evaluated, consumption categories, and data analysis. The combined odds ratio (OR) was 1.68 (95% confidence interval (CI) = 0.98-2.90; I(2) = 55.4%) for the association between salted/salty meat and intestinal metaplasia (4 studies) and the OR was 1.53 (95% CI = 0.72-3.24; I(2) = 76.8%) for salt preference. There was a positive, nonstatistically significant association between intestinal metaplasia and urinary sodium excretion. The heterogeneity of methodological options and results preclude quantitative synthesis or its proper interpretation, even if the available evidence may suggest a positive association between salt and intestinal metaplasia.

E-mail guillard@univ-montp2.fr; International audience; The effect of three cooling rates (rapid, intermediate and slow CR) on the moisture barrier properties and on the physical state of acetylated and high melting point hydrophobic self-supported moisture barriers has been investigated. The selected CR were representative of industrial processing conditions and the selected barrier materials of common effective GRAS substances (acetomonopalmitin, white beeswax, two commercial blends of beeswax and acetylated glycerides and a blend of palmitic/stearic acids). Variations of CR affected crystallisation kinetics, SFC in an extend depending on the fat chemical composition and degree of undercooling, crystal size and ratio of polymorphs present in the materials. It did not have major influence on the contact angles with water measured at the surface of the materials and on the mass-volume area properties of the material. The resultant effect on the macroscopic moisture barrier properties of the materials were evaluated using water vapour permeability (WVP) measurements. The CR had no significant effect on the WVP, except for one blend of acetylated fat and beeswax for which a slow CR may have favoured the healing of imperfections. The variations of WVP between all materials and CRs were mainly attributed to variation in materials polarity using multivariable analysis

Cardiomyopathy is a manifestation of mitochondrial cytopathies, but rarely constitutes the dominant feature, especially in adults. We report the case of a 59-year-old male with a personal and maternal history of diabetes and deafness, who presented with cardiomyopathy and kidney disease. We diagnosed the patient as having a mitochondrial cytopathy resulting from the 3243A>G mutation on the tRNALeu(UUR) gene in the mitochondrial DNA. The family history, broad spectrum of clinical manifestations and fluctuant clinical course provided clues to the diagnosis. We discuss the possible mechanisms underlying the phenotypic variability and fluctuant clinical course of mitochondrial disorders and the potential usefulness of coenzyme Q10 and L-carnitine in 3243A>G mutation patients.

International audience; Four acetostearin products with increasing acetylation degree were synthesized by chemical inter-esterification followed by fractionation/blending stages. Their physical properties and functional barrier properties were studied and compared to the properties of technical tristearin. Increasing acetylation degree (AD) modified the triacylglycerols crystal habits and probably led to an increase in acyl chain fluidity, which induced, at macroscopic levels, a decrease in solid fat content (SFC), in melting point, in surface and bull: material hydrophobicity, and an increased moisture effective diffusivity. Water vapor permeability (WVP) coefficients of the materials were partially influenced by the AD factor, but also by the development of macroscopic cracks in lipids presenting high SF C. Acetylated stearin up to 47% (acetyl mol/mol of esterified chain) presented the lowest WVP at 20 degrees C resulting from an adequate balance between hydrophobicity and mechanical properties of the material.

Outer membrane proteins (Omp) in bacteria, porins, have an important role on antibiotic translocation trough bacterial membranes. OmpF is one of the most important E. coli porins in the permeation of beta-lactam and quinolone antibiotics. Moreover, these antibiotics might penetrate the bacterial outer membrane through porins or through the lipid/protein interface. We focused on the characterization of E. coli membranes, once this knowledge is crucial for studies of antibiotic translocation and useful to understand and counteract bacterial antibiotic resistance, one of the biggest concerns of nowadays. Steady-state fluorescence anisotropy is a good technique to detect changes in lipid environments through the use of fluorescent probes. We used two fluorescent probes, DPH and TMA-DPH (which provide information from the core and from the interface regions of the bilayer, respectively), and determined the temperature dependence of these two anisotropy probes, incubated or incorporated into POPE/POPG (0.75:0.25), POPE/POPG/Cardiolipin (0.67:0.23:0.10) and E. coli total lipid extract liposomes (LUVs) and OmpF proteoliposomes. POPE/POPG and POPE/POPG/Cardiolipin mixtures are conventionally used to mimic the E. coli membranes, while the E. coli total lipid extract is a natural system. In this study we determine the phase transition temperatures characteristics of each membrane mimetic system and still prove the correct insertion of the protein in the membrane. When comparing the transition temperatures obtained for each mimetic system, our study suggest that the ternary lipid mixture is a better system to mimic the E. coli membranes than the binary system.

O objetivo geral desta Dissertação foi produzir e caracterizar filmes ativos à base de gelatina e/ou quitosana incorporados com emulsão dupla água-em-óleo-em-água (A/O/A) contendo extrato hidroetanólico da folha de pitangueira (Eugenia uniflora L.) na fase aquosa interna, como veículo de princípios ativos, além de caracterizar as emulsões água-em-óleo (A/O) e água-em-óleo-em-água (A/O/A) produzidas. A emulsão A/O/A foi produzida em duas etapas, sendo que a primeira etapa consistiu na produção das emulsões A/O e a segunda etapa consistiu na produção das emulsões A/O/A, variando as proporções de fases e concentrações dos emulsificantes (poliricinoleato de poliglicerol (PGPR) e Tween 80) em ambas as emulsões. As emulsões A/O e A/O/A foram caracterizadas em relação ao diâmetro de gotas e estabilidade física. As emulsões A/O e A/O/A com melhores propriedades foram ainda caracterizadas em relação ao comportamento reológico, sendo que a emulsão A/O/A ótima foi também caracterizada em relação às atividades antioxidante e antimicrobiana, juntamente com o EHFP. Os filmes ativos foram produzidos usando gelatina, quitosana e blenda gelatina/quitosana como biopolímeros, e a emulsão A/O/A, e foram caracterizados em relação às suas principais propriedades físicas, e em relação às atividades antioxidantes e antimicrobianas. As emulsões A/O se apresentaram com diâmetro médio de gotas nanométrico, baixa polidispersão, distribuições mono e bimodal, e elevada estabilidade física, com a emulsão A/O 20/80 com 3 g PGPR/100 g óleo a escolhida como emulsão A/O ótima, apresentando comportamento reológico de fluído newtoniano. As emulsões A/O/A apresentaram distribuição de diâmetro médio de gotas trimodal, com populações de gotas de diâmetro nanométrico, submicrométrico e micrométrico, mas com diâmetro médio de gotas submicrométrico e elevada polidispersão. A emulsão A/O/A 40/60 com 3 g/Tween 80/100 g água foi escolhida como ótima, apresentando comportamento reológico de fluído pseudoplástico. Além disso, a emulsão A/O/A foi capaz de manter a atividade antioxidante e antimicrobiana do EHFP. A solubilidade, a permeabilidade ao vapor de água, o ângulo de contato, o brilho e a barreira à transmissão da luz UV/Visível diminuíram em todos os filmes ativos pela incorporação da emulsão A/O/A, ao passo que a opacidade aumentou e a intensidade dos picos de difração dos filmes à base de quitosana e blenda gelatina/quitosana foi afetada. A rigidez e resistência dos filmes à base de gelatina ou quitosana aumentaram com a adição da emulsão A/O/A, ao passo que diminuíram para o filme à base de blenda gelatina/quitosana. Além disso, a presença da emulsão A/O/A só diminuiu a temperatura de transição vítrea do filme à base de quitosana, não afetando os demais. Os filmes ativos apresentaram aumento na atividade antioxidante devido à incorporação da emulsão A/O/A, mas não ocorreu a formação de halos de inibição contra as bactérias testadas, apenas inibição do crescimento na região de contato com os filmes à base de quitosana e blenda gelatina/quitosana. A emulsão A/O/A mostrou-se um sistema eficaz no encapsulamento do EHFP, sendo possível realizar a incorporação dela em filmes ativos à base de gelatina, quitosana e blenda gelatina/quitosana, cujas propriedades foram afetadas, no geral, de maneira positiva. The general objective of this Dissertation was to produce and characterize active films based on gelatin and/or chitosan incorporated with water-in-oil-in-water (W/O/W) double emulsion containing pitangueira (Eugenia uniflora L.) leaf hydroethanolic extract leaf in the internal aqueous phase as a vehicle of active ingredients, in addition to characterizing the water-in-oil (W/O) and water-in-oil-in-water (W/O/W) emulsions. The W/O/W emulsion was produced in two steps, the first step being the production of W/O emulsions and the second step the production of W/O/W emulsions, varying of the ratio phases and concentrations of the emulsifiers (Polyglycerol polyricinoleate and Tween 80) in both emulsions. W/O and W/O/W emulsions were characterized in relation to the average droplet diameter and physical stability. The W/O and W/O/W emulsions with the best properties were characterized in relation to the rheological behavior, and the optimal W/O/W emulsion was also characterized in relation to antioxidant and antimicrobial activities, together with PLHE. The active films were produced using gelatin, chitosan or gelatin/chitosan blend as biopolymers, and the W/O/W emulsions, and were characterized in relation to their main physical properties, and in relation to antioxidant and antimicrobial activities. The W/O emulsions had a nanometer mean droplet diameter, low polydispersity, mono and bimodal distributions, and high physical stability, with the W/O emulsion 20/80 with 3 g PGPR/100 g oil as the best W/O emulsion, presenting rheological behavior of Newtonian fluid. The W/O/W emulsions had a trimodal mean droplet diameter distribution, with nanometer, submicrometer and micrometer droplet populations, but with submicrometer average droplet diameter and high polydispersity. W/O/W emulsion 40/60 with 3 g/Tween 80/100 g water was chosen as optimal, showing rheological behavior of pseudoplastic fluid. In addition, the emulsion W/O/W was able to maintain the antioxidant and antimicrobial activity of PLHE. Solubility, water vapor permeability, contact angle, gloss and barrier to UV/Visible light transmission decreased in all active films by incorporation of the W/O/W emulsion, while opacity increased and the intensity of the diffraction peaks of the chitosan and gelatin/chitosan blend films was affected. The stiffness and strength of gelatin or chitosan based films increased with the addition of the W/O/W emulsion, while decreasing for gelatin/chitosan blend film. In addition, the presence of emulsion W/O/W only decreased the glass transition temperature of the chitosan-based film, not affecting the others. Although little, the active films showed antioxidant activity due to the incorporation of the emulsion W/O/W, but there was no formation of inhibition halos against the tested bacteria, only growth inhibition in the contact region with the chitosan and gelatin/chitosan blend based films. The emulsion W/O/W proved to be an effective system for encapsulating the PLHE, being possible to incorporate it in active films based on gelatin, chitosan and gelatin/chitosan blend, whose properties were generally positively affected.

Introdução: O diabetes mellitus (DM) está associado a complicações que comprometem a qualidade de vida e a sobrevida dos indivíduos. Além disso, acarreta elevados custos para o controle metabólico e o tratamento de suas complicações, sendo assim caracterizado como um problema de saúde pública. A regulação da digestão e da absorção intestinal dos carboidratos, com vista a manter a homeostase da glicose plasmática, constituem importantes estratégias de proteção em condições clínicas como o diabetes tipo 2 (DM2), obesidade e síndrome metabólica. Os compostos fenólicos compreendem um grupo complexo de fitoquímicos bioativos presentes nos vegetais. Estudos in vitro e in vivo têm demonstrado que os compostos fenólicos inibem a atividade de carbohidrases (α-amilase e α-glicosidase) e o transporte intestinal de glicose mediado pelos transportadores SGLT1 e GLUT2. O cerrado brasileiro compreende uma larga biodiversidade, porém, apesar de muitas espécies terem sido identificadas, o seu potencial nutritivo e funcional ainda é pouco conhecido. Dentre estas espécies nativas é destacado o jatobá-do-cerrado. O jatobá-do-cerrado é uma leguminosa nativa brasileira, cuja a polpa farinácea que envolve suas sementes apresenta quantidades significativas de compostos fenólicos, podendo ter um potencial efeito sobre o metabolismo da glicose. Objetivos: Verificar os efeitos dos compostos fenólicos da farinha de jatobá-do-cerrado na digestão de carboidratos e na captação de glicose em células intestinais Caco-2. Metodologia: Os compostos fenólicos da farinha de jatobá foram obtidos por extração sequencial com as soluções de etanol (60%) e acetona (70%). Em seguida, o extrato foi digerido utilizando enzimas (α-amilase, pepsina e pancreatina) em pH fisiológico. Os compostos fenólicos presentes no extrato antes e após a digestão foram identificados por cromatografia líquida de ultra performance - espectrômetro de massas (UPLC-MS/MS). Foi avaliada a capacidade de inibição dos extratos de jatobá digeridos em relação à atividade das enzimas α-amilase e α-glicosidase. Células intestinais Caco-2 foram incubadas com diferentes concentrações (0,05 mg/mL - 0,1 mg/mL) de extratos de farinha de jatobá digeridos em diferentes tempos (30 min, 2h e 12 h) para a avaliação da captação de glicose e da expressão gênica dos transportadores de glicose SGLT1 e GLUT2. Resultados: 44 compostos fenólicos foram identificados, dentre eles, a principal classe presente são os flavonoides. Compostos como o ácido cafeico, o kaempferol, quercetina-3- rutinosideo e a quercetrina estavam presentes no extrato antes da digestão. O conteúdo de compostos fenólicos do extrato foi reduzido após a digestão, entretanto o mesmo ainda apresentou compostos de relevância biológica como o ácido p-cumárico, ácido 3-o-feruloilquinico, theaflavina, crisina e grandinina que já apresentaram efeito positivo sobre o metabolismo da glicose in vitro em outros trabalhos. Os extratos fenólicos de jatobá após a digestão in vitro inibiram significativamente a atividade das enzimas α-amilase (76 e 91%) e α- glicosidase (53 e 77%). Os extratos também demonstraram inibir significativamente tanto a captação de glicose independente de sódio quanto a expressão gênica dos transportadores de glicose SGLT1 e GLUT2 de maneira dose-dependente. Conclusão: Este é o primeiro trabalho que identificou os compostos fenólicos presentes na farinha de jatobá. A partir do exposto, podemos concluir que a farinha de jatobá apresenta potencial benefício a saúde devido ao seu conteúdo de compostos fenólicos e a capacidade destes compostos de regular a digestão e a absorção de carboidratos in vitro. Introduction: Diabetes mellitus (DM) is associated with complications that decrease the quality of life and survival of individuals. In addition, it entails high costs for metabolic control and treatment of its complications, thus being characterized as a public health problem. The regulation of digestion and intestinal absorption of carbohydrates to maintain plasma glucose homeostasis are important strategies for protection in chronic diseases such as type 2 diabetes (DM2), obesity and metabolic syndrome. Phenolic compounds are a complex group of chemical substances present in plants. In vitro and in vivo studies have shown that phenolic compounds are able to inhibit the activity of carbohydrases (α-amylase and α-glycosidase) and the intestinal transport of glucose mediated by SGLT1 and GLUT2 transporters. Brazilian Cerrado present a large biodiversity, but although many species have been identified, its nutritional and functional potential is still little known. Among these native species is the jatobá-docerrado. Jatobá-do-cerrado is a brazilian native legume, whose farinaceous pulp that surrounds its seeds presents significant amounts of phenolic compounds and may have a potential effect on glucose metabolism. Objectives: To verify the effects of phenolic compounds from jatobá-do-cerrado flour in the digestion of carbohydrates and uptake of glucose in Caco-2 intestinal cells. Methods: Phenolic compounds of jatobá flour were obtained by sequential extraction with solutions of ethanol (60%) and acetone (70%). The extract was digested using enzymes (α-amylase, pepsin and pancreatin) at physiological pH. The phenolic compounds present in the extract before and after the digestion were identified by liquid chromatography of ultra-performance - mass spectrometer (UPLCMS / MS). The ability of inhibition of the extracts of jatobá digested in relation to the activity of α-amylase and α-glycosidase enzymes was evaluated. Caco-2 intestinal cells were incubated with different concentrations of jatobá flour extracts (0.1 mg / mL - 0.05 mg / mL) for different time (30 min, 2 h and 12 h) to the evaluation of facilitated uptake (sodium-free buffer) and gene expression of SGLT1 and GLUT2 glucose transporters. Results: 44 phenolic compounds have been identified, among them a major class present are flavonoids. Compounds such as caffeic acid, quercetin-3-rutinoside and quercetrine were present in the extract before in vitro digestion. The content of phenolic compounds of the extract after digestion was reduced. However, the extract presents compounds with biological activity such as p-coumaric acid, 3-o-feruloylquinic acid , theaflavin, chrysin and grandinine, which already presented positive effects on glucose metabolism in vitro in other studies. Phenolic extracts of jatobá after in vitro digestion inhibited the activity of α-amylase (76 and 91%) and α-glycosidase (53 and 77%). The extracts also shown to inhibit both glucose uptake and gene expression of glucose transporters SGLT1 and GLUT2 in a dose-dependent manner. Conclusion: This is the first work that identified the phenolic compounds present in jatobá flour. Thus, we can conclude that the jatobá flour presents potential health benefit by modulate digestion and the absorption of carbohydrates in vitro.

O objetivo do presente estudo foi avaliar a atividade antioxidante de extrato de folhas de oliveira (EFO) (Olea europaea L.) por diferentes metodologias analíticas in vitro e in situ, para verificação de efeito em sistemas biológicos. O extrato foi obtido a partir de folhas secas de oliveira, previamente micronizadas, em metanol/água (80/20%) na proporção 1:20 (m/v), após remoção de compostos solúveis em n-hexano. Após liofilização, no EFO foi avaliado o poder redutor por Folin-Ciocalteau, conteúdo de flavonoides totais, teor de oleuropeina, poder de redução do íon férrico (FRAP) e atividade antioxidante sobre DPPHo, ABTSo+, ânion superóxido (O2o-), ácido hipocloroso (HOCl) e óxido nítrico (NOo). O extrato foi também avaliado quanto ao efeito protetor sobre danos oxidativos em eritrócitos humanos. O ácido ascórbico foi utilizado como referência. O experimento foi repetido seis vezes (n = 6) e os ensaios realizados em duplicata. O poder redutor do extrato e o conteúdo de flavonoides totais e oleuropeína foram 131,7 ± 9,4 mg equivalente de ácido gálico/g extrato seco (ms), 19,4 ± 1,3 mg equivalente de quercetina/g ms e 25,5 ± 5,2 mg oleuropeína/g ms, respectivamente. O ensaio de FRAP apresentou 281,8 ± 22,8 mg equivalente de trolox/g ms. O EFO foi efetivo na inibição dos radicais DPPHo e ABTSo+, dependente da concentração de extrato, com valores de IC50 de 13,8 ± 0,8 e 16,1 ± 1,2 µg/mL, respectivamente. Com relação à atividade antioxidante sobre espécies reativas de importância biológica, o EFO apresentou forte capacidade de inibição de O2o- (IC50 = 52,6 ± 2,1 µg/mL) e NOo (IC50 = 48,4 ± 6,8 µg/mL), quando comparado ao ácido ascórbico. Porém, a inibição de HOCl não foi tão eficiente (IC50 = 714,1 ± 31,4 µg/mL). O EFO inibiu a hemólise induzida em eritrócitos de maneira dependente da concentração (IC50 = 7,8 ± 1,1 µg/mL), assim como a peroxidação lipídica e a formação de meta-hemoglobina, com valores de IC50 de 38,0 ± 11,7 e 186,3 ± 29,7 µg/mL, respectivamente. Os resultados obtidos neste estudo sugerem que extrato de folhas de oliveira possui efetiva atividade antioxidante em sistemas biológicos, pelo efeito sequestrador de determinadas espécies reativas que participam dos processos bioquímicos, e pela prevenção de danos oxidativos em eritrócitos humanos. Portanto, sua ingestão pode estar relacionada com a prevenção de estresse oxidativo in vivo, com consequentes benefícios à saúde. The aim of this study was to evaluate the antioxidant activity of olive leaf extract (OLE) (Olea europaea L.) by different in vitro and in situ analytical methodologies for determination of its effect in biological systems. The extract was obtained from dried olive leaves, previously micronized, in methanol/water (80/20%) in ratio 1:20 (w/v), after removal of n-hexane soluble compounds. After lyophilization, OLE was analyzed for reducing power by Folin-Ciocalteau, total flavonoid content, oleuropein content, ferric reducing antioxidant power (FRAP) and antioxidant activity against DPPHo, ABTSo+, superoxide anion (O2o-), hypochlorous acid (HOCl), and nitric oxide (NOo). The extract was also analyzed against oxidative damage in human erythrocytes. Ascorbic acid was used as reference. The experiment was repeated six times (n = 6) and the assays were performed in duplicate. The reducing power and total flavonoid and oleuropein contents of extract were 131.7 ± 9.4 mg galic acid equivalent/g dry extract (dw), 19.4 ± 1.3 mg quercetin equivalent/g dw, and 25.5 ± 5.2 mg oleuropein/g dw, respectively. The FRAP assay was 281.8 ± 22.8 mg trolox equivalent/g dw. The OLE was effective in inhibition of DPPHo and ABTSo+ radicals, in a concentration dependent manner, with IC50 values of 13.8 ± 0.8 and 16.1 ± 1.2 µg/mL, respectively. In relation to the antioxidant activity against reactive species of biological importance, the OLE showed high ability to inhibit O2o- (IC50 = 52.6 ± 2.1 µg/mL) and NOo (IC50 = 48.4 ± 6.8 µg/mL), when compared to ascorbic acid. However, inhibition of HOCl was not as efficient (IC50 = 714.1 ± 31.4 µg/mL). The OLE inhibited induced erythrocyte hemolysis in a concentration dependent manner (IC50 = 7.8 ± 1.1 µg/mL) as well as lipid peroxidation and the formation of methemoglobin, with IC50 values of 38.0 ± 11.7 and 186.3 ± 29.7 µg/mL, respectively. The results suggest that olive leaf extract has effective antioxidant activity in biological systems, based on its scavenging effect of certain reactive species that participate in biochemical processes, and the prevention of oxidative damage in human erythrocytes. So, the intake of olive leaf extract may be related to the prevention of in vivo oxidative stress, with health benefits.
