Insulin resistance is a critical pathophysiological process in the onset and advancement of type 2 diabetes mellitus. It is well-recognized that alterations in the metabolism of lipids and aberrant fat buildup effectively trigger the development of resistance to insulin. Adjusting one's eating habits and managing weight appropriately are crucial for treating, controlling, and reducing the risk of T2DM because obesity and a lack of physical exercise are the primary factors responsible for the worldwide rise in T2DM. Omega-3 fatty acid is one of the polyunsaturated fatty acids (PUFA) that include long-chain omega-3 fatty acids such as eicosapentaenoic acid and docosahexaenoic acid, commonly found in fish oils. Omega-3 and omega-6 polyunsaturated fatty acids (PUFAs; 3 and 6 PUFAs) are essential for human health because they serve as metabolic precursors of eicosanoids, a class of signaling molecules that are essential for controlling a body's inflammation. Since humans are unable to produce any of the omega-3 or omega-6 PUFAs, they both constitute imperative nutritional ingredients. Long-standing concerns about long-chain omega-3 fatty acids' impact on diabetes management have been supported by experimental investigations that found significant increases in fasting glucose following omega-3 fatty acid supplementation and foods rich in PUFA and omega-3 fatty acid. Cellular explanations to explain the connection between inflammation and IR include mitochondrial dysfunction, endoplasmic reticulum (ER) stress, and oxidative stress. Modifications in the lipid composition of mitochondrial membranes and/or receptor-mediated signaling may be part of the mechanism behind the activation of mitochondrial fusion by fish oil/omega-3 PUFA. E. sinensis, normally harvested in October and November, is an economic aquatic product in China. Pond culture has been widely applied for the production of E. sinensis, wherein a stable food supply for crabs is provided. In order to improve the nutritional quality of E. sinensis products, this study evaluated the effect of the local pond culture on the nutritive profiles of E. sinensis and screened out the best harvest time for the nutrient-rich crabs, thereby guiding the local crab industry to improve its aquaculture mode and harvest strategy. The results indicated that pond culture enhanced the levels of protein, amino acids, and specific organic acid derivatives, and reduced the levels of peptides and polyunsaturated fatty acids (PUFAs). Compared with E. sinensis harvested in October, peptide levels were significantly increased, whereas sugar, phenolic acid, and nucleotide levels were decreased in those harvested in November. Overall, the study revealed that the nutritive profile of the pond-reared E. sinensis was significantly modulated by a high-protein diet, thus lacking the diversity of metabolites. BackgroundObservational studies have indicated an association between polyunsaturated fatty acids (PUFAs) and chronic pain, but the potential causal link remains controversial. Here, we aimed to investigate whether a causal relationship exists between the concentration of circulating PUFAs and chronic pain as well as the direction of this association.MethodsWe collected statistical data from relevant genome-wide association studies to explore the causal link between four PUFAs, along with the ratio of omega-6 fatty acids (FAs) to omega-3 FAs (omega-6:3 ratio), and chronic pain in eight specific body parts. We used the inverse-variance weighting (IVW) method for two-sample Mendelian randomization (MR) analysis and conducted supplementary analyses using four other methods (MR-Egger, weighted median, weighted mode, and simple mode). To verify the robustness of the MR study, we performed multiple sensitivity analyses.ResultsThe results revealed a negative correlation between omega-3 FAs [IVW, OR 95% CI: 0.952 (0.914, 0.991), p = 0.017] and docosahexaenoic acid (DHA) [IVW, OR 95% CI: 0.935 (0.893, 0.978), p = 0.003] with abnormal and pelvic pain. Furthermore, a positive correlation was observed between the omega-6:3 ratio [IVW, OR 95% CI: 1.057 (1.014, 1.101), p = 0.009] with abdominal and pelvic pain. Additionally, we found a negative correlation between omega-3 FAs [IVW, OR 95% CI: 0.947 (0.902, 0.994), p = 0.028] and lower back pain or sciatica. However, no causal relationship was found between the concentration of circulating PUFAs and pain in other body parts, including the face, throat and chest, joints, limbs, lower back, and gynecological parts. The robustness of these MR results was verified through multi-validity and retention method analyses.ConclusionOur analysis suggests that higher circulating concentrations of omega-3 FAs and DHA and a lower omega-6:3 ratio are associated with a reduced risk of abdominal and pelvic pain. Additionally, a higher concentration of circulating omega-3 FAs is linked to a reduced risk of lower back pain and/or sciatica. These findings have major implications for the targeted prevention and treatment of chronic pain using PUFAs.

BACKGROUND/AIM:Cytochrome P450 epoxygenase is a major enzyme involved in the metabolism of ω-3 polyunsaturated fatty acids (PUFAs) to produce biologically active ω-3 epoxy fatty acids (ω-3 epoxides). In general, all epoxy PUFAs including ω-3 epoxides are quickly metabolized/inactivated by soluble epoxide hydrolase (sEH) to form diol products. The aims of this study were to determine the effect and mechanism of fat-1 transgene, and ω-3 PUFA combined with sEH gene knockout or inhibitor on inhibiting pancreatic cancer and the related mechanisms involved. MATERIALS AND METHODS:PK03-mutant KrasG12D murine pancreatic carcinoma cells were inoculated into mouse models including fat-1, sEH-/- and C57BL/6J mice. The mice were fed with AIN-76A diet with or without ω-3 PUFA supplementation or treated with sEH inhibitor. In addition to tumor growth (tumor size and weight), cell proliferation, mutant Kras-mediated signaling, inflammatory reaction and angiogenesis were analyzed immunohisto-chemically and by western blot assay. ω-3 PUFA metabolism, particularly focusing on ω-3 epoxy fatty acids (ω-3 epoxides), was measured using a liquid chromatography with tandem mass spectrometry (LC-MS/MS) approach. RESULTS:Significant decreases of weight and size of the PK03 pancreatic carcinoma were observed in the fat-1 transgenic mice treated with sEH inhibitor compared to those of C57BL/6J control mice fed with AIN-76A diet (weight: 0.28±0.04 g vs. 0.58±0.06 g; size: 187.0±17.5 mm3 vs. 519.3±60.6 mm3). In a separate experiment, sEH-/- mice fed ω-3 PUFA supplement and C57BL/6J mice treated with sEH inhibitor and fed ω-3 PUFA supplement exhibited a significant reduction in the weight and size of the pancreatic carcinoma compared to C57BL/6J control mice (weight: 0.26±.26 g and 0.39±.39 g vs. 0.69±0.11 g, respectively; size: 274.2±36.2 mm3 and 296.4±99.8 mm3 vs. 612.6±117.8 mm3, respectively). Moreover, compared to the pancreatic tumors in C57BL/6J control mice, the tumors in fat-1 transgenic mice treated with sEH inhibitor showed a significant less inflammatory cell infiltrate (62.6±9.2/HPF (high power field) vs. 8.0±1.2/HPF), tumor cell proliferation (48.5±1.7% vs. 16.5±1.6%), and angiogenesis (micro-vessel density (MVD): 35.0±1.0 vs. 11.1±0.5) immunohistochemically, as well as significantly increased caspase-3 labeled apoptosis (0.44±0.06% vs. 0.69±0.06%, respectively). Using western blot approach, significant inhibition of mutant Kras-activated signals including phosphorylated Serine/threonine kinases (cRAF), Mitogen-activated protein kinase kinase (MEK), and extracellular signal-regulated kinase (ERK) were identified in pancreatic carcinoma of fat-1 transgenic mice treated with sEH inhibitor. Eicosanoic acid metabolic profiling of the serum specimens detected a significant increase of the ratios of epoxides to dihydroxy fatty acid (DiHDPE) for docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), and epoxides/dihydroxy octadecenoic acid (DiHOME) for arachidonic acid (ARA) and linoleic acid (LA), as well as a significant increase of epoxy metabolites of DHA, EPA, ARA and LA in fat-1 transgenic mice treated with a sEH inhibitor. CONCLUSION:ω-3 epoxy products from ω-3 PUFA metabolism play a crucial role in inhibiting pancreatic cancer growth, and use of ω-3 PUFAs combined with sEH inhibition is a strategy with high potential for pancreatic cancer treatment and prevention.

In addition to being a source of high-quality protein, many aquatic products are also rich in oil, especially long-chain polyunsaturated fatty acids such as eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), which play an important role in human physiology and health. Aquatic oil has high nutritional value, for example krill oil and microalgae oil have broad prospects for development. However, aquatic oil is easy to be oxidized, and its extraction and processing technology have an important impact on its quality. The extraction and refining of aquatic oil and the enrichment of DHA and EPA were reviewed, the advantages and disadvantages of different methods and the feasibility of applying them to industrial production were also analyzed. The nutrition and safety of aquatic oil, as well as the methods to improve the oxidative stability of aquatic oil were introduced. The development and utilization of aquatic oil resources has become an inevitable trend, and aquatic oil products will be more diversified.

Sesame (Sesanum indicum L.) is one of the primary annual oilseeds grown in Africa and Asia. Sesame seed oil (SSO) is of great economic and human nutrition interest worldwide. Due to its composition in phytochemical antioxidants and profile in unsaturated fatty acids, sesame is used as a biological source of essential fatty acids. It contains bioactive compounds such as lignans (sesamin, sesamol, sesamolin), tocopherols and phytosterols. The oleic/linoleic fatty acids ratio of sesame makes it important for human health. SSO has bioactive compounds that can help prevent certain cardiovascular, metabolized and coronary diseases. The ω-3 and ω-6 fatty acids in SSO are precursors to eicosanoids that regulate the immune system and inflammatory functions. The essential fatty acids contained in this oil are essential for cell construction and highly recommended during the first trimester of pregnancy. The consumption of SSO allows both a decrease in the LDL-cholesterol complex and an increase in the HDL-cholesterol complex. It regulates blood sugar and may have favorable effects on people with liver cancer and those developing fatty liver disease. In this review, the nutritional value, antioxidant properties, and health benefits of SSO have been compiled to provide collective information of nutritional and medical interest.

This systematic review and meta-analysis aimed at summarizing earlier findings on the association of n-6 PUFAs levels in diets or blood with blood pressure. PubMed/Medline, Scopus, and Web of Science were searched for observational studies. Publications with data on the risk of hypertension, or the correlation between n-6 PUFAs or mean values of serum n-6 PUFAs levels in normotensive and hypertensive were included. Twenty-two studies (16 cross-sectional studies, 5 cohorts and one case-control) were eligible. Combining 14 extracted effect sizes showed that higher circulatory/dietary n-6 PUFAs tended to be associated with 10% lower risk of HTN (95% CI: 0.81, 1.00), whereas combining 23 effect sizes illustrated no difference in circulatory/dietary n-6 PUFAs mean levels between normotensive and hypertensive subjects. According to subgroup analysis based on fatty acid types, total n-6 PUFAs (OR = 0.82, 95% CI: 0.70, 0.97) and linoleic acid (OR = 0.56, 95% CI: 0.39, 0.82) were inversely related to the risk of HTN. Circulatory/dietary n-6 PUFAs were correlated neither with systolic nor with diastolic blood pressure. Higher circulatory/dietary n-6 PUFAs tend to be associated with lower odds of HTN. Background: Human heart changes its energetic substrates from lactate and glucose to fatty acids during the neonatal period. Noticing the lack of fatty acids in media for the culture of cardiomyocytes derived from human pluripotent stem cells (hiPS-CM), researchers have supplemented mixtures of fatty acids to hiPS-CM and reported the enhancement in the maturation of hiPS-CM. In our previous studies, we separately supplemented two polyunsaturated fatty acids (PUFAs), docosahexaenoic acid (DHA) or arachidonic acid (AA), to rat fetal cardiomyocytes and found that the supplementations upregulated the expressions of mRNAs for cardiomyocyte differentiation, fatty acid metabolism, and cellular adhesion. The enhancement in cellular contractility was attributed to the improvement in intercellular connection rather than a direct enhancement of the contractile force. Methods: This study reports the successive results of the effects of DHA or AA supplementation on hiPS-CM. In addition to the contractile force and mRNA measurements used in the previous study, we further investigated the effect of different cellular aggregations on the contractile force output by means of finite element analysis, measured glucose and fatty acids metabolites, and assessed cTNT and MLC2v expressions through immunofluorecsence evaluation. Results: It showed that the sole supplementation of albumin-conjugated DHA or AA can be taken up by hiPS-CM without other uptake-enhancing factors, and the supplementations may activate the CD36_ERRγ metabolic pathway. DHA or AA supplementation increased the cellular contractile ratio on collagen gels and AA supplementation stimulated hiPS-CM aggregation to form cellular clusters. The enhancement effect on the hiPS-CM contractile force was modest since the increase in contractile force was not significant. AA supplementation was more effective than DHA supplementation because it significantly upregulated mRNA expressions of P300 and CD36. However, finite element analysis showed that the formation of clusters on a collagen gel attenuated the contractile force exerted by the gel on its surroundings. Conclusion: DHA and AA, as having been supplemented in infant formulas, have no direct and significant enhancement effect on the performance of the hiPS-CM when they were supplemented individually, although they were able to enter the cellular metabolic system. The AA supplementation showed some auxiliary effect on the maturation of hiPS-CM, which is worthy of further investigation under the consideration of membrane composition alteration and remodeling of membrane molecules. Background: In dogs, dietary omega 3 polyunsaturated fatty acids (n-3 PUFA) affect the fatty acid (FA) profile of blood plasma, erythrocyte membrane (EM), and semen, but their correlation has not yet been investigated. Aim: In this study, we evaluated the association between dietary PUFA and their profile in blood plasma, EM, and semen of dogs, with the possibility to predict the semen profile using the values of the three first. Methods: Twelve male dogs received the same standard commercial diet for 4 weeks. The FA profile was analyzed by gas chromatography in paired diet, blood (plasma and EM determinations), and semen samples. Data were analyzed with SAS Proc Corr version 9.4. Pearson's correlation coefficient (significant if p < 0.05) was used to assess the association of dietary FA profiles with those in blood plasma, EM, and semen. Results: There was a positive correlation between dietary eicosapentaenoic acid (EPA) and blood plasma (r = 0.97), EM (r = 0.94) and semen (r = 0.92) EPA, and between dietary docosahexaenoic acid (DHA) and arachidonic acid (ARA) and semen DHA (r = 0.93) and ARA (r = 0.92), respectively. There was a negative correlation between dihomogamma-linolenic acid (DGLA) in the diet and EM DGLA (r = -0.94). Conclusion: The dietary EPA is correlated with blood plasma, EM, and semen EPA concentrations, and dietary DHA and ARA are associated with semen DHA and ARA concentrations in dogs. These findings suggest that dietary EPA, DHA, and ARA concentrations could be helpful to predictive markers for such concentrations in the semen of dogs. [ABSTRACT FROM AUTHOR]

Apolipoprotein D (ApoD) is lipocalin able to bind hydrophobic ligands. The APOD gene is upregulated in a number of pathologies, including Alzheimer's disease, Parkinson's disease, cancer, and hypothyroidism. Upregulation of ApoD is linked to decreased oxidative stress and inflammation in several models, including humans, mice, Drosophila melanogaster and plants. Studies suggest that the mechanism through which ApoD modulates oxidative stress and regulate inflammation is via its capacity to bind arachidonic acid (ARA). This polyunsaturated omega-6 fatty acid can be metabolised to generate large variety of pro-inflammatory mediators. ApoD serves as a sequester, blocking and/or altering arachidonic metabolism. In recent studies of diet-induced obesity, ApoD has been shown to modulate lipid mediators derived from ARA, but also from eicosapentaenoic acid and docosahexaenoic acid in an anti-inflammatory way. High levels of ApoD have also been linked to better metabolic health and inflammatory state in the round ligament of morbidly obese women. Since ApoD expression is upregulated in numerous diseases, it might serve as a therapeutic agent against pathologies aggravated by OS and inflammation such as many obesity comorbidities. Simple Summary: This review discusses the synthesis and significance of fatty acids, glycerophospholipids, and triacylglycerol in glioblastoma. The focus is on all enzymes involved in the synthesis of these lipids, highlighting their roles in the tumorigenesis of glioblastoma. Due to the fact that the role of many of these enzymes in glioblastoma tumorigenesis remains unexplored, we conducted a bioinformatic analysis based on the GEPIA database to indicate their possible functions and significance. Specific properties of certain enzymes were also described to indicate their functions in the tumorigenesis of glioblastoma better. One area of glioblastoma research is the metabolism of tumor cells and detecting differences between tumor and healthy brain tissue metabolism. Here, we review differences in fatty acid metabolism, with a particular focus on the biosynthesis of saturated fatty acids (SFA), monounsaturated fatty acids (MUFA), and polyunsaturated fatty acids (PUFA) by fatty acid synthase (FASN), elongases, and desaturases. We also describe the significance of individual fatty acids in glioblastoma tumorigenesis, as well as the importance of glycerophospholipid and triacylglycerol synthesis in this process. Specifically, we show the significance and function of various isoforms of glycerol-3-phosphate acyltransferases (GPAT), 1-acylglycerol-3-phosphate O-acyltransferases (AGPAT), lipins, as well as enzymes involved in the synthesis of phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylserine (PS), phosphatidylinositol (PI), and cardiolipin (CL). This review also highlights the involvement of diacylglycerol O-acyltransferase (DGAT) in triacylglycerol biosynthesis. Due to significant gaps in knowledge, the GEPIA database was utilized to demonstrate the significance of individual enzymes in glioblastoma tumorigenesis. Finally, we also describe the significance of lipid droplets in glioblastoma and the impact of fatty acid synthesis, particularly docosahexaenoic acid (DHA), on cell membrane fluidity and signal transduction from the epidermal growth factor receptor (EGFR). In the United States, pregnant women have low concentrations of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), which are essential for fetal development. Although maternal blood provides accurate polyunsaturated fatty acid (PUFA) concentrations, venipuncture is expensive and not always accessible. PUFA-containing foods consumption, both omega-3 ad omega-6 is supposed to reflect in the status (plasma, RBC, adipose tissue) of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA). De novo synthesis of DHA and EPA during pregnancy is supposed to be higher compared to pre and/or post-pregnancy periods. Thus, this study aimed to determine the association between maternal self-reported dietary intake of foods high in DHA and EPA, along with vegetable oils as a source of omega-6 fatty acids, with maternal blood DHA and EPA concentrations. Pregnant women (13–16 weeks gestation) were recruited and asked to complete a food-frequency questionnaire (FFQ) and blood draw at enrollment and 36 weeks. Circulating concentrations of DHA and EPA were quantified and change scores were calculated. Correlations were done to determine associations between FFQ results and EPA/DHA maternal blood concentrations. Regression analyses were run to examine significant predictors of the main outcomes. Overall, PUFA-food consumption and RBC's DHA levels decreased from early to late pregnancy; self-reported PUFA-rich food consumption positively correlated with DHA and EPA levels. DHA concentration was predicted by self-reported PUFA-rich oils (sunflower/soy/corn/olive) consumption, but EPA concentration was predicted by maternal BMI. These findings suggest that EPA and DHA consumption decreased across pregnancy and the FFQ can be utilized as an effective method for estimating PUFA blood concentration during pregnancy. Peroxisomes are essential organelles involved in lipid metabolisms including plasmalogen biosynthesis and β‐oxidation of very long‐chain fatty acids. Peroxisomes proliferate by the growth and division of pre‐existing peroxisomes. The peroxisomal membrane is elongated by Pex11β and then divided by the dynamin‐like GTPase, DLP1 (also known as DRP1 encoded by DNM1L gene), which also functions as a fission factor for mitochondria. Nucleoside diphosphate kinase 3 (NME3) localized in both peroxisomes and mitochondria generates GTP for DLP1 activity. Deficiencies of either of these factors induce abnormal morphology of peroxisomes and/or mitochondria, and are associated with central nervous system dysfunction. To investigate whether the impaired division of peroxisomes affects lipid metabolisms, we assessed the phospholipid composition of cells lacking each of the different division factors. In fibroblasts from the patients deficient in DLP1, NME3, or Pex11β, docosahexaenoic acid (DHA, C22:6)‐containing phospholipids were found to be decreased. Conversely, the levels of several fatty acids such as arachidonic acid (AA, C20:4) and oleic acid (C18:1) were elevated. Mouse embryonic fibroblasts from Drp1‐ and Pex11β‐knockout mice also showed a decrease in the levels of phospholipids containing DHA and AA. Background: Arachidonic acid (ARA) is associated with colorectal cancer (CRC), a major public health concern. However, it is uncertain if ARA contributes to the development of colorectal polyps which are pre-malignant precursors of CRC. Objective: The study aimed to investigate the association between lifelong exposure to elevated ARA and colorectal polyp incidence. Methods: Summary-level GWAS data from European, Singaporean, and Chinese cohorts (n = 10,171) identified 4 single-nucleotide polymorphisms (SNPs) associated with blood ARA levels (p < 5 × 10−8). After pruning, 1 SNP was retained (rs174547; p = 3.0 × 10−971) for 2-stage Mendelian randomization. Results: No association between ARA and colorectal polyp incidence was observed (OR = 1.00; 95% CI: 0.99, 1.00; p value = 0.50) within the UK Biobank (1,391 cases; 462,933 total). Conclusions: Blood levels of ARA do not associate with colorectal polyp incidence in a general healthy population. Although not providing direct evidence, this work supports the contention that downstream lipid mediators, such as PGE2 rather than ARA itself, are key for polyp formation during early-stage colorectal carcinogenesis.

Spring Viremia of Carp Virus (SVCV) is a highly pathogenic pathogen that infects carp fish, and SVCV infection frequently causes a severe inflammatory response in the affected fish. The viral infection is extremely lethal in carp larvae, often leading to heavy economic losses. Cholic acid (CA) is a primary bile acid synthesized directly from cholesterol in the liver and has been found to have an inhibitory effect on viral infections. To substantiate this antiviral potential, 500 μM CA was tested against SVCV replication in vitro and showed a significant inhibitory effect on viral replication. Further experimental tests demonstrated that CA exerted antiviral effects during its replication phase but not at viral adsorption and entry stages. In vivo experiments demonstrated that CA increased the survival rate of SVCV-infected zebrafish by 40 %, reduced viral load in tissues and reduced histopathological lesions. Comparison of the liver transcriptome of zebrafish between CA treatment and control (NC) groups revealed that CA treatment was able to promote the synthesis of polyunsaturated fatty acids, with the most significant changes in docosahexaenoic acid (DHA). Inhibition assay on fatty acid desaturase 2 (Fads2), the first rate-limiting enzyme for the synthesis of polyunsaturated fatty acid, showed that the antiviral effect of CA on SVCV disappeared. These new findings suggest that CA may exert its antiviral effects by regulating the synthesis of polyunsaturated fatty acid. In addition, this study also verified that DHA has antiviral activity against SVCV. Cellular residues after lipid extraction are the primary waste by-products of microbial polyunsaturated fatty acid production. To mitigate the discharge of cellular residues and reduce the cost of DHA and ARA production, this study examined the utilization of enzymatically hydrolyzed Aurantiochytrium cellular residues (ACRs) and Mortierella alpina cellular residues (MCRs) as nitrogen sources. Results demonstrated that ACRs and MCRs could partially substitute yeast extract (YE) without adverse effects on DHA and ARA fermentation. Moreover, the implementation of a new fermentation medium incorporating ACRs, MCRs, and YE as mixed nitrogen sources resulted in DHA and ARA yields of 17.78 and 5.77 g/L, respectively. These values represented increases of 10.37% and 9.28% compared to traditional cultural methods, while simultaneously reducing the usage of YE by 80% and 60%, respectively. Therefore, the strategy of recycling waste cellular residues presents a novel approach for reducing the costs and environmental impact associated with microbial fermentations.

ABSTRACT: Although “zero waste” valorization concepts are gaining increasing attention, colostrum, a byproduct of milk production, remains underused due to technological challenges. Information about the fat fraction and the size of fat globules is needed to address these challenges, but such information is currently lacking. This study aimed to fill this gap in the knowledge by measuring the size distribution of bovine colostrum fat globules (CFG) and analyzing its relationships with postpartum milkings, parity, and fatty acids (FA) profile. Four sequential postpartum colostrum samples were collected from 44 cows and analyzed for the abovementioned parameters. The results indicated that CFG size increases almost twice during postpartum milkings (from ∼5 to ∼10 µm), whereas lactation has little, if any, effect on CFG size. The FA profile analyses showed that the content of most FA in the fourth postpartum milking was different from the previous milkings. The correlation analyses between CFG size and FA profile also demonstrated that the fourth milking was clearly distinguishable from the first 3 postpartum milkings. For example, the saturated FA content from the first 3 milkings had a positive correlation with smaller CFG (and a negative correlation with larger CFG), whereas the fourth milking demonstrated no correlations. Based on these CFG size and FA profile analyses, the results of this study suggest that the first 3 postpartum milkings can be considered as colostrum, whereas the fourth milking represents transition milk. Information about CFG size distribution enables modification of the FA profile of colostrum products and the ability to create better valorization technologies for colostrum-based food and feed supplements.

Abstract Background To evaluate the association and dose–response relationship between polyunsaturated fatty acid (PUFA) intake and incidence of type 2 diabetes (T2D) in adults. Methods PubMed, Embase, Cochrane Library, and Web of Science databases were searched for cohort studies that examined the association between PUFA and T2D incidence published up to September 6, 2021. Relative risk (RR) or hazard ratio (HR) was used as the effect indicator, each effect size was expressed by 95% confidence interval (CI). The presence of heterogeneity of effect size between studies was assessed by the Q-test and I2 statistics. If I2 ≥ 50%, the random-effects model was applied, otherwise the fixed effects model was used. Sensitivity analysis was performed for all models. Potential publication bias was assessed. We conducted linear and nonlinear dose–response meta-analyses, calculated summary relative risk (SRR). Results Twenty-five articles were selected including 54,000 patients in this study. Our estimates observed no linear associations between total PUFA and the incidence of T2D. The purpose of this study was to assess long chain polyunsaturated fatty acid (LCPUFA) status in relation to socio-behavioral outcomes in children with Attention Deficit/Hyperactivity Disorder (AD/HD). In a case-control design, plasma phospholipid fatty acid content was assessed in children aged 5–12 years with AD/HD and in typically functioning children. Dietary intakes of LCPUFAs arachidonic acid (AA; 20:4n6) and docosahexaenoic acid (DHA; 22:6n3) were quantified using a four-day food record, polymorphisms were determined in FADS1 and FADS2, and socio-behavioral outcomes were assessed using the Conners 3 Parent Rating Scales in a cross section of children with AD/HD. Compared to typically functioning children, plasma AA and DHA were 40% lower in children with AD/HD. Median intake of AA, but not DHA, was higher in children with AD/HD compared to typically functioning children. Polymorphisms in FADS1 (rs174546) and FADS2 (174575) were associated with higher plasma linoleic acid (LA; 18:2n6) level. Plasma DHA level was inversely associated with inattention score. Despite having an elevated intake of AA, children diagnosed with AD/HD have a reduction in plasma AA level which may be due in part to polymorphisms in the fatty acid desaturase (FADS) gene cluster or increased conversion to AA-derived metabolites. Increasing intake of DHA may ameliorate symptoms of inattention in AD/HD.

Food aversion and food avoidance are significant challenges to overcome for patients with eating disorder such as anorexia nervosa. The epoxide hydrolase 2 gene (EPXH2) has been uncovered as a novel anorexia nervosa risk gene. We have also discovered EPHX2-associated eicosanoids derived from polyunsaturated fatty acids to be aberrant in patients with anorexia nervosa, suggesting that genetically moderated lipid metabolism may be an underlying factor in AN pathogenesis. The data presented in this article are related to the research article entitled Personalized polyunsaturated fatty acids as a potential adjunctive treatment for anorexia nervosa [1]. In this data article, we provide both fasting and non-fasting (postprandial) concentration of eicosanoids in remitted patients with eating disorder and healthy controls. The data provides information on quantitative bioactive lipid mediators in fasting as well as non-fasting states, allowing inference of lipid metabolism associated with food consumption. The data set is made available to enable critical or extended analyzes.

The cyclooxygenases COX-1 and COX-2 oxygenate arachidonic acid (AA) to prostaglandin H2 (PGH2). COX-2 also oxygenates the endocannabinoids 2-arachidonoylglycerol (2-AG) and arachidonoylethanolamide (AEA) to the corresponding PGH2 analogs. Both enzymes are targets of nonsteroidal anti-inflammatory drugs (NSAIDs), but NSAID-mediated COX inhibition is associated with gastrointestinal toxicity. One potential strategy to counter this toxicity is to also inhibit fatty acid amide hydrolase (FAAH), which hydrolyzes bioactive fatty acid ethanolamides (FAEs) into fatty acids and ethanolamine. Here, we investigated the mechanism of COX inhibition by ARN2508, an NSAID that inhibits both COXs and FAAH with high potency, target selectivity, and decreased gastrointestinal toxicity in mouse models, presumably due to its ability to increase levels of FAEs. A 2.27-Å-resolution X-ray crystal structure of the COX-2·(S)-ARN2508 complex reveals that ARN2508 adopts a binding pose similar to that of its parent NSAID flurbiprofen. However, ARN2508's alkyl tail is inserted deep into the top channel, an active site region not exploited by any previously reported NSAID. As for flurbiprofen, ARN2508's potency is highly dependent on the configuration of the α-methyl group. Thus, (S)-ARN2508 is more potent than (R)-ARN2508 for inhibition of AA oxygenation by both COXs and 2-AG oxygenation by COX-2. Also, similarly to (R)-flurbiprofen, (R)-ARN2508 exhibits substrate selectivity for inhibition of 2-AG oxygenation. Site-directed mutagenesis confirms the importance of insertion of the alkyl tail into the top channel for (S)-ARN2508's potency and suggests a role for Ser-530 as a determinant of the inhibitor's slow rate of inhibition compared with that of (S)-flurbiprofen.

ObjectiveRegulation of fatty acid (FA) metabolism is central to adipocyte dysfunction during diet-induced obesity (DIO). Long-chain acyl-CoA synthetase-4 (ACSL4) has been hypothesized to modulate the metabolic fates of polyunsaturated FA (PUFA), including arachidonic acid (AA), but the in vivo actions of ACSL4 are unknown. The purpose of our studies was to determine the in vivo role of adipocyte ACSL4 in regulating obesity-associated adipocyte dysfunction.MethodsWe developed a novel mouse model with adipocyte-specific ablation of ACSL4 (Ad-KO) using loxP Cre recombinase technology. Metabolic phenotyping of Ad-KO mice relative to their floxed littermates (ACSL4floxed) was performed, including body weight and body composition over time; insulin and glucose tolerance tests; and energy expenditure, activity, and food intake in metabolic cages. Adipocytes were isolated for ex vivo adipocyte oxygen consumption by Clark electrode and lipidomics analysis. In vitro adipocyte analysis including oxygen consumption by Seahorse and real-time PCR analysis were performed to confirm our in vivo findings.ResultsAd-KO mice were protected against DIO, adipocyte death, and metabolic dysfunction. Adipocytes from Ad-KO mice fed high-fat diet (HFD) had reduced incorporation of AA into phospholipids (PL), free AA, and levels of the AA lipid peroxidation product 4-hydroxynonenal (4-HNE). Additionally, adipocytes from Ad-KO mice fed HFD had reduced p53 activation and increased adipocyte oxygen consumption (OCR), which we demonstrated are direct effects of 4-HNE on adipocytes in vitro.ConclusionThese studies are the first to elucidate ACSL4's in vivo actions to regulate the incorporation of AA into PL and downstream effects on DIO-associated adipocyte dysfunction. By reducing the incorporation of AA into PL and free fatty acid pools in adipocytes, Ad-KO mice were significantly protected against HFD-induced increases in adipose and liver fat accumulation, adipocyte death, gonadal white adipose tissue (gWAT) inflammation, and insulin resistance (IR). Additionally, deficiency of adipocyte ACSL4 expression in mice fed a HFD resulted in increased gWAT adipocyte OCR and whole body energy expenditure (EE).

Spinocerebellar ataxias (SCAs) are autosomal dominant diseases characterized by cerebellar atrophy and ataxia. The SCA subtype SCA34 is caused by specific mutations in the gene ELOVL4, which encodes a fatty acid (FA) elongase that synthesizes ultra-long-chain (ULC; ≥C26) FAs. However, the pathogenesis and molecular mechanism that confers dominant inheritance remains unknown. Here, a cell-based assay demonstrated that each of the five known SCA34 mutants produced shorter ULC polyunsaturated FA-containing phosphatidylcholines (ULC-PCs) than wild-type protein, in the following order of severity: Q180P and T233M > W246G > I171T and L168F. Next, we generated knock-in mouse embryonic stem cells that contained heterozygous Q180P, heterozygous W246G, or homozygous W246G mutations. Neuronal differentiation-dependent production of ULC-PCs was reduced in heterozygous Q180P and homozygous W246G cells relative to control cells, and we observed shortening of the FA moiety in all mutant cells. This FA shortening was consistent with our prediction that amino acid residues substituted by SCA34 mutations are located in the transmembrane helices that interact with the ω-end region of the FA moiety of the substrate acyl-CoA. Hence, reduced levels and shortening of ULC-PCs in neurons may cause SCA34, and incomplete elongation of ULC polyunsaturated acyl-CoAs by mutated ELOVL4 may induce dominant inheritance. The development of colorectal cancer typically involves the accumulated influences of genetic alterations, medical issues, lifestyle, and diet. Dietary fatty acids appear to affect the tumorigenesis and progression of colorectal cancer. Despite conflicting results, the current consensus on the effects of very long-chain polyunsaturated fatty acids on colorectal cancer is that low levels of eicosapentaenoic acid and docosahexaenoic acid, and high levels of arachidonic acid are associated with an increased risk of colorectal cancer. Altered levels of arachidonic acid in membrane phospholipids can change the levels of prostaglandin E2, which affect the biological activities of cancer cells in multiple stages. Arachidonic acid and other very long-chain polyunsaturated fatty acids can affect tumorigenesis in prostaglandin E2-independent manners as well, including stabilization of β-catenine, ferroptosis, ROS generation, regulation of transcription factors, and de novo lipogenesis. Recent studies have revealed an association between the activities of enzymes synthesizing very long-chain polyunsaturated fatty acids and tumorigenesis and cancer progression, although the mechanisms are still unknown. In this study, PUFA effects on tumorigenesis, the endogenous very long-chain polyunsaturated fatty acid synthesis pathway, metabolites of arachidonic acid and their effects on tumorigenesis and progression of CRC, and current knowledge that supports the association of the enzymes involved in the polyunsaturated fatty acid synthesis pathway with colorectal cancer tumorigenesis and progression are reviewed. Singlet oxygen (¹O2) is a non-radical reactive oxygen species (ROS), a photosynthetic byproduct in plants. Like other chloroplastic radicals, such as superoxide and hydroxyl radicals, ¹O2 can damage biological molecules including nucleic acids, lipids, and proteins, exacerbating photoinhibition and oxidative stress. Since the photosystem II reaction center (PSII RC) is a primary place of ¹O2 production, PSII damage, quality control, and reassembly occur as light-driven default processes. In addition, the chloroplast has evolved systems to quench excess light energy and to directly detoxify ¹O2 as counteractive measures. However, upon the first publication of a research article elucidating the genetic basis of ¹O2-triggered stress responses by Klaus Apel's group in 2004, the classical view of ¹O2 as a toxic agent has dramatically changed. Since then, many scientists started the incredible journey of ¹O2-signaling research with more discoveries of ¹O2-overproducing mutants under certain conditions. This chapter summarizes the current knowledge of the biogenesis of photosynthetic and non-photosynthetic ¹O2, PSII damage and repair cycle, and the ¹O2-dependent oxidative modification of biomolecules and related retrograde signaling pathways in plants. In particular, we highlight recent progress in ¹O2 signaling pathways mediated by two spatially separated chloroplast ¹O2 sensors, β-carotene and EXECUTER proteins, as well as possible other branches of the ¹O2 signaling network, providing insights into the importance of ¹O2 in plant stress responses, such as acclimation and programmed cell death. Introduction: Apelin could be one of the last protective defenses before developing obesity-related disorders, including insulin resistance, type 2 diabetes, and hypertension, which can be modified by dietary intake. The present study investigated the association of habitual intake of total fatty acids (TFAs), saturated-, monounsaturated-, polyunsaturated FAs, n-3, and n-6 FAs with Apelin expression in visceral adipose tissue (VAT) and subcutaneous adipose tissue (SAT). Methods: We obtained VAT and SAT from 168 participants (64 nonobese and 104 obese) who had undergone open abdominal surgery. Dietary intake information was gathered with a valid and reliable food frequency questionnaire. The mRNA expression of the Apelin gene was analyzed by realtime PCR. Results: Apelin serum levels were increased in the obese subjects compared to the nonobese group (p = 0.016). The SAT and VAT Apelin mRNA levels were significantly elevated in the obese participants compared to the nonobese ones (p < 0.05). Based on BMI status, only obese subjects indicated a positive association between SAT and VAT Apelin expression and TFA intake (p < 0.001). However, this association was observed between SAT and VAT Apelin gene expression and polyunsaturated fatty acid (PUFA) and n-3 FA intakes in both obese and nonobese groups (p < 0.05). Conclusion: High Apelin gene expression was associated with TFA intake in obese subjects in both fat tissues. However, habitual intake of PUFA and n-3 FA was associated with Apelin gene expression in obese and nonobese individuals. It has been reported that dietary n-3 polyunsaturated fatty acids (n-3 PUFAs) exert therapeutic potential for the preservation of functional β-cell mass. However, the effect of dietary n-3 PUFA deficiency on pancreatic injury and whether the supplementation of n-3 PUFA could prevent the development of pancreatic injury are still not clear. In the present study, an n-3 PUFA deficiency mouse model was established by feeding them with n-3 PUFA deficiency diets for 30 days. Results showed that n-3 PUFA deficiency aggravated streptozotocin (STZ)-induced pancreas injury by reducing the insulin level by 18.21% and the HOMA β-cell indices by 31.13% and the area of islet by 52.58% compared with the STZ group. Moreover, pre-intervention with DHA and EPA for 15 days could alleviate STZ-induced pancreas damage by increasing the insulin level by 55.26% and 44.33%, the HOMA β-cell indices by 118.81% and 157.26% and reversed the area of islet by 196.75% and 205.57% compared to the n-3 Def group, and the effects were significant compared to γ-linolenic acid (GLA) and alpha-linolenic acid (ALA) treatment. The possible underlying mechanisms indicated that EPA and DHA significantly reduced the ration of n-6 PUFA to n-3 PUFA and then inhibited oxidative stress, inflammation and islet β-cell apoptosis levels in pancreas tissue. The results might provide insights into the prevention and alleviation of pancreas injury by dietary intervention with PUFAs and provide a theoretical basis for their application in functional foods. Docosahexaenoic acid (DHA) is an important omega-3 polyunsaturated fatty acid (PUFA) that plays a critical physiological role in human health. Schizochytrium sp. is considered an excellent strain for DHA production, but the synthesis of DHA is limited by the availability of nicotinamide adenine dinucleotide phosphate (NADPH). In this study, the endogenous glucose-6-phosphate dehydrogenase (G6PD) gene was overexpressed in Schizochytrium sp. H016. Results demonstrated that G6PD overexpression increased the availability of NADPH, which ultimately altered the fatty acid profile, resulting in a 1.91-fold increase in DHA yield (8.81 g/L) and increased carbon flux by shifting it from carbohydrate and protein synthesis to lipid production. Thus, G6PD played a vital role in primary metabolism. In addition, G6PD significantly increased DHA content and lipid accumulation by 31.47% and 40.29%, respectively. The fed-batch fermentation experiment results showed that DHA production reached 17.01 g/L in the overexpressing G6PD strain. These results elucidated the beneficial effects of NADPH on the synthesis of PUFA in Schizochytrium sp. H016, which may be a potential target for metabolic engineering. This study considered the change in survival rate, body weight and fatty acid composition of mealworm during refrigerated storage. The mealworms were kept in refrigerator (temperature : 4.5 ± 0.27°C, humidity : 22.1 ± 0.64%) for 0, 10, 20, 30 and 45 days. The mealworms were 200 individuals per group and the three groups were prepared for each storage period. We measured the survival rate, body weight and fatty acid composition of mealworm for each storage period. Survival rate and body weight decreased with increasing refrigerated storage time. The proportion of unsaturated fatty acids and polyunsaturated fatty acids increased with increasing refrigerated storage time. Therefore, we need to consider that survival rate, body weight and fatty acid composition change with refrigerated storage time in case the mealworm is used as live feed. [ABSTRACT FROM AUTHOR]

The objective of this study was to determine the multivariate relationship among linoleic acid, alpha-linolenic acid, and their main rumen biohydrogenation (BH) intermediates and products in bovine rumen fluid using canonical correlation analysis (CCA). A dataset consisting of 1177 observations generated by 107 in vitro rumen incubation systems of pure and mixed linoleic acid (18:2-c9, c12) and alpha-linolenic acid (18:3-c9, c12, c15) was gathered. Two canonical variates were defined: A: composed of the nine main BH intermediates and products (18:2-c9, t11; 18:2-t11, c15; 18:1-t11; 18:1-t9; 18:1-t6; 18:1-c11; 18:1-c6; 18:1-c9; 18:0) of 18:2-c9, c12 and 18:3-c9, c12, c15 and B: composed of 18:2-c9, c12 and 18:3-c9, c12, c15. Two canonical functions between A and B with significant canonical correlations (R1=0.990 and R2=0.738; p <0.01) were obtained. However, only the first function was selected for CCA. Exploration of canonical loadings for first function, revealed the following quantitative significance (absolute value) order for fatty acids (FA) within their respective canonical variates: A: 18:0(0.958)>18:1-t9(0.837)>18:1-c11(0.835)>18:1-c6(0.824)>18:1-t11(0.747)>18:1-c9(0.738)>18:1-t6(0.415)>18:2-t11, c15(0.387)> 18:2-c9, t11(0.239); B: 18:2-c9, c12(0.667)>18:3-c9, c12, c15(0.488). The CCA showed that 18:2-c9, c12 has a greater contribution than that of 18:3-c9, c12, c15 on the production of the aforementioned BH intermediates, in which 18:0, as well as the groups of 18:1 cis and trans-FA were mainly affected. The primary production of fjords across the Arctic and Subarctic is undergoing significant transformations due to the climatically driven retreat of glaciers and ice sheets. However, the implications of these changes for upper trophic levels remain largely unknown. In this study, we employ both bulk and compound-specific stable isotope analyses to investigate how shifts at the base of fjord food webs impact the carbon and energy sources of consumers. Focusing on two rapidly changing fjords in Southern Greenland, we used the migratory Arctic char as an indicator species, sampling populations along environmental gradients within the fjords, building upon the assumption that char populations feed primarily close to their natal stream, thereby integrating a dietary gradient. Our analysis of bulk stable isotopes in Arctic char tissue confirmed this premise, revealing a consistent change in resource use from the outer to the inner fjord, which nonetheless served as preferred feeding grounds. Essential amino acid analysis further indicated shifts in carbon and nitrogen sources, consistent with changes in nutrient use near glacier inputs characterized by low turbidity and high iron levels. Notably, these changes in the source of primary production were associated with shifts in trophic positions and the transfer of polyunsaturated fatty acids, with Arctic char in glacier-influenced inner fjords feeding at lower trophic level (size-corrected) and accumulating higher levels of high-quality docosahexaenoic acid (DHA). These findings highlight the usefulness of new analytical tools in revealing that glacial retreat can substantially alter food web dynamics, enhancing both carbon flow and the nutritional quality of fish in fjord ecosystems. The two Southern Greenland fjords studied could represent the future of other fjords, where retreating glaciers become land-terminating and glacial inputs decrease. Based on the observation that urea, water, and ethyl esters (EE) can form gypsum-like mixtures, this study explored the feasibility of employing water as a solvent for urea in the urea complexation method to enrich n-3 polyunsaturated fatty acids with docosahexaenoic acid (DHA)-containing ethyl esters (DHA- EE) from Crypthecodinium cohnii as the material. Under the conditions of a urea/DHA-EE ratio of 3, a water/DHA-EE ratio of 0.75, a mixing temperature of 65℃, and a cooling temperature of 20℃, a concentrate containing over 90% DHA was achieved. This demonstrated that using water as a solvent for urea, instead of polar organic solvents, is feasible and efficient for enriching DHA in urea complexation process.

Abstract Background The relationship of consumption of dietary fat and fatty acids with esophageal squamous cell carcinoma (ESCC) risk remains unclear. This study aimed to explore the relationship of dietary fat and fatty acids intake with ESCC risk. Methods This case-control study included 879 incident cases and 892 community-based controls recruited from Southwest China. A food frequency questionnaire was adopted to collect information about dietary information, and intake of fat, saturated fatty acid (SFA), monounsaturated fatty acid (MUFA), polyunsaturated fatty acid (PUFA), and total fatty acid (TFA) was calculated. Odds ratios (ORs) with 95% confidence intervals (95% CIs) were estimated using the logistic regression model. Results When comparing the highest with lowest intake quintiles, MUFA (OR: 0.33, 95% CI: 0.21–0.51), PUFA (OR: 0.32, 95% CI: 0.20–0.51), and TFA (OR: 0.44, 95% CI: 0.28–0.70) were related to a reduced risk of ESCC after adjusting for confounders; for non-drinkers rather than drinkers, the intake of SFA was significantly related to a 61% (OR: 0.39, 95% CI: 0.19–0.81) reduced risk of ESCC when comparing the highest with the lowest intake quintiles. Dietary fat was not related to the risk of ESCC. Conclusions This study suggested that the more intake of MUFA and PUFA, the lower risk of ESCC, whereas the protective effect of TFA was only observed among non-drinkers. Strategic nutritional programs should consider food rich in unsaturated fatty acids to mitigate the occurrence of ESCC.

To assess the response of two safflower genotypes to auxin foliar application under late-season drought stress using a factorial split-plot experiment with the randomised complete block design, a two-year experiment (2016-2017 and 2017-2018) was conducted in Iran. The watering regime as specified in two levels including, regular irrigation and drought stress in main plots and two safflower genotypes and auxin foliar application in two levels including non-foliar application (control) and auxin foliar application at a concentration of 30 ppm were categorised in subplots. Drought stress at the seed filling stage reduced the safflower seed and oil yield. Moreover, a significant drought-induced decrease in linoleic acid, palmitic acid and behenic acid content, as well as an increase in oleic acids, was observed in two safflower genotypes. Among two safflower genotypes, the early maturing genotype less affected by drought and Goldasht had higher seed yield than the Padideh by 814 kg/ha, respectively. Auxin foliar application alleviates the adverse effects of drought, which led to an 18% increase in seed yield and components. However, the auxin application had no effect on the safflower seed oil content but palmitic acid content was affected by auxin foliar application. Generally, safflower along with auxin application can be recommended to develop safflower cultivation in semi-arid areas.

Background: Polyunsaturated fatty acids (PUFAs) remain part of the diet and are essential for growth and development. Furthermore, omega − 3 fatty acids boost various cardiovascular disease risk factors as well as lower blood pressure and cholesterol levels. The effects of PUFAs on glycemia in type 2 diabetes patients are unclear. In the present study, the anti-diabetic and anti-hypertensive potential of eicosapentenoic acid (EPA) and docosahexaenoic acid (DHA)—two polyunsaturated fatty acids—were examined. Material and Methods: Using 3T3-L1 pre-adipocyte cells fed with PUFAs, the antioxidant capacity of EPA and DHA was assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay/test. The DPPH activity of EPA and DHA was 49.72 and 50.51%, respectively, indicating a reduction in oxidative stress. The number, size, and total lipid content of adipocytes in adipose tissue were used to study the anti-diabetic effect of EPA and DHA. Both PUFAs were revealed to have a much lower capacity for cell lysis of 3T3-L1 pre-adipocytes when compared to propylene glycol monomethyl ether acetate (PMA). In 3T3-L1 pre-adipocyte cells that had been treated with EPA and DHA, the gene expression profiles for ATP synthase 6 were examined. Results: The results demonstrated a similar trend of reducing total lipid content in 3T3-L1 pre-adipocyte cells treated with EPA and DHA. The amount of cell lysis was then examined for 3T3-L1 pre-adipocyte cells exposed to DHA and EPA, and the results showed 38.45% and 41.26%, respectively. In the 3T3-L1 pre-adipocyte cells, treatment with PUFAs, EPA, and DHA dramatically lowered total lipid content after 48 hours. The study also revealed that exposing 3T3-L1 pre-adipocyte cells to EPA at 90 g/ml for 48 hours reduced the total lipid content by a significant amount. Conclusion: According to the findings, EPA and DHA therapy reversed oxidative stress in mitochondria and upregulated the ATP synthase 6 gene. This discovery shows how EPA and DHA have anti-diabetic and hypertension properties.

Trans fatty acids (C18) can increase blood cholesterol levels and the risk of cardiovascular disease, which cause adverse effects on the human body. EPA and DHA are important long-chain polyunsaturated fatty acids with a variety of biological activities and they are of great significance for maintaining human health. EPA and DHA will undergo trans-isomerization during the refining process. Compared with cis EPA and cis DHA, the physiological functions of the trans EPA and trans DHA have undergone significant changes. The sources, detection technology and physiological function of trans EPA and trans DHA were reviewed in order to provide a scientific basis and theoretical basis for the comprehensive utilization and development of EPA and DHA.

Abstract Background Triacylglycerol (TAG) is an important storage lipid in organisms, depending on the degree of unsaturation of fatty acid molecules attached to glycerol; it is usually used as the feedstock for nutrition or biodiesel. However, the mechanism of assembly of saturated fatty acids (SFAs) or polyunsaturated fatty acids (PUFAs) into TAGs remains unclear for industrial oleaginous microorganism. Results Diacylglycerol acyltransferase (DGAT) is a key enzyme for TAG synthesis. Hence, ex vivo (in yeast), and in vivo functions of four DGAT2s (DGAT2A, DGAT2B, DGAT2C, and DGAT2D) in industrial oleaginous thraustochytrid Aurantiochytrium sp. SD116 were analyzed. Results revealed that DGAT2C was mainly responsible for connecting PUFA to the sn-3 position of TAG molecules. However, DGAT2A and DGAT2D target SFA and/or MUFA. Conclusions There are two specific TAG assembly routes in Aurantiochytrium. The “saturated fatty acid (SFA) TAG lane” primarily produces SFA-TAGs mainly mediated by DGAT2D whose function is complemented by DGAT2A. And, the “polyunsaturated fatty acid (PUFA) TAG lane” primarily produces PUFA-TAGs via DGAT2C. In this study, we demonstrated the functional distribution pattern of four DGAT2s in oleaginous thraustochytrid Aurantiochytrium, and provided a promising target to rationally design TAG molecular with the desired characteristics.

Microalgal oils are natural and edible potential bioresources rich in PUFAs. Using microalgal oils to produce prepared functional lipids to replace natural animal and vegetable functional lipids not only can solve the bottleneck in the quality and supply for natural functional lipids, but also hopefully obtain new functional lipids products.The preparation of functional lipids by enzymatic method, lipid content and fatty acid composition of common microalgae were summarized, and the research progress on the preparation of functional lipids such as monoglycerides, structural triacylglycerides and functional phospholipid rich in PUFAs by enzymatic modification of microalgal oil was introduced. Preparation of functional lipids by enzymatic modification of microalgal oil is an new way for high-value utilization of microalgal oil.

BackgroundArtificial insemination (AI) is an effective reproductive technique to improve the performance of cashmere goats and prevent the spread of diseases, and the quality of the semen determines the success of AI. The potential of Moringa oleifera leaf powder (MOLP) and Moringa oleifera leaf ethanolic extract (MOLE) to improve semen quality has been reported, but the underlying mechanisms remain unclear. For the purpose, 18 mature male cashmere goats were randomly assigned into three groups: the control (CON), MOLP, and MOLE groups. The CON group received distilled water orally; the MOLP group was orally treated with 200 mg/kg body weight (BW) MOLP; and the MOLE group was orally treated with 40 mg/kg BW MOLE.ResultsResults showed that MOLE contained long-chain fatty acids and flavonoids. Treatment with MOLP and MOLE increased the activities of the serum catalase, superoxide dismutase, and glutathione peroxidase (P < 0.05), enhanced the total antioxidant capacity (P < 0.05), and reduced the serum malondialdehyde level (P < 0.05). At the same time, MOLE increased the contents of serum gonadotropin releasing hormone and testosterone (P < 0.05). Moreover, MOLE significantly increased sperm concentration, motility, and viability (P < 0.05). Meanwhile, MOLE raised the Chao1 index (P < 0.05) and altered the composition of the rumen microbiota; it also raised the relative abundance of Treponema (P < 0.05) and Fibrobacter (P < 0.05) and reduced the relative abundance of Prevotella (P < 0.1). Correlation analysis revealed the genus Prevotella was significantly negatively correlated with sperm concentration, as well as sperm motility and viability. Furthermore, MOLE significantly increased the rumen levels of the steroid hormones testosterone and dehydroepiandrosterone (P < 0.05), as well as the polyunsaturated fatty acids (PUFAs) alpha-Linolenic acid, gamma-Linolenic acid, docosapentaenoic acid, and 9-S-Hydroperoxylinoleicacid (P < 0.05).ConclusionsOral MOLE supplementation can improve semen quality by increasing the antioxidant capacity and altering the rumen microbiota and metabolites of cashmere goats. Moreover, the MOLP supplementation could enhance the antioxidant capacity of cashmere goats.

This paper presents the data on the growth pattern, amino acid, and fatty acid profile of five (5) selected indigenous marine microalgae (Chaetoceros sp.; Isochrysis sp.; Skeletonema sp.; Nannochloropsis sp.; and Tetraselmis sp.) of Bay of Bengal, Bangladesh. The microalgae species were cultured in f/2 Guillard's medium with maintaining standard physico-chemical parameters. The growth pattern was determined for all the microalgae as a prerequisite for further necessary experimental works. Biological treatment of saline and viscous food waste is hindered because of the overly concentrated substrates and contaminative growth. To address this issue, the salinity-tolerant Aurantiochytrium sp. L3W was cultivated on saline and viscous yakisoba (Y) and okonomiyaki (O) liquid food wastes supplied by a sauce factory in Hiroshima (Japan) at various viscosities to produce polyunsaturated fatty acids (PUFAs). The salinity of the growth medium may reduce the contaminative growth under non-sterile conditions. It was found that an increased viscosity of 1.34 Pa·s inhibited the growth of strain L3W. The liquid waste samples were diluted to retrieve the effective biomass growth. Cultivation in 25-times-diluted samples at the original acidic pH under non-sterile conditions revealed the docosahexaenoic acid (DHA) levels of 50 mg/g (Y) and 55 mg/g (O). Despite non-sterile conditions, neutral pH 7 adjustment induced a remarkable PUFA production. The resultant biomass, collected from the 25-times-diluted liquid waste samples, contained DHA and eicosapentaenoic acid (EPA) of 105 and 0.11 mg/g (Y); and of 50 and 0.06 mg/g (O), respectively. Furthermore, in the context of carbon and nitrogen consumption, the biomass yield analysis indicated that the sample Y contained more suitable nitrogen for the strain L3W as compared to the control medium. Deep-water shrimps are recognized as non-conventional culinary delicacies owing to the presence of high-quality polyunsaturated fatty acids, even though they remain one of the predominantly unexplored marine fishery resources. In this study, variation of fatty acid profiles of Aristeus alcocki , Solenocera choprai , Heterocarpus chani and Plesionika quasigrandis , caught along the south-west coast of India (Arabian Sea) during the months spanning from September 2018–April 2019, were compared. Among the deep-water shrimps studied, A. alcocki contained a greater amount of n -3 fatty acids, such as docosahexaenoic and eicosapentaenoic acid along with optimal n -3/ n -6 polyunsaturated fatty acid ratios beneficial for human nutrition (up to 8 during the winter months) when compared with other shrimps. In general, fatty acid concentrations exhibited variations, particularly with regard to polyunsaturated fatty acids, such as eicosapentaenoic acid and docosahexaenoic acid. These n -3 fatty acids were predominant during December–February of the studied year, but concentrations were noticeably lower during March and April. Females had considerably higher (P < 0.05) C 20–22 n -3 fatty acid concentrations along with nutritionally balanced polyunsaturated/saturated fatty acid ratio than males. Results of the two-way multivariate analysis of variance (MANOVA) revealed statistically significant differences in the fatty acid profiles between the species and months. The interaction effects of months with species were also highly significant (Wilk's lambda = 0.000001; F = 187.7, P < 0.0001). Lesser atherogenicity (<3) and thrombogenicity (<0.5) indices coupled with considerably greater n -3/ n -6 fatty acid ratios recognized these deep-water species as a possible source of highly nutritional human food. Background: Although numerous studies have investigated the association of dietary intake of omega-3 fatty acids with cognitive function and the risks of dementia, the relationship between fish oil supplementation and incident dementia in a large population-based cohort study has not yet well studied. Materials and methods: A total of 211,094 community-dwelling older persons over 60 years from the UK Biobank cohorts enrolled between 2006 and 2010 that reported regularly taking fish oil and had no dementia at baseline, was included in the present study. All participants completed an electronic questionnaire regarding habitual use of supplements including fish oil. Results: Overall, 83,283 (39.5%) participants reported regularly taking fish oil at baseline. Of 211,094 participants with the median age was 64.1 years, 5,274 participants developed dementia events during a median follow-up of 11.7 years, with 3,290 individuals derived from fish oil non-users. In the multivariable adjusted models, the adjusted hazard ratios (HRs) associated with fish oil supplementation for all-cause dementia, vascular dementia, frontotemporal dementia, and other dementia were 0.91 [CI = 0.84-0.97], 0.83 [CI = 0.71-0.97], 0.43 [CI = 0.26-0.72], 0.90 [CI = 0.82-0.98], respectively (all P < 0.05). However, no significant association between fish oil supplementation and Alzheimer's disease was found (HR = 1.00 [CI = 0.89-1.12], P = 0.977). In the subgroup analyses, the associations between use of fish oil and the risk of all-cause dementia (P for interaction = 0.007) and vascular dementia were stronger among men (P for interaction = 0.026). Conclusion: Among older adults, regular fish oil supplementation was significantly associated with a lower risks of incident all-cause dementia, as well as vascular dementia, frontotemporal dementia and other dementia but not Alzheimer's disease. Unconventional and underutilized plant oils are increasing popular and helping to meet demand for food, health and other industrial application. In recent years, the search for alternative sources that deliver healthful benefits, low‐environmental impact and ethical responsibility have become a priority to both scientists and industry. Oil from njangsa seed (Ricinodendron heudelotti) was recovered using four selected enzymes—hemicellulase, protease, pectinase, and amylase in an enzyme‐assisted aqueous extraction (EAE) and the recovery, quality indices and fatty acid composition was compared to solvent extraction using hexane (HE). Njangsa seed oil (NSO) recovered from the EAE ranged between 29 and 36% and were significantly (p < 0.05) lower than HE (46%), and the quality indices (free fatty acid, peroxide, para‐anisidine, and thiobarbituric acid values) were consistently lower and better than HE‐NSO. The fatty acid profile of EAE‐NSO was qualitatively and quantitatively comparable to HE‐NSO, with differences among the enzymes used. Alpha‐eleostearic acid was the most abundant fatty acid and was consistently ca 41% among the samples. Among the volatile compounds analyzed on the polar and nonpolar columns, 13 (2 alcohols, 9 aldehydes, and 2 esters) were detected in EAE‐NSO using hemicellulase, and 35 (4 aldehydes, 1 ester, and 30 hydrocarbons) in HE‐NSO. The most abundant flavor compounds identified were pentanal, methylcyclopentane, cyclohexane, 3‐methylpentane, (E,E)‐2,4‐nonadienal, (E)‐2‐heptenal, hexanal, 1‐butanol, and butanal. This study suggests that the amount, quality, and content of oil and volatile compounds in NSO can be influenced by the choice of extractant. [ABSTRACT FROM AUTHOR]
