Altering the food intake, exercise, and sleep patterns have a great influence on the homeostasis of the biological clock. This leads to accelerated aging of the articular cartilage, susceptibility to arthropathy and other aspects. Deficiency or overexpression of certain circadian clock‐related genes accelerates the cartilage deterioration and leads to phenotypic variation in different joints. The process of joint cartilage development includes the formation of joint site, interzone, joint cavitation, epiphyseal ossification center, and cartilage maturation. The mechanism by which, biological clock regulates the cell‐cycle, growth, metabolism, and other biological processes of chondrocytes is poorly understood. Here, we summarized the interaction between biological clock proteins and developmental pathways in chondrogenesis and provided the evidence from other tissues that further predicts the molecular patterns of these protein‐protein networks in activation, proliferation, and differentiation. Fibrosis‐related diseases carry with them a high mortality rate and their morbidity increases with age. Recent findings indicate that induced senescence in myofibroblasts can limit or reduce myocardial fibrosis, cirrhosis, and idiopathic pulmonary fibrosis, while also accelerating wound healing. However, more senescent cells are accumulated as organisms age, which exacerbates aging‐related diseases. These two contradictory theories inspired us to summarize papers on the restrictive effect of senescence on fibrosis and to input the key findings into simple software that we developed to assist with data organization and presentation. In this review, we illustrate that senescent cells secrete more matrix metalloproteinases to solubilize excess collagen, while chemokines and cytokines activate immune cells to eliminate senescent cells. In the elderly, it is perhaps more effective to limit fibrosis by inducing myofibroblast senescence and then removing senescent cells that are not cleared via normal mechanisms by antisenescence therapies. Aim Of Study: The survival rate in high-grade glioma (HGG) patients receiving a combined regimen of radiotherapy (RT) and temozolomide after tumor resection was increased. However, cognitive deficits and depression after the treatments challenge the treatments. The aim of this study was to evaluate the cognition and depression in postoperative patients with HGG following RT and chemotherapy.Materials and Methods: Six-five eligible patients were included in the study. Cognition and depression were examined at baseline (after surgery before RT), every 3 months during follow-up using mini-mental state examination and Zung Self-Rating Depression Scale (SDS), respectively.Results: Our results showed that cognition was not significantly affected after treatments (F = 1.19, P = 0.32). However, significant differences between baseline and follow-ups were found regarding SDS scores (F = 3.26, P =0.0.01). SDS score at the 3rd month was significantly higher than that at baseline (t = -3.16, P = 0.002).Conclusion: This prospective study showed that although cognition was not significantly affected, the treatment caused depression, particularly at the 3rd month. This paper builds a novel framework for the cordon-based road reservation system to manage road traffic in urban areas, and the rules of reserving the right of way are established. Both the simulation experiment and the analytical modeling approaches offer evidence that the reservation system performs better than the basic system and some types of pricing systems. Considering two different types of vehicles (i.e., reserved and unreserved vehicles) and a general link cost function, we conduct three types of experiment simulations, including the basic system, the reservation system, and the pricing system. Although the severity of traffic congestion is mitigated inside the cordon both in the reservation and pricing systems, the reservation system shows the best network performance from the traffic indicators provided by the simulation software. In the analytical modeling approach, the Frank–Wolfe​ algorithm with a modified Bureau of Public Road function is used to analyze the road reservation scheme, and results agree with the expected effectiveness of the reservation both in the aspect of the total system travel time and the equilibrium of the traffic flow in the road network. In the United States, several land use and land cover (LULC) data sets are available based on satellite data, but these data sets often fail to accurately represent features on the ground. Alternatively, detailed mapping of heterogeneous landscapes for informed decision-making is possible using high spatial resolution orthoimagery from the National Agricultural Imagery Program (NAIP). However, large-area mapping at this resolution remains challenging due to radiometric differences among scenes, landscape heterogeneity, and computational limitations. Various machine learning (ML) techniques have shown promise in improving LULC maps. The primary purposes of this study were to evaluate bagging (Random Forest, RF), boosting (Gradient Boosting Machines [GBM] and extreme gradient boosting [XGB]), and stacking ensemble ML models. We used these techniques on a time series of Sentinel 2A data and NAIP orthoimagery to create a LULC map of a portion of Irion and Tom Green counties in Texas (USA). We created several spectral indices, structural variables, and geometry-based variables, reducing the dimensionality of features generated on Sentinel and NAIP data. We then compared accuracy based on random cross-validation without accounting for spatial autocorrelation and target-oriented cross-validation accounting for spatial structures of the training data set. Comparison of random and target-oriented cross-validation results showed that autocorrelation in the training data offered overestimation ranging from 2% to 3.5%. The XGB-boosted stacking ensemble on-base learners (RF, XGB, and GBM) improved model performance over individual base learners. We show that meta-learners are just as sensitive to overfitting as base models, as these algorithms are not designed to account for spatial information. Finally, we show that the fusion of Sentinel 2A data with NAIP data improves land use/land cover classification using geographic object-based image analysis. [ABSTRACT FROM AUTHOR]

Rotavirus (RV) is the main pathogen that causes severe diarrhea in infants and children under 5 years of age. No specific antiviral therapies or licensed anti-rotavirus drugs are available. It is crucial to develop effective and low-toxicity anti-rotavirus small-molecule drugs that act on novel host targets. In this study, a new anti-rotavirus compound was selected by ELISA, and cell activity was detected from 453 small-molecule compounds. The anti-RV effects and underlying mechanisms of the screened compounds were explored. In vitro experimental results showed that the small-molecule compound ML241 has a good effect on inhibiting rotavirus proliferation and has low cytotoxicity during the virus adsorption, cell entry, and replication stages. In addition to its in vitro effects, ML241 also exerted anti-RV effects in a suckling mouse model. Transcriptome sequencing was performed after adding ML241 to cells infected with RV. The results showed that ML241 inhibited the phosphorylation of ERK1/2 in the MAPK signaling pathway, thereby inhibiting IκBα, activating the NF-κB signaling pathway, and playing an anti-RV role. These results provide an experimental basis for specific anti-RV small-molecule compounds or compound combinations, which is beneficial for the development of anti-RV drugs. [ABSTRACT FROM AUTHOR]

Cerebrospinal fluid (CSF) flow patterns and their relationship with arterial pulsation can depict the function of glymphatic system (GS). We propose an improved multi-directional diffusion-sensitized driven-equilibrium (iMDDSDE) prepared heavily T2-weighted 3D FSE (iMDDSDE-HT2) magnetic resonance imaging (MRI) method to noninvasively assess the mobility (MO) of CSF distributed in the ventricles and perivascular spaces (PVS). This method could obtain 3D high resolution (1 mm isotropic) imaging of CSF MO with full brain coverage within five min and distinguish the CSF MO across different pulse phases using a peripheral pulse unit (PPU). The MO curves had the largest amplitude value in the PVS of middle cerebral artery (11.11 × 10−9 m2/s) and the largest amplitude growth rate in the posterior cerebral artery (189%). The average coefficient of variations (CVs) in non-pulse trigger and pulse phase 1 and 3 were 0.11, 0.10 and 0.09 respectively. The MO in older healthy participants was lower compared to the young participants, and the MO in cerebral major artery stenosis patients with acute ischemia stroke (AIS) were lower compared to those without AIS in several ventriclar ROIs (P < 0.05). A novel empirical mode decomposition method was adopted to investigate the dissociative or interactive neural impact of depression and motor impairments in Parkinson's disease (PD). Resting-state fMRI data of 59 PD subjects were first decomposed into characteristic frequency bands, and the main effects of motor severity and depression and their interaction on the energy of blood-oxygen-level-dependent signal oscillation in specific frequency bands were then evaluated. The results show that the severity of motor symptoms is negatively correlated with the energy in the frequency band of 0.10-0.25 Hz in the bilateral thalamus, but positively correlated with 0.01-0.027 Hz band energy in the bilateral postcentral gyrus. The severity of depression, on the other hand, is positively correlated with the energy of 0.10-0.25 Hz but negatively with 0.01-0.027 Hz in the bilateral subgenual gyrus. Notably, the interaction between motor and depressive symptoms is negatively correlated with the energy of 0.10-0.25 Hz in the substantia nigra, hippocampus, inferior orbitofrontal cortex, and temporoparietal junction, but positively correlated with 0.02-0.05 Hz in the same regions. These findings indicate unique associations of fMRI band signals with motor and depressive symptoms in PD in specific brain regions, which may underscore the neural impact of the comorbidity and the differentiation between the two PD-related disorders. To explore the stability and repeatability of diffusion-weighted imaging (DWI) of normal pancreas with different field of views (FOV) on 5.0 T magnetic resonance imaging (MRI) system. Twenty healthy subjects underwent two sessions of large FOV (lFOV) and reduced FOV (rFOV) DWI sequence scanning. Two radiologists measured the apparent diffusion coefficient (ADC) values and the signal-to-noise ratio (SNR) of the pancreatic head, body, and tail on DWI images, simultaneously, using a 5-point scale, evaluate the artifacts and image quality. One radiologist re-measured the ADC on DWI images again after a 4-week interval. The test-retest repeatability of two scan sessions were also evaluated. Intra-observer and inter-observer at lFOV and rFOV, the ADC values were not significantly different (P > 0.05), intraclass correlation coefficients (ICCs) and coefficient of variations were excellence (ICCs 0.85–0.99, CVs < 8.0%). The ADC values were lower with rFOV than lFOV DWI for the head, body, tail, and overall pancreas. The consistency of the two scan sessions were high. The high stability and repeatability of pancreas DWI has been confirmed at 5.0 T. Scan durations are reduced while resolution and image quality are improved with rFOV DWI, which is more preferable than lFOV for routine pancreas imaging. [ABSTRACT FROM AUTHOR]

The longitudinal movement of the expansion joint affects the wear of the bearing and the fatigue damage of the expansion joint itself. A long-term monitoring on a long-span suspension bridge has been set up to reveal the mechanism and the response characteristic of the longitudinal movement of the expansion joint. This paper presents the field measurement results and analysis of the longitudinal movement. The results show that the temperature variation results in the fluctuation of the longitudinal movement of the expansion joint. Whereas, the high-frequency components are dominated by the vehicle and ambient excitations. Moreover, the RMS movement and the temperature show a significant linear relationship. Then, the frequency-domain analysis is performed to investigate the spectral characteristic of the longitudinal movement. It is shown that the quasi-static effect of the vehicle governs the longitudinal movement at 10-min interval, and the dynamic effect of the vehicle plays an important role in the cumulative movement of the expansion joint. At last, the sway vibration of the girder is discussed through the finite element model of the suspension bridge, and the effects of the central buckle on the longitudinal movement of the expansion joint are investigated. CF101 (IB-MECA) is an adenosine A3 receptor agonist that has anti-inflammatory and pain-relieving properties. Adenosine A3 receptor activation can delay the process of Osteoarthritis(OA) and prevent the occurrence of OA. However, the mechanism of CF101 on OA is still unknown. This study aimed to investigate the effect of CF101 on rats induced by anterior cruciate ligament-transection (ACLT) and rat chondrocytes induced by IL-1ß. ACLT-induced OA rats were administered CF101, and autophagy levels were measured to determine whether CF101 had an autophagy-mediated protective effect on articular cartilage. Furthermore, the mechanism by which CF101 protected articular cartilage in IL-1ß-induced chondrocytes mimicking OA was investigated. In rats treated with ACLT, CF101 was able to delay the progression of OA, as well as reduce inflammation and type II collagen degradation factors. In addition, in vitro experiments revealed that CF101 reduced type II collagen degradation factors in OA chondrocytes. In rats treated with ACLT and OA chondrocytes, CF101 enhanced autophagy and increased the ratio of AMP/ATP and AMPK protein levels while decreasing mTOR expression. Treatment of OA chondrocytes with 3-MA prior to treatment with CF101 resulted in inhibition of autophagy factor levels, as well as increased levels of inflammatory factors and type II collagen degradation compared to the CF101 group. These findings demonstrated that CF101 could protect articular cartilage against OA by enhancing the ratio of ATP/AMP and altering the AMPK/mTOR pathway to enhance autophagy and reduce inflammation. • A strategy for rationally selecting solvent systems of spherical agglomeration. • Wettability is emphasized to be the key role in spherical agglomeration. • Wetting and adhesion were studied by the Lifshitz-van der Waals acid-base approach. • New solvent systems were selected by the strategy for diverse substances. A strategy for rationally selecting solvent systems of spherical agglomeration is proposed for the first time. It emphasizes that followed by crystallization and phase separation, spherical agglomeration can be achieved only when the solvent system can induce wetting and adhesion which are estimated by the Lifshitz-van der Waals acid-base approach. Especially for wettability of bridging liquid, solvent and anti-solvent, the strategy indicates that it is of great importance for a successful solvent system. Valid systems therefore are selected in the first place while invalid ones are removed effectively according to the wettability difference. Based on the strategy, valid solvent systems for cefotaxime sodium and benzoic acid spherical agglomeration were selected successfully from 720 and 2184 systems respectively without missing the reported systems. The strategy also predicted no valid systems for potassium chloride, maltitol and trisodium phosphate in the given solvent combinations, which agreed with the experiment results. Objective: To assess the performance of hybrid multi-dimensional magnetic resonance imaging (HM-MRI) in quantifying hematoxylin and eosin (H&E) staining results, grading and predicting isocitrate dehydrogenase (IDH) mutation status of gliomas., Materials and Methods: Included were 71 glioma patients (mean age, 50.17 ± 13.38 years; 35 men). HM-MRI images were collected at five different echo times (80-200 ms) with seven b-values (0-3000 s/mm 2 ). A modified three-compartment model with very-slow, slow and fast diffusion components was applied to calculate HM-MRI metrics, including fractions, diffusion coefficients and T2 values of each component. Pearson correlation analysis was performed between HM-MRI derived fractions and H&E staining derived percentages. HM-MRI metrics were compared between high-grade and low-grade gliomas, and between IDH-wild and IDH-mutant gliomas. Using receiver operational characteristic (ROC) analysis, the diagnostic performance of HM-MRI in grading and genotyping was compared with mono-exponential models., Results: HM-MRI metrics F Dvery-slow and F Dslow demonstrated a significant correlation with the H&E staining results (p < .05). Besides, F Dvery-slow showed the highest area under ROC curve (AUC = 0.854) for grading, while Dslow showed the highest AUC (0.845) for genotyping. Furthermore, a combination of HM-MRI metrics F Dvery-slow and T2 Dslow improved the diagnostic performance for grading (AUC = 0.876)., Discussion: HM-MRI can aid in non-invasive diagnosis of gliomas., (© 2024.
