Abstract

Stem cells have recently emerged as an important candidate for cell therapy. However, some major limitations still exist such as a small quantity of cell supply, senescence, and insufficient differentiation efficiency. Therefore, there is an unmet need to control stem cell behavior for better clinical performance. Since native microenvironment factors including stem cell niche, genetic factors, and growth factors direct stem cell fate cooperatively, user-specified in vitro settings are required to understand the regulatory roles and effects of each factor, thereby applying the factors for improved cell therapy. Among others, various types of biomaterials and transfection method have been employed as key tools for development of the in vitro settings. This review focuses on the current strategies to improve stemness maintenance, direct differentiation, and reprogramming using biomaterials and genetic factors without any aids from additional biochemicals and growth factors.

Abstract

To investigate the features of macrophage-like cell (MLC) characterized by en face optical coherence tomography (OCT) in Behçet's uveitis (BU). The extent of fluorescein vascular leakage (FVL) was graded on a scale of 0-3 (0=none, 1=mild, 2=moderate, 3=severe) for the optic nerve head (ONH), macula and peripheral retina. The 3μm en face OCT slabs on inner limiting membrane of ONH or macular region was used to visualize the MLCs. The MLC densities of BU group in ONH and macular region were significantly higher than the control group (both p<0.001). The ONH and macular MLC density were significantly higher in eyes with higher FVL grade and they were positively correlated with FVL score (all p<0.001). MLC density was elevated in Behçet's uveitis and it may serve as a noninvasive indicator for the severity of fluorescein leakage and retinal inflammation in Behcet's uveitis. [ABSTRACT FROM AUTHOR]

Abstract

Mesenchymal stem cells (MSCs) play a crucial role in tissue engineering, as their differentiation status directly affects the quality of the final cultured tissue, which is critical to the success of transplantation therapy. Furthermore, the precise control of MSC differentiation is essential for stem cell therapy in clinical settings, as low-purity stem cells can lead to tumorigenic problems. Therefore, to address the heterogeneity of MSCs during their differentiation into adipogenic or osteogenic lineages, numerous label-free microscopic images were acquired using fluorescence lifetime imaging microscopy (FLIM) and stimulated Raman scattering (SRS), and an automated evaluation model for the differentiation status of MSCs was built based on the K-means machine learning algorithm. 134. Optimization for better Immunohistochemistry assay and more comprehensive pathological interpretation for PD-L1 expression in Classical Hodgkin lymphoma

Abstract

Bckgoround:Treating the relapsed or refractory Classical Hodgkin Lymphomas were still challenging, previous studies indicated they can benefit from immune checkpoint inhibitors, over expression of PD-L1 can also be predictive marker for anti -PD -1 therapeutic efficacy, however, harmonization with different IHC assay s and explanation of the PD -L1 immunostaining results remains controversial in CHL. In this study, we sought to optimize the PD-L1 Immunohistochemistry (IHC) assay in classical Hodgkin lymphoma (CHL). Methods: All tests were performed on tumor tissue microarray established from 54 CHLs. Three IHC assays (405.9A11, SP142, 22C3) for PD-L1 expression were compared semi-quantitatively with RNAscope assay (No. 310035, ACD), and the difference for their expression on background immune cells (ICs), their associations with densities of TIL/TAM makers and their implication on survival were also analyzed. Results: 405.9A11 assay demonstrated best specificity on HRS cells and sensitivity on ICs. Thus, positive expression of PD-L1 was more frequent on ICs (85.2%) than on HRS cells (48.1%). Different subgroups of background IC cells including tumor associated macrophages (TAMs) were assessed and scored by CD4, CD8, FOXP3, and CD163. PD-L1 expression on ICs were most associated with elevated densities of TAMs, and in concordance with TAMs(P=0.026), PD-L1 expression on ICs ( P=0.067)indicated a negative effect on survival.Conclusions: 405.9A11 assay was proved to be most convincible for demonstrating PD-L1 expression，pathologists should report PD-L1 expression in a combined manner including both positive status of HRS cells and positive percentage of ICs.

148. Induction of Neuronal Differentiation of Rat Muscle-Derived Stem Cells in Vitro Using Basic Fibroblast Growth Factor and Ethosuximide

Abstract

Several studies have demonstrated that basic fibroblast growth factor (bFGF) can induce neural differentiation of mesenchymal stem cells. In this study, we investigated the neural differentiation of muscle-derived stem cells (MDSCs) following treatment with bFGF and ethosuximide, a small molecule used as an anticonvulsant in humans. Stem cells isolated from rat skeletal muscle (rMDSCs) were pre-induced by culturing with 25 ng/mL bFGF for 24 h and then were transferred to a medium supplemented with or without 4 mM ethosuximide. Neuronal differentiation was assessed by immunocytochemical and western blotting analyses of marker expression. Immunocytochemistry of rMDSCs treated with bFGF and ethosuximide identified abundant cells expressing neuronal markers (TuJ1, neuron-specific class III β-tubulin; NeuN, neuronal nuclear antigen; and NF-MH; neurofilament M and H). Olig2 (oligodendrocyte transcription factor 2)-positive cells were also observed, indicating the presence of oligodendrocyte lineage cells. These findings were substantiated by western blotting analysis of marker proteins. In particular, the expression of NeuN and TuJ1 was significantly higher in rMDSCs treated with ethosuximide and bFGF than in cells stimulated with bFGF alone (NeuN, p < 0.05 and TuJ1, p < 0.001). Expression of the astrocyte marker GFAP (glial fibrillary acidic protein) was not detected in this study. Collectively, the results showed that treatment with bFGF and ethosuximide induced effective transdifferentiation of rMDSCs into cells with a neural-like phenotype. Notably, rMDSCs treated with a combination of bFGF plus ethosuximide showed enhanced differentiation compared with cells treated with bFGF alone, implying that ethosuximide may stimulate neuronal differentiation.
