Chapter 2 of the book "James Joyce" is presented. The chapter looks at the influence of Parnellism and Fenianism on the middle-class family of author James Joyce in Dublin, Ireland. His father John Stanislaus was appointed as a rate collector with the Dublin civil service following the approval of the British chief secretary of Ireland but he lost the post due to drinking problems. The Joyce family belonged to the stratum of Dublin Catholic society that was empowered by Parnellism.

104. Dynamic Electrodeposition of Manganese Dioxide: Temporal Variation in the Electrodeposition Mechanism

Abstract

The time resolved mechanism of electrodeposition and the effect of this changing mechanism on the nucleation and growth of solid manganese dioxide has been investigated in both acidic and neutral electrolytes on the rotating ring disc electrode (RRDE). The fate of the Mn3+ intermediate is a key feature of this electrodeposition mechanism, the formation of which is dependent on the substrate, which in this case is either platinum, MnO2 or MnOOH. On the platinum surface, for all electrolytes, soluble Mn3+ is produced initially. The stability of this soluble Mn3+ species determines the initial morphology, and rate of change of mechanism for the process. In a neutral electrolyte, nucleation and growth of MnO2 occurs primarily through the precipitation of a 2D film of MnOOH on the platinum, which rapidly covers the surface. Nucleation in an acidic H2SO4 system occurs primarily via a disproportionation route which forms 3D MnO2 hemispheroids that cover the substrate slowly. Subsequent growth of MnO2 in both electrolytes then proceeds via formation of a MnOOH film, which is subsequently oxidized in the solid state to form MnO2. MnOOH oxidation to MnO2 appears kinetically limited, which is overall a limiting factor in the electrodeposition process.

Abstract

This essay is about the relation between the treatment of the body in Beckett's major French texts of 1945–6 and the Vichy regime. It examines Vichy conceptions of physical life and their effect on Beckett's texts, considering those texts as responses to them. It addresses the ideological construction of the body in France 1940–4 and its connection with Vichy pastoralism, folkloric regionalism, natalism, familialism, and paternalism; the historical materiality of mutilated, impoverished, ‘inferior’, and expelled bodies and their significance under Vichy; and the influence on the Vichyite conception of the body of Le Play, Barrès, and above all Maurras, his nationalism, provincialism, and reactionary aesthetics. Beckett's letters show him to have been dismissive of these influences. Premier amourand the Nouvellesrepeatedly evoke certain features of the experience of bodily life under Vichy. They also conduct a war on Vichyite, Pétainist, and Maurrasian body politics and its moral terrorism, not least because the ideological construction of the body in Vichy France was strikingly close to that in de Valera's Ireland. The texts are a weird, ironical hymn to incapacity, to the ‘second-rate’ or ‘defective’ body. This in turn dictates the specific character of Beckett's break with representation at this time.

108. Are drug metabolites able to cause T-cell-mediated hypersensitivity reactions?

Abstract

Introduction:Adverse drug reactions with an immune pathogenesis are a problem in the clinic and an impediment to drug development. T lymphocytes are believed to play a role in the pathogenesis; however, the nature of the drug interaction with immune receptors remains an area of debate.Areas covered:This article reviews recent advances in our understanding of drug hypersensitivity focusing specifically on the way in which drugs are displayed in MHC molecules. Most drugs associated with a high incidence of reactions have been shown to form protein-reactive metabolites. Hence, the relationship between drug metabolism and T-cell activation is discussed in detail.Expert opinion:The role of metabolism in pathogenesis of immunological drug reactions has only been studied with a small number of drugs where synthetic metabolites are available for functional studies. In each case, metabolite-responsive T cells have been detected. However, the field is skewed by the fact that most research is conducted using the parent compound in metabolically inert cell systems. We propose that research efforts are directed towards the synthesis of drug metabolites and/or drug–protein conjugates. Furthermore, analytical methods need to be developed to relate metabolite exposure to the T-cell response. For now, our understanding of the chemical basis of drug hypersensitivity is incomplete.

126. Drug-Induced Hypersensitivity Syndrome (DIHS)/Drug Reaction with Eosinophilia and Systemic Symptoms (DRESS): Clinical Features and Pathogenesis

Abstract

Drug-induced hypersensitivity syndrome/drug reaction with eosinophilia and systemic symptoms (DIHS/DRESS) is one example of a severe delayed T-cell–mediated adverse drug reaction. DIHS/DRESS presents with fever, widespread rash and facial edema, organ involvement, and hematological abnormalities, including eosinophilia and atypical lymphocytosis. DIHS/DRESS is associated with relapse 2 to 4 weeks after acute symptoms, often coinciding with reactivation of prevalent chronic persistent human herpesviruses such as human herpesvirus 6, EBV, and cytomegalovirus. The mortality of DIHS/DRESS is up to 10% and often related to unrecognized myocarditis and cytomegalovirus complications, with longer-term consequences that contribute to morbidity including autoimmune diseases such as thyroiditis. It is essential that all potential drug causes, including all new drugs introduced within the 8 weeks preceding onset of DIHS/DRESS symptoms, are identified. All potential drug culprits, as well as drugs that are closely related structurally to the culprit drug, should be avoided in the future. Systemic corticosteroids have remained the mainstay for the treatment of DIHS/DRESS with internal organ involvement. Steroid-sparing agents, such as cyclosporine, mycophenolate mofetil, and monthly intravenous immune globulin, have been successfully used for treatment, and careful follow-up for cytomegalovirus reactivation is recommended. Strong associations between HLA class I alleles and DIHS/DRESS predisposition include HLA-B∗13:01 and dapsone, HLA-B∗58:01 and allopurinol, and HLA-B∗32:01 and vancomycin. These have opened a pathway for prevention, risk stratification, and earlier diagnosis. Single-cell sequencing and other studies of immunopathogenesis promise to identify targeted treatment approaches.

127. Practical Implementation of Genetics: New Concepts in Immunogenomics to Predict, Prevent, and Diagnose Drug Hypersensitivity

Abstract

Delayed drug hypersensitivities are CD8+ T-cell mediated reactions associated with up to 50% mortality. Human leukocyte antigen (HLA) alleles are known to predispose disease, specific to drug, reaction, and patient ethnicity, with pre-treatment screening recommended for a handful of the strongest associations to identify and prevent drug use in high-risk patients. However, an incomplete predictive value implicates other HLA-imposed risk factors, and low carriage of many identified HLA-risk alleles combined with the high cost of sequence-based typing has limited economic viability for similar recommendation of screening across drugs and healthcare systems. To mitigate, an expanding armoury of low-cost polymerase chain reaction-based screens is being developed, and HLA-imposed risk factors are being discovered. These include (i) polymorphic variants of metabolic and endoplasmic reticulum aminopeptidase enzymes, towards multi-allelic screening with increased predictivity, (ii) regulation by immune checkpoint inhibitors, enabling de-tolerised animal models of human disease, and (iii) immunodominant T-cell receptors (TCR) on clonally-expanded CD8+ T-cells. For the latter, HLA-risk restricted TCR provides immunogenomic strategies and samples from a single patient to identify (i) novel HLA-risk associations in underserved minority populations, (ii) tissue-relevant effector biomarkers towards earlier diagnosis and treatment, and (iii) HLA-TCR-presented immunogenic structures to aid future drug development.

Abstract

Serine phosphorylation of the FcγRI CY domain directs its migration to lipid rafts and its interaction with protein 4.1G, suggesting a tethering role for protein 4.1G. The high‐affinity IgG receptor (CD64, FcγRI) has several special capacities, including the receptor‐stimulated cleavage of the cell surface B cell‐activating factor of the TNF superfamily (TNFSF13B). With the use of the yeast two‐hybrid system, we and others have shown that FcγRI interacts with protein 4.1G (EPB41L2). Our mutational analyses identified two required 4.1G‐interacting regions in the FcγRI CY and one FcγRI‐interacting site in the C‐terminus of protein 4.1G. Herein, we explore mechanism(s) that may regulate the interaction between protein 4.1G and FcγRI CY and influence FcγRI membrane mobility and function. We show that FcγRI CY interacts with protein 4.1G in vitro and that FcγRI coimmunoprecipitates protein 4.1G in freshly isolated human PBMC. With the use of immunostaining, we show that FcγRI colocalizes with protein 4.1G in unstimulated U937 cells, in which the FcγRI CY is constitutively serine‐phosphorylated, but significant uncoupling occurs following FcγRI cross‐linking, suggesting phosphoserine‐regulated interaction. In vitro, protein 4.1G interacted preferentially with CK2‐phosphorylated FcγRI CY, and compared with WT FcγRI, a nonphosphorylatable FcγRI mutant receptor was excluded from lipid rafts, suggesting a key role for protein 4.1G in targeting phosphorylated FcγRI to rafts. These data are consistent with a phosphoserine‐dependent tethering role for protein 4.1G in maintaining FcγRI in lipid rafts and provide insight into the unique phosphoserine‐based regulation of receptor signaling by FcγRI CY.

Abstract

FcαRI (CD89) is a human IgA FcR expressed on cells of myeloid lineage such as neutrophils, monocytes, tissue macrophages, eosinophils, and subpopulations of dendritic cells. FcαRI mediates cell activation through Src family kinases and downstream tyrosine‐based phosphorylation pathways. However, the role of IgA and the expression and role of its cognate receptor FcαRI (CD89) in platelet activation are undefined. In the current study, we demonstrate that human platelets express FcαRI mRNAs and proteins. Furthermore, we show that the platelet FcαRI is associated with the FcR γ‐chain, and cross‐linking of FcαRI leads to Syk phosphorylation. Clustering of FcαRI induces pre‐mRNA splicing and protein production of tissue factor and IL‐1β, suggesting novel roles for human platelet FcαRI and serum IgA in thrombosis and inflammation.

131. “ALL PROPAGATED WITH THE BEST Intentions”: Greene, THE Us, AND INDOCHINA 1951–1955

Abstract

Graham Greene’s The Quiet American is an “epochal novel.” It announces the inception of a new epoch at a decisive historical moment: on March 15, 1954, US National Security Council Directive 5412 came into force. NSC 5412 more or less coincided with the onset of the Cold War and made a political imperative of the absolute deniability of covert actions. It thereby ushered in a specifically modern form of innocence. Greene’s novel is about an early version of that innocence. Modern innocence conceals the “principle of implosion” according to which ferocious opposites reverse into one another. Greene gives us his own version of implosion, the collapse of any private moral space putatively valorizable apart from global antagonisms. Modern innocence might seem principally identifiable with the Cold War era. However, Orwell’s analysis of doublethink gives the concept larger scope, as do Le Carré’s masterful analyses of its persistence in post-1997 British culture.

OBJECTIVE To define the molecular basis of the autosomal dominant progressive external ophthalmoplegia and parkinsonism in a large family with a dominantly transmitted multiple mitochondrial DNA deletion disorder. DESIGN Microsatellite analysis and screening of the progressive external ophthalmoplegia 1 (PEO1), adenine nucleotide translocator 1 (ANT1, and polymerase γ-1 (POLG1 genes. RESULTS We identified 3 novel heterozygous POLG1 substitutions in the same family. Autosomal dominant progressive external ophthalmoplegia segregated with 1532G>A in exon 8 and an intronic variant c.2070 + 158G>A in cis. The one patient with parkinsonism had an additional heterozygous substitution in exon 7 in trans (1389G>T). Both coding region mutations were predicted to alter conserved amino acids in the linker region of polymerase γ. None of the substitutions were found in 192 ethnically matched control chromosomes, 108 patients with progressive external ophthalmoplegia, nor 140 cases of sporadic idiopathic Parkinson disease. CONCLUSION Both autosomal dominant progressive external ophthalmoplegia and parkinsonism can because caused by mutations that directly affect the polymerase domain of polymerase γ.Arch Neurol. 2007:64;553-557 --

133. THREE DIALOGUES AND BECKETT'S TRAGIC ETHICS

Abstract

This essay seeks to read the third of the Three Dialoguesand particularly its conclusion in the light of my recent case for an account of Beckett founded on a (much adapted) version of Alain Badiou's. Here I seek to give my argument a new dimension by focusing on Beckett's allusion to "the Sufist alHaqq." I explicate this allusion with reference to its religious and historical context, its place in Beckett's text, and the tone and discursive mode of the Three Dialogues. I conclude by suggesting, in the light of this explication, that Badiou's reading of Beckett needs to be inflected according to what I call a tragic ethics.

135. Converting waste gases from pulp mills into value‐added chemicals

A combined sorption/catalytic process has been discovered that converts the methanol and mercaptans in Kraft pulp mill waste gas condensates into formaldehyde. A pilot plant was operated at Georgia‐Pacific's Brunswick, GA, mill for two years to optimize both sorbents and catalyst. Patents were issued. Applications within Georgia‐Pacific and external licensing opportunities for the process are being investigated. The process gives pulp and paper mills a profitable alternative for converting their waste gases to valuable chemicals, as opposed to incineration or biodegradation in treatment ponds.

Abstract

Although the cytoplasmic domain of the human FcgammaRIa alpha-chain lacks tyrosine-based phosphorylation motifs, it modulates receptor cycling and receptor-specific cytokine production. The cytoplasmic domain of FcgammaRIa is constitutively phosphorylated, and the inhibition of dephosphorylation with okadaic acid, an inhibitor of type 1 and type 2A protein serine/threonine phosphatase, inhibits both receptor-induced activation of the early tyrosine phosphorylation cascade and receptor-specific phagocytosis. To explore the basis for these effects of the cytoplasmic domain of FcgammaRIa, we developed a series of human FcgammaRIa molecular variants, expressed in the murine macrophage cell line P388D1, and demonstrate that serine phosphorylation of the cytoplasmic domain is an important regulatory mechanism. Truncation of the cytoplasmic domain and mutation of the cytoplasmic domain serine residues to alanine abolish the okadaic acid inhibition of phagocytic function. In contrast, the serine mutants did not recapitulate the selective effects of cytoplasmic domain truncation on cytokine production. These results demonstrate for the first time a direct functional role for serine phosphorylation in the alpha-chain of FcgammaRIa and suggest that the cytoplasmic domain of FcgammaRI regulates the different functional capacities of the FcgammaRIa-receptor complex.

142. Single-nucleotide polymorphisms of T cell receptor ζ chain in patients with systemic lupus erythematosus

Abstract

Signaling molecules from the T cell receptor ζ/Fcε receptor γ (TCRζ/FcRγ) family play a critical role in the function of Fcγ receptors and the TCR and are located on human chromosome 1, where lupus susceptibility genes are located. This study was undertaken to investigate the possibility of polymorphisms and/or mutations of TCRζ in systemic lupus erythematosus (SLE). We amplified the whole coding region of TCRζ by reverse transcriptase–polymerase chain reaction (PCR) and directly sequenced the PCR products with a dye primer technique to facilitate heterozygote detection. An alternative splicing form of TCRζ, with a CAG codon (glutamine) inserted at the splice junction of exons 4 and 5, was found both in SLE and in non-SLE subjects. Both splice isoforms of TCRζ occurred in human mixed peripheral blood mononuclear cells, natural killer cells, and Jurkat T cells. In TCRζ, 2 silent and 2 missense mutations were found, but neither coding change occurred in the immunoreceptor tyrosine–activation motif. No unique mutations were found in Caucasian, African American, Hispanic, Chinese, or Japanese SLE patients living in North America. The uncommon and equal occurrence of novel single-nucleotide polymorphisms in both SLE patients and normal subjects makes it improbable that they play important roles in genetic susceptibility to SLE.

143. Calculation of the spectral momentum density for electrons in a topologically disordered model of amorphous graphite

Abstract

The spectral momentum density (SMD) has been computed for electrons in a two-dimensional topologically disordered continuous random network (CRN) model of amorphous graphite, where odd-membered rings have destroyed all remnants of a crystalline lattice. The basis of this approach is an operator, referred to as the network momentum operator (NMO), constructed from the structural relationships of the CRN model: each atom is three-fold coordinated and is a member of only five-, six- or seven-membered rings. This operator generates a relationship between energy and momentum in an analogous way to that in which the translation operator does in crystalline materials. In particular, any Hamiltonian, H, for the disordered system can be separated into a conservative Hamiltonian, H0, which commutes with the NMO, and a potential, V, which only mixes states from different irreducible representations of the NMO. The potential Vhas been shown to be exponentially localized to the odd-membered rings in the CRN model, and therefore the NMO can be thought of as a mapping from topological to substitutional disorder. The SMD is computed using the conservative or effective medium Hamiltonian H0. The result is similar to a spherical average of the crystalline band structure for energy states close to the Δ point in the Brillouin zone of the crystalline material, but is qualitatively different for states closer to the Brillouin zone boundaries. The computed SMD is discussed in relation to results from inelastic electron scattering experiments on samples of evaporated and ion-sputtered amorphous carbon.

146. Prevalence of skin wounds in working donkeys in Bukombe, Tanzania

Abstract

Preventable wounds are a common welfare issue in working donkeys in many countries. In the Bukombe District of Tanzania, there are estimated to be 3000 working donkeys, used primarily to transport loads for direct income generation. For historical reasons, oxen‐yoke carts are used; their design is inappropriate for donkeys and results in serious neck wounds. The project aim was to assess the prevalence and nature of wounds in working donkeys. In November 2018, 148 donkeys owned by 48 owners were examined, and data were collected. The study revealed that one or more wounds were present in 56.1% of the population and yoke‐related, dorsal neck wounds comprised 79.5% of these. These wounds ranged in surface area from 1 cm2to 300 cm2. Clinically, 96.6% of all wound types were superficial and the majority of these (51.1%) were granulating. These data will enable the future evaluation of targeted interventions aimed at reducing the prevalence of these specific wounds.
