The mixed ion effect (MIE) in Li/Na–NaZr2(PO4)3 with a three-dimensional diffusion path was observed. The conductivity minimum at an ionic ratio 0.8 was lower by about three orders of magnitude than for either end member. Hexagonal parameters a, c and 7Li MAS/NMR spectral intensity against the ionic ratio showed a linear change. The spectral width of 31P MAS NMR at chemical shift 5.9 ppm against the ionic ratio showed a minimum at the same ionic ratio with the conductivity data. The behavior suggests that the conductivity minimum results from the inhomogeneous distribution of two unlike ions and a blocking of diffusion path. [Copyright &y& Elsevier]

Dynein from inner arms of Chlamydomonasflagella contains sevendistinct subspecies, a through g. Several lines of evidence suggest thesesubspecies play important roles in generating flagellar beating and thatthe different subspecies are functionally diverse. To evaluate theirroles and diversity, the mechanical properties of subspecies-c, which isa single-headed motor, were examined using optical trap nanometry. Apolystyrene bead coated with a small number of subspecies-c moleculeswas captured with the optical trap and brought into contact with amicrotubule fixed to a coverslip. Movements of the bead were measured bya quadrant photodiode sensor with sub-nanometer- and millisecond-resolution.Beads carrying a single active subspecies-c molecule moved processivelyalong the microtubules in 8-nm steps but slipped backwards under highloads. Force-velocity relationships of single subspecies-c molecules werealmost linear and the shapes of the normalized curves at 5 μM and 100μM ATP were similar. These results indicate that dynein subspecies-cfunctions in a very different way from conventional motor proteins, suchas myosin and kinesin, and has properties that could produceself-oscillation in vivo. The ionic conductivity against the ionic ratio showed a shallow minimum in Na/Ag-NaZr2(PO4)3 with a three-dimensional diffusion path. The ionic conductivity of the mixed ion system Na/Ag was calculated by means of the path probability method (PPM) using a two-dimensional honeycomb lattice model, resulting from the weak interaction between two different ions.23Na MAS NMR spectra showed the second-order quadrupole interaction with the electric field gradient. Despite an emphasis on the prognostic impact of serum soluble interleukin-2 receptor (sIL-2R) at diagnosis in patients with diffuse large B-cell lymphoma (DLBCL), whether the prognostic impact of elevated sIL-2R is linear remains unclear. To verify the presence of a non-linear association between sIL-2R level at diagnosis and overall survival (OS) in patients with newly diagnosed DLBCL, we conducted a multi-center, observational retrospective study. Among 488 analyzable patients, Cox proportional hazards modeling identified serum sIL-2R level at diagnosis as an independent predictor of OS. Multivariate Cox hazard modeling with restricted cubic spline model demonstrated that the relationship between serum sIL-2R level and OS was clearly non-linear (P for effect of sIL-2R = 0.002; P for non-linearity = 0.015). Mortality risk increased gradually as sIL-2R levels increased and plateaued at approximately 5,000 U/mL. Segmented regression analysis revealed that the trend in negative prognostic impact from a gradual increase in serum sIL-2R level changed significantly, with a breakpoint at approximately 2,000 U/mL. Multivariate receiver operating characteristic curves showed a significant improvement in prediction ability when serum sIL-2R level was added to the International Prognostic Index (IPI). Serum sIL-2R level at diagnosis was not only a prognostic factor, but also improved predictive accuracy for OS when incorporated with the IPI. Background: The association between surgical performance ratings and clinical outcomes in robotic surgery is poorly understood. Additionally, no studies have reported on the relationship between the surgeon's initial case-skill evaluation and the learning curve in robot-assisted surgery. We evaluated whether an objective surgical technique evaluation score for initial robot-assisted radical prostatectomy (RARP) was associated with clinical outcomes and surgeons' learning curves., Methods: Six surgeons who were trained in and started to perform RARP at our institution were included. Anonymized, unedited videos of each surgeon's 10th RARP case were evaluated by three reviewers, using modified Objective Structured Assessment of Technical Skill (OSATS) scores. We then divided the surgeons into two groups on the basis of these OSATS scores. We retrospectively compared the clinical outcomes and learning curves of the console time of the two groups for consecutive RARPs, performed from March 2018 to July 2023., Results: We analyzed 258 RARPs (43 cases/surgeon), including 129 cases performed by high-OSATS score surgeons (18.2-19.3 points) and 129 cases performed by low-OSATS score surgeons (11.9-16.0 points). Overall, the high-OSATS score group had significantly shorter operation and console times than the low-OSATS score group did (both P < 0.01) and their patients' rate of continence recovery by 3 months post-RARP was significantly higher (P = 0.03). However, complications, blood loss, and positive margins did not differ between the groups (P = 0.08, P = 0.51, and P = 0.90, respectively). The high-OSATS score group had a significantly shorter console time than the low-OSATS score group did after the 11-20 cases., Conclusions: The OSATS score in early RARP cases can predict subsequent surgical outcomes and surgeons' learning curves., (© 2024. Introduction Prolonged standing during surgery is a cause of musculoskeletal disorders in surgeons. Wearable chairs have reportedly reduced musculoskeletal strain effectively when used in industry. However, discomfort and instability issues may remain. This study examines whether using a wearable chair for surgical procedures imposes negative effects, such as mental or physical strain on surgeons prior to its clinical implementation. Methods This prospective cross-over study compared mental strain and subjective sensations in simulated suturing tasks with and without using Archelis®, a wearable chair (Archelis Inc, Yokohama, Japan). Six surgeons participated in the study. Mental strain was examined using heart rate variations calculated during tasks. Four subjective sensations (fatigue, comfort, balance, and workability) after each task and differences in body localized pain pre- and post-task were compared using a 10 centimetres (cm)-visual analog scale (VAS) score questionnaire. Results Results showed no significant differences in mental strain nor subjective sensations with or without the wearable chair. The mean VAS scores for all four subjective sensations with the wearable chair were relatively positive. There was a slight yet insignificant post-task increase in VAS mean scores for body-localized pain in the lower legs. Conclusions Significant negative effects on surgeons from the wearable chair were not observed during simulated suturing procedures. Mechanical stress significantly affects the physiological functions of cells, including tissue homeostasis, cytoskeletal alterations, and intracellular transport. As a major cytoskeletal component, microtubules respond to mechanical stimulation by altering their alignment and polymerization dynamics. Previously, we reported that microtubules may modulate cargo transport by one of the microtubule-associated motor proteins, dynein, under compressive mechanical stress. Despite the critical role of tensile stress in many biological functions, how tensile stress on microtubules regulates cargo transport is yet to be unveiled. The present study demonstrates that the low-level tensile stress-induced microtubule deformation facilitates dynein-driven transport. We validate our experimental findings using all-atom molecular dynamics simulation. Our study may provide important implications for developing new therapies for diseases that involve impaired intracellular transport.

The effect of treatment with efgartigimod in seronegative myasthenia gravis (MG) remains unclear. This retrospective study aimed to evaluate symptomatic changes and safety of treatment with efgartigimod in patients with generalized MG (gMG) double-seronegative for acetylcholine receptor antibody and muscle-specific kinase antibody. We reviewed the medical records of double-seronegative gMG treated with 10 mg/kg efgartigimod once/week per cycle (4 weeks) from June 2022 to June 2023. A total of 16 patients were included. MG-activities of daily living (ADL) scores improved from 9.2 to 7.4. Mean prednisolone dose was reduced from 5.4 to 4.1 mg/day. The duration before MG-ADL deterioration after the end of a cycle was 6.1 weeks. Five patients had mild adverse events. This paper describes a method for recording X-ray diffraction patterns from a small amount of fibrous protein materials while being oriented by using a micro shear-flow cell. This cell consists of two concentrically arranged glass tubes. The inner tube is stationary, while the outer one rotates at a high speed. The gap between the two tubes is about 100 μm, into which the suspension of fibrous protein materials is injected. By using synchrotron-radiation X-ray microbeams (diameter, 10 μm), clear diffraction images from oriented protein materials can be recorded. The required volume of the sample is only about 10 μl. This method can also be combined with the laser-flash photolysis of caged compounds.
