108. A cyclic effect of cAMP and calcium signaling contributes to jujube growth and development

Abstract

3′,5′-Cyclic adenosine monophosphate (cAMP) is an important metabolite that is specifically enriched in jujube. However, the effect of cAMP on jujube cellular responses has not been comprehensively studied. Here, we established jujube cell suspension cultures and investigated the calcium influx in response to cAMP treatment through protoplast isolation and fluorescence intensity. Firstly, cAMP treatment could promote jujube growth and increase the content of endogenous cAMP. Using transcriptome analysis with transgenic Arabidopsis plants overexpressing adenylate cyclase (ZjAC) as a positive control, we identified 60 calcium-related differential expressed genes (DEGs) that contributed to the calcium signaling and inter- or intra-cellular responses. Pharmacological treatments such as cAMP and the calcium ionophore A23187 could induce ZjAC expression, the accumulation of cAMP and calcium influx in jujube cells, while ethylene glycol tetraacetic acid (EGTA) or bithionol treatment inhibited these changes. Moreover, the calcium channels and transporters in calcium influx, such as the ZjCNGC2 channel and the mitogen activated protein (MAP) kinase pathway, could be activated by cAMP treatment. In summary, our findings demonstrated that cAMP biosynthesis is dependent on calcium influx and the amplifying effect between calcium and cAMP may be involved in intracellular signal induction, which might contribute to the growth and development of jujube.

Abstract

Background: Manganese (Mn) and iron (Fe) are essential trace elements for humans, yet excessive exposure to Mn or Fe can accumulate in the central nervous system (CNS) and cause neurotoxicity. The purpose of this study was to investigate the effects of Mn and Fe exposure, alone or in combination, on inducing oxidative stress-induced neurological damage in rat cortical and SH-SY5Y cells, and to determine whether combined exposure to these metals increases their individual toxicity. Methods: SH-SY5Y cells and male Sprague-Dawley rats were used to observe the effects of oxidative stress-induced neurological damage induced by exposure to manganese and iron alone or in combination. To detect the expression of anti-oxidative stress-related proteins, Nrf2, HO-1, and NQO1, and the apoptosis-related proteins, Bcl2 and Bax, and the neurological damage-related protein, α-syn. To detect reactive oxygen species generation and apoptosis. To detect the expression of the rat cortical protein Nrf2. To detect the production of proinflammatory cytokines. Results: We demonstrate that juvenile developmental exposure to Mn and Fe and their combination impairs cognitive performance in rats by inducing oxidative stress causing neurodegeneration in the cortex. Mn, Fe, and their combined exposure increased the expression of ROS, Bcl2, Bax, and α-syn, activated the inflammatory factors IL-6 and IL-12, inhibited the activities of SOD and GSH, and induced oxidative stress-induced neurodegeneration both in rats and SH-SY5Y cells. Combined Mn-Fe exposure attenuated the oxidative stress induced by Mn and Fe exposure alone by increasing the expression of antioxidant factors Nrf2, HO-1, and NQO1. Conclusion: In both in vivo and in vitro studies, manganese and iron alone or in combination induced oxidative stress, leading to neuronal damage. In contrast, combined exposure to manganese and iron mitigated the oxidative stress induced by exposure to manganese and iron alone by increasing the expression of antioxidant factors. Therefore, studies to elucidate the main causes of toxicity and establish the molecular mechanisms of toxicity should help to develop more effective therapeutic modalities in the future.

Abstract

Global climate change, extreme climate events, earthquakes and their accompanying natural disasters pose significant risks to humanity. Yet due to the nonlinear feedbacks, strategic interactions and complex structure of the Earth system, the understanding and in particular the predicting of such disruptive events represent formidable challenges for both scientific and policy communities. During the past years, the emergence and evolution of Earth system science has attracted much attention and produced new concepts and frameworks. Especially, novel statistical physics and complex networks-based techniques have been developed and implemented to substantially advance our knowledge for a better understanding of the Earth system, including climate extreme events, earthquakes and Earth geometric relief features, leading to substantially improved predictive performances. We present here a comprehensive review on the recent scientific progress in the development and application of how combined statistical physics and complex systems science approaches such as, critical phenomena, network theory, percolation, tipping points analysis, as well as entropy can be applied to complex Earth systems (climate, earthquakes, etc.). Notably, these integrating tools and approaches provide new insights and perspectives for understanding the dynamics of the Earth systems. The overall aim of this review is to offer readers the knowledge on how statistical physics approaches can be useful in the field of Earth system science.

Abstract

The gravitational wave detector of higher sensitivity and greater bandwidth in kilohertz window is required for future gravitational wave astronomy and cosmology. Here we present a new type broadband high frequency laser interferometer gravitational wave detector utilizing polarization of light as signal carrier. Except for Fabry-Perot cavity arms we introduce dual power recycling to further amplify the gravitational wave signals. A novel method of weak measurement amplification is used to amplify signals for detection and to guarantee the long-term run of detector. Equipped with squeezed light, the proposed detector is shown sensitive enough within the window from 300Hz to several kHz, making it suitable for the study of high frequency gravitational wave sources. With the proposed detector added in the current detection network, we show that the ability of exploring binary neutron stars merger physics be significantly improved. Abstract

Changes in CO$_2$ emissions during the COVID-19 pandemic have been estimated from indicators on activities like transportation and electricity generation. Here, we instead use satellite observations together with bottom-up information to track the daily dynamics of CO$_2$ emissions during the pandemic. Unlike activity data, our observation-based analysis can be independently evaluated and can provide more detailed insights into spatially-explicit changes. Specifically, we use TROPOMI observations of NO$_2$ to deduce ten-day moving averages of NO$_x$ and CO$_2$ emissions over China, differentiating emissions by sector and province. Between January and April 2020, China's CO$_2$ emissions fell by 11.5% compared to the same period in 2019, but emissions have since rebounded to pre-pandemic levels owing to the fast economic recovery in provinces where industrial activity is concentrated.

Abstract

The Indian summer monsoon rainfall (ISMR) has a decisive influence on India's agricultural output and economy. Extreme deviations from the normal seasonal amount of rainfall can cause severe droughts or floods, affecting Indian food production and security. Despite the development of sophisticated statistical and dynamical climate models, a long-term and reliable prediction of the ISMR has remained a challenging problem. Towards achieving this goal, here we construct a series of dynamical and physical climate networks based on the global near surface air temperature field. We uncover that some characteristics of the directed and weighted climate networks can serve as efficient long-term predictors for ISMR forecasting. The developed prediction method produces a forecast skill of 0.5 with a 5-month lead-time in advance by using the previous calendar year's data. The skill of our ISMR forecast, is comparable to the current state-of-the-art models, however, with quite a short (i.e., within one month) lead-time. We discuss the underlying mechanism of our predictor and associate it with network-delayed-ENSO and ENSO-monsoon connections. Moreover, our approach allows predicting the all India rainfall, as well as forecasting the different Indian homogeneous regions' rainfall, which is crucial for agriculture in India. We reveal that global warming affects the climate network by enhancing cross-equatorial teleconnections between Southwest Atlantic, Western part of the Indian Ocean, and North Asia-Pacific with significant impacts on the precipitation in India. We find a hotspots area in the mid-latitude South Atlantic, which is the basis for our predictor. Remarkably, the significant warming trend in this area yields an improvement of the prediction skill.

Abstract

AIM: To investigate the short-term visual quality outcomes after femtosecond laser small incision lenticule extraction(SMILE)and evolution implantable collamer lens(EVO-ICL)implantation for the correction of moderate myopia.METHODS: Prospective control study. A total of 51 cases(51 eyes)with moderate myopia who underwent SMILE or EVO-ICL implantation surgery at Weifang Eye Hospital from April 2021 to February 2022 were selected. They were divided into SMILE group(30 patients, 30 eyes)and EVO-ICL group(21 patients, 21 eyes)according to the surgical methods. The changes of visual acuity [uncorrected distance visual acuity(UDVA), corrected distance visual acuity(CDVA)], diopter [spherical equivalent(SE)] and related parameters of optical quality analysis system(OQAS Ⅱ)were observed before surgery and at 1wk, 1 and 3mo after surgery, and the quality of vision(QoV)questionnaire was completed.RESULTS: At 3mo after surgery, the safety index(postoperative CDVA/preoperative CDVA)of SMILE gruop and EVO-ICL group were 1.20(1.00, 1.20)and 1.20(1.00, 1.38), respectively, the efficacy index(postoperative UDVA/preoperative CDVA)were 1.00(1.00, 1.20)and 1.00(1.00, 1.20), respectively, and the percentage of SE within ±0.50D was 87% and 100%, respectively. In SMILE group, the objective scattering index(OSI)was increased after surgery, while modulation transfer function cutoff frequency(MTF cutoff), contrast visual acuity(VA)100%, and VA20% at 1wk and 1mo after surgery, and Strehl ratio(SR)and VA9% at each time point after surgery were all decreased compared with those before surgery(all P0.05). The most common visual symptoms after SMILE and EVO-ICL implantation were visual haze and halos, respectively.CONCLUSION: Both SMILE and EVO-ICL implantation have good safety, efficacy and predictability in the short term after the correction of moderate myopia. Both groups had visual symptoms after surgery, but the overall satisfaction of patients was high. Furthermore, EVO-ICL implantation has better objective visual quality performance.

Abstract

AIM:To observe the changes in corneal aberrations and the characteristics of visual quality after transepithelial photorefractive keratectomy(T-PRK)and femtosecond small incision lenticule extraction(SMILE)in the correction of low myopia.METHODS: Prospective cohort study. A total of 32 cases(32 eyes)with low myopia who underwent T-PRK surgery and 45 cases(45 eyes)of SMILE surgery at Weifang Eye Hospital from April 2021 to April 2022 were selected. The uncorrected visual acuity(UCVA), best corrected visual acuity(BCVA), spherical equivalent(SE), corneal higher-order aberrations(HOAs)and objective visual quality were compared between the two groups.RESULTS:All patients completed the surgery successfully without complications such as infection. At 3mo postoperatively, the safety index was 1.13±0.16 and 1.16±0.17(P=0.48)and the efficacy index was 1.10±0.20 and 1.15±0.18(P=0.27)in the T-PRK and SMILE groups, respectively. The percentage of UCVA(LogMAR)≤0 in the T-PRK and SMILE groups was 94% and 98%, respectively. The percentage of the residual SE within ±0.5D was 88% and 87% in the two groups, respectively. The HOAs and spherical aberration in both groups were significantly increased(P≤0.01), and the increase was not statistically significant between the two groups(P=0.31, 0.89). There was no significant change in horizontal coma, horizontal trefoil and vertical trefoil in both groups(P>0.05). The vertical coma in SMILE group was significantly increased(P0.05), and the increase was significantly greater in SMILE group than in T-PRK group(P0.05).CONCLUSION:Both T-PRK and SMILE showed good safety, efficacy, and visual quality in correcting low myopia, while SMILE induced more vertical coma than T-PRK.

Abstract

The El Nino Southern Oscillation (ENSO) is one of the most prominent interannual climate phenomena. An early and reliable ENSO forecasting remains a crucial goal, due to its serious implications for economy, society, and ecosystem. Despite the development of various dynamical and statistical prediction models in the recent decades, the ``spring predictability barrier'' (SPB) remains a great challenge for long (over 6-month) lead-time forecasting. To overcome this barrier, here we develop an analysis tool, the System Sample Entropy (SysSampEn), to measure the complexity (disorder) of the system composed of temperature anomaly time series in the Nino 3.4 region. When applying this tool to several near surface air-temperature and sea surface temperature datasets, we find that in all datasets a strong positive correlation exists between the magnitude of El Nino and the previous calendar year's SysSampEn (complexity). We show that this correlation allows to forecast the magnitude of an El Nino with a prediction horizon of 1 year and high accuracy (i.e., Root Mean Square Error $=0.23^\circ C$ for the average of the individual datasets forecasts). For the on-going 2018 El Nino event, our method forecasts a weak El Nino with a magnitude of $1.11\pm 0.23^\circ C$. Our framework presented here not only facilitates a long--term forecasting of the El Nino magnitude but can potentially also be used as a measure for the complexity of other natural or engineering complex systems.

Abstract

We experimentally realize the internal and external entanglement tradeoff, which is a new kind of entanglement monogamy relation different from that usually discussed. Using a source of twin photons, we find that the external entanglement in polarization of twin photons, and the path-polarization internal entanglement of one photon, limit each other. In the extreme case, when the internal state is maximally entangled, the external entanglement must be vanishing, that illustrate entanglement monogamy. Our results of the experiment coincide with the theoretical predictions, and therefore provide a direct experimental observation of the internal and external entanglement monogamy relation., Comment: 10 pages, 7 figures

Abstract

Sustainable financial markets play an important role in the functioning of human society. Still, the detection and prediction of risk in financial markets remain challenging and draw much attention from the scientific community. Here we develop a new approach based on combined network theory and machine learning to study the structure and operations of financial product markets. Our network links are based on the similarity of firms' products and are constructed using the Securities Exchange Commission (SEC) filings of US listed firms. We find that several features in our network can serve as good precursors of financial market risks. We then combine the network topology and machine learning methods to predict both successful and failed firms. We find that the forecasts made using our method are much better than other well-known regression techniques. The framework presented here not only facilitates the prediction of financial markets but also provides insight and demonstrate the power of combining network theory and machine learning.

Abstract

We experimentally test the recently predicted anisotropic invariance properties of pure three-qubit states, via generation and measurement of polarisation-path entangled three-qubit states. These properties do not require aligned reference frames and can be determined from measurements on any two of the qubits. They have several applications, such as a universal ordering of pairwise quantum correlations, strong monogamy relations for Bell inequalities and quantum steering, and a complementarity relation for Bell nonlocality versus 3-tangle, some of which we also test. The results indicate that anisotropic invariance, together with the three qubit Bloch vector lengths, can provide a robust and complete set of invariants for such states under local unitary transformations., Comment: 6 pages, 1 figure

Abstract

Wavefunction is the foundation of quantum theory, which is assumed to give a complete description of a quantum system. For a long time, wavefunction is introduced as an abstract element of the theory and there lacks effective ways to measure it directly. The situation, however, is somewhat changed when Lundeen et al. reported the direct measurement of the quantum wavefunction via weak measurements, which gives the wavefunction a clearly operational definition [Nature 474, 188 (2011)]. The weak measurement method requires sequential measurements of conjugate observables position and momentum with the position measurement is weak enough. Surprisingly, the recent research by Vallone and Dequal shows that performing sequential strong measurements realizes the same target, in which case no approximation has to be made compared to the case of weak measurements[Phys. Rev. Lett. 116, 040502 (2016)]. Here we experimentally report the direct measurement of the two-dimensional transverse wavefunction of photons via strong measurements for the first time, which implies that an accurate and clear operational definition can be given to wavefunction. We have measured the Gaussian and Laguerre-Gaussian of l = 1 spatial wavefunctions of photons with R-square are 0.97 and 0.93 respectively. As a potentially important application, we show that the direct measurement of two-dimensional wavefunction provides an alternative way to realize digital holography of three-dimensional objects. The results presented here will not only deepen our understanding of abstract wavefunction but also have significant applications in quantum information processing and quantum imaging., Comment: 5 pages

Abstract

This study aimed to compare the predictive value of six selected anthropometric indicators for benign prostatic hyperplasia (BPH). Males over 50 years of age who underwent health examinations at the Health Management Center of the Second Xiangya Hospital, Central South University (Changsha, China) from June to December 2020 were enrolled in this study. The characteristic data were collected, including basic anthropometric indices, lipid parameters, six anthropometric indicators, prostate-specific antigen, and total prostate volume. The odds ratios (ORs) with 95% confidence intervals (95% CIs) for all anthropometric parameters and BPH were calculated using binary logistic regression. To assess the diagnostic capability of each indicator for BPH and identify the appropriate cutoff values, receiver operating characteristic (ROC) curves and the related areas under the curves (AUCs) were utilized. All six indicators had diagnostic value for BPH (all P ≤ 0.001). The visceral adiposity index (VAI; AUC: 0.797, 95% CI: 0.759–0.834) had the highest AUC and therefore the highest diagnostic value. This was followed by the cardiometabolic index (CMI; AUC: 0.792, 95% CI: 0.753–0.831), lipid accumulation product (LAP; AUC: 0.766, 95% CI: 0.723–0.809), waist-to-hip ratio (WHR; AUC: 0.660, 95% CI: 0.609–0.712), waist-to-height ratio (WHtR; AUC: 0.639, 95% CI: 0.587–0.691), and body mass index (BMI; AUC: 0.592, 95% CI: 0.540–0.643). The sensitivity of CMI was the highest (92.1%), and WHtR had the highest specificity of 94.1%. CMI consistently showed the highest OR in the binary logistic regression analysis. BMI, WHtR, WHR, VAI, CMI, and LAP all influence the occurrence of BPH in middle-aged and older men (all P ≤ 0.001), and CMI is the best predictor of BPH.

Abstract

Aquilaria sinensis is an important non-timber tree species for producing high-value agarwood, which is widely used as a traditional medicine and incense. Agarwood is the product of Aquilaria trees in response to injury and fungal infection. The APETALA2/ethylene responsive factor (AP2/ERF) transcription factors (TFs) play important roles in plant stress responses and metabolite biosynthesis. In this study, 119 AsAP2/ERF genes were identified from the A. sinensis genome and divided into ERF, AP2, RAV, and Soloist subfamilies. Their conserved motif, gene structure, chromosomal localization, and subcellular localization were characterized. A stress/defense-related ERF-associated amphiphilic repression (EAR) motif and an EDLL motif were identified. Moreover, 11 genes that were highly expressed in the agarwood layer in response to whole-tree agarwood induction technique (Agar-Wit) treatment were chosen, and their expression levels in response to methyl jasmonate (MeJA), salicylic acid (SA), or salt treatment were further analyzed using the quantitative real time PCR (qRT-PCR). Among the 11 genes, eight belonged to subgroup B-3. All 11 genes were significantly upregulated under salt treatment, while eight genes were significantly induced by both MeJA and SA. In addition, the gene clusters containing these upregulated genes on chromosomes were observed. The results obtained from this research not only provide useful information for understanding the functions of AP2/ERF genes in A. sinensis but also identify candidate genes and gene clusters to dissect their regulatory roles in agarwood formation for future research. Le bois d'aloes resulte de la blessure et de l'infection par un champignon des arbres d'Aquilaria. Les facteurs de transcription (FT) AP2/ERF jouent des roles importants dans les reponses aux stress et dans la synthese des metabolites. Dans ce travail, 119 genes AsAP2/ERF ont ete identifies au sein du genome de l'A. sinensis et divises en sous-familles (ERF, AP2, RAF et Soloist). Leurs motifs conserves, structure genique, emplacement chromosomique et localisation subcellulaire ont ete caracterises. Un motif EAR (<< ERF-associated amphiphilic repression >>) associe au stress ou a la defense ainsi qu'un motif EDLL ont ete identifies. De plus, 11 genes qui etaient fortement exprimes au sein de la couche de bois responsable de la production de bois d'aloes, en reponse a la technique d'induction sur arbres entiers (traitement Agar-Wit), ont ete choisis et leur niveau d'expression en reponse a divers traitements (MeJA, SA ou salinite) a ete analyse plus a fond par qRT-PCR. Parmi ces 11 genes, huit appartenaient au sous-groupe B-3. Tous les genes etaient exprimes significativement plus fortement en reponse au stress salin, tandis que huit de ces genes etaient induits a la fois par le MeJA et le SA. De plus, les amas geniques contenant ces genes surexprimes ont fait l'objet d'observations. Les resultats de ces travaux fournissent non seulement de l'information utile pour comprendre les fonctions des genes AP2/ERF chez l'A. sinensis, mais apportent egalement des genes candidats et des amas geniques pour dissequer leurs roles dans la regulation de la formation du bois d'aloes.
