Researchers interested in the effects of early experiences of caregiving adversity have employed neuroscientific methods to illuminate whether and how such environmental input impacts on brain development, and whether and how such impacts underpin poor socioemotional outcomes in this population. Evidence is compelling in documenting negative effects on the individual’s neurodevelopment following exposure to adverse or disadvantaged environments such as institutionalization or maltreatment. Neuroimaging research focused specifically on attachment-relevant processing of socioemotional stimuli and attachment outcomes among children looked-after is scarcer, but largely consistent. This review begins by summarizing the key general brain structural and functional alterations associated with caregiving deprivation. Then, neuroscientific evidence that is more directly relevant for understanding these children’s attachment outcomes, both by employing social stimuli and by correlating children’s neural markers with their attachment profiles, is reviewed. Brief interpretations of findings are suggested, and key limitations and gaps in the literature identified.

Seagrass ecosystems, vital as primary producer habitats for maintaining high biodiversity and delivering numerous ecosystem services, face increasing threats from climate change, particularly marine heatwaves. This study introduces a pioneering methodology that integrates Dynamic Bayesian Networks of ecosystem resilience with climate projections, aiming to enhance our understanding of seagrass responses to extreme climate events. We developed cutting-edge metrics for measuring shoot density and biomass in terms of population and site extinction, presented as annual ratios relative to their respective baselines. These metrics include associated uncertainties and projected recovery times. This innovative approach was applied in a case study focusing on Zostera muelleri in Gladstone Harbour, Australia. Utilising five downscaled climate models with a 10 km resolution, our study encompasses a range of Shared Socioeconomic Pathways and emissions trajectories, offering a comprehensive perspective on potential future scenarios. Our findings reveal significant variations in seagrass resilience and recovery times across different climate scenarios, accompanied by varying degrees of uncertainty. For instance, under the optimistic SSP1-1.9 scenario, seagrass demonstrated a capacity for recovery heat stress, with shoot density ratios improving from 0.2 (90% Prediction Interval 0.219, 0.221) in 2041 to 0.5 (90% PI 0.198, 1.076) by 2044. However, this scenario also highlighted potential site extinction risks, with recovery gaps spanning 12 to 18 years. In contrast, the more pessimistic SSP5-8.5 scenario revealed a significant decline in seagrass health, with shoot density ratios decreasing from 0.42 (90% PI 0.226, 0.455) in 2041 to just 0.2 (90% PI 0.211, 0.221) in 2048, and no recovery observed after 2038. This study, through its novel integration of climate models, Dynamic Bayesian Networks, and Monte Carlo methods, offers a groundbreaking approach to ecological forecasting, significantly enhancing seagrass resilience assessment and supporting climate adaptation strategies under changing climatic conditions. This methodology holds great potential for application across various sites and future climate scenarios, offering a versatile tool for integrating Dynamic Bayesian Networks ecosystem models.

Invasive species can generate negative impacts environmentally, socially, and economically. The interplay between human and natural systems renders management a complex problem that must be addressed by decision-makers. Perceptions of invasive species issues varies depending on an individual’s access to information. Although invasive species and their management are often discussed in formal higher education, not all members of the population have access to a formal educational setting. Informal educational experiences may be a mechanism to reach out to community members in a more accessible and perhaps engaging way than traditional higher education classroom experiences. The purpose of this study was to determine the effect of an informal, place-based educational program on perceptions and knowledge of invasive species and their management within the community. Place-based education is a pedagogy connection local places with intimate knowledge of the systems, processes, and outcomes that give it meaning to human and non-human species. First, we organized and administered short walks in two central Texas, USA, parks with conversational lessons and hands-on learning experiences. Second, we administered a follow-up retrospective-reflective survey to measure participants’ knowledge and perceptions of invasive species management and postwalk/lesson changes in them. We also compared between the treatment group of participants versus a control group within the community who did not participate in the informal lessons. Fifty-two people participated in both the educational program walks and testing over the course of 1 year, and 63 people were included in the follow-up retrospective-reflective survey control group. Results indicate a statistically significant positive change in knowledge and perception categories within the treatment group. Post hoc results indicate a positive change in knowledge and perceptions in our sample as a function of treatment group members’ age and income, respectively. The methodology used in this study was simple and inexpensive. Hence, our approach could be easily replicated in other areas to educate community members.

Juvenile justice-involved youth with special education eligibility may have distinct needs from other justice-involved youth that place them at higher risk of re-offending. This study examines the extent to which the comorbidity of risk factors, such as school challenges and mental and emotional health problems, is related to recidivism among probation youth with a diagnosis eligible for special education. Data came from the Washington State Juvenile Court Assessment provided to 4,317 youth adjudicated to probation for at least 3 months. We used independent sample t-tests and chi-square tests to assess the difference in mental health and school problems (e.g., suspension/expulsion history) between those with and without special education needs. Multiple regression models estimated the unique and cumulative role of special education status, mental health, and school problems in future recidivism. In the study sample, 39.6% (n = 1,708) of the youth had diagnoses eligible for special education; over 42% of these youth had two or more qualifying diagnoses. Controlling for demographics, mental health, and self-regulation skills, our findings suggest that probation youth with special education needs, compared to the rest of the probation youth, were more likely to recidivate. School exclusion increased the number of recidivisms significantly more for justice-involved youth with special education needs than those without special education needs. The findings of the study illuminate important factors for continued justice-involvement as well as insights into service and treatment planning for youth serving probation in the community, especially for those who are eligible for special education. Paragangliomas are neuroendocrine tumors of the autonomic nervous system that are variably clinically functional and have a potential for metastasis. Up to 40% occur in the setting of a hereditary syndrome, most commonly due to germline mutations in succinate dehydrogenase (SDHx) genes. Immunohistochemically, paragangliomas are characteristically GATA3-positive and cytokeratin-negative, with loss of SDHB expression in most hereditary cases. In contrast, the rare paragangliomas arising in the cauda equina (CEP) or filum terminale region have been shown to be hormonally silent, clinically indolent, and have variable keratin expression, suggesting these tumors may represent a separate pathologic entity. We retrospectively evaluated 17 CEPs from 11 male and 6 female patients with a median age of 38 years (range 21-82), none with a family history of neuroendocrine neoplasia. Six of the 17 tumors demonstrated prominent gangliocytic or ganglioneuromatous differentiation. By immunohistochemistry, none of the CEPs showed GATA3 positivity or loss of SDHB staining; all 17 CEPs were cytokeratin positive. Genome-wide DNA methylation profiling was performed on 12 of the tumors and compared with publicly available genome-wide DNA methylation data. Clustering analysis showed that CEPs form a distinct epigenetic group, separate from paragangliomas of extraspinal sites, pheochromocytomas, and other neuroendocrine neoplasms. Copy number analysis revealed diploid genomes in the vast majority of CEPs, whereas extraspinal paragangliomas were mostly aneuploid with recurrent trisomy 1q and monosomies of 1p, 3, and 11, none of which were present in the cohort of CEP. Together, these findings indicate that CEPs likely represent a distinct entity. Future genomic studies are needed to further elucidate the molecular pathogenesis of these tumors.

Background: People with severe to profound intellectual disabilities have particular needs for others to help them regulate stress. Such support may be hampered by difficulties in detecting and interpreting expressions of stress. This study aimed to describe and identify stress-related behaviours and sensitive, stress-regulating responses to these behaviours. Method: Video recordings of psychotherapeutic interactions between four clients with severe intellectual disabilities and behavioural problems and two attachment therapists were analysed using a stepwise spiral of analysis approach. Stress-related behaviours were identified with a behaviour rating scale for arousal and valence. Results: Distinct stress-related behaviours were subdivided into stress-specific, non-stress-specific, and client-specific behaviours. Additionally, examples of responsive behaviours to clients' expressions of varying arousal and valence were found. Conclusions: A wide variety of expressions indicative of different levels of arousal were found, with few expressions specific for stress. The descriptions of responsive behaviours might support parents and caregivers in external stress regulation.

Background: During the initial phase of the COVID-19 pandemic, many people with disabilities living in home care facilities could not receive visitors. The use of virtual social contact has been recommended by health authorities. This systematic review examined the scientific evidence of the use and feasibility of information and communication technology (ICT) for social contact by people with intellectual disabilities living in care facilities, and potential effects on well-being. Methods: Five databases were searched using traditional systematic screening and machine-learning supported screening. Findings are presented in a narrative synthesis using thematic analysis. Results: Nine studies were included. We described three themes: means of ICT used for social contact; effects on well-being; and benefits, barriers, and preconditions. Conclusions: Engaging in virtual social contact may be feasible for people with severe to mild intellectual disabilities, but there is little concrete evidence that this can be used as an alternative for in-person contact.

Over the years, microscopic evaluation of hematoxylin-eosin-stained slides from patient samples has been the gold standard for cancer diagnosis and grading of malignancy. The Nottingham Grading System (NGS) is widely [...], * Context.--Breast carcinoma grade, as determined by the Nottingham Grading System (NGS), is an important criterion for determining prognosis. The NGS is based on 3 parameters: tubule formation (TF), nuclear pleomorphism (NP), and mitotic count (MC). The advent of digital pathology and artificial intelligence (AI) have increased interest in virtual microscopy using digital whole slide imaging (WSI) more broadly. Objective.--To compare concordance in breast carcinoma grading between AI and a multi-institutional group of breast pathologists using digital WSI. Design.--We have developed an automated NGS framework using deep learning. Six pathologists and AI independently reviewed a digitally scanned slide from 137 invasive carcinomas and assigned a grade based on scoring of the TF, NP, and MC. Results.--Interobserver agreement for the pathologists and AI for overall grade was moderate ([kappa] = 0.471). Agreement was good ([kappa] = 0.681), moderate ([kappa] = 0.442), and fair ([kappa] = 0.368) for grades 1, 3, and 2, respectively. Observer pair concordance for AI and individual pathologists ranged from fair to good ([kappa] = 0.313-0.606). Perfect agreement was observed in 25 cases (27.4%). Interobserver agreement for the individual components was best for TF ([kappa] = 0.471 each) followed by NP ([kappa] = 0.342) and was worst for MC ([kappa] = 0.233). There were no observed differences in concordance amongst pathologists alone versus pathologists + AI. Conclusions.--Ours is the first study comparing concordance in breast carcinoma grading between a multi-institutional group of pathologists using virtual microscopy to a newly developed WSI AI methodology. Abstract Aerosols have implications to climate and biogeochemical cycles in the global oceans. At sites under indirect influence of dust emitted by the Patagonian semi-desert, a debate exists on the potential fertilization effects of iron enriched aerossol. Considering this subject we conducted measurements of aerosols optical properties using a Microtops II sun photometer to access aerosol size distributions and other intrinsic properties oversea from Atlantic Southern mid-latitudes to Antarctica. Oceanographic cruises were developed between December 2010 to April 2011 and October 2011 to April 2012, in the context of the Brazilian Antarctic Program, and between November 2011 to December 2011. This survey was taken as part of the Global Maritime Aerosol Network (MAN/NASA). Our data of AOD (500 nm) along the South American coast depicts a steady decrease southwards following the decreased latitudinal continental extent. However, the influence of the aerosols blown from Patagonia semi-desert region was clear from latitude 53⁰S to 64⁰S. The predominance of aerosol fine mode was observed in Central Atlantic and close to the Drake Passage. An unexpected aerosol coarse mode predominance was found close to the Antarctic Peninsula. We attribute that to a possible weathering of rock outcrops due to the strong westerly winds in that region.

The Antarctic Peninsula is experiencing one of the highest warming rates globally. In polar regions, macroalgae thrive under extreme environmental conditions, which could worsen because of future climate change scenarios, including increased ultraviolet exposure, extremely low light availability, and fluctuating temperatures, particularly in the intertidal zones. To investigate the potential role of photoprotective and antioxidant mechanisms in response to future increases in sea surface temperatures caused by climate change, we conducted laboratory experiments using three intertidal macroalgae model species: Adenocystis utricularis (Ochrophyta, Phaeophyceae), Pyropia endiviifolia (Rhodophyta, Bangiophyceae), and Monostroma hariotii (Chlorophyta, Ulvophyceae). These algae were collected in Punta Artigas (King George Island, Antarctica) and acclimated at 2°C for 48 h. They were then assessed in laboratory experiments for up to 5 days under two treatments: (1) control conditions at 2°C and (2) elevated tem.perature conditions at 8°C, representing the most negative increment in SSTs estimated by the end of the 21st century. Carbon, nitrogen, pigments (chlorophylls and carotenoids), mycosporine-like amino acids (MAAs), and phenolic compounds were quantified after 3 and 5 days of exposure. For M. hariotii, elevated temperatures led to an increase in the C/N ratio, total antioxidant capacity, and levels of nitrogen, total carotenoids, chlorophyll-a, pigments (chlorophyll-b and violaxanthin), and phenolic compounds. For A. utricularis, elevated temperatures led to elevated C/N ratio and levels of chlorophyll-a and carotenoids (fucoxanthin and β-carotene). For P. endiviifolia, elevated temperatures resulted in elevated levels of carotenoids (lutein and β-carotene), phenolic compounds, and MAAs (porphyra-334, shinorine, and palythine). Thus, our study suggests that increasing water temperatures due to global warming can enhance the photoprotective abilities of three Antarctic intertidal macroalgae (M. hariotii, A. utricularis, and P. endiviifolia), with each species showing specific responses.

The development of new compounds to treat Chagas disease is imperative due to the adverse effects of current drugs and their low efficacy in the chronic phase. This study aims to investigate nitroisoxazole derivatives that produce oxidative stress while modifying the compounds’ lipophilicity, affecting their ability to fight trypanosomes. The results indicate that these compounds are more effective against the epimastigote form of T. cruzi, with a 52 ± 4% trypanocidal effect for compound 9. However, they are less effective against the trypomastigote form, with a 15 ± 3% trypanocidal effect. Additionally, compound 11 interacts with a higher number of amino acid residues within the active site of the enzyme cruzipain. Furthermore, it was also found that the presence of a nitro group allows for the generation of free radicals; likewise, the large size of the compound enables increased interaction with aminoacidic residues in the active site of cruzipain, contributing to trypanocidal activity. This activity depends on the size and lipophilicity of the compounds. The study recommends exploring new compounds based on the nitroisoxazole skeleton, with larger substituents and lipophilicity to enhance their trypanocidal activity.

Systemic sclerosis (SSc) is a clinically heterogeneous autoimmune disease characterized by mutually exclusive autoantibodies directed against distinct nuclear antigens. We examined HLA associations in SSc and its autoantibody subsets in a large, newly recruited African American (AA) cohort and among European Americans (EA). In the AA population, the African ancestry-predominant HLA-DRB1*08:04 and HLA-DRB1*11:02 alleles were associated with overall SSc risk, and the HLA-DRB1*08:04 allele was strongly associated with the severe antifibrillarin (AFA) antibody subset of SSc (odds ratio = 7.4). These African ancestry-predominant alleles may help explain the increased frequency and severity of SSc among the AA population. In the EA population, the HLA-DPB1*13:01 and HLA-DRB1*07:01 alleles were more strongly associated with antitopoisomerase (ATA) and anticentromere antibody-positive subsets of SSc, respectively, than with overall SSc risk, emphasizing the importance of HLA in defining autoantibody subtypes. The association of the HLA-DPB1*13:01 allele with the ATA+ subset of SSc in both AA and EA patients demonstrated a transancestry effect. A direct correlation between SSc prevalence and HLA-DPB1*13:01 allele frequency in multiple populations was observed (r = 0.98, P = 3 × 10-6). Conditional analysis in the autoantibody subsets of SSc revealed several associated amino acid residues, mostly in the peptide-binding groove of the class II HLA molecules. Using HLA α/β allelic heterodimers, we bioinformatically predicted immunodominant peptides of topoisomerase 1, fibrillarin, and centromere protein A and discovered that they are homologous to viral protein sequences from the Mimiviridae and Phycodnaviridae families. Taken together, these data suggest a possible link between HLA alleles, autoantibodies, and environmental triggers in the pathogenesis of SSc.
