106. Potent lipophilic 3-hydroxy-2-pyridinium aldoxime reactivators of phosphorylated cholinesterases

Abstract

Organophosphate compounds (OPs) poisoning can occur due to exposure to OP pesticides (e.g., parathion) or nerve agents (e.g., sarin, cyclosarin, VX, tabun). OPs irreversibly inhibit acetylcholinesterase (AChE) causing the accumulation of acetylcholine in synapses and cholinergic crisis, which can lead to respiratory arrest and death or result in long- term neurological impairments in survivors. Reactivation of OP-inhibited AChE is one of the possible therapy approaches, but standard oxime reactivators (2- PAM, HI-6, obidoxime) are not potent enough for every OP and achieve low brain concentrations due to a permanent charge (quaternary nitrogen). New class of oximes, 3- hydroxy-2-pyridine aldoximes, without permanent charge have been synthesized and tested in detail. They were proven to be potent reactivators of AChE inhibited by VX, sarin, cyclosarin, tabun, and paraoxon with the affinity of AChE in the micromolar range. Moreover, molecular docking studies predicted optimal orientation of the oximes in the active site of OP-inhibited AChE. Furthermore, these oximes were proven to be lipophilic and by analysing various physicochemical properties that describe their size, charge, lipophilicity, and polarity we predicted their penetration through the blood-brain barrier (BBB). The permeability of these oximes across the membrane was confirmed in vitro using an artificial membrane that mimics the BBB. In addition, most of the oximes were shown to be metabolically stable when incubated with human microsomes and were not cytotoxic for neuroblastoma and astrocytoma cell lines. Finally, the lead oxime crossed the BBB in vivo in mice after intramuscular application proving its potential to be used as a centrally active antidote.

Abstract

Organophosphate compounds (OPs) are still a plausible tool in wars and terroristic attacks where they are used as nerve agents (e.g. sarin, cyclosarin, VX, tabun). In addition, OP pesticides (e.g. paraoxon) cause more than a million acute poisonings per year worldwide. Their toxic mechanism is the inhibition of acetylcholinesterase (AChE) enzyme followed by the accumulation of acetylcholine in the synapses and cholinergic crisis. This can lead to respiratory arrest and death, whereas survivors experience long-term neurological impairments. Reactivation of OP-inhibited AChE is one of the approaches in therapy, but standard oxime reactivators (2-PAM, HI-6, obidoxime) are not potent enough for every OP and achieve low brain concentrations due to a permanent charge (quaternary nitrogen). Representatives of 3-hydroxy-2-pyridine aldoxime compounds, devoid of a permanent charge, were proven to be potent reactivators of AChE inhibited by VX, sarin, cyclosarin, tabun and paraoxon, and also showed the potency to efficiently detoxify OPs in whole human blood. Their blood-brain barrier penetration was predicted based on their physicochemical properties and in vitro PAMPA assay. In addition, logD7.4 values determined by chromatography confirmed their relatively high lipophilicity. To test whether the lipophilic character predisposes these oximes to exert toxic effects in the cells or be metabolized via the cytochrome P450 system, we used a cytotoxicity test in different cell lines and an incubation test with liver microsomes, respectively. The oximes were proven not to be toxic at physiologically relevant concentrations and to be metabolically stable. Abstract

International audience; Non‐protein thiols such as GSH, Hcy, or Cys, are present at different concentration levels in the body depending on the thiol species and their localization (plasma, cell, urine etc···). They play vital roles in controlling redox‐mediated biological processes, and abnormal levels of biothiols are related to a variety of syndromes. In few years, fluorometric methods have gain increasing attention enabling a rapid, sensitive and spatiotemporal detection of biothiols. Herein, is reported a novel two‐step access to a water‐soluble fluorogenic probe based on the 7‐aminocoumarin dye conjugated to a pyrazolone moiety. Upon the addition of biothiols, the fluorescence is turned‐on at 485 nm by a reversible thio‐Michael addition process, which enables the detection of biothiols in the low nanomolar range.

Abstract

International audience; Acetylcholinesterase is a well‐known enzyme, important in neurosciences and toxicology. Detection of AChE illustrates the complexity of quantifying proteins, even enzymes that have remarkable properties and substrates. AChE remains a model to propose novel strategies to detect proteins by light. Here we derived fluorescent probes from huprine, one of the most evolved AChE inhibitors. Two Near Infra‐Red (NIR) fluorescent probes HupNIR1 and HupNIR2 based on the huprine scaffold and cyanine 5.0 dyes have been synthesized and evaluated in situ for the detection of acetylcholinesterases in different tissues. As anticipated by the initial properties of huprine, both probes displayed a high affinity and selectivity for AChE vs BChE, with IC50 values in the nanomolar range and no non‐specific binding in the tissues. HupNIR2 appears the best probe for AChE with a great selectivity and sensitivity for AChE even in a low AChE region like striatum. Moreover, the binding of HupNIR2 is affected when AChE is inhibited with toxic molecules such as organophosphates. This work provided a new tool to visualize AChE activity in biological tissues.

126. New uncharged potent reactivators of AChE and BChE inhibited by nerve agents

Abstract

Intoxication by organophosphorus nerve agents (OPNA ; e.g. sarin, cyclosarin, tabun, VX) leads to the irreversible inhibition of enzymes acetylcholinesterase (AChE, E.C. 3.1.1.7.) and butyrylcholinesterase (BChE, E.C. 3.1.1.8). This inhibition causes acetylcholine accumulation in the peripheral and central nervous system synapses, leading to a cholinergic crisis (miosis, headache, dyspnoea, ocular impairments, excessive secretion, fasciculations, etc.). The most prominent symptoms with a possible lethal outcome are respiratory failure and seizures, but those who survive can also experience long-term neurological impairments, e.g. cognitive and behavioural incapacitations. The antidote treatment comprises an antimuscarinic drug, an oxime reactivator and an anticonvulsive drug. Standard reactivators (e.g. 2-PAM, HI-6, LüH-6) are not efficient for every OPNA, and since they have positively charged quaternary nitrogen in their structures, they cross the brain-blood barrier poorly. We synthesised and evaluated novel uncharged and therefore possibly centrally active reactivators. They showed a highly promising in vitro reactivation profile for VX-inhibited AChE/BChE, sarin- inhibited AChE and cyclosarin-inhibited BChE. Using molecular docking, the possible interactions that these oximes form with the amino acid residues in the active site gorge of the enzymes near the covalently linked OPNA, were detected. Additionally, in silico determined physicochemical properties (e.g. lipophilicity, polar surface area, number of hydrogen bond donors and acceptors, rotatable bonds, pKa values) implied that these novel oximes were likely to be centrally active. On the other side, the higher lipophilicity was connected with a higher probability of accumulation in different tissues and therefore could have led to adverse effects. Further evaluation of these novel reactivators’ in vivo efficacy is needed, as well as evaluations of their pharmacokinetic and toxicological profiles. 127. Novel centrally active antidote for intoxication by sarin and VX nerve agents

Abstract

Sarin and VX are some of the best known organophosphorus nerve agents (OPNA), which show their toxic effect through the irreversible inhibition of the enzymes acetylcholinesterase (AChE, E.C. 3.1.1.7.) and butyrylcholinesterase (BChE, E.C. 3.1.1.8) that normally degrade the neurotransmitter acetylcholine (ACh). This inhibition compromises normal cholinergic nerve signal transduction in the peripheral and central nervous system (PNS and CNS) synapses, leading to a cholinergic crisis with minor to medium symptoms (miosis, dyspnoea, ocular impairments, excessive secretion of body fluids, fasciculations, etc.), as well as to life- threatening impairments (respiratory failure and seizures). Survivors of attacks with these nerve agents can also experience long-term neurological impairments, e.g. cognitive and behavioural incapacitations. The antidote treatment comprises an antimuscarinic drug, an oxime reactivator of the OPNA-inhibited enzyme and an anticonvulsive drug. Standard reactivators (e.g. 2-PAM, HI-6, obidoxime) are not efficient for every OPNA, and since they have positively charged quaternary ammonium in their structures, they are ill-equipped to cross the brain-blood barrier (BBB). Novel uncharged and therefore possibly centrally active reactivators were synthesised and evaluated by in vitro and in silico methods. The in vitro reactivation assay showed that they are promising reactivators of hAChE/hBChE inhibited by VX, as well as good reactivators of hAChE inhibited by sarin. In silico- determined physicochemical parameters enabled the prediction of the amount of non-ionic species of specific antidotes at physiological pH, as well as the calculation of the CNS drug score. The cell line cytotoxicity profile showed that, at the most common therapeutic dose, they do not induce significant cell death. Induction of reactive oxygen species (ROS) production in cells was also tested to evaluate if they would have a synergistic or antagonistic effect toward one of the secondary mechanisms of neurotoxicity induced by the nerve agents (i.e. oxidative stress induction). Further in vivo BBB permeability potential and therapeutic efficacy is to be determined to conclude on the overall effectiveness of this design approach for reactivators of cholinesterases inhibited by OPNA. Medical practice today utilises an antimuscarinic atropine and the oxime reactivator of OPNA-inhibited main target acetylcholinesterase (AChE, EC 3.1.1.7). The approved pyridinium oximes (2-PAM, HI-6, obidoxime) are not efficient for every OPNA and they cannot cross the blood-brain barrier as they are charged. Our research was focused on several uncharged lipophilic oximes (JR595, JR585, GM508, CG193) designed to be passively transported across the blood-brain barrier and act in the central nervous system. Our primary testing in vitro showed that they can efficiently reactivate AChE inhibited by a range of OPNAs. However, little is still known about their in vivo beneficial effects that are not linked to reactivation and adverse effects that may be related to their structural characteristics. Namely, the preliminary testing showed higher chemosensitivity of the human neuronal cells (SH-SY5Y) to this kind of compounds. To get an insight into uncharged oximes, we thoroughly investigated their adverse effects on SH-SY5Y cells as the primary targets of these antidotes in humans. We studied the basic cytotoxicity of these oximes in terms of cell viability and cellular reactive oxygen species status. These two outcomes were also studied with OPNA compounds, which were applied in nanomolar concentrations in order to confirm oxidative stress, which is one of the proposed mechanisms of the neuronal damage in OP intoxication. Our results suggest that there is a thin line between beneficial and adverse effects of uncharged oximes on the cellular level. Therefore, our future studies will try to elucidate the mechanism behind the observed effects in vitro and to evaluate the toxicity of uncharged oximes in vivo. Abstract

Hydroxamic acids are outstanding zinc chelating groups that can be used to design potent and selective metalloenzyme inhibitors in various therapeutic areas. Some hydroxamic acids display a high plasma clearance resulting in poor in vivo activity, though they may be very potent compounds in vitro. We designed a 57-member library of hydroxamic acids to explore the structure–plasma stability relationships in these series and to identify which enzyme(s) and which pharmacophores are critical for plasma stability. Arylesterases and carboxylesterases were identified as the main metabolic enzymes for hydroxamic acids. Finally, we suggest structural features to be introduced or removed to improve stability. This work thus provides the first medicinal chemistry toolbox (experimental procedures and structural guidance) to assess and control the plasma stability of hydroxamic acids and realize their full potential as in vivo pharmacological probes and therapeutic agents. This study is particularly relevant to preclinical development as it allows obtaining compounds equally stable in human and rodent models.

Abstract

The combination of the scaffolds of the cholinesterase inhibitor huprine Y and the antioxidant capsaicin results in compounds with nanomolar potencies toward human acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) that retain or improve the antioxidant properties of capsaicin. Crystal structures of their complexes with AChE and BChE revealed the molecular basis for their high potency. Brain penetration was confirmed by biodistribution studies in C57BL6 mice, with one compound (5i) displaying better brain/plasma ratio than donepezil. Chronic treatment of 10 month-old APP/PS1 mice with 5i(2 mg/kg, i.p., 3 times per week, 4 weeks) rescued learning and memory impairments, as measured by three different behavioral tests, delayed the Alzheimer-like pathology progression, as suggested by a significantly reduced Aβ42/Aβ40 ratio in the hippocampus, improved basal synaptic efficacy, and significantly reduced hippocampal oxidative stress and neuroinflammation. Compound 5iemerges as an interesting anti-Alzheimer lead with beneficial effects on cognitive symptoms and on some underlying disease mechanisms.
