A highly efficient denitrifying bacterium aHD7 was screened from activated sludge. After static culture at 30 °C for 3 days, the denitrification rate of the aHD7 reached 91.7%, and during denitrification, nitrite had lower accumulation, with its concentration basically maintained at 1.8 μmg · L-1. The microscopy observation demonstrated that the aHD7 was a gram-negative bacillus, with an average size of 0.5 μm×(1.5-2.5) μm. Based on its biochemical/morphological characteristics and homologic analysis of 16S rDNA sequence, the aHD7 was identified as Pseudomonas mendocina. The investigation on the factors affecting the denitrification capacity of aHD7 showed that at the initial concentration of nitrate nitrogen being less than 276.95 mg · L-1, the denitrification rate was almost 100%, and when the initial concentration of nitrate nitrogen was as high as 553.59 μmg · L-1, the denitrification rate could reach 66.8%, with little nitrite accumulated. Ethanol was the most suitable carbon source. C/N ratio 6-8 and pH value 6-9 benefited the denitrification. Abstract: Reactions of the title complex (1), which features the rigid doubly-bridged biscyclopentadienyl ligand and the unusually long Mo–Mo bond in the structure, with a variety of classic substrates are explored. Complex 1 reacts with I2 in CH2Cl2 to give the expected Mo–Mo cleaved product [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)6I2 (2A) with a Mo–Mo distance of 5.329Å. While the same reaction in benzene forms the product 2B, which has the same structural formula as 2A but a different conformation, in which the Mo–Mo distance is 6.442Å. Reaction of 1 with Ph2E2 (E=S, Se) in refluxing toluene or under UV irradiation yields products with the carbonyls completely removed, [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(EPh)2(μ-EPh)2 (3a–b). Reaction of 1 with [Et2NC(S)S]2 results in the di-substituted product [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)4(Et2NC(S)S)2 (4). Reaction of 1 with C2Ph2 affords the acetylene-bridged derivative [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)4(μ-C2Ph2) (5). Reaction of 1 with an excess of PPh3 in refluxing toluene only provides the mono-substituted derivative [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)5(PPh3) (6), while the reaction of 1 with excess P(OPh)3 under the same conditions produces not only the mono-substituted derivative [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)5(P(OPh)3) (7), but also the di-substituted derivative [(η 5-C5H3)2(CMe2)(SiMe2)]Mo2(CO)4(P(OPh)3)2 (8). The reactivity and properties of the doubly-bridged molybdenum complex 1 and the corresponding non-bridged and singly-bridged analogs are compared. Reactions of phenylethynyl lithium with substituted cyclopentenones gave the corresponding pendant phenylethynyl substituted cyclopentadienes. Subsequent deprotonation and transmetallation with TiCl4·2THF, ZrCl4, and Cp∗ZrCl3 yielded the alkyne-functionalized metallocene complexes [C5Me4(C≡CPh)]2MCl2 [M=Ti (1), Zr (2)], Cp∗[C5Me4(C≡CPh)]ZrCl2 (3), and Cp∗[C5H2R′2(C≡CPh)]ZrCl2 [R′=Me (4), Ph (5)]. These complexes were fully characterized by 1H NMR, 13C NMR, MS spectra, and elemental analysis. The molecular structure of 2 was determined by single crystal X-ray diffraction analysis. Density-functional theory based on first principles is used to investigate oxidation of a semiconducting graphene nanoribbon with armchair edges (9-AGNR) by oxygen. The calculated results demonstrate that the oxygen is favorably physisorbed on the inner of 9-AGNR, while the oxygen is chemisorbed at the edge of 9-AGNR. Compared to the oxygen chemisorbed (cyclo- addition) 9-AGNR, two epoxy groups formed from cyclo-additions at the edge of 9-AGNR is energetically preferred. It is also found that the uniaxial strain generated by the cooperative alignment of two epoxy groups can significantly change the band gaps of 9-AGNR, leading to a change in the band gap for 9-AGNR as the concentration of epoxy groups varies. The pronounced change in the electronic properties, in particular, the band gaps, of 9-AGNR, as oxygen is adsorbed on 9-AGNR, may indicate that a semiconducting AGNR could have a potential application as a chemical (oxygen) sensor. Aim: To discriminate 32 phenethyl-amines between antagonists and agonists, and predict the activities of these compounds. Methods: The support vector machine (SVM) is employed to investigate the structure-activity relationship (SAR)/quantitative structure-activity relationship (QSAR) of phenethyl-amines based on molecular descriptors. Results: By using the leave-one-out cross-validation (LOOCV) test, 1 optimal SAR and 2 optimal QSAR models for agonists and antagonists were attained. The accuracy of prediction for the classification of phenethyl-amines by using the LOOCV test is 91.67%, and the accuracy of prediction for teh classification of phenethyl-amines by using the independent test is 100%; the results are better than those of the Fisher, the artificial neural network (ANN), and the K-nearest neighbor models for this real world data. The RMSE (root mean square error) of antagonists' QSAR model is 0.5881, and the RMSE of agonists' QSAR model is 0.4779, which are better than those of the multiple linear regression, partial least squares, and ANN models for this real world data. In this work, support vector classification (SVC) algorithm was used to build structure-activity relationship (SAR) model of the 5-hydroxytryptamine type 3 (5-HT3) receptor antagonists with 26 compounds. In a benchmark test, SVC was compared with several techniques of machine learning currently used in the field. The prediction performance of the model was discussed on the basis of the leave-one-out cross-validation. The results show that the accuracy of prediction of SVC model was higher than those of back propagation artificial neural network (BP ANN), K-nearest neighbor (KNN) and Fisher methods. Ti-based bulk metallic glasses (Ti4oZr25Cu9Ni8Be18)100-xNbx with x = 0 to 5at.% are prepared by copper-mold casting. The glass formation ability is almost unchanged by addition of Nb. The compression plasticity is, however, apparently changed, from 3% at x = 0 to 13% at x = 3, about 330% increases at the strain rate of 1 × 10[?]4s[?]1. The increment of the plasticity can be attributed to the segregation of Nb in the area of shear bands during the compression processing. Two-dimensional (2D) organic nanomaterials are fascinating because of their unique properties and pentential applications in future optoelectronic devices. Polyaniline (PANI) has attracted much attention for its high conductivity, good environmental stability and unusual doping chemistry. We report on liquid-phase exfoliation of layered PANI films grown by electrochemical polymerization. Atomic force microscopy images demonstrate that few- or even mono-layer PANI nanosheets can be fabricated. The PANI nanosheets can be transferred onto a variety of surfaces, providing a promising route to their incorporation into a variety of devices for further studies and various applications. To explore the coupling of dry-wet seasonal variations of soil respiration with their environmental factors in the alpine meadow under the background of increasing nitrogen (N) deposition, we conducted an experiment in the typical degraded Poa pratensis meadow in the Napahai, Yunnan. There were four treatments, i.e ., control (0 g·m -2 ·a -1 ), low (5 g·m -2 ·a -1 ), medium (10 g·m -2 ·a -1 ), and high (15 g·m -2 ·a -1 ) levels. We examined the effects of aboveground biomass, plant diversity, and soil physicochemical properties on soil respiration. The results showed that N deposition significantly promoted soil respiration. Compared with that in the control, soil respiration rates increased by 21.9%-53.9% and 27.3%-51.2% in dry and wet seasons, respectively. The maximum value of soil respiration rate was recorded in the medium N treatment. N deposition dramatically elevated aboveground biomass (52.2%-66.4%). Plant diversity declined with increasing N addition levels, with the maximum value (13.5%-24.2%) being recorded in high treatment in wet season. The values of ammonium nitrogen, organic matter, microbial biomass carbon and nitrogen, temperature and moisture in the three N treatments were elevated by 14.3%-333.5% compared with the control, while those of soil pH were decreased by 9.0%-34.6%. Results of the structural equation modelling showed that plant biomass, Shannon diversity, microbial biomass, soil temperature, and moisture showed a positive effect on soil respiration, while bulk density had a negative effect. Soil nitrogen pool and pH were main factors driving soil CO 2 emissions, accounting for 55.7% and 45.1% of the variations, respectively. Therefore, short-term atmospheric N deposition stimulated soil respiration primarily via altering soil pH and nitrogen pool components in the degraded alpine meadow.

A previously reported o-nitrobenzaldehyde (ONBA) degrading bacterium Pseudomonas sp. ONBA-17 was further identified and characterized. Based on results of DNA base composition and DNA-DNA hybridization, the strain was identified as P. putida. Its degradation effect enhanced with increase of inoculum amount and no lag phase was observed. Higher removal rate was achieved under shaking conditions. All tested ONBA with different initial concentrations could be completely degraded within 5 d. In addition, degradative enzyme(s) involved was confirmed as intra-cellular distributed and constitutively expressed. Effects of different compounds on relative activity of degradative enzyme(s) within cell-free extract were also evaluated. Finally, 2-nitrobenzoic acid and 2, 3-dihydroxybenzoic acid were detected as metabolites of ONBA degradation by P. putida ONBA-17, and relevant metabolic pathway was preliminary proposed. This study might help with future research in better understanding of nitroaromatics biodegradation.

Aim: To summarize our experience in the application of Crurasoft® for antireflux surgery and hiatal hernia (HH) repair and to introduce the work of Chinese doctors on this topic., Methods: Twenty-one patients underwent HH repair with Crurasoft® reinforcement. Gastroesophageal reflux disease (GERD) and HH-related symptoms including heartburn, regurgitation, chest pain, dysphagia, and abdominal pain were evaluated preoperatively and 6 mo postoperatively. A patient survey was conducted by phone by one of the authors. Patients were asked about "recurrent reflux or heartburn" and "dysphagia". An internet-based Chinese literature search in this field was also performed. Data extracted from each study included: number of patients treated, hernia size, hiatorrhaphy, antireflux surgery, follow-up period, recurrence rate, and complications (especially dysphagia)., Results: There were 8 type I, 10 type II and 3 type III HHs in this group. Mean operative time was 119.29 min (range 80-175 min). Intraoperatively, length and width of the hiatal orifice were measured, (4.33 ± 0.84 and 2.85 ± 0.85 cm, respectively). Thirteen and eight Nissen and Toupet fundoplications were performed, respectively. The intraoperative complication rate was 9.52%. Despite dysphagia, GERD-related symptoms improved significantly compared with those before surgery. The recurrence rate was 0% during the 6-mo follow-up period, and long-term follow-up disclosed a recurrence rate of 4.76% with a mean period of 16.28 mo. Eight patients developed new-onset dysphagia. The Chinese literature review identified 12 papers with 213 patients. The overall recurrence rate was 1.88%. There was no esophageal erosion and the rate of dysphagia ranged from 0% to 24%., Conclusion: The use of Crurasoft® mesh for HH repair results in satisfactory symptom control with a low recurrence rate. Postoperative dysphagia continues to be an issue, and requires more research to reduce its incidence.

A fenvalerate-degrading bacterial strain F-7 was isolated from long-term contaminated sludge. Based on morphological, physiological and biochemical characterization, and phylogenetic analysis of 16S rRNA gene sequence, strain F-7 was identified as Sphingomonas sp. The bacterium could utilize fenvalerate as the sole source of carbon. An amount measuring 100 mg L(-1) fenvalerate was completely degraded within 72 h and 3-phenoxybenzoic acid (3-PBA) was detected as a major metabolite. The result indicates that S. sp. F-7 might metabolize fenvalerate by hydrolysis of carboxylester linkage. It was capable of degrading permethrin, fenpropathrin, beta-cypermethrin, cyhalothrin, deltamethrin, bifenthrin and 3-PBA. Further studies demonstrated that the strain was multi-resistant to heavy metals and antibiotics. In addition, degradative enzymes involved were confirmed as intracellular distributed and constitutively expressed. Furthermore, application of the strain was found to accelerate the removal of fenvalerate in soil. This is the first report of fenvalerate degrading strain isolated from S. sp. These results might help with future research in better understanding of pyrethroid biodegradation and highlight S. sp. F-7 might have potential for practical application in bioremediation of fenvalerate-contaminated sites.

Objective: To study the inducing effect of trichosanthin on the apoptosis of mouse prostatic cancer RM-1 cells and its mechanism., Methods: MTT assay was used to identify the effect of trichosanthin on RM-1 cells in vitro. The cells apoptosis was detected by Hoechest 33258 fluorescent staining and flow cytometry. The levels of bax proteins were detected by western blot analysis as well as their apoptosis rate., Results: The IC50 value of trichosanthin for RM-1 cell was 117.32 mg/L. Cell cycle was analysised by flow cytometry, trichosanthin induced an arrest in G0/G1 phase. Hoechest 33258 fluorescent staining showed typical nucleolus changes in apoptosis cells. Expression of bax was up-regulated at protein levels in a time-dependent manner., Conclusion: Trichosanthin can induce apoptosis in RM-1 cells. The induction of apoptosis is a very important mechanism of trichosanthin to inhibit prostatic cancer.

Objective: To investigate the expression and clinical significance of cyclin H and cyclin-dependent kinase 7 (CDK7) in human hemangiomas., Methods: Immunohistochemistry technique was used to measure the expression of cyclin H and CDK7 proteins in proliferative, involuting hemangiomas and normal skin tissues. Immunohistochemical technique for factor VIII-related antigen was used to prove that the cells which expressed cyclin H and CDK7 were endothelial cells. Average optical density and positive area of the expression of cyclin H and CDK7 proteins in proliferative, involuting hemangiomas and normal skin tissues were measured by image analysis (HPIAS-1000)., Results: The expression of cyclin H and CDK7 protein in proliferating hemangiomas was significantly higher than that in involuting hemangiomas and normal skin tissues (P < 0.01). But no significant difference was found in the expression of cyclin H and CDK7 protein between involuting hemangiomas and normal skin tissues (P > 0.05)., Conclusions: cyclin H and CDK7 may play an important role in the generation and development of human hemangiomas.

Structure propensities of amino acids are important determinants in guiding proteins' local and global structure formation. We constructed a phage display library--a hexa-HIS tag upstream of a CXXC (X stands for any of the 20 natural amino acids) motif appending N-terminal to the minor capsid protein pIII of M13KE filamentous phage--and developed a novel directed-evolution procedure to select for amino acid sequences forming increasingly stable beta-turns in the disulfide-bridged CXXC motif. The sequences that emerged from the directed-evolution cycles were in good agreement with type II beta-turn propensities derived from surveys of known protein structures, in particular, Pro-Gly forming a type II beta-turn. The agreement strongly supported the notion that beta-turn formation plays an active role in initiating local structure folding in proteins.

On the bases of exploring measurement mechanism and complex compound components of indirect determination of cysteine acid by flame atomic absorption spectrometry with zinc sulfide, the stability constants of the complex compound of zinc cysteinate were studied successfully, after the numeric measurements and theoretical investigations under the best optimized acidity for the determination. In conclusion, at the best optimized acidity pH 9.7, the stability constant of alkalescence zinc cysteinate is between 2.998 x 10(30) and 3.858 x 10(30), with the average value beta w = 3.286 x 10(30) and log beta w = 30.517, and the standard deviation is 0.315 x 10(30). While the stability constant of zinc cysteinate is between 7.853 x 10(21) and 1.011 x 10(22), with the average value beta w = 8.607 x 10(21) and log beta w = 21.935, and the standard deviation is 0.315 x 10(21). It is shown that the atomic absorption spectrometry can not only be used for the determination of trace elements and the indirect determination of organic compound, but also can carry out the study of physical configuration and the determination of physical constant of complex ion.

To investigate the modulatory action of endogenous and exogenous nitric oxide (NO) on survival of alveolar macrophages (AMs) in different cellular states, AMs from normal rats (normal AMs) and from bleomycin (BLM)-treated rats (BLM AMs) were incubated by sodium nitroprusside (SNP, NO donor) and L-arginin (L-Arg, NO precursor), respectively. The survival of AMs was evaluated by apoptosis and cell cycles. The molecular mechanisms were investigated by the contents of Bcl-2, Bax proteins in AMs. The results are as follows: (1) The degree of BLM AMs apoptosis was higher than that of normal AMs; the number of BLM AMs in G(0)/G(1) phases was less than that of normal AMs; there was no significant difference in S+G(2)M phases between the number of BLM AMs and that of normal AMs. (2) Down-regulation of Bcl-2 and up-regulation of Bax occurred in BLM AMs, compared to those in normal AMs. (3) Apoptosis of AMs, either normal AMs or BLM AMs, was induced by both SNP and L-Arg, when compared to their respective control; only the number of BLM AMs in S+G(2)M phases was increased by L-Arg. (4) SNP and L-Arg induced a down-regulation of Bcl-2 and an up-regulation of Bax proteins in normal AMs, but did not induce the same change pattern in BLM AMs. (5) The Bax in BLM AMs was down-regulated by L-Arg. It is concluded that NO can induce the apoptosis of BLM AMs and normal AMs; that Bcl-2 and Bax are implicated in NO-induced apoptosis of normal AMs, whereas they are not involved in that of BLM AMs, suggesting the differential molecular mechanisms underlying the NO-induced apoptosis of normal AMs and BLM AMs; and that endogenous NO promotes proliferation of BLM AMs, which might be associated with down-regulation of Bax.

Objective: To explore the effects of the combined method of abdominal axial flap transposition and penile elongation for the treatment of the remnant penis., Methods: Fifty-two cases of the remnant penis treated with the combined method from 1984 April to February 2004 were analyzed retrospectively. Follow-up ranged from 0.5 to 20 years postoperatively., Results: The lengths (both in normal and erectile conditions) and the circumferences of the penis gained after operation were (5.6 +/- 1.4) cm, (6.8 +/- 2.5 cm and (6.9 +/- 2.3) cm respectively. The recovery rates of the sensory function were 94.2% and 100% in the glans (immediately and 3 months after operation) and 32.7%, 51.9% and 75% in the flap area (3, 6 and 12 months postoperatively). The two-point distinguishing sense in the glans and the flap area was (5.1 +/- 0.9) mm and(7.9 +/- 1.3) mm 5 years after operation. Early complications included distant flap necrosis (3 cases), disruption of the wound (2 cases), part necrosis of the skin graft in the abdominal wall (2 cases) and poor contours occurred in 4 cases in the later period because of the thickness of the flaps. All of them were corrected with satisfactory results., Conclusion: The combined method of abdominal axial flap transposition and penile elongation was recommendable for the treatment of the remnant penis because of its positive effects and less complications.

Objective: To investigate the expression of p73 and c-fos protein and its significance in the development of children hemangioma., Methods: The quantitative expressions of p73 and c-fos protein in hemangioma and normal skin were detected by immunohistochemistry., Results: The expressions of p73 and c-fos protein were strong in proliferative hemangioma while they were very weak in involutional hemangioma and normal skin. There were significant differences between the proliferative and involutional hemangioma or the normal skin in the expressions of p73 and c-fos (P < 0.01). No statistical significances of p73 or c-fos P73 expressions were observed between involutional hemangioma and normal skin (P > 0.05)., Conclusions: P73 and c-fos may play an important role in the development and involution of skin hemangioma.

Objective: To investigate the changes of nitric oxide concentration in the distal portion of a random pattern skin flap and the influence of the exogenous L-arginine on the survival of the random pattern skin flap., Methods: A random pattern skin flap (7 cm x 2 cm) was cranially designed and elevated on the back of a Wistar rat. An image analysis technology was used to evaluate the survival rate of the skin flap, while a biochemistry method was used to test the concentrations of the NO in the tissue., Results: The survival area of the flap in the L-arginine-treated group was significantly enlarged (63.83 +/- 5.13)% (P < 0.01) in seven days postoperatively, compared with the control group (43.26 +/- 2.86)%. The NO concentration in the tissue was no statistic difference between all of the groups immediately after the operation (P > 0.05). But, the NO concentration in the control was decreasing at the beginning and then increasing slightly to reach the high level in 12 hours after the operation. It was thereafter slumped down to the baseline in 72 hours after the surgery. Although the changes in the L-arginine-treated group were quite similar to the control excepting of the extent, the NO concentration was kept in a higher level in the sequential time after the operation (P < 0.01, P < 0.01, P < 0.05 and P < 0.01)., Conclusions: The NO concentration in skin flap tissue after the elevation was going up slightly for a short time. The exogenous L-arginine could promote the NO concentration in the random pattern skin flap to protect it from ischemic injury.

Aim: To observe the role of endogenous peroxynitrite (ONOO-) in pulmonary injury and fibrosis induced by bleomycin A5 (BLM-A5) in rats., Methods: Pulmonary injury and fibrosis of rats were evaluated by testing the level of lipid peroxides (LPO) in out-going pulmonary blood (OPB), and by observing histological changes, including type III and type I collagen changes in lung which were examined with Sirius red staining under polarized light. The peroxynitrite expression was detected by immunohistochemistry for nitrotyrosine (NT), a marker of the peroxynitrite production., Results: (1) The level of LPO was elevated in OPB of rats on d 14 after intratracheal administration of BLM-A5. Thickened alveolar wall and macrophage infiltration were seen, and fibroblasts were near by the interstitial macrophages. Increased amounts of type III collagen and type I collagen were deposited in disoriented fashion. (2) High expression of ONOO- was detected in alveolar epithelial cells and pulmonary interstitial macrophages. (3) The above changes were reduced by aminoguanidine (AG), an inhibitor of nitric oxide synthase (iNOS)., Conclusion: Endogenous ONOO- mediated BLM-A5-induced pulmonary toxicity. The therapeutic potential of AG for pulmonary injury and fibrosis was realized partly by reducing ONOO- formation.

Objectives: To investigate the tissue ultrastructure changes of small testis and sex hormone and their correlation., Methods: The patients were divided into small tests (n = 8) and control group(n = 12). FSH, LH, T were determined by radioimmunassay. Diameter and wall thickness of convoluted seminiferous tubule were investigated with light microscope and electro microscopy on small testis tissue morphology and ultrastructure., Results: FSH, LH, T of small testis and control group were (21.05 +/- 9.15) IU/L vs (6.74 +/- 3.52) IU/L, (22.88 +/- 6.25) IU/L vs (6.60 +/- 1.48) IU/L and (0.30 +/- 0.04) nmol/L vs (17.55 +/- 9.25) nmol/L, respectively. Seminiferous tubule diameter and wall thickness were(37.33 +/- 6.80) microns vs (198.46 +/- 29.84) microns and (10.30 +/- 1.82) microns vs (2.95 +/- 0.20) microns. Small testis tissue ultrastructure changed significantly., Conclusions: Pathologic changes of small testis tissue in many parts such as seminiferous tubule, germinal epithelium, Sertoli cell, Leydig cell, limiting membrance and blood vessel may relate with genetics and immunoreaction.
