Abstract

Monolithic materials prepared from a mixture of n-lauryl methacrylate (LMA) and ethylene glycol dimethacrylate (EGDMA) dedicated to nano-liquid chromatography separation were synthesized using in situ UV polymerization in 75 µm inner diameter capillary tubing. A mixture of cyclohexanol and ethylene glycol was used as a porogen to control porosity. While the preparation conditions yielded satisfactory analytical results, values of pertinent parameters turned out to be critical for obtaining columns with efficient separation. In particular, the impact of two key parameters was studied here in an attempt to identify optimal preparation conditions: (a) different concentrations of the crosslinker EGDMA and (b) different porogen compositions while the monomer to porogen ratio was kept constant. Resulting monolithic phases were characterized in terms of permeability, mean pore diameter and swelling using three different eluents (water, acetonitrile and a mixture at maximum viscosity). First, the LMA/EGDMA monolithic phases present peculiar morphology and hydrodynamic properties for 37% by weight of EGDMA, as reflected by the peak observed for their permeability and mean pore diameter. Swelling experiments revealed the coexistence of two phases in the monolithic structure: a highly crosslinked rigid phase which was insensitive to swelling in the presence of solvent and a weakly crosslinked flexible phase exhibiting significant swelling, with a transition to such a biphasic behavior taking place at 37% by weight of EGDMA. The effects of porogen composition and network swelling properties are presented based on a combination of the Flory − Huggins theory of isotropic mixing in polymer solutions and the Flory − Rehner theory of rubber elasticity in the affine network approximation. © 2016 Society of Chemical Industry

Abstract

Our work is focused on two applications of fine tunable microfluidic systems, first to optimize heterogeneous size nanoparticle synthesis and second to build catalytic microreactors for advanced organic reactions. The first part of our work consists in the use of an original microfluidic setup for gold nanoparticle synthesis, which allows a high control of the reaction parameters as the reactants flow, the concentration, the temperature, and the reaction time. We show that using such microfluidic systems permit a better control of the reaction parameters for producing homodispersed 1–2 nm gold nanoparticle. The second part of our work deals with the incorporation of gold nanoparticles into silica capillaries to build catalytic microreactors dedicated to fine chemical reactions. Our strategy consists in the immobilization of gold nanopadiegolirticles onto the inner surface (2D dispersion) or into the inner volume (3D dispersion) of functionalized silica microcapillaries. Characterizations show that by different functionalization procedures, those gold nanoparticles are well anchored inside the microcapillary.

The physical chemistry and the free radical chemistry of flavan-3-ols are extensively investigated by laser flash photolysis and cyclic voltammetry studies. The results are very useful in discussing biological and biochemical phenomenon and give a new insight into the antioxidative properties of flavan-3-ols.

110. Comparative Proteomic and Transcriptomic Analysis of Follistatin-Induced Skeletal Muscle Hypertrophy

Abstract

Skeletal muscle, the most abundant body tissue, plays vital roles in locomotion and metabolism. Myostatin is a negative regulator of skeletal muscle mass. In addition to increasing muscle mass, Myostatin inhibition impacts muscle contractility and energy metabolism. To decipher the mechanisms of action of the Myostatin inhibitors, we used proteomic and transcriptomic approaches to investigate the changes induced in skeletal muscles of transgenic mice overexpressing Follistatin, a physiological Myostatin inhibitor. Our proteomic workflow included a fractionation step to identify weakly expressed proteins and a comparison of fast versus slow muscles. Functional annotation of altered proteins supports the phenotypic changes induced by Myostatin inhibition, including modifications in energy metabolism, fiber type, insulin and calcium signaling, as well as membrane repair and regeneration. Less than 10% of the differentially expressed proteins were found to be also regulated at the mRNA level but the Biological Process annotation, and the KEGG pathways analysis of transcriptomic results shows a great concordance with the proteomic data. Thus this study describes the most extensive omics analysis of muscle overexpressing Follistatin, providing molecular-level insights to explain the observed muscle phenotypic changes.

111. Scale-up Study of Benzoic Acid Alkylation in Flow: From Microflow Capillary Reactor to a Milliflow Reactor

Abstract

The esterification reaction is one of the most important reaction in organic chemistry, and the development of scalable continuous flow conditions is of high interest. Here we describe a scaling-up study of benzoic acid alkylation by MeI in the presence of superbases from a capillary reactor (282 mg·h–1at 75 °C) to a low flow reactor (LFR, 6.82 g·h–1at 80 °C). First, the optimization of the base demonstrated that 1,8-diazabicyclo[5.4.0]-undec-7-ene (DBU) was a good compromise in terms of basicity and price. A comparison between batch and LFR reaction showed similar kinetic rates. We also observed a maximum reactivity for a flow rate of 3 mL·min–1which corresponds to an optimum mixing of the entering fluids. A colorimetric study permits to show a visual change in the flow regime (dispersed to stratified) by varying the flow rate with a maximum conversion obtained at 3 mL·min–1corresponding to a 105 s residence time, demonstrating that the studied reaction is sensitive to mixing conditions and that LFR presents some mixing limitations for low flow rates. After thermal optimization (80 °C), the reaction has been improved to a productivity of 6.82 g/h at the lab scale using DBU as the base and Eyring/Arrhenius plots permitted to extract activation energies.

Abstract

Newly designed gels for electrophoresis protein separation were synthesized from acrylamide, N,N′-methylenebis (acrylamide) and dextran mixtures. Radical polymerization was initiated by ammonium persulfate and N,N,N′,N′-tetramethylethylenediamine. The time dependence of absorbance during polymerization was monitored using UV-visible spectroscopy. The exothermic polymerization process exhibited a sharp rise of temperature reminiscent of the Trommsdorff effect. The swelling kinetics of the synthesized gels was examined in deionized water and buffer solutions. One of the challenges was to find an alternative to commercial products, sold as mixtures with no detailed chemical contents, commonly used in sodium dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE) for protein separation. For this reason, a systematic comparison was made of the properties of one of the most commonly used commercial gels, Duracryl™ from Genomics Solution Inc., and those of the synthesized polyacrylamide/dextran gels. Abstract

International audience; Protein identification is one of the major task of Proteomics researchers. Protein identification could be resumed by searching the best match between an experimental mass spectrum and proteins from a database. Nevertheless this approach can not be used to identify new proteins or protein variants. In this paper an evolutionary approach is proposed to discover new proteins or protein variants thanks a "de novo sequencing" method. This approach has been experimented on a specific grid called Grid5000 with simulated spectra and also real spectra.
