A cDNA clone, designated pUP-44, whose longest open reading frame codes for a protein that is homologous to the wheat α-amylase inhibitors, has been isolated from a library obtained from developing barley endosperm. The deduced sequence for the mature protein, which is 122 residues long, is preceded by a sequence of 30 residues which has the typical features of a signal peptide. A closely corresponding protein, designated BDAI-1, has been isolated from mature endosperm. BDAI-1 behaves as a dimer and inhibits the α-amylase from the insect Tenebrio molitor at concentrations that have no effect on salivary or pancreatic α-amylases. [ABSTRACT FROM AUTHOR]

The CM proteins are a group of major salt-soluble endosperm proteins encoded by a disperse multigene family. The effects of high-lysine mutations on the net accumulation in barley endosperm of three members of this group (CMa, CMb, and CMe) have been investigated. Genes CMa, CMb and CMe are located in chromosomes 1, 4, and 3 respectively. Protein CMe has been found to be identical with a previously described trypsin inhibitor. The three proteins have been quantified in the different genetic stocks by HPLC, The different high-lysine mutations have different effects on the expression patterns of the three genes: CMe is markedly decreased and CMa and CMb are increased in mutant Risø 1508, whereas all three proteins are decreased in Risø 527 and increased in Ris0 7 with respect to the wild-type Bomi; CMa and CMb are increased and CMe is unaffected in mutant Risø 56 with respect to the wild-type Carlsberg II; and protein CMe is markedly decreased in Hiproly barley as compared with its sister line CI4362, The implications of these results in connection with the evolution of CM proteins and with the characterization of high-lysine mutations are discussed. - Abstract
The N‐terminal amino acid sequences of two chloroform/methanol soluble globulins from barley and one form wheat are reported. They are homologous with N‐terminal sequences previously reported for α‐amylase and trypsin inhibitors from cereals and 2 S storage proteins from castor bean and rape. Three albumins were also purified from Aegilops squarrosaand Triticum monococcum. These had N‐terminal amino acid sequences most closely related to the α‐amylase and trypsin inhibitors. The relationships of this superfamily of seed proteins are discussed.

A tetrameric inhibitor that is active against α‐amylase from the larvae of the insect Tenebrio molitor, but inactive against the enzyme from human saliva and against the endogenous one, has been described in barley endosperm. The subunits of the inhibitor have been identified as the previously characterized proteins CMa, CMb and CMd, of which only CMa was inhibitory by itself.

A barley salt-soluble protein of 14.5 kDa, which inhibits the α-amylase from the insect Tenebrio molitor, has been identified as a major IgE-binding component of sera from baker's asthma patients. The N-terminal amino acid sequence of this protein indicates that it is a member of a previously described family of α-amylase/trypsin inhibitors.

[EN] Several members of an inhibitor family of heterologous α-amylases and of trypsin have been isolated from wheat, barley and rye endosperms. Comparisons of the amino acid sequences of these inhibitors together with the chromosomal of their structural genes in the three species, indicate that they are encoded by a disperse multi-gene family that must have originated both by translocation and intrachromosomal duplications events that took place in common ancestors of wheat, barley and probably rye. Homology among some members of the inhibitor family and various proteins of maize, ragi, castor bean and others, indicate that this protein family extends to other phylogenetically distant species.

- Abstract
[EN] The historical stone heritage that we inherit must be passed on to future generations, not only in the same conditions that we found it but, if possible, in better ones. Construction also demands better and more durable materials, often stone. The protection of these materials requires knowledge of the types of rocks and their physical properties. The characterization of these properties is often standardized to ensure the quality and reproducibility of the protocols. These must be approved by entities whose purpose is to improve the quality and competitiveness of companies and to protect the environment. Standardized water absorption tests could be envisaged to test the effectiveness of certain coatings in protecting natural stone against water penetration, but we found that some steps of these protocols neglect any surface modification of the stones, and hence may not be completely effective when a hydrophilic protective coating (i.e., graphene oxide) is present. In this work, we analyze the UNE 13755/2008 standard for water absorption and propose alternative steps to adapt the norm for use with coated stones. The properties of coated stones may invalidate the interpretation of the results if the standard protocol is applied as is, so here we pay special attention to the characteristics of the coating applied, the type of water used for the test, the materials used, and the intrinsic heterogeneity of the specimens

[ES] El presente proyecto contempla la adecuación del pavimento de la pista de vuelos del Aeropuerto de Son Bonet, demoliendo y levantando el pavimento existente, excavación del fondo de caja y posteriormente proceder a la nueva pavimentación. A continuación, se ejecutará la señalización horizontal y la reposición de los posibles servicios afectados. Todas las actuaciones seguirán las normas y recomendaciones indicadas en la Orden FOM/2086/2011, de 8 de julio, por la que se actualizan las normas técnicas contenidas en el Anexo al Real Decreto 862/2009, de 14 de mayo, por el que se aprueban las normas técnicas de diseño y operación de aeródromos de uso público y se regula la certificación de los aeropuertos de competencia del Estado., [EN] This project contemplates the runway repavement at Son Bonet Airport, demolishing and raising the existing pavement, excavation and later, paving the new runway. Next, the surface marking and the replacement of the possible affected services will be executed. All actions will follow the standards and recommendations indicated in Order FOM/2086/2011, of July 8, which updates the technical standards contained in the Annex to Royal Decree 862/2009, of May 14, which the technical standards for the design and operation of aerodromes for public use are approved and the certification of airports under the jurisdiction of the State is regulated.

[ES] La Directiva Europea sobre ruido ambiental, establece un enfoque común para evitar o reducir los efectos nocivos de la exposición al ruido. Para analizar la molestia debida a la contaminación acústica en León, se llevó a cabo un estudio mediante una encuesta entre 509 habitantes. En el estudio, las personas más sensibles al ruido también lo consideran un agente muy contaminante. Se constata que el ruido afecta a diversas actividades, como las interferencias en la comunicación y la actividad intelectual. Asimismo, se presentan las acciones consideradas más eficaces por los habitantes, para mitigar el ruido en su ciudad., [EN] The Environmental Noise Directive establishes a common approach to prevent or reduce the harmful effects of noise exposure. To analyze the nuisance due to noise pollution in Leon (Spain), we carried out a study using a survey of 509 people. It shows that people who consider them self as most sensitive to noise, also think about it as a highly polluting agent. Also, some daytime activities are affected, like interference with talks and intellectual activity. It also presents an overview of the actions considered more effective by the inhabitants to mitigate the noise in their city

[ES] La Universidad de León apuesta por la preservación del patrimonio cultural de la provincia y por proteger y divulgar la riqueza de León. Su grupo de investigación Nanoshield trabaja para aminorar los efectos que el clima adverso tiene en la roca patrimonial más representativa, la caliza. En colaboración con el Instituto de Carboquímica de Zaragoza, se han sintetizado una serie de nanomateriales que ya han sido probados exitosamente en laboratorio y se están ensayando en exteriores, lo que podrá demostrar la efectividad de los productos. Con estos nuevos materiales se podrá proteger la superficie de iglesias, palacios, esculturas, etc. En concreto, los derivados del grafeno ya han sido patentados a nivel europeo. También se pone a disposición de particulares técnicas de espectroscopía y diferentes ensayos, como de absorción y de resistencia, para evaluar el estado del bien y protegerlo eficientemente. Pero, además de trabajar por la preservación de los bienes, también han apostado por promocionarlos y hacerlos más accesibles para todos. Para lograrlo, se ha comenzado una campaña de escaneo de edificios, tanto en el exterior como en el interior, que se completará con imágenes de vuelos con dron, ofreciendo la riqueza de León en forma de visitas virtuales.

[EN] Most concrete studies are concentrated on mechanical properties especially strength properties either directly or indirectly (fresh and durability properties). Hence, the ratio of split tensile strength to compressive strength plays a vital role in defining the concrete properties. In this review, the impact of design parameters on the strength ratio of various grades of Self-Compacting Concrete (SCC) with recycled aggregate is assessed. The design parameters considered for the study are Water to Cement (W/C) ratio, Water to Binder (W/B) ratio, Total Aggregates to Cement (TA/C) ratio, Fine Aggregate to Coarse Aggregate (FA/CA) ratio, Water to Solid (W/S) ratio in percentage, superplasticizer (SP) content (kg/cu.m), replacement percentage of recycled coarse aggregates (RCA), replacement percentage of recycled fine aggregates (RFA), fresh density and loading area of the specimen. It is observed that the strength ratio of SCC with recycled aggregates is affected by design parameters.

[EN] University education in times of COVID-19 was forced to seek alternative teaching/learning methods to the traditional ones, having to abruptly migrate to the online modality, changes that have repercussions on student satisfaction. That is why this study aims to compare the level of student satisfaction in face-to-face and “forced” online modalities under COVID-19. A quantitative, cross-sectional methodology was applied to two groups of students: Under a face-to-face modality (n = 116) and under an online modality (n = 120), to which a questionnaire was applied under a Likert scale, with four dimensions: Course design structure, content, resources, and instructor. Non-parametric statistics, specifically the Mann–Whitney U-test, were used to compare the groups. The results showed that there are significant differences in the level of satisfaction of students in the face-to-face and online “forced” modalities (p = 0.01984 < 0.05), and the dimensions of the level of satisfaction that presented significant differences were course design structure (p = 0.04523 < 0.05) and content (p = 0.00841 < 0.05). The research shows that students in the face-to-face modality express a higher level of satisfaction, which is reflected in the dimension design structure of the course, specifically in its workload indicator, as well as in the dimension content, in its indicators, overlapping with other courses and materials.

[ES] En ocasiones, la transformación de espacios urbanos en productos de consumo turístico, no ha tenido en cuenta la protección medioambiental y la funcionalidad. Muchas de estas áreas se han convertido en zonas peatonales, donde numerosas fuentes de ruido pueden causar molestia entre la población, conduciendo al abandono de las mismas. Los mapas estratégicos de ruido evalúan globalmente la exposición al mismo, dejando dichas zonas al margen del estudio. En este sentido, se propone el análisis de la zona peatonal de León (España). Su incorporación a los mapas se realizará a partir de una clasificación urbana y medidas de corta duración., [EN] Sometimes, transformation of urban spaces in tourism consumption products, has not taken into account environmental protection and urban functionality. [ES] El presente proyecto tiene como objetivo la redacción de un documento teórico técnico que proporcione al promotor los datos necesarios para el diseño, ejecución y puesta en marcha de una serie de instalaciones. Estas instalaciones serán: Ventilación, calefacción, climatización y ampliación de las cámaras de conservación de congelados y túnel de congelación. La ventilación se calcula acorde a las necesidades impuestas por la normativa en toda la fábrica. La calefacción y climatización se hace a través de una UTA en la zona administrativa, aprovechando así la instalación de ventilación existente. Se modifican algunos cerramientos establecidos en el proyecto previo para cumplir con la normativa en materia de ahorro energético. La cámara de conservación y el túnel de congelación se amplían debido a que su capacidad no es suficiente para el aumento de producción. El resto de maquinaria es capaz de cumplir con la demanda debido a que no trabajaban a plena capacidad. Se calculan también todas las conducciones necesarias y los elementos para su correcto funcionamiento. La parcela donde se ubicará el proyecto es de propiedad del promotor y se localiza en el Polígono Industrial de Onzonilla (León). La elaboración de este proyecto está supeditada al cumplimiento de la legislación vigente y de los condicionantes impuestos por el promotor. Todo lo anteriormente expuesto se refleja en los siguientes documentos que componen el proyecto: memoria, planos, pliego de condiciones, presupuesto y estudio de seguridad y salud., [EN] This project´s objetive is developing a technical theoretical document that provides the promoter with the necessary data for the design, execution and start up of a series of facilities. These facilities will be: Ventilation, heating, air conditioning and expansion of the frozen storage chambers and freezing tunnel. Ventilation is calculated according to the needs imposed by regulations throughout the factory. The heating and air conditioning is done through a UTA in the administrative area, thus taking advantage of the existing ventilation installation. Some enclosures established in the previous project are modified to comply with the regulations on energy saving. The conservation chamber and the freezing tunnel are expanded due to the fact that their capacity is not enough to increase production. The rest of the machinery is capable of cope with the demand because they were not working at full capacity. All the necessary conductions and the elements for its correct operation are also calculated. The plot where the project will be located is the property of the developer and is located in the Onzonilla Industrial Estate (León). The preparation of this project is subject to compliance with current legislation and the conditions imposed by the promoter. All of the above is reflected in the following documents that make up the project: report, plans, specifications, budget and health and safety study.

[ES] En el presente proyecto se mostrará las medidas y operaciones realizadas para el desarrollo completo de un centro logístico cuyo uso es exclusivamente la conservación de plátanos y su correcta maduración. Para ello se deberá llevar a cabo el cumplimiento de la normativa destinada a este sector. En primer lugar, se ha seleccionado la localización siguiendo una serie de pautas y consideraciones. También se han tenido en cuenta las condiciones del promotor, importantes para el correcto desarrollo del centro. Con esas condiciones se ha desarrollado el sistema de cámaras frigoríficas y la distribución de la planta industrial. En este proyecto se encontrarán los planos obtenidos en función de todas las instalaciones que complementan a este centro. La finalidad principal de este proyecto es realizar los cálculos para que la conservación de la mercancía del producto sea perfecta. El producto elegido son los plátanos ya que estos requieren una temperatura de conservación alta, una atmosfera controlada y unos niveles de etileno correctos para que su desarrollo se realice en condiciones óptimas. En cuanto a la elección del refrigerante, se ha tenido en cuenta la normativa actual, así como las posibles variaciones que pueda sufrir. Es por ello por lo que la elección del refrigerante es fundamental. La selección de este se ha realizado también, teniendo en cuenta el impacto medioambiental que este tendrá. Otro punto importante es, dadas las necesidades frigoríficas de cada cámara, la elección de los componentes necesarios para su correcto funcionamiento, así como el diseño y dimensionamiento de las tuberías por donde circula el refrigerante para evitar pérdidas de presión excesivas que influyan en el desarrollo y rendimiento de la instalación. Los elementos de las cámaras se han tratado de elegir de una forma rentable buscando siempre que cumplan las necesidades exigidas, pero con un consumo de potencia controlado para que así el consumo de electricidad no sea elevado. [ES] El presente proyecto tiene como objetivo, exponer toda la información necesaria para llevar a cabo la ejecución de una nave industrial acondicionada para uso de oficinas y situada en el Polígono Industrial de León. La nave industrial contará con varias salas alquilables a empresas externas, como pueden ser algunas de las oficinas, la sala de proyecciones o el almacén, poniendo a disposición una gran plantilla de profesionales que desempeñarán la gestión del edificio. La nave industrial estará ubicada en la Parcela 04 del polígono 78330 del Polígono Industrial de León, en la zona perteneciente al término municipal de Santovenia de la Valdoncina. La parcela seleccionada pone a disposición 4400 m2 de superficie, de los cuales 2000 m2 se destinarán íntegramente para la nave industrial y el resto serán distribuidos entre los aparcamientos, la zona de carga y descarga, la zona peatonal y los retranqueos del edificio. El edificio contará con multitud de instalaciones de suministro, para asegurar el máximo confort en el interior del edificio, como pueden ser las instalaciones de electricidad, iluminación, fontanería, saneamiento y calefacción. Toda la documentación descrita en el presente proyecto, cumple estrictamente la normativa vigente aplicable a cada uno de los ámbitos del mismo. El presupuesto general del proyecto, teniendo en cuenta todos los aspectos, asciende a una cantidad de 1.928.019,37€.

[ES] Se trata de un almacén frigorífico con un sistema de picking automatizado para productos congelados en el que se recepciona, almacena, empaqueta y se expide la mercancía. El proyecto desarrollara la instalación frigorífica utilizando amoniaco para alcanzar la temperatura de régimen. El almacén se dividirá en dos zonas, la zona de recepción/expedición, con una temperatura de régimen de 0ºC y la zona del sistema automatizado de picking con -23ºC. La primera zona constara con dos instalaciones de expansión directa simple con R152a y la segunda constara de una instalación inundada de R717 (amoniaco) de doble compresión y con un sistema de desescarche por gas caliente.

Este trabajo analiza los riesgos de los diferentes procesos de soldadura que hoy en día se realizan de forma manual, aunque muchos de ellos ya han sido automatizados, en función de las características del trabajo se aplican de forma manual. En los procesos automatizados al no existir la presencia del trabajador el riesgo es eliminado. En la actualidad tenemos cuatro métodos que son manuales: la soldadura oxiacetilénica, la soldadura de tungsteno y gas inerte, la soldadura con electrodo revestido y la soldadura por arco metálico y gas. Existen diferencias entre los riesgos de cada método. La soldadura acetilénica al usar gases inflamables y oxígeno puro quizá la convierte en la más peligrosa. La soldadura con arco de tungsteno es capaz de producir grandes cantidades de ozono. En sus inicios se usaban electrodos de tungsteno con torio que al ser afilados emitían partículas radioactivas. La soldadura con electrodo revestido emite demasiados humos metálicos. La soldadura por arco metálico y gas es la menos peligrosa en cuanto a gases, pero es más costoso su equipo. Los humos de soldadura dependen principalmente de cuatro factores: del material base, recubrimiento del metal base, del metal de aporte y los gases protectores y del aire circundante

[ES] En el presente proyecto se mostrará las medidas y operaciones realizadas para el desarrollo completo de un centro logístico cuyo uso es exclusivamente la conservación de plátanos y su correcta maduración. Para ello se deberá llevar a cabo el cumplimiento de la normativa destinada a este sector. En primer lugar, se ha seleccionado la localización siguiendo una serie de pautas y consideraciones. También se han tenido en cuenta las condiciones del promotor, importantes para el correcto desarrollo del centro. Con esas condiciones se ha desarrollado el sistema de cámaras frigoríficas y la distribución de la planta industrial. En este proyecto se encontrarán los planos obtenidos en función de todas las instalaciones que complementan a este centro. La finalidad principal de este proyecto es realizar los cálculos para que la conservación de la mercancía del producto sea perfecta. El producto elegido son los plátanos ya que estos requieren una temperatura de conservación alta, una atmosfera controlada y unos niveles de etileno correctos para que su desarrollo se realice en condiciones óptimas. En cuanto a la elección del refrigerante, se ha tenido en cuenta la normativa actual, así como las posibles variaciones que pueda sufrir. Es por ello por lo que la elección del refrigerante es fundamental. La selección de este se ha realizado también, teniendo en cuenta el impacto medioambiental que este tendrá. Otro punto importante es, dadas las necesidades frigoríficas de cada cámara, la elección de los componentes necesarios para su correcto funcionamiento, así como el diseño y dimensionamiento de las tuberías por donde circula el refrigerante para evitar pérdidas de presión excesivas que influyan en el desarrollo y rendimiento de la instalación. Los elementos de las cámaras se han tratado de elegir de una forma rentable buscando siempre que cumplan las necesidades exigidas, pero con un consumo de potencia controlado para que así el consumo de electricidad no sea elevado. Este proyecto contará también, con un estudio de entidad propia donde se verán reflejados los estudios de impacto medioambiental, estudios geotécnicos, programación de la ejecución y puesta en marcha y en último lugar un estudio de seguridad y salud en la construcción. [ES] El presente proyecto tiene como objetivo la redacción de un documento teórico técnico que proporcione al promotor los datos necesarios para el diseño, ejecución y puesta en marcha de una serie de instalaciones. Estas instalaciones serán: Ventilación, calefacción, climatización y ampliación de las cámaras de conservación de congelados y túnel de congelación. La ventilación se calcula acorde a las necesidades impuestas por la normativa en toda la fábrica. La calefacción y climatización se hace a través de una UTA en la zona administrativa, aprovechando así la instalación de ventilación existente. Se modifican algunos cerramientos establecidos en el proyecto previo para cumplir con la normativa en materia de ahorro energético. La cámara de conservación y el túnel de congelación se amplían debido a que su capacidad no es suficiente para el aumento de producción. El resto de maquinaria es capaz de cumplir con la demanda debido a que no trabajaban a plena capacidad. Se calculan también todas las conducciones necesarias y los elementos para su correcto funcionamiento. La parcela donde se ubicará el proyecto es de propiedad del promotor y se localiza en el Polígono Industrial de Onzonilla (León). La elaboración de este proyecto está supeditada al cumplimiento de la legislación vigente y de los condicionantes impuestos por el promotor. Todo lo anteriormente expuesto se refleja en los siguientes documentos que componen el proyecto: memoria, planos, pliego de condiciones, presupuesto y estudio de seguridad y salud. [EN] This project´s objetive is developing a technical theoretical document that provides the promoter with the necessary data for the design, execution and start up of a series of facilities. These facilities will be: Ventilation, heating, air conditioning and expansion of the frozen storage chambers and freezing tunnel. Ventilation is calculated according to the needs imposed by regulations throughout the factory. The heating and air conditioning is done through a UTA in the administrative area, thus taking advantage of the existing ventilation installation. Some enclosures established in the previous project are modified to comply with the regulations on energy saving. The conservation chamber and the freezing tunnel are expanded due to the fact that their capacity is not enough to increase production. The rest of the machinery is capable of cope with the demand because they were not working at full capacity. All the necessary conductions and the elements for its correct operation are also calculated. The plot where the project will be located is the property of the developer and is located in the Onzonilla Industrial Estate (León). The preparation of this project is subject to compliance with current legislation and the conditions imposed by the promoter. All of the above is reflected in the following documents that make up the project: report, plans, specifications, budget and health and safety study.

136. Adaptation of a Standard Method for Water Absorption Testing of Stone Materials: The Case of a Hydrophilic Protective Coating.

- Abstract
The historical stone heritage that we inherit must be passed on to future generations, not only in the same conditions that we found it but, if possible, in better ones. Construction also demands better and more durable materials, often stone. The protection of these materials requires knowledge of the types of rocks and their physical properties. The characterization of these properties is often standardized to ensure the quality and reproducibility of the protocols. These must be approved by entities whose purpose is to improve the quality and competitiveness of companies and to protect the environment. Standardized water absorption tests could be envisaged to test the effectiveness of certain coatings in protecting natural stone against water penetration, but we found that some steps of these protocols neglect any surface modification of the stones, and hence may not be completely effective when a hydrophilic protective coating (i.e., graphene oxide) is present. In this work, we analyze the UNE 13755/2008 standard for water absorption and propose alternative steps to adapt the norm for use with coated stones. The properties of coated stones may invalidate the interpretation of the results if the standard protocol is applied as is, so here we pay special attention to the characteristics of the coating applied, the type of water used for the test, the materials used, and the intrinsic heterogeneity of the specimens.

- Abstract
Introduction: The COVID-19 pandemic has resulted in unparalleled changes to patient care, including the suspension of cancer surgery. Concerns regarding COVID-19-related risks to patients and healthcare workers with the re-introduction of major complex minimally invasive and open surgery have been raised. This study examines the COVID-19 related risks to patients and healthcare workers following the re-introduction of major oesophago-gastric (EG) surgery., Patients and Methods: This was an international, multi-centre, observational study of consecutive patients treated by open and minimally invasive oesophagectomy and gastrectomy for malignant or benign disease. Patients were recruited from nine European centres serving regions with a high population incidence of COVID-19 between 1 May and 1 July 2020. The primary endpoint was 30-day COVID-19-related mortality. All staff involved in the operative care of patients were invited to complete a health-related survey to assess the incidence of COVID-19 in this group., Results: In total, 158 patients were included in the study (71 oesophagectomy, 82 gastrectomy). Overall, 87 patients (57%) underwent MIS (59 oesophagectomy, 28 gastrectomy). A total of 403 staff were eligible for inclusion, of whom 313 (78%) completed the health survey. Approaches to mitigate against the risks of COVID-19 for patients and staff varied amongst centres. No patients developed COVID-19 in the post-operative period. Two healthcare workers developed self-limiting COVID-19., Conclusions: Precautions to minimise the risk of COVID-19 infection have enabled the safe re-introduction of minimally invasive and open EG surgery for both patients and staff. Further studies are necessary to determine the minimum requirements for mitigations against COVID-19., (© 2021. The Author(s).)

Background: The best alimentary and biliopancreatic limb (BPL) lengths in the Roux-en-Y gastric bypass (RYGB) still remain unclear. The aim of this study was to compare the effect of a BPL of 70 vs 120 cm, with a constant AL of 150 cm on long-term weight loss, remission of comorbidities, and supplementation needs after RYGB., Patients and Methods: A prospective randomized study of morbidly obese patients undergoing RYGB was performed. Patients were randomized into two groups: those patients undergoing RYGB with a BPL of 70 cm (BPL 70 cm) and those ones undergoing RYGB with a BPL of 120 cm (BPL 120 cm). BMI, excess BMI loss (EBMIL), remission of comorbidities and specific vitamin and mineral supplementation needs at 1, 2, and 5 years were analyzed., Results: Two hundred fifty-three patients were included in each group. There were no significant differences in BMI, EBMIL and the remission of diabetes mellitus, hypertension, and dyslipidemia between groups at 1, 2, and 5 years after surgery. Patients from group BPL 120 cm required greater specific supplementation of vitamin B12, folic acid, and vitamin A during all the follow-up., Conclusion: A RYGB with 120 cm BPL does not achieve greater weight loss or remission of comorbidities than a RYGB with 70 cm BPL but is associated with greater deficiencies of vitamin B12, vitamin A, and folic acid., Trial Registration: ClinicalTrials.gov Identifier NCT03607305. https://clinicaltrials.gov/.

Background: Profilins are commonly involved in polysensitization of allergic patients; therefore, appropriate markers should be used in component-resolved diagnosis., Objective: To evaluate the immunological equivalence between profilins from pollens and plant-derived foods, to be used in component-resolved diagnosis., Methods: Specific immunoglobulin (Ig) G antibodies against pollen and fruit profilins, as well as sera from patients allergic to mustard, melon, or olive pollen, were used. Purified profilins from mustard seeds, fruit melon, and chenopod and birch pollen were assayed in immunoblotting, enzyme-linked immunosorbent assay (ELISA), and ELISA inhibition assays., Results: Significant correlation was found in the response of purified profilins by ELISA and immunoblotting for both specific IgG and IgE. The highest levels of IgE binding were obtained for olive pollen-allergic patients, which could be related to the route of sensitization. The responses of individual patients to profilins were also similar and independent of the sensitizing source. The inhibition between pairs of allergens was generally higher than 70%, indicating that profilins share most of the IgE epitopes. Modeling of mimotopes in the conformational structure of the implicated profilins supports their strong cross-reactivity obtained experimentally., Conclusions: No correlation exists between the level of IgE response of individual patients to specific profilins and the corresponding theoretical sensitizing source, suggesting that the sensitization could be attributable to any profilin present in the environment of the patients. - Abstract
Background: Lipid transfer proteins (LTPs) are clinically relevant plant food panallergens and have been proposed as ideal tools to study true food allergy. Pru p 3, the major peach allergen in the Mediterranean area, is among the best-characterized allergenic members of the LTP family. Its diagnostic value for Rosaceae fruit allergy has been demonstrated both in vivo and in vitro., Objective: We sought to locate major IgE-binding epitopes of Pru p 3., Methods: A serum pool and individual sera from patients with peach allergy and positive skin prick test results to Pru p 3 were used. Three-dimensional modeling was achieved by using experimentally available structures of Pru p 3 homologues as templates. Theoretical prediction of potential IgE-binding regions was performed by selecting specific residues on the molecular surface displaying prominent electrostatic potential features. Point mutants of Pru p 3 were constructed by standard polymerase chain reaction procedures with the appropriate primers. Mutants were expressed in P pastoris by means of the pPIC 9 vector and purified from the corresponding supernatants by gel-filtration chromatography followed by RP-HPLC. IgE binding by Pru p 3 mutants was tested by immunodetection and quantified by ELISA and ELISA inhibition assays. Synthetic peptides (10 mer; 5 amino acids overlapping) covering the full Pru p 3 sequence were used to detect IgE epitopes by (125)I-anti-IgE immunodetection., Results: Pru p 3 showed a 3-dimensional structure comprising 4 alpha-helixes and a nonstructured C-terminal coil (residues 73 to 91). Regions around amino acids in positions 23 to 36, 39 to 44, and 80 to 91, particularly residues R39, T40, and R44, K80 and K91, were predicted as potential antibody recognition sites according to their relevant surface and electrostatic properties. Point mutants K80A and K91A were found to have an IgE-binding capacity similar to that of recombinant Pru p 3, but the triple mutant R39A/T40A/R44A showed a substantial decrease (approximately 5 times) of IgE binding. IgE immunodetection of synthetic peptides led to the identification of Pru p 3 sequence regions 11 to 25, 31 to 45, and 71 to 80 as major IgE epitopes., Conclusions: Main IgE-binding regions of the Pru p 3 amino acid sequence were identified. The three major ones comprised the end of an alpha-helix and some residues of the following interhelix loop. These data can help to search for Pru p 3 hypoallergenic forms.
